Abstract

L'invention se rapporte à des procédés servant à diminuer les fixations non spécifiques à des billes dans des essais basés sur une double bille ainsi qu'à des biodisques optiques apparentés et des systèmes d'unité de disque. Ces procédés consistent à identifier la présence d'un agent ciblé dans un spécimen biologique, et à mélanger des billes de capture auxquelles est fixée au moins une sonde de transport, des billes reporteurs possédant chacune au moins une sonde de signalisation et un spécimen biologique. On exécute ce mélange dans des conditions de fixation rendant possible la formation d'un complexe à double bille si l'agent ciblé est présent dans le spécimen. La bille reporteur et la bille de capture sont fixées chacune à l'agent ciblé. L'invention concerne également la dénaturation de cet agent ciblé et sa conservation sous sa forme dénaturée au moyen d'un tampon d'hybridation spécial. Un agent de dénaturation est l' isothiocyanate de guanidine. Les procédés consistent, de plus, à isoler ce complexe à double bille du mélange afin d'obtenir un isolat, à exposer cet isolat à un champ de capture sur un disque et à détecter la présence du complexe à double bille dans le disque, ce qui indique la présence de l'agent ciblé dans le spécimen. Ces procédés peuvent également consister à disloquer de façon sélective la fixation non spécifique entre des billes de capture et des billes reporteurs au moyen d'un agent de digestion. L'invention met également en application un procédé servant à disloquer la fixation non spécifique entre des billes de capture et des billes reporteurs au moyen d'un tampon de lavage contenant un agent chimique. On applique ces procédés à la détection de cibles médicales.
