Abstract

In der Erfindung wird rekombinante Proteinase K beschrieben. Weiterhin wird ein Verfahren zur Herstellung von rekombinanter Proteinase K offengelegt, welches gekennzeichnet ist durch: a) Transformation einer Wirtszelle mit einer rekombinanten Nukleinsäure, welche für die zymogene Vorstufe der Proteinase K codiert, b) Kultivierung der Wirtszelle in einer Weise, daß die zymogene Vorstufe von Proteinase K in Form von 'inclusion bodies' in der Wirtszelle entsteht, c) Isolierung der 'inclusion bodies' und Naturierung unter Bedingungen, bei denen der Proteaseteil der zymogenen Vorstufe in ihrer natürlichen Konformation entsteht, d) Aktivierung und Aufreinigung der naturierten Proteinase K.
