本発明は、 力ルポニル基含有化合物を還元して、 医薬、 農薬等の中間体原料 として産業上有用な化合物である光学活性アルコール類に変換する活性を有す るポリペプチド、 該ポリペプチドをコードする D N A、 該 D N Aをベクターに '組み込んで得られる組換え体 D N A、 該組換え体 D N Aを保有する形質転換体 に関し、 さらに該形質転換体、 該形質転換体培養物または該形質転換体処理物 を用いた光学活性アルコール類の製造方法に関する。 背景技術
( E ) — 7— [ 2'—シクロプロピル一 4一 (4一フルォロ: ェ -ル) 一キノ リンー 3一ィル] 一 3 , 5—ジヒ ドロキシーヘプトー 6一ェン酸エステ/レ類を 化学的に製造する方法としては、 例えば、 特許文献 1に以下に示す製造ルート が知られている。

また、 特許文献 2には、 光学活性なシッフ塩基を用いて製造する方法も報告 されている。
またさらに、 特許文献 3には、 (R ) _3_tert -ブチルジメチルシリルォキシ— 6ージメ トキシホスフィ二/レー 5ーォキソへキサン酸メチノレエステノレを用いて 極低温にて製造する方法も報告されている。
一方、 より安価にまた副生産物の少ない方法として、 微生物の菌体及ぴ ま たは該菌体処理物を用いて、 カルボエル基を有する化合物から立体選択的に還 元し、 光学活性なアルコール体を生成する方法を上記化合物の製造に応用する 方法が考えられるが、 カルボニル基の側鎖にキノリン骨格を有する化合物を用 いる方法として、 非特許文献 1には、 以下に示す反応をミクロバタテリゥム キャンホクエマドエンシス (Microbacterium campoquemadoensis) MB5614株を 用いて行えることが記載されている。

しかしながら、 (E ) - 7 - [ 2—シクロプロピル一 4— ( 4—フルオロフ ェニル) ーキノリンー 3—ィル] — 3, 5—ジォキソヘプトー 6—ェン酸エス テル類のように、 カルボ-ル基の α位にォレフィンが存在する上に、 カルボ二 ル基が連続して存在する化合物については、 微生物を用いて立体選択的に還元 できるという例は知られていなかった。

従って、 (3 R, 5 S) 一 (E) — 7— [2—シクロプロピル一 4一 (4一 フルオロフェニル) 一キノ リ ン一 3—ィル] 一 3, 5—ジヒ ドロキシヘプトー 6一ェン酸エステル類を工業的に安価に製造できる新規な製造方法を開発する ことが望まれていた。
本発明者らは、 上記課題を解決するために、 (3 R, 5 S) 一 (E) - 7 - [ 2—シク ロプロ ピノレー 4一 ( 4—フルォロフエ-ノレ) 一キノ リ ン一 3—ィ ル] 一 3, 5—ジヒ ドロキシヘプトー 6—ェン酸エステル類の製造方法につい て鋭意検討した結果、 原料となるカルボニル基含有化合物の還元反応を触媒す る新規酵素を単離し、 組換え菌を用いて発現させた該酵素を、 原料となるカル ボニル基含有化合物に作用させることにより、 高い光学純度かつ高濃度で目的 物を得ることができることを見い出し、 本発明を完成するに至った。
すなわち、 本発明の要旨は、
(1) 下記 (A) または (B) の何れかのアミノ酸配列を有するポリべプチ ド、。
(A) 配列番号 1に記載のアミノ酸配列。
(B) 配列番号 1に記載のアミノ酸配列において、 1から複数個のアミノ酸が 欠失、 付加または置換されているアミノ酸配列であって、 カルボ-ル還元酵素 活性を有するポリペプチドのアミノ酸配列。 ;
(2) 下記 (a) から (e) の何れかの塩基配列を有する DNA。
(a ) 配列番号 1に記載のアミノ酸配列からなるポリべプチドをコードする塩 基配列。
(b) 配列番号 1に記載のアミノ酸配列において、 1から複数個のアミノ酸が 欠失、 付加または置換されているアミノ酸配列からなるポリべプチドであって、 カルボ-ル還元酵素活性を有するポリぺプチドをコードする塩基配列。
( c ) 配列番号 2に記載の塩基配列。 (d) 配列番号 2に記載の塩基配列において、 1から複数個の塩基が欠失、 付 加または置換されている塩基配列であって、 カルボニル還元酵素活性を有する ポリぺプチドをコ一ドする塩基配列。

以下に、 本発明を詳細に説明する。
本発明のポリペプチドは、 配列番号 1に記載のアミノ酸配列を有するもの、 又はそのホモログであってカルボ-ル還元酵素活性を有するものである。
本明細書において、 力ルポ-ル還元酵素活性とは、 カルボニル基含有化合物 中のカルボュル基を不斉還元して光学活性なアルコール類とする活性をいう。 このような活性は、 力ルポュル基含有化合物を基質として含有し、 さらに N A D P Hを補酵素として含有する反応系において、 酵素として、 目的のポリぺプ チド、 形質転換体細胞、 形質転換体細胞培養物または形質転換体細胞処理物を 作用させて N A D P H減少初速度を測定することにより測定することができる。 本発明のポリぺプチドは、 本発明によってそのァミノ酸配列および該ァミノ 酸配列をコードする塩基配列が明らかになつたので、 後述するように本発明の ポリぺプチドのァミノ酸配列の一部又は全部をコードする塩基配列を元にして 作製したプローブを用いて、 カルボュル基の還元活性を有する任意の微生物か ら還元酵素をコードする D N Aを単離した後、 それを元に通常の遺伝子工学的 手法を用いて得ることができる。 また、 本発明を完成するにあたってなされた ように、 カルボニル基の還元活性を有する微生物、 すなわち、 カルボニル還元 酵素をコードする D N Aを有する微生物、 例えば、 ォガタエァ属酵母の培養物 より精製することも出来る。
ォガタエァ属酵母としては、 例えば、 ォガタエア ミヌタ (Ogataea minut a) が特に本発明のカルボニル還元酵素の産生能に優れ、 この酵母は、 例えば I FO 1 4 7 3株として財団法人発酵研究所より入手する事が出来る。
微生物の培養物からの本発明のポリぺプチドの取得方法としては、 通常の酵 素の精製方法を用いることができ、 例えば、 以下の方法で行うことができる。 上記微生物を YM培地等の真菌の培養に用いられる一般的な培地で培養するこ とで十分に増殖させた後に回収し、 DTT (D i t h i o t h r e i t o l ) 等の還元剤や、 フエ-ルメタンスルホ-ルフルオリ ド (p h e n y l m e t h a n s u l f o n y l f l u o r i d e ; PMS F) の様なプロテアーゼ阻 害剤を加えた緩衝液中で破砕して無細胞抽出液とする。 無細胞抽出液から、 タ ンパク質の溶解度による分画 (有機溶媒による沈殿や硫安などによる塩析な ど) や、 陽イオン交換、 陰イオン交換、 ゲル濾過、 疎水クロマトグラフィー、 キレート、 色素、 抗体等を用いたアブイ二ティークロマトグラフィー等を適宜 組み合わせることにより精製することが出来る。 例えば、 DEAEセファロー スを用いた陰イオン交換クロマトグラフィー、 ォクチルセファロースを用いた 疎水ク口マトグラフィー、 Q—セファロースを用いた陰イオン交換クロマトグ ラフィー、 スーパーデッタス 200を用いたゲル濾過等を経て電気泳動的にほ ぼ単一バンドまで精製することが出来る。
このように精製されたォガタエア ミヌタ (Ogataea minuta) に由来する本 発明のポリべプチドは、 典型的には以下の性状を備えたものである。
(1) 至適 p H
5. 0 ~ 6. 0
(2) 分子量
ドデシル硫酸ナトリウム一ポリアクリルアミ ドゲル電気泳動 (以下、 S D S— PAGEと省略) による分子量測定で約 2 7, O O OD aである。
さらにこの酵素は以下の性状で特徴付ける事が出来る。
(3) 安定 p H範囲
P H 5. 5— 6. 5 の範囲で比較的安定である
(4) 作用適温の範囲 至適温度は 6 0 - 70°Cである。
(5) 温度安定性
40°Cまで比較的安定である。
(6) 阻害
本酵素は水銀 ( I ) イオン、 鉛 ( I I ) イオンにより阻害を受ける。
上記の方法で単離された本発明のポリぺプチドは、 単なるカルボエル基含有 化合物だけでなく、 ジカルボ-ル化合物も不斉還元する能力を有する優れた酵 素である。
本発明のポリべプチドのホモログとは、 カルボエル還元酵素活性を害さない 範囲内において配列番号 1に記載のァミノ酸配列に 1から複数個のアミノ酸が 欠失、 付加または置換されたアミノ酸配列を有するものである。 ここで複数個 とは、 具体的には 20個以下、 好ましくは 1 0個以下、 より好ましくは 5個以 下である。
また、 本発明のポリペプチドのホモログとは、 配列番号 1に示されるァミノ 酸配列と少なく とも 5 0 %、 好ましくは少なく とも 6 0 %または 70 %、 より好 ましくは 8 0%以上のホモロジ一を有するアミノ酸配列を有するポリペプチド をいう。
ちなみに上記ポリペプチドのホモロジ一検索は、 例えば、 DNA Databank of JAPAN (DDB J)を対象に、 FASTA programや BLAST programなどを用いて行うこと ができる。 配列番号 1に記載のアミノ酸配列を用いて DDBJを対象に BLAST prog ramを用いてホモロジ一検索を行った結果、 既知のタンパク質の中でもつとも 高いホモ口ジ一を示したのは、 シゾサッカロミセス ボンべ (Schizosaccharo myces pombe) の probable short chain dehydrogenase (T41540) の 3 7. 4 % であった。
また、 本発明の DNAは、 上記ポリペプチドをコードする DNAまたはその ホモログであって、 カルボ-ル還元酵素活性を有するポリべプチドをコ一ドす る DNAである。
上記ポリペプチドをコードする DN Aとしては、 具体的には配列番号 2に記 載の塩基配列を含むものが挙げられる。
本発明のポリペプチドをコードする DNAは、 例えば、 以下のような方法に よって単離することができる。
まず、 本発明のポリペプチドを上記の方法等にて精製後、 N末端アミノ酸配 列を解析し、 さらに、 リジルエンドべプチダーゼ、 V 8プロテアーゼなどの酵 素により切断後、 逆相液体クロマトグラフィーなどによりぺプチド断片を精製 後、 プロティンシーケンサ一によりァミノ酸配列を解析することにより複数の ァミノ酸配列を決める。
決定したァミノ酸配列を元に P CR用のプライマーを設計し、 カルボ-ル還 元酵素生産微生物株の染色体 DNAもしくは、 c DNAライブラリーを鎳型と し、 アミノ酸配列から設計した P CRプライマーを用いて P CRを行うことに より本発明の DNAの一部を得ることができる。 さらに、 得られた DNA断片 をプローブとして、 カルボ-ル還元酵素生産微生物株の染色体 DN Aの制限酵 素消化物をファージ、 プラスミ ドなどに導入し、 大腸菌を形質転換して得られ たライブラリ一や c DNAライブラリ一を利用して、 コ口エーハイブリダイゼ ーシヨン、 プラークハイブリダィゼーシヨンなどにより、 本発明の DNAを得 ることができる。 また、 P CRにより得られた DNA断片の塩基配列を解析し、 得られた配列から、 既知の DNAの外側に伸長させるための P C Rプライマー を設計し、 カルボ-ル還元酵素生産微生物株の染色体 DN Aを適当な制限酵素 で消化後、 自己環化反応により DN Aを铸型として逆 P CRを行うことにより
(Genetics 120, 621-623 (1988)) 、 また、 RACE法 (Rapid Amplificati on of cDNA End 「 P C R実験マニュアル j p25- 33, HB J出版局) などによ り本発明の DNAを得ることも可能である。
なお本発明の DN Aは、 以上のような方法によってクローニングされるゲノ ム DNA、 あるいは c DNAの他、 本発明によりその塩基配列が明らかになつ たため、 配列番号 2に基づく化学合成等によって得ることもできる。
本発明のポリぺプチドをコ一ドする DNAホモログとは、 カルボエル還元酵 素活性を害さない範囲内において配列番号 1に記載のァミノ酸配列に 1個から 複数個のアミノ酸が欠失、 付加または置換されたァミノ酸配列を含むポリぺプ チドをコードする DN Aを含む。 上記ポリぺプチドをコードする DN Aホモ口 グとしては、 具体的には配列番号 2に記載の塩基配列がコードするポリぺプチ ドにおいて、 カルボエル還元酵素活性を害さない範囲内において、 配列番号 2 に記載の塩基配列に 1から複数個のァミノ酸が欠失、 付加または置換された塩 基配列を含むものが挙げられる。 ここで複数個とは、 具体的には 6 0個以下、 好ましくは 3 0個以下、 より好ましくは 1 0個以下である。
当業者であれば、 配列番号 2に記載の DN Aに部位特異的変異導入法 (Nu c l e i c A c i d R e s . 1 0, p p . 648 7 (1 9 8 2) 、 Me t h o d s i n E n z ymo l . 1 00, p p . 448 (1 9 8 3) 、 Mo l e c u 1 a r C l o n i n g 2 n d E d t . , C o l d S p r i n g

P r a c t i c a l Ap p r o a c h I R L P r e s s p . 20 0 ( 1 9 9 1) ) 等を用いて適宜置換、 欠失、 揷入及び/または付加変異を導 入することにより本発明の DNAホモログを得ることが可能である。
また、 本発明の DN Aのホモログは、 本発明のポリペプチドをコードする D NAまたはその一部をプローブとして用いて、 カルボニル基の還元活性を有す る任意の微生物から調整した DN Aに対し、 コロニーハイプリダイゼーション 法、 プラークハイプリダイゼーシヨン法、 あるいはサザンブロットハイブリダ ィゼーション法等によりストリンジェントな条件下でハイブリダイゼーション を行い、 ハイブリダィズする DN Aを得ることによつても取得できる。 本発明 のポリペプチドをコードする DNAの 「一部」 とは、 プローブとして用いるの に十分な長さの DN Aのことであり、 具体的には 1 5 b p以上、 好ましくは 5 O b p以上、 より好ましくは 1 00 b p以上のものである。
各ノヽイブリグイ ITーシヨン【ま、 Molecular Cloning: A laboratory Mannual, 2nd Ed. , Cold Spring Harbor Laboratory, Cold Spring Harbor, NY. , 1989. ( 以後 〃モレキュラークローユング第 2版〃 と略す) 等に記載されている方 法に準じて行うことができる。 本明細書において 「ストリンジェントな条件下でハイプリダイズする塩基配 列」 とは、 D N Aをプローブとして使用し、 ストリンジェントな条件下、 コロ ニーハイプリダイゼーション法、 プラークハイブリダイゼーション法、 あるい はサザンプロットハイプリダイゼーション法等を用いることにより得られる D N Aの塩基配列を意味し、 ス ト リ ンジェントな条件としては、 例えば、 コ口- 一ハイプリダイゼーション法およびプラークハイブリダイゼーション法におい ては、 コロニーあるいはプラーク由来の D N Aまたは該 D N Aの断片を固定化 したフィルターを用いて、 0 . 7〜1 . O Mの塩化ナトリウム存在下 6 5 °Cで ハイプリダイゼーシヨ ンを行った後、 0 . 1 ~ 2 X S S C溶液 ( 1 X S S Cの 組成は、 1 5 0 mM塩化ナトリウム、 1 5 mMクェン酸ナトリウム) を用い、 6 5 °C条件下でフィルターを洗浄する条件を挙げることができる。
上記のようにして単離された、 本発明のポリぺプチドをコードする D N Aを 公知の発現ベクターに発現可能に揷入することにより、 カルボエル還元酵素発 現ベクターが提供される。 また、 この発現べクタ一で形質転換した形質転換体 を培養することにより、 カルボニル還元酵素を該形質転換体から得ることがで きる。 発現ベクターとしては、 本発明の D N Aを発現可能なベクターであれば 特に限定されず、 形質転換する宿主微生物の種類などに応じて適宜選択可能で ある。 あるいは、 形質転換体は、 公知の宿主の染色体 D N Aに本発明の D N A を発現可能に組み込むことによつても得ることができる。
形質転換体の作製方法としては、 具体的には、 微生物中において安定に存在 するプラスミ ドベクターやファージベクター中に、 本発明の D N Aを導入し、 構築された発現ベクターを該微生物中に導入するか、 もしくは、 直接宿主ゲノ ム中に本発明の D N Aを導入し、 その遺伝情報を転写 ·翻訳させることができ る。
このとき、 本発明の D N Aが宿主微生物中で発現可能なプロモーターを含ん でいない場合には、 適当なプロモーターを本発明の D N A鎖の 5' -側上流に、 より好ましくはターミネータ一を 3' -側下流にそれぞれ組み込むことができる。 このプロモーター及ぴターミネータ一としては、 宿主として利用する微生物中 において機能することが知られているプロモーター及びターミネータ一であれ ば特に限定されず、 これら各種微生物において利用可能なベクター、 プロモー ター及びターミネータ一などに関しては、 例えば 「微生物学基礎講座 8遺伝子 工学 ·共立出版」 、 特に酵母に関しては、 Adv. Biochem. Eng. 43, 75-102 ( 1 990)、 Yeast 8, 423-488 ( 1992)、 などに詳細に記述されている。
本発明の力ルポ-ル還元酵素を発現させるための形質転換の対象となる宿主 微生物としては、 宿主自体が本反応に悪影響を与えない限り特に限定されるこ とはなく、 具体的には以下に示すような微生物を挙げることができる。
エシュリヒア(Escherichia)属、 バチルス(Baci l lus)属、 シユードモナス(Ps eudomonas)属、 セラチア(Serratia)属、 ブレビパクテリゥム(Brevibacterium) 属、 コ リネノ クテリ ゥム(Corynebacterium)属、 ス ト レプトコッカス(Streptoc occus)属、 ラタ トバチルス(Lactobaci l lus)属などに属する宿主ベクター系の 確立されている細菌;
口 ドコッカス(Rhodococcus)属、 ス トレプトマイセス(Streptomyces)属など に属する宿主ベクター系の確立されている放線菌;
サッカロマイセス (Saccharomyces)属、 クライべ口マイセス (Kluyveromyces) 属、 シゾサッカロマイセス(Schi zosaccharorayces)属、 チゴサッカロマイセス (Zygosaccharomyces) fe、 ャロウィァ (Yarrowia) 、 トリコスホロン (Trichosp oron)属、 ロ ドスポジジゥム (Rhodosporidium)属、 ノヽンゼヌラ (Hansenula)属、 ピキア(Pi chia)属、 キャンディダ(Candi da)属などに属する宿主ベクター系の 確立されている酵母;
ノイロスポラ(Neurospora)属、 ァスぺノレギゾレス(Aspergi l lus)属、 セファ口 スポリゥム(Cephalosporium)属、 ト リコデノレマ(Tri chodertna)属などこ属する 宿主ベクター系の確立されているカビ。
上記微生物の中で宿主として好ましくは、 ェシェリヒア(Escheri chi a)属、 バチノレス(Baci llus)属、 ブレビバタテリゥム(Brevibacterium)属、 コリネバタ テリゥム(Corynebacterium)属であり、 特に好ましくは、 ェシエリヒア(Escher ichia)属、 コ];ネノ クテ];ゥム(Corynebacterium)属である。 形質転換体の作製のための手順、 宿主に適合した組換えベクターの構築およ ぴ宿主の培養方法は、 分子生物学、 生物工学、 遺伝子工学の分野において慣用 されている技術に準じて行うことができる (例えば、 Sambrookら、 モレキユラ 一 · クローユング、 Cold Spring Harbor Laboratories) D
以下、 具体的に、 好ましい宿主微生物、 各微生物における好ましい形質転換 の手法、 ベクター、 プロモーター、 ターミネータ一などの例を挙げるが、 本発 明はこれらの例に限定されない。
ェシエリ ヒア属、 特に大腸菌ェシエリ ヒア ' コリ (Escherichia coli)におい ては、 プラスミ ドベクターとしては、 pBR、 pUC系プラスミ ドが挙げられ、 lac ( ]3 -ガラク トシダーゼ)、 trp (トリプトファンオペロン)、 tac、 trc (lac, tr Pの融合)、 λファージ P L、 P Rなどに由来するプロモーターなどが挙げられ る。 また、 ターミネータ一としては、 trpA由来、 ファージ由来、 rrnBリボソ一 マゾレ R N A由来のターミネータ一などが挙げられる。
バチルス属においては、 ベクターとしては、 pUBl lO系プラスミ ド、 pC194系 プラスミ ドなどを挙げることができ、 また、 染色体にインテグレートすること もできる。 プロモーター及びターミネータ一としては、 アルカリプロテアーゼ、 中性プロテアーゼ、 ひ一アミラーゼ等の酵素遺伝子のプロモーターやターミネ 一ターなどが利用できる。
シユードモナス属においては、 ベクターとしては、 シユードモナス ' プチダ (Pseudomonas put i da) シユードモナス · セ/ヽシァ (Pseudomonas cepacia)な どで確立されている一般的な宿主ベクター系や、 トルエン化合物の分解に関与 するプラスミ ド、 T0Lプラスミ ドを基本にした広宿主域ベクター(RSF1010など に由来する自律的複製に必要な遺伝子を含む) pKT240 (Gene, 26, 273-82 (198 3) )を挙げることができる。
ブレビバタテリゥム属、 特にプレビパクテリゥム · ラタ トフアーメンタム(B revibacterium lactofermentum) こお ヽて fま、 ベクターとして【ま、 pAJ43 (Gene 39, 281 (1985) )などのプラスミ ドベクターを挙げることができる。 プロモー ター及びターミネータ一としては、 大腸菌で使用されている各種プロモーター 脑 62

及ぴターミネータ一が利用可能である。 コリネパクテリゥム属、 特にコリネバ クテリゥム ■ ク、'ノレタミカム (Corynebacterium glutamicum)【こお ヽて ίま、 ベクタ 一としては、 pCSl l (特開昭 57- 183799号公報)、 pCB10 l (Mol. Gen. Genet. 196, 175 (1984)などのプラスミ ドベクターが挙げられる。
サッカロマイセス(Saccharomyces)属、 特にサッ力ロマイセス ·セレビジァ ェ(Saccharomyces cerevis iae) におレヽて ίま、 ベクターとしては、 YRp系、 YEp 系、 YCp系、 Yip系プラスミ ドが挙げられる。 また、 アルコール脱水素酵素、 グ リセルアルデヒ ド一 3—リン酸脱水素酵素、 酸性フォスファターゼ、 —ガラ ク トシダーゼ、 ホスホグリセレー トキ^ "一ゼ、 エノラーゼといった各種酵素遺 伝子のプロモーター、 ターミネータ一が利用可能である。
シゾサッカロマイセス(Schizosaccharomyces)属におレヽては、 ベクターとし ては、 Mol. Cell, Biol. 6, 80 ( 1986)に記載のシゾサッカロマイセス ' ポン ベ由来のプラスミ ドベクターを挙げることができる。 特に、 PAUR224は、 宝酒 造から市販されており容易に利用できる。
ァスぺノレギノレス(Aspergi llus)属においては、 ァスぺノレギノレス ■ 二ガー(Aspe rgi l lus niger; ヽ アスペスレギノレス ■ オリジー (Aspergi l lus oryzae) などカ カビの中で最もよく研究されており、 プラスミ ドゃ染色体へのインテグレーシ ョンが利用可能であり、 菌体外プロテア一ゼゃァミラーゼ由来のプロモーター が利用可能である (Trends in Biotechnology 7, 283-287 (1989) ) 。
また、 上記以外でも、 各種微生物に応じた宿主ベクター系が確立されており、 それらを適宜使用することができる。 また、 微生物以外でも、 植物、 動物にお いて様々な宿主 ·ベクター系が確立されており、 特に蚕を用いた昆虫などの動 物中 (Nature 315, 592-594 (1985) ) や菜種、 トウモロコシ、 ジャガイモなど の植物中に大量に異種タンパク質を発現させる系、 及び大腸菌無細胞抽出液や 小麦胚芽などの無細胞タンパク質合成系を用いた系が確立されており、 好適に 利用できる。
さらに本発明は、 上記の方法などで得られる本発明の形質転換体細胞、 該形 質転換体細胞培養物、 該形質転換体細胞処理物を、 反応基質である下記一般式 ( I ) 、 ( I I ) 及び (III) で表される化合物に作用させることにより、 該 化合物の力ルポエル基を不斉還元させ、 光学活性アルコールを製造する方法に 関する。 形質転換体細胞、 該形質転換体細胞培養物、 該形質転換体細胞処理物 はいずれかを単独で用いてもよく、 組み合わせて用いてもよい。

本発明の製造方法に用いられる原料である、 上記式 (I) 〜 (III) で表され る化合物において、 Rは、 水素原子、 アルキル基またはァリール基を示す。 アルキル基としては、 メチル基、 ェチル基、 イソプロピル基、 シクロプロピ ル基、 ブチル基、 イソブチル基、 t一ブチル基、 シクロへキシル基、 ベンジノレ 基、 フエネチル基等の、 アルキル基またはァリール基で置換されていてもよい 直鎖、 分岐若しくは環状のアルキル基である。
ァリール基としては、 フエニル基、 メシチル基、 ナフチル基等の、 アルキル 基で置換されていても良いフエニル基又はナフチル基が挙げられる。
上記 Rとして好ましくは、 Ci Caのアルキル基、 ベンジル基又はフエニル 基であり、 より好ましくは C! C のアルキル基であり、 特に好ましくはメチ ル基又はェチル基である。
本発明の製造方法において、 上記式 (II) および (ΠΙ) で表される化合物 は少なく とも下記式 (π' ) および下記式 (Π ) で表される光学活性体を含 む。 上記式 (II) および (III) で表される化合物のうち下記式 (II' ) および 下記式 (Π ) の含有量が多いことが好ましい。 さらに上記式 (II) および
(in) で表される化合物は下記式 (π') および下記式 (ΠΓ ) からなること が好ましい。 下記式 (Ι ) および下記式 (III' ) で表される化合物において Rは前記と同義である。

なお、 本発明の製造方法により式 ( I ) 〜 (ΙΠ) 及び (Ι ) 〜 (Π ) で 表される化合物から得られる産物は混合物でもよいが、 少なく とも下記式 (I V)を含む。 下記式 (IV) で表される化合物において Rは前記と同義である。

式 ( I ) 〜 (III) 及ぴ (Ι ) ~ (III' ) で表される化合物は、 特開平 1一 2798 66号公報、 特開平 8— 1 2 758 5号公報、 特開平 5— 1 7884 1号公報等に記載された方法と公知の方法を組み合わせることによって、 任意 に製造することができる。 これらの化合物は 1種あるいは 2種以上を組み合わ せて原料として使用することができる。
原料として、 式 (I) で表される化合物を用いて式 (IV) で表される化合物 を製造する場合には、 その製造中間体として、 式 (Π') で表される化合物を 経由する場合と、 式 (ΙΙΓ) で表される化合物を経由する場合がある。
ついては、 式 ( I ) で表される化合物から、 あらかじめ式 (II' ) で表され る化合物及び式 (Π ) で表される化合物を製造し、 それを単離し、 さらに式
(IV) で表される化合物へ誘導してもよいし、 式 (ΙΙ') で表される化合物及 び式 (ΙΙ ) で表される化合物を単離することなく、 そのまま式 (IV) で表 される化合物を製造してもよい。
また、 反応基質は、 通常、 基質濃度が 0. 0 1〜 90 w/v° 好ましくは 0. 1〜 30 wZ V。/。の範囲で用いられる。 これらは、 反応開始時に一括して 添加しても良いが、 酵素の基質阻害があった場合の影響を減らすと言う点や生 成物の蓄積濃度を向上させるという観点からすると、 連続的もしくは間欠的に 添加することが望ましい。 本発明の製造方法において、 カルボニル基含有化合物に上記形質転換体を作 用させるに当たっては、 該形質転換体細胞をそのまま、 該形質転換体細胞培養 物、 あるいは該形質転換体細胞を公知の方法によつて処理して得られる該形質 転換体細胞処理物、 例えば、 該形質転換体細胞をアセ トン、 ジメチルスルホキ シド (DMSO) 、 トルエン等の有機溶媒や界面活性剤により処理したもの、 凍結乾燥処理したもの、 物理的または酵素的に破砕したもの等の菌体処理物、 該形質転換体細胞中の本発明の酵素画分を粗製物あるいは精製物として取り出 したもの、 さらには、 これらをポリアタリルァミ ドゲル、 カラギーナンゲル等 に代表される担体に固定化したものを用いることができる。
また、 本発明の製造方法においては、 補酵素 NAD P+もしくは NAD PHを 添加するのが好ましく、 通常、 0. 00 ImM〜: L 0 OmM、 好ましくは 0. 0 1 〜 1 OmM添加する。
上記補酵素を添加する場合には、 NAD PHから生成する NAD P+を NAD PHへの再生させることが生産効率向上のため好ましく、 再生方法としては、 1 ) 宿主微生物自体の NAD P+還元能を利用する方法、 2) NADP+から N AD PHを生成する能力を有する微生物やその処理物、 あるいは、 グルコース 脱水素酵素、 ギ酸脱水素酵素、 アルコール脱水素酵素、 アミノ酸脱水素酵素、 有機酸脱水素酵素 (リンゴ酸脱水素酵素など) などの NAD PHの再生に利用 可能な酵素 (再生酵素) を反応系内に添加する方法、 3) 形質転換体を製造す るに当たり、 NAD PHの再生に利用可能な酵素である上記再生酵素類の遺伝 子を本発明の DN Aと同時に宿主に導入する方法が挙げられる。
このうち、 上記 1) の方法においては、 反応系にグルコースやエタノール、 ギ酸などを添加する方が好ましい。
また、 上記 2) の方法においては、 上記再生酵素類を含む微生物、 該微生物 菌体をアセ トン処理したもの、 凍結乾燥処理したもの、 物理的または酵素的に 破砕したもの等の菌体処理物、 該酵素画分を粗製物あるいは精製物として取り 出したもの、 さらには、 これらをポリアタリルァミ ドゲル、 カラギーナンゲル 等に代表される担体に固定化したもの等を用いてもよく、 また市販の酵素を用 PC蘭膽 62

いても良い。
この場合、 上記再生酵素の使用量としては、 具体的には、 本発明のカルボ二 ル還元酵素に比較して、 酵素活性で 0 . 0 1〜 1 0 0倍、 好ましくは 0 . 5〜 2 0倍程度となるよう添加する。
また、 上記再生酵素の基質となる化合物、 例えば、 グルコース脱水素酵素を 利用する場合のグルコース、 ギ酸脱水素酵素を利用する場合のギ酸、 アルコー ル脱水素酵素を利用する場合のエタノールもしくはイソプロパノールなど、 の 添加も必要となるが、 その添加量としては、 反応原料である力ルポュル基含有 化合物に対して、 0 . 1 ~ 2 0倍モル当量、 好ましくは 1 ~ 5倍モル当量添加 する。
また、 上記 3 ) の方法においては、 本発明の D N Aと上記再生酵素類の D N Aを染色体に組み込む方法、 単一のベクター中に両 D N Aを導入し、 宿主を形 質転換する方法及ぴ両 D N Aをそれぞれ別個にベクターに導入した後に宿主を 形質転換する方法を用いることができるが、 両 D N Aをそれぞれ別個にベクタ 一に導入した後に宿主を形質転換する方法の場合、 両ベクター同士の不和合性 を考慮してベクターを選択する必要がある。
単一のベクター中に複数の遺伝子を導入する場合には、 プロモーター及びタ 一ミネ一ターなど発現制御に関わる領域をそれぞれの遺伝子に連結する方法や ラタ トースオペ口ンのような複数のシストロンを含むオペ口ンとして発現させ ることも可能である。
本発明の製造方法は、 反応基質及び本発明の形質転換体細胞、 該形質転換体 細胞培養物または該形質転換体処理物、 並びに、 必要に応じて添加された各種 補酵素及びその再生システムを含有する水性媒体中もしくは該水性媒体と有機 溶媒との混合物中で行われる。 上記水性媒体としては、 リン酸ナトリウムゃリ ン酸カリゥムなどを用いた緩衝液などを用いることができ、 その緩衝液に有機 溶媒や、 界面活性剤などの通常の酵素反応に用いられる任意成分を適宜加えた ものを用いることができる。 有機溶媒としては、 ジメチルスルホキシド (D M S O ) ゃテトラヒ ドロフラン (T H F ) 等の水溶性溶媒や酢酸プチルやへキサ ン等の非水溶性有機溶媒などが挙げられ、 反応基質の溶解度が高い物を好まし く使用することができる。 界面活性剤としては Tw e e n 8 0やシュガーエス テルなどが挙げられる。
本発明の方法は、 通常、 4〜60°C、 好ましくは 1 0〜4 5°Cの反応温度で、 通常 p H 3〜 l 1、 好ましくは p H 5〜 8で行われる。 また、 膜リアクターな どを利用して行うことも可能である。
本発明の方法により生成する光学活性アルコールは、 反応終了後、 反応液中 の菌体ゃタンパク質を遠心分離、 膜処理などにより分離した後に、 酢酸ェチル、 トルエンなどの有機溶媒による抽出、 蒸留、 カラムクロマトグラフィー、 晶析 等のなどを適宜組み合わせることにより精製を行うことができる。 実施例
以下に実施例を挙げて本発明を更に具体的に説明するが、 その要旨を越えな い限り本発明の技術分野における通常の変更をすることができる。
尚、 (E) - 7 - [ 2—シクロプロピル一 4一 (4一フルオロフェエル) - キノ リン一 3—ィノレ] 一 3, 5—ジヒ ドロキシヘプトー 6—ェン酸エステノレ類 は、 目的とする 3 R, 5 S体の他に異性体として、 3 S, 5 R体、 3 R, 5 R 体及び 3 S, 5 S体が存在し、 (E) — 7— [ 2—シクロプロピル一 4— (4 —フルオロフェニル) 一キノ リ ン一 3—ィル] - 3, 5—ジヒ ドロキシヘプト 一 6—ェン酸ェチルエステル (以後、 DO LEと略す) を例として示すと、 そ の構造式は以下の通りである。

<実施例 1 > カルボエル還元酵素の単離
ォガタエア ミヌタ 変種ノンフアーメンタス (Ogataea minuta var nonfe rmentans) I F O 1 4 7 3株を 20 Lの YM培地 (グルコース 24g、 酵母ェキ ス 3g、 麦芽エキス 3g、 ペプトン 5g/L、 pH 6.0) で培養し、 遠心分離により菌 体を調製した。 得られた湿菌体のうち 3 00 gを 5 0mMリン酸カリゥム緩衝液 (p H 7. 0、 1 mM DTT) で澱懸し、 ダイノーミル (Dy n o— M i 1 1社製) により破砕後、 遠心分離により菌体残渣を除去し、 無細胞抽出液を得 た。 この無細胞抽出液に 12%w/vのポリエチレングリコール (P EG) 6 000 を添加し、 遠心分離により上清を得た。 その上清を標準緩衝液 (l OraMリン酸 カリウム緩衝液 (ρ Η 7. 0) 、 1 mM DTT) で平衡化した DEAEセフ ァロース 6 F F (2. 6 cmx 28 c m) に添加した。 標準緩衝液でカラムを 洗浄した後、 さらに 0. 07M塩化ナトリウムを含む標準緩衝液で洗浄後、 0. 0 7-0. 1 8M塩化ナトリウムの勾配溶出を行った。 溶出した画分を回収し、 カルボニル還元酵素活性測定を行った。
カルボニル還元酵素活性測定は以下の反応液組成:酵素活性測定画分 200μ L、 に NAD PH 8 mMの水酸化カリ ウム 0. 1 mMの水溶液 1 0 μ L、 0. 1Mリン酸緩衝液 (ρΗ7.0) 25/iL、 20 g / L DOXE (DM S O溶液) 1 0 μ L添加したものを 30°C、 一夜振とう反応させることで行った。 活性検出は 以下方法で行った。 反応終了後の反応液に酢酸ェチルを 0. 5 mL加え激しく 混合し、 遠心分離にて有機層と水層とに分離した。 有機層を別の容器に移し、 溶媒を濃縮遠心機で留去し、 乾固された生成物を酢酸ェチル 0. O l mLで溶 解し、 薄層クロマトグラフィー (TL C) をおこなった。 T LCはシリカゲル プレート (メルク社製 silica gel 60 F 2M)を用い、 展開溶媒はへキサン Z酢酸 ェチルニ丄 丄を用いた。 展開終了後、 UVランプにて生成物の確認をした。 化合物 ( I ) は R i = 0. 76〜0. 8 6、 化合物 ( I I ) およぴ化合物 ( I 1 1 ) は0. 5 4〜0. 6 1、 化合物 ( I V) (式中、 R =ェチル基の化合物 : DO L E) は R f = 0 , 3 3である。
カルボニル還元酵素活性は、 0. 1 3 M塩化ナトリウム付近にピークが見ら れた。 溶出した活性画分を回収し、 硫安を終濃度 0. 3 Mになるまで添加し、 0. 3 M硫安を含む標準緩衝液 (1 OmMリン酸カリウム緩衝液 (p H 7. 0 ) 、 1 mM D T T) で平衡化したォクチルセファロース C L 4 B ( 0. 8 craX 2 0 cm) に添加し、 硫安濃度を 0. 3〜 0Mの勾配溶出を行った。 力ルポニル還 元酵素活性は、 0M硫安付近にピークが見られた。
溶出したピーク部分を回収し、 限外濾過により標準溶液まで脱塩、 濃縮した 後同緩衝液で平衡化した M o n o Q HR 5 / 5に添加し、 同緩衝液及ぴ 0 · 0 5M 塩化ナトリウムを含む同緩衝液で洗浄後、 0. 0 5 M〜0. 2 5M塩 化ナトリウムの濃度勾配溶出を行った。 カルボ-ル還元酵素活性は、 0. 1 7 M 塩化ナトリウム画分に溶出したため、 この画分を回収し、 濃縮した。 濃縮 した酵素液をスーパーデッタス 2 0 0 HR 1 0 Z 3 0を用い、 0. 1 5 M塩 化ナトリゥムを含む標準緩衝液でゲル濾過を行った。 ゲル濾過により得られた 活性画分の分子量は、 約 1 0. 7万 D aであった。
また、 上記活性画分をポリアク リルアミ ドゲル電気泳動 (S D S— P AG E) により解析した結果、 ほぼ単一バンドであり、 その分子量は約 2. 7万 Ό aであった。
また、 該酵素を種々のケトンやアルデヒ ドと反応させることにより、 具体的 には、 基質 lOOmMのイソプロパノール溶液を 5 L、 NAD P H 8 mM及び 水酸化カリウム 0. 1 mMの水溶液 1 0 /x Lを 0. 1Mリン酸緩衝液 (pH7.0) で総量 2 5 0 μ Lの反応液としたものの中での NAD Ρ Η減少初速度を測定す ることにより、 その還元活性を測定した。 このときの 2, 2, 2—トリフルォ ロアセ トフェノンでの吸光度変化量を 1 0 0としたときの比活性を表 1に示し た。

該酵素の至適 p Hは、 : Hの異なる複数の緩衝液中で、 2, 2, 2—トリフ ルォロアセトフヱノン還元活性を測定することで求めた。 具体的には、 0. 1Mの リン酸カリゥム緩衝液、 酢酸ナトリゥム緩衝液及びクェン酸緩衝液において、 それぞれ pHの異なるものを用意し、 活性測定した。 反応の至適 pHは 5. 0 - 6. 5であった。
また、 反応温度だけを変化させて、 2, 2, 2—トリフル'ォロアセトフエノ ン還元活性を測定し、 力ルポ-ル還元酵素の作用至適温度を測定したところ、 至適温度は 6 0〜 7 0°Cであった。
該酵素の; p H安定性は、 0. 1Mのリン酸カリゥム緩衝液、 酢酸ナトリゥム緩衝 液及びクェン酸緩衝液において異なる p Hのものを複数用意し、 それぞれ 30°C、 30分間インキュベートし、 残存活性を測定したところ、 p H 5. 5〜6. 5に おいて最も安定であった。
該酵素の温度安定性は、 p H 7. 0で 2 0 mMのリン酸緩衝液中、 3 0 °C、
4 0 °C、 5 0 °C、 6 0 °C、 7 0 °C及ぴ 8 0 °Cの温度でそれぞれ 10分間放置した 後、 2, 2, 2—トリフルォロアセトフエノン還元活性を測定することにより 調べた。 それによると本発明の酵素は 4 0°Cまで 7 9%以上の残存活性を示し、
5 0 °C以上で急速に失活しはじめ、 7 0°Cでは活性が全くなくなることがわか つた。
また、 該酵素を種々の試薬中で 30°C、 10分間処理した後、 2, 2, 2—トリ フルォロアセトフエノン還元活性を測定したところ、 該酵素は、 硝酸水銀や塩 化鉛のような水銀 ( I ) イオン、 鉛 ( I I ) イオン存在下で活性が顕著に阻害 された。
<実施例 2 > カルボエル還元酵素の部分ァミノ酸配列の解析
実施例 1で得られたカルボニル還元酵素を含む画分を脱塩、 濃縮後、 リジル エンドぺプチダーゼを用いて、 30°Cで終夜処理を行った。 消化したペプチドを 逆相 H P L C (アマシャム-フアルマシア製 uRPC C2/C18、 46mm X 1 0 0 m m) を用い、 0. 1% トリフルォロ酢酸 (TFA) 中でァセトニト リルのグラジェン ト溶出によりペプチドを分離し、 分取した。 分取したペプチドピ^ "ク 3種を Fr aC20,27及び 30とし、 それぞれェドマン法によりアミノ酸配列の解析を行った。 Frac20,27及び 30のそれぞれのアミノ酸配列は配列番号 3、 4及び 5に示した。 同様にカルボニル還元酵素を含む画分を脱塩後ェドマン法により N末端ァミノ 酸の解析を行い、 結果を配列番号 6で示した。 く実施例 3 > ォガタエア ミヌタ 変種ノンフアーメンタス (Ogataea rainu ta var nonf ermentans) I F O 147 3株由来の本発明の D N Aの配列解析 及び形質転換体の作製
ォガタエア ミヌタ 変種ノンファーメンタス (Ogataea minuta var nonfe rmentans) I FO 1 4 73株を YM培地で培養し、 菌体を調製した。
菌体からの総 RN Aの精製は、 Ma g E x t r a c t o r (東洋紡社製) を 用いて行った。 精製した RN Aをもとにして S u p e r S c r i p t l l (L i f e T e c h n o l o g y社製) を用いて F i r s t— S t r a n d c DNAを合成した。 方法はそれぞれ添付の説明書通りに行った。
実施例 2で得られた N末端アミノ酸配列を元にセンスデジエネレイ ト(degene rate)プライマー及び配列番号 3、 4及び 5のアミノ酸配列を元にそれぞれァ ンチセンスのデジエネレイ ト(degenerate)プライマーを計 4種類合成した。 そ れぞれの塩基配列を配列番号 7、 8、 9、 10に示した。 N末端と 3つの部分アミ ノ酸配列の組み合わせ (3通り) でプライマーを選び、 プライマーを各 400 pmo l、 dNTP 0. 4 mm o 1、 上記で合成したォガタエア ミヌタ 変 種ノンファーメンタス (Ogataea minuta var nonf ermentans) I F O 1 4 7 3株の c DNA 1 L、 TaKaRa Taq用 1 0 X緩衝液 (宝酒造社製) 5 μ L及ぴ 耐熱性 DNAポリメラーゼ 2ユニット(宝酒造社製、 商品名 「TaKaRa Taqj ) を含む 5 0 Lの反応液を用い、 変性 (94°C、 30秒) 、 ァニール (50°C、 30 秒) 、 伸長 (72°C、 1分) を 30サイクル、 PTC- 200 Peltier Thermal Cycler (M J Research社製)を用いて行った。 P C R反応液の一部をァガロースゲル電気 泳動により解析した結果、 配列番号 7のプライマーと配列番号 8のプライマー、 配列番号 7のプライマーと配列番号 9のプライマー、 配列番号 7のプライマー と配列番号 1 0との組み合わせにおいて特異的と思われるバンドが検出できた。 上記で得られた 3種類の DN A断片を、 ァガロースゲル電気泳動を行い、 目 的とするパンドの部分を切り出し Gel Extraction kit (QIAGEN社製)にて精製 して回収した。 得られた DN A断片を、 pGEM- T Vector Systera(Promega|± 製)により TAクローニングし、 大腸菌 DH5o;株 (東洋紡社製) を形質転換した。 形質転換株を 1%バタ ト トリプトン、 0. 5%パク ト酵母エキス及び 1%塩化ナ トリウムを含む培地 (以下、 LB培地と略す) にアンピシリン (l O O wg/mL) を加え、 1. 5 %バタ トァガーでプレート化したもの上で生育させ、 いくつか のコロニーを用いて、 ベクターの配列より合成された配列番号 1 1及び 1 2を プライマーとしてコロニーダイレク ト P CRを行い、 揷入断片のサイズを確認 した。 目的とする DN A断片が揷入されていると考えられるコロニーを 100/i g /mLアンピシリ ンを含む液体 L B培地で培養し、 Mini- Prep (QIAGEN製) により プラスミ ドを精製した。 配列番号 7のプライマーと配列番号 8のプライマー、 配列番号 7のプライマーと配列番号 9のプライマー、 配列番号 7のプライマー と配列番号 1 0のプライマーとの組み合わせから得られたプラスミ ドをそれぞ れ phir2ト 23、 phir2ト 25、 phir2ト 27とした。
精製したプラスミ ドを用いて、 揷入 DNAの塩基配列をダイターミネータ一 法により解析した。 決定された phir21-23、 phir21- 25、 phir21- 27の各挿入断 片部分の塩基配列をそれぞれ配列番号: 1 3、 14、 1 5として示した。
上記塩基配列を元に設計したプライマーを用いて、 ォガタエア ミヌタ 変 種ノンファーメ ンタス (Ogataea rainuta var nonf ermentans) I F O 1 4 7 3株から調製された総 RNAに対し、 5 ' RACE法及ぴ 3 ' RACE法を行 つた。
5 ' RAC E法は、 配列番号 1 6、 1 7で示される塩基配列のプライマーを Gene Specific Primer (以下 GSPと略す) 1, 2として用い、 5, RACE S y s t em f o r R a i d Amp l i f i c a t i o n o f c DN A E n d s、 v e r 2. 0 (L i f e T e c h n o l o g y社製)にて行つ た後、 それをテンプレートとして再度配列番号 1 8で示される塩基配列のブラ イマ一を GSPとして 5, RACE法を行った。
3 ' RACE法については、 配列番号 1 9で示される塩基配列のプライマー PC漏膽 62

を GSPとして用い、 3 ' RAC E S y s t e m f o r R a i d Am p l i f i c a t i o n o f c DNA E n d s ( L i f e T e c h n o 1 o g y社製)を用いて行った。 なお、 各 R AC E法はおのおの添付されて いる取扱説明書に基づいておこなった。
得られた DNA断片は pGEM- T Vector System (Promega社製)により TAクロー- ングし、 大腸菌 DH5a株 (東洋紡社製) を形質転換した。 形質転換株を 1 00 μ g/mLアンピシリンを含む L B培地プレート上で生育させ、 いくつかのコ口- 一を用いて、 ベクターの配列より合成された配列番号 1 1、 1 2をプライマー として用いてコロニーダイレク ト P CRを行い、 揷入断片のサイズを確認した。 目的とする DN A断片が挿入されていると考えられるコロニーを 1 00 xg/mL アンピシリンを含む液体 L B培地で培養し、 Mini- Prep (QIAGEN製) によりプ ラスミ ドを精製した後に、 ダイターミネータ一法により DNA塩基配列の解析 を行った。
上記 5, RAC E及び 3, R AC Eで得られた phir21- 25の 5'上流の塩基配 列及び 3'下流の塩基配列と、 配列番号 14に記載の塩基配列情報とを元に作成 した DNA配列について Genetyx (ソフトウェア開発株式会社製) を用いて 0RF 検索を行い、 本発明の DNA配列及びコードするタンパク質のァミノ酸配列を 配列番号: 20に示される塩基配列及ぴアミノ酸配列に仮決定した。
配列番号 2 0に記載のアミノ酸配列を用いて DDBJを対象に BLAST programを 用いてホモロジ一検索を行った結果、 既知のタンパク質の中でもっとも高いホ モロジ一を示したのは、 シゾサッカロミセス ボンべ (Schizosaccharomyces porabe) の probable snort chain dehydrogenase (T41540J の 3 7. 4%であつ た。 引き続き、 上記配列番号 2 0に記載の配列を元に、 クローユング用のブラ イマ一として配列番号: 2 1に記載の塩基配列及ぴ配列番号: 22に記載の塩 基配列を合成し、 上記プライマーを各 5 Opmol、 dNTP20 Onmol, ォガタエア ヌタ 変種ノンファーメンタス (Ogataea minuta var nonfermentans) I F O 1 4 7 3株の cDNA 1/zL 、 KOD- DNApolymerase用 1 0 X緩衝液 (東洋紡 社製) 5 L、 KOD- DNA polymerase 2. 5ュ-ット(東洋紡社製)を含む 5 0 μ L PC漏膽 62

の反応液を用い、 変性 (96°C、 3 0秒)、 ァエール ( 5 4 °C、 3 0秒) 、 伸長 ( 7 4。C、 1分) を 3 0サイクノレ、 PTC-200 Peltier Thermal Cycler (MJ Res earch社製) を用いて行った。 P C R反応液の一部をァガロースゲル電気泳動 により解析した結果、 特異的と思われるバンドが検出できた。
上記で検出されたバンド部分を QIAGEN Gel Extraction kit (キアゲン社 製) にて回収した。 回収した D NA断片を制限酵素 Hindlllと EcoRIで消化し、 ァガロースゲル電気泳動を行い、 目的とするパンドの部分を切り出し、 再度、 Qiagen Gel Extraction kit (キアゲン社製) により精製後回収した。 得ら れた DN A断片を、 EcoRI、 及び Hindlllで消化した pKK223- 3 (フアルマシア社 製) と Takara Ligation Kitを用いて、 ライゲーシヨ ンし、 大腸菌 JM109株を形 質転換した。
形質転換体をアンピシリ ン ( 5 0 z g/mL) を含む L B培地プレート上で生育 させ、 いくつかのコロニーを用いて、 ベクターの配列より合成された配列番号 2 3で示される塩基配列のプライマー及ぴ配列番号 2 4で示される塩基配列の プライマーを用いてコロニーダイレク ト P C Rを行い、 挿入断片のサイズを確 認した。
目的とする DNA断片が揷入されていると考えられる形質転換体を 5 0 μ g/ mLのアンピシリンを含む液体 L B培地で培養し、 Qiagen 500 (キアゲン社製) を用いてプラスミ ドを精製し、 PKK223- 30CR1とした。
プラスミ ドに揷入した DNAの塩基配列をダイターミネータ一法により解析 したところ、 揷入された DNA断片は、 配列番号: 2で表される塩基配列から なる遺伝子の 5'上流にクローニングのための 6塩基が、 3'下流にォガタエア ミヌタ 変種ノンファーメンタス (Ogataea minuta var nonfermentans) I F O 1 4 7 3株由来のダブルストップコドン (TAGTAAT) とクローエングのた め 6塩基が付与された DN A断片であった。
KK 2 3 3 - 3 0 C R 1に揷入された D N A断片の塩基配列を配列番号: 2 5に、 該 D N A断片がコードするタンパク質のアミノ酸配列を配列番号: 1 に示した。 62

<実施例 5 > 本発明の DNAによって形質転換した大腸菌を用いた D OX E からの DO L Eの製造
実施例 4で得られた形質転換体をアンピシリ ン (S O ^g/mL) 、 O.lmM イソ プロピル —D—チォガラタ トピラノシド (IPTG) を含む L B培地で 37°C で 1 7時間生育させ、 得られた菌体ブロス 2mLを遠心分離により集菌後、 Ιδθμ Lの lOOmMリン酸カリウム緩衝液 (pH7. 0) に懸濁した後に、 下記に示す方法 により、 (E) — 7— [2—シクロプロピル一 4一 (4一フルオロフェニル) 一キノ リ ン一 3—ィル] 一 3 , 5一ジォキソヘプトー 6—ェン酸ェチルエステ ル (DOXE) を基質として還元活性を確認した。
上記菌体懸濁物に 2g/L NADP+ (オリエンタル酵母社製) 10Μ L、 トルエン 1 / Lを添加後、 5分間ボルテックスミキサーで攪拌した。 50%グルコース 10μ L、 グルコースデヒ ドロゲナーゼ溶液 (天野製薬社製; 25 U/mL) 10 β L, 上記基質を DMS Oに 20g/Lで溶解させたもの 50μ L (基質 1 m g分) を添 加後 40°Cで 20時間振とう反応させた。 反応終了後の反応懸濁液にァセトニト リル 1. 75 mLを添加して希釈後遠心し、 上清上の生成物を高速液体クロマ トグラフィー (HP L C) を用いて測定した。 HP L Cの条件は以下の通りで ある。
カラム: MCIGEL CHP2MGM (三菱化学社製)
溶離液: メタノール Zァセトニトリル Z水 Zリン酸 =8ooZiooZioo/ 1 流速: 0.6ml/tnin
検出: U V 254 nm
温度: 60°C
この結果、 収量は 3 1 9 gであり、 収率は 3 1. 9%であった。
また、 光学純度の測定は、 反応終了後の反応液に酢酸ェチルを 0. 5mL加 え激しく混合し、 遠心分離にて有機層と水層とに分離した。 有機層を別の容器 に移し、 溶媒を濃縮遠心機で留去し、 乾固された生成物を酢酸ェチル 0. 0 1 mLで溶解し、 薄層クロマトグラフィー (TLC) をおこなった。 TLCはシ リカゲルプレート (メルク社製 silica gel 60 F 2M)を用い、 展開溶媒はへキサ ン Z酢酸ェチル = 1/1を用いた。
展開終了後、 UVランプにて生成物の確認をした。 化合物 ( I ) は R f = 0· 7 6〜 0. 8 6 、 化合物 ( I I ) および化合物 ( I I I ) は 0. 5 4〜0. 6 1、 化合物 ( I V) (式中、 R=ェチル基の化合物: 以下、 DO L Eと略 す) は R i = 0. 3 3である。 DOLEの T LC上のスポットを搔き取りイソ プロパノール 0. 2 5 mLで溶出し、 遠心分離後上清を高速液体クロマトダラ フィー (HP LC) を用いて光学純度および T L C搔き取りサンプルの濃度を 分析した。
HP LCの条件は以下の通りである。
カラム : ダイセル CHIRALCEL AD
溶離液:へキサン/エタノール Zトリフルォロ酢酸 = 900/1 00/1 流速: 1 ral/min
検出: UV 254 nm
温度:室温
その結果、 光学純度は 1 00 %e. e.、 anti体は 0. 6。/。であった。
また該遺伝子を含まないプラスミ ド pKK223- 3を持つ大腸菌を 0. IraMIPTGを 添加した L B培地で終夜培養した菌体を、 同様に反応させてみたが上記生成物 は認められなかった。 く実施例 6 > 本発明の DNAによって形質転換した大腸菌を用いた 5 S一 M O L Eからの DO L Eの製造
実施例 4で得られた形質転換体をアンピシリン (5 0 xg/mL) 、 0. ImM イソ プロピル — D—チォガラタ トピラノシド (IPTG) を含む LB培地で 3 7。C で 1 7時間生育させ、 得られた菌体ブロス 2mLを遠心分離により集菌後、 180 z Lの lOOraMリン酸カリウム緩衝液 (pH7. 0) に懸濁した後に、 下記に示す方法 により、 5 S- (E) -7 - 〔2 -シクロプロピル- 4- (4 -フルオロフェュル) - キノ リン - 3 -ィル〕 -5 -ヒ ドロキシ- 3- ォキソ- ヘプト- 6- ェン酸ェチルエステ ル (5 S— MO L E) を基質として還元活性を確認した。
上記菌体懸濁物に 2g/L NADP+ (オリエンタル酵母社製) 10^ L、 トルエン 1 i Lを添加後、 5分間ポルテックスミキサーで攪拌した。 50%グルコース 10// L、 グルコースデヒ ドロゲナーゼ溶液 (天野製薬社製; 2 5 U/mL) 10 μ L、 上記基質を DM S Oに 20g/Lで溶解させたもの50M L (基質 1 m g分) を添 加後 4 0°Cで 20時間振とう反応させた。 反応終了後の反応懸濁液をァセトニト リルで希釈後遠心し、 上清上の生成物を高速液体クロマトグラフィー (H P L C) を用いて測定した。 H P L Cの条件は以下の通りである。
カラム : コスモシノレ 5 C 1 8 M S— I I 4. 6 X 2 5 0 mm (ナカライ 社製)
溶離液: メタノール/ァセトェトリル /水/リン酸 = 3 5 0 / 1 5 0 / 5 0 0/ 1
流速: 0. 6 ml/min
検出: U V 2 5 4 nm
温度: 5 0 °C
この結果、 収量は 807/z gであり、 収率は 80.7%、 a n t i体は 15.3% であった。
また、 光学純度の測定は、 反応終了後、 反応液に酢酸ェチルを 0. 5 m L加 え激しく混合し、 遠心分離にて有機層と水層とに分離した。 有機層を別の容器 に移し、 溶媒を濃縮遠心機で留去し、 乾固された生成物を酢酸ェチル 0. 0 1 m Lで溶解し、 薄層クロマトグラフィー (T L C) をおこなった。 T L Cはシ リカゲルプレート (メルク社製 silica gel 60 F 254)を用い、 展開溶媒はへキサ ン Z酢酸ェチル = 1/1を用いた。
展開終了後、 UVランプにて生成物の確認をした。 化合物 ( I ) は R f = 0. 7 6〜 0. 8 6 、 化合物 ( I I ) および化合物 ( I I I ) は 0. 5 4〜0. 6 1、 化合物 ( I V) (式中、 R =ェチル基の化合物:以下、 D O L Eと略 す) は R f = 0. 3 3である。 D O L Eの T L C上のスポッ トを搔き取りイソ プロパノール 0. 2 5 mLで溶出し、 遠心分離後上清を高速液体クロマトダラ フィー (HP LC) を用いて光学純度および T L C搔き取りサンプルの濃度を 分析した。
HP L Cの条件は以下の通りである。
カラム : ダイセル CHIRALCEL AD
溶離液:へキサン Zエタノール = 9 5/5
流速: 1 ml/min
検出 : UV 254 nm
温度: 50 °C
その結果、 光学純度は 97%e. e.、 anti体は 15.2%であった。
また該遺伝子を含まないプラスミ ド PKK223- 3を持つ大腸菌を 0. lmMIPTGを 添加した L B培地で終夜培養した菌体を、 同様に反応させてみたが上記生成物 は認められなかった。 産業上の利用の可能性
医薬、 農薬等の中間体原料として産業上有用な化合物である光学活性アルコ 一ル類を高光学純度かつ高収率で得ることができる製造方法を提供する。

Claims

請求の範囲
1. 下記 (A) または (B) の何れかのアミノ酸配列を有するポリペプチド。
(A) 配列番号 1に記載のアミノ酸配列。
(B) 配列番号 1に記載のアミノ酸配列において、 1から複数個のアミノ酸が 欠失、 付加または置換されているアミノ酸配列であって、 力ルポエル還元酵素 活性を有するポリぺプチドのァミノ酸配列。
2. 下記 (a) から (e) の何れかの塩基配列を有する DNA。
(a ) 配列番号 1に記載のアミノ酸配列からなるポリべプチドをコードする塩 基配列。
(b) 配列番号 1に記載のアミノ酸配列において、 1から複数個のアミノ酸が 欠失、 付加または置換されているアミノ酸配列からなるポリぺプチドであって、 カルボニル還元酵素活性を有するポリぺプチドをコードする塩基配列。
(c) 配列番号 2に記載の塩基配列。
(d) 配列番号 2に記載の塩基配列において、 1から複数個の塩基が欠失、 付 加または置換されている塩基配列であって、 力ルポュル還元酵素活性を有する ポリペプチドをコードする塩基配列。
(e ) 配列番号 2に記載の塩基配列もしくはその相補配列またはそれらの一部 とストリンジェントな条件下でハイプリダイズする塩基配列であって、 カルボ ニル還元酵素活性を有するポリべプチドをコードする塩基配列。
3. 請求項 2に記載の DNAをベクターに組み込んで得られる組換え体 DN

4. 請求項 3に記載の組換え体 DNAを保有する形質転換体。
5. 請求項 2に記載の DN Aを染色体 DN Aに組み込んで得られる形質転換 体
