明 細 書 口腔用組成物 技術分野
本発明は、 保型性、 分散性、 歯磨後のジュースの味を変えず、 特に経時安定性 に優れた口腔用組成物に関する。 また、 本発明は、 カチオン性殺菌剤の歯面滞留 性に優れた口腔用組成物に関する。 背景技術
従来、 口腔用組成物の保型性や口腔内での分散性はカルボキシメチルセル口一 ス、 メチノレセノレロース、 ヒ ドロキシェチノレセノレロース、 アラビアガム、 キサンタ ンガム、 カラギーナン、 ァノレギン酸ナトリウム、 ポリアクリル酸ナトリウム等の 通常良く用いられる粘結剤を配合することによって得られている。 さちに、 特開 平 5— 5 8 8 6 1号により、 微粒化セルロースを配合させることによって、 従来 よりも良好な保型性を維持し、 口腔内での分散性に優れた性質を有する口腔用組 成物が提案された。 しかしながら、 この口腔用糸且成物では長期の保管により固液 分離を生じることなど、 実用上問題があり、 また、 界面活性剤としてアルキル硫 酸ナトリウムが用いられており、 これは、 歯磨後のジュースの味を変えることが 知られている。
また、 カチオン性殺菌剤は、 優れた口腔組織吸着性を有し、 殺菌力およぴプラ ーク形成抑制作用が高く、 そのため、 歯周疾患、 齲蝕等の口腔疾患の予防を目的 として種々の口腔用組成物に配合されている。
しかしながら、 カチオン性殺菌剤は、 それが有する電荷ゆえに、 他に配合され るァユオン性の成分と静電的コンプレックスを形成して単位当たりの殺菌活性が 低下するという問題があった。 それに対して、 これまでに非イオン性または両ィ オン性界面活性剤ゃ非ィォン性粘結剤を配合してカチオン性殺菌剤の単位当たり の活性の低下を防止する試みがなされてきたが、 十分な効果は得られていない。 一方、 カチオン性殺菌剤といえども口腔内における殺菌効果は一過性の効果で ある場合が多いため、 殺菌剤の歯面滞留性の向上を図ることによっても殺菌剤の 活性を全体的に向上させ得ることが考えられる。
本発明の第一の目的は、 良好な保型性を維持し、 口腔内での分散性に優れ、 歯 磨後のジュース効果の味を変えず、 しかも長期保管中に固液分離を生じない、 ま た、 'これらの特徴に加えてカチオン性殺菌剤の歯面滞留性が向上した口腔用組成 物を提供することにある。
また、 本突明の第二の目的は、 カチオン性殺菌剤の歯面滞留性を向上させるこ とにより、 カチオン性殺菌剤の滞留殺菌力を向上させ、 齲蝕と歯周疾患を効果的 に予防し得る口腔用組成物を提供することにある。
上記の前者の事情に鑑み、 本発明者らは鋭意研究を重ねた結果、 結晶セルロー スと特定の界面活性剤とを組合せて配合することにより、 良好な保型性を維持し、 口腔内での分散性に優れ、 歯磨後のジュースの味を変えず、 しかも長期保管中に 固液分離を生じない口腔用組成物が得られることを見い出し、 本発明の第一の態 様を完成するに至った。
また、 上記の後者の事情に鑑み、 本発明者らは鋭意研究を重ねた結果、 カチォ ン性殺菌剤と結晶セルロースとを特異的に組合せることにより、 カチオン性殺菌 剤の歯面滞留効果が顕著に向上することを見い出し、 本発明の第二の態様を完成 するに至った。
すなわち、 本発明は、 その第一の態様として、
1 . 結晶セルロースと、 アルキルグリコシド、 ポリグリセリン脂肪酸エステル、 ショ糖脂肪酸ェステルおよびべタインからなる群より選ばれる 1種以上の界面活 性剤とを含む口腔用組成物;
2 . 結晶セルロースを 0 . 2— 1 0重量。/。配合する前記 1に記載の口腔用組 成物;
3 . 界面活性剤がアルキルグリコシドである前記 1または 2に記載の口腔用 組成物
4 . アルキルダリコシドのアルキル鎖長が C 8— C 1 6である前記 3に記載 の口腔用組成物; 5 . 界面活性剤がポリグリセリン脂肪酸エステルまたはショ糖脂肪酸エステ ルである前記 1または 2に記載の口腔用組成物;
6 . ポリグリセリン脂肪酸エステルまたはショ糖脂肪酸エステルの脂肪酸部 分のアルキル鎖長が C 8— C 1 6である前記 5に記載の口腔用組成物;
7 . 界面活性剤がベタィンである前記 1または 2に記載の口腔用組成物;
8 . ベタインが脂肪酸ァミドプロピルべタインである前記 Ίに記載の口腔用 組成物;
9 . 脂肪酸ァミドプロピルべタインの脂肪酸部分のアルキル鎖長が C 8— C 1 6である前記 8に記載の口腔用組成物;および
1 0 . さらに、 カチオン性殺菌剤を含む前記 1一 9いずれか 1に記載の口腔用 組成物を提供する。
本発明の第一の態様によれば、 保型性、 口腔内での分散性、 齒磨後のジュース の味を変えず、 特に経時安定性に優れた、 またはそれらの特徴に加えてカチオン 性殺菌剤の歯面滞留性が向上した口腔用組成物を提供することができる。
また、 本発明は、 その第二の態、様として、
1 1 . カチオン性殺菌剤と結晶セルロースを含む口腔用組成物;
1 2 . カチオン性殺菌剤が第四級アンモニゥム塩である前記 1 1に記載の口 腔用組成物;
1 3 . カチオン性殺菌剤がビグァニド系殺菌剤である前記 1 1に記載の口腔 用組成物;
1 4 . カチオン性殺菌剤が塩化セチルビリジニゥム、 塩ィ匕ベンザルコニゥム、 塩化べンゼトニゥム、 塩酸ク口ノレへキシジンおょぴダルコン酸クロルへキシジン よりなる群から選択される 1種以上である前記 1 1に記載の口腔用組成物;
1 5 . カチオン性殺菌剤を 0 .0 0 1— 1 0重量0 /0含む前記 1 1— 1 4いず れか 1に記載の口腔用組成物;
1 6 . 結晶セルロースを 0 . 2— 1 0重量0/。含む前記 1 1 - 1 5いずれか 1 に記載の口腔用組成物;
1 7 . さらに、 非イオン性または両イオン性界面活性剤から選択される 1種 以上の界面活性剤を含む前記 1 1— 1 6いずれか 1に記載の口腔用組成物; 1 8 . 界面活性剤が、 アルキル鎖長 C 8 - 1 6のアルキルグリコシドである 前記 1 7に記載の口腔用組成物;および
1 9 . 界面活性剤が、 脂肪酸部分のアルキノレ鎖長が C 8— 1 6である脂肪酸 アミ ドプロピルべタインである前記 1 7に記載の口腔用組成物を提供する。
本発明の第二の態様によれば、 カチオン性殺菌剤の歯面滞留効果が顕著に向上 し、 歯周疾患および齲蝕等の口腔疾患を効果的に予防し得る口腔用組成物を提供 することができる。 発明を実施するための最良の形態
以下、 本発明の第一および第二の態様を順次説明する。
本発明の第一の態様で用いる結晶セルロースは、 通常、 商業的に入手できるも のであれば特に制限されないが、 結晶セルロースの平均粒径は 1 0 m以下が好 ましく、 中でも 2— 6 μ πιが最も好ましい。 結晶セルロースの平均粒径が 1 0 /x mを超えると口腔用組成物の口腔内での分散性が悪くなる。 さらに結晶セルロー スの配合量は口腔用組成物全量に対して 0 . 2— 1 0重量%が好ましい。 結晶セ ルロースの配合量が 0 . 2重量%より少ないと十分な保型性が得られず、 一方 1 0重量%より多いと口腔用組成物の粘性が大きくなりすぎるので好ましくない。 本発明の第一の態様で用いる界面活性剤は、 アルキルグリコシド、 ポリグリセ リン脂肪酸エステル、 ショ糖脂肪酸エステルまたはべタインであり、 これらは 1 種だけでも 2種以上を併用してもよい。 界面活性剤の配合量は口腔用組成物全量 に対して 0 . 5— 5重量%が好ましい。 界面活性剤の配合量が 0 . 5重量%より少 ないと口腔用組成物の発泡性が小さくなり使用感が低下し、 一方 5重量% り多 いと界面活性剤原料由来の味や臭いが無視できなくなるので好ましくない。 これら界面活性剤のうち、 本発明で用いるアルキルグリコシドは、 特に制限さ れないが、 アルキル鎖長は C 8— C 1 6が好ましい。 アルキル鎖長が C 8より短 いと苦味が生じ、 一方 C 1 6より長いと発泡性が低下するので口腔用糸且成物とし ての使用が耐えがたくなることがある。 この鎖長範囲に入る具体例としては、 デ シルグリコシド、 ラウリルグリコシド、 ミリスチルダリコシド等があり、 P L A N T A C AR E 1 2 0 0、 P L AN T A C A R E 2 0 0 0 (コグニス) 、 O r a m i x NS 10 O r am i x NS 26 (SEPP I C) 等が商業的に 入手できる。
また、 本発明の第一の態様で用いるポリグリセリン脂肪酸エステルは、 特に制 限されないが、 脂肪酸部分のアルキル鎖長は C 8— C 16が好ましい。 アルキル 鎖長が C 8より短いと苦味が生じ、 一方 C 16より長いと発泡性が低下する傾向 がみられる。 また、 ポリグリセリン部分の重合度は 4以上が好ましい。 重合度が 3以下だと発泡性が低下する傾向がある。 かかるポリダリセリン脂肪酸エステル の具体例としては、 デカグリセリンモノラウリン酸エステル、 テトラグリセリン モノラウリン酸エステル、 デカグリセリンモノミリスチン酸エステル、 テトラグ リセリンモノミリスチン酸エステル等があり、 N I KKOL De c a g l y n

また、 本努明の第一の態様で用いるショ糖脂肪酸エステルは、 特に制限されな いが、 脂肪酸部分のアルキル鎖長は C 8— C 16が好ましい。 C8より短いと苦 味が生じ、 一方 C 16より長いと発泡性が低下し、 油味が生じることがある。 か かるショ糖脂肪酸エステルの具体例としては、 ショ糖ラウリン酸エステル、 ショ 糖パルミチン酸エステル等があり、 DKエステル Sシリーズ (第一工業製薬) 、 リヨ一ドーシュガーエステル (三菱化学) 等が商業的に入手できる。
また、 本発明の第一の態様で用いるベタイン界面活性剤は、 特に制限されない

さらに、 本発明の第一の態様で用いるカチオン性殺菌剤は、 特に制限されない 力 第四級アンモニゥム塩およびビグアニド系殺菌剤が好ましく、 例えば塩化セ チルピリジニゥム、 塩化ベンザルコニゥム、 塩化べンゼトニゥム、 塩化ジステア リルジメチルアンモニゥム、 塩化ステアリルジメチルベンジルアンモニゥム、 塩 化ステアリノレト リメチルアンモニゥム、 塩ィヒセチルトリメチルアンモ-ゥム、 塩 化ラウリルトリメチルアンモニゥム、 塩化ラウリルピリジニゥム等の第四級アン モニゥム塩や、 塩酸クロルへキシジン、 酢酸クロルへキシジン、 ダルコン酸クロ ルへキシジン、 塩酸ァレキシジン、 酢酸ァレキシジン、 ダルコン酸ァレキシジン 等のビグアニド系殺菌剤等のカチオン性殺菌剤が含まれる。 これらの中でも、 塩 化セチルピリジニゥムおよび塩化べンザルコニゥムがより好ましく、 塩ィ匕セチル ピリジ-ゥムが特に好ましい。 これらのカチオン性殺菌剤は単独または 2種以上 を組合せて配合し得る。 また、 カチオン性殺菌剤の配合量は、 口腔用組成物全量 に対して、 好ましくは 0 . 0 0 1— 1 0重量 °/0、 より好ましくは 0 . 0 1— 1重 量%である。 配合量が 0 . 0 0 1重量%よりも.少ないと殺菌作用が期待できず、 一方 1 0重量%よりも多いと口腔粘膜に対する刺激が強くなり、 安全性上好まし くない。
本発明の第一の態様の口腔用組成物は練歯磨、 潤性歯磨、 液状歯磨、 口腔用パ スタ、 ジエル、 スプレー、 泡などの剤型に調製することができる。 本宪明の第一 の態様の口腔用組成物にはそれら剤型の相違により、 それらの種類に応じた適宜 な成分、 例えば有効成分、 発泡剤または洗浄剤、 研磨剤、 粘結剤、 湿潤剤、 保存 剤、 香味剤、 甘味料、 ; p H調整剤等の適当な成分を本発明の効果を損なわない範 囲で配合することができる。
このうち、 有効成分としては、 トリクロサンやイソプロピルメチルフエノール 等の非イオン'性殺菌剤、 フッ化ナトリウム、 フッ化カリウム、 フッ化アンモェゥ ム、 フッ化スズ、 モノフルオルリン酸ナトリゥム等のフッ化物、 アミラーゼ、 プ 口テアーゼ、 リゾチーム、 デキストラナーゼ等の酵素、 ビタミン B、 C、 E等の ビタミン類、 力リゥム塩等が挙げられる。

研磨剤としては、 リン酸水素カルシウム■ 2水和物および無水物、 リン酸カル シゥム、 第 3リン酸カ^/シゥム、 第 3リン酸マグネシウム、 ピロリン酸カ シゥ ム、 ハイドロキシァパタイト、 不溶性メタリン酸ナトリゥム、 含水ケィ酸、 無水 ケィ酸、 シリカゲル、 沈降性シリカ、 ケィ酸アルミニウム、 ケィ酸ジルコニウム、 ケィ酸カルシウム、 炭酸カルシウム、 炭酸マグネシウム、 アルミナ、 水酸化アル ミニゥム、 硫酸カルシウム、 ポリメタクリル酸メチル等が挙げられる。
粘結剤としては、 本発明で用いる結晶セルロース以外に、 カルボキシメチルセ ノレロースナトリ ゥム、 力ルポキシメチノレヒドロキシェチルセノレロースナトリゥム 等のァニオン性粘結剤、 ヒドロキシェチルセルロース、 ヒドロキシプロピルセル ロース等のセノレロース誘導体、 キサンタンガム、 トラガントガム、 カラャガム、 ァラビヤガム、 カラギーナン等の'天然ゴム類や、 塩ィ匕 Q— [ 2—ヒドロキシー 3— (トリメチノレアンモニォ) プロピル] ヒ ドロキシェチルセルロース等のカチ オン性粘結剤が挙げられるが、 本発明の第一の態様の口腔用組成物がカチオン性 殺菌剤を含む場合には、 ァニオン性粘結剤を配合することは好ましくない。
湿潤剤としては、 グリセリン、 プロピレングリコール、 1 , 3—ブチレングリ コール、 ソルビトール、 ポリエチレングリコール、 キシリ トール、 ポリプロピレ ングリコール等が挙げられる。

香味剤としては、 メントール、 カルボン、 オイゲノール、 サリチル酸メチル、 メチノレオィゲノーノレ、 チモーノレ、 ァネトーノレ、 リモネン、 オシメン、 n一デシノレ アルコーノレ、 シトロネ一ノレ、 <¾一テルピネオール、 メチノレアセタート、 シトロネ 二ルァセタート、 シネオール、 リナロール、 ェチルリナロール、 ヮニリン、 タイ ム、 ナツメグ、 スペアミント油、 ペパーミント油、 レモン油、 オレンジ油、 セー ジ油、 ローズマリー油、 珪皮油、 シソ油、 冬緑油、 丁子油、 ユーカリ油、 ピメン ト油、 ティーツリー油、 タバナ油等が挙げられる。
甘味料としては、 サッカリンナトリウム、 アセスルファームカリウム、 ステビ ォサイド、 ネオヘスペリジルジヒドロカルコン、 グリチルリチン、 ペリラルチン、 タウマチン、 ァスパラチルフエ二ルァラニルメチルエステル、 メ トキシシンナミ ックアルデヒド、 キシリット等が挙げられる。
p H調整剤としては、 クェン酸およびその塩、 リン酸およびその塩、 リンゴ酸 およびその塩、 ダルコン酸およびその塩、 マレイン酸およびその塩、 ァスパラギ ン酸およびその塩、 ダルコン酸およびその塩、 コハク酸おょぴその塩、 グルクロ ン酸およびその塩、 フマル酸およびその塩、 グルタミン酸およびその塩、 アジピ ン酸およびその塩、 塩酸、 水酸化ナトリウム、 水酸化カリウム、 ケィ酸ナトリウ ム等が挙げられる。
これらの成分は単独または 2種以上を組合せて本発明の第一の態様の口腔用組 成物に含ませることができる。
つぎに、 本発明の第二の態様で用いるカチオン性殺菌剤は、 特に制限されない 力 第四級アンモニゥム塩およびビグアニド系殺菌剤が好ましく、 例えば塩化セ チルピリジニゥム、 塩化ベンザルコニゥム、 塩化べンゼトニゥム、 塩化ジステア リルジメチルアンモニゥム、 塩化ステアリルジメチルペンジルアンモニゥム、 塩 化ステアリルトリメチルアンモニゥム、 塩化セチルトリメチルアンモ-ゥム、 塩 ィ匕ラウリノレトリメチルアンモニゥム、 塩化ラウリルピリジニゥム等の第四級アン モニゥム塩や、 塩酸クロルへキシジン、 酢酸クロルへキシジン、 ダルコン酸クロ ノレへキシジン、 塩酸ァレキシジン、 酢酸ァレキシジン、 ダルコン酸ァレキシジン 等のビグァニド系殺菌剤等のカチオン性殺菌剤が含まれる。 これらのカチオン性 殺菌剤は単独または 2種以上を組合せて配合し得る。 また、 カチオン性殺菌剤の 配合量は、 口腔用組成物全量に対して、 好ましくは 0 . 0 0 1— 1 0重量%、 よ り好ましくは 0 . 0 1— 1重量%である。 配合量が 0. 0 0 1重量%よりも少ない と殺菌作用が期待できず、 一方 1 0重量。 /0よりも多いと口腔粘膜に対する刺激が 強くなり、 安全性上好ましくない。
また、 本発明の第二の態様で用いる結晶セルロースは、 通常、 商業的に入手で きるものであればよく、 この結晶セルロースの配合量は、 口腔用組成物全量に対 して、 好ましくは 0 . 2— 1 0重量0 /0、 より好ましくは 0 . 5— 5重量%である。 結晶セルロースの配合量が 0 . 2重量。 /。よりも少ないと殺菌剤の歯面滞留性を向 上させる効果が小さくなり、 一方 1 0重量%ょりも多いと組成物の粘性が大きく なりすぎるので好ましくない。 また、 結晶セルロースの平均粒径は組成物中で均 —に分散させる観点より 1 0 μ πι以下が好ましく、 2— 6 μ πιがより好ましい。 また、 0 . 1 / mよりも平均粒径が小さなものは現実的に入手困難である。
また、 本発明の第二の態様で用いる界面活性剤としては、 非イオン性界面活性 剤、 両性界面活性剤、 カチオン性界面活性剤が好ましい。 ァニオン性界面活性剤 を用いると製剤中でのカチオン性殺菌剤の安定性を損なう場合がある。 より好ま しくは、 非イオン性界面活す生剤、 両性界面活性剤である。 非イオン性界面活性剤 としては、 例えば、 アルキルグリコシド、 ショ糖脂肪酸エステル、 マルトース脂 肪酸エステル、 ラクトース脂肪酸エステル等の糖脂肪酸エステル、 ポリオキシェ チレンアルキルエーテル類、 脂肪酸アル力ノールァミド類、 ポリォキシエチレン ソルビタンモノラウレート、 ポリオキシエチレンソルビタンモノステアレート等 のポリォキシェチレンソルビタン脂肪酸エステル、 ポリォキシェチレンヒマシ油、 ソルビタン脂肪酸エステル、 デカグリセリンモノラウリン酸エステル、 ペンタグ リセリンジステアリン酸エステル等のポリグリセリン脂肪酸エステル、 ポリォキ シエチレンポリォキシプロピレン共重合体等が挙げられる。 両性界面活性剤とし ては、 例えば、 N—ラウリルジァミノェチルダリシン、 N—ミリスチルジァミノ ェチルダリシン等の N—アルキルジァミノェチルダリシン、 脂肪酸ァミ ドプロピ ノレべタイン、 N—アクレキノレー N—カノレポキシメチノレアンモニゥムベタイン、 2— アルキル一 1ーヒドロキシェチルイミダゾリンべタインナトリゥム等が挙げられ る。 中でも、 アルキルグリコシド、 ショ糖脂肪酸エステル、 ポリオキシエチレン ヒマシ油、 ポリグリセリン脂肪酸エステル、 ポリォキシエチレンポリォキシプロ ピレン共重合体、 N—アルキルジァミノェチルダリシン、 脂肪酸ァミ ドプロピル ベタィンが好ましい。 中でも特にアルキルグリコシドゃ脂肪酸ァミ ドプロピルべ タインが好ましい。 さらにアルキルグリコシドのアルキル鎖長は C 8 _ C 1 6、 特に C 1 0— C 1 4が好ましく、 脂肪酸ァミ ドプロピルベタィンの脂肪酸部分の アルキル鎖長は C 8— C 1 6、 特に C 1 2— C 1 4が好ましい。 これら界面活性 剤の配合量は、 口腔用組成物全量に対して、 好ましくは 0 . 5— 5重量%である。 本発明の第二の態様の口腔用組成物は練歯磨、 潤性歯磨、 液状歯磨、 口腔用パ スタ、 ジエル類などの剤型に調製することができる。 本発明の第二の態様の口腔 用組成物にはそれら剤型の相違により、 それら種類に応じた適宜な成分、 例えば 有効成分、 研磨剤、 粘結剤、 湿潤剤、 保存剤、 香味剤、 甘味料、 P H調整剤等の 適当な成分を本発明の効果を損なわない範囲で配合することができる。
このうち、 有効成分としては、 前記の第四級アンモニゥム塩ゃビグアニド系等 のカチオン性殺菌剤のほか、 トリクロサンやイソプロピルメチルフエノール等の 非イオン性殺菌剤、 フッ化ナトリウム、 フツイ匕カリウム、 フッ化アンモニゥム、 フッ化スズ、 モノフルオルリン酸ナトリウム等のフッ化物、 アミラーゼ、 プロテ ァーゼ、 リゾチーム、 デキストラ^ "一ゼ等の酵素、 ビタミン B、 C、 E等のビタ ミン類、 硝酸カリウム、 乳酸アルミニウム等の収斂剤等が挙げられる。
研磨剤としては、 リン酸水素カルシウム · 2水和物おょぴ無水和物、 リン酸力 ノレシゥム、 第 3リン酸カルシウム、 第 3リン酸マグネシウム、 ピロリン酸カルシ ゥム、 ハイドロキシァパタイト、 不溶性メタリン酸ナトリウム、 含水ケィ酸、 無 水ケィ酸、 シリカゲル、 沈降性シリカ、 ケィ酸アルミニウム、 ケィ酸ジノレコユウ ム、 ケィ酸カルシウム、 炭酸カルシウム、 炭酸マグネシウム、 ァノレミナ、 水酸化 ァノレミニゥム、 硫酸カルシウム、 ポリメタクリル酸メチル等が挙げられる。 これ らの中でも、 リン酸水素カルシウム . 2水和物および無水和物、 リン酸カルシゥ ム、 第 3リン酸カルシウム、 第 3リン酸マグネシウム、 ピロリン酸カルシウム、 ハイドロキシァパタイ ト、 炭酸カルシウムまたは炭酸マグネシウムが好ましい。 粘結剤としては、 本発明で用いる結晶セルロース以外に、 ヒドロキシェチルセ ノレロース、 ヒドロキシプロピルセルロース等のセノレロース誘導体、 カラギーナン、 キサンタンガム、 トラガントガム、 カラャガム、 ァラビヤガム、 ジエランガム等 の天然ゴム類、 ポリビュルアルコール、 ポリアクリル酸ナトリゥム等の合成粘結 剤、 増粘性シリカ、 ビーガム等の無機粘結剤等が挙げられる。

表 1の結果より、 界面活性剤にプルロニック F 88を用いた比較例 1 - 3にお いて、 ヒドロキシェチルセルロース配合量を増加させても、 他の粘結剤であるキ サンタンガムを併用しても、 固液分離が生じた。 また界面活性剤を HCO— 60 にしても固液分離を生じた。
一方、 実施例 1一 4において、 界面活性剤にラゥリルグリコシド、 ポリグリセ リンラウリン酸エステル、 ショ糖ラゥリン酸エステル、 ヤシ油脂肪酸ァミドプロ ピルベタインを用レ、た口腔用組成物は室温で 1ヶ月保管しても固液分離は見られ ず、 経時安定性が優れていた。 実施例 5

試験方法 ァパタイト粉末への塩ィヒセチルピリジニゥム滞留量の測定 紫外線滅菌したヒト唾液 2m 1にヒドロキシァパタイト (DNA G r a d e B i o-Ge l HTP ; B I O— RAD製) 50 m gを 37°Cにて 1 5時間浸 漬して人工ペリクルをァパタイト表面に形成させ、 その後遠心 (3000 r pm、 10分間) し、 上清を捨てた。 次に塩ィ匕セチルピリジニゥム (CPC) 0.3重 量%、 種々の粘結剤および界面活性剤を種々の量配合した実施例 14一 18およ び比較例 5— 8の口腔用組成物の 4倍希釈スラリ一の上清 2 m】に上記ヒドロキ シァパタイトを 37°Cにて 15分間浸漬した。 遠心 (3000 r pm、 10分 間) 後、 上清を捨て、 蒸留水 2m 1を添加し攪拌後、 遠心 (3000 r pm、 1 0分間) した。 さらに上清を捨て、 蒸留水 2m 1を添加し攪拌後遠心 (3000 r m 10分間) し、 上清を捨てた。 次に下記の抽出液を用いヒドロキシァパ タイトに吸着している CPCを抽出し、 HPLCで定量してヒドロキシァパタイ ト 5 Omgに滞留した CP C量を求めた。 結果をまとめて表 2に示す。 抽出液を示す。 pH3の 0.02Mクェン酸緩衝液 1 Lあたり 2.88 gのラウリ ノレ硫酸ナトリウムを溶解させた溶液:ァセトニトリル = 1 : 3

表 2の結果より、 塩ィ匕セチルピリジニゥムおよぴ結晶セノレロースを特異的に組 合せた実施例 14一 18の口腔用組成物では、 同量の塩化セチルビリジニゥムぉ よび他のセルロース誘導体を組合せた比較例 5一 8の口腔用組成物と比較して、 塩化セチルビリジニゥムの歯面滞留量が顕著に増加することが明らかとなった。 また、 これらの成分に加えて配合し得る界面活性剤としては、 アルキルグリコシ ドゃベタインが他の界面活性剤と比較して塩ィ匕セチルピリジニゥム滞留量を増加 させ、 好ましいことが明らかとなった。 実施例 19

得られた口腔用組成物は、 カチオン性殺菌剤の歯面滞留量を向上させ、 歯周疾 患および齲蝕等の口腔疾患を効果的に予防し得た。 産業上の利用の可能性
本発明の第一の態様により、 保型性、 口腔内での分散'性、 歯磨後のジュースの 味を変えずに、 特に経時安定性に優れた、 また、 カチオン性殺菌剤の歯面滞留効 果が向上した口腔用組成物を提供することができる。
また、 本発明の第二の態様により、 カチオン性殺菌剤の歯面滞留効果が顕著に 向上し、 歯周疾患および齲蝕等の口腔疾患を効果的に予防し得る口腔用組成物を 提供することができる。

Claims

請 求 の 範 囲
I . 結晶セルロースと、 アルキルグリコシド、 ポリグリセリン脂肪酸エステル、 ショ糖脂肪酸エステルおよびべタインからなる群より選ばれる 1種以上の界面活 性剤とを含む口腔用組成物。
2 . 結晶セルロースを 0 . 2— 1 0重量%配合する請求項 1に記載の口腔用組 成物。
3 . 界面活性剤がアルキルグリコシドである請求項 1または 2に記載の口腔用 組成物。
4. アルキルグリコシドのアルキル鎖長が C 8— C 1 6である請求項 3に記载 の口腔用組成物。
5 . 界面活性剤がポリグリセリン脂肪酸エステルまたはショ糖脂肪酸エステル である請求項 1または 2に記載の口腔用組成物。
6 . ポリグリセリン脂肪酸エステルまたはショ糖脂肪酸エステルの脂肪酸部分 のアルキル鎖長が C 8— C 1 6である請求項 5に記載の口腔用組成物。
7 . 界面活性剤がべタインである請求項 1または 2に記載の口腔用組成物。
8 . ベタインが脂肪酸ァミドプロピルべタインである請求項 7に記載の口腔用 組成物。
9 . 脂肪酸ァミドプロピルベタィンの脂肪酸部分のアルキル鎖長が C 8— C 1 6である請求項 8に記載の口腔用組成物。
1 0 . さらに、 力チオン性殺菌剤を含む請求項 1一 9レ、ずれか 1項に記載の口 腔用組成物。
I I . カチォン性殺菌剤と結晶セルロースを含む口腔用組成物。
1 2. カチオン性殺菌剤が第四級ァンモニゥム塩である請求項 1 1に記載の口 腔用組成物。
1 3 . カチオン性殺菌剤がビグァニド系殺菌剤である請求項 1 1に記載の口腔 用糸且成物。
1 4 . カチオン性殺菌剤が塩化セチルピリジニゥム、 塩化ベンザノレコニゥム、 塩化べンゼトニゥム、 塩酸クロルへキシジンおよびダルコン酸クロルへキシジン よりなる群から選択される 1種以上である請求項 1 1に記載の口腔用組成物。
15. カチォン性殺菌剤を 0.001— 10重量%含む請求項 1 1 _ 14いず れか 1項に記載の口腔用組成物。
16. 結晶セルロースを 0.2 _ 10重量%含む請求項 1 1 - 15いずれか 1 項に記載の口腔用組成物。
17. さらに、 非ィオン性または両ィオン性界面活性剤から選択される 1種以 上の界面活性剤を含む請求項 1 1一 16いずれか 1項に記載の口腔用組成物。
18. 界面活性剤が、 ァルキル鎖長 C 8— 16のアルキルグリコシドである請 求項 17に記載の口腔用組成物。
19. 界面活性剤が、 脂肪酸部分のアルキル鎖長が C 8- 16である脂肪酸ァ ミドプロピルベタィンである請求項 1 7に記載の口腔用組成物。
