Verfahren und Vorrichtung zur biotechnologischen Herstellung von Wertstoffen bei dem einem Bioreaktor ein Medium zugefüttert und einem Fermentationsprozess unterzogen wird und bei dem über eine nachgeschaltete Querstromfiltrationsanlage der Wertstoff als gefiltertes Permeat und/oder als konzentriertes Retentat geerntet wird und Rückstände dem Bioreaktor bis zur Ernte als Retentat wieder zugeführt werden, wobei neben dem Medium weitere Stoffe dem Bioreaktor kontrolliert zugefüttert werden können, wobei das konzentrierte Retentat und das Permeat kontrolliert geerntet werden können, und wobei über eine digitale Kontrolleinheit der Fermentationsprozess und die Filtration in einem integrierten System aufeinander abgestimmt geregelt werden.

Description

Verfahren und Vorrichtung zur biotechnologischen Herstellung von Wertstoffen
Beschreibung
Die Erfindung betrifft ein Verfahren zur biotechnologischen Herstellung von Wertstoffen, bei dem einem Bioreaktor ein Medium zugefüttert und einem Fermentationsprozess unterzogen wird, und bei dem über eine nachgeschaltete Querstromfiltra- tionsanlage der Wertstoff als gefiltertes Permeat und / oder als konzentriertes Retentat geerntet wird und Rückstände dem Bioreaktor bis zur Ernte als Retentat wieder zugeführt werden.
Die Erfindung betrifft weiterhin eine Vorrichtung zur bio- technologischen Herstellung von Wertstoffen, im Wesentlichen bestehend aus einem Bioreaktor mit einem vorgeschalteten ersten Zufütterbehälter für ein Medium und einer nachgelagerten Querstromfiltrationsanlage, deren Permeatleitung mit einem ersten Erntebehälter verbunden ist und deren Retentatleitung in den Bioreaktor zurückführt.
Ein Verfahren zur biotechnischen Herstellung von Wertstoffen ist beispielsweise aus der EP 0 307 737 Bl bekannt. Insbesondere für die Herstellung rekombinanter Proteine ergibt sich jedoch ein Widerspruch zwischen einer möglichst hohen Zellproduktivität (Hochzelldichtekultivierung) und einer langen Standzeit der Membranen (Cross-Flow-Membranen) von Querstrom- mikrofiltrationsanlagen. Insbesondere kann es bei einer Erhöhung des Permeatfluxes über einen bestimmten Grenzwert bei gegebener Biomassekonzentration in der Produktlösung zu einem dramatischen Anstieg des Transmembrandruckes und damit zu ei- nem Zusetzen der Membranporen, zu einem sogenannten Membran- fouling, kommen.
Aufgabe der vorliegenden Erfindung ist es daher, den Fermen- tierungs- und Filtrationsprozess so zu verbessern, dass bei möglichst hoher Zellproduktivität eine möglichst lange Standzeit der Membranen der Querstromfiltrationsanlage erreicht werden kann.
Diese Aufgabe wird in Verbindung mit dem Oberbegriff des Anspruches 1 dadurch gelöst, dass neben dem Medium weitere Stoffe dem Bioreaktor kontrolliert zugefüttert werden können, dass das konzentrierte Retentat und das Permeat kontrolliert geerntet werden können und dass über eine Kontrolleinheit der Fermentationsprozess und die Filtration in einem integrierten System aufeinander abgestimmt geregelt werden.
Dadurch, dass der Fermentationsprozess und die Filtration in einem integrierten System aufeinander abgestimmt geregelt werden, so dass insbesondere die Zufütterung von Stoffen und die Ernte kontrolliert erfolgen kann, wird zuverlässig erreicht, dass kritische Werte, die die Standzeit der Membranen verringern könnten, vermieden werden. Insbesondere ist es so möglich, den Überströmdruck durch den die Produktionslösung, die die WertStoffe enthält, an der Membran vorbeigeführt wird, größer zu halten als den Transmembrandruck quer zur Membran, wodurch sich die Standzeit der Membran erhöht.
Gemäß einer bevorzugten Ausführungsform der Erfindung kann das integrierte System von der digitalen Kontrolleinheit gesteuert einer in-situ-Reinigung und Sterilisation unterzogen werden. Dadurch wird eine schnelle und sichere Reinigung und Sterilisation ermöglicht. Gemäß einer weiteren bevorzugten Ausführungsform der Erfindung werden als Wertstoffe rekombinante Proteine hergestellt, wobei das Permeat eine zellfreie Ernte und das Retentat eine zellbehaftete Ernte ergibt. Der Prozessablauf kann dabei im Rahmen einer sequentiellen integrierten Prozessführung erfolgen. In einer Batchphase können dabei dem Bioreaktor zugeführte Zellen an das Medium adaptieren und in einer anschließenden Fed Batchphase die Zellen durch Zufütterung mit konstanter Wachstumsrate angezogen werden. In einer anschließen- den Produktionsphase erfolgt durch Zugabe eines Induktionsstoffes die Induktion der Produktbildung und die eigentliche Herstellung der rekombinanten Proteine. Die Konzentration des Induktionsstoffes kann dabei vorteilhaft über eine Fließdiffusionsanalyse gemessen und über Zufütterung aus einer Vorla- ge geregelt werden. In einer an die Produktionsphase anschließenden Produkterntephase wird dann ein Teil des Bioreaktors zellfrei abgeerntet. Die Zellmasse des Retentats wird in einer Zellerntephase abgeerntet, der sich eine Medium- refreshphase mit einer Zufütterung anschließen kann. Nach der Mediumrefreshphase beginnt der eigentliche zyklische Prozess, bei dem außer in der Produkternte nur der Retentat- aber nicht der Permeatstrom fließen soll, mit der Produktionsphase neu.
Gemäß einer weiteren bevorzugten Ausführungsform der Erfindung werden die rekombinanten Proteine unter Verwendung der Hefe Pichie pastoris hergestellt. Die Hefe ist ähnlich leicht zu kultivieren wie E.coli, sie ist aber als Eukaryot viel besser für eine korrekte Faltung der rekombinanten Proteine geeignet. Weiterhin ist sie fähig, Proteine zu glycolisieren, was wichtig für deren strukturelle Vollständigkeit, Löslichkeit und biologische Aktivität ist. Darüber hinaus können Hefenproteine über die Zellwand ausscheiden (Sekretion) damit die Trennung der gewünschten Produkte von zellulären Bestand- teilen stark erleichtern. Nach einer weiteren bevorzugten Ausführungsform der Erfindung wird zur Induktion der Sequenzen des Zellproteins als Induktionsstoff Methanol in das Medium des Bioreaktors zugegeben. Dabei wird die Methanolkonzentration auf einem konstanten Le- vel gehalten.
Da die Sequenzen des Zielproteins in den nativen Genabschnitt zur Expression einer Alkoholoxidase (AOX) von P. pastoris integriert werden, erfolgt durch eine Zugabe von Methanol in das Medium deren Induktion.
Dadurch, dass die Methanolkonzentration auf einem möglichst konstanten Level im unteren Gramm / Liter-Bereich gehalten wird, wird eine Überfütterung, die toxisch wirken könnte, vermieden. Durch eine Online-Messung und Regelung der Methanolkonzentration über die erwähnte Fließdiffusionsanalyse wird der konstante Level der Methanolkonzentration ermöglicht.
Nach einer weiteren bevorzugten Ausführungsform der Erfindung wird in der Fed Batchphase und / oder in der Produktionsphase zur Produktionssteigerung Glycerol zugefüttert.
Gemäß einer weiteren bevorzugten Ausführungsform der Erfin- düng verläuft der Prozessablauf im Rahmen einer kontinuierlichen integrierten Prozessführung. Dabei laufen die Produktionsphase, die Produkterntephase und die Zellerntephase parallel ab. Damit wird eine permanente Produkt- und Turbidostat- zellernte, letztere zur Aufrechterhaltung der Membranfunktio- nalität, ermöglicht.
Da für P. pastoris geeignete sekretorische Gensequenzen zur Verfügung stehen, können die gewünschten Produkte mittels integrierten Bioprozess hergestellt werden. Dabei können sowohl prozessvorbereitende Schritte (up stream) , beginnend mit der Konstruktion produktionsgeeigneter Expressionssysteme bis hin zur Vorkulturführung, als auch nachfolgender Primäraufbear- beitungsschritte (down stream) in die eigentlich Reaktionsführung, d.h. Zellkultivierung und Produktbildung, eingebun- den werden. Durch diese Prozessführung werden die umweltbelastenden Aufbearbeitungsschritte einer Proteinprozessierung mit E.coli vermieden. Die Produkternte während des Kultivierungsablaufes kann hier direkt in nachfolgenden Feinreini- gungsschritten für die korrekt prozessierten Proteine über- führt werden.
Die beispielsweise aus der EP 0 307 737 Bl bekannte Vorrichtung weist die für die bekannten Verfahren beschriebenen Nachteile auf.
Weitere Aufgabe der Erfindung ist es daher, die bekannten Vorrichtungen so zu verbessern, dass der bei der Herstellung rekombinanten Proteine bekannte Widerspruch einer möglichst hohen Zellproduktivität und einer langen Standzeit der Memb- ranen gelöst wird.
Diese Aufgabe wird in Verbindung mit dem Oberbegriff des Anspruches 17 dadurch gelöst, dass mindestens ein zweiter Zu- fütterbehälter mit einem Induktionsstoff dem Bioreaktor vor- geschaltet ist, dass ein zweiter Erntebehälter für eine zellbehaftete Ernte des Retentats über eine Ernteleitung mit dem Biorektor verbunden ist und dass eine Kontrolleinheit zur Messung und Regelung des Fermentations- und Filtrationsprozesses angeordnet ist.
Durch die (digitale) Kontrolleinheit zur Messung und Regelung des Fermentations- und Filtrationsprozesses wird ein optimaler Prozessverlauf erzielt, der bei einer hohen Zellproduktivität eine lange Standzeit der Membranen ermöglicht. Gemäß einer bevorzugten Ausführungsform der Erfindung weist zur Messung der Konzentration des Induktionsstoffes im Biore- aktor- die Kontrolleinheit ein Analysesystem auf, das über einen im Bioreaktor angeordneten Sensor die Konzentration des Induktionsstoffes misst und durch Steuerung einer dem zweiten Zufütterbehälter vorgeschalteten Fütterpumpe die Induktionsstoffkonzentration im Bioreaktor regelt. Insbesondere bei einer kontinuierlichen integrierten Prozessführung ist das Analysesystem dabei als ein Fließdiffusionsanalysesyste ausge- bildet. Dadurch wird vorteilhaft eine kontinuierliche Messung und Regelung ermöglicht.
Gemäß einer weiteren bevorzugten Ausführungsform der Erfindung weist zur Messung einer Zellkonzentration im Bioreaktor die Kontrolleinheit ein zweites Analysesystem auf, das über einen im Bioreaktor angeordneten zweiten Sensor die Zellkonzentration misst und durch Steuerung einer dem zweiten Erntebehälter vorgeschalteten Erntepumpe die Zellkonzentration im Bioreaktor regelt.
Die Kontrolleinheit kann sämtliche regeltechnischen Aufgaben, die typisch für einen Fermentationsprozess sind, übernehmen, beispielsweise Messung und Regelung von Temperatur, pH-Wert, p02-Wert über Begasungsrate und Gaszusammensetzung, Rühr- erdrehzahl, Schaumbekämpfung u.s.w.. Die Kontrolleinheit ü- bernimmt auch die Regelung der Parameter der automatisierten Querstromfiltrationsanlage, wie Permeatstrom, Retentatstrom und die automatische in-situ-Reinigung und Sterilisation des integrierten Systems.
Als Querstromfiltrationsanlagen kommen Mikrofiltrations- und Ultrafiltrationsanlagen oder Kombinationen aus Mikro- und Ultrafiltrationsanlagen in Frage. Weitere Einzelheiten der Erfindung ergeben sich aus der nachfolgenden ausführlichen Beschreibung und den beigefügten Zeichnungen, in denen bevorzugte Ausführungsformen der Erfindung beispielsweise veranschaulicht sind .

Eine Vorrichtung zur biotechnologischen Herstellung von Wertstoffen besteht im Wesentlichen aus einem Bioreaktor 1 mit einem vorgeschalteten ersten Zufütterbehälter 2, einem zweiten Zufütterbehälter 3, einem dritten Zufütterbehälter 4 und einer nachgelagerten Querstromfiltrationsanlage 5, sowie einer Kontrolleinheit 6.
Der erste Zufütterbehälter 2 ist über eine erste Zufütterleitung 7 und eine erste Zufütterpumpe 8 mit dem Bioreaktor 1 verbunden. Der zweite Zufütterbehälter 3 ist über eine zweite Zufütterleitung 9 und eine zweite Zufütterpumpe 10 mit dem Bioreaktor 1 verbunden. Der dritte Zufütterbehälter 4 ist ü- ber eine dritte Zufütterleitung 11 und eine dritte Zufütterpumpe 12 ebenfalls mit dem Bioreaktor 1 verbunden.
Die Querstromfiltrationsanlage 5 ist dem Bioreaktor 1 nachge- lagert und über eine Förderleitung 13 mit dem Biorektor 1 verbunden. Zwischen Bioreaktor 1 und Querstromfiltrationsanlage 5 ist eine Förderpumpe 14 angeordnet. Über eine Permeat- leitung 15 und eine Permeatpumpe 25 ist die Querstromfiltrationsanlage 5 mit einem ersten Erntebehälter 16 verbunden. Die Querstromfiltrationsanlage 5 ist über eine Retentatlei- tung 17 mit dem Bioreaktor 1 verbunden. Ein zweiter Erntebehälter 18 ist für eine zellbehaftete Ernte des Retentats über eine Ernteleitung 19 und eine Harvestpumpe 20 mit dem Bioreaktor 1 verbunden.
Die digitale Kontrolleinheit 6 ist über Messleitungen 21 mit den Behältern 2, 3, 4, 16, 18 zugeordneten Wägevorrichtungen 22 verbunden. Über Steuerleitungen 23 ist die Kontrolleinheit 6 mit den Pumpen 8, 10, 12, 14, 20 verbunden.
In Figur 2 ist der Prozessablauf einer sequenziell integrierten Prozessführung für die Herstellung rekombinanter Proteine unter Einsatz der Hefe P. pastoris dargestellt. Aufgetragen sind das Reaktorvolumen VL des Bioreaktors 1 und die Zellkon- zentration cxι (Biotrockenmasse) . In einer Batchphase 29 t e [0,tχ] adaptieren die Zellen an das Medium und werden bis ca. 15 gl-1 angezüchtet. Das Reaktorvolumen VL nimmt durch Probenahme ab.
In einer Fed Batchphase 30 t e [ti, t2] werden die Zellen durch Zufütterung von Glycerol 37 bis 25 gl-1 mit konstanter (substratlimitierter) Wachstumsrate angezogen. In der Produktionsphase 31 t ε [t2, t3] erfolgt durch Zugabe von Methanol 36 als Induktionsstoff zunächst die Induktion der Produktbildung und die eigentliche Herstellung der rekombinanten Proteine.
Die Methanolkonzentration wird mit einem Analysesystem 24 der Kontrolleinheit 6, das als Fließdiffusionsanalyse-System (FDA) ausgebildet ist, gemessen und über Zufütterung aus dem zweiten Zufütterbehälter 3 - der Methanolvorlage - durch Steuerung der zweiten Zufütterpumpe 10 geregelt.
Zur Produktionssteigerung kann in dieser Phase eine geringe Menge Glycerol 37 aus dem dritten Zufütterbehälter 4 durch Steuerung der dritten Zufütterpumpe 12 über die dritte Zufüt- terleitung 11 dem Bioreaktor 1 zugegeben werden. Durch Zugabe von Medium 35 aus dem ersten Zufütterbehälter 2 über die erste Zufütterleitung 7 in den Bioreaktor 1 steigt das Reaktorvolumen VL an und die Zellen wachsen vermindert weiter (im Beispiel bis 30 gl"1) . In der Produkterntephase 32 t e [t3, t4] wird ein Viertel (Menge ist in Grenzen wahlfrei) des Bioreaktors 1 zellfrei als Permeat der Querstromfiltrationsanlage 5 abgeerntet. Das Permeat 38 fließt dabei über die Per- meatpumpe 25 und die Permeatleitung 15 in den ersten Erntebehälter 16. Dadurch steigt die Zellkonzentration bis 40 gl-1 an.
Ab te wird wieder mit dem gleichen Produktions- und Erntezyklus wie bei t2 mit ca. 25 gl"1 gestartet. In einer Zellerntephase 33 t e [t , t5] wird über die Harvestpumpe 20 und die Ernteleitung 19 Zellmasse bzw. Retentat aus dem Bioreaktor 1 in den zweiten Erntebehälter 18 abgelassen. In einer Mediu - refreshphase 34 t e [ts, t6] wird methanol- und glycerolfrei- es Medium 35 aus dem ersten Zufütterbehälter 2 dem Bioreaktor 1 zugegeben. Ab te beginnt der eigentliche zyklische Prozess, bei dem außer in der Produkternte 32 nur der Retentat- aber nicht der Permeatstro fließen soll.
Die hier angegebenen Daten für Zeiten, Prozente u.s.w. sind nur für den untersuchten Prozess gültig. Sie können stark variieren. Eine Ausdehnung auf Zellkulturen ist möglich.
Bei einer kontinuierlichen integrierten Prozessführung laufen die drei Phasen 31, 32, 33 des Produktions- und Erntezyklus t e [t2, tζ] parallel ab. Hierbei handelt es sich um ein ver- maschtes Regelungsproblem, das über die digitale Kontrolleinheit 6 gemessen und geregelt wird.
In Figur 3 ist eine Vorrichtung zur biotechnologischen Herstellung von Wertstoffen mit einer kontinuierlichen integrierten Prozessführung dargestellt .' Hierzu weist die in Figur 3 nicht weiter dargestellte Kontrolleinheit 6 ein Analysesys- tem 24 auf, das über einen im Bioreaktor 1 angeordneten Sensor 26 die Konzentration des Induktionsstoffes, im Beispiel die Methanolkonzentration, misst und durch Steuerung der dem zweiten Zufütterbehälter 3 vorgeschalteten Zufütterpumpe 10 die Induktionsstoff- bzw. Methanolkonzentration im Bioreaktor 1 geregelt. Das Analysesystem 24 ist dabei als ein Fließdiffusionsanalyse-System (FDA) ausgebildet. Über die Fließdiffusionsanalyse wird der Istwert der Methanolkonzentration gemessen und einem ersten Regler 39 zugeführt, der den Istwert mit dem Sollwert eines ersten Sollwertstellers 40 vergleicht und ein Steuersignal an die zweite Zufütterpumpe 10 gibt.
Zur Messung der Zellkonzentration im Bioreaktor 1 weist die Kontrolleinheit 5 ein zweites Analysesystem 27 auf. Das zweite Analysesystem 27 misst über einen im Bioreaktor 1 angeord- neten zweiten Sensor 28 die Zellkonzentration und regelt durch Steuerung einer dem zweiten Erntebehälter 18 vorgeschalteten Harvestpumpe 20 (Ernt'epumpe) die Zellkonzentration im Bioreaktor 1. Über einen mit dem zweiten Sensor 28 verbundenen Analysator 41 wird der Istwert der Zellkonzentration analysiert bzw. gemessen und einem zweiten Regler 42 zugeführt, der den Istwert mit dem Sollwert eines zweiten Sollwertstellers 43 vergleicht und ein Steuersignal an die Harvestpumpe 20 gibt.
Zur Regelung der Mediumzugabe in den Bioreaktor 1 erhält ein dritter Regler 44 von der Wägevorrichtung 22 des Bioreaktors 1 ein Istsignal und vergleicht das Istsignal bzw. den Istwert mit dem Sollwert eines dritten Sollwertstellers 45 und gibt ein entsprechendes Steuersignal an die erste Zufütterpumpe 8.

Claims

Patentansprüche
1. Verfahren zur biotechnologischen Herstellung von Wertstoffen bei dem einem Bioreaktor ein Medium zugefüttert und einem Fermentationsprozess unterzogen wird und bei dem über eine nachgeschaltete Querstromfiltrationsanlage der Wertstoff als gefiltertes Permeat und / oder als konzentriertes Retentat geerntet wird und Rückstände dem Bioreaktor bis zur Ernte als Retentat wieder zugeführt werden, dadurch gekennzeichnet, dass neben dem Medium weitere Stoffe dem Bioreaktor (1) kontrolliert zugefüttert werden können, dass das konzentrierte Retentat und das Permeat kontrolliert geerntet werden können, und dass über eine Kontrolleinheit (6) der Fermentationsprozess und die Filtration in einem integrierten System aufein- ander abgestimmt geregelt werden.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass das integrierte System von der Kontrolleinheit (6) gesteuert einer in-situ-Reinigung und Sterilisation unterzogen werden kann.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass als Wertstoffe rekombinante Proteine hergestellt werden, wobei das Permeat eine zellfreie Ernte und das Retentat eine zellbehaftete Ernte ergibt.
4. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der Prozessablauf im Rahmen einer sequentiellen integrierten Prozessführung erfolgt.
5. Verfahren nach einem der Ansprüche 3 oder 4, dadurch gekennzeichnet, dass in einer Batchphase (29) dem Bioreaktor
(1) zugeführte Zellen an das Medium adaptieren und in einer anschließenden Fed Batchphase (30) die Zellen durch Zufütte- rung mit konstanter Wachstumsrate angezogen werden.
6. Verfahren nach einem der Ansprüche 3 bis 5, dadurch gekennzeichnet, dass in einer Produktionsphase (31) durch Zugabe eines Induktionsstoffes die Induktion der Produktbildung und die eigentliche Herstellung der rekombinanten Proteine erfolgt.
7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, dass die Konzentration des Induktionsstoffes über eine Fließdiffu- sionsanalyse gemessen und über Zufütterung aus einem zweiten Zufütterbehälter (3) geregelt wird.
8. Verfahren nach einem der Ansprüche 4 bis 7, dadurch gekennzeichnet, dass in einer Produkterntephase (32) ein Teil des Bioreaktors (1) zellfrei abgeerntet wird.
9. Verfahren nach Anspruch 8, dadurch gekennzeichnet, dass in einer Zellerntephase (33) Zellmasse des Retentats abgeerntet und eine Mediumrefreshphase (34) mit einer Zufütterung von Medium (35) angeschlossen wird.
10. Verfahren nach Anspruch 9, dadurch gekennzeichnet, dass nach der Mediumrefreshphase (34) der zyklische Prozess, bei dem außer in der Produkternte nur der Retentat- aber nicht der Permeatstrom fließen soll, mit der Produktionsphase (31) neu beginnt.
11. Verfahren nach einem der Ansprüche 3 bis 10, dadurch gekennzeichnet, dass die rekombinanten Proteine unter Verwendung der Hefe Pichie pastoris hergestellt werden.
12. Verfahren nach Anspruch 11, dadurch gekennzeichnet, dass zur Induktion der Sequenzen des Zellproteins als Induktionsstoff Methanol (36) in das Medium (35) des Bioreaktors (1) zugegeben wird.
13. Verfahren nach Anspruch 12, dadurch gekennzeichnet, dass die Methanolkonzentration auf einem konstanten Level gehalten wird.
14. Verfahren nach einem der Ansprüche 11 bis 13, dadurch gekennzeichnet, dass in der Fed Batchphase (30) und / oder in der Produktionsphase (31) zur Produktionssteigerung Glycerol (37) zugefüttert wird.
15. Verfahren nach einem der Ansprüche 1 bis 3 und 5 bis 14, dadurch gekennzeichnet, dass der Prozessablauf im Rahmen ei- ner kontinuierlichen integrierten Prozessführung verläuft.
16. Verfahren nach Anspruch 15, dadurch gekennzeichnet, dass die Produktionsphase (31) , die Produkterntephase (32) und die Zellerntephase (33) parallel ablaufen.
17. Vorrichtung zur biotechnologischen Herstellung von Wertstoffen im Wesentlichen bestehend aus einem Bioreaktor mit einem vorgeschalteten ersten Zufütterbehälter für ein Medium und einer nachgelagerten Querstromfiltrationsanlage, deren Permeatleitung mit einem ersten Erntebehälter verbunden ist und deren Retentatleitung in den Bioreaktor zurückführt, dadurch gekennzeichnet, dass mindestens ein zweiter Zufütterbehälter (3) mit einem Induktionsstoff dem Bioreaktor (1) vorgeschaltet ist, dass ein zweiter Erntebehälter (18) für eine zellbehaftete Ernte des Retentats über eine Ernteleitung (19) mit dem Bioreaktor (1) verbunden ist, und dass eine Kontrolleinheit (6) zur Messung und Regelung des Fermentationsund Filtrationsprozesses angeordnet ist.
18. Vorrichtung nach Anspruch 17, dadurch gekennzeichnet, dass zur Messung der Konzentration des Induktionsstoffes im Bioreaktor (1) die Kontrolleinheit (6) ein Analysesystem (24) aufweist, das über einen im Bioreaktor (1) angeordneten Sensor die Konzentration des Induktionsstoffes misst und durch Steuerung einer dem zweiten Zufütterbehälter (3) vorgeschalteten zweiten Zufütterpumpe (9) die Induktionsstoffkonzentra- tion im Bioreaktor (1) regelt.
19. Vorrichtung nach Anspruch 18, dadurch gekennzeichnet, dass das Analysesystem (24) als ein Fließdiffusionsanalyse- System (FDA) ausgebildet ist.
20. Vorrichtung nach einem der Ansprüche 17 bis 18, dadurch gekennzeichnet, dass zur Messung einer Zellkonzentration im Bioreaktor (1) die Kontrolleinheit (6) ein zweites Analysesystem (27) aufweist, das über einen im Bioreaktor (1) angeordneten zweiten Sensor (28) die Zellkonzentration misst und durch Steuerung einer dem zweiten Erntebehälter (18) vorgeschalteten Harvestpumpe (20) die Zellkonzentration im Biore- aktor (1) regelt.
21. Vorrichtung nach einem der Ansprüche 17 bis 20, dadurch gekennzeichnet, dass zur Regelung der Mediumzugabe in den Bioreaktor (1) ein dritter Regler (44) über eine Wägevorrich- tung (22) des Bioreaktors (1) mit einer Zufütterpumpe (8) verbunden ist.
