Verfahren zur Herstellung von chiralen substituierten Diolen und deren Derivaten der Formel (I), in der R H oder einen gegebenenfalls substituierten C5-C20-Aryl-, C5-C20-Heterocyclus- oder Cl-C20-Alkylrest und Rl einen gegebenenfalls substituierten C5-C20-Aryl-, C5-C20-Heterocyclus- oder Cl-C20-Alkylrest bedeutet, R2 für H, einen gegebenenfalls substituierten Cl-C20-Alkyl-, C3-C7-Heterocyclus-, Silyl-, C5-C20-Aryl-, C5-C20-Arylsulfonyl- oder Cl-C20-Alkylsulfonylrest steht, R3 H oder eine O-Schutzgruppe sein kann und X Sauerstoff, Schwefel, Stickstoff oder Phosphor bedeutet, bei welchem a) eine chirale Hydroxycarbonsäure oder Hydroxycarbonsäureester der Formel (II), in der R4 H oder eine Cl-C6-Alkyl bedeutet, mit einer O-Schutzgruppenverbindung zu der Verbindung der Formel (III), in der R3 die O-Schutzgruppe bedeutet umgesetzt wird, sodann b) mit einem Alkalibor- oder -aluminiumhydrid zu der Verbindung der Formel (IV) reduziert wird, bei der sodann c) gegebenenfalls entweder das Sauerstoffatom aktiviert und anschliessend durch einen Schwefelatom-, Stickstoffatom- oder Phosphoratom-hältigen Rest ausgetauscht wird, oder d) die gegebenenfalls durch Umsetzung mit einem Alkylierungs- oder Arylierungsreagens in eine Verbindung der Formel (V), in der R'2 einen gegebenenfalls substituierten C1-C20-Alkyl-, C3-C7-Heterocyclus-, Silyl-, C5-C20-Aryl-, C5-C20-Arylsulfonyl- oder C1-C20-Alkylsulfonylrest und R3 die O-Schutzgruppe bedeuten, überführt wird und e) im Anschluss an Schritt b), c) oder d) gegebenenfalls die O-Schutzgruppe entfernt wird.
Description
Verfahren zur Herstellung chiraler substituierter Diole und Diolanaloger Derivate
Die Erfindung betrifft ein Verfahren zur Herstellung von chiralen substituierten Diolen und analogen Derivaten in hoher optischer Reinheit und hoher Ausbeute. Optisch aktive Diole und deren Derivate finden als strukturbestimmende Bausteine Anwendung in verschiedenen pharmazeutischen Wirkstoffen wie z. B. CCR5 Antagonisten zur Behandlung von HIV, Arthritis, Dermatitis, Organabstoßung, Asthma und multiple Sklerose.
Aus WO 00/66558 ist beispielsweise die Herstellung von racemischen 2-Methoxy-1- (4'-trifluormethylphenyl)ethanol aus p-Trifiuormethylstyrol über das entsprechende Epoxid bekannt. Der Nachteil bei diesem Verfahren liegt einerseits darin, dass das Produkt in racemischer Form und andererseits in geringer Ausbeute von lediglich 55% erhalten wird. Ein weiterer Nachteil ist die schlechte Verfügbarkeit des Styrolde- rivates.
Aufgabe der Erfindung war es, ein geeignetes Verfahren zur Herstellung von chiralen substituierten Diole und deren Derivaten zu finden, das die gewünschten Endverbindungen auf einfache Weise in hoher chemischer und optischer Ausbeute liefert.
Unerwarteterweise konnte diese Aufgabe durch ein einfaches Eintopfverfahren ausgehend von kostengünstigen Rohstoffen gelöst werden.
Gegenstand der Erfindung ist demnach ein Verfahren zur Herstellung von chiralen substituierten Diolen und deren Derivaten der Formel (I)
in der R H oder einen gegebenenfalls ein- oder mehrfach durch Halogen, Ci-Cβ- Alkyl-, Halo-Cι-C6-Alkyl, CrC6-Alkoxy, Halo-C C6-Alkoxy, Aryloxy, Acylamino, gegebenenfalls substituiertes Phenyl oder Benzyl substituierten C5-C2o-Aryl-, C5-C2o- Heterocyclus- oder CrC2o-Alkylrest bedeutet,
R1 einen gegebenenfalls ein- oder mehrfach durch Halogen, C C6-Alkyl-, Halo-d- C6-Alkyl, C C6-Alkoxy, Halo-CrC6-Alkoxy, Aryloxy, Acylamino, gegebenenfalls substituiertes Phenyl oder Benzyl substituierten C5-C2o-Aryl-, C5-C2o-Heterocyclus- oder CrC2o-Alkylrest bedeutet,
R2 für H, CrC2o-Alkyl-, C5-C2o-Aryl oder einen gegebenenfalls ein- oder mehrfach durch Halogen, CrCe-Alkyl, Halo-C C6-Alkyl, C C6-Alkoxy, Halo-d-Ce-Alkoxy, Aryloxy, Acylamino, Phenyl oder Benzyl substituierten Cι-C20-Alkyl-, C3-C7- Heterocyclus-, Silyl-, C5-C20-Aryl-, C5-C2o-Arylsulfonyl- oder CrC20-Alkylsulfonylrest steht,
R3 H oder eine O-Schutzgruppe sein kann und X Sauerstoff, Schwefel, Stickstoff oder Phosphor bedeutet, das dadurch gekennzeichnet ist, dass a) eine chirale Hydroxycarbonsäure oder ein chiraler Hydroxycarbonsäureester der Formel (II)
in der R1 und R wie oben definiert sind und R H oder d-Cβ-Alkyl bedeutet in einem organischen, aprotischen Lösungsmittel mit einer O-Schutzgruppen- verbindung bei einer Temperatur von -20°C bis +90°C zu der Verbindung der Formel (III) o'R3

b) die Verbindung der Formel (III) in einem organischen Lösungsmittel aus der Gruppe der Ether, Alkohole oder der Kohlenwasserstoffe mit einem Reduktionsmittel aus der Gruppe der Alkalibor- oder -aluminiumhydride bei einer Temperatur von - 20°C bis +100°C zu der Verbindung der Formel (IV)
in der R und R1 wie oben definiert sind und R3 die O-Schutzgruppe bedeutet, reduziert wird, bei der sodann c) gegebenenfalls entweder das Sauerstoffatom bei einer Temperatur von -20°C bis +100°C zuerst durch Umsetzung mit einer Sulfonsäureverbindung in Gegenwart einer Base aktiviert und anschließend durch einen Schwefelatom, Stickstoffatom oder Phosphoratom-hältigen Rest ausgetauscht wird, sodass eine Verbindung der Formel (I) erhalten wird, in der X S, N oder P bedeutet, R, R1 und R2 wie oben definiert sind und R3 die O-Schutzgruppe ist oder d) gegebenenfalls durch Umsetzung mit einem Alkylierungs- oder Arylierungsrea- gens aus der Gruppe der Alkyl- oder Arylhalogenide, -sulfate oder -sulfonate, Acylhalogenide, Silylhalogenide oder Aryldiazoniumsalze bei einer Temperatur von -20°C bis +100°C in eine Verbindung der Formel (V)
in der R und R1 wie oben definiert sind, R2 ' Cι-C2u-Alkyl-, C5-C20-Aryl oder einen gegebenenfalls ein- oder mehrfach durch Halogen, CrCβ-Alky!, Halo-Ci-Cβ-Alkyl, C C6-Alkoxy, Halo-Ci-Cβ-Alkoxy, Aryloxy, Phenyl oder Benzyl substituierten C C20-Alkyl-, C3-C7-Heterocyclus-, Acyl-, Silyl-, C -C20-Aryl-, C5-C20-Arylsulfonyl- oder CrC2o-Alkylsulfonylrest und R3 die O-Schutzgruppe bedeuten, überführt wird und
e) im Anschluss an Schritt b), c) oder d) gegebenenfalls die O-Schutzgruppe bei einer Temperatur von -20°C bis +70°C mit einem Alkohol in Gegenwart einer Säure, oder mittels H2/Pd oder basisch entfernt wird, sodass die gewünschten Endverbindungen der Formel (I), in der R3 H bedeutet, erhalten werden.
Gemäß dem erfindungsgemäßen Verfahren werden Verbindungen der Formel (I) hergestellt.
In der Formel (I) bedeuten
R H oder einen gegebenenfalls ein- oder mehrfach durch Halogen, Ci-Ce-Alkyl, Halo- C C6-Alkyl, d-C6-Alkoxy, Halo-Ci-Ce-Alkoxy, Aryloxy, Acyl-amino, gegebenenfalls substituiertes Phenyl oder Benzyl substituierten C5-C2o-Aryl-, Cs-do-Heterocyclus- oder d-C2o-Alkylrest und
R1 einen gegebenenfalls ein- oder mehrfach durch Halogen, CrC6-AlkyI, Halo-d- Cδ-Alkyl, CrCβ-Alkoxy, Halo-Ci-Ce-Alkoxy, Aryloxy, Acyl-amino, gegebenenfalls substituiertes Phenyl oder Benzyl substituierten C5-C20-Aryl-, Cs-do-Heterocyclus- oder Cι-C20-Alkylrest.
Unter C5-C20-Aryl sind aromatische Reste zu verstehen, wie etwa Phenyl, Biphenyl, Naphthyl, Indenyl, Fluorenyl u.s.w.. Bevorzugte Arylreste sind Phenyl oder Naphthyl. Unter C5-C20-Heterocyclus sind cyclische Reste zu verstehen, die mindestens ein S-, O- oder N-Atom im Ring enthalten. Dies sind beispielsweise Furyl, Pyridyl, Pyrimidyl, Thienyl, Isothiazolyl, Imidazolyl, Tetrazolyl, Pyrazinyl, Benzofuranyl, Benzothiophe- nyl, Chinolyl, Isochinolyl, Benzothienyl, Isobenzofuryl, Pyrazolyl, Indolyl, Isoindolyl, Benzoimidazolyl, Purinyl, Carbazolyl, Oxazolyl, Thiazolyl, 1 ,2,4-Thiadiazolyl, Isoxazo- lyl, Pyrrolyl, Chinazolinyl, Pyridazinyl, Phthalazinyl, Morpholinyl, Triazolyl, Imidazoli- dinyl, Chinoxalinyl, Piperazinyl, Piperidinyl, Tetrahydropyranyl u.s.w. Funktionelle O- oder N-Gruppen können dabei nötigenfalls geschützt werden. Bevorzugt Heterocyclusreste sind dabei Cs-Ce-Heterocyclen mit einem Heteroatom aus der Gruppe O, S oder N.
Unter Cι-C2o-Alkyl sind dabei gesättigte oder ein- oder mehrfach ungesättigte, lineare, verzweigte oder cyclische, polycyclische, überbrückte, primäre, sekundäre oder tertiäre Hydrocarbonreste zu verstehen, wie etwa Methyl, Ethyl, Propyl, i-Propyl, Pro- penyl, Butyl, i-Butyl, t-Butyl, Butenyl, Butinyl, Pentyl, Cyclopentyl, i-Pentyl, neo- Pentyl, Pentenyl, Hexyl, i-Hexyl, Cyclohexyl, Cyclohexylmethyl, 3-Methylpentyl, 2,2- Dimethylbutyl, 2,3-Dimethylbutyl, Octyl, Cyclooctyl, Decyl, Cyclodecyl, Dodecyl, Cyc- lododecyl, 4-lsopropyl-1-methyl-cyclohexyl, Norbomyl, Cholestan u.s.w.. Bevorzugt sind dabei lineare, verzweigte oder cyclische, gesättigte d-Cι2-Alkylreste und besonders bevorzugt Ci-Ce-Alkylreste.
Bevorzugt bedeutet in der Formel (I) der Rest R H oder CrC8-Alkyl, einen Phenyl- oder Naphthylrest und
R1 Ci-Ce-Alkyl, einen Phenyl- oder Naphthylrest oder einen C5-C6-Heterocyclus mit einem Heteroatom aus der Gruppe O, S oder N.
Besonders bevorzugt bedeutet R H und R1 einen Phenyl- oder Naphthylrest.
Die Reste können dabei ein- oder mehrfach durch Halogen, C C6-Alkyl, Halo-d-C6- Alkyl, CrC6-Alkoxy, Halo-CrC6-Alkoxy, Aryloxy, Acylamino, gegebenenfalls substituiertes Phenyl oder Benzyl substituiert sein.
Halo-Ci-Cθ-Alkyl- und Halo-d-C6-Alkoxyreste sind dabei ein- oder mehrfach durch Halogen, wie etwa Fluor, Chlor Brom oder lod, bevorzugt durch Fluor, substituierte Alkyl- oder Alkoxyreste.
Bevorzugte Substituenten sind dabei Halogen, Cι-C -Alkyl, Halo-CrC4-Alkyl oder gegebenenfalls substituiertes Phenyl. Besonders bevorzugt sind Fluor, Chlor, Tri- fluormethyl oder Phenyl.
R2 bedeutet in der Formel (I) H, d-C2u-Alkyl-, C5-C20-Aryl oder einen gegebenenfalls ein- oder mehrfach durch Halogen, Ci-Ce-Alkyl, Halo-Cι-C6-Alkyl, CrC6-Alkoxy, Ha- lo-Ci-Cβ-Alkoxy, Aryloxy, Phenyl oder Benzyl substituierten Cι-C20-Alkyl-, C3-C7- Heterocyclus-, Silyl-, C5-C2o-Aryl-, C -C2o-Arylsulfonyl- oder d-C2o-Alkylsulfonylrest
Bevorzugt bedeutet R2 H, einen Cι-C -Alkyl-, Phenyl-, Naphthyl-, C5-C6- Heterocyclus-, Silyl-, Cβ-Cio-Arylsulfonyl- oder C C6-Alkylsulfonylrest, wobei die Reste gegebenenfalls ein- oder mehrfach durch Halogen, bevorzugt durch Fluor oder Chlor, C C4-Alkyl, Halo-C C4-Alkyl, C C4-Alkoxy, Halo-C C4-Alkoxy oder Phenyl substituiert sein können.
Besonders bevorzugt bedeutet R2 H, d-C2-Alkyl, Phenyl, Phenylsulfonyl, oder d- C4-Alkylsulfonyl, wobei die Reste ein- oder zweifach durch Fluor, Chlor, C C2-Alkyl, Trifluormethyl oder Phenyl substituiert sein können.
R3 steht in der Formel (I) für H oder eine O-Schutzgruppe. Als O-Schutzgruppe eignen sich alle gängigen sauer, basisch oder hydrogenolytisch abspaltbaren O- Schutzgruppen, wie etwa solche vom Ketal- oder Ethertyp oder Silylschutzgruppen. Bevorzugte O-Schutzgruppen sind dabei Tetrahydropyranyl, Dimethylmethoxy- methyl, Dimethylethoxymethyl, Methylethoxymethyl, Benzylether, Allylether, Tri- methylsilyl, tert.Butyl-dimethylsilyl u.s.w..
X bedeutet in der Formel (I) ein Heteroatom aus der Gruppe O, S, N oder P. Bevorzugt steht X für O, N oder S, besonders bevorzugt für O.
Das N- und das S-Atom können dabei gegebenenfalls durch Ci-Ce-Alkyl substituiert sein.
Beispiele für erfindungsgemäß herstellbare Verbindungen der Formel (I) sind (S)- oder (R)-2-Methoxy-1-(4'-trifluormethylphenyl)ethanol, (S)- oder (R)-2-Methoxy-1- phenylethanol, (S)- oder (R)-2-Methoxy-1-(2-Tetrahydropyranyloxy)-1-(4'-trifluor- methylphenyl)ethan, (S)- oder (R)-2-(2-Tetra-hydropyranyloxy)-2-(4'-trifluormethyl- phenyl)ethanol, (S)- oder (R)-2-Methylthio-1-phenylethanol, (S)- oder (R)-2- Methylthio-1-(4'-trifluormethylphenyl)ethanol, S)- oder (R)-2-(N,N-dimethylamino)-1- phenylethanol u.s.w..
Als Ausgangsverbindung zur Herstellung der gewünschten Endverbindungen der Formel (I) wird erfindungsgemäß eine chirale Hydroxycarbonsäure oder ein chiraler Hydroxycarbonsäureester der Formel (II) in der R und R1 wie in der Formel (I) definiert sind und R4 H oder CrC6-Alkyl bedeutet.
Die chiralen Hydroxycarbonsäuren und -ester können dabei in ihrer (S)- oder (R)- Form eingesetzt werden.
Die Verbindungen der Formel (II) sind käuflich erwerbbar. Sie können jedoch auch durch Umsetzung von einem Aldehyd oder Keton der Formel (VI)
in der R und R1 wie oben definiert sind, in einem organischen Lösungsmittel aus der Gruppe der Ether, Ketone oder der aromatischen Kohlenwasserstoffe bei einer Temperatur von -10 bis +50°C in Gegenwart eines Cyanidgruppendonors und einer (S)- oder (R)-Hydroxynitrillyase (HNL) zu der Verbindung der Formel (VII)
in der R und R1 wie oben definiert sind, und a) anschließender enzym-katalysierter Hydrolyse oder durch Hydrolyse mit HCI bei Temperaturen von 10-100°C zu den entsprechenden chiralen Hydroxycarbonsäuren oder b) mit einem CrCβ-Alkohol in Gegenwart einer Säure, wie etwa HCI oder H2SO u.s.w. bei 40°C bis zur Rückflusstemperatur, gegebenenfalls unter Druck, zu dem entsprechenden chiralen Hydroxycarbonsäureester ohne Verlust an optischer Reinheit hergestellt werden.
Die Hydroxycarbonsäure bzw. der Ester der Formel (II) muss dabei nicht aus dem Reaktionsgemisch isoliert werden, sondern kann ohne Isolierung und Aufreinigung als Lösung in das erfindungsgemäße Verfahren eingesetzt werden.
Ein weiterer Gegenstand der vorliegenden Erfindung ist demnach ein Gesamtverfahren zur Herstellung von Verbindungen der Formel (I), das dadurch gekennzeichnet ist, dass ein Aldehyd oder Keton der Formel (VI)
in der R und R1 wie oben definiert sind, in einem organischen Lösungsmittel aus der Gruppe der Ether, Ketone oder der aromatischen Kohlenwasserstoffe bei einer Temperatur von -10 bis +50°C in Gegenwart eines Cyanidgruppendonors und einer (S)- oder (R)-Hydroxynitrillyase zu der Verbindung der (VII)
in der R und R1 wie oben definiert sind, umgesetzt wird, die anschließend durch enzym-katalysierte Hydrolyse oder durch Hydrolyse mit HCI bei Temperaturen von 10-100°C in die entsprechende chirale Hydroxycarbonsäure der Formel (II) oder durch Umsetzung mit einem CrCe-Alkohol in Gegenwart einer Säure bei 40°C bis zur Rückflusstemperatur, gegebenenfalls unter Druck, in den entsprechenden chiralen Hydroxycarbonsäureester der Formel (II) überführt wird, worauf die Schritte a) bis e) zur Herstellung der gewünschten Verbindung der Formel (I) durchgeführt werden.
Die Aldehyde und Ketone der Formel (VI) werden dabei durch enzymatische Addition einer Cyanidgruppe in das entsprechende (S)- oder (R)-Cyanhydrin überführt. Die enzymatische Addition kann dabei analog dem Stand der Technik, beispielsweise analog EP 0 951 561 , EP 0 927 766, EP 0 632 130, EP 0547 655, EP 0 326 063 u.s.w., erfolgen.
Das entsprechende (R)- und (S)- Cyanhydrin der Formel (VII) wird sodann, beispielsweise nach Extraktion oder nach Abfiltrieren des Enzyms und Abdestillieren des Lösungsmittels, ohne weitere Reinigung analog dem Stand der Technik, beispielsweise wie in Angew. Chem. 1994, 106, S 1615 oder in Tetrahedron Letters, Vol. 31 , No. 9, pp1249-1252, 1990 beschrieben, mit Salzsäure hydrolysiert. Die Hydrolyse kann jedoch auch enzymatisch, wie etwa in J. org. Chem. 2002, 67, pp 6542- 6545 oder in J. Am. Chem. Soc. 2002, 124, pp 9024-9025 beschrieben, erfolgen.
Die so erhaltenen rohen (R)- und (S -α-Hydroxycarbonsäuren oder -carbonsäure- ester der Formel (II), die in etwa die gleiche optische Reinheit wie die korrespondierenden (R)- und (S)- Cyanhydrine aufweisen, können erfindungsgemäß ohne Isolierung und Aufreinigung in die nächste Stufe, Schritt a) als Lösung eingesetzt werden.
In Schritt a) wird die Verbindung der Formel (II) mit einer geeigneten O- Schutzgruppenverbindung zu der Verbindung der Formel (III) umgesetzt. Als O-Schutzgruppe eignen sich alle gängigen, sauer, basisch oder hydrogenolytisch abspaltbaren O-Schutzgruppen. Bevorzugt sind solche vom Ketal- oder Ethertyp oder Silylschutzgruppen, wie etwa Tetrahydropyranyl, Dimethylmethoxymethyl, Di- methylethoxymethyl, Methylethoxymethyl, Benzylether, Allylether, Trimethylsilyl, teil. Butyl-dimethylsilyl u.s.w..
Als bevorzugte Schutzgruppenverbindungen werden demnach 3,4-Dihydro-2H- pyran, Ethylvinyether, Isopropenylethylether, Isopropenylmethylether, Trimethylsilyl- chlorid , teil. Butyl-dimethylsilylchlorid u.s.w. eingesetzt.
Die Schutzgruppenverbindung wir dabei in einer Menge von 1.0 bis 2 Äquivalenten, bevorzugt von 1.05 bis 1.5 Äquivalenten eingesetzt.
Die Umsetzung erfolgt dabei in einem organischen, aprotischen Lösungsmittel. Bevorzugt werden solche aus der Gruppe der Ether, wie etwa Methyl-tert.butyl-Ether (MTBE), Tetrahydrofuran (THF), u.s.w. oder aus der Gruppe der Kohlenwasserstoffe, wie etwa Toluol u.s.w..
Die Reaktionstemperatur liegt bei -20°C bis +90°C, bevorzugt bei -10 bis +30°C und besonders bevorzugt bei 0 bis 20°C.
Um die Umsetzung zu beschleunigen, wird dem Reaktionsgemisch ein geeigneter Katalysator zugegeben. Geeignete Katalysatoren sind dabei wasserfreie Mineralsäuren, bevorzugt Schwefelsäure, oder Sulfonsäuren, wie etwa p-Toluolsulfonsäure (pTSA). Der Katalysator wird in einer Menge von etwa 0.001 bis 0.05 Äquivalenten, bevorzugt von 0.01 bis 0.03 Äquivalenten zugesetzt.
Im Falle von Silylschutzgruppen wird hingegen eine geeignete Base, wie etwa Triethylamin, in einer Menge von mindestens 1 Moläquivalent bis zu 3 Moläquivalenten zugesetzt.
Die Verbindung der Formel (III) muss nicht aus dem Reaktionsgemisch entfernt werden, sondern das Reaktionsgemisch wird direkt in der nächsten Stufe, Schritt b), eingesetzt.
In Schritt b) wird die Verbindung der Formel (III) in einem organischen Lösungsmittel aus der Gruppe der Ether, Alkohole oder der Kohlenwasserstoffe mit einem Reduktionsmittel aus der Gruppe Alkalibor- oder -aluminiumhydride bei einer Temperatur von -20°C bis +100°C zu der Verbindung der Formel (IV) reduziert. Als organische Lösungsmittel eignen sich hier aliphatische oder cyclische Ether, wie etwa Tetrahydrofuran (THF), MTBE, u.s.w. oder Kohlenwasserstoffe, wie etwa Tolu- ol, Heptan, u.s.w. und im Falle von Alkaliborhydriden Alkohole, wie etwa Isopropanol, Polyethylenglkole, u.s.w..
Geeignete Reduktionsmittel sind Alkalibor- oder -aluminiumhydride, wie etwa Na-, K- oder Li-Borhydride oder Na-, K- oder Li-Aluminiumhydride. Bevorzugt werden Aluminiumhydride, besonders bevorzugt Li-Aluminiumhydrid eingesetzt. Das Reduktionsmittel wird dabei in einer Menge von 0.5 bis 4 Äquivalenten, bevorzugt von 0.6 bis 2.5 Äquivalenten, bevorzugt als Suspension in dem zur Umsetzung eingesetzten Lösungsmittel, zugesetzt.
Die Reaktionstemperatur liegt für Schritt b) bei -20°C bis +100°C, bevorzugt bei -20°C bis +25°C und besonders bevorzugt bei -10°C bis +10°C.
Gegebenenfalls kann Schritt b) auch unter Druck durchgeführt werden. Nach erfolgter Umsetzung - der Reaktionsfortschritt kann beispielsweise über Dünnschichtchromatographie verfolgt werden - wird bevorzugt durch Zugabe von Wasser und verdünnter Natronlauge oder durch saure Extraktion mit Zitronensäure ein gut filtrierbares Salz des Reduktionsmittels hergestellt. Anschließend wird die organische Phase abgetrennt, gegebenenfalls mit Wasser gewaschen und gegebenenfalls, beispielsweise azeotrop, getrocknet.
Die Verbindung der Formel (IV), in der R und R1 wie oben definiert sind und R3 die O-Schutzgruppe ist, kann gegebenenfalls bereits dem gewünschten Endprodukt der Formel (I), in der R und R1 wie oben definiert sind, R2 gleich H ist und R3 die O- Schutzgruppe ist, entsprechen.
Im Anschluss an Schritt b) kann gegebenenfalls noch die O-Schutzgruppe entfernt werden (Schritt e)), sodass die gewünschten Endverbindungen der Formel (I), in der R3 H bedeutet, R und R1 wie oben definiert sind und R2 gleich H ist, erhalten werden.
Die Abspaltung der Schutzgruppe erfolgt in Abhängigkeit von der Art der Schutzgruppe gemäß dem Stand der Technik, beispielsweise mit einem Alkohol, wie etwa Methanol oder Ethanol in Gegenwart einer Säure, mit H2/Pd oder basisch mit einer geeigneten Base in einem Alkohol.
Als Säuren eignen sich bevorzugt Mineralsäuren, wie etwa Schwefelsäure, Sulfonsäuren und für die Abspaltung der Silylgruppen Trifluoressigsäure oder ein Fluorid- donor, wie etwa Tetrabutylammoniumfluorid u.s.w.
Der Alkohol wird dabei in einer Menge von 2 Bis 10 Äquivalenten, bevorzugt von 2.5 bis 4 Äquivalenten und die Säure in einer Menge von 0.005 bis 0.5, bevorzugt von 0.001 bis 0.1 eingesetzt.
Die Reaktionstemperatur liegt wiederum bei -20°C bis +70°C, bevorzugt bei 0 bis 30°C.
Die Abspaltung mit H2/Pd erfolgt in einem Lösungsmittel aus der Gruppe der Alkohole, Ether, Carbonsäuren oder Kohlenwasserstoffe bei einer Temperatur von 20 bis 100°C, wobei H2 in bis zu 5 bar Überdruck verwendet wird.
Die basische Abspaltung erfolgt mittels einer geeigneten Base, wie etwa einem Alka- licarbonat, beispielsweise mit K2CO3, in einem Alkohol, wie etwa Methanol. Die Reaktionstemperatur liegt dabei bei 20 bis 50°C.
Die so erhaltene Verbindung der Formel (IV) kann aber auch entweder gemäß Schritt c) oder gemäß Schritt d) weiterverarbeitet werden.
Gemäß Schritt c) wird bei der Verbindung der Formel (IV) gegebenenfalls das Sauerstoffatom durch zuerst durch Umsetzung mit einer Sulfonsäureverbindung in Gegenwart einer Base aktiviert und anschließend durch einen Schwefelatom-, Stickstoffatom- oder Phosphoratom-hältigen Rest ausgetauscht, sodass eine Verbindung der Formel (I) in der X S, N oder P bedeutet, R, R1 und R2 wie oben definiert sind und R3 die O-Schutzgruppe ist, erhalten wird.
Als Sulfonsäureverbindung eignen sich beispielsweise Tosylchlorid oder -anhydrid, Methansulfonsäurechlorid oder -anhydrid, Trifluormethansulfonsäurechlorid oder -anhydrid u.s.w. Die Sulfonsäureverbindung wird dabei in einer Menge von 1.0 bis 2.0 Äquivalenten zugesetzt.
Als Säurefänger wird zudem eine Base in der Menge von 1.0 bis 2.0 Äquivalenten, bevorzugt von 1.0 bis 1.3 Äquivalenten zugegeben. Als Base kommen org. Stickstoffbasen, wie etwa Triethylamin, Diisopropylethylamin, 1 ,4-Diazabicyclo-[2,2,2]- octan u.s.w. in Frage.
Anschließend erfolgt die Umsetzung mit 1 bis 2 Äquivalenten einer entsprechenden S-, N- oder P-haltigen Verbindung aus der Gruppe der sekundären Amine, der Phosphinderivate, Thiole, wie etwa Alkalialkylthiolate, Thiophenolate, u.s.w., oder der Sulfide oder einem entsprechenden Salz.
Die Verbindungen enthalten dabei das Strukturelement X-R2, wobei X N, S oder P bedeutet und R2 wie oben definiert ist. Das N- und das S-Atom können dabei gegebenenfalls durch Ci-Cβ-Alkyl substituiert sein.
Die Reaktionstemperatur liegt wiederum bei -20 bis +100°C, bevorzugt bei 0 bis 20°C.
Im Anschluss an Schritt c) kann gegebenenfalls noch wie oben beschrieben die O- Schutzgruppe entfernt werden, sodass die gewünschten Endverbindungen der Formel (I), in der R3 H bedeutet, X für S, N oder P steht und R, R1 und R2 wie oben definiert sind, erhalten werden.
Gemäß Schritt d) wird die Verbindung der Formel (IV) gegebenenfalls durch Umsetzung mit einem Alkylierungs- oder Arylierungsreagens aus der Gruppe der Alkyl- o- der Arylhalogenide, -sulfate oder sulfonate, Acylhalogenide, Silylhalogenide oder A- ryldiazoniumsalze in eine Verbindung der Formel (V)
in der R und R1 wie oben definiert sind, R2 ' Cι-C2ü-Alkyl-, Cs-do-Aryl oder einen gegebenenfalls ein- oder mehrfach durch Halogen, Cι-C6-Alkyl, Halo-d-C6-Alkyl, d- Cβ-Alkoxy, Halo-Cι-C6-Alkoxy, Aryloxy, Phenyl oder Benzyl substituierten CrC20- Alkyl-, C3-C7-Heterocyclus-, Silyl-, C5-C20-Aryl-, C5-C2o-Arylsulfonyl- oder C C20- Alkylsulfonylrest und R3 die O-Schutzgruppe bedeuten, überführt. Als Alkylierungs- oder Arylierungsreagens werden Verbindungen aus der Gruppe der Alkyl- oder Arylhalogenide, -sulfate oder sulfonate, Acylhalogenide, Silylhalogenide oder Aryldiazoniumsalze eingesetzt.
Geeignete Verbindungen sind beispielsweise Dialkylsulfate, wie etwa Dimethylsulfat, Benzoldiazoniumsalze, wie etwa Benzoldiazoniumchlorid, Trimethylsilylchlorid, tert.Butyl-dimethylsilylchlorid u.s.w.
Das Alkylierungs- bzw. Arylierungsreagens wird dabei in einer Menge von 1.0 bis 4.0 Äuqivalenten, bevorzugt von 1.4 bis 2.5 Äqivalenten zugesetzt.
Die Umsetzung erfolgt in Gegenwart von 1.0 bis 4.0 Aquvivalenten, bevorzugt von 1.4 bis 2.5 Äqivalenten einer nicht nukleophilen Base, wie etwa Natriumhydrid u.s.w. oder wässrige NaOH unter Phasentransferkatalyse.
Die Reaktionstemperatur liegt bei -20°C bis +100°C, bevorzugt bei 0°C bis 80°C Als Lösungsmittel eignen sich wiederum aliphatische oder cyclische Ether, wie etwa Tetrahydrofuran (THF), MTBE, u.s.w. oder Kohlenwasserstoffe, wie etwa Toluol, Heptan, u.s.w..
Die Aufarbeitung des Reaktionsgemisches erfolgt durch Zugabe von Wasser und
Phasentrennung.
Die Verbindung der Formel (V) muss nicht isoliert werden, sondern die organische
Phase kann direkt gegebenenfalls nach Waschen mit Wasser und azeotropen
Trocknen in die nächste Stufe eingesetzt werden.
Im Anschluss an Schritt d) kann gegebenenfalls noch wie oben beschrieben die O- Schutzgruppe entfernt werden, sodass die gewünschten Endverbindungen der Formel (I), in der R3 H bedeutet, R, R1 wie oben definiert ist und R2 gleich R2 'ist, erhalten werden.
Die O-Schutzgruppe kann aber auch bereits vor Schritt d) entfernt werden, sodass die gebildete OH-Gruppe gegebenenfalls gleichzeitig mit dem Alkylierungs- oder Arylierungsreagens reagiert, wodurch Verbindungen der Formel (VIII)
erhalten werden, in der R und R1 wie oben definiert sind und R2 'und R3 ' gleich sind d-C20-Alkyl-, C5-C20-Aryl oder einen gegebenenfalls ein- oder mehrfach durch Halogen, Ci-Ce-Alkyl, Halo-Cι-Ce-Alkyl, C C6-Alkoxy, Halo-Ci-Ce-Alkoxy, Aryloxy, Phenyl oder Benzyl substituierten Cι-C20-Alkyl-, C3-C7-Heterocyclus-, Silyl-, C5-C2o-Aryl-, C5- C2o-Arylsulfonyl- oder CrC20-Alkylsulfonylrest bedeuten.
Die Umsetzung erfolgt wie oben beschrieben, wobei jedoch die doppelte Menge an Base und Alkylierungs- oder Arylierungsreagens zugegeben werden.
Die Isolierung und Aufarbeitung des jeweiligen gewünschten Endproduktes kann durch in Abhängigkeit vom Aggregatzustand durch übliche Methoden, wie etwa Destillation, Extraktion, Kristallisation u.s.w. erfolgen.
Durch das erfindungsgemäße Verfahren werden die gewünschten Endprodukte der Formel (I) im Vergleich zum Stand der Technik in hohen chemischen Ausbeuten von bis zu über 65%, berechnet vom Aldehyd der Formel (VI) bzw. von bis zu über 80%, berechnet von der Hydroxycarbonsäure bzw. -ester der Formel (II) und hohen optischen Reinheiten bis zu 99.9% erhalten. Vorteilhaft dabei ist, dass bei dem erfindungsgemäßen Verfahren kein bis höchstens ein Zwischenprodukt isoliert werden muss und dass es sich um ein Eintopfverfahren handelt, das zudem von kostengünstigen Edukten ausgeht. Ein weiterer Vorteil ist, dass sich die optische Reinheit der Hydroxycarbonsäure oder des Carbonsäureesters der Formel (II) während der Umsetzung bis zum gewünschten Endprodukt nicht verringert.
Beispiel 1 : (S)-2-Hydroxy-2-(4'-trifluormethylphenyl)acetonitril
38.3g 4'-Trifluormethylbenzaldehyd wurden in 55ml Diisopropylether gelöst und auf 5°C gekühlt. S-Hydroxynitrillyase (2000 lU/g Aldehyd) wurde mit 54ml Citratpuffer (pH 5.5, 0.01 M) vereint und zum Aldehyd gegeben. Unter starkem Rühren wurden 17,2ml HCN (2 Äquivalente) während einer Stunde chargiert. Nach 3h Reaktionszeit bei 5°C wurden 120ml Diisopropylether zugegeben, 5 Minuten gerührt und Phasen getrennt. Die organische Phase wurde abgetrennt, die wässrige Phase wurde noch zweimal mit je 80ml Diisopropylether extrahiert. Die organischen Phasen wurden über Natriumsulfat getrocknet und a) am Rotavapor zur Trockene eingedampft oder b) in Lösung weiter umgesetzt.
Umsatz 99%; Ausbeute a) 95% (S)-2-Hydroxy-2-(4'-trifluormethylphenyl)acetonitril Optische Reinheit: 96% ee
Beispiel 2: (S)-2-Hydroxy-2-(4'-trifluormethylphenyl)essigsäure
Zu einer Lösung von 5g (S)-2-Hydroxy-2-(4'-trifluormethylphenyl)acetonitril in 30ml Diispropylether wurde 1 ml HCI conc gegeben, auf 70°C erwärmt und 3h bei dieser Temperatur gehalten. Anschließend wurde mit 100ml Wasser versetzt, Phasen getrennt und die organische Phase mit Wasser und gesättigter NaCI Lösung geschüttelt. Umsatz: 95%. Das Lösungsmittel wurde auf MTBE getauscht, das Produkt wurde nicht isoliert, sondern in Lösung weiter umgesetzt.
Beispiel 3: (S)-2-(2-Tetrahydropyranyloxy)-2-(4'-trifluormethylphenyl)essigsäure
Zu einer Lösung von (S)-2-Hydroxy-2-(4'-trifluormethylphenyl)essigsäure in MTBE wurden 0.02 Äquivalente pTSA gegeben und auf 15°C gekühlt. 1.1 Äquivalente 3,4- Dihydro-2H-pyran wurden langsam zugegeben und die Temperatur dabei auf 15°C gehalten. Nach vollendeter Zugabe wird noch 3h bei Raumtemperatur gerührt. Reak-
tionskontrolle erfolgte über Dünnschichtchromatographie. Umsatz: 95%. Die Reaktionsmischung wurde direkt weiter umgesetzt.
Beispiel 4: (S)-2-(2-Tetrahydropyranyloxy)-2-(4,-trifIuormethylphenyl)ethanol
Eine Lösung von (S)-2-(2-Tetrahydropyranyloxy)-2-(4'-trifluormethylphenyl)essig- säure in MTBE wurde zwischen -10 und 0° zu einer Suspension von 1.2 Äquivalenten Lithiumaluminiumhydrid in MTBE getropft. Der Reaktionsfortschritt wurde über Dünnschichtchromatographie verfolgt. Nach erfolgter Umsetzung wurde durch se- quenzielle Zugabe von Wasser, 15%iger NaOH und wiederum Wasser gequencht und ein gut filtrierbares Salz erzeugt. Die organische Phase wurde abgetrennt, mit Wasser gewaschen und gegebenenfalls azeotrop getrocknet. Umsatz: 95%.
Beispiel 5: (S)-2-Methoxy-1 -(2-Tetrahydropyranyloxy)-1 -(4'-trifluormethylphenyl)- ethan
a)
(S)-2-(2-Tetrahydropyranyloxy)-2-(4'-trifluormethylphenyl)ethanol gelöst in MTBE wurde zusammen mit 50%iger NaOH, 2 Äquivalenten an Dimethylsulfat und 3% Adogen® 464 kräftig gerührt. Der Umsatz wurde über Dünnschichtchromatographie verfolgt. Nach erfolgter Reaktion wurde noch eine Stunde auf 70°C erwärmt um restliches Dimethylsulfat zu vernichten. Anschließend wurde Wasser zugegeben und die Phasen getrennt. Die organische Phase wurde mit Wasser gewaschen und azeotrop getrocknet. Umsatz 95%. Das Produkt wurde nicht isoliert, sondern gleich weiter umgesetzt.
b)
Eine azeotrop getrocknete Lösung von (S)-2-(2-Tetrahydropyranyloxy)-2-(4'-trifluor- methylphenyl)ethanol in MTBE wurde auf 10°C gekühlt und mit 2 Äquivalenten Natriumhydrid versetzt. Im Anschluss daran wurden 2 Äquivalente Dimethylsulfat langsam
zugegeben. Nach erfolgter Zugabe wurde noch eine Stunde bei Raumtemperatur gerührt. Der Reaktionsfortschritt wurde über Dünnschichtchromatographie verfolgt. Nach vollständiger Umsetzung wurde Wasser zugegeben und 1h lang auf 80°C erhitzt um restliches Dimethylsulfat zu zerstören. Die Phasen wurden getrennt und die organische Phase mit Wasser gewaschen und azeotrop getrocknet und aufkonzentriert. Umsatz: 98%. Das Produkt wurde nicht isoliert, sondern direkt weiter umgesetzt.
c)
Eine azeotrop getrocknete Lösung von (S)-2-(2-Tetrahydropyranyloxy)-2-(4'-trifluor- methylphenyl)ethanol und Triethylamin in MTBE wurde auf 10°C gekühlt und mit 1.2 Äquivalenten Toluolsulfonsäurechlorid versetzt. Anschließend wurden 1.5 Äquivalente Natriummethylat langsam zugegeben. Nach vollständigem Umsatz wurde Wasser verdünnt und die Phasen getrennt. Die organische Phase wurde mit Wasser gewaschen, azeotrop getrocknet und aufkonzentriert. Umsatz: 97%. Das Produkt wurde nicht isoliert, sondern direkt weiter umgesetzt.
Beispiel 6: (S)-2-Methylthio-1 -(2-Tetrahydropyranyloxy)-1 -(4'-trifluormethylphenyl)- ethan
Eine azeotrop getrocknete Lösung von (S)-2-(2-Tetrahydropyranyloxy)-2-(4'-trifluor- methylphenyl)ethanol und Triethylamin in MTBE wurde auf 10°C gekühlt und mit 1.2 Äquivalenten Toluolsulfonsäurechlorid versetzt. Anschließend wurden 1.5 Äquivalente Natriummethanthiolat langsam zugegeben. Nachdem die Reaktion vervollständigt war wurde mit reichlich Wasser verdünnt und die Phasen getrennt. Die organische Phase wurde mehrmals mit Wasser gewaschen und über Natriumsulfat getrocknet. Umsatz: 99%. Das Produkt wurde nicht isoliert, sondern direkt weiter umgesetzt.
Beispiel 7: (S)-2-Methoxy-1-(4'-trifluormethylphenyl)ethanol
Eine Lösung von (S)-2-Methoxy-1-(2-Tetrahydropyranyloxy)-1-(4'-trifluormethyl- phenyl)ethan in MTBE wurde mit 3 Äquivalenten Methanol und 0.01 Äquivalent an pTSA versetzt und 3 Stunden bei Raumtemperatur gerührt. Die Reaktion wird über Dünnschichtchromatographie verfolgt. Methanol und MTBE werden gegen Toluol getauscht und die erhaltene Lösung mit Natriumhydrogencarbonat und Wasser gewaschen. Das Produkt wurde als Lösung in Toluol erhalten. Die Quantifizierung erfolgte über 1 H-NMR. Umsatz: 95%. Die Gesamtausbeute belief sich (berechnet vom Aldehyd) auf 65%. Optische Reinheit: 96% ee.

1. Verfahren zur Herstellung von chiralen substituierten Diolen und deren Derivaten der Formel (I)

in der R und R1 wie oben definiert sind und R H oder eine Cι-C6-Alkyl bedeutet, in einem organischen, aprotischen Lösungsmittel mit einer O- Schutzgruppenverbindung bei einer Temperatur von -20°C bis +90°C zu der Verbindung der Formel (III)
in der R, R1 und R4 wie oben definiert sind und R3 die O-Schutzgruppe bedeutet umgesetzt wird, worauf b) die Verbindung der Formel (III) in einem organischen Lösungsmittel aus der Gruppe der Ether, Alkohole oder der Kohlenwasserstoffe mit einem Reduktionsmittel aus der Gruppe der Alkalibor- oder -aluminiumhydride bei einer Temperatur von -20°C bis +100°C zu der Verbindung der Formel (IV)
in der R und R1 wie oben definiert sind und R3 die O-Schutzgruppe bedeutet, reduziert wird, bei der sodann c) gegebenenfalls entweder das Sauerstoffatom bei einer Temperatur von -20°C bis +100°C zuerst durch Umsetzung mit einer Sulfonsäureverbindung in Gegenwart einer Base aktiviert und anschließend durch einen Schwefelatom-, Stickstoffatom- oder Phosphoratom-hältigen Rest ausgetauscht wird, sodass eine Verbindung der Formel (I) erhalten wird, in der X S, N oder P bedeutet, R, R1 und R2 wie oben definiert sind und R3 die O-Schutzgruppe ist oder d) gegebenenfalls durch Umsetzung mit einem Alkylierungs- oder Arylierungsreagens aus der Gruppe der Alkyl- oder Arylhalogenide, -sulfate oder sulfonate, Acylhalogenide, Silylhalogenide oder Aryldiazoniumsalze bei einer Temperatur von -20°C bis +100X in eine Verbindung der Formel (V)

2. Verfahren zur Herstellung von Verbindungen der Formel (I) gemäß Anspruch 1 , dadurch gekennzeichnet, dass ein Aldehyd oder Keton der Formel (VI)
in der R und R1 wie in Anspruch 1 definiert sind, in einem organischen Lösungsmittel aus der Gruppe der Ether, Ketone oder der aromatischen Kohlenwasserstoffe bei einer Temperatur von -10 bis +50°C in Gegenwart eines Cyanidgruppendo- nors und einer (S)- oder (R)-Hydroxynitrillyase zu der Verbindung der Formel (VII)
in der R und R1 wie oben definiert sind, umgesetzt wird, die anschließend mittels enzym-katalysierter Hydrolyse oder durch Hydrolyse mit HCI bei Temperaturen
von 10-100°C in die entsprechende chirale Hydroxycarbonsäure der Formel (II) oder durch Umsetzung mit einem d-C6-Alkohol in Gegenwart einer Säure bei einer
Temperatur von 40°C bis zur Rückflusstemperatur, gegebenenfalls unter Druck, in den chiralen Hydroxycarbonsäureester der Formel (II) überführt wird, worauf die
Schritte a) bis e) gemäß Anspruch 1 zur Herstellung der gewünschten Verbindung der Formel (I) durchgeführt werden.
3. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die O-Schutzgruppe gegebenenfalls bereits vor Schritt d) entfernt wird, sodass die gebildete OH- Gruppe in Schritt d) gleichzeitig mit dem Alkylierungs- oder Arylierungsreagens reagiert, wodurch Verbindungen der Formel (VIII)
erhalten werden, in der R und R1 wie oben definiert sind und R2 'und R3 ' gleich sind und Cι-C2u-Alkyl-, C5-C2u-Aryl oder einen gegebenenfalls ein- oder mehrfach durch Halogen, Ci-Ce-Alkyl, Halo-C C6-Alkyl, Ci-Ce-Alkoxy, Halo-Ci-Ce-Alkoxy, Aryloxy, Phenyl oder Benzyl substituierten Cι-C20-Alkyl-, C3-C -Heterocyclus-, Silyl-, C5-C2o-Aryl-, C -C2o-Arylsulfonyl- oder d-C2o-Alkylsulfonylrest bedeuten.
4. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass in Schritt a) als Schutzgruppen Schutzgruppen vom Ketal- oder Ethertyp oder Silylschutzgruppen in einer Menge von 1.0 bis 2 Äquivalenten eingesetzt werden.
δ.Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass in Schritt a) zur Beschleunigung der Umsetzung eine wasserfreie Mineralsäure oder eine Sulfonsäu- re als Katalysator in einer Menge von etwa 0.001 bis 0.05 Äquivalenten zugesetzt
wird und im Falle von Silylschutzgruppen eine Base in einer Menge von 1 bis 3 Moläquivalenten zugesetzt wird .
6. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass in Schritt b) als Lösungsmittel Ether aus der Gruppe Tetrahydrofuran oder Methyl-tert.butylether oder Kohlenwasserstoffe aus der Gruppe Toluol oder Heptan und im Falle von Alkaliborhydriden auch Alkohole aus der Gruppe Isopropanol oder Polyethylenglykole eingesetzt werden.

Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die Aktivierung in Schritt c) mittels 1.0 bis 2.0 Äquivalenten an einer Sulfonsäureverbindung aus der Gruppe Tosylchlorid oder -anhydrid, Methansulfonsäurechlorid oder -anhydrid, oder Trifluormethansulfonsäurechlorid oder -anhydrid in Gegenwart von 1.0 bis 2.0 Äquivalenten an einer organischen Stickstoffbase aus der Gruppe Triethylamin, Diisopropylethylamin oder 1 ,4-Diazabicyclo-[2,2,2]-octan erfolgt.
8Nerfahren nach Anspruch 1 , dadurch gekennzeichnet, dass in Schritt c) im Anschluss an die Aktivierung die Umsetzung mit 1 bis 2 Äquivalenten einer S-, Ν- oder P-haltigen Verbindung aus der Gruppe der Sulfide, Thiole, sekundäre Amine oder Phosphinderivate oder deren Salze, die das Strukturelement X-R2 enthalten, erfolgt.
9. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass in Schritt d) 1.0 bis 4.0 Äquivalente an Alkylierungs- oder Arylierungsreagens aus der Gruppe der der Alkyl- oder Arylhalogenide, -sulfate oder sulfonate eingesetzt werden.
10. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass Schritt d) in Gegenwart von 1.0 bis 4.0 Äquivalenten an einer nicht nukleophilen Base oder von wäss- riger ΝaOH unter Phasentransferkatalyse erfolgt.
1. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die saure O- Schutzgruppenentfernung mittels 2 bis 10 Äquivalenten an Cι-C6-Alkohol in Gegenwart von 0.005 bis 0.5 Äquivalenten einer Säure aus der Gruppe Schwefelsäure, oder Sulfonsäure erfolgt.
