Abstract

La présente invention se rapporte à des procédés permettant de favoriser la formation d'un apport de sang collatéral au niveau d'un site ischémique dans un tissu, par culture de cellules à fixation précoce dérivées de la croissance de moelle osseuse obtenue par aspiration in vitro, collecte des cellules à fixation précoce produites par la culture de moelle osseuse et injection du milieu conditionné produit par la culture de ces cellules à fixation précoce dans un site ischémique du coeur ou d'un membre. Les cellules à fixation précoce préférées pouvant être utilisées dans les procédés de cette invention sont des cellules stromales dérivées de la moelle. Toute moelle osseuse d'un donneur peut être utilisée pour la préparation du milieu conditionné. Eventuellement, les cellules à fixation précoce peuvent être transfectées au moyen d'un transgène favorisant l'angiogenèse et codant le facteur 1 alpha induisant l'hypoxie, un facteur de croissance des fibroblastes et/ou une monoxyde d'azote-synthase. L'invention se rapporte également à des milieux conditionnés contenant des cytokines angiogéniques produites à partir de ces cellules et destinées à être injectées dans un tissu, par exemple dans le coeur ou un muscle de membre périphérique, nécessitant la formation d'un apport de sang collatéral.
