Abstract

タンパク質の失活を防ぎ、大掛かりな装置を必要とせず、タンパク質を再現性よく固定化することができ、しかも微小な流路内でもタンパク質を固定化することのできる、新しいタンパク質の固定方法とタンパク質チップを提供する。また、固定化されるタンパク質を細胞接着タンパク質とすることで、細胞をも対象にして基板上の任意の領域に固定化することのできる細胞の固定方法と細胞チップも提供する。

Description

明 細 書 タンパク質の固定方法とタンパク質チップならびに
細胞の固定方法および細胞チップ 技術分野
この出願の発明は、 タンパク質の固定方法とそれによつて作成されるタンパク 質チップに関するものであり、 また、 細胞の固定方法とそれによつて作成される 細胞チップに関するものである。 さらに詳しくは、 この出願の発明は、 タンパク 質の失活を防ぎ、 大掛かりな装置を必要とせず、 タンパク質を再現性よく固定化 することができ、しかも微小な流路内でもタンパク質を固定化することのできる、 新しいタンパク質の固定方法とタンパク質チップに関するものである。
さらにまた、 この出願の発明は、 細胞をも対象にして基板上の任意の領域に固 定することのできる、 新しい細胞の固定方法と細胞チップに関するものである。 背景技術
ヒトゲノムの解読が終了し、 そのデータを利用した病理診断や創薬等を目的と したタンパク質の設計図である DNAを研究 ·解析するため、 DNAチップの研究開 発が進んでいる。 その一方で、 ヒトゲノムの解読データを利用した効果的な病理 診断や創薬等を実現するためには、 この DNAの情報だけでは不十分である。 これ は、 タンパク質が、 生命活動において種々の高次機能を有する物質であることに 起因している。 そのため、 このような特徴を有するタンパク質の機能や情報を調 ベることが必要となっている。
そのためのツールとして、 プロテインチップ (タンパク質チップ) が期待され ており、 盛んにその研究開発が行われている。 そして、 このプロテインチップに おいて、 その基盤技術としてタンパク質を基板上に再現性よく固定するための技 術開発も求められている。
現在、 プロテインチップに関する技術としては、 たとえば、 抗原抗体反応を利 用したタンパク質チップが提案されている (文献 1 )。 このタンパク質チップは、 インクジェットプリン夕の原理で、 平面状の基板上に抗原 (または、 抗体) が配 列 (スポッティング) されており、 前記配列された抗原 (または、 抗体) と、 蛍 光試薬等で標識された抗体 (または抗原) とによる抗原抗体反応を利用して形成 されている。
また、 電気化学的な手法を利用してタンパク質を解析するタンパク質チップも 提案されている (文献 2 )。 このタンパク質チップの場合には、電極としての機能 を有する基板を用いて、 この基板表面に予めタンパク質を固定させ、 そして基板 表面のタンパク質と特異的な結合を形成する電気化学活性物質で標識した試料夕 ンパク質を電気化学的に検出することで目的とするタンパク質を解析している。 さらに、 この出願の発明の発明者は、 細胞を基板上に固定する方法として、 電 気化学的な接着方法も提案している (文献 3 )。 この方法は、細胞非接着性タンパ ク質であるアルブミンを疎水性相互作用で基板に固定化して細胞非接着表面とし、 基板近傍に配置した電極で電気化学的に活性酸化種を生成し、 細胞非接着表面を 細胞接着表面へと改質することで、 細胞接着性タンパク質を介して基板上に細胞 を固定することができ、 細胞のパターン固定を可能としている。
しかしながら、 上記文献 1のタンパク質チップおよび文献 2のタンパク質チッ プのいずれにおいても、 < 1 >あらかじめ固定化しておく必要があるため、固定、 乾燥、 保存の過程においてタンパク質の失活が起こる可能性が高い、 < 2 >比較 的大掛かりな装置を用いる必要がある、 < 3 >決まつた量の夕ンパク質を再現性 よく固定することができない、 さらに、 < 4〉微小な流路内等への固定化ができ ない、 等という問題があった。
また、 上記文献 3の方法による細胞接着領域を形成した基板を利用して、 タン パク質のパターン固定を試みたところ、 タンパク質を安定に固定することはでき なかった。 実際、 文献 3による基板に、 蛍光標識したタンパク質 (プロテイン A や抗体等) 溶液に浸漬し、 蛍光観察したところ、 明確な蛍光シグナルを得ること ができなかった。 アルブミン以外のタンパク質非吸着性物質を用いた場合も、 同 様に安定にタンパク質を基板に固定することはできないという問題があった。 そこで、 この出願の発明は、 以上のとおりの背景から、 従来技術の問題点を解 決し、 試験の都度にタンパク質を固定し、 時間をおかずに試験に供することを可 能とし、これによつて、タンパク質の失活を防ぎ、大掛かりな装置を必要とせず、 夕ンパク質を再現性よく固定化することができ、 しかも微小な流路内でもタンパ ク質を固定化することのできる、 新しいタンパク質の固定方法とタンパク質チッ プを提供することを課題としている。
また、 この出願の発明は、 上記の固定化されるタンパク質を細胞接着タンパク 質とすることで、 細胞をも対象にして基板上の任意の領域に固定化することので きる、 新しい細胞の固定方法と細胞チップを提供することを課題としている。 文献

この出願の発明は、 前記の課題を解決するものとして、 第 1には、 正電荷を帯 びた基板表面に負電荷を帯びた夕ンパク質非吸着性物質を積層して形成した夕ン パク質非吸着性表面を形成する工程と、 タンパク質非吸着性表面を局所的にタン パク質吸着性表面に改質する工程と、 局所的に改質した領域にタンパク質を吸着 させる工程とを含むことを特徴とするタンパク質の固定方法を提供する。
また、 この出願の発明は、 第 2には、 正電荷を帯びた基板表面は、 カチオン性 ポリマーで形成されているタンパク質の固定方法を、 第 3には、 カチオン性ポリ マーは、 ポリエチレンィミンであるタンパク質の固定方法を、 第 4には、 負電荷 を帯びたタンパク質非吸着性物質は、 グリコサミノダリカン、 アルブミンおよび フイブリノ一ゲンからなる群の内少なくとも 1つであるタンパク質の固定方法を、 第 5には、 グリコサミノダリカンは、 へパリン、 へパリン誘導体およびヒアルロ ン酸からなる群の内少なくとも 1つであるタンパク質の固定方法を提供する。 第 6には、 夕ンパク質非吸着性基板表面の夕ンパク質吸着性基板表面への改質 は、 基板近傍に配置した電極に酸化電位もしくは酸化電流を印加して生成される 活性化学種によって行われるタンパク質の固定方法を提供し、 第 7には、 活性化 学種は、 ハロゲン化物イオンを酸化させて生成された活性ハロゲン種であるタン パク質の固定方法を、 第 8には、 活性八ロゲン種は、 次亜臭素酸 (HOBr) または 次亜塩素酸 (H0C1) であるタンパク質の固定方法を提供する。

そして、 この出願の発明は、 第 15 には、 局所的に固定されたタンパク質は、 抗体であるタンパク質チップを、第 16には、局所的に固定されたタンパク質は、 プロティン Αまたはプロティン Gであり、 このプロティン Aまたはプロティン G に抗体を結合させて、 抗体が基板表面に固定されているタンパク質チップを提供 し、 第 17 には、 局所的に固定されたタンパク質は、 酵素であるタンパク質チッ プを提供する。
第 18 には、 局所的に固定されたタンパク質は、 細胞接着タンパク質である夕 ンパク質チップを、 第 19 には、 細胞接着タンパク質は、 フイブロネクチン、 コ ラーゲンおよびラミニンからなる群から少なくとも 1つが選択されるタンパク質 チップを提供する。
さらに、 この出願の発明は、 第 20には、 上記第 18または第 19の発明のタン パク質チップ上に細胞を接着させることを特徴とする細胞の固定方法を提供し、 第 21には、 上記第 20の発明の細胞の固定方法により作成された細胞チップであ つて、 細胞が局所的に固定されていることを特徴とする細胞チップを提供する。 図面の簡単な説明

この出願の発明は、 上記のとおりの特徴をもつものであるが、 以下にその実施 の形態について詳しく説明する。
この出願の発明は、 タンパク質非吸着性の基板表面の局所を改質してタンパク 質を吸着させるタンパク質の固定方法とこの方法により作成されるタンパク質チ ップであることを特徴としている。 具体的には、 このタンパク質の固定方法は、 正電荷を帯びた基板表面に負電荷を帯びたタンパク質非吸着性物質を積層して形 成したタンパク質非吸着性表面を形成する工程と、 タンパク質非吸着性表面を局 所的にタンパク質吸着性表面に改質する工程と、 局所的に改質した領域にタンパ ク質を吸着させる工程とを含むことを特徴としている。
ここで、 「局所的」 とは、 通常はサブ mから数 1 0 0 mのサイズであること を意味する。 そして、 基板の任意の箇所に限定してタンパク質非吸着性の基板表 面をタンパク質吸着性の基板表面に改質することで、 任意の夕ンパク質を基板に 吸着させることができる領域をパターンニングできる。
また、 正電荷を帯びた基板表面とするためには、 カチオン性ポリマーを基板表 面に被覆させ形成させることが好ましく、 カチオン性ポリマーとしては、 ポリエ チレンィミン、 ポリビニルァミン、 ポリアリルァミン、 ポリオル二チン、 ポリリ ジン等を用いることが好ましい。 この出願の発明において基板表面をタンパク質非吸着性にするために、: 基板表 面に固定する負電荷を帯びた夕ンパク質非吸着性物質は、 夕ンパク質が基板に吸 着することを阻害する活性をもたらすものであれば、 特に限定されるものではな い。 たとえば、 具体例としては、 アルブミン、 フイブリノ一ゲン、 グリコサミノ ダリカン等が使用でき、 また、 グリコサミノダリカンとしては、 たとえば、 へパ リン、 へパリン誘導体、 ヒアルロン酸等が使用できる。
すなわち、 この出願の発明は、 タンパク質は、 通常、 負電荷を帯びていること から、 タンパク質を吸着固定させたい箇所には、 正電荷を帯びた基板表面 (たと えば、 カチオン性ポリマーによって形成) を露出させてタンパク質接着性表面へ と改質し、 吸着固定させたくない箇所はタンパク質非吸着性の表面としている。 こうすることで、 負電荷の夕ンパク質を正電荷の基板上に静電相互作用で吸着固 定することができる。 一方で、 表面が負電荷を帯びるようにするだけで、 タンパ ク質非吸着性表面とすることができるわけでないことも判明した。 そして、 静電 的反発だけでなく、 タンパク質非吸着性物質固有の生理活性により、 タンパク質 の吸着が阻害されていることも考慮できる。
この出願の発明におけるタンパク質非吸着性の基板表面のタンパク質吸着性の 基板表面への改質については、 たとえば、 基板近傍にマイクロ電極等の電極を配 置し、 この電極に酸化電位もしくは酸化電流を印加することで、 電気化学的に活 性化学種を局所的に生成させ、 この活性化学種の利用によって、 タンパク質が吸 着することのできる領域を作ることができる。 すなわち、 基板の任意の箇所で、 電極に酸化電位や酸化電流 (あるいは、 印加時間が短いのなら酸化パルスともい うことがある) を印加することによって、 活性化学種が生成され、 その箇所のみ をタンパク質吸着性に変化 (改質) させることができ、 タンパク質が基板に吸着 することができるようになる。 この場合の電極のサイズは小さいため、 これら一 連の反応は上記のとおり局所的に行われる。 もちろん、 この電極は、 任意の箇所 に配置することができ、 また複数の電極を配列させるように配置することもでき る。 さらに、 その移動や動作等は、 電極と連結されているコンピュータによって 制御することもできる。 この活性化学種によって、 タンパク質非吸着性からタン パク質吸着性へと改質されるものとしては、 たとえば、 上記のとおり、 アルブミ ン、 へパリン、 へパリン誘導体、 フイブリノ一ゲン、 ヒアルロン酸等が挙げられ る。
なお、 この活性化学種としては、 ハロゲン化物イオンを酸化させて生成した活 性ハロゲン種であることが好ましい。 ハロゲン化物イオンは、 周期表 17族 (7B) に属するハロゲン元素のイオンのことであり、 いずれのハロゲン化物イオンも利 用することができる。 具体的には、 フッ素 )、 塩素 (Cl)、 臭素 (Br)、 ヨウ素(I) またはァス夕チオン (At)のことである。 活性ハロゲン種としては、 次亜臭素酸 (HOBr) または次亜塩素酸 (H0C1) のいずれかであることがさらに好ましい。 そして、 この出願の発明は、 上記のとおりのタンパク質の固定方法を逐次的に 行うことで、夕ンパク質の固定領域を所要のパターンに配列させることもできる。 特に、 上記のとおり、 複数の電極を所要のパターンに配置して、 逐次的に酸化電 位もしくは酸化電流を印加することで、 複数の、 しかも任意のタンパク質を基板 に吸着させることができる。 このとき、 タンパク質は、 同一のものが複数吸着さ れていてもよいし、種類の異なる複数種類のタンパク質が吸着されていてもよい。 また、 このとき、 活性化学種を利用する場合には、 さらに活性酸化種に対して 非感受性であるタンパク質非吸着性の基板表面修飾剤 (すなわち、 活性酸化種で タンパク質吸着性に改質されない基板表面修飾剤) を、 基板表面に配列固定する ことで、 活性酸化種の生成条件 (すなわち、 酸化電位もしくは酸化電流の条件) に依存せずに、 より効率よく、 しかも正確に再現性よく、 活性酸化種によって、 タンパク質非吸着性からタンパク質吸着性へと改質する(活性酸化種感受性)、ァ ルブミン、 へパリン、 へパリン誘導体、 フイブリノ一ゲン、 ヒアルロン酸等が固 定された領域のみに夕ンパク質を固定することができる。
なお、 活性酸化種に対して非感受性であるタンパク質非吸着性の基板表面修飾 剤は、 ポリエチレングリコール重合体 (PEG重合体) およびメ夕クリロイルォキ シェチルホスホリルコリン重合体 (MPCポリマー) の内少なくともいずれかであ ることが好ましい。 また、 これら基板表面修飾剤は、 通常、 長期間安定である。 具体的には、 既にタンパク質が吸着されている基板に対して、 タンパク質が吸 着されていない任意の箇所に配置された電極に酸化電位 (酸化パルス等) を印加 して、 活性化学種を生成させる。 この活性化学種が、 再び基板の任意の箇所 (局 所) をタンパク質吸着性の基板表面へと改質し、 そこに新たなタンパク質を吸着 させることによって、 タンパク質を任意のパターンで基板に吸着させることがで きる。 上記のとおり、 この際の新たに吸着させるタンパク質の種類は、 特に限定 されるものではなく、 先に吸着されているタンパク質の種類とは、 同一のもので も、 異なるものでもよい。 つまり、 研究、 実験等の目的に合わせて、 タンパク質 の種類は、 適宜に採択することができる。
たとえば、 この出願の発明は、 基板に吸着 (固定) させるタンパク質として抗 体を用いてタンパク質チップを作成し、 このタンパク質チップを抗体チップとし て免疫測定 (ィムノアツセィ) に応用することができる。 また、 このような抗体 チップを作成する工程で、 プロティン Aまたはプロティン Gを吸着させた後に抗 体を結合させてもよい。このプロティン Aやプロティン Gは、抗体の定常領域 (Fc 部位) と結合するので、 抗体を効果的に配向させることができ、 作成される抗体 チップを用いたィムノアツセィの感度と精度を向上させることができる。
さらに、 上記のとおりの固定方法を逐次的に行って、 抗体の固定領域が配列し た夕ンパク質チップを作成することもできる。
この出願の発明は、 基板に吸着させるタンパク質としては、 フイブロネクチン やコラーゲンもしくはラミニン等の細胞接着夕ンパク質を用いることで、 細胞が 固定された細胞チップの作成への利用が可能となる。 そして、 このとき、 基板表 面修飾剤を併用することで、 種々の細胞を培養するマルチ細胞パ夕一ニング培養 や、 長期パターン培養へ応用可能であると期待できる。
この出願の発明に用いることができる 「細胞(培養細胞)」 は、 いかなる由来の 細胞でもよく、 たとえば植物細胞、 昆虫細胞、 動物細胞等が使用でき、 また異種 由来の細胞同士あるいは細胞とコラーゲンゲル膜、 繭糸、 ナイロンメッシュ等の 非細胞との融合細胞でもよい。
もちろん、 培養細胞は、 初代細胞や株化細胞でもよい。 特に動物細胞であるこ とが好ましい。動物細胞における初代細胞としては、ニヮトリ胚由来細胞 (PSG)、 ラット初代心筋細胞、ラット初代肝細胞、マウス初代骨髄細胞、プ夕初代肝細胞、 ゥシ血管内皮細胞、 ヒト初代臍帯血細胞、 ヒト初代骨髄造血細胞、 後根神経節細 胞 (DRG)等のヒト初代神経細胞等が例示される。 また、 株化細胞では、 チヤィニ ーズハムスター卵巣細胞由来の CH0細胞、 ヒト子宮ガン由来の HeLa細胞、 ヒト 肝ガン由来の ι7細胞や HepG2細胞等、 後根神経節細胞 (DRG)等の神経細胞ある いは、 心筋細胞や内皮細胞等も使用できる。 また、 これら細胞にプラスミド導入 やウィルス感染等の遺伝子操作により得られた細胞もこの出願の発明に用いるこ とができる。
また、 これら細胞は、 接着性細胞あるいは浮遊性細胞でもよいが、 接着性細胞 であることが、 この出願の発明の効果をより顕著に得ることができるため好まし い。
さらに、 この出願の発明は、 基板に吸着させるタンパク質として酵素を用いて タンパク質チップとすることで、 酵素反応を利用する生化学分析チップへの応用 もできる。 この酵素についても、 特に制限されるものではなく、 たとえば、 ペル ォキシダーゼやチロシキナーゼ、 グルコースをはじめとする各種糖類の脱水素酵 素等、 各種の酵素が使用できる。
この出願の発明は、 活性化学種を生成するための電気化学反応を制御して進行 させるために必要な、 作用電極および対極等からなる電極系を微小流路チップ内 に内蔵することができ、 微小流路内にも効率よく夕ンパク質を固定化させること ができる。
この出願の発明における基板として用いる材料としては、 たとえば、 カチオン 性ポリマ一そのものはもちろん、 半導体やガラス板、 プラスチック板、 金属薄膜 がコ一ティングされた基板等、 種々のものを利用することができ、 特に限定され るものではない。
以上のような、 この出願の発明によって、 タンパク質の失活を防ぎ、 従来と比 ベて小型かつ安価な電気化学システムを用いるため、 大掛かりな装置を必要とせ ず、 タンパク質を再現性よく固定化することができ、 しかも微小な流路内でも夕 ンパク質を固定化することができる。 このように、 迅速、 かつ、 簡便に基板にタンパク質を固定することが可能とな ることで、 病理診断、 創薬、 食品や環境の分析 ·検査、 医用工学やセンサ工学等 の幅広い分野領域に革新的なツールとして提供することができ、 産業的にも、 経 済的にも大きな効果をもたらすことができる。 そして、 固定化されるタンパク質 を細胞接着夕ンパク質とすることで、 細胞を基板上の任意の領域に固定化するこ とも可能となり、 その活用範囲はより広がることになる。
次に実施例を説明し、 さらに詳しくこの出願の発明について説明する。 もちろ ん、 以下の例によって発明が限定されることはない。 実施例
実施例 1 :電気化学的処理の利用
< I >タンパク質非吸着性物質として、 へパリンを用いた場合
( 1 ) 基板の前処理
ガラス板を基板として用いた。 この基板を洗浄し、 10mg/mLポリエチレンイミ ン (PEI) 水溶液に 2時間浸し、 PEI層を形成させた。 次に、 2mg/mLへパリン水 溶液に 30分間浸すことによって、 へパリンを静電相互作用によって基板表面に 固定させ、 基板表面をタンパク質非吸着性にした。
( 2 ) 電気化学的処理
25 mM KBr を含むリン酸緩衝液中で、 臭化物イオンの酸化電位 (1. 7 V vs. Ag/AgCl) を印加した Ptディスク型マイクロ電極 (電極径: 20 m) を基板近傍 に配置し、 活性ハロゲン種を生成させた。 この処理で、 PEI 層が基板表面が露出 し、 後に固定するタンパク質のリンカ一層として機能する。 ここで、 Ptディスク 型マイクロ電極は作用極(WE)であり、銀/塩化銀電極 (Ag/AgCl)を参照極( E)、 白金板を対極 (CE) とする三極式の電極系にポテンシヨスタツトを接続して行わ れた。
( 3 ) タンパク質の固定
次に、 公知の方法で蛍光標識したプロテイン Aの 0. 025mg/mL溶液に、 基板を 20分間浸潰し、洗浄後蛍光観察によりプロティン Aが局所的に固定されているこ とを確認した。 なお、 図 1に示したとおり、 電位を印加する時間 (5秒間、 10秒 間、 20秒間、 30秒間) で、 プロテイン Aの吸着範囲 (タンパク質の固定サイズ) が制御可能である。
基板に吸着 (固定) させるタンパク質として、 上記のプロテイン A以外にも、 各種抗体、 各種細胞接着タンパク質が局所固定できた。 たとえば、 図 2に示した とおり、 Mouse IgG (抗体) を局所固定することができ、 図 1と同様に、 その吸 着範囲は、 印加時間の長さに比例して拡大することを確認できた。
< 1 1 >タンパク質非吸着性物質として、 アルブミンを用いた場合
上記実施例 1と同様の実験手順で、 タンパク質の基板への固定を行った。
結果は、図には示していないが、実施例 1と同様に、タンパク質の固定ができ、 また、 電位を印加する時間で、 プロテイン Aの吸着範囲 (タンパク質の固定サイ ズ) が制御可能であることを確認できた。
実施例 2 :複数種類のタンパク質のパターンニング
実施例 1の (2 ) および (3 ) の操作を 2回繰り返し、 2種類のタンパク質を 同一の基板上に固定した。 具体的には、 まず、 プロテイン Aの局所固定を介して 蛍光標識した Mouse IgGの固定化を行い (0. 025mg/mL Mouse IgG溶液に 20分間 浸漬)、 その後 2 mg/mL ゥシ血清アルブミン溶液に基板を 30分間浸した。 次に、 再度プロティン Aの局所固定を行った後に蛍光標識した Human IgGの固定化を行 つた (0. 025mgM Human IgG溶液に 20分間浸漬)。
結果は、 図 3 (a) (b) に示したとおり、 同一の基板上に、 Mouse IgGおよび Human IgGそれぞれを固定化させることができた。
実施例 3 :電極系を一体化した基板を用いる活性ノヽロゲン種の生成
実施例 1および 2は、 基板表面に近接させた Pt ディスク型マイクロ電極 (電 極径: 20 /zm) を用いて電気化学処理を行っている。 一方で、 別の基板上に電極 を作成しておき、 これを実施例 1 ( 1 ) と同様に前処理を施したガラス基板にス ぺーサ一を用いて対向させて電気化学処理を行う方法が、 実用上望まれることが ある。 さらに、 通常は別途用意される参照電極も電極基板に搭載されることが望 ましい。 そこで、 実施例 3として、 1対の白金の電極がパターン化された基板の みを用いて活性ハロゲン種を電解生成し得ることを示した。
まず、 図 4に示したとおり、 基板電極の一方の電極を作用極 )、 もう一方 の電極を対極 (CE)、 別途挿入した銀塩化銀電極 (Ag/AgCl) を参照極 (RE) とし て、 0. 1 M KC1を含むリン酸緩衝液 (0. 1 M、 H 7. 5) 中でサイクリックポル夕モ メトリー (CV) を行った。 この図 4では、 25 niM KBr存在下の場合では、 実線で 示したとおり、 臭化物ィオン酸化の寄与が支配的である酸化電流が観測された。 また、 非存在化の場合では、 点線で示したとおり、 塩化物イオン酸化の寄与が支 配的である酸化電流が観測された。
次いで、 図 5に示したとおり、 基板電極の一方の電極を作用極 (WE)、 もう一 方の電極を参照極(RE) (兼対極) として同様の溶液中で CVを行ったところ、 KBr 存在下では、 臭化物イオン酸化の寄与が支配的である酸化電流、 非存在化では塩 化物イオン酸化の寄与が支配的である酸化電流が観測され、 得られる CV形状は 図 4の場合と同等であった。 すなわち、 外部に参照極を別途用意せずとも、 基板 上の白金電極を參照極として用いる二極式の電気化学系でハロゲン化物イオンの 酸化反応が制御可能であつた。
この結果は、電極系の全てが微小な基板上に集積できることを示すものであり、 この出願の発明のタンパク質チップや、 その関連装置の構造を大幅に簡素化する 効果を有する。
実施例 4:微小流路内へのタンパク質の固定
本実施例における、 微小流路が形成された基板は、 図 6に例示したとおり、 実 施例 3の電極基板と、 実施例 1 ( 1 ) と同様にへパリンを固定化したガラス基板 を PETフィルムのスぺーサーを介して挟み込むことによつて流路を形成させた。 この流路に 25 mM KBrを含むリン酸緩衝液を充填し、 作用電極に臭化物イオン の酸化電位 (2. 2 V vs. Pt) を印加してタンパク質吸着領域を形成した。 次に、 蛍光標識したプロティン Aの 0. 025mg/mL溶液を 10分間充填し、 洗浄した後蛍光 顕微鏡を用いて観察した。
結果は、 図 7に示したとおり、 へパリンを固定化した基板上には、 電極基板の 電極位置と対応してタンパク質 (プロテインお を固定させることができた。 微小流路と組み合わせたタンパク質チップは、 用いる溶液を少量とし、 操作の 連続性と再現性も向上させる効果を有する望ましい形態である。 この実施例は、 これまで極めて困難であった微小流路内への局所へのタンパク質固定を初めて実 施したものであり、 タンパク質チップが有する問題点を解決する効果を有する。 実施例 5 :細胞のパターンエング
基本的な操作は、実施例 1の (2 ) ( 3 ) の操作と同様である。基板の表面に局 所的に接着させるタンパク質として、 細胞接着タンパク質であるフイブロネクチ ン (Fibronect in) をパターニング接着させた。
そして、フイブロネクチンをパターンニング接着させた箇所に培養細胞として、 HeLa細胞 (HeLa cel l) を接種して培養すると、 図 8に示したとおり、 このフィ ブロネクチンのパターン上にのみ選択的に HeLa細胞の接着を誘導することがで きた。 図 8 (a) は、 フイブロネクチンを蛍光標識して可視化している。 図 8 (b) は、 この基板上で HeLa細胞を培養した結果の位相差顕微鏡写真像である。
また、 図には示していないが、 同様の方法で、 他の培養細胞、 たとえば、 神経 細胞や心筋細胞、 内皮細胞等の付着性細胞でもパターンニングが可能だつた。 実施例 6 :微小流路内における細胞のパターンニング
図 9に例示したように、 実施例 4で利用した装置と同様の装置、 つまり電極基 板と、 へパリンを固定化したガラス基板を PETフィルムのスぺーサーを介して挟 み込むことによって流路を形成させたものを用いた。 そして、 細胞接着タンパク 質であるフイブロネクチンを固定した後、 培養細胞として HeLa細胞を接種して 培養した。
結果は、 図 9に示したとおりであった。 この図 9に示したとおり、 基板の微小 流路内の局所に、 細胞 (HeLa細胞) を接着することができた。 このことから、 細 胞診断等の細胞チップを用いる応用には、 この結果のような 「その場固定」 が重 要であることを考慮すると、 この出願の発明の細胞チップを細胞診断等にも十分 に活用することができる。 また、 この出願の発明の細胞チップは、 基礎生物学も しくは創薬等の応用研究の立場から、 腫瘍細胞と T細胞との結合等の細胞間相互 作用を研究、 解析等のためのツールとしても活用できる。 実施例 7 :微小流路を有する基板 (微小流路チップ) へのタンパク質の固定
( 1 ) 微小流路チップの作製
図 1 0に示したように、 一連の微細加工技術を用いてガラス基板上に Pt をパ ターニングして、 電極を内蔵する電極基板を作製した。 この電極基板とガラス基 板をスぺーサ一 (シリコンラバー:厚さ 50 τώ を介して、 挟み込むことによ つて流路を形成した。 この流路内への送液は、 インレットの細管を目的の溶液の 入ったリザ一パーに連結し、 アウトレットで吸引することで行った。 インレット のリザ一パーには参照極として銀塩化銀電極を設置した。
( 2 ) 流路内へのタンパク質の固定化とィムノアッセィ
<Α>抗体のパターンニング
作製した微小流路チップに、 図 1 1に例示したように、 以下のステップでサン ドイツチイムノアツセィを行った。
ポリエチレンィミン (ΡΕΙ) 水溶液 (5 mg/mL) を流路内に導入し、 30分インキ ュべー卜した。
次に、 へパリン水溶液 (2 mg/mL) を流路内に導入し、 20分インキュベートす ることで、 へパリンを基板表面に静電相互作用によって固定した。
次に、 25 mM KBr を含むリン酸緩衝液を流路内に充填した後, 作用電極に臭化 物イオンの酸化電位 (1. 7 V vs. Ag/AgCl) を、 5秒または 10秒印加し、 基板に 固定した PEI/へパリン層を改質した。
次に、 プロテイン A溶液 (25 a g/mL) を流路内に充填し、 30分インキュベー トすることで、 基板に固定した。
次に、一次抗体溶液(ャギ由来抗マウス IgG : 25 ti g/mL) を流路内に導入し、 30分ィンキュベー卜することで抗体を基板上に固相化した。
次に、 牛血清アルブミン (BSA) 溶液 (5 mg/mL) を流路内に導入し、 30分イン キュベ一卜することでブロッキングした。
次に、 抗原溶液 (マウス IgG : 25 g/mL) を流路内に導入し、 30分インキュ ベー卜した。
次に、 蛍光標識二次抗体溶液 (FITC標識ャギ由来抗マウス IgG : 25 g ) を流路内に導入し、 30分インキュベートした後、 蛍光観察した。
結果は、 図 1 2に示したとおり、 へパリンをパターン固定 (パターンニング) した箇所のみで、抗体反応を確認することができた。また、反応時間に依存して、 反応強度も向上することも確認できた。
< B〉マルチ抗体のパターンニング
作製した微小流路チップに、 図 1 3 (a) に例示したように、 以下のステップ でサンドイッチィムノアツセィを行った。 なお、 操作手順は、 基本的には、 上記
<A>とほぼ同様である。
ポリエチレンィミン (PEI) 水溶液 (5 mg/mL) を流路内に導入し、 30分インキ ュベー卜した。
次に、 へパリン水溶液 (2 mg/mL) を流路内に導入し、 20分インキュベートす ることで、 へパリンを基板表面に静電相互作用によって固定した。
次に、 25 niM KBr を含むリン酸緩衝液を流路内に充填した後, 作用電極に臭化 物イオンの酸化電位 (1. 7 V vs. Ag/AgCl) を 10秒印加し、 基板に固定した PEI/ へパリン層を改質した。
次に、 プロテイン A溶液 (25 z /mL) を流路内に充填し、 30分インキュベー トすることで、 基板に固定した。
次に、 一種類目の抗体 (Cy3-labeled mouse: 25 ^ g/mL) を流路内に導入し、 30分ィンキュベ一トすることで抗体を基板上に固相化した。
次に、 牛血清アルブミン (BSA) 溶液 (5 mg/mL) を流路内に導入し、 30分イン キュベートすることでプロッキングした。
次に、 PEI/へパリン層の改質操作を行い、 次いで、 プロテイン Aの基板への固 定操作も再度行った。
次に、 二種類目の抗体 (Cy2-labeled human IgG: 25 n g/mL) を流路内に導 入し、 30分インキュベートした後、 蛍光観察した。
その結果、 図 1 3 (b) (c) (d) に示したように、一種類目の抗体(Cy3-labeled mouse) および二種類目の抗体 (Cy2-labeled human IgG) いずれにおいても、 へ パリンをパターン固定 (パ夕一ンニング) した箇所のみで、 抗体反応を確認する ことができた。
実施例 8 :基板表面修飾剤を用いてのタンパク質の固定
基板表面修飾剤であるポリエチレングリコール (PEG) や MPCポリマーは、 活 性酸化種である次亜臭素酸 (HOBr) に対する耐性を有する長期間安定な表面修飾 剤である。
この基板表面修飾剤である PEGや MPCポリマーのパターニング方法について説 明する。 なお、 ポリジメチルシロキサン (PMS) スタンプは、 フォトリソグラフ ィ一によりフォトレジスト (膜厚 9 xm) をパターニングしたガラス基板を铸型 として作製した。
図 1 4に示したように、 この PDMS スタンプをガラス基板に置き、 PEG (Poly (ethylene glycol) dimethacrylate) または MPCポリマーの溶液を、 毛細 管現象を利用してスタンプの凹んだ部分と基板表面の隙間に流し込んだ。ここで、 PEG の溶液は平均分子量 550 の Poly (ethylene glycol) dimethacrylate (99. 5wt %) と光重合開始剤の 2_hydroxy-2-methylpropiophenoiie (0. 5wt%) の 混合溶液を用いた。 また、 MPCポリマーは 5wt%のエタノ一ル溶液である。
PEGの溶液を流し込んだ場合には、 UV照射 (365腿, 15 mW/cm2, 20 sec) でポ リマーを硬化させた後、 PDMSスタンプを取り除いた。 MPCポリマ一の溶液の場合 には、 基板を 20分乾燥させた後、 スタンプを取り除いた。
図 1 5は、 PEGおよび PCポリマ一をパターエングしたガラス基板上で細胞を 培養した様子を示した図である。細胞は、 HeLa細胞またはゥシ大動脈由来血管内 皮細胞を用いて、 公知の培養条件で培養した。 結果、 ガラス基板が露出している 領域にのみ細胞の接着 ·伸展が観察された。 特に、 ゥシ大動脈由来血管内皮細胞 は、 このパターンを維持したまま 1ヶ月を超える長期培養が可能であった。
( 1 ) 微小流路チップにおけるタンパク質の固定
表面修飾剤として MPCポリマーを微小流路チップの基板にパターン固定し、 ま た、 活性酸化種で改質するへパリンをパターン固定した。
図 1 6に示すように、 基板上に作製した MPCポリマーとへパリンの境界領域を マイクロ電極で生成した次亜臭素酸で処理した。 すると、 PEI/へパリン層のみが 改質され、 したがって、 これらの基板表面修飾剤で流路チップ上を予めパター二 ングしておくことで、意図しない領域へのタンパク質の吸着を防ぐことができる。
( 2 ) 微小流路チップにおけるィムノアッセィ
次に、 図 1 7に示すように、 予め MPCポリマーをパターニングしたガラス基板 を用いて微小流路チップを作製した。
そして、 この微小流路チップに実施例 7 ( 2 ) で示した操作を適用し、 ィムノ アツセィを行った。
結果は、 MPCポリマーで被覆していない領域でのみ蛍光が検出され、 上述した ように意図しない領域へのタンパク質吸着を抑制できた。 これらの基板表面修飾 剤は長期間安定であるため、 図 1 8に示したように、 種々の細胞を培養するマル チ細胞パターニング培養や、 長期パターン培養へ応用可能であると期待できる。 産業上の利用可能性
以上、 詳しく説明したとおり、 この出願の発明によって、 タンパク質の失活を 防ぎ、 大掛かりな装置を必要とせず、 タンパク質を再現性よく固定化することが でき、 しかも微小な流路内でもタンパク質を固定化することができ、 さらに、 夕 ンパク質の固定領域の配列させることもできる。
また、 この出願の発明によって、 上記の固定化されるタンパク質を細胞接着夕 ンパク質とすることで、 細胞をも対象にして基板上の任意の領域に固定化するこ ともできる。

Claims

請求の範囲
1 . 正電荷を帯びた基板表面に負電荷を帯びた夕ンパク質非吸着性物質を積層 して形成したタンパク質非吸着性表面を形成する工程と、 タンパク質非吸着性表 面を局所的にタンパク質吸着性表面に改質する工程と、 局所的に改質した領域に タンパク質を吸着させる工程とを含むことを特徵とするタンパク質の固定方法。
2. 正電荷を帯びた基板表面は、 カチオン性ポリマーで形成されている請求項 1に記載のタンパク質の固定方法。
3 . カチオン性ポリマーは、 ポリエチレンィミンである請求項 1または 2に記 載のタンパク質の固定方法。
4. 負電荷を帯びたタンパク質非吸着性物質は、 グリコサミノダリカン、 アル ブミンおょぴフイブリノ一ゲンからなる群の内少なくとも 1つである請求項 1か ら 3いずれかに記載のタンパク質の固定方法。
5. グリコサミノダリカンは、 へパリン、 へパリン誘導体およびヒアルロン酸 からなる群の内少なくとも 1つである請求項 4に記載のタンパク質の固定方法。
6. タンパク質非吸着性基板表面のタンパク質吸着性基板表面への改質は、 基 板近傍に配置した電極に酸化電位もしくは酸化電流を印加して生成される活性化 学種によって行われる請求項 1から 5いずれかに記載のタンパク質の固定方法。
7 . 活性化学種は、 ハロゲン化物イオンを酸化させて生成された活性ハロゲン 種である請求項 6に記載のタンパク質の固定方法。
8. 活性ハロゲン種は、 次亜臭素酸 (HOBr) または次亜塩素酸 (H0C1) である 請求項 7に記載のタンパク質の固定方法。
9. 請求項 1から 8いずれかに記載のタンパク質の固定方法を逐次的に行うこ とによって、 タンパク質の固定領域を配列することを特徴とするタンパク質の固 定方法。
1 0. 活性化学種に対して非感受性であるタンパク質非吸着性の基板表面修飾 剤を基板表面に配列固定して、 タンパク質の固定領域を設定する請求項 6から 9 いずれかに記載の夕ンパク質の固定方法。
1 1 . 基板表面修飾剤は、 ポリエチレングリコール重合体およびメタクリロイ ルォキシェチルホスホリルコリン重合体の少なくともいずれかである請求項 1 0 に記載の夕ンパク質の固定方法。
1 2. 請求項 1から 1 1いずれかに記載のタンパク質の固定方法により作成さ れた夕ンパク質チップであって、 夕ンパク質非吸着性の基板表面が夕ンパク質吸 着性の基板表面に局所的に改質され、 タンパク質が局所的に固定されていること を特徴とするタンパク質チップ。
1 3. 基板には、 微小流路および電極系が内蔵されている請求項 1 2に記載の タンパク質チップ。
1 4. 内蔵されている電極系は、 一対の電極からなる二極式であり、 対極が白 金である請求項 1 3に記載のタンパク質チップ。
1 5. 局所的に固定されたタンパク質は、 抗体である請求項 1 2から 1 4いず れかに記載の夕ンパク質チップ。
1 6. 局所的に固定されたタンパク質は、 プロテイン Aまたはプロテイン Gで あり、 このプロテイン Aまたはプロテイン Gに抗体を結合させて、 抗体が基板表 面に固定されている請求項 1 2から 1 4いずれかに記載のタンパク質チップ。
1 7. 局所的に固定されたタンパク質は、 酵素である請求項 1 2から 1 4いず れかに記載の夕ンパク質チップ。
1 8. 局所的に固定されたタンパク質は、 細胞接着タンパク質である請求項 1 2から 1 4いずれかに記載のタンパク質チップ。
1 9. 細胞接着タンパク質は、 フイブロネクチン、 コラーゲンおよびラミニン からなる群から少なくとも 1つが選択される請求項 1 8に記載のタンパク質チッ プ。
2 0. 請求項 1 8または 1 9に記載のタンパク質チップ上に細胞を接着させる ことを特徵とする細胞の固定方法。
2 1. 請求項 2 0に記載の細胞の固定方法により作成された細胞チップであつ て、 細胞が局所的に固定されていることを特徴とする細胞チップ。
