Abstract

Gegenstand der vorliegenden Anmeldung sind 150 neue essentielle Gene und deren Genprodukte von Bacillus licheniformis und hinreichend ähnliche Gene und Proteine. Diese Faktoren übernehmen in vivo jeweils elementare Lebensprozesse, beispielsweise die Replikation (zum Beispiel DNA-Polymerease, Helicase oder Gyrase), die Transkription (zum Beispiel RNA-Polymerase), die Proteinbiosynthese (ribosomale Proteine, Aminocyl-tRNA-Synthetasen, Initiations- und Elongationsfaktoren), die Sekretion von Proteinen (zum Beispiel Translocase) oder den Energie-Stoffwechsel. Ein Fehlen dieser Gene ist für die betreffenden Zellen unmittelbar letal und kann nicht durch eventuelle Verbindungen aus dem Nährmedium ausgeglichen werden. Somit können die zugehörigen Gene als Selektionsmarker genutzt werden. Dadurch lassen sich insofern verbesserte biotechnologische Produktionsverfahren durch Mikroorganismen etablieren, als eine Selektion über diese Gene ohne Antibiotika auskommt und zudem nicht durch Bestandteile üblicher Nährmedien ausgeglichen werden kann.
