緑茶葉酵素処理法
技術分野
[0001] 本発明は、原料緑茶茶葉を、抽出に先立ち、酵素溶液と反応させることにより含有 成分を変換し、緑茶茶葉の品質を改質し、該茶葉を抽出処理することにより、香味の 優れた高香味緑茶抽出液を製造する方法に関する。
背景技術
[0002] 緑茶抽出液の製造において、緑茶の抽出の際に各種酵素を作用させて、抽出を促 進し、その抽出液の香味若しくは品質を向上させようとする試みは以前より数多く提 案されている。しかし、このような酵素処理を用いた抽出方法においては、通常、茶 抽出工程で酵素処理が行われるので、酵素処理の効果を上げるためには、比較的 長い処理時間が必要となる。その一方で、処理の長時間化は、抽出工程での品質劣 化や、生産効率の低下につながる。したがって、その酵素処理は、通常、短時間でし か設定できず、従来の酵素処理は、抽出の促進等、特定範囲のものに限られ、その 効果にも限界があった。
[0003] 例えば、特開 2003— 210110号公報には、低温かつ短時間で効率的に茶葉から 有用成分を抽出し、苦渋味が少なぐ澱が生成しないコクと旨みを有する茶抽出液の 製造方法が開示されている。具体的には、抽出に際して、セルラーゼ、へミセルラー ゼ、ぺクチナーゼ等の酵素を用いて酵素分解抽出処理を行ない、可溶性固形分の 抽出効率を高める方法が示されている。該公報に開示される茶抽出液の製造方法 では、比較的長時間の酵素分解抽出処理時間が採用されている。すなわち、該方法 における酵素分解抽出処理では、 0. 5〜5時間、実施例では 2時間攪拌しながら酵 素分解抽出処理が実施されている。これは通常緑茶飲料を製造する場合の抽出ェ 程よりはるかに長い時間をかけており、抽出時の香味劣化等、茶抽出液の品質への 影響が懸念される。また、長時間の酵素分解抽出処理は、飲料製造工場での生産 効率を下げることになることから、該方法は、実用的とは考えにくい。
[0004] このような茶抽出液の製造における、酵素分解抽出処理の問題を解消する方法と して、次のような 2つの方法が提案されている。まず、その一つの方法としては、酵素 処理を、飲料製造工程上の抽出段階で使用するのではなぐ製造法の自由度が比 較的高ぐ抽出時間も長く設定できる「緑茶エキス」の製造の際に使用する方法であ る。たとえば、特開 2003— 144049号公報には、緑茶に対してプロテアーゼとタンナ ーゼを併用処理して旨みゃコク味が強ぐ渋味の少ない緑茶エキスを製造する方法 が開示されている。この方法では、酵素反応を行いながら、同時に抽出を行なってい る。具体的な実施例としては、上記酵素を添加して、 16時間酵素処理を行い、抽出 を行なうことが示されている。この方法は、プロテアーゼとタンナーゼを併用処理して 、茶葉中のタンパク質とタンニンの結合の分解とタンパク質の分解を行なって、旨み ゃコク味の強い抽出エキスを製造するものである。該方法は、抽出時間を長く設定で きる「緑茶エキス」の製造の際に適用できる方法である。
[0005] もう一つは、予め製茶の段階で酵素処理を行なっておいた茶葉を用いて、抽出を 行なう方法である。特開昭 51— 115999号公報には、採取した生茶葉に対してタン ナーゼ処理し、これを焙じることによって、冷水でも抽出可能な乾燥茶葉或いは該乾 燥茶葉から抽出、乾燥されたインスタント茶粉末の製造方法が開示されている。また 、特開平 5— 308901号公報には、茶生葉に対して、蒸熱や釜入り工程の後に、ぺク チナーゼゃセルラーゼなどの細胞壁消化酵素で処理を行なレ、、該酵素処理茶葉を 、通常の加熱乾燥処理工程に付して、水でも抽出可能な茶葉の製造を行なう方法が 開示されている。これらはいずれも抽出を促進し、冷水抽出が可能な、抽出効率の 向上を主眼においた発明である。その酵素処理の効果にも限界があって、茶含有成 分を変換し、香味の改良を行なっているものではない。
[0006] 一方で、近年、茶類の製造方法において、茶葉を分解酵素等を用いて茶葉の原料 成分を変え、味覚や風味を改良した茶類の製造方法が開示されている。例えば、特 開 2003— 153651号公報には、茶類の渋みの多い生葉原料又は乾燥茶葉に、タン ニン分解、多糖類分解、蛋白質分解をおこなう少なくとも一種以上の分解酵素を添 カロして、酵素処理し、茶葉原料中の成分を変換する方法を提案している。具体的に は、緑茶等の生茶葉を蒸煮し、冷却したもの、或いは、烏龍茶原料茶葉を萎凋処理 を施したものに、タンニン分解酵素であるタンナーゼ、多糖類分解酵素であるガマナ ーゼ、蛋白質分解酵素であるコクラーゼを添加して酵素処理を行なっている。これを 通常の製茶工程に付して、茶葉の原料成分を変え、茶の渋味を調整して、甘味と旨 味のある茶製品を製造することが開示されている。し力しながら、この製茶工程にお ける酵素処理も、分解酵素を茶葉に噴霧、混合攪拌して、原料成分中のタンニンや 、多糖類、及び蛋白質を分解処理するにとどまるものであり、茶葉中の香味成分を変 換して、高香味の茶抽出液を製造し得るような酵素処理を行なっているものではない

発明の開示
発明が解決しょうとする課題
[0008] 本発明の課題は、原料緑茶茶葉を酵素溶液と反応させて含有成分を変換し、緑茶 茶葉の品質を改質し、該茶葉を抽出処理することにより、香味の優れた高香味緑茶 抽出液を製造する方法を提供することにある。また、本発明の課題は、原料緑茶茶 葉の含有香味成分を効果的に変換し得る酵素処理方法を提供し、原料緑茶茶葉に 高香味を付与して、該茶葉から香味に特徴付けられた緑茶抽出液を製造する方法を 提供することにある。
課題を解決するための手段
[0009] 本発明者は、上記課題を解決すベぐ鋭意検討する中で、乾燥緑茶葉を粉砕した 或いは緑茶葉製造工程で粉砕された粉砕緑茶葉の状態で、該粉砕緑茶葉に対して 、酵素溶液を 1: 0. 2〜: 1: 5の重量比という水分含量が非常に制限された状態で混 合して、反応を行なうことにより、反応が、飛躍的に進行することを見い出した。この特 定条件での粉砕緑茶葉の酵素処理により、従来の方法では得られなかった、乾燥緑 茶葉中の香味成分の効果的な変換が可能となった。更に、該方法を用いて、緑茶茶 葉の品質を改質し、該茶葉を抽出処理することにより、香味の優れた高香味緑茶抽 出液を製造することが可能となった。 [0010] すなわち、本発明の緑茶抽出液の製造方法は、乾燥緑茶葉を粉碎した或いは緑 茶葉製造工程で粉砕された、粉碎緑茶葉と、酵素溶液を 1 : 0. 2〜: 1 : 5の重量比で 混合して反応を行なったのち、水もしくは温水で抽出することにより、香味の優れた緑 茶抽出液を製造する方法からなる。本発明において、乾燥緑茶葉の粉砕度は、 20メ ッシュパス成分が 50重量%以上の粉砕度であることが好ましい。また、本発明におい ては、粉砕緑茶葉と、酵素溶液との反応終了後、一旦乾燥するか、若しくは湿潤状 態のまま密封容器に充填した後殺菌することにより、処理後の加工茶葉を流通、保 存の便に供することができる。本発明において、緑茶抽出液の香味成分の変換のた めに、粉砕緑茶葉と、酵素溶液との反応に使用する特に優れた酵素として、ポリフエ ノールォキシダーゼ、 β—グリコシダーゼ、リパーゼ、クロロゲン酸エステラーゼ、ヌク レアーゼ、プロテアーゼ、ラタターゼ、インべノレターゼ、ぺクチナーゼ、キシラナーゼ 及びデァミナーゼを挙げることができる。
[0011] すなわち具体的には本発明は、(1)乾燥緑茶葉を粉砕した或いは緑茶葉製造ェ 程で粉砕された、粉碎緑茶葉と、酵素溶液を 1 : 0. 2〜: 1 : 5の重量比で混合して反応 を行なったのち、水もしくは温水で抽出することを特徴とする香味の優れた緑茶抽出 液の製造法や、(2)粉碎緑茶葉と、酵素溶液の重量比が、 1 : 0. 2〜: 1 : 3の重量比で あることを特徴とする上記(1)記載の香味の優れた緑茶抽出液の製造法や、 (3)酵 素溶液の量が、茶葉と混合した際に、当該混合物から水分が分離してこない範囲で あることを特徴とする上記(1)又は(2)記載の香味の優れた緑茶抽出液の製造法や 、(4)乾燥緑茶葉の粉碎度が、 20メッシュパス成分が 50重量%以上の粉砕度である ことを特徴とする上記(1)〜(3)のいずれか記載の香味の優れた緑茶抽出液の製造 法や、(5)粉砕緑茶葉と、酵素溶液を 1 : 0. 2〜: 1 : 5の重量比で混合して反応を行な つたのち、加熱処理を行うことを特徴とする上記(1)〜(4)のいずれか記載の香味の 優れた緑茶抽出液の製造法からなる。
[0012] また本発明は、(6)粉砕緑茶葉と、酵素溶液との反応に使用する酵素が、ポリフエノ 一ルォキシダーゼ、 β—グリコシダーゼ、リノくーゼ、クロロゲン酸エステラーゼ、ヌクレ ァーゼ、プロテアーゼ、ラタターゼ、インべノレターゼ、ぺクチナーゼ、キシラナーゼ及 びデアミナーゼから選択される 1又は 2以上の酵素であることを特徴とする上記(1)〜 (5)のいずれか記載の香味の優れた緑茶抽出液の製造法や、(7)乾燥緑茶葉を粉 砕した或いは緑茶葉製造工程で粉砕された、粉碎緑茶葉と、酵素溶液を 1 : 0. 2〜1 : 5の重量比で混合して反応を行なったのち、一旦乾燥するか、若しくは湿潤状態の まま密封容器に充填した後殺菌することによって調製した、高香味緑茶抽出液調製 用の緑茶茶葉や、(8)上記(7)記載の緑茶茶葉を、水若しくは温水で抽出することを 特徴とする香味の優れた緑茶抽出液の製造法や、(9)上記(1)〜(6)のいずれか記 載の緑茶抽出液の製造法又は上記(8)記載の緑茶抽出液の製造法によって製造さ れた抽出液を、そのまま若しくは他の飲料原料と混合した後、殺菌、充填したことを 特徴とする容器詰緑茶飲料力 なる。
発明の効果
[0013] 本発明の緑茶抽出液の製造方法は、原料緑茶茶葉の含有香味成分を酵素溶液と の反応を促進して、効果的に変換することを可能とする。そして、従来の方法では難 しかった、荒茶段階以降の原料緑茶茶葉の含有香味成分の変換を可能として、原料 緑茶茶葉に高香味を付与して、該茶葉から香味に特徴付けられた緑茶抽出液を製 造する方法を提供する。本発明により、新しい香味を有する抽出液を創出できる。 発明を実施するための最良の形態
[0014] 本発明は、乾燥緑茶葉を粉砕した或いは緑茶葉製造工程で粉砕された、粉砕緑 茶葉と、酵素溶液を 1 : 0. 2〜: 1 : 5の重量比で混合して反応を行なったのち、水もしく は温水で抽出することを特徴とする香味の優れた緑茶抽出液の製造法からなる。
[0015] 本発明の緑茶抽出液の製造法に用いられる乾燥緑茶葉は、通常、緑茶葉とよばれ るものであっていわゆる荒茶段階にまで加工されていればよい。このとき水分量は、 概ね 6重量%以下である。本発明においては、該乾燥緑茶葉を、粉砕して粉砕緑茶 葉の状態で、酵素溶液と反応させる。粉砕緑茶葉を調製するには、乾燥緑茶葉を通 常の粉碎機を用いて粉砕すればよい。また、粉砕緑茶葉は、緑茶葉の製造工程で 粉砕された、粉茶とよばれる製茶工程の副産物を用いてもよい。粉碎緑茶葉の粉碎 の細かさ(粉砕度)は、 20メッシュパス成分が 50重量%以上(50〜: 100重量%)の粉 砕度であることが好ましぐ 70重量%以上が最も好ましい。
[0016] 本発明の緑茶抽出液の製造法に用いられる酵素溶液としては、基本的には、茶葉 に対して反応して抽出液の香味変化が確認できるものであって、食品衛生法上使用 が認められている酵素であれば、何れの酵素でも用いることができる。具体的には、 アミラーゼ、ダルコアミラーゼ、 CGTase、デキストラナーゼ、セルラーゼ、ダルカナー ゼ、グルコースイソメラーゼ、キシラナーゼ、へミセルラーゼ、キシラナーゼ、マンナナ ーゼ、ぺクチナーゼ、ラタターゼ、インベルターゼなどの糖質に作用する酵素、プロテ ァーゼ、ぺプチダーゼ、デァミナーゼ、トランスグルタミナーゼなどのタンパク質.ぺプ チドに作用する酵素、リパーゼ、エステラーゼなどの脂質に作用する酵素、そのほか のカタラーゼ、ォキシダーゼ、ヌクレアーゼ、グルタミナーゼなどを、挙げることができ る。その中でも特徴ある香味の付与という観点では j3—グリコシダーゼ(WO2003/ 056930号公報)、ポリフエノールォキシダーゼ、リパーゼ、クロロゲン酸エステラーゼ 、ヌクレアーゼ、プロテアーゼ、ラタターゼ、インベルターゼ、ぺクチナーゼ、キシラナ ーゼ及びデァミナーゼが好ましい。これらは必ずしも単一で使用する必要はなぐ効 果を見て、 2種以上を組み合わせて使用することができる。また、数種の酵素が混ざ つた複合酵素の形で使用しても良い。酵素の使用量は、反応による効果が生じれば 特に限定はない。その具体的範囲は、その活性の強さや製剤中の賦形剤の含量で 大きく変わってくるので一概には決めかねる力 一般には茶葉に対して 0. 01重量% 〜50重量%、好ましくは 0. 1重量%〜10重量%程度でぁる。
本発明の緑茶抽出液の製造方法において酵素溶液との反応は、水分含量を制限 した状態でおこなわれる。これは従来の緑茶葉に対する酵素処理が、同時に成分を 抽出することを目的としていたために、比較的大量の水分量下でおこなわれていたこ とと大きく異なる。すなわち、本発明の緑茶抽出液の製造法における酵素溶液との反 応に際しての水分量は、茶葉中に酵素が分散して反応が十分に進行するのに必要 な量であり、かつ反応中に茶成分が水分に移行しない量、具体的には茶葉重量の 0 . 2倍から 5倍程度(粉砕緑茶葉に対して酵素溶液を 1 :0. 2〜1 :5の重量比)である。 なお、本発明でいう「酵素溶液」は予め水に酵素を溶解したもののみならず、茶葉と 水とを混合したのちに酵素を添加、混合すること、或いは、逆に茶葉と酵素を混合し たのちに水を添加、混合することもできる。つまり、結果として茶葉の 0. 2〜5倍量の 酵素溶液を添加することになればよい。 [0018] 好ましい酵素溶液量の上限は、使用する茶葉の性質によって変化するが、茶葉と 混合した際に「当該混合物から水分が分離してこない範囲」であることが望ましい。本 発明における「当該混合物から水分が分離してこない範囲」とは、以下の方法で定義 すること力 Sできる。すなわち、該「水分が分離してこない範囲」とは、原料茶葉 10gに 対し所定量の酵素溶液を混合したものを 50°Cで 2時間放置した後、 200メッシュステ ンレス円形金網(目開き 75 x m、直径 75mm)に載せて均一に広げて 5分間放置し た際に、分離した水分が 5ml以下であることを指す。この方法を本発明において「分 離水分判定法」と呼ぶこととする。また、乾燥茶葉と酵素溶液の重量比が 1 :3以下で あれば水分の分離がより少ないので好ましい。更には、本発明の一形態である、反 応後に一旦乾燥させた後に抽出するような場合には、乾燥効率の面から、より少ない 水分含量、すなわち茶葉と酵素溶液の重量比力 S1: 2以下、最も好ましくは 1 : 1以下が よい。一方、好ましい酵素量の下限は、茶葉に対する重量が 0. 5倍のときであって、 それ以上であると酵素溶液との反応がより効果的に進む。
[0019] このとき特に低水分量の場合には酵素処理の原料となる乾燥茶葉自体が有する水 分量が影響する。本発明でいう茶葉重量の 0. 2倍量とは、水分量 6%以下の乾燥茶 葉を使用した場合の比率を指す。したがって、それ以上の水分量を有する乾燥茶葉 を使用する場合には、超過相当分の水分量を勘案して更に酵素溶液量を減らしても よいことになる。酵素反応において、該水分量を保持するために、反応中変化がない ように密封した環境で行なうことが好ましい。また、反応速度の制御の目的で、適宜 攪拌等の物理操作を組み入れることができる。
[0020] 本発明の緑茶抽出液の製造法における酵素処理に際しては、用いる酵素により、 酵素液の pHの調節を行なうことが好ましい。例えば、特定のポリフエノールォキシダ ーゼ、 β—グリコシダーゼ、プロテアーゼ、インベルターゼなどの場合には、酸性条 件下で、酵素反応を行なうのが好ましぐァスコルビン酸やクェン酸、乳酸などの酸性 物質との共存下で反応させると反応中の品質劣化を抑制できることから望ましい。酵 素処理に際しての、反応温度、反応時間、用いる酵素量等のその他の酵素処理条 件については、用いる酵素に応じて、適宜、最適の条件を定めることができる。ただし 、反応温度は、微生物管理上 45°C以上、更には 50°C以上が望ましい。なお、酵素 処理終了後は、加熱、マイクロ波照射など、公知の方法により酵素を失活させた方が
、香味の安定化という点で望ましい。加熱処理の好適例としては 100°Cで 10分から 6 0分間相当があげられる。好ましくは 100°Cで 20分力 40分間相当である。
[0021] 本発明の緑茶抽出液の製造法において、抽出は、通常の緑茶抽出液の製造にお ける抽出操作に従って行われる。すなわち、茶葉の 10倍〜 50倍程度の抽出水を用 いる。その温度は常温から沸騰水まで適宜使用できる。時間も数分力 数時間まで 選択できる。短時間の場合には、ろ過、遠心分離などの固液分離工程を経て、緑茶 飲料の調合液に使用できるし、比較的長時間かけて抽出する場合にはいわゆる緑 茶エキスとして飲料原料として使用するのが普通である。抽出液からそのまま緑茶飲 料を製造する場合には、通常の方法に則り、必要な原材料を混合して、調合液を製 造した後、殺菌を行い、 PET、缶等の容器に充填し、製品化する。一方、緑茶エキス にする場合には、通常の後処理工程を経た後に、そのまま、或いは、濃縮した後に、 殺菌し、缶などの容器に充填し、製品化する。場合によっては、スプレードライ、フリ ーズドライなどの公知の乾燥手段を用いて、乾燥、粉末化することも可能である。
[0022] 酵素溶液と反応した茶葉は通常の方法、すなわち、加熱乾燥、減圧乾燥、真空乾 燥、凍結乾燥などの方法により、水分を 5重量%以下程度まで減らすことにより通常 の製茶した茶葉と同様の取り扱いが可能になる。また、湿潤状態のままレトルトバウチ や金属缶のような密封容器に充填したのちに殺菌することでも同様に扱えるようにな る。これらの場合には、それぞれ酵素処理した複数の酵素処理茶葉を混合したもの を抽出に供することもできるし、また、酵素処理した茶葉を、通常の緑茶葉に適量混 合して使用しても良い。このように、酵素処理茶葉を組み合わせることにより、抽出液 の香味を自由に設計できるのも本発明の優れた点である。
[0023] 本発明においては、酵素溶液との反応が、飛躍的に進行するため、香味成分の変 換に充分な反応が達成でき、しかも、該反応が、抽出操作とは異なる工程で行なわ れるため、緑茶抽出液の抽出は、酵素溶液との反応条件に左右されずに実施でき、 その結果、香味の豊かな緑茶抽出液を得ることができる。
[0024] 以下、実施例により本発明をより具体的に説明するが、本発明の技術的範囲はこ れらの例示に限定されるものではない。 実施例

(実施例 1): 20メッシュパス成分が 80重量%以上になるまでに粉砕した荒茶葉 10g に対し、ポリフエノールォキシダーゼ(ラッカーゼダイワ Y120 ;大和化成) lOOmgとビ タミン C300mgを添加したイオン交換水 20gをよく混合した。その後、水分蒸発防止 のためにラップしたうえで、 35°Cで 12時間放置した。
(比較例 1):酵素を添加しない以外は全く同様に調製したものを 35°Cで 12時間放置 した。

(比較例 2):実施例 1で使用した荒茶葉 10gを粉砕せずに重量比 30倍の 70°Cの熱 水で 6分間抽出した後、濾過して茶葉を取り除き、緑茶抽出液を得た。この抽出液を 、 300mlに調整したのち、ポリフエノールォキシダーゼを 100mg、ビタミン Cを 300m gになるように添カ卩した上で、 35°C、 12時間放置した。
(比較例 3):比較例 2と全く同じように調製したものを 55°Cで、 30分間放置した。 (比較例 4):比較例 2と全く同じように調製したものを 55°Cで、 2時間放置した。
(比較例 5):酵素をカ卩えない以外は、上記比較例と全く同様に調製したものを 35°C1 2時間放置した。
(比較例 6):茶葉を粉碎しない以外は、実施例 1と全く同様にして、酵素溶液と反応さ せた後、緑茶飲料を得た。 [0028] (評価)
比較例 2— 5の各抽出液については、ビタミン C,重曹を用いて茶葉使用率 1%、 pH 6. 5になるような緑茶飲料を調製した後、比較例 6の緑茶飲料と合わせて官能評価 をおこなった。酵素無添カ卩の比較例 5に対して、比較例 2は香味の変化は認められた が、実施例 1に比べると香りの質ははるかに悪ぐ実施例 1で感じられた甘さもほとん ど感じられなかった。反応時間を短くした、比較例 3及び 4でも同様な傾向であった。 これらのことから明らかなように、抽出液に酵素処理しても実施例 1で得られた優れた 香気の抽出液は得られない。また、比較例 6も、香味の変化が少なぐ香りの質も悪 力、つた。香味成分を変換して高香味を得るには、茶葉の粉砕が必須であることが示さ れた。

原料となる茶葉を、 20メッシュパス成分が 80重量%以上になるまでに粉砕した煎茶 に変えた以外は、実施例 1及び比較例 1と全く同様にして、実施例 2と比較例 7の緑 茶飲料を得た。なお、同様に調製した茶葉一酵素溶液混合物は、いずれも分離水分 判定法により好ましい水分範囲と判定された。

評価の結果、実施例 1及び比較例 1と同様な傾向の結果が得られたが、その効果は 実施例 1の方が、実施例 2よりも感じる甘さがより上品であり高評価であった。

(実施例 3、比較例 8):煎茶の粉茶(20メッシュパス成分が 90重量%以上)、酵素を βーグリコシダーゼ(天野ェンザィム)に変えた以外は実施例 1、比較例 1と全く同様 にして実施例 3、比較例 8の緑茶飲料を得た。なお、同様に調製した茶葉一酵素溶 液混合物は、いずれも分離水分判定法により好ましい水分範囲と判定された。
(実施例 4、実施例 5):更に、ァスコルビン酸を添加しない、もしくは等量のクェン酸に 変えて、実施例 4、実施例 5の緑茶飲料を得た。

これらの官能評価をおこなった。比較例 8に比べて、実施例 3は清涼感のあるダリー ン感のある花様の香りを非常に強く感じた。実施例 4はその傾向が少し抑えられ、一 方、実施例 5は香りの強さゆえに感じた合成的、薬品的な匂いが消えてさわやかな香 りになり非常に好ましかった。

(実施例 6、 7、及び比較例 9):反応を 55°C4時間で行うこと以外は、実施例 3、 4およ び比較例 8と同様にして実施例 6、 7および比較例 9を得た。
(評価)
実施例 3、 4、比較例 8と同様の香味傾向が確認できた。ただし、香気の質は実施例 3 , 4および比較例 8よりもさわやかで上品なものであった。

粉砕した煎茶 10g (20メッシュパス成分が 60重量%以上)に対し、表 1に示す各酵素 150mgを添加した、若しくは添加しないイオン交換水 20gをよく混合したあと、水分 蒸発防止のためにラップしたうえで、 35°Cで 12時間放置した。なお、同様に調製し た茶葉一酵素溶液混合物は、いずれも分離水分判定法により好ましい水分範囲と判 定された。これらの処理茶葉を 100°Cのオーブンで 20分間置いて酵素を失活させる と同時に殺菌をおこなって加工茶葉を得た。加工茶葉を 75°Cの熱水 300mlに入れ て時々攪拌しながら 4分間抽出した。これを固液分離した後、得られた抽出液をビタミ ン Cと重曹を用いて調整をおこなレ、 ρΗ6· 5の緑茶飲料 lOOOgを調製した。 7名のパ ネリストにより官能評価をおこなった。

[実施例 14〜: 16、比較例 11]
(実施例 14、 15、 16) : 20メッシュパス成分が 70重量%以上になるまでに粉砕した煎 茶葉 10gに対し、 j3—グ!;コシダーゼ(天野ェンザィム) 50mg、 100mg、 200mgを それぞれ添加したイオン交換水 20gをよく混合した。その後、水分蒸発防止のために ラップしたうえで、 40°Cで 8時間放置した。なお、同様に調製した茶葉一酵素溶液混 合物は、いずれも分離水分判定法により好ましい水分範囲と判定された。これらの処 理茶葉を 100°Cのオーブンで 10分間置いて酵素を失活させると同時に殺菌をおこ なって加工茶葉を得た。これを真空乾燥機中で乾燥して乾燥加工茶葉 (水分含量 3 %以下)を得た。この乾燥加工茶葉を 60°Cの熱水 300mlに入れて時々攪拌しながら 10分間抽出した。これを固液分離した後、得られた抽出液をビタミン Cと重曹を用い て調整をおこない pH6. 5の緑茶飲料 1000gを調製した。更に、これを通常の方法 に従い容器 (ビン)につめてからレトルト殺菌をおこなレ、、容器詰め飲料である実施例 14〜: 16を調製した。
(比較例 11):酵素を添加しなレ、以外は全く同様にして比較例 11を調製した。

上記、実施例 14〜: 16、比較例 11の容器詰め飲料について、官能評価をおこなった 。比較例 11は、香りが平坦で弱力 たのに対し、実施例ではフローラルグリーンの華 やかな香気が感じられた。その強さは酵素の量が増えるにしたがって強くなつた。酵 素量が一番少ない実施例 14でも比較例との香味差は十分に感じられた。加工茶葉 を一旦乾燥させた後に抽出作業をおこない、更に、抽出液を容器詰めにして殺菌し ても本発明の効果が十分に発揮できた。

(実施例 17) : 20メッシュパス成分が 90重量%以上の荒茶葉 10gに対して、ラタター ゼ(ラタターゼ F「ァマノ」 ) 500mgとビタミン C300mgを溶力、した酵素溶 f夜 20gを混 ぜた後に、 60°Cで 18時間放置した。なお、同様に調製した茶葉一酵素溶液混合物 は、いずれも分離水分判定法により好ましい水分範囲と判定された。
反応終了後、 100°Cで 20分間加熱して反応茶葉を得た。この処理茶葉を 80°Cの熱 水 300mlに入れて時々攪拌しながら 10分間抽出した。茶葉を分離したのちに、得ら れた抽出液に対して、ビタミン Cと重曹を用いて、 pH6. 5の実施例 17の緑茶飲料 10 00gを調製した。 (比較例 12):酵素を添加しなレ、以外は全く同様にして比較例 12を調製した。

実施例 17は、比較例 12に比べて、甘い香りが目立ち、華やかでフルーティな香気を 感じて高評価だった。

(実施例 18):酵素をインベルターゼ (スミチーム INV) 250mgに変えて、反応温度を 50°Cに変えた以外は全く同様にして、実施例 18の緑茶抽出液を得た。

実施例 18は、比較例 13と比べて、シャープで切れのある花様の香りが目立った。 マスカットのような青ぐかつナチュラルな香気が感じられて好評価だった。

(実施例 19、 20、 21、 22) : 20メッシュパス成分が 80重量%以上の煎茶 10gに対し て、各種プロテアーゼをカ卩えた酵素溶液 20gをカ卩えてよく混合し、 50°C15時間反応 させた。なお、同様に調製した茶葉一酵素溶液混合物は、いずれも分離水分判定法 により好ましい水分範囲と判定された。プロテアーゼとしてプロテアーゼ M「ァマノ」 G150mg (実施例 19)を使用する場合にはビタミン C300mgを同時に溶解した。プロ テアーゼ N「ァマノ」 G150mg (実施例 20)、サモアーゼ Y320 (大和化成) 150m g (実施例 21)及びパンチダーゼ NP— 2 (ヤクルト薬品工業) 300mg (実施例 22)使 用の際にはビタミン Cは添加しなかった。反応終了後、ビタミン Cを使用していない酵 素溶液を使用したものにっレ、ては、ビタミン Cをそれぞれ粉末の形態で 300mg添カロ して混合した。そして、 100°C、 20分間加熱処理を行なった。
(比較例 14、 15、 16、 17):粉砕していない茶葉を使用する以外は、実施例 19— 22 と全く同様にして、比較例 14—17を調製した。
(比較例 18):酵素を添加しない以外は、全く同様にして比較例 18を調製した。
[0043] これら処理茶葉を 80°Cの熱水 300mlに入れて時々攪拌しながら 10分間抽出した 。茶葉を分離したのちに、得られた抽出液に対して、ビタミン Cと重曹を用いて、 pH6 . 5の実施例 19— 22及び比較例 14— 18の緑茶飲料 lOOOgを調製した。 (比較例 19):実施例 19— 22で使用した煎茶 lOgを未粉砕のまま、 80°Cの熱水 300 mlに入れて、時々攪拌しながら 10分間抽出した。茶葉を取り除いて、再度 300mlに 調整した後、プロテアーゼ M「ァマノ」 G150mg、ビタミン C300mgを添カロして、 50°C 15時間放置した。反応終了後、ビタミン Cと重曹を用いて pH6. 5の緑茶飲料 lOOOg を調製した。

粉砕した茶葉を用レ、た実施例 19 _ 22は、未粉砕煎茶を用レ、た比較例 14 _ 17に比 ベて、はるかに旨味とコク味が強かった。比較例 14— 17は、実施例 19— 22に比べ て劣化香味が目立ち、また、茶葉の褐色化も進んで飲料にした場合の液色の点でも 問題があった。酵素をカ卩えていない比較例 18は、旨味、コク味が感じられなかった。 抽出液に酵素を加えた比較例 19は、旨味、コク味がほとんど感じられず、また、反応 中に風味劣化を起こすとレ、う問題があった。
[0045] [実施例 23— 32、比較例 20— 24]
プロテアーゼ(プロテアーゼ M「ァマノ」 GlOOmgもしくはインベルターゼ (スミチ一 ム INV200mg)とビタミン C300mgを含み、液量を変化させた酵素溶液を調製した。 これらを 20メッシュパス成分が 90重量%以上の荒茶葉 (水分量 5重量%) 10gに対し てそれぞれよく混合して 50°C15時間反応させた。なお、同様に調製した茶葉一酵素 溶液混合物は、いずれも分離水分判定法により好ましい水分範囲と判定された。反 応終了後、 100°C20分間加熱処理した。これら処理茶葉を 80°Cの熱水 300mlに入 れて時々攪拌しながら 10分間抽出した。茶葉を分離したのちに、得られた抽出液に 対して、ビタミン Cと重曹を用いて、 pH6. 5の緑茶飲料 1000gを調製した。また酵素 を添加しないで溶液量を 15gにした以外は、全く同様にして比較例を調製した。

プロテアーゼについての評価結果を表 2に、インベルターゼについての評価結果を 表 3に示した。プロテアーゼはコク味の強度を指標にして、それぞれの反応の進み具 合を評価した。表 2中、コク味評価は、次の指標で示す:—;感じなレ、、土殆ど感じな レ、、 +感じる、 + +強く感じる、 + + +非常に強く感じる。また、インベルターゼは花 様の香気の強さを指標にして、それぞれの反応の進み具合を評価した。表 3中、花 様の香気の強さは、次の指標で示す:花様香気評価 一;感じない、土殆ど感じなレ' 、 +感じる、 + +強く感じる、 + + +非常に強く感じる。

200mg、ビタミン C600mgを水に溶力 た酵素溶液 40gを加えてよく混合して、 50 。Cで 15時間反応させた。なお、同様に調製した茶葉一酵素溶液混合物は、いずれ も分離水分判定法により好ましい水分範囲と判定された。反応後の茶葉をカウパック レトルトパゥチ NACF— 101に充填して空気を抜いて密封し、 121。C20分間のレトル ト殺菌をおこなった。殺菌後の茶葉を全量用いて、 80°Cの熱水 600gで 5分間抽出し たのち、茶葉を除去してから冷却し、茶葉使用率 1 %となるように希釈し、ビタミン Cと 重曹で pH6. 5に調整した。更に、ビン容器に充填して、レトルト殺菌をおこない実施 例 33の緑茶飲料を調製した。酵素を —グリコシダーゼ 300mgに変えた以外は全く 同様にして、また酵素を全く使用しない以外は全く同様にして、それぞれ実施例 34、 比較例 25の飲料を調製した。

プロテアーゼ処理した実施例 33は比較例 25にはない強い旨みとコク味を感じ、 β一 グリコシダーゼ処理した実施例 34は華やかな香りを感じた。処理茶葉を湿潤状態の ままレトルト殺菌しても効果は保持されてレ、た。

各種酵素を用いて、茶葉に対する酵素溶液量 (倍率)と「分離水分判定法」及び香気 との関係について試験した。分離水分判定試験として、煎茶粉砕茶葉 (20メッシュパ ス 80重量%以上:以降 Α)と荒茶粉碎茶葉(20メッシュパス 90重量%以上:以降 Β) 1 Ogに対して、水分量、酵素種類を変えて 50°C15時間反応を行なった。同様に調製 した茶葉一酵素溶液混合物の分離水分判定結果を表 4に示す。表中、分離水分判 定結果、〇のものは本発明の実施例を、 Xのものは比較例を示す。
[0052] 反応終了後、 100°Cで 20分間加熱処理した。これら処理茶葉を 80°Cの熱水 300 mlに入れて時々攪拌しながら 10分間抽出した。茶葉を分離したのちに、得られた抽 出液に対して、ビタミン Cと重曹を用いて、 pH6. 5の緑茶飲料 lOOOgを調製した。官 能評価により、それぞれの酵素反応の大きさを評価した。結果を、表 4に示す。

粉砕したほうじ茶 10g (20メッシュパス成分が 60重量%以上)に対して,プロテアーゼ (プロテアーゼ M「ァマノ」 G) 100mgとビタミン C300mgを溶かした酵素溶液 20gを 混ぜた後に、 50°Cで 6時間反応させた。なお、同様に反応させた茶葉-酵素溶液混 合物はいずれも分離水分判定法により好ましい水分範囲と判定された。反応終了後 、 100°Cで 20分間加熱して反応茶葉を得た。この反応茶葉を 75°Cの温水 400mlに 入れて時々攪拌しながら 6分間抽出した。茶葉を分離した後に、得られた抽出液に 対してビタミン Cと重曹を用いて、 pH6. 5の実施例 50の緑茶飲料 1000gを調製した 酵素を添加しない以外は全く同様にして比較例 29を調製した。

実施例 50は、比較例 29よりも味の厚みに幅がでて、旨みの強いほうじ茶になり好ま しかった。

Claims

請求の範囲
[1] 乾燥緑茶葉を粉砕した或いは緑茶葉製造工程で粉砕された、粉砕緑茶葉と、酵素 溶液を 1: 0. 2〜: 1: 5の重量比で混合して反応を行なったのち、水もしくは温水で抽 出することを特徴とする香味の優れた緑茶抽出液の製造法。
[2] 粉砕緑茶葉と、酵素溶液の重量比が、 1: 0. 2〜: 1: 3の重量比であることを特徴とす る請求項 1記載の香味の優れた緑茶抽出液の製造法。
[3] 酵素溶液の量が、茶葉と混合した際に、当該混合物から水分が分離してこない範囲 であることを特徴とする請求項 1又は 2記載の香味の優れた緑茶抽出液の製造法。
[4] 乾燥緑茶葉の粉碎度が、 20メッシュパス成分が 50重量%以上の粉砕度であることを 特徴とする請求項 1〜3のいずれか記載の香味の優れた緑茶抽出液の製造法。
[5] 粉砕緑茶葉と、酵素溶液を 1: 0. 2〜: 1: 5の重量比で混合して反応を行なったのち、 加熱処理を行うことを特徴とする請求項 1〜4のいずれか記載の香味の優れた緑茶 抽出液の製造法。
[6] 粉砕緑茶葉と、酵素溶液との反応に使用する酵素が、ポリフエノールォキシダーゼ、 βーグリコシダーゼ、リパーゼ、クロロゲン酸エステラーゼ、ヌクレアーゼ、プロテア一 ゼ、ラタターゼ、インべノレターゼ、ぺクチナーゼ、キシラナーゼ及びデァミナーゼから 選択される 1又は 2以上の酵素であることを特徴とする請求項 1〜5のいずれか記載 の香味の優れた緑茶抽出液の製造法。
[7] 乾燥緑茶葉を粉砕した或いは緑茶葉製造工程で粉砕された、粉砕緑茶葉と、酵素 溶液を 1: 0. 2〜: 1: 5の重量比で混合して反応を行なったのち、一旦乾燥するか、若 しくは湿潤状態のまま密封容器に充填した後殺菌することによって調製した、高香味 緑茶抽出液調製用の緑茶茶葉。
[8] 請求項 7記載の緑茶茶葉を、水若しくは温水で抽出することを特徴とする香味の優れ た緑茶抽出液の製造法。
[9] 請求項:!〜 6のいずれか記載の緑茶抽出液の製造法又は請求項 8記載の緑茶抽出 液の製造法によって製造された抽出液を、そのまま若しくは他の飲料原料と混合した 後、殺菌、充填したことを特徴とする容器詰緑茶飲料。
