Abstract

L'invention concerne de nouveaux azahétérocycles annelés possédant la formule générale 1 et comportant un fragment de pyrimidine sous forme de bases libres ou de sels pharmaceutiquement acceptables ainsi que des compositions pharmaceutiques les comprenant, de même que leur utilisation dans des cocktails thérapeutiques pour traiter des personnes et des animaux à sang chaud souffrant de maladies liées à l'activité des phosphoinositolkinases (PI3K) de classe I (et de leurs isoformes mutantes), notamment de maladies oncologiques. L'invention concerne aussi l'utilisation de ces composés en tant qu'instruments pharmacologiques destinés aux recherches in vitro et in vivo des mécanismes d'action biochimique des PI3K kinases, notamment de celles de classe I et de leurs isoformes mutantes. Dans la formule, X représente un atome d'oxygène ou un atome de soufre ou un groupe avec un atome de carbone du groupe CH éventuellement substitué, Z représente un atome d'oxygène, R(1) représente un atome d'hydrogène ou un alkyle C(1)-C(6) éventuellement substitué, ou Z représenteun atome d'azote qui, avec l'atome de carbone auquel il est lié, referme via Z et R(1) le noyau d'imidazoline éventuellement substitué; R(2)et R(3) représentent indépendamment l'hydrogène, un alkyle éventuellement substitué, un aryle éventuellement substitué ou un hétérocyclyle éventuellement substitué, si Y=N; ou R(2)et R(3) représentent indépendamment un alkyle éventuellement substitué, un aryle éventuellement substitué ou ou un hétérocyclyle éventuellement substitué; si Y est groupe avec un atome de carbone du groupe CH éventuellement substitué et Y=O,S, ou R(2) représente l'hydrogène et R(3) représente l'aminoalkyle éventuellement substitué ou l'azahétérocyclyle éventuellement substitué, si Y est groupe avec un atome de carbone du groupe CH éventuellement substitué et X=O,S, ou R(2) représente l'hydrogène, et R(3) représente un aryle éventuellement substitué,si R(1) est l'aminoalkyle substitué, l'hydroxyalkyle substitué ou l'azahétérocyclyle éventuellement substitué, Y est un groupe avec un atome de carbone du groupe CH éventuellement substitué, et X=O,S; et R(4) est l'hydrogène ou l'alkyle inférieur.
