Le procédé comprend les étapes 1) à 4) suivantes : 1) la mise à disposition d'un support auquel est reliée une protéine de liaison à la biotine, et d'une protéine biotinylée comprenant de la biotine et une protéine capable de se lier spécifiquement à une substance à détecter et liée à la biotine ; 2) la liaison de la protéine biotinylée au support mis à disposition à l'étape 1) pour produire un support auquel la protéine biotinylée est liée ; 3) l'ajout du composant (a) représenté ci-dessous et du composant (b-i) ou (b-ii) représenté ci-dessous au support auquel est liée la protéine biotinylée préparé à l'étape 2) et le mélange des composants et du support ensemble ; et 4) la détection de la substance qui a été liée spécifiquement à la protéine biotinylée : (a) un échantillon biologique ; (b-i) à la fois un extrait de désintégration cellulaire qui est préparé à partir d'une cellule homogène à une cellule hôte utilisée pour l'expression de la protéine de liaison à la biotine et ainsi de suite à l'étape 1) et une protéine de liaison à la biotine ; et (b-ii) un extrait de désintégration cellulaire préparé à partir d'une cellule qui est homogène à une cellule hôte utilisée pour l'expression de la protéine de liaison à la biotine et ainsi de suite à l'étape 1) et qui a été modifiée par génie génétique pour exprimer la protéine de liaison à la biotine.
