Abstract
Description
(1)甜茶エキスに無機塩を添加したものを電気透析し、濃縮液として得られる甜茶エッセンスと、ビフィドバクテリウム属細菌を含有することを特徴とする発酵食品。
(2)ビフィドバクテリウム属細菌を含有する発酵食品の製造において、甜茶エキスに無機塩を添加したものを電気透析し、濃縮液として得られる甜茶エッセンスを任意の段階で配合することを特徴とするビフィドバクテリウム属細菌を含有する発酵食品の製造方法。
(3)ビフィドバクテリウム属細菌を含有する発酵食品に、甜茶エキスに無機塩を添加したものを電気透析し、濃縮液として得られる甜茶エッセンスを配合することを特徴とするビフィドバクテリウム属細菌の生残性改善方法。
(4)甜茶エキスに無機塩を添加したものを電気透析し、濃縮液として得られることを特徴とする甜茶エッセンス。
(5)甜茶エキスに無機塩を添加したものを電気透析し、濃縮液を得ることを特徴とする甜茶エッセンスの製造方法。
(6)甜茶エキスに無機塩を添加したものを電気透析し、濃縮液を得ることを特徴とする甜茶エキスの風味改善方法。
甜茶エキスの製造:
甜茶葉に粉砕等の処理を施した後、甜茶葉の15倍量の水と甜茶葉の5%相当量のクエン酸を添加してpHを3.8に調整し、20℃で60分間の抽出を行った。更に、得られた抽出液をエバポレーターで5倍濃縮し、Brix.13の甜茶エキスを得た。
甜茶エッセンスの製造(1):
甜茶葉に粉砕等の処理を施した後、甜茶葉の15倍量の水と甜茶葉の5%相当量のクエン酸を添加してpHを3.8に調整し、20℃で60分間の抽出を行った。この抽出液に、塩化マグネシウムの濃度が0.05mol/Lとなるように塩化マグネシウム六水和物を添加した。次に、これを電気透析装置(電気透析膜:AC220-50、製品名:マイクロアシライザーS-3、株式会社アストム社製)の脱塩室に入れ、濃縮室に抽出液の17%に相当する水を入れて、脱塩室の電気伝導度が平衡(2ミリジーメンス毎センチメートル(mS/cm))となるまで電気透析処理し、濃縮液を回収した。更に、この濃縮液をエバポレーターで5倍に濃縮し、Brix.12の甜茶エッセンス1を得た。
乳製品の製造(1):
20%脱脂粉乳培地を基本培地とし、これに実施例1で調製した甜茶エッセンス1を、0.2%添加し、120℃で3秒間加熱殺菌して培養培地を調製した。これらの培地にビフィドバクテリウム・ブレーベ YIT12272のスターターを1%、ラクトコッカス・ラクチス YIT2027のスターターを0.1%およびストレプトコッカス・サーモフィルス YIT2021のスターターを0.01%接種して37℃で24時間培養を行い、培養物を得た。この培養物を15MPaで均質化したもの40質量部に、10%砂糖溶液を100℃で5分間殺菌した物を60質量部添加し、更にヨーグルト香料(株式会社ヤクルトマテリアル製)を0.1%添加して乳製品を製造した。また、比較のため、基本培地に、甜茶エッセンス1に代えて甜茶エキスを0.2%添加した培地で上記と同様に製造した乳製品も得た。乳製品中の甜茶エッセンス1または甜茶エキスの濃度は0.08%である。
生残率(%)=保存後の生菌数÷製造時の生菌数×100

5 : 苦みを全く感じない
4 : 苦みをほとんど感じない
3 : 苦みをやや感じる
2 : 苦みを感じる
1 : 苦みを強く感じる
甜茶エッセンスの製造(2):
実施例1において、塩化マグネシウム六水和物に代えて塩化ナトリウム、塩化カリウム、塩化カルシウム二水和物またはクエン酸三カリウム一水和物をその塩の無水物の濃度が0.05mol/Lとなるように使用する以外は同様にして甜茶エッセンス2~5を製造した。
乳製品の製造(2):
実施例2において、甜茶エッセンス1に代えて実施例3で製造した甜茶エッセンス2~5を同量使用する以外は同様にして乳製品を得た。これらの乳製品について、実施例2と同様にして製造時および10℃で21日間保存後における乳製品中の生菌数を測定し、生残率を求め、風味を評価した。これらの結果を表2に示した。
甜茶エッセンスの製造(3):
実施例1において、塩化マグネシウム六水和物を、塩化マグネシウムの濃度が0.05mol/Lとなるように使用したのに代えて、塩化マグネシウム六水和物を塩化マグネシウムの濃度が0.01、0.02、0.1、0.2または0.5mol/Lとなるように使用する以外は同様にして甜茶エッセンス6~10を製造した。
乳製品の製造(3):
実施例2において、甜茶エッセンス1に代えて実施例5で製造した甜茶エッセンス6~10を同量使用する以外は同様にして乳製品を得た。これらの乳製品について、実施例2と同様にして製造時および10℃で21日間保存後における乳製品中の生菌数を測定し、生残率を求め、風味を評価した。これらの結果を表3に示した。
乳製品の製造(4):
20%脱脂粉乳培地に実施例1で調製した甜茶エッセンス1を、0.01、0.02、0.05、0.1、0.2または0.5%添加し、120℃で3秒間加熱殺菌して培養培地を調製した。これらの培地にそれぞれビフィドバクテリウム・ブレーベ YIT12272のスターターを1%およびラクトコッカス・ラクチス YIT2027のスターターを0.01%接種して37℃で24時間培養を行い、培養物Aを得た。また、16%脱脂粉乳培地にストレプトコッカス・サーモフィルス YIT2021のスターターを0.5%接種して37℃で24時間培養を行い、培養物Bを得た。一方、還元麦芽糖水あめ8%、ポリデキストロース4%、ガラクトオリゴ糖液糖4%、ペクチン0.5%、コラーゲンペプチド0.2%、ビタミンB製剤0.05%、スクラロース0.007%、乳化鉄(ピロリン酸鉄)0.6%を水に溶解した後、121℃で3秒間殺菌してシロップを得た。
乳製品の製造(5):
実施例7において、ビフィドバクテリウム・ブレーベ YIT12272に代えてビフィドバクテリウム・ブレーベ YIT4065を使用する以外は、同様にして乳製品を製造した。これらの乳製品は実施例7で得られた乳製品と同様に、甜茶エッセンスを添加することにより、乳製品の風味にはほとんど影響を及ぼすことなく、ビフィドバクテリウム属細菌の生残性改善効果が認められた。
乳製品の製造(6):
実施例7において、ビフィドバクテリウム・ブレーベ YIT12272に代えてビフィドバクテリウム・ブレーベ YIT10001を使用する以外は、同様にして乳製品を製造した。これらの乳製品は実施例7で得られた乳製品と同様に、甜茶エッセンスを添加することにより、乳製品の風味にはほとんど影響を及ぼすことなく、ビフィドバクテリウム属細菌の生残性改善効果が認められた。
乳製品の製造(7):
実施例7において、ビフィドバクテリウム・ブレーベ YIT12272に代えてビフィドバクテリウム・ビフィダム YIT4007を使用する以外は、同様にして乳製品を製造した。これらの乳製品は実施例7で得られた乳製品と同様に、甜茶エッセンスを添加することにより、乳製品の風味にはほとんど影響を及ぼすことなく、ビフィドバクテリウム属細菌の生残性改善効果が認められた。
乳製品の製造(8):
実施例7において、ビフィドバクテリウム・ブレーベ YIT12272に代えてビフィドバクテリウム・ビフィダム YIT10347を使用する以外は、同様にして乳製品を製造した。これらの乳製品は実施例7で得られた乳製品と同様に、甜茶エッセンスを添加することにより、乳製品の風味にはほとんど影響を及ぼすことなく、ビフィドバクテリウム属細菌の生残性改善効果が認められた。
Claims (9)
- 甜茶エキスに無機塩を添加したものを電気透析し、濃縮液として得られる甜茶エッセンスと、ビフィドバクテリウム属細菌を含有することを特徴とする発酵食品。
- 無機塩が、カリウム塩、ナトリウム塩、カルシウム塩およびマグネシウム塩から選ばれる1種または2種以上である請求項1記載の発酵食品。
- 無機塩が、マグネシウム塩である請求項1記載の発酵食品。
- 無機塩の添加量が、0.02~0.2mol/Lである請求項1~3のいずれかに記載の発酵食品。
- ビフィドバクテリウム属細菌を含有する発酵食品の製造において、甜茶エキスに無機塩を添加したものを電気透析し、濃縮液として得られる甜茶エッセンスを任意の段階で配合することを特徴とするビフィドバクテリウム属細菌を含有する発酵食品の製造方法。
- ビフィドバクテリウム属細菌を含有する発酵食品に、甜茶エキスに無機塩を添加したものを電気透析し、濃縮液として得られる甜茶エッセンスを配合することを特徴とするビフィドバクテリウム属細菌の生残性改善方法。
- 甜茶エキスに無機塩を添加したものを電気透析し、濃縮液として得られることを特徴とする甜茶エッセンス。
- 甜茶エキスに無機塩を添加したものを電気透析し、濃縮液を得ることを特徴とする甜茶エッセンスの製造方法。
- 甜茶エキスに無機塩を添加したものを電気透析し、濃縮液を得ることを特徴とする甜茶エキスの風味改善方法。
