Description
Verfahren zur Reduktion eines Secodion-Derivats
mit einer Alkohol-Dehydrogenase
Die Erfindung betrifft ein Verfahren zur Reduktion eines Secodion-Derivats der allgemeinen Formel 1 mit einem Enzym aus der Klasse der Alkohol-Dehydrogenasen (ADHs).

Die Secodion-Derivate der allgemeinen Formel 1 werden auch als Secosteroide oder 8,14- Seco-gonatetraen-14,17-dione bezeichnet Mit R = Ethyl und R' = Methyl bezeichnet man die Substanz als Ethylsecodion (Ethyl-3-methoxy-8,14-seco-gona-1 ,3, 5(10), 9(11 )-tetraen-14,17- dion) und mit R = Methyl und R' = Methyl als Methylsecodion (Methyl-3-methoxy-8,14-seco- gona-1 ,3,5(10),9(11 )-tetraen-14, 17-dion).
Die Secodione sind Schlüsselsubstanzen in der Synthese von enantiomerenreinen Steroiden aus der Klasse der Estrogene und Gestagene. Beispiele sind Levonorgestrel, Estradiol, 3-Keto-desogestrel und Deso estrel.

Der Schlüsselschritt zur Herstellung der enantiomerenreinen Steroide aus den Secodionen der allgemeinen Formel 1 ist die selektive Reduktion nur einer Ketongruppe in den entsprechenden Alkohol in stereoselektiver Weise. Bei der stereoselektiven einfachen Reduktion eines Secodions der allgemeinen Formel 1 zu einem Seconol sind theoretisch 4 unterschiedliche räumliche Konfigurationen möglich, je nachdem ob die Hydroxylgruppe an der Position 14 oder 17 ebildet wird und ob die - oder ß-Konfiguration entsteht.

Die Alkohol-Dehydrogenase-katalysierte Reduktion von Secodionen der allgemeinen Formel 1 kann neben den einfach reduzierten Seconolen der allgemeinen Formeln 2-5 auch doppelt reduzierte Secodiole der allgemeinen Formel 6 bilden, die für die weitere Umwandlung unerwünscht sind.

Von wirtschaftlich besonderem Interesse ist das 17-ß-Seconol der allgemeinen Formel 2, aus welchem durch weitere chemische Umwandlungen die oben genannten Steroide hergestellt werden können.
Die Synthese von optisch aktivem Seconol wird im industriellen Maßstab durch mikrobielle Ganzzelltransformation mit Wildtyp-Organismen der Gattung Saccharomyces oder Pichia durchgeführt (H. Heidepriem et al. Justus Liebigs Annalen der Chemie (1968), 712, 155-67;
H. Gibian et al. Tetrahedron Letters (1966), 7, 2321-30; V. Bihari et al. Current Science (1984), 53, 124-6).
In DE 2120676 wird die fermentative Reduktion von ausgewählten Seco-Steroiden unter Verwendung der spezifischen Mikroorganismen Pichia farinosa NRRL Y-1 18 oder Pichia farinosa CBS 185 beschrieben.
US 3,549,499 beschreibt die Synthese von optisch aktiven Seco-Steroiden durch mikrobiologische Reduktion. Diese Verbindungen können zur Herstellung von physiologisch aktiven Steroiden verwendet werden.
US 3,481 ,974 betrifft einen mikrobiologischen Prozess zur stereospezifisch selektiven Reduktion einer der Carbonyl-Gruppen eines 13-Alkyl-3-alkoxy-8,14-secogona-
I , 3,5(10),9(1 1 )-tetraen-14,17-dions zur Herstellung des 17ß-Hydroxy-lsomers. Geeignete Mikroorganismen sind Mitglieder der Gattungen Humicola, Rhodotorula und Schizosaccharomyces.
In WO 2008/068030 wird ein Verfahren beschrieben, in dem die Transformation mit rekombinanten Enzymen aus der Klasse der Alkohol-Dehydrogenasen durchgeführt wird. Eine genaue Angabe über die Produktreinheit oder den Stereoisomerenüberschuss wird in WO 2008/068030 nicht gemacht.
Der Vorteil von rekombinanten Enzymen gegenüber Wildtyp-Zellen ist, dass die Reaktionsbedingungen nicht an ein Überleben der Zellen geknüpft sind. So können z.B. höhere Substratkonzentrationen benutzt werden. Zudem entfällt bei der Verwendung von freien Enzymen die Notwendigkeit der Diffusion, bzw. des Transports des Substrats in die Zelle. Ein weiterer Vorteil ist die einfachere Produktion des Biokatalysators in gleichbleibender Qualität. Bei der Verwendung von Mikroorganismen sowie mit den Enzymen aus WO2008068030 entsteht jedoch ein hoher Anteil an Nebenprodukten, insbesondere von doppelt reduziertem Secodiol und daraus abgeleiteten Folgeprodukten. Durch die Bildung dieser Nebenprodukte wird die Ausbeute an einfach reduziertem Seconol gesenkt und die Aufarbeitung des Reaktionsgemisches wird erschwert. Der Prozess wird also insgesamt weniger wirtschaftlich.
Ferner sind das Substrat und das Reaktionsprodukt in Wasser nur schwer löslich. Für eine effektive enzymatische Katalyse muss das Substrat im wässrigen System verfügbar gemacht werden. Dies wird in WO2008068030 durch die Verwendung von hohen Konzentrationen (40 - 67%) sekundärer Alkohole wie Isopropanol oder 4-Methyl-2-pentanol gelöst. Diese haben jedoch den Nachteil, dass viele Enzyme bei diesen Konzentrationen irreversibel inaktiviert werden oder der Alkohol nicht von dem ADH-Enzym akzeptiert wird.
Der Erfindung liegt die Aufgabe zugrunde, die aus dem Stand der Technik bekannten enzymatischen Verfahren zur Reduktion von Secodion-Derivaten zu verbessern.
Diese Aufgabe wird durch den Gegenstand der Patentansprüche gelöst.
Es wurde überraschend ein Verfahren gefunden, mit dem das 17-ß-Seconol der allgemeinen Formel 2 mit hoher Stereoselektivität und in hoher Reinheit erhalten werden kann.
Es wurden überraschend ADHs identifiziert, die eine besonders hohe Stereoselektivität bzgl. des 17-ß-Seconols der allgemeinen Formel 2 besitzen und dieses in hoher Produktreinheit bei verschiedenen Reaktionsbedingungen liefern.
Es wurde ferner überraschend gefunden, dass durch die Verwendung milder Tenside wie beispielsweise Triton-X-100 in Kombination mit geringen Konzentrationen an Cosolventien wie Isopropanol oder Dimethylsulfoxid (DMSO) ein Reaktionssystem erhalten wird, in dem das Substrat homogen verteilt und effektiv enzymatisch umgesetzt werden kann. Überraschenderweise bleibt unter diesen Reaktionsbedingungen die Aktivität vieler Enzyme erhalten.

Produktreinheit
Die Produktreinheit wird mittels HPLC mit einer reversed phase C18 Säule (Gemini 5 μ, 150 x 4.6 mm, Phenomenex) analysiert. Es wird isokratisch eluiert mit einem Gemisch aus Wasser und 65% Acetonitril. Die Detektion erfolgt mittels einer UV-Lampe bei 265 nm. Die Produktzuordnung erfolgt soweit verfügbar über Referenzverbindungen und wird mittels HPLC-MS bestätigt. Es können 3 Produktpeaks identifiziert werden: Seconol (alle Stereoisomere) bei 4,32 min; Secodiol bei 3,34 min; und Peak 3 bei 4,64 min. Für Peak 3 kann die gleiche molare Masse wie für überreduziertes Secodiol gemessen werden. Peak 3
kann somit durch eine Folgereaktion von Secodiol erklärt werden. Die Produktreinheit von einfach reduziertem Seconol wird demnach wie folgt berechnet:

Der Anteil des gewünschten 17-ß-Seconol 2 im Produkt wird durch Auswertung eines HPLC Chromatograms an einer chiralen HPLC-Phase ermittelt. Es wird eine Chiralpak AY-H (Chiral Technologies Europe) benutzt mit dem Eluenten n-Heptan/lsopropanol/Diethylamin = 90:10:0,1. Die Peaks der 4 einfach reduzierten Steroisomeren können mit dieser Analytik klar getrennt werden und eluieren in folgender Reihenfolge 17a-, 14ß-, 17ß- und 14a- Seconol. Der Stereoisomerenüberschuss wird aus den jeweiligen Peakflächen nach folgender Formel berechnet:

Der Umsatz von Secodion zum einfach reduzierten Seconol wird demnach wie folgt berechnet:

Alkohol-Dehydrogenasen gehören zur Enzymfamilie der Oxidoreduktasen. Alkohol- Dehydrogenasen zeichnen sich dadurch aus, dass sie sowohl die Oxidation von Alkoholen zu den entsprechenden Carbonylverbindungen als auch die korrespondierende Rückreaktion katalysieren.
Tensid
Tenside sind amphiphile Moleküle, die in wässrigen Systemen schwer lösliche Substanzen in homogener Emulsion bzw. Suspension halten können.
Unter dem Begriff "NADP" wird Nicotinamid-adenin-dinucleotid-phosphat und unter dem Begriff "NADPH" reduziertes Nicotinamid-adenin-dinucleotid-phosphat verstanden. Der Begriff "NAD" steht für Nicotinamid-adenin-dinucleotid und der Begriff "NADH" für reduziertes Nicotinamid-adenin-dinucleotid.
Die Erfindung betrifft ein Verfahren zur enzymatischen Reduktion eines Secodion-Derivats der allgemeinen Formel 1
(1 ) mit einer Alkohol-Dehydrogen at der allgemeinen Formel 2

R für -H oder eine -(CVC4)-Alkylgruppe steht;
R' ausgewählt ist aus -H, -(C C6)-Alkyl, -(C1-C4)-Alkenyl, -C(=0)R",
-CRdRe-Aryl, -Tetrahydropyranyl und -(CH2)i-2-X-Rf; worin
R" für -Phenyl; unsubstituiertes, ein oder mehrfach durch Halogen substituiertes -(C C6)-Alkyl oder für -O-Allyl steht;
Ra, Rb und Rc jeweils unabhängig voneinander für -(CrC6)-Alkyl oder Phenyl stehen;
Rd und Re jeweils unabhängig voneinander für -H, für unsubstituiertes oder durch -OMe einfach substituiertes -Phenyl stehen;
R' für -(C1-C4)-Alkyl, Phenyl oder -(CH2)i-2SiRaRbRc steht;
X für O oder S steht;
Aryl für unsubstituiertes oder durch -OMe einfach oder mehrfach substituiertes Phenyl steht; und wobei das 17-ß-Seconol-Derivat der allgemeinen Formel 2 bevorzugt mit einer Produktreinheit von > 90% und einem Stereoisomeren-Anteil von > 90% erhalten wird.
Vorzugsweise wird das 17-ß-Seconol-Derivat der allgemeinen Formel 2 mit einem Stereoisomerenüberschuss von >95%, besonders bevorzugt >98%, und am bevorzugtesten >99% erhalten.
Vorzugsweise wird das 17-ß-Seconol-Derivat der allgemeinen Formel 2 mit einer Produktreinheit von >90%, vorzugsweise -.95%, besonders bevorzugt ä98%, und am bevorzugtesten >99% erhalten.
Vorzugsweise steht R in den allgemeinen Formeln 1 und 2 jeweils für Methyl oder Ethyl.
Vorzugsweise steht R' in den allgemeinen Formeln 1 und 2 jeweils für Methyl.
In einer bevorzugten Ausführungsform stehen R und R' in den allgemeinen Formeln 1 und 2 jeweils für Methyl, d.h. die Verbindung der allgemeinen Formel 1 ist Methylsecodion (Methyl- 3-methoxy-8,14-seco-gona-1 ,3,5(10), 9(1 1 )-tetraen-14,17-dion).
In einer anderen bevorzugten Ausführungsform steht R in den allgemeinen Formeln 1 und 2 jeweils für Ethyl und R' jeweils für Methyl, d.h. die Verbindung der allgemeinen Formel 1 ist Ethylsecodion (Ethyl-3-methoxy-8,14-seco-gona-1 ,3,5(10),9(1 1 )-tetraen-14,17-dion).
Erfindungsgemäß wird das Secodion-Derivats der allgemeinen Formel 1 mit einer Alkohol- Dehydrogenase reduziert.
Vorzugsweise umfasst die Alkohol-Dehydrogenase eine Aminosäuresequenz mit mindestens 60% oder mindestens 65%, bevorzugt mindestens 70% oder mindestens 75%, besonders bevorzugt mindestens 80% oder mindestens 85%, noch bevorzugter mindestens 90% oder mindestens 92%, ganz besonders bevorzugt mindestens 93% oder mindestens 94%, am bevorzugtesten mindestens 95%, mindestens 96%, mindestens 97% oder mindestens 98%, und insbesondere mindestens 99%, Homologie zu wenigstens einer der Aminosäuresequenzen ausgewählt aus der Gruppe bestehend aus (Seq ID No: 2), (Seq ID No: 4), (Seq ID No: 6), (Seq ID No: 8) und (Seq ID No: 10). Diese Aminosäuresequenzen werden zum Beispiel durch die DNA-Sequenzen (Seq ID No: 1 ), (Seq ID No: 3), (Seq ID No: 5), (Seq ID No: 7) bzw. (Seq ID No: 9) codiert.
Im Sinne der Erfindung wird die Homologie einer Sequenz bevorzugt berechnet als Identität mittels BLASTP 2.2.20+ (Stephen F. Altschul, Thomas L. Madden, Alejandro A. Schäffer, Jinghui Zhang, Zheng Zhang, Webb Miller, and David J. Lipman (1997)), "Gapped BLAST and PSI-BLAST: a new generation of protein database search programs", Nucleic Acids Res. 25:3389-3402; Stephen F. Altschul, John C. Wootton, E. Michael Gertz, Richa Agarwala, Aleksandr Morgulis, Alejandro A. Schäffer, and Yi-Kuo Yu (2005) ".
In einer bevorzugten Ausführungsform weist die Alkohol-Dehydrogenase zu wenigstens einer der Aminosäuresequenzen ausgewählt aus der Gruppe bestehend aus Seq ID No 2, Seq ID No 4, Seq ID No 6, Seq ID No 8 und Seq ID No 10 eine Homologie von mindestens 60% oder mindestens 65%, bevorzugt mindestens 70% oder mindestens 75%, besonders bevorzugt mindestens 80% oder mindestens 85%, noch bevorzugter mindestens 90% oder mindestens 92%, ganz besonders bevorzugt mindestens 93% oder mindestens 94%, am bevorzugtesten mindestens 95%, mindestens 96%, mindestens 97% oder mindestens 98%, und insbesondere mindestens 99%, auf.

In einer bevorzugten Ausführungsform weist die Alkohol-Dehydrogenase in ihrer Proteinsequenz eines oder mehrere der folgenden Sequenzmotive auf:
L1 NNAG, wobei 1 = I, L oder V, oder FVSNAG.
Eine Unit Alkohol-Dehydrogenase wird definiert als Umsetzung von 1 μηιοΙ 4-Chloro-3-oxo- Buttersäure-Ethylester (COBE) innerhalb von 1 min bei 30 °C in 50 mM Tris/HCL Puffer bei pH 7,0; 10 mM COBE, 2 mM MgCI2, 0,25 mM NADH, 0,25 mM NADPH.
In einer bevorzugten Ausführungsform weist die Alkohol-Dehydrogenase nach Inkubation über 48 h bei 30 °C in 50 mM Tris/HCL Puffer bei pH 7,0; 2 mM MgCI2; 0, 1 % DMSO; 0,01 % Triton-X-100; 3 g/L Ethylsecodion; 1 mM NADH noch eine Restaktivität von > 50 %, bevorzugter > 60 %, besonders bevorzugt 70 %, ganz besonders bevorzugt > 80 % und insbesondere bevorzugt £ 85 % auf.
In einer bevorzugten Ausführungsform wird die verwendete Alkoholdehydrogenase rekombinant mit Hilfe eines Mikroorganismus hergestellt. Beispiele für geeignete Wirte zur rekombinanten Herstellung der Alkohol-Dehydrogenase sind Escherichia coli, Pseudomonas sp., Bacillus sp., Pichia pastoris, Saccharomyces cerevisiae oder Aspergillus sp.
In einer bevorzugten Ausführungsform wird die verwendete Alkoholdehydrogenase mit einer Ausbeute von > 25000 U / L Kultur, bevorzugter > 50000 U / L Kultur, besonders bevorzugt > 100000 U / L Kultur, ganz besonders bevorzugt > 200000 U / L Kultur und insbesondere bevorzugt > 300000 U / L Kultur hergestellt.
In einer bevorzugten Ausführungsform wird die verwendete Alkoholdehydrogenase mit einer Konzentration von < 10.000 U / L Reaktion, bevorzugter < 5.000 U / L Reaktion, besonders bevorzugt -5 3.000 U / L Reaktion, ganz besonders bevorzugt ^ 2.000 U / L Reaktion und insbesondere bevorzugt < 1 .500 U / L Reaktion eingesetzt.
Vorzugsweise wird das erfindungsgemäße Verfahren in einem wässrigen organischen Zweiphasensystem durchgeführt.
Vorzugsweise wird das erfindungsgemäße Verfahren bei einem pH- Wert von 5 bis 10, besonders bevorzugt 6 bis 8 durchgeführt.
In einer bevorzugten Ausführungsform umfasst die Reaktionsmischung einen Puffer, besonders bevorzugt einen Kaliumphosphat-, Tris/HCI-, Glycin- oder Triethanolamin-Puffer,
insbesondere einen Tris/HCI-Puffer. Vorzugsweise enthält der Puffer ferner Magnesium- Ionen und/oder Zinkionen.
Vorzugsweise wird das Secodion-Derivat der allgemeinen Formel 1 in einer Konzentration von > 1 g/L, bevorzugter > 9 g/L, besonders bevorzugt > 25 g/L, ganz besonders bevorzugt > 35 g/L und insbesondere bevorzugt ä 40 g/L eingesetzt.
Vorzugsweise wird bei der Umsetzung des Secodion-Derivates zum gewünschten Seconol ein Umsatz von > 70 %, bevorzugter > 80 %, besonders bevorzugt ä 90 %, ganz besonders bevorzugt 2: 95 % und insbesondere bevorzugt 2. 97 % erreicht.
In einer bevorzugten Ausführungsform wird das Secodion-Derivat der allgemeinen Formel 1 in einer Konzentration von > 40 g/L und die Alkoholdehydrogenase in einer Konzentration von < 1500 U / L Reaktion eingesetzt und ein Umsatz von > 97 % erreicht.
Vorzugsweise wird das Secodion-Derivat der allgemeinen Formel 1 in Gegenwart einer substöchiometrischen Menge eines NADH oder NADPH Co-Faktors reduziert.
Vorzugsweise wird der NADH oder NADPH Co-Faktor mit Hilfe einer weiteren enzymatischen Reaktion kontinuierlich regeneriert.
Das NADH bzw. NADPH kann als solches oder in seiner oxidierten Form, d.h. als NAD bzw. NADP, eingesetzt und mit Hilfe der weiteren enzymatischen Reaktion in situ generiert werden.
In einer bevorzugten Ausführungsform wird der NADH oder NADPH Co-Faktor durch Oxidation wenigstens eines Alkohols, vorzugsweise eines monofunktionellen, sekundären (C3-C8)-Alkohols, in Gegenwart der Dehydrogenase regeneriert.
In einer anderen bevorzugten Ausführungsform wird der NADH oder NADPH Co-Faktor durch Oxidation von Glucose zu Gluconolacton in Gegenwart einer Glucose-Dehydrogenase (GDH) regeneriert; d.h. die enzymatische Reduktion des Secodion-Derivats der allgemeinen Formel 1 wird ferner in Gegenwart einer Glucose-Dehydrogenase durchgeführt.
Eine Unit GDH-Aktivität wird definiert als die Umsetzung von 1 μιηοΙ Glucose innerhalb von 1 min bei 30 °C in 50 mM Kaliumphosphatpuffer pH 7,4, 200 mM Glucose, 1 mM NAD.
In einer bevorzugten Ausführungsform wird die GDH mit einer Konzentration von
< 5.000 U / L Reaktion, bevorzugter < 2.000 U / L Reaktion, besonders bevorzugt
< 1.000 U / L Reaktion, ganz besonders bevorzugt < 500 U / L Reaktion und insbesondere bevorzugt < 200 U / L Reaktion eingesetzt.
In weiteren bevorzugten Ausführungsformen wird die Cofaktor-Regenerierung durch Oxidation von Formiat in Gegenwart von Formiat-Dehydrogenase, durch Oxidation von Phosphit durch Phosphit-Dehydrogenase oder durch Oxidation von Glucose-6-Phosphat durch Glucose-6-Phosphat-Dehydrogenase durchgeführt.
Vorzugsweise wird die enzymatische Reduktion in Gegenwart wenigstens eines Tensids, besonders bevorzugt in Gegenwart eines nicht-ionischen Tensids durchgeführt.
Geeignete nicht-ionische Tenside sind beispielsweise
- Alkylglycoside, insbesondere Polyoxyethylen-Sorbitanfettsäureester (Polysorbate), wie z.B. Polyoxyethylen(20)sorbitanmonolaurat (Handelsname u.a. Tween® 20), Polyoxy- ethylen(4)sorbitanmonolaurat (Handelsname u.a. Tween® 21 ), Polyoxyethylen(20)- sorbitanmonopalmitat (Handelsname u.a. Tween® 40), Polyoxyethylen(20)sorbitan monostearat (Handelsname u.a. Tween® 60), Polyoxyethylen(20)sorbitantristearat (Handelsname u.a. Tween® 65), Polyoxyethylen(20)sorbitanmonooleat (Handelsname u.a. Tween® 80), Polyoxyethylen(5)sorbitan monooleat (Handelsname u.a. Tween® 81 ), und Polyoxyethylen(20)sorbitantrioleat (Handelsname u.a. Tween® 85); vorzugsweise Fettsäuremonoester des Polyoxyethylensorbitans gemäß der allgemeinen Formel 7
(7), worin (w+x+y+z) eine Zahl im Bereich von 15 bis 60, vorzugsweise 17 bis 40, und insbesondere von 19 bis 21 ist; und Alkylen eine gegebenenfalls ungesättigte (C6-C30)-Alkylengruppe, besonders bevorzugt (C8-C24)-Alkylengruppe, und am bevorzugtesten (Ci0-Ci6)-Alkylengruppe ist;

Besonders bevorzugte nicht-ionische Tenside sind ausgewählt aus der Gruppe bestehend aus den Polyalkylenglycolethern; Alkylglycosiden, insbesondere Polyoxyethylen-Sorbitan- fettsäureestern; Octylphenolethoxylaten und Nonylphenolethoxylaten.
Ganz besonders bevorzugt sind Alkylphenolethoxylate der allgemeinen Formel 11, worin R4 für (C8-C9)-Alkyl steht und sich in para-Position relativ zum Polyoxyethylen-Rest befindet; und p eine Zahl zwischen 8 und 12 bedeutet. Insbesondere bevorzugt ist Triton X-100.
Vorzugsweise weist das Tensid einen HLB-Wert (hydrophilic-lipophilic balance) von mindestens 8, besonders bevorzugt mindestens 10, und insbesondere mindestens 12 auf.
Beispiele für diese Art von Tensiden sind die vorstehend beschriebenen Tenside Triton X- 100, "Tween® 20", "Tween® 40", "Tween® 60", "Tween® 80", "Brij® 30", "Brij® 35", "Brij® 56" und "Brij® 58".
In einer bevorzugten Ausführungsform hat das Tensid einen HLB-Wert im Bereich von 12±4, vorzugsweise 12±3,5, besonders bevorzugt 12±3, noch bevorzugter 12±2,5, ganz besonders bevorzugt 12±2, am bevorzugtesten 12±1 ,5, und insbesondere 12±1.
In einer anderen bevorzugten Ausführungsform hat das Tensid einen HLB-Wert im Bereich von 14±4, vorzugsweise 14±3,5, besonders bevorzugt 14±3, noch bevorzugter 14±2,5, ganz besonders bevorzugt 14±2, am bevorzugtesten 14±1 ,5, und insbesondere 14±1.
Vorzugsweise beträgt der Gehalt des Tensids in der Reaktionsmischung mindestens 0,0001 Vol.-%, besonders bevorzugt mindestens 0,0005 Vol.-%, ganz besonders bevorzugt mindestens 0,001 Vol.-%, und insbesondere mindestens 0,005 Vol.-%.
Vorzugsweise beträgt der Gehalt des Tensids in der Reaktionsmischung höchstens 25 Vol.-%, besonders bevorzugt höchstens 20 Vol.-%, ganz besonders bevorzugt höchstens
15 Vol.-%, noch bevorzugter höchstens 10 Vol.-%, am bevorzugtesten höchstens 7,5 Vol.-%, und insbesondere höchstens 5 Vol.-%.
In einer bevorzugten Ausführungsform beträgt der Gehalt des Tensids in der Reaktionsmischung 0,0001 bis 1 Vol.-%, und insbesondere bevorzugt 0,001 bis 0,5 Vol.-%.
In einer anderen bevorzugten Ausführungsform beträgt der Gehalt des Tensids in der Reaktionsmischung 0,01 bis 10 Vol.-%, besonders bevorzugt 0,1 bis 5 Vol.-%, und insbesondere 0,1 bis 1 Vol.-%.
In einer weiteren bevorzugten Ausführungsform beträgt der Gehalt des Tensids in der Reaktionsmischung 0,1 bis 10 Vol.-%, besonders bevorzugt 1 bis 10 Vol.-%, und insbesondere 2,5 bis 7,5 Vol.-%.
In einer bevorzugten Ausführungsform wird die enzymatische Reduktion in Gegenwart wenigstens eines Alkohols, vorzugsweise eines monofunktionellen (C2-C8)-Alkohols, insbesondere eines monofunktionellen, sekundären (C3-C8)-Alkohols und/oder Dimethyl- sulfoxid als Cosolvens durchgeführt. Soweit ein Alkohol als Cosolvens vorhanden ist, dient dieser vorzugsweise auch zur Regenerierung des NADH oder NADPH Co-Faktors.
Vorzugsweise beträgt der Gehalt des Cosolvens in der Reaktionsmischung höchstens 90 Vol-%, besonders bevorzugt höchstens 60 Vol.-%, ebenfalls bevorzugt höchstens 30 Vol.-%, weiterhin bevorzugt höchstens 25 Vol.-%, am bevorzugtesten höchstens 20 Vol.-%, und insbesondere höchstens 15 Vol.-%.
In einer bevorzugten Ausführungsform wird die enzymatische Reduktion in Gegenwart wenigstens eines Alkohols durchgeführt.
Bevorzugte Alkohole sind monofunktionelle (C2-C8)-Alkohole, besonders bevorzugt sind monofunktionelle, sekundäre (C3-C8)-Alkohole vorzugsweise ausgewählt aus der Gruppe bestehend aus Isopropanol (2-Propanol), 2-Butanol, 2-Pentanol, 3-Pentanol, 3-Methyl-2- butanol, 2-Hexanol, 3-Hexanol, 3-Methyl-2-pentanol, 4-Methyl-2-pentanol, 2-Methyl-3- pentanol, 3,3-Dimethyl-2-butanol, 2-Heptanol, 2-Octanol und Cyclohexanol. Ganz besonders bevorzugt ist Isopropanol.
Vorzugsweise beträgt der Gehalt des Alkohols in der Reaktionsmischung höchstens 30 Vol-%, besonders bevorzugt höchstens 25 Vol.-%, am bevorzugtesten höchstens 20 Vol.-%, und insbesondere höchstens 15 Vol.-%.
Vorzugsweise beträgt der Gehalt des Alkohols in der Reaktionsmischung mindestens 1 Vol-%, besonders bevorzugt mindestens 2,5 Vol.-%, am bevorzugtesten mindestens 5 Vol-%, und insbesondere mindestens 7,5 Vol.-%.
In einer anderen bevorzugten Ausführungsform wird die enzymatische Reduktion in Gegenwart von Dimethylsulfoxid durchgeführt.
Vorzugsweise beträgt der Gehalt des Dimethylsulfoxids in der Reaktionsmischung höchstens 20 Vol-%, besonders bevorzugt höchstens 15 Vol.-%, ganz besonders bevorzugt höchstens 10 Vol.-%, am bevorzugtesten höchstens 7,5 Vol.-% und insbesondere höchstens 5 Vol.-%.
Verglichen mit dem mit Ganzzellkatalyse mit Wildtyp-Organismen der Gattung Saccharomyces durchgeführten, aus H. Heidepriem et al. Justus Liebigs Annalen der Chemie (1968), 712, 155-67 und H. Gibian et al. Tetrahedron Letters (1966) bekannten Verfahren, hat das erfindungsgemäße Verfahren den Vorteil, dass ein weitaus geringerer Anteil an doppelt reduzierten Nebenprodukten entsteht. Ferner hat die Verwendung von rekombinanten Enzymen gegenüber Wildtyp-Zellen generell den Vorteil, dass die Reaktionsbedingungen nicht an ein Überleben der Zellen geknüpft sind, höhere Substratkonzentrationen benutzt werden können, keine Zellgrenzen überwunden werden müssen und höhere Produkt-Bildungsraten erzielt werden.
Das erfindungsgemäße Verfahren hat ferner gegenüber den aus dem Stand der Technik bekannten Verfahren, sowohl gegenüber denen mit Ganzzellkatalyse als auch gegenüber denen mit rekombinanten Enzymen, den Vorteil, dass das reduzierte Produkt mit hoher Stereoselektivität und Reinheit erhalten und somit der Prozess effektiver gestaltet werden kann.
Ein weiterer Vorteil des erfindungsgemäßen Verfahrens ist, dass das schwerlösliche Substrat und das reduzierte Produkt, insbesondere bei Einsatz einer Kombination eines Cosolvens und eines Tensids homogen in der wässrigen Reaktionslösung suspendiert werden können. Folglich können die eingesetzte Konzentrationen an sekundären Alkoholen, wie z.B. Isopropanol, 4-Methyl-2-pentanol oder 2-Heptanol, stark reduziert oder sogar ganz auf den Einsatz eines monofunktionellen, sekundären Alkohols verzichtet werden und die
Akzeptanz des Reaktionssystems durch das ADH-Enzym erhöht werden. Unter solchen milden Bedingungen können im wässrigen System Substratkonzentrationen von bis zu 40 g/l erzielt werden.
Ein weiterer Aspekt der Erfindung betrifft ein Verfahren zur enzymatischen Reduktion eines Secodion-Derivats der allgemeinen Formel 1
mit einer Alkohol-Dehydrogenase zu einem 17-ß-Seconol-Derivat der allgemeinen Formel 2

Vorzugsweise wird auch bei diesem Verfahren das 17-ß-Seconol-Derivat der allgemeinen Formel 2 mit einer Produktreinheit von > 90% und einem Stereoisomeren-Anteil von > 90% erhalten.
Ein weiterer Aspekt der Erfindung betrifft eine Verwendung einer Alkohol-Dehydrogenase zur enzymatischen Reduktion eines Secodion-Derivats der allgemeinen Formel 1
(2 ) jeweils für -H oder eine -(C C4)-Alkylgruppe steht; ausgewählt ist aus -H, -(CrC6)-Alkyl, -(d-C )-Alkenyl, -C(=0)R",
-CRdRe-Aryl, -Tetrahydropyranyl und -(CH2)i-2-X-Rf; worin
R' für -Phenyl; unsubstituiertes, ein oder mehrfach durch Halogen substituiertes -(C-|-C6)-Alkyl oder für -O-Allyl steht;
Ra, Rb und Rc jeweils unabhängig voneinander für -(d-CßJ-Alkyl oder Phenyl stehen;
Rd und Re jeweils unabhängig voneinander für -H, für unsubstituiertes oder durch -OMe einfach substituiertes -Phenyl stehen;
R für -(C C4)-Alkyl, Phenyl oder -(CH2)1-2SiRaRbRc steht;
X für O oder S steht; und
Aryl für unsubstituiertes oder durch -OMe einfach oder mehrfach substituiertes Phenyl steht; und besonders bevorzugt R für Methyl oder Ethyl und R' für Methyl steht.
Es wurde weiterhin überraschend gefunden, dass die Synthese von Estrogenen bzw. Gestagenen möglich ist durch ein Verfahren umfassend das Verfahren zur enzymatischen Reduktion eines Secodion-Derivats der allgemeinen Formel 1 mit einer Alkohol- Dehydrogenase zu einem 17-ß-Seconol-Derivat der allgemeinen Formel 2.
Ein weiterer Aspekt der Erfindung betrifft somit ein Verfahren zur Herstellung eines Estrogens oder eines Gestagens umfassend das Verfahren zur enzymatischen Reduktion eines Secodion-Derivats der allgemeinen Formel 1 mit einer Alkohol-Dehydrogenase zu einem 17-ß-Seconol-Derivat der allgemeinen Formel 2.
Die nachfolgenden Beispiele dienen zur näheren Erläuterung der Erfindung, sind jedoch nicht einschränkend auszulegen.
Beispiele
Beispiel 1 - Herstellung von Enzymen
Die Enzymvarianten wurden mit Hilfe herkömmlicher Verfahren hergestellt und im Hinblick auf ihre Eigenschaften untersucht. Das Herstellverfahren beruhte auf Transformation von Escherichia coli Stämmen mit einem DNA Plasmid, welches die Information und Regulation für die Produktion des Enzyms beinhaltete. Die kodierende Sequenz des Enzyms oder der Enzymvariante wurde dabei unter die Kontrolle eines induzierbaren Promoters gestellt. Dadurch ließ sich durch Zugabe eines Induktors (i.d.R. Isopropyl-ß-D-thiogalactopyranosid
(IPTG)) steuern. Der transformierte E. coli Stamm wurde dann in einem herkömmlichen Nährmedium (z.B. LB (lysogeny broth) mit 10 g/L NaCI; Lennox-Broth mit 5 g/L NaCI, Minimalmedium M9) kultiviert und, nachdem mit IPTG induziert wurde und die Enzymproduktion maximal war, durch Abtrennung der Biomasse durch z.B. Zentrifugation geerntet. Aus der Biomasse konnte dann das Enzym nach entsprechender Zelllyse gewonnen werden. Dabei wurden Zentrifugations-, Fällungs-, Ultrafiltrations- und/oder Chromatographieverfahren eingesetzt.
Beispiel 2 - Ganzzelltransformation mit Wildtyp-Orqanismen.
Es wurden zwei mikrobielle Referenzsysteme hinsichtlich der Umsetzung und Produktreinheit getestet. Hierfür wurden die Stämme Saccharomyces uvarum (DSMZ: 70412) und Saccharomyces carlsbergensis (DSMZ: 6580) ausgewählt (vgl. dazu H. Gibian et al. Tetrahedron Letters (1966), 21 2321 -30). Die Hefestämme wurden in einem Nährmedium (2 % Glucose, 2 % Saccharose, 0.8 % Hefe-Extrakt, pH 6.5) bei 26 ± 2 °C kultiviert. Nach Erreichen einer optischen Dichte (600 nm) von 8-12 wurde der pH-Wert auf pH 6.5 eingestellt und 3 g/L Ethylsecodion zugegeben. Anschließend wurden die Kulturen für weitere 60 h bei 26 ± 2 °C geschüttelt.
Nach Extraktion der Kulturen mit tert-Butylmethylether (MTBE) wurde eine achirale HPLC - Analytik zur Bestimmung der Produktreinheit durchgeführt.

Es zeigte sich, dass mit diesem Verfahren überreduzierte Nebenprodukte auftreten. Beispiel 3 - Screening der ADH-Kollektion
Für das erste Screening wurde die Reaktion unter folgenden Reaktionsbedingungen durchgeführt: 50 mM Tris/HCL Puffer bei pH 7,0; 2 mM MgCI2; 1 % DMSO; 0,01 % Triton-X- 100; 3 g/l Ethylsecodion; 1 mM NADH bzw. NADPH; 200 mM Glucose; GDH-03 und das entsprechende ADH-Enzym. Die Reaktion wurde über Nacht bei 30°C inkubiert. Die Enzyme wurden alle in E. coli gemäß Beispiel 1 hergestellt und in der Reaktion eingesetzt. Die Analyse erfolgte zuerst mit achiraler HPLC zur Bestimmung der Produktbildung und der
Produktreinheit. Die interessanten Kandidaten wurden ferner zur Bestimmung des Stereoisomerenüberschusses mit chiraler HPLC analysiert.
Beispiel 4 - Umsetzung von Ethylsecodion mit Isopropanol Cofaktorreqenerierung
In diesem Ansatz wurde Isopropanol zur Cofaktorregenerierung eingesetzt. Die Zusammensetzung der Reaktionsmischung war: 50 mM Tris/HCL Puffer bei pH 7,0 bzw pH 9,0 für CmADH9; 2 mM MgCI2; 10% Isopropanol; 3 g/l Ethylsecodion; 1 mM NAD bzw NADP und das entsprechende ADH-Enzym. Die Enzyme wurden wie in Beispiel 1 beschrieben hergestellt. Es wurden neben erfindungsgemäßen Enzymen zum Vergleich die in WO2008/068030 benutzten Enzyme mit den dort offenbarten Nukleotid-Sequenzen Seq ID No 6 (CaADH), Seq ID No 7 (RxADH), Seq ID No 8 (CmADH8), Seq ID No 9 (CmADH9) sowie Seq ID No 10 (CmADHIO) bzw. Protein-Sequenzen Seq ID No 1 (CaADH), Seq ID No 2 (RxADH), Seq ID No 3 (CmADH8), Seq ID No 4 (CmADH9) sowie Seq ID No 5 (CmADHIO) untersucht. Die RxADH konnte nicht löslich und somit nur inaktiv hergestellt werden. Es wurde dementsprechend keine Umsetzung von Secodion festgestellt. Es wurde die Umsetzung von Ethylsecodion untersucht.

Es zeigte sich, dass die Umsetzung mit dem erfindungsgemäßen Enzym gemäß Seq ID 4 sowohl eine hohe Produktreinheit als auch einen hohen Stereoisomerenüberschuss liefert. Die untersuchten Enzyme aus WO2008/068030 hingegen zeichnen sich dadurch aus, dass sie in Bezug auf Produktreinheit und Stereoisomerenüberschuss deutlich unterlegen sind.
Beispiel 5 - Umsetzung von Ethylsecodion mit Glucose/GDH zur Cofaktorregenerierung
In diesem Ansatz wurde das Glucose/GDH System zur Cofaktorregenerierung benutzt. Als Cosolvenz wurde DMSO eingesetzt. Die Zusammensetzung der Reaktionsmischung war: 50 mM Tris/HCL Puffer bei pH 7,0 bzw pH 9,0 für CmADH9; 2 mM MgCI2; 1 % DMSO; 0,01 % Triton-X-100; 3 g/l Ethylsecodion; 1 mM NAD bzw NADP, 200 mM Glucose, GDH-03 und das entsprechende ADH-Enzym. Die Enzyme wurden wie in Beispiel 1 beschrieben
hergestellt. Es wurden neben erfindungsgemäßen Enzymen zum Vergleich die in WO2008/068030 benutzten Enzyme untersucht (siehe Beispiel 4).

Es zeigte sich, dass nur in den Umsetzungen mit Enzym gemäß Seq ID 2 oder Seq ID 4 sowohl eine hohe Produktreinheit als auch ein hoher Stereoisomerenüberschuss erreicht werden konnte. Die untersuchten Enzyme aus WO2008/068030 hingegen zeichnen sich auch in diesem Reaktionssystem dadurch aus, dass sie in Bezug auf Produktreinheit und Stereoisomerenüberschuss deutlich unterlegen sind.
Beispiel 6 - Präparative Umsetzung mit Seq ID 2
In einem präparativen Ansatz wurden 600 mg Ethylsecodion in einem Gesamtvolumen von 15 ml unter folgendem Bedingungen umgesetzt (40g/l): 100 mM Triethanolamin/HCL Puffer pH 7,0; 2 mM MgCI2; 5 % DMSO; 5 % Triton-X-100; 1 mM NAD, 200 mM Glucose, GDH-03 und ADH gemäß Seq ID NO 2. Während der Reaktion wurde der pH-Wert durch automatische Titration von 5 M NaOH-Lösung in einem pH-Stat-Gerät konstant auf 7,0 gehalten. Alternativ dazu kann das Verfahren auch unter Einsatz von 0,25 mM NAD und Titration mittels 1 M NaOH durchgeführt werden. Die Reaktion wurde 30h bis 39 h bei 30°C gerührt. Das Produkt wurde mit MTBE extrahiert und auf Umsatz, Produktreinheit und Stereoisomerenüberschuss analysiert. Es wurden 97,4% des Ethylsecodions zu 17-ß- Ethylseconol umgesetzt. Die Produktreinheit betrug >99,9% und der Stereoisomerenüberschuss >99,9%.
Beispiel 7 - Umsetzung von Methylsecodion mit ADH Seq ID 2
Die Reaktion wurde in analoger weise mit dem Substrat Methylsecodion durchgeführt. Die Bedingungen waren: 100 mM Tris/HCL Puffer bei pH 7,0; 2 mM MgCI2; 5 % DMSO; 0,1 % Triton-X-100; 3 g/l Methylsecodion; 1 mM NAD, 200 mM Glucose, GDH-03 und ADH Seq ID 2. Der Umsatz betrug 92,2% und die Produktreinheit 95%.
Beispiel 8 - Umsetzung von Methylsecodion mit ADH Seq ID 4
Die Reaktion von Methylsecodion mit ADH Seq ID 4 wurde unter folgenden Bedingungen durchgeführt: 100 mM Tris/HCL Puffer bei pH 7,0; 2 mM MgCI2; 10% Isopropanol; 0,1 % Triton-X-100; 3 g/l Methylsecodion; 1 mM NADP und ADH Seq ID 4. Der Umsatz betrug 87,5% und die Produktreinheit 99%.
Claims
Patentansprüche:
Ein Verfahren zur enzymatischen Reduktion eines Secodion-Derivats der allgemeinen Formel 1
(1 ) mit einer Alkohol-Dehydrogenase zu einem 17-ß-Seconol-Derivat der allgemeinen Formel 2
(2) wobei jeweils
R für -H oder eine -(C C4)-Alkylgruppe steht; und
R' ausgewählt ist aus -H, -(C C6)-Alkyl, -(C1-C4)-Alkenyl, -C(=0)R",
-SiRaRbRc, -CRdRe-Aryl, -Tetrahydropyranyl und -(CH2)i-2-X-Rf; worin
R" für -Phenyl; unsubstituiertes, ein oder mehrfach durch Halogen substituiertes -(C C6)-Alkyl oder für -O-Allyl steht;
Ra, Rb und Rc jeweils unabhängig voneinander für -(C C6)-Alkyl oder Phenyl stehen;
Rd und Re jeweils unabhängig voneinander für -H, für unsubstituiertes oder durch -OMe einfach substituiertes -Phenyl steht;
Rf für -(d-C^-Alkyl, Phenyl oder -(CH^^SiR^R0 steht; X für O oder S steht; und
Aryl für unsubstituiertes oder durch -OMe einfach oder mehrfach substituiertes Phenyl steht.
2. Das Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass das 17-ß-Seconol- Derivat der allgemeinen Formel 2 mit einem Stereoisomeren-Anteil von > 90% und einer Produktreinheit von > 90% erhalten wird.
3. Das Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass R für Methyl oder Ethyl steht.
4. Das Verfahren nach einem der vorstehenden Ansprüche, dadurch gekennzeichnet, dass R' für Methyl steht.
5. Das Verfahren nach einem der vorstehenden Ansprüche, dadurch gekennzeichnet, dass die Alkohol-Dehydrogenase eine Aminosäuresequenz mit mindestens 60% Homologie zu wenigstens einer der Aminosäuresequenzen ausgewählt aus der Gruppe bestehend aus (Seq ID No: 2), (Seq ID No: 4), (Seq ID No: 6), (Seq ID No: 8) und (Seq ID No: 10) umfasst.
6. Das Verfahren nach einem der vorstehenden Ansprüche, dadurch gekennzeichnet, dass das Secodion-Derivat der allgemeinen Formel 1 in Gegenwart einer substöchio- metrischen Menge eines NADH oder NADPH Co-Faktors reduziert wird.
7. Das Verfahren nach einem der vorstehenden Ansprüche, dadurch gekennzeichnet, dass das Secodion-Derivat der allgemeinen Formel 1 in Gegenwart einer Glucose- Dehydrogenase reduziert wird.
8. Das Verfahren nach einem der vorstehenden Ansprüche, dadurch gekennzeichnet, dass die enzymatische Reduktion in Gegenwart wenigstens eines Tensids durchgeführt wird.
9. Das Verfahren nach einem der vorstehenden Ansprüche, dadurch gekennzeichnet, dass die enzymatische Reduktion in Gegenwart wenigstens eines nicht-ionischen Tensids, vorzugsweise ausgewählt aus der Gruppe bestehend aus Polyalkylen- glycolethern, Polyoxyethylen-Sorbitanfettsäureestern, Octylphenolethoxylaten und Nonylphenolethoxylaten durchgeführt wird.
10. Das Verfahren nach Anspruch 8 oder 9, dadurch gekennzeichnet, dass der Gehalt des Tensids in der Reaktionsmischung mindestens 0,001 Vol.-%, beträgt.
1 1. Das Verfahren nach einem der vorstehenden Ansprüche, dadurch gekennzeichnet, dass die enzymatische Reduktion in Gegenwart eines sekundären (C3-C8)-Alkohols und/oder von Dimethylsulfoxid als Cosoivens durchgeführt wird.
12. Das Verfahren nach einem der vorstehenden Ansprüche, dadurch gekennzeichnet, dass der Gehalt des Cosoivens in der Reaktionsmischung höchstens 20 Vol.-% beträgt.
Verwendung einer Alkohol-Dehydrogenase definiert wie in einem der Ansprüche 5 6 zur enzymatischen Reduktion eines Secodion-Derivats der allgemeinen Formel 1
(1 ) nem 17-ß-Seconol-Derivat der allgemeinen Formel 2
(2) wobei jeweils
R für -H oder eine -(d-C4)-Alkylgruppe steht; und
R' ausgewählt ist aus -H, -(d-Q -Alkyl, -(C C4)-Alkenyl, -C(=0)R",
-SiRaRbRc, -CRdRe-Aryl, -Tetrahydropyranyl und -(CHz^-X-R'; worin R" für -Phenyl; unsubstituiertes, ein oder mehrfach durch Halogen substituiertes -(Ci-C6)-Alkyl oder für -O-Allyl steht;
Ra, Rb und Rc jeweils unabhängig voneinander für -(C C6)-Alkyl oder Phenyl stehen;
Rd und Re jeweils unabhängig voneinander für -H, für unsubstituiertes oder durch -OMe einfach substituiertes -Phenyl steht;
Rf für -(d-C4)-Alkyl, Phenyl oder -(CH2)i-2SiRaRbRc steht;
X für O oder S steht; und
Aryl für unsubstituiertes oder durch -OMe einfach oder mehrfach substituiertes Phenyl steht.
Ein Verfahren zur Herstellung eines Estrogens oder eines Gestagens umfassend das Verfahren nach einem der Ansprüche 1 bis 12.
