Description

アスコルビン酸の影響抑制方法
 本発明は、糖化ヘモグロビンの測定におけるアスコルビン酸の影響抑制方法、及び、糖化ヘモグロビンの測定用試薬に関する。
 糖化ヘモグロビンは、ヘモグロビンにグルコースが結合した糖化物である。ヘモグロビンはα鎖、β鎖からなる四量体の構造を取っているが、β鎖のN末端が糖化したものはヘモグロビンA1cと呼ばれ、血糖値が高い程、増加することから、糖尿病マーカーとして、臨床検査において測定されている。
 糖化ヘモグロビンの測定方法としては、HPLC法等のクロマトグラフィ法、電気泳動法、ラテックス免疫凝集法など抗体を利用する免疫測定法、糖化タンパク質に作用する酵素と、糖化ペプチド及び/又は糖化アミノ酸に作用する酵素とを用いる酵素測定法等が知られている。
 糖化ヘモグロビンの酵素測定法としては、先ず、ヘモグロビン含有試料中のヘモグロビンを変性剤により変性させ、変性したヘモグロビンにタンパク質分解酵素を作用させ、次いで生成した糖化ペプチドに糖化ペプチド酸化酵素を作用させ、生成した過酸化水素をペルオキシダーゼ等の過酸化活性物質の存在下に、酸化発色型色原体と反応させて色素に導き、生成した色素の吸光度から糖化ヘモグロビンを測定する方法(吸光度法)等が知られている。
 しかしながら、この吸光度法による糖化ヘモグロビンの測定においては、試料中に存在するアスコルビン酸の影響が問題となる。従来、過酸化水素測定系を用いた試料中の測定対象物の測定において、アスコルビン酸の影響が問題となっており、アスコルビン酸の影響抑制という課題に対して、例えばアスコルビン酸酸化酵素を用いる方法(特許文献1参照)、鉄、コバルト、セレン、銅、水銀、ニッケル等の有機錯体を用いる方法(特許文献2~4参照)、ヨウ素酸塩を用いる方法(特許文献5参照報)、ヨウ素酸塩と共に、ヨウ素酸塩の補助剤として、リン酸化合物、ほう酸化合物、クエン酸化合物、リンゴ酸化合物、酒石酸化合物、及びシュウ酸化合物から選ばれた少なくとも1種の化合物を用いる方法(特許文献6)等が知られているが、糖化ヘモグロビンの測定におけるアスコルビン酸の影響抑制効果は十分なものではなかった。

 本発明の目的は、糖化ヘモグロビン含有試料中の糖化ヘモグロビンの測定におけるアスコルビン酸の影響抑制方法、及び、糖化ヘモグロビン測定用試薬を提供することにある。
 本発明者らは本課題について鋭意検討したところ、糖化ヘモグロビン含有試料にタンパク質分解酵素を作用させた後、生成した糖化ペプチドを糖化ペプチド酸化酵素と反応させ、生成した過酸化水素を測定することにより、試料中の糖化ヘモグロビンを測定する方法において、糖化ペプチドと糖化ペプチド酸化酵素との反応を、ハロゲン酸化物と、窒素原子及び硫黄原子を含む特定の5員環複素環化合物との存在下に行うことにより、試料中のアスコルビン酸の影響を抑制できる、という知見を見出し、本発明を完成させた。すなわち、本発明は、以下の[1]~[7]に関する。
[1] 糖化ヘモグロビン含有試料にタンパク質分解酵素を作用させた後、生成した糖化ペプチドを糖化ペプチド酸化酵素と反応させ、生成した過酸化水素を測定することにより、試料中の糖化ヘモグロビンを測定する方法において、糖化ペプチドと糖化ペプチド酸化酵素との反応を、ハロゲン酸化物と、一般式(I)

 本発明により、糖化ヘモグロビンの測定におけるアスコルビン酸の影響抑制方法、及び、糖化ヘモグロビン測定用試薬が提供される。

 本発明の、糖化ヘモグロビン含有試料中の糖化ヘモグロビンの測定におけるアスコルビン酸の影響抑制方法は、糖化ヘモグロビン含有試料にタンパク質分解酵素を作用させた後、生成した糖化ペプチドを糖化ペプチド酸化酵素と反応させ、生成した過酸化水素を測定することにより、試料中の糖化ヘモグロビンを測定する方法において、糖化ペプチドと糖化ペプチド酸化酵素との反応を、ハロゲン酸化物と、一般式(I)

 本発明の測定方法における糖化ヘモグロビン含有試料は、糖化ヘモグロビン及びヘモグロビンを含有し、本発明のアスコルビン酸の影響抑制方法を可能とする試料であれば特に制限はなく、例えば全血、血球、血球に血漿が混在した試料、これらの試料を溶血処理した試料等が挙げられる。溶血処理としては、全血、血球、血球に血漿が混在した試料を溶血させる処理であれば特に制限はなく、例えば物理的方法、化学的方法、生物学的方法等が挙げられる。物理的方法としては、例えば蒸留水等の低張液を用いる方法、超音波を用いる方法等が挙げられる。化学的方法としては、例えばメタノール、エタノール、アセトン等の有機溶媒を用いる方法、ポリオキシエチレン系界面活性剤を用いる方法等が挙げられる。生物学的方法としては、例えば抗体や補体を用いる方法等が挙げられる。
 本発明における糖化ヘモグロビンは、ヘモグロビンにグルコース等の糖が結合して生成したものであり、ヘモグロビンA1a、ヘモグロビンA1b、ヘモグロビンA1c等が挙げられ、ヘモグロビンA1cが好ましい。
 本発明におけるハロゲン酸化物としては、本発明のアスコルビン酸の影響抑制方法を可能とするハロゲン酸化物であれば特に制限はないが、例えば、ヨウ素酸若しくはその塩、臭素酸若しくはその塩、過ヨウ素酸若しくはその塩等が挙げられ、ヨウ素酸若しくはその塩、臭素酸若しくはその塩が好ましい。塩としては、例えばリチウム塩、ナトリウム塩、カリウム塩、アンモニウム塩、カルシウム塩、マグネシウム塩等が挙げられる。
 ハロゲン酸化物の具体例(製品)としては、例えばヨウ素酸、ヨウ素酸ナトリウム、ヨウ素酸カリウム、臭素酸、臭素酸ナトリウム、臭素酸カリウム、過ヨウ素酸、過ヨウ素酸ナトリウム、過ヨウ素酸カリウム等が挙げられる。
 本発明の糖化ヘモグロビンの測定方法において、ハロゲン酸化物の反応液中の濃度としては、本発明の糖化ヘモグロビンの測定方法を可能とする濃度であれば、特に制限はないが、通常0.005~20 mmol/Lであり、0.01~10 mmol/Lが好ましい。
 本発明における化合物(I)におけるRは置換若しくは非置換のアルキルを表す。置換若しくは非置換のアルキルにおけるアルキルとしては、例えば炭素数1~20の直鎖アルキル、炭素数3~20の分岐アルキル等が挙げられる。炭素数1~20の直鎖アルキルとしては、例えばメチル、エチル、プロピル、ブチル、ペンチル、ヘキシル、ヘプチル、オクチル、ノニル、デシル、ウンデシル、ドデシル(ラウリル)、トリデシル、テトラデシル(ミリスチル)、ペンタデシル、ヘキサデシル(セチル)、ヘプタデシル、オクタデシル(ステアリル)、ノナデシル、イコシル等が挙げられる。炭素数3~20の分岐アルキルとしては、例えばイソプロピル、イソブチル、イソペンチル、イソヘキシル、イソヘプチル、イソオクチル、イソノニル、イソデシル、イソウンデシル、イソドデシル、イソトリデシル、イソテトラデシル、イソペンタデシル、イソヘキサデシル、イソヘプタデシル、イソオクタデシル、イソノナデシル、イソイコシル、オクチルドデシル等が挙げられる。置換アルキルにおける置換基としては、例えばフェニル基、水酸基、スルホ基、シアノ基、ハロゲン原子等が挙げられる。ハロゲン原子としては、例えば塩素原子、臭素原子、ヨウ素原子等が挙げられる。
 化合物(I)の具体例(製品)としては、例えば2-オクチル-4-イソチアゾリン-3-オン(東京化成社製、ケミクレア社製)、2-メチル-4-イソチアゾリン-3-オン(シグマアルドリッチ社製)等が挙げられる。
 本発明における化合物(II)におけるRは置換若しくは非置換のアルキルを表す。置換若しくは非置換のアルキルにおけるアルキルとしては、例えば炭素数1~20の直鎖アルキル、炭素数3~20の分岐アルキル等が挙げられる。炭素数1~20の直鎖アルキルとしては、例えばメチル、エチル、プロピル、ブチル、ペンチル、ヘキシル、ヘプチル、オクチル、ノニル、デシル、ウンデシル、ドデシル(ラウリル)、トリデシル、テトラデシル(ミリスチル)、ペンタデシル、ヘキサデシル(セチル)、ヘプタデシル、オクタデシル(ステアリル)、ノナデシル、イコシル等が挙げられる。炭素数3~20の分岐アルキルとしては、例えばイソプロピル、イソブチル、イソペンチル、イソヘキシル、イソヘプチル、イソオクチル、イソノニル、イソデシル、イソウンデシル、イソドデシル、イソトリデシル、イソテトラデシル、イソペンタデシル、イソヘキサデシル、イソヘプタデシル、イソオクタデシル、イソノナデシル、イソイコシル、オクチルドデシル等が挙げられる。置換アルキルにおける置換基としては、例えばフェニル基、水酸基、スルホ基、シアノ基、ハロゲン原子等が挙げられる。ハロゲン原子としては、例えば塩素原子、臭素原子、ヨウ素原子等が挙げられる。
 化合物(II)の具体例(製品)としては、例えば2-メチルチアゾリン(東京化成社製)、2-メチルチアゾール(東京化成社製)、2-エチルチアゾール(東京化成社製)、2-プロピルチアゾール(東京化成社製)2-イソブチルチアゾール(東京化成社製)等が挙げられる。
 本発明のアスコルビン酸の影響抑制方法において、化合物(I)及び化合物(II)の反応液中の濃度としては、本発明の糖化ヘモグロビンの測定方法を可能とする濃度であれば、特に制限はないが、通常0.005~20 mmol/Lであり、好ましくは0.01~10 mmol/Lである。
 糖化ヘモグロビン含有試料にタンパク質分解酵素を作用させて糖化ペプチドを生成させる反応は、糖化ペプチドを生成し得る条件であれば、いかなる条件下でも行うことができる。当該反応、すなわち、糖化ヘモグロビン含有試料中の糖化ヘモグロビンとタンパク質分解酵素との反応は、水性媒体中で行われることが好ましい。水性媒体としては後述の水性媒体等が挙げられる。また、当該反応は、糖化ヘモグロビン含有試料中の糖化ヘモグロビンの変性剤の存在下に行うこともできる。当該変性剤により、糖化ヘモグロビンはタンパク質分解酵素との反応が可能な状態となることから、糖化ヘモグロビンとタンパク質分解酵素との反応は、変性剤の存在下に行うことが好ましい。変性剤としては、糖化ヘモグロビンをタンパク質分解酵素との反応が可能な状態にし得る変性剤であれば特に制限はなく、例えば陽イオン性界面活性剤、陰イオン性界面活性剤、両性界面活性剤、非イオン性界面活性剤等が挙げられる。
 陽イオン性界面活性剤としては、例えば第四級アンモニウム塩、ピリジニウム塩等が挙げられる。
 第四級アンモニウム塩としては、炭素数8~20の直鎖アルキルを少なくとも1つ有する第四級アンモニウム塩が好ましい。炭素数8~20の直鎖アルキルとしては、例えばオクチル、ノニル、デシル、ウンデシル、ドデシル(ラウリル)、トリデシル、テトラデシル(ミリスチル)、ペンタデシル、ヘキサデシル(セチル)、ヘプタデシル、オクタデシル(ステアリル)、ノナデシル、イコシル等が挙げられる。
 第四級アンモニウム塩の具体例(製品)としては、例えばデシルトリメチルアンモニウム クロライド、デシルトリメチルアンモニウム ブロマイド、ドデシルトリメチルアンモニウム クロライド、ドデシルトリメチルアンモニウム ブロマイド、テトラデシルトリメチルアンモニウム ブロマイド(以下、C14TMAと記す)、ヘキサデシルトリメチルアンモニウム クロライド、ヘキサデシルトリメチルアンモニウム ブロマイド、ジデシルジメチルアンモニウム クロライド、ジデシルジメチルアンモニウム ブロマイド、ジドデシルジメチルアンモニウム クロライド、ジドデシルジメチルアンモニウム ブロマイド(いずれも、東京化成社製)等が挙げられる。
 ピリジニウム塩としては、N-アルキルピリジニウム塩、N-アルケニルピリジニウム塩等が挙げられる。アルキルとしては、例えば炭素数1~20の直鎖アルキル、炭素数3~20の分岐アルキル等が挙げられる。炭素数1~20の直鎖アルキルとしては、例えばメチル、エチル、プロピル、ブチル、ペンチル、ヘキシル、ヘプチル、オクチル、ノニル、デシル、ウンデシル、ドデシル(ラウリル)、トリデシル、テトラデシル(ミリスチル)、ペンタデシル、ヘキサデシル(セチル)、ヘプタデシル、オクタデシル(ステアリル)、ノナデシル、イコシル等が挙げられる。炭素数3~20の分岐アルキルとしては、例えばイソプロピル、イソブチル、イソペンチル、イソヘキシル、イソヘプチル、イソオクチル、イソノニル、イソデシル、イソウンデシル、イソドデシル、イソトリデシル、イソテトラデシル、イソペンタデシル、イソヘキサデシル、イソヘプタデシル、イソオクタデシル、イソノナデシル、イソイコシル、オクチルドデシル等が挙げられる。
 アルケニルとしては、例えば炭素数2~20のアルケニル等が挙げられる。炭素数2~20のアルケニルとしては、例えばビニル、プロペニル、アリル、ブテニル、ペンテニル、ヘキセニル、ヘプテニル、オクテニル、ノネニル、デセニル、ウンデセニル、ドデセニル、テトラデセニル、ペンタデセニル、ヘキサデセニル、ヘプタデセニル、オクタデセニル、オレイル、ノナデセニル、イコセニル等が挙げられる。
 また、アルキルとして、置換アルキルを用いることもでき、アルケニルとして、置換アルケニルを用いることもできる。置換アルキル及び置換アルケニルにおける置換基としては、例えばフェニル基、水酸基、スルホ基、シアノ基、ハロゲン原子等が挙げられる。ハロゲン原子としては、例えば塩素原子、臭素原子、ヨウ素原子等が挙げられる。
 さらに、ピリジン環上の2~6位に置換基を有するN-アルキルピリジニウム塩、N-アルケニルピリジニウム塩も用いることもできる。ピリジン環上の2~6位の置換基としては、例えば炭素数1~20の直鎖アルキル、炭素数3~20の分岐アルキル、炭素数2~20のアルケニル等が挙げられる。炭素数1~20の直鎖アルキルとしては、例えば前述の炭素数1~20の直鎖アルキル等が挙げられる。炭素数3~20の分岐アルキルとしては、例えば前述の炭素数3~20の分岐アルキル等が挙げられる。炭素数2~20のアルケニルとしては、例えば前述の炭素数2~20の直鎖アルケニル等が挙げられる。
 ピリジニウム塩の具体例(製品)としては、例えば1-ドデシルピリジニウムクロライド(以下、C12pyと記す;東京化成社製)、1-セチルピリジニウムクロライド(東京化成社製)、1-セチル-4-メチルピリジニウムクロライド(東京化成社製)、N-オクタデシル-4-スチルバゾールブロミド(東京化成社製)等が挙げられる。
 陰イオン性界面活性剤としては、例えば硫酸エステル塩、カルボン酸塩、スルホン酸塩、リン酸エステル塩、スルホコハク酸塩、N-メチルタウリン塩、N-アルカノイル-N-メチルタウリン塩等が挙げられる。
 両性界面活性剤としては、例えば第三級アミンオキサイド、アルキルカルボキシベタイン等が挙げられる。
 非イオン性界面活性剤としては、例えばポリオキシエチレンアルキルアミン、ポリオキシエチレンアルケニルアミン、ポリオキシエチレンアルキルエーテル、ポリオキシエチレンアルケニルエーテル、ポリオキシエチレンアルキルフェニルエーテル、エチレンジアミンテトラポリオキシエチレン、ポリグリセリン脂肪酸エステル等が挙げられ、ポリオキシエチレンアルキルアミン、ポリオキシエチレンアルキルエーテルが好ましい。
 ポリオキシエチレンアルキルアミンにおけるアルキルとしては、例えば炭素数8~20のアルキル等が挙げられる。炭素数8~20のアルキルとしては、例えばオクチル、ノニル、デシル、ウンデシル、ドデシル(ラウリル)、トリデシル、テトラデシル(ミリスチル)、ペンタデシル、ヘキサデシル(セチル)、ヘプタデシル、オクタデシル(ステアリル)、ノナデシル、イコシル等が挙げられる。
 ポリオキシエチレンアルケニルアミンにおけるアルケニルとしては、例えば炭素数8~20のアルケニル等が挙げられる。炭素数8~20のアルケニルとしては、例えばオクテニル、ノネニル、デセニル、ウンデセニル、ドデセニル、テトラデセニル、ペンタデセニル、ヘキサデセニル、ヘプタデセニル、オクタデセニル、オレイル、ノナデセニル、イコセニル等が挙げられる。
 ポリオキシエチレンアルキルエーテルにおけるアルキルとしては、例えば炭素数8~20のアルキル等が挙げられる。炭素数8~20のアルキルとしては、例えば前述の炭素数8~20のアルキル等が挙げられる。
 ポリオキシエチレンアルケニルエーテルにおけるアルケニルとしては、例えば炭素数8~20のアルケニル等が挙げられる。炭素数8~20のアルケニルとしては、例えば前述の炭素数8~20のアルケニル等が挙げられる。
 ポリオキシエチレンアルキルフェニルエーテルにおけるアルキルとしては、例えば炭素数8~20のアルキル等が挙げられる。炭素数8~20のアルキルとしては、例えば前述の炭素数8~20のアルキル等が挙げられる。
 糖化ヘモグロビン含有試料中の糖化ヘモグロビンと、タンパク質分解酵素との反応における反応温度は、通常10~50℃であり、20~40℃が好ましく、反応時間は、通常1分間~3時間であり、2.5分間~1時間が好ましい。タンパク質分解酵素の濃度としては、ヘモグロビン含有試料中の糖化ヘモグロビンと、タンパク質分解酵素との反応が進行する濃度であれば特に制限はなく、通常50~25,000 kU/Lであり、好ましくは250~10,000 kU/Lである。
 タンパク質分解酵素としては、ヘモグロビン含有試料中の糖化ヘモグロビンに作用し、糖化ヘモグロビンから糖化ペプチドを生成させる酵素であれば特に制限はなく、例えばセリンプロテアーゼ(キモトリプシン、スブチリシン等)、システインプロテアーゼ(パパイン、カスパーゼ等)、アスパラギン酸プロテアーゼ(ペプシン、カテプシンD等)、メタロプロテアーゼ(サーモリシン等)、N-末端スレオニンプロテアーゼ、グルタミン酸プロテアーゼ等が挙げられる。本発明においては、市販のタンパク質分解酵素も使用することができ、市販品としては例えばプロテアーゼP「アマノ」3G、プロテアーゼK「アマノ」(以上、天野エンザイム社製)、アクチナーゼAS、アクチナーゼE(以上、科研ファルマ社製)、サーモリシン(大和化成社製)、スミチームMP(新日本化学工業社製)等が挙げられる。
 上記タンパク質分解酵素を作用させる反応における変性剤の濃度としては、糖化ヘモグロビン含有試料中の糖化ヘモグロビンと、タンパク質分解酵素との反応が進行する濃度であれば特に制限はなく、通常0.0001~10%であり、0.0005~5%が好ましい。
 糖化ヘモグロビン含有試料中の糖化ヘモグロビンと、タンパク質分解酵素との反応により、糖化ペプチドが生成する。生成した糖化ペプチドは、糖化ペプチド酸化酵素と反応し、過酸化水素が生成する。糖化ペプチドと糖化ペプチド酸化酵素との反応は、水性媒体中で行われることが好ましい。水性媒体としては後述の水性媒体等が挙げられる。
 糖化ペプチドと糖化ペプチド酸化酵素との反応における反応温度は、通常10~50℃であり、20~40℃が好ましく、反応時間は、通常1分間~3時間であり、2.5分間~1時間が好ましい。糖化ペプチド酸化酵素の濃度としては、フルクトシルヘモグロビンと糖化ペプチド酸化酵素との反応が進行する濃度であれば特に制限はなく、通常0.1~30 kU/Lであり、好ましくは0.2~15 kU/Lである。
 糖化ペプチド酸化酵素としては、糖化ペプチドに作用して過酸化水素を生成させる酵素であれば特に制限はなく、例えば糸状菌由来、酵母由来、放線菌由来、バクテリア由来、古細菌由来の糖化ペプチド酸化酵素等が挙げられる。本発明においては、市販の糖化ペプチド酸化酵素も使用することができ、市販品としては例えばFPOX-CE(キッコーマン社製)、FPOX-EE(キッコーマン社製)、FPOX-CET(キッコーマン社製)等が挙げられる。
 生成した過酸化水素の測定方法としては、例えば電極を用いる方法、過酸化水素測定用試薬を用いる方法等が挙げられ、過酸化水素測定用試薬を用いる方法が好ましい。過酸化水素測定用試薬は、過酸化水素を検出可能な物質に変換するための試薬である。検出可能な物質としては、例えば色素、光(発光)、蛍光等が挙げられ、色素が好ましい。
 検出可能な物質が色素の場合、過酸化水素測定用試薬は、ペルオキシダーゼ等の過酸化活性物質と酸化発色型色原体を含む試薬等が挙げられる。酸化発色型色原体としては、酸化カップリング型色原体、ロイコ型色原体等が挙げられ、ロイコ型色原体が好ましい。ロイコ型色原体としては、例えばフェノチアジン系色原体、トリフェニルメタン系色原体、ジフェニルアミン系色原体、o-フェニレンジアミン、ヒドロキシプロピオン酸、ジアミノベンジジン、テトラメチルベンジジン等が挙げられ、フェノチアジン系色原体が好ましい。フェノチアジン系色原体としては、例えば10-N-カルボキシメチルカルバモイル-3,7-ビス(ジメチルアミノ)-10H-フェノチアジン(CCAP)、10-N-メチルカルバモイル-3,7-ビス(ジメチルアミノ)-10H-フェノチアジン(MCDP)、10-N-(カルボキシメチルアミノカルボニル)-3,7-ビス(ジメチルアミノ)-10H-フェノチアジン ナトリウム塩(DA-67)等が挙げられる。フェノチアジン系色原体の中でも、10-N-(カルボキシメチルアミノカルボニル)-3,7-ビス(ジメチルアミノ)-10H-フェノチアジン ナトリウム塩(DA-67)が特に好ましい。トリフェニルメタン系色原体としては、例えばN,N,N’,N’,N’’,N’’-ヘキサ(3-スルホプロピル)-4,4’,4’’-トリアミノトリフェニルメタン(TPM-PS)等が挙げられる。ジフェニルアミン系色原体としては、例えばN-(カルボキシメチルアミノカルボニル)-4,4’-ビス(ジメチルアミノ)ジフェニルアミン ナトリウム塩(DA-64)、4,4’-ビス(ジメチルアミノ)ジフェニルアミン、ビス〔3-ビス(4-クロロフェニル)メチル-4-ジメチルアミノフェニル〕アミン(BCMA)等が挙げられる。
 検出可能な物質が光(発光)の場合、過酸化水素測定用試薬は、ペルオキシダーゼ等の過酸化活性物質と化学発光物質を含む試薬等が挙げられる。化学発光物質としては、例えばルミノール、イソルミノール、ルシゲニン、アクリジニウムエステル等が挙げられる。
 検出可能な物質が蛍光の場合、過酸化水素測定用試薬は、ペルオキシダーゼ等の過酸化活性物質と蛍光物質を含む試薬等が挙げられる。蛍光物質としては、例えば4-ヒドロキシフェニル酢酸、3-(4-ヒドロキシフェニル)プロピオン酸、クマリン等が挙げられる。
 本発明における、糖化ヘモグロビン含有試料中の糖化ヘモグロビンの測定方法は、糖化ヘモグロビン含有試料中の総ヘモグロビン量に対する糖化ヘモグロビン量の割合として算出する方法も包含する。
 総ヘモグロビン量は、公知の方法、例えばシアンメトヘモグロビン法、オキシヘモグロビン法、SLS-ヘモグロビン法等によって決定することができる。総ヘモグロビン量は、糖化ヘモグロビン含有試料そのもののみならず、糖化ヘモグロビン含有試料に、ハロゲン酸化物と、化合物(I)及び化合物(II)からなる群より選ばれる物質とを添加して得られる試料、糖化ヘモグロビン含有試料に、ハロゲン酸化物、化合物(I)及び化合物(II)からなる群より選ばれる物質、及び、タンパク質分解酵素を添加して得られる試料に対して、シアンメトヘモグロビン法、オキシヘモグロビン法、SLS-ヘモグロビン法を適用することによっても決定することができる。
(2)糖化ヘモグロビン含有試料中の糖化ヘモグロビン測定試薬
 本発明の、糖化ヘモグロビン含有試料中の糖化ヘモグロビン測定試薬は、タンパク質分解酵素、糖化ペプチド酸化酵素、ハロゲン酸化物、化合物(I)及び化合物(II)からなる群より選ばれる物質、及び、過酸化水素測定用試薬を含む試薬である。本発明の糖化ヘモグロビン測定試薬は、本発明のアスコルビン酸の影響抑制方法に使用される。
 本発明の測定試薬におけるタンパク質分解酵素、糖化ペプチド酸化酵素、ハロゲン酸化物、化合物(I)、化合物(II)、及び、過酸化水素測定試薬としては、例えば、それぞれ、前述のタンパク質分解酵素、糖化ペプチド酸化酵素、ハロゲン酸化物、化合物(I)、化合物(II)、及び、過酸化水素測定試薬等が挙げられる。
 本発明の測定試薬におけるタンパク質分解酵素の濃度は、通常50~25,000 kU/Lであり、好ましくは250~10,000 kU/Lである。本発明の測定試薬における糖化ペプチド酸化酵素の濃度は、通常0.1~30 kU/Lであり、好ましくは0.2~15 kU/Lである。
 本発明の測定試薬におけるハロゲン酸化物の濃度は、通常0.005~20 mmol/Lであり、好ましくは0.01~10 mmol/Lである。
 本発明の測定試薬における化合物(I)の濃度は、通常0.005~20 mmol/Lであり、0.01~10 mmol/Lが好ましい。
 本発明の測定試薬における化合物(II)の濃度は、通常0.005~20 mmol/Lであり、0.01~10 mmol/Lが好ましい。
 本発明の測定試薬には、必要に応じて、水性媒体、変性剤、安定化剤、防腐剤、塩類、妨害物質の影響抑制剤、可溶化剤、有機溶媒等が含有されてもよい。
 水性媒体としては、例えば脱イオン水、蒸留水、緩衝液等が挙げられ、緩衝液が好ましい。
 水性媒体のpHは、例えばpH4~10である。水性媒体として緩衝液を用いる場合には、設定するpHに応じた緩衝剤を用いることが望ましい。緩衝液に用いる緩衝剤としては、例えばトリス(ヒドロキシメチル)アミノメタン緩衝剤、リン酸緩衝剤、ホウ酸緩衝剤、グッドの緩衝剤等が挙げられる。
 グッドの緩衝剤としては、例えば2-モルホリノエタンスルホン酸(MES)、ビス(2-ヒドロキシエチル)イミノトリス(ヒドロキシメチル)メタン(Bis-Tris)、N-(2-アセトアミド)イミノ二酢酸(ADA)、ピペラジン-N,N’-ビス(2-エタンスルホン酸)(PIPES)、N-(2-アセトアミド)-2-アミノエタンスルホン酸(ACES)、3-モルホリノ-2-ヒドロキシプロパンスルホン酸(MOPSO)、N,N-ビス(2-ヒドロキシエチル)-2-アミノエタンスルホン酸(BES)、3-モルホリノプロパンスルホン酸(MOPS)、N-〔トリス(ヒドロキシメチル)メチル〕-2-アミノエタンスルホン酸(TES)、2-〔4-(2-ヒドロキシエチル)-1-ピペラジニル〕エタンスルホン酸(HEPES)、3-〔N,N-ビス(2-ヒドロキシエチル)アミノ〕-2-ヒドロキシプロパンスルホン酸(DIPSO)、N-〔トリス(ヒドロキシメチル)メチル〕-2-ヒドロキシ-3-アミノプロパンスルホン酸(TAPSO)、ピペラジン-N,N’-ビス(2-ヒドロキシプロパンスルホン酸)(POPSO)、3-〔4-(2-ヒドロキシエチル)-1-ピペラジニル〕-2-ヒドロキシプロパンスルホン酸(HEPPSO)、3-〔4-(2-ヒドロキシエチル)-1-ピペラジニル〕プロパンスルホン酸[(H)EPPS]、N-〔トリス(ヒドロキシメチル)メチル〕グリシン(Tricine)、N,N-ビス(2-ヒドロキシエチル)グリシン(Bicine)、N-トリス(ヒドロキシメチル)メチル-3-アミノプロパンスルホン酸(TAPS)、N-シクロヘキシル-2-アミノエタンスルホン酸(CHES)、N-シクロヘキシル-3-アミノ-2-ヒドロキシプロパンスルホン酸(CAPSO)、N-シクロヘキシル-3-アミノプロパンスルホン酸(CAPS)等が挙げられる。
 緩衝液の濃度は、通常0.001~2.0 mol/Lであり、0.005~1.0 mol/Lが好ましい。
 変性剤としては、例えば前述の変性剤等が挙げられる。本発明の測定試薬における変性剤の濃度は、通常0.0001~10%であり、0.0005~5%が好ましい。
 安定化剤としては、例えばエチレンジアミン四酢酸(EDTA)、シュークロース、塩化カルシウム、酢酸カルシウム、硝酸カルシウム、フェロシアン化カリウム、牛血清アルブミン(BSA)等が挙げられる。防腐剤としては、例えばアジ化ナトリウム、抗生物質等があげられる。塩類としては、例えば塩化ナトリウム、硝酸ナトリウム、硫酸ナトリウム、炭酸ナトリウム、塩化カリウム、硝酸カリウム、硫酸カリウム、炭酸カリウム等が挙げられる。妨害物質の影響抑制剤としては、例えば過酸化物の影響を抑制するためのピルビン酸等が挙げられる。可溶化剤としては、例えばポリオキシエチレンアルキルエーテルやポリオキシエチレンアルキルフェニルエーテル等の非イオン性界面活性剤等が挙げられる。有機溶媒としては、例えばロイコ型色原体の水性媒体への溶解補助のためのジメチルホルムアミド(DMF)、ジメチルスルホキサイド(DMSO)、ジオキサン、アセトン、メタノール、エタノール等が挙げられる。
 本発明の糖化ヘモグロビン測定試薬は、キットの形態を取ってもよい。糖化ヘモグロビン測定キットとしては、例えば2試薬系キット、3試薬系キット等が挙げられる。糖化ヘモグロビン測定キットが2試薬系キットである場合、タンパク質分解酵素、糖化ペプチド酸化酵素、ハロゲン酸化物、化合物(I)及び化合物(II)からなる群より選ばれる物質、及び、過酸化水素測定用試薬の各要素の配置は、キットの保存安定性の観点から、当業者が適宜、選択することができるが、タンパク質分解酵素及び糖化ペプチド酸化酵素が別々の試薬に含まれるキットが好ましい。
 以下、実施例により本発明をより詳細に説明するが、これらは本発明の範囲を何ら限定するものではない。尚、本実施例においては、下記メーカーの試薬及び酵素を使用した。
 Bis-Tris(同仁化学研究所社製)、MOPS(同仁化学研究所社製)、塩化カルシウム2水和物(関東化学社製)、ピルビン酸ナトリウム(関東化学社製)、DMSO(ナカライテスク社製)、DA-67(和光純薬工業社製)、トリトンX-405(非イオン性界面活性剤;シグマアルドリッチ社製)、1-ドデシルピリジニウムクロライド(C12py)(変性剤;東京化成社製)、テトラデシルトリメチルアンモニウムブロマイド(C14TMA)(変性剤;東京化成社製)、ヨウ素酸カリウム(ハロゲン酸化物;東京化成社製)、臭素酸カリウム(ハロゲン酸化物;東京化成社製)、2-オクチル-4-イソチアゾリン-3-オン[化合物(I);ケミクレア社製]、プロクリン950{2-メチル-4-イソチアゾリン-3-オン[化合物(I)]の9.5%水溶液;シグマアルドリッチ社製}、2-メチルチアゾリン[化合物(II);東京化成社製]、アクチナーゼE(タンパク質分解酵素;科研製薬社製)、FPOX-CET(糖化ペプチド酸化酵素;キッコーマン社製)、ペルオキシダーゼ(東洋紡績社製)。
 以下の第1試薬及び第2試薬からなる、第1表に示すHbA1c測定キット(キット1~3;キットa~e)を調製した。

 HbA1c測定キットとして、実施例1のキット1を、試料として、糖尿病が疑われる16名の被験者由来の全血を用いて、以下の手順により、各試料におけるHbA1c濃度(量)の総ヘモグロビン濃度(量)に対する割合[HbA1c(%)]を決定した。
(1)総ヘモグロビン濃度決定のための検量線の作成
 測定キットとして、総ヘモグロビン測定用キットであるネスコートヘモキット-N(シアンメトヘモグロビン法)(アルフレッサファーマ社製)を用いて、検体として、ネスコートヘモキット-Nに付属の標準液(ヘモグロビン濃度:15.3 g/dL)を用いて、ヘモグロビン濃度と吸光度との関係を示す検量線を作成した。
(2)HbA1c濃度決定のための検量線の作成
 ラテックス免疫凝集法と、血球画分の総ヘモグロビン値とから、HbA1c濃度が2.97 μmol/L、7.80 μmol/Lと値付けされた2つの血球画分について、実施例1のキット1を用いて測定し、各血球画分に対する吸光度を測定した。当該血球画分の代わりに生理食塩水を用いて、生理食塩水に対するHbA1c濃度を測定した。当該血球画分に対するそれぞれの吸光度から、生理食塩水に対する吸光度を差し引いて算出した値を、当該血球画分に対するブランク補正吸光度とした。当該血球画分に対するブランク補正吸光度と、生理食塩水に対するブランク補正吸光度(0 Abs)とから、HbA1c濃度(μmol/L)と吸光度との間の関係を示す検量線を作成した。
(3)各血球画分におけるヘモグロビン濃度の決定
 各試料に対して、25℃、3,000 rpmで5分間遠心分離を行い、血球画分を得た。各血球画分について、ネスコートヘモキット-Nを用いて測定し、得られた測定値と(1)の検量線とから、各血球画分中のヘモグロビン濃度(μmol/L)を決定した。
(4)各血球画分におけるHbA1c濃度の決定
 各血球画分について、実施例1のキット1を用いて測定し、得られた測定値と(2)の検量線とから、各血球画分中のHbA1c濃度(μmol/L)を決定した。
(5)HbA1c(%)(=HbA1c濃度のヘモグロビン濃度に対する割合)の決定
 上記(3)で決定した各血球画分におけるヘモグロビン濃度(μmol/L)と、上記(4)で決定した各血球画分におけるHbA1c濃度(μmol/L)とから、以下の式により、日本糖尿病学会(Japan Diabetes Society;JDS)値のHbA1c(%)を算出した。

(6)同一血球画分中のHbA1c(%)の免疫測定法での決定
 上記(5)でのHbA1c(%)の決定に使用した血球画分と同一の血球画分を用いて、各血球画分中のHbA1c(%)を、デタミナーL HbA1c(協和メデックス社製)を用いる免疫測定法により、デタミナーL HbA1cの添付文書に記載の方法に従って決定した。
(7)本発明の測定キットを用いる測定方法と免疫測定法との相関
 本発明の測定キットを用いる測定方法により、上記(5)で決定したHbA1c(%)と、免疫測定法を用いて、上記(6)で決定したHbA1c(%)とから、本発明の測定方法と免疫測定法との間の相関関係を検証し、相関係数を決定した。
 同様に、実施例1のキット1の代わりに、実施例1のキット2~3、キットa~e、実施例2のキット4~6、キットf~j、実施例3のキット7~8、キットkの各キットを用いて、デタミナーL HbA1c(協和メデックス社製)を用いた測定との間の相関係数を決定した。その結果を第4表に示す。

 第4表に示す通り、実施例1~3の各キットを用いるHbA1cの測定方法と、免疫測定法との間に良好な相関関係が認められた。従って、実施例1~3の各キットを用いることにより、試料中のHbA1cを正確に測定できることを確認した。
 HbA1c測定キットとして、実施例1のキット1を、試料として、以下に方法により調製されたアスコルビン酸の影響確認用試料を用いて、以下の手順により、各試料におけるHbA1c濃度(μmol/L)を決定した。
(1)アスコルビン酸の影響確認用試料の調製
 ヒト血液を遠心分離して得られた血球画分について、総ヘモグロビン測定用キットであるネスコートヘモキット-N(シアンメトヘモグロビン法)(アルフレッサファーマ社製)を用いてヘモグロビン濃度を決定した後、当該血球画分を精製水で希釈すると共に、溶血させて、ヘモグロビン濃度6.4 mg/mLのヘモグロビン溶液を調製した。
 調製した当該ヘモグロビン溶液にアスコルビン酸を添加し、アスコルビン酸濃度が0.5 mg/dL, 1 mg/dL, 2 mg/dL、及び、5 mg/dLの各ヘモグロビン溶液を調製し、アスコルビン酸の影響確認用試料とした。また、調製した当該ヘモグロビン溶液そのものを、アスコルビン酸濃度が0 mg/mLであるアスコルビン酸の影響確認用試料とした。
(2)HbA1c濃度決定のための検量線の作成
 実施例4の(2)の方法により、HbA1c濃度(μmol/L)と吸光度との間の関係を示す検量線を作成した。
(3)アスコルビン酸の影響確認用試料中のHbA1c濃度の決定
 上記(1)で調製したアスコルビン酸の影響確認用試料の各試料について、実施例1のキット1を用いて測定し、得られた測定値と(2)の検量線とから、各試料中のHbA1c濃度(μmol/L)を決定した。アスコルビン酸濃度が0 mg/dLであるアスコルビン酸の影響確認用試料におけるHbA1c濃度(μmol/L)を100としたときの、アスコルビン酸濃度が0.5 mg/dL、1 mg/dL、2 mg/dLであるアスコルビン酸の影響確認用試料の各試料中のHbA1c濃度(μmol/L)の相対値を決定した。
 実施例1のキット1の代わりに、実際例1のキット2~3、キットa~eの各キットを用いて同様の手順により測定を行い、各試料におけるHbA1c濃度(μmol/L)の相対値を決定した。決定された相対値を第5表に示す。相対値が100に近い程、アスコルビン酸の影響を受け難いことを意味する。

 第5表より明らかなように、ヨウ素酸カリウムと、化合物(I)又は化合物(II)とを含むキット1~3においては、ヨウ素酸カリウム、及び、化合物(I)又は化合物(II)の少なくとも1つを含まないキット(キットa~e)に比較して、HbA1c濃度の相対値が100に近く、アスコルビン酸の影響を受け難いことが分かる。
 実施例1のキット1の代わりに、実施例2のキット4~6、キットf~jの各キットを用いる以外は実施例5と同様の方法により、アスコルビン酸の影響確認用試料の各試料におけるHbA1c濃度(μmol/L)の相対値を決定した。決定されたHbA1c濃度(μmol/L)の相対値を第6表に示す。

 第6表より明らかなように、ヨウ素酸カリウムと、化合物(I)又は化合物(II)とを含むキット4~6においては、ヨウ素酸カリウム、及び、化合物(I)又は化合物(II)のいずれか又は両方を含まないキット(キットf~j)に比較して、HbA1c濃度の相対値が100に近く、アスコルビン酸の影響を受け難いことが分かる。
 実施例1のキット1の代わりに、実施例3のキット7~8、キットk、g~jの各キットを用いる以外は実施例5と同様の方法により、アスコルビン酸の影響確認用試料の各試料におけるHbA1c濃度(μmol/L)の相対値を決定した。決定されたHbA1c濃度(μmol/L)の相対値を第7表に示す。

 第7表より明らかなように、臭素酸カリウムと、化合物(I)又は化合物(II)とを含むキット7~8においては、臭素酸カリウム、及び、化合物(I)又は化合物(II)のいずれか又は両方を含まないキット(キットk、g~j)に比較して、HbA1c濃度の相対値が100に近く、アスコルビン酸の影響を受け難いことが分かる。
 従って、ハロゲン酸化物と、化合物(I)又は化合物(II)とを含むキットを用いたHbA1cの測定においては、アスコルビン酸の影響が顕著に抑制されていることが分かる。
 本発明により、糖化ヘモグロビン含有試料中の糖化ヘモグロビンの測定におけるアスコルビン酸の影響抑制方法、糖化ヘモグロビン含有試料中の糖化ヘモグロビン測定用試薬が提供される。本発明のアスコルビン酸の影響抑制方法、及び、糖化ヘモグロビン測定用試薬は、糖尿病の診断等に有用である。
 

Claims (7)

  1.  糖化ヘモグロビン含有試料にタンパク質分解酵素を作用させた後、生成した糖化ペプチドを糖化ペプチド酸化酵素と反応させ、生成した過酸化水素を測定することにより、試料中の糖化ヘモグロビンを測定する方法において、糖化ペプチドと糖化ペプチド酸化酵素との反応を、ハロゲン酸化物と、一般式(I)

    (式中、Rは置換若しくは非置換のアルキルを表す)で表される物質、及び、一般式(II)

    (式中、Rは置換若しくは非置換のアルキルを表し、---は単結合又は二重結合を表す)で示される物質からなる群より選ばれる物質との存在下に行うことを特徴とする、試料中の糖化ヘモグロビンの測定におけるアスコルビン酸の影響抑制方法。
  2.  ハロゲン酸化物が、ヨウ素酸若しくはその塩、及び、臭素酸若しくはその塩からなる群より選ばれるハロゲン酸化物である、請求項1記載の方法。
  3.  糖化ヘモグロビンが、ヘモグロビンA1cである請求項1又は2記載の方法。
  4.  タンパク質分解酵素、糖化ペプチド酸化酵素、ハロゲン酸化物、一般式(I)

  5.  ハロゲン酸化物が、ヨウ素酸若しくはその塩、及び、臭素酸若しくはその塩からなる群より選ばれるハロゲン酸化物である、請求項4記載の試薬。
  6.  糖化ヘモグロビンが、ヘモグロビンA1cである請求項4又は5記載の試薬。
  7.  試料中の糖化ヘモグロビンの測定におけるアスコルビン酸の影響を抑制するための請求項4~6のいずれかに記載の試薬。
