천연 폴리페놀 화합물 함유 조성물 및 경구 섭취용 조성물이 개시된다. 보다 상세하게는, 수분산성 또는 수용해성이 우수한 천연 폴리페놀 화합물 함유 조성물 및 이를 포함하는 경구 섭취용 조성물이 개시된다.
세계적인 웰빙 트렌드의 확산과 고령화의 영향으로 인하여 세계 건강기능식품 시장의 규모는 최근 5년간 연평균 6% 수준의 고성장세를 지속하고 있다. 이러한 건강기능식품 시장의 높은 성장세는 「건강 지향적 소비자」의 증가에 따라 지속되고, 나아가 더 확대될 것으로 예상된다. 또한, 우리나라 건강기능식품 시장의 규모도 빠른 추세로 증가하고 있어 건강기능식품 시장에 대한 중요성이 점차 커지고 있는 실정이다.
또한, 종래에는 단일 기능성 제품이 주를 이루었으나 최근에는 단일 기능성 제품 중심에서 벗어나 다기능성 제품으로 시장이 확대되거나, 다양한 건강기능식품이 등장하는 등 패러다임이 변화하고 있다. 이러한 건강기능식품에는 자연 상태에서 얻은 천연물이 다수 포함되어 있다. 그러나, 대다수의 천연물은 강력한 항산화 기능 등 다양한 효능을 갖고 있기는 하지만, 물에 대한 용해도가 매우 낮아서 경구 섭취시 생체이용률이 떨어지는 문제점이 있다.
본 발명의 일 구현예는 수분산성 또는 수용해성이 우수한 천연 폴리페놀 화합물 함유 조성물을 제공한다.
본 발명의 다른 구현예는 상기 천연 폴리페놀 화합물 함유 조성물을 포함하는 경구 섭취용 조성물을 제공한다.

본 발명의 일 구현예에 따른 천연 폴리페놀 화합물 함유 조성물은 수분산성 또는 수용해성이 우수하여, 경구 섭취용 조성물 분야에 이용될 경우 생체이용률이 높은 이점을 갖는다.
이하에서는 본 발명의 일 구현예에 따른 천연 폴리페놀 화합물 함유 조성물을 상세히 설명한다.
본 발명의 일 구현예에 따른 천연 폴리페놀 화합물 함유 조성물은 수난용성 천연 폴리페놀 화합물(PSNPPC: poorly water-soluble natural polyphenolic compound) 및 사포닌을 포함하고, 수분산성(water dispersibility) 또는 수용해성(water solubility)을 갖는다. 본 명세서에서, 「수분산성」이란 상기 수난용성 천연 폴리페놀 화합물 함유 조성물이 수중에서 에멀젼 상태로 존재하는 성질을 의미하고, 「수용해성」이란 상기 천연 폴리페놀 화합물 함유 조성물이 수중에서 용액 상태로 존재하는 성질을 의미한다.
상기 수난용성 천연 폴리페놀 화합물은 0.004~0.087의 용해도를 갖는 것일 수 있다. 본 명세서에서. 「용해도」란 인간의 체온과 유사한 37℃의 물 1mL에 용해되는 용질(수난용성 천연 폴리페놀 화합물)의 양(mg)을 의미한다.
상기 수난용성 천연 폴리페놀 화합물은 생리조절활성기능(physiological function)을 갖는 것일 수 있다.
상기 수난용성 천연 폴리페놀 화합물은 커큐민(curcumin), 실리마린(silymarin), 레스베라트롤(resveratrol) 또는 이들의 조합을 포함할 수 있다. 상기 커큐민은 강황뿌리 추출물에서 유래된 것일 수 있고, 상기 실리마린은 밀크씨슬(milk thistle) 추출물에서 유래된 것일 수 있으며, 상기 레스베라트롤은 포도류 등의 추출물에서 유래된 것일 수 있다.
상기 사포닌은 면역력 강화 기능 및 신진대사 촉진 기능과 같은 생리조절활성기능을 가지고 있을 뿐만 아니라, 상기 수난용성 천연 폴리페놀 화합물의 수분산성 또는 수용해성을 증가시키는 역할을 수행한다.

상기 사포닌의 함량은 상기 수난용성 천연 폴리페놀 화합물 100중량부에 대하여 25~400중량부일 수 있다. 상기 사포닌의 함량이 상기 범위이내이면, 상기 수난용성 천연 폴리페놀 화합물의 수분산성 또는 수용해성을 높여 그의 생체이용률을 향상시킬 수 있다.
상기와 같은 구성을 갖는 본 발명의 일 구현예에 따른 천연 폴리페놀 화합물 함유 조성물은 물에 분산되거나 용해될 경우 수난용성 천연 폴리페놀 화합물이 물리화학적으로 변화되지 않고 유효성분을 그대로 보존할 수 있으며, 제조방법이 간단하기 때문에 다양한 용도로 사용될 수 있다.
본 발명의 다른 구현예는 상기 천연 폴리페놀 화합물 함유 조성물을 포함하는 경구 섭취용 조성물을 제공한다.
상기 경구 섭취용 조성물은 정제, 캡슐제, 액제 또는 음료의 형태로서, 건강기능식품 또는 의약품일 수 있다. 본 명세서에서, "액제"는 일반적인 액상 형태 또는 점조성을 나타낼 수 있는 액상제제로서 통상 10~30mL 용량의 앰플이나 바이얼 형태의 용기에 담겨 유통되는 것을 의미하고, "음료"는 일반 드링크 형태로서 통상 75~120mL 용량의 용기에 담겨 유통되는 것을 의미한다.
상기 정제는 직접 타정법(direct compression)과 같은 통상적인 제조방법에 의해 제조될 수 있다. 구체적으로, 상기 정제는 상기 경구 섭취용 조성물을 균질하게 혼합한 후 정제 성형 압축기로 압축하여 제조될 수 있다.
상기 캡슐제는 상기 경구 섭취용 조성물을 균질하게 혼합한 후, 경질 캡슐에 충진함으로써 제조될 수 있다.
상기 액제 및 음료 등은 상기 경구 섭취용 조성물을 정제수에 투입하여 특유의 향과 감미가 있는 음료를 제조하고, 그 결과물을 용기에 충전하고 포장하여 쉽게 음용이 가능하게 할 수 있다.
상기 경구 섭취용 조성물 중 상기 사포닌의 함량은 0.4~50중량%일 수 있다. 상기 사포닌의 함량이 상기 범위이내이면, 상기 수난용성 천연 폴리페놀 화합물의 생체이용률을 높일 수 있으며, 우수한 생리조절활성기능을 제공할 수 있는 경구 섭취용 조성물을 얻을 수 있다.
이하, 실시예들을 들어 본 발명에 대하여 더욱 상세히 설명하지만, 본 발명이 이러한 실시예들에 한정되는 것은 아니다.
실시예
실험예 1: 수난용성 천연 폴리페놀 화합물(PSNPPC)의 용해도 측정
3종의 수난용성 천연 폴리페놀 화합물(프랑스의 NATUREX사 제품)의 물에 대한 용해도를 각각 측정하여, 그 결과를 하기 표 1에 나타내었다.

실험예 2: 첨가제 첨가시 수난용성 천연 폴리페놀 화합물(PSNPPC)의 용해도
3종의 수난용성 천연 폴리페놀 화합물(PSNPPC)에 11종의 첨가제를 하기 표 2에서와 같이 1:0.25(624mg : 156mg), 1:1(625mg : 625mg) 또는 1:4(625mg : 2500mg)의 비율로 혼합하여 분말상의 혼합물을 제조한 후, 상기 혼합물을 45mL의 물에 투입하여 교반하였다. 이후, 상기 결과물을 약 5ml 취한 후, 이를 여과하고 적절히 희석하여 고성능 액체 크로마토그래피법(HPLC) 분석용 검액으로 사용하였다. 즉, 상기 검액의 구성성분 및 각 구성성분의 함량을 HPLC로 정량하였다. 이후, HPLC로 검출된 수난용성 천연 폴리페놀 화합물의 함량을 이용하여 물 1mL에 대한 각 화합물의 용해도를 계산하였다. 또한, 상기 용해도 측정결과를 하기 표 2에 나타내었다. 하기 표 2에 열거된 첨가제의 출처는 다음과 같다: 차 사포닌 (Hunan Amazing Grace biotechnology Co., Ltd., China), 서양인삼 사포닌 (Ningbo Dekang Biochem Co., Ltd., China), 고려인삼 엽사포닌　(Ningbo Dekang Biochem Co., Ltd., China), 고려인삼 근사포닌 (Hong-ziu Biotech Co., Ltd., China), 대두 사포닌 (Xi'an Greenherb Biotech Co., Ltd., China), PEG 400 (Samchun pure chemical, Korea), Tween 80 (Sigma Aldrich, U.S.A), β-Aescin amorphors (Nuri PharmTech Co., Ltd., Korea), Hawthorn Fruit P.E. (Shannxi M.R Natural product Co., Ltd., China), Quillaja saponaria saponin (Xi'an sinuote Bio-tech Co., Ltd., China), Tribulus terrestris saponin (Ningbo Tianhong Biotech Co., Ltd., China).

상기 표 2 및 표 1을 참조하면, 차 사포닌, 서양인삼 사포닌, 고려인삼 엽사포닌, 고려인삼 근사포닌 및 대두 사포닌을 첨가한 경우가 Tween®80을 제외한 다른 첨가제를 첨가한 경우에 비해 수난용성 천연 폴리페놀 화합물의 용해도(즉, 물에 대한 용해도) 증가 효과가 우수한 것으로 나타났다. Tween®80은 용해도 개선에는 효과적이지만, 생리조절활성기능을 갖지 않는다.
실시예 1: 정제의 제조
레스베라트롤 및 고려인삼 근사포닌 등을 사용하여 하기 표 3의 조성을 갖는 정제를 제조하였다.

Claims (9)

  1. 수난용성 천연 폴리페놀 화합물; 및
    사포닌을 포함하고,
    수분산성 또는 수용해성을 갖는 천연 폴리페놀 화합물 함유 조성물.
  2. 제1항에 있어서,
    상기 수난용성 천연 폴리페놀 화합물은 0.004~0.087의 용해도를 갖는 것인 천연 폴리페놀 화합물 함유 조성물.
  3. 제2항에 있어서,
    상기 수난용성 천연 폴리페놀 화합물은 커큐민(curcumin), 실리마린(silymarin), 레스베라트롤(resveratrol) 또는 이들의 조합을 포함하는 천연 폴리페놀 화합물 함유 조성물.
  4. 제1항에 있어서,
    상기 사포닌은 차 사포닌(tea saponin), 서양인삼 사포닌(American ginseng saponin), 고려인삼 엽사포닌(Panax ginseng saponin(leaf)), 고려인삼 근사포닌(Panax ginseng saponin(root)), 대두 사포닌(Soy saponin) 또는 이들의 조합을 포함하는 천연 폴리페놀 화합물 함유 조성물.
  5. 제1항에 있어서,
    상기 천연 폴리페놀 화합물 함유 조성물의 용해시, 상기 수난용성 천연 폴리페놀 화합물의 용해도는 0.006~1.087인 천연 폴리페놀 화합물 함유 조성물.
  6. 제5항에 있어서,
    상기 사포닌의 함량은 상기 수난용성 천연 폴리페놀 화합물 100중량부에 대하여 25~400중량부인 천연 폴리페놀 화합물 함유 조성물.
  7. 제1항 내지 제6항 중 어느 한 항에 따른 천연 폴리페놀 화합물 함유 조성물을 포함하는 경구 섭취용 조성물.
  8. 제7항에 있어서,
    상기 경구 섭취용 조성물은 정제, 캡슐제, 액제 또는 음료의 형태로서, 건강기능식품 또는 의약품인 경구 섭취용 조성물.
  9. 제7항에 있어서,
    상기 경구 섭취용 조성물 중 상기 사포닌의 함량은 0.4~50중량%인 경구 섭취용 조성물.
