Definitions
- the present invention relates to new, multi-part, capsules for pharmaceutical preparations intended for oral administration of two incompatible drugs.
- Capsules with pharmaceuticals are widely used in the treatment and diagnosis of diseases.
- Hard capsules or bivalves consist of two cylindrical parts telescopically inserted into each other: a body with a hemispherical base and a lid of the same shape, but shorter. The inner diameter of the lid is actually equal to the outer diameter of the housing. When connected, two parts make up a container of standard sizes that are filled after passing through the entire molding process cycle and gaining rigidity.
- Capsules are usually made from gelatin.
- the capsules are made from well-digestible water-soluble plastics by humans, which, when taken orally, release the active ingredients in certain sections of the gastrointestinal tract.
- EP 0 460 921 describes two-part capsules of chitosin and starch, cereal flour, oligosaccharides, methyl acrylate, methacrylic acid, acetate, succinate or hydroxypropyl methyl cellulose phthaleate.
- the material of the capsule is characterized in that its contents are released only in the large intestine.
- GB 938828 describes capsules for radioactive substances used in therapy or diagnosis. Such capsules are made from water-soluble gelatin, methyl cellulose, polyvinyl alcohol or non-toxic water-soluble thermoplastics.
- RU 961 15307 A (10.27.1998 PCT GB 94/02703, A61 K 9/48) claims a pharmaceutical form in the form of two-phase capsules comprising a capsule having one compartment containing at least two filling compositions, characterized in that it is prevented mixing compositions with each other.
- the two fill compositions are separated by a physical partition that contains an additional fill composition of a material different from the material of the capsule.
- the material of the partition contains a material having a temperature melting 25 ° C or 37 ° C.
- the pharmaceutical form may include a first filling composition containing a pharmaceutically active substance for a short release during non-parenteral administration, and a second filling composition containing a pharmaceutically active substance for a sustained release during non-parenteral administration.
- the pharmaceutical form may include a first filling composition containing a biologically active substance, and a second filling composition containing a substance adapted to adjust the intestinal pH to a pH of from 7.5 to 9.
- an encapsulated drug is claimed.
- the capsule body has inside two or more compartments separated by a partition for pharmaceutical compositions.
- the closest constructive analogue to the invention is a capsule [SU 1637654, (US 3647.491 / 14), A 61 J 1/06, A61 K 9/48, A61 K 9/58, 03.23.91) having a hard shell made in the form telescopically connected to the cap and housing, having side cylindrical walls, an open end and a closed end region.
- the cylindrical surfaces of the side walls of the lid and the casing are of equal length for the inner surface of the side wall of the lid to overlap the outer surface of the side wall of the casing, and the region of the closed end has a conical or hemispherical shape.
- the disadvantage of the capsules cited in the prior art is that they provide release of the contents of the capsules in one particular section of the gastrointestinal tract.
- the objective of the invention is to isolate incompatible pharmaceuticals in one capsule, which can provide a sustained release of a particular pharmaceutical in the right part of the gastrointestinal tract.
- the claimed capsule for two incompatible drugs having a hard shell made in the form of telescopically connected two parts of the body and cover having cylindrical side walls, an open end and a closed end region, the cylindrical surfaces of the side walls being of equal length and the closed end region is hemispherical, which according to the invention is additionally provided with a partition made with a bottom for overlapping the open end of the housing, made with a protrusion, the outer diameter of which is equal to the inside the diameter of the septum, the case, the lid and the septum are gastro-soluble or enteric-soluble with a prolonged release of the drug of one of the capsule parts, or one part of the capsule is gastro-soluble and the other is enteric-soluble.
- Fig.1 shows a capsule, General view
- figure 2 is the same, a longitudinal section
- in Fig.Z - cover in longitudinal section
- figure 4 - body in longitudinal section
- figure 5 is a partition, in longitudinal section.
- the capsule has a hard shell (figure 1), made in the form of telescopically connected coaxial cover 1, the housing 2 and the partition 3.
- the cover 1 has a lateral cylindrical wall 4 (Fig.Z), the open end 5, the region 6 of the closed end.
- the housing 2 (FIG. 4) has a cylindrical side wall 7 (FIG. 3), an open end 8, a closed end region 9.
- the cylindrical walls 4 and 7 have surfaces of equal length for loading the drug into both parts of the capsule. Areas 6 and 9 of the closed ends of the lid and housing are hemispherical.
- the housing 2 from the side of the open end is made with a protrusion, the outer diameter of which is equal to the inner diameter of the partition 3.
- the cover 1, the housing 2 and the partition are telescopically coaxially connected to each other.
- the capsule is made in the generally accepted way by immersing in the gelatin solution fingers, having the form of a cover, a body and a partition for drying gelatin on the fingers, followed by cutting the blanks of the desired length.
- the material for the manufacture of the capsule may be other pharmaceutically acceptable hydrophilic polymers.
- a plasticizer sorbitol
- essential oils for aesthetic perception - dyes or flavoring substances.
- acetylphthalyl cellulose is introduced into the capsule mass or an enteric film coating is applied to the surface of the capsule.
- Prologged hard gelatine capsules are prepared by introducing special ingredients (acrylic polymers, cellulose derivatives) into the product filler. Used combinations of substances prevent the rapid release of the active components of the capsule.
- the capsule is filled as follows.
- the housing 1 is filled with a metered amount of one medicine, tightly telescopically coaxially fit a septum 3 on it with the open end, then the lid is filled with a metered amount of another incompatible medicine and the housing is sealed telescopically coaxially coaxially with a sealed end. If additional sealing is necessary, the lid is glued to the capsule body (with a melted gelatin tape or with a solution of polyvinyl alcohol).
- the capsule for two incompatible drugs may be gastro-soluble (gastro-soluble) or enteric-soluble (enteric) with a prolonged release of one of its parts, or gastro-soluble one part and enteric-soluble other.
