Abstract
Description
従って、1-ケストースをヒトや動物に摂取させて、腸内の酪酸の濃度や量、あるいは酪酸産生菌の数を増加させることにより、腸内炎症を予防または治療することができる。また、1-ケストースは、腸内炎症の治療または予防用医薬品を製造するために使用することができる。
(1)1-ケストースを経口摂取させたラットの飼育
1-ケストースを0、0.5、1.0、2.5および5質量%となるように配合した飼料を日本クレア社に委託して調製した。飼料の組成を下記に示す。40匹のSDラット(日本エスエルシー社)を8匹ずつ5つの群に分け、A~E群とした。A群には1-ケストースを配合しない飼料を、B~E群には1-ケストースを配合した飼料をそれぞれ自由摂取させながら、30日間飼育した。飼育条件は温度23±1℃、明期12時間(8:00~20:00)および暗期12時間(20:00~8:00)とした。
本実施例1(1)の各群のラットを解剖して盲腸を摘出し、盲腸の内容物を採取して重量を測定した。次に、盲腸内容物における短鎖脂肪酸(酪酸、プロピオン酸、イソ酪酸、酢酸、イソ吉草酸および吉草酸)の濃度を、ガスクロマトグラフ質量分析計(GC/MS)を用いて測定した。また、短鎖脂肪酸濃度の測定結果を基に、盲腸内容物における短鎖脂肪酸の量を算出した。続いて、各群毎に短鎖脂肪酸濃度および短鎖脂肪酸量の平均値を算出して、棒グラフに表した。その結果を図1~図3に示す。

実施例1(1)のA群およびE群のラットの盲腸内容物について、下記の4種の微生物の量をリアルタイムPCR法により測定した。
1.ビフィドバクテリウム(Bifidobacterium)属、
2.クロストリジウム クラスター14a(Closrtidium cluster XIVa)およびクロストリジウム クラスター14b(Closrtidium cluster XIVb)(酪酸産生菌群)、
3.ラクトバシラス(Lactobacillus)属、
4.アッカーマンシア・ムシニフィラ(Akkermansia muciniphila)。

Claims (11)
- 1-ケストースを有効成分とする腸内酪酸増加剤。
- 1日あたり0.04g/kg体重以上の1-ケストースがヒトまたは動物に対して摂取されるように用いられることを特徴とする、請求項1に記載の腸内酪酸増加剤。
- 腸内炎症、脂肪肝、糖尿病、大腸癌または肥満の予防または治療に用いられることを特徴とする、請求項1または請求項2に記載の腸内酪酸増加剤。
- 1-ケストースを有効成分とする腸内酪酸増加用食品組成物。
- 1-ケストースを有効成分とする酪酸産生菌増殖剤。
- 1-ケストースを有効成分とする酪酸産生菌増殖用食品組成物。
- ヒトまたは動物に1-ケストースを摂取させることにより前記ヒトまたは動物の腸内における酪酸の量または濃度を増加させる工程を有する、腸内酪酸の増加方法。
- ヒトまたは動物に1-ケストースを摂取させることにより前記ヒトまたは動物の腸内における酪酸産生菌の数を増加させる工程を有する、酪酸産生菌の増殖方法。
- 酪酸産生菌を含む培地に1-ケストースを添加して培養する工程を有する、酪酸産生菌の増殖方法。
- 腸内炎症、脂肪肝、糖尿病、大腸癌もしくは肥満を罹患している、または、罹患する可能性があるヒトもしくは動物に、1-ケストースを摂取させることにより、前記ヒトもしくは動物の腸内における酪酸の量もしくは濃度または酪酸産生菌の数を増加させる工程を有する、腸内炎症、脂肪肝、糖尿病、大腸癌または肥満の治療または予防方法。
- 腸内炎症、脂肪肝、糖尿病、大腸癌または肥満の治療または予防用医薬品を製造するための1-ケストースの使用。
