Definitions

  • the present invention relates to a low molecular collagen peptide and a method for producing a food composition using the same, and more particularly, a method for producing a low molecular collagen peptide utilizing a weak acid containing hydrogen calcium and a companion containing the low molecular collagen peptide prepared by the method It relates to a method for producing a composition for animal nutrition.
  • Collagen is a protein found in most animals and is a major protein that makes up the flesh and connective tissue of mammals in particular. It is rich in 25% to 35% of the proteins that make up the body.
  • the increased fibrous collagen is mainly present in fibrous tissues such as tendons, ligaments and skin, and also exists in the cornea, cartilage, bone, blood vessels, digestive tract, and spinal disc.
  • Collagen acts as a major component of the endocardium.
  • Collagen has a molecular weight of 300,000 and three ⁇ -molecules with a molecular weight of 100,000 form a ⁇ -ray. It is a triple helix structure of 1.5 nm in diameter and 300 nm in length. ⁇ chains are strongly bonded by some hydrogen bonds, and when microfibrils are gathered, they become collagen fibrils by covalent cross-linking, and they become fibers.
  • Collagen like other proteins, the structure, synthesis amount, etc. are determined according to the genetic information. Molecular biology and biochemistry studies on collagen have recently made significant advances and their structure has been elucidated in detail. Searching the literature on the topic of collagen reveals that more than 10,000 publications are published annually. To date, more than 13 types have been reported, depending on the type of molecule, and more than 22 types of polypeptide chains. In general, the difference with other proteins is that collagen protein does not function as a single molecule, and only plays a role in the living body when many molecules are combined to form a higher structure.
  • Collagen is also a protein that is the largest quantitative and functional part of the human body, meaning the extracellular matrix, more specifically the major constituent of connective tissue. do.
  • Human tissues containing large amounts of collagen include bone tissue such as skin, bone and cartilage, tendons, ligaments, and the like. Aging of such tissues causes human aging diseases such as wrinkles and sagging skin, and osteoporosis, which is known to be due to a decrease in the amount and function of collagen in the body due to a decrease in collagen synthesis of tissues.
  • a feature of the collagen amino acid composition is that glycine makes up about one third of the total.
  • 2/9 of the whole is composed of proline or hydroxyproline (Hydroxy-proline). Hydroxyproline is produced when proline on the translational peptide of the collagen gene is hydroxylated at position 4 carbon by molecular oxygen.
  • the content of amino acids depends on the type of animal, especially the upper limit of the body's body temperature, and 2/9 is the case of constant temperature animals such as birds and mammals. That is, the thermal stability of the collagen structure depends on the imino acid content, in particular on the amount of hydroxyproline.
  • Hydroxyproline is rarely found in proteins other than collagen (somewhat present in elastin), and the amount of collagen can be measured by quantifying hydroxyproline, since only proline contained in the peptide is hydrated to form hydroxyproline. (Hyproproline content is about 10%, so 100 ⁇ g of hydroproline, about 1 mg as collagen).
  • hydroxyproline is used as an indicator of the total amount of collagen protein synthesis, the amount of degradation and the like.
  • the hydroxyproline content in collagen varies not only with the form (gene) of collagen, but also with the conditions (environmental conditions) when animal tissues and cells synthesize.
  • the amount of lysine and arginine is higher than that of glutamic acid or aspartic acid, and the charge of the whole collagen peptide has a positive charge in neutral pH. That is, the isoelectric point can be seen as basic. More glycine and less hydrophobic amino acids result in a smaller volume of molecules (0.75 for globular proteins, about 0.70 for collagen), and an average amino acid molecular weight of less than 100 (for globular proteins, usually 110).
  • the amount of collagen produced decreases with age. Collagen production is the highest in 20s, and 70% is known to decrease rapidly afterwards. Therefore, collagen is not a substance produced in the body, so it is necessary to take it as food.
  • Collagen can be divided into high molecular collagen and low molecular collagen according to the molecular weight.
  • Polymer collagen generally has a molecular weight of 5,000 Da.
  • polymer collagen with a molecular weight of 5,000 or more is used for general foods and the like.
  • low molecular weight collagen has a molecular weight of 1,500 Da. The following things can be expected to have high body absorption and high functionality such as skin elasticity. In general, low molecular weight collagen is used in health food, cosmetics and general medicine.
  • Collagen is a component of the human body but is also present in large quantities in meat and fish.
  • pig shells, chicken wings, doggantang, chicken feet are known to be present a lot of people are eating a lot.
  • collagen is ingested through meat products of animal foods, but a small amount of collagen in plant foods is present.
  • Mainly dried persimmons, tofu chungcha, persimmon leaf tea, etc. There is a side that is difficult to expect collagen effect through the intake of such foods.
  • the connective tissue of the dermis which is closely related to skin aging, is mainly composed of collagen and elastin, and collagen is the main protein of connective tissue, accounting for 70-80% of the dry weight of the dermis, giving elasticity and strength to the skin. It plays a large role in maintaining moisturizing. Degradation and content of collagen due to photoaging and natural aging will adversely affect the skin such as wrinkle formation, rough skin, reduced elasticity and dryness. As such, collagen plays an important role in maintaining the elasticity and moisturizing of the skin, which can be attributed to proteins essential for cell composition and production. Therefore, in terms of nutrition, high quality protein intake is closely related to skin health.
