Abstract
본 발명은 코어와 그의 표면에 결합된 시알산, 시알릴락토스 또는 이들의 유도체를 포함하는 결합체 및 그의 용도에 관한 것이다. 본 발명에서 제공하는 결합체는 인플루엔자 바이러스의 표면에 존재하는 헤마글루티닌과 결합하여 인플루엔자 바이러스의 감염과정을 억제함으로써, 인플루엔자 바이러스 감염증을 예방 또는 치료할 수 있을 뿐만 아니라, 항 바이러스 제제에 대하여 내성을 나타내는 인플루엔자 바이러스에 대하여도 그의 감염증을 예방 또는 치료할 수 있으므로, 인플루엔자 바이러스 감염증의 예방 또는 치료용 제제의 개발에 널리 활용될 수 있다.
Description
본 발명은 코어와 그의 표면에 결합된 시알산, 시알릴락토스 또는 이들의 유도체를 포함하는 결합체 및 그의 용도에 관한 것이다.
인플루엔자 바이러스(Influenza virus)는 오르소믹소 계통(Family Orthomyxoviridae)에 속하는 RNA 바이러스로서 혈청형은 A형, B형, C형 등 3가지로 구분된다. 그 중 B형과 C형은 사람에서만 감염이 확인되고 있으며, A형은 사람, 말, 돼지, 기타 포유류 그리고 다양한 종류의 가금과 야생조류에서 감염이 확인되고 있다. A형 인플루엔자 바이러스의 혈청형은 바이러스 표면의 두 가지 단백질인 혈구응집소(Hemagglutinin; HA)와 뉴라미니다제(Neuraminidase:NA)의 종류에 따라 구분되는데, 지금까지 144종류(HA 단백질 16종과 NA 단백질 9종)가 알려져 있다. HA는 바이러스가 체세포에 부착하는 역할을 하며, NA는 바이러스가 세포 내로 침투할 수 있도록 한다. 이러한 인플루엔자 바이러스는 매년 겨울이면 새로운 항원성을 지닌 인플루엔자 바이러스가 유행하며 사람을 포함한 가축, 조류에 거쳐 바이러스가 유행하며 이러한 바이러스의 모니터링은 매우 중요하다. 따라서 이에 따른 특이성과 민감성이 높은 프라이머와 프로브의 개발이 요구되고 있다.
최근 관심이 집중되고 있는 신종 인플루엔자 A(H1N1) 바이러스는 "신종 플루" 또는 "신종 플루 바이러스"라고도 하는데, 이는 사람, 돼지, 조류 인플루엔자 바이러스의 유전물질이 혼합되어 있는 새로운 형태의 바이러스로서 2009년 4월 처음 발견되었다. 바이러스의 전파 경로에 대해서는 아직까지 명확하게 밝혀지지 않았지만, 기존의 계절 인플루엔자 바이러스와 유사하게 비말(飛沫) 감염, 곧 감염된 사람의 기침이나 재채기 등을 통하여 주로 사람 대 사람으로, 감염자와 가까운 접촉자 사이(약 2m이내)에서 전파되는 것으로 알려져 있다. 또한, 음식의 경우 70℃ 이상으로 가열 조리하면 음식에 존재하는 바이러스가 사멸하는 것으로 알려져 있다.
상기 신종 플루 바이러스의 잠복기는 대략 1~7일 사이로 추정되고, 증상으로는 확진 환자에게서 발열, 오한, 두통, 기침, 인후통, 콧물, 호흡곤란 등의 상기도 증상, 근육통, 관절통, 피로감, 구토 또는 설사 등이 나타난다고 알려져 있다. 보통 증상이 발생하기 하루 전부터 발생 후 7일까지 전염력이 있는 것으로 보고되었으며, 어린이의 경우는 10일 이상으로 길어질 수도 있다. 상기 신종 플루 바이러스의 감염으로 인한 피해를 감소시키기 위하여, 신종 플루 바이러스의 감염을 조기에 진단할 수 있는 방법을 개발하려는 연구가 활발히 진행되어왔다. 예를 들어, 한국공개특허 제2011-0064174호에는 타종 인플루엔자 바이러스를 제외한 신종 인플루엔자 바이러스 A/ H1N1에만 특이적인 단클론 항체와 이를 생산하는 융합 세포, 상기 단클론 항체를 포함한 진단 키트 및 이를 이용한 신종 인플루엔자 바이러스 A/H1N1을 진단하는 방법이 개시되어 있고, 한국공개특허 제2011-0096940호에는 신종 인플루엔자 바이러스의 헤마글루티닌(HA) 항원에 대한 항체를 이용한 신속 면역크로마토그라피법을 이용하여 A형 신종 인플루엔자 바이러스(Influenza A, H1N1)를 진단할 수 있는 진단 키트가 개시되어 있으며, 한국공개특허 제2011-0127034호에는 인플루엔자 A형 바이러스를 검출하기 위한 인플루엔자 A형 바이러스 공통 PCR 프라이머 및 돼지 유래 인플루엔자(신종플루, A/Korea/01/2009(H1N1)) 바이러스 검출 특이 PCR 프라이머를 이용하여 신종플루(A/Korea/01/ 2009(H1N1)) 바이러스를 신속하고 정확하게 검출할 수 있는 방법이 개시되어 있다.
그러나, 이처럼 바이러스의 감염을 진단한다 하여도 감염된 인플루엔자 바이러스를 치료할 수 있는 제제로는 오셀타미비르(Oseltamivir, 상품명 타미플루), 자나미비어(Zanamivir, 상품명 리렌자) 등에 불과하고, 이러한 제제를 사용하여도 이에 대한 내성을 갖는 바이러스가 발생됨에 따라, 상기 바이러스를 치료하는 방법의 개발은 여전히 미미한 실정이다.
본 발명자들은 인플루엔자 바이러스 감염증을 효과적으로 치료하는 방법을 개발하고자 예의 연구노력한 결과, 코어(core)와 그의 표면에 결합된 시알산, 시알릴락토스 또는 이들의 유도체를 포함하고, 상기 시알산, 시알릴락토스 또는 이들의 유도체는 서로 동일한 간격으로 배열된 것을 특징으로 하는 결합체를 사용할 경우, 인플루엔자 바이러스 감염증을 치료할 수 있을 뿐만 아니라, 종래의 항 바이러스 제제에 대하여 내성을 나타내는 바이러스에 의한 감염까지도 치료할 수 있음을 확인하고, 본 발명을 완성하였다.
본 발명의 하나의 목적은 코어와 그의 표면에 결합된 시알산, 시알릴락토스 또는 이들의 유도체를 포함하고, 상기 시알산, 시알릴락토스 또는 이들의 유도체는 서로 동일한 간격으로 배열된 것을 특징으로 하는 결합체를 제공하는 것이다.
본 발명의 다른 목적은 상기 결합체를 포함하는 인플루엔자 바이러스 감염증 예방 또는 치료용 약학 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 결합체를 포함하는 인플루엔자 바이러스 감염증 예방 또는 개선용 식품 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 결합체를 인플루엔자 바이러스 감염이 의심되거나 또는 인플루엔자 바이러스에 감염된 개체에 투여하는 단계를 포함하는 인플루엔자 바이러스 감염증을 예방 또는 치료하는 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 결합체를 분리된 인플루엔자 바이러스에 처리하는 단계를 포함하는, 인플루엔자 바이러스의 감염을 억제하는 방법을 제공하는 것이다.
본 발명에서 제공하는 결합체는 인플루엔자 바이러스의 표면에 존재하는 헤마글루티닌과 결합하여 인플루엔자 바이러스의 감염과정을 억제함으로써, 인플루엔자 바이러스 감염증을 예방 또는 치료할 수 있을 뿐만 아니라, 항 바이러스 제제에 대하여 내성을 나타내는 인플루엔자 바이러스에 대하여도 그의 감염증을 예방 또는 치료할 수 있으므로, 인플루엔자 바이러스 감염증의 예방 또는 치료용 제제의 개발에 널리 활용될 수 있다.

본 발명의 용어 “코어(core)”란, 시알산, 시알릴락토스 또는 이들의 유도체와 결합하여, 상기 시알산, 시알릴락토스 또는 이들의 유도체가 서로 동일한 간격으로 배열될 수 있게 하는 지지체로서의 역할을 수행하는 물질을 의미한다.
본 발명에 있어서, 상기 코어를 구성하는 물질은 시알산, 시알릴락토스 또는 이들의 유도체와 결합할 수 있는 반응기를 포함하는 한 특별히 이에 제한되지 않으나, 일 예로서, 폴리아미도아민(polyamidoamine, PAMAM), 폴리하이드록시부티레이트, 폴리하이드록시발레르에이트, 폴리라이신, 폴리락트산, 폴리글리콜리드, 폴리카프로락톤, 폴리프로필렌퓨머레이트, 폴리다이옥세논, 폴리뉴클레오티드, 이들의 공중합체을 단독으로 또는 조합하여 사용할 수 있다.
본 발명에 있어서, 상기 코어는 그의 표면에 복수개의 시알산, 시알릴락토스 또는 이들의 유도체가 결합하여, 서로 동일한 간격으로 배열되어야 하기 때문에, 일정 수준 이상의 직경을 갖고 있는 것이 바람직하다. 상기 코어의 직경은 그의 표면에 복수개의 시알산, 시알릴락토스 또는 이들의 유도체가 결합하여, 서로 동일한 간격으로 배열될 수 있는 한, 특별히 이에 제한되지 않으나, 일 예로서, 3.0 내지 6.0 nm가 될 수 있고, 다른 예로서, 3.5 내지 5.5 nm가 될 수 있으며, 또 다른 예로서, 4.0 내지 6.0 nm가 될 수 있고, 또 다른 예로서, 4.5 내지 5.4 nm가 될 수 있다.

본 발명의 일 실시예에서는, 상기 코어 구성물질에 포함된 반응기가 규칙적으로 배열되어, 이에 결합된 시알산, 시알릴락토스 또는 이들의 유도체가 동일한 간격으로 배열될 수 있는, 덴드리머 형태의 코어를 제작하였다.
본 발명의 용어 "덴드리머(dendrimer)"란, 기반 화합물이 나무(dendron)가지 모양의 분자사슬로 중첩되어 구형의 구조를 이루는 거대분자를 의미하는데, 정확한 분자량과 구조를 갖는 나노 크기의 입자 형성이 용이하고, 최외각에 반응기를 다수 보유하고 있기 때문에, 화학적 또는 물리적으로 독특한 특성을 나타내며, 표면의 밀집된 말단기에 다양한 유도체와 반응기를 도입하는 것이 가능하다. 특히, 덴드리머는 선형 고분자 형태에 비해서 조절이 쉽고, 구조 예측이 용이하여 다양한 분야에 적용하는 것이 가능하다.
덴드리머를 형성하는 기반 화합물의 중첩 수준에 따라, 덴드리머의 세대를 특정할 수 있다. 예를 들어, 하나의 기반 화합물이 상호간에 결합되지 않은 상태로 중합된 경우 1세대 덴드리머(G1)라 표현할 수 있고, 상기 1세대 덴드리머에 포함된 기반 화합물의 말단에 1개의 기반 화합물이 추가로 결합된 경우 2세대 덴드리머(G2)라 표현할 수 있으며, 상기 2세대 덴드리머에 포함된 기반 화합물의 말단에 1개의 기반 화합물이 추가로 결합된 경우 3세대 덴드리머(G3)라 표현할 수 있고, 상기 3세대 덴드리머에 포함된 기반 화합물의 말단에 1개의 기반 화합물이 추가로 결합된 경우 4세대 덴드리머(G4)라 표현할 수 있으며, 상기 4세대 덴드리머에 포함된 기반 화합물의 말단에 1개의 기반 화합물이 추가로 결합된 경우 5세대 덴드리머(G5)라 표현할 수 있다. 즉, 기반 화합물이 1개 추가로 결합됨에 따라, 세대수로 1세대씩 증가한다.
본 발명에 있어서, 상기 덴드리머는 상술한 코어를 구성하는 물질을 기반 화합물로 사용할 수 있는데, 상기 기반 화합물은 표면에 복수개의 시알산, 시알릴락토스 또는 이들의 유도체가 결합하여, 서로 동일한 간격으로 배열될 수 있는 코어를 형성할 수 있는 한 특별히 이에 제한되지 않으나, 일 예로서, 폴리아미도아민, 폴리라이신, 폴리뉴클레오티드 등이 될 수 있고, 다른 예로서, 폴리아미도아민이 될 수 있다.
본 발명의 용어 “시알산(sialic acid)”이란, 시알린산 또는 뉴라민산 이라고도 호칭되며, 피루브산과 만노사민의 알돌축합체인 노이람산이 n-아세틸화, n-글리콜화 또는 o-아세틸화된 화학구조를 갖는 아미노당의 일종을 의미한다.
본 발명에 있어서, 상기 시알산은 바이러스의 헤마글루티닌(HA)과 결합하여 그의 기능을 억제하는 리간드로서 사용될 수 있는데, 상기 리간드로는 시알산 뿐만 아니라 그의 유도체가 사용될 수도 있다.
본 발명의 용어 “유도체(derivative)”란, 목적 화합물의 일부를 화학적으로 변화시켜서 얻어지는 유사한 화합물을 의미하는데, 대체로 목적 화합물 중의 수소원자 또는 특정 원자단이 다른 원자 또는 원자단에 의하여 치환된 화합물이 될 수 있다.
본 발명에 있어서, 상기 유도체는 시알산 또는 시알릴락토스의 유도체로 해석될 수 있는데, 상기 시알산 또는 시알릴락토스의 유도체는 바이러스의 HA와 결합하여 그의 기능을 억제할 수 있는 한, 특별히 이에 제한되지 않으나, 일 예로서, 시알산에 당류가 결합된 시알릴올리고당이 될 수 있고, 다른 예로서, 3'-시알릴락토스(3'-Sialyllactose), 6'-시알릴락토스(6'-Sialyllactose), 시알릴락토-N-테트라오스(sialyl lacto-N-tetraose), 디시알릴락토-N-테트라오스(disialyl lacto-N-tetraose) 등이 될 수 있다.
본 발명에서 제공하는 결합체에 포함된 시알산, 시알릴락토스 또는 이들의 유도체 간의 간격은 특별히 이에 제한되지 않으나, 1.0 nm 이상이 될 수 있고, 1.2 nm 이상이 될 수 있으며, 1.6 nm 이상이 될 수 있고, 1.9 nm 이상이 될 수 있으며, 2.0 nm 이상이 될 수 있고, 2.1 nm 이상이 될 수 있으며, 2.4 nm 이상이 될 수 있고, 3.1 nm 이상이 될 수 있으며, 1.0 내지 4.0 nm 가 될 수 있고, 1.2 내지 4.0 nm 가 될 수 있으며, 1.6 내지 4.0 nm 가 될 수 있고, 1.9 내지 4.0 nm 가 될 수 있으며, 2.0 내지 4.0 nm 가 될 수 있고, 2.1 내지 4.0 nm 가 될 수 있으며, 2.4 내지 4.0 nm 가 될 수 있고, 3.1 내지 4.0 nm 가 될 수 있다.
하나의 예시로서, 폴리아미도아민 덴드리머를 코어로서 포함하고, 6'-시알릴락토스(6SL)를 상기 코어에 결합되는 시알산 유도체로서 포함하는 결합체의 경우, 상기 6SL 간의 간격은 특별히 이에 제한되지 않으나, 일 예로서, 1.0 내지 4.0 nm가 될 수 있고, 다른 예로서, 1.6 내지 3.5 nm가 될 수 있으며, 또 다른 예로서, 2.0 내지 3.1 nm가 될 수 있고, 또 다른 예로서, 2.4 또는 3.1 nm가 될 수 있다.

하나의 예시로서, 폴리아미도아민(PAMAM) 덴드리머를 코어로서 포함하고, 6'-시알릴락토스(6SL)를 상기 코어에 결합되는 시알산 유도체로서 포함하는 결합체로는, 2 내지 5세대의 PAMAM 덴드리머의 말단 아민그룹에 10 내지 128개의 6SL이 결합된 형태(6SL-PAMAM)의 결합체가 될 수 있다.
상기 6SL-PAMAM 결합체는 사용된 PAMAM 덴드리머의 세대에 따라, 이에 결합된 6SL의 갯수가 변화될 수 있는데, 일 예로서, 3세대의 PAMAM 덴드리머를 포함하는 6SL-PAMAM 결합체는 상기 덴드리머의 말단 아민그룹에 10 내지 32개의 6SL이 결합될 수 있고, 다른 예로서, 4세대의 PAMAM 덴드리머를 포함하는 6SL-PAMAM 결합체는 상기 덴드리머의 말단 아민그룹에 20 내지 64개의 6SL이 결합될 수 있으며, 또 다른 예로서, 5세대의 PAMAM 덴드리머를 포함하는 6SL-PAMAM 결합체는 상기 덴드리머의 말단 아민그룹에 20 내지 128개의 6SL이 결합될 수 있다.

먼저, 4세대의 PAMAM 덴드리머를 포함하는 6SL-PAMAM 결합체의 경우, 일 예로서 상기 덴드리머의 말단 아민그룹에 20 내지 64개의 6SL이 결합될 수 있고, 다른 예로서 20 내지 40개의 6SL이 결합될 수 있으며, 또 다른 예로서 20 내지 31개의 6SL이 결합될 수 있고, 또 다른 예로서 20 내지 26개의 6SL이 결합될 수 있으며, 또 다른 예로서 20개의 6SL이 결합될 수 있다.

본 발명의 일 실시예에 의하면, 에틸렌 디아민 코어(G2 내지 G5)를 갖는 PAMAM 덴드리머의 아미노기로 6SL의 환원당의 알데히드 그룹을 환원성 아미노화 함으로써 다양한 형태의 6SL-PAMAM 결합체를 제조하고(도 1a 및 1d). 이에 포함된 6SL의 수를 결정하였다(표 4).
이어, 상기 6SL-PAMAM 결합체의 인플루엔자 바이러스 감염억제능을 시험관 조건에서 분석한 결과, 일정한 크기의 덴드리머가 필요하고(도 2a), 6SL 리간드 간의 간격과 H1N1 바이러스의 감염억제 효율이 연관됨을 확인하였다(표 5, 도 2b). 또한, 뉴라미니다제(neuraminidase, NA)의 억제제를 상기 6SL-PAMAM 결합체와 병용처리할 경우, H1N1 바이러스 감염억제 효과가 증가됨을 확인하였다(표 6).
다음으로, HA에 대한 6SL-PAMAM 결합체의 결합 친화력을 분석한 결과, 6SL 리간드 간의 간격이 2.4 nm 이상인 6SL-PAMAM 결합체(S3-G4, S4-G4 및 S7-G5)는 모두 HA 단백질과 높은 결합친화도를 나타냄을 확인하였다(도 3a).
다른 실시양태로서, 본 발명은 상기 결합체를 포함하는 인플루엔자 바이러스 감염증 예방 또는 치료용 약학 조성물을 제공한다.
본 발명의 용어 "인플루엔자 바이러스(influenza virus)"란, 바이러스성 호흡기 질환의 주요 원인 병원체를 의미하는데, NP(nuleocapsid)와 M(matrix) 단백질의 항원성 차이에 의해 크게 A, B 및 C형으로 구분된다. 이 중 A형은 다시 헤마글루티닌(hemagglutinin) 단백질 항원 특성에 따라 H1형에서 H16형까지, 뉴라미다아제(Neuramidase) 단백질 항원 특성에 따라 N1형에서 N9형의 아형으로 분류된다
본 발명에 있어서, 상기 인플루엔자 바이러스는 특별히 이에 제한되지 않으나, 일 예로서 인플루엔자 A 바이러스 또는 인플루엔자 B 바이러스가 될 수 있고, 다른 예로서 신종 인플루엔자 A(H1N1) 바이러스가 될 수 있다.
본 발명의 용어 "신종 인플루엔자 A(H1N1) 바이러스"란, 본 발명의 명세서에서 "신종 플루 바이러스", "H1N1" 또는 "H1N1 바이러스"라고도 표시하고, A형 인플루엔자 바이러스가 변이를 일으켜 생성되며, 바이러스 표면의 두 가지 단백질인 혈구응집소(Hemagglutinin; HA)와 뉴라미니다제(Neuraminidase:NA)의 종류에 의해 결정되는 혈청형은 H1N1이고, 사람에게 감염되어 새로운 호흡기 질환을 유발시키는 바이러스를 의미한다.
본 발명의 용어 “인플루엔자 바이러스 감염증”이란, 개체에 인플루엔자 바이러스가 감염되어 나타나는 병리학적 질환을 의미한다. 예를 들어, A형 인플루엔자 바이러스의 변이주의 감염에 의해 발생되는 질환인 "신종 플루"는 발열, 오한, 두통, 기침, 인후통, 콧물, 호흡곤란 등의 상기도 증상, 근육통, 관절통, 피로감, 구토, 설사 등의 증상을 나타낸다고 알려져 있다.
본 발명의 용어, "예방"이란, 본 발명에서 제공하는 상기 결합체 또는 이를 포함하는 약학 조성물의 투여에 의해 인플루엔자 바이러스 감염증의 증상을 억제시키거나 또는 지연시키는 모든 행위를 의미한다.
본 발명의 용어, "치료"란, 본 발명에서 제공하는 상기 결합체 또는 이를 포함하는 약학 조성물의 투여에 의해 인플루엔자 바이러스 감염증의 증상을 호전시키거나 이롭게 변경시키는 모든 행위를 의미한다.
본 발명에서 제공하는 약학 조성물에 포함된 결합체의 함량은 인플루엔자 바이러스 감염증을 예방 또는 치료하는 효과를 나타낼 수 있는 한 특별히 이에 제한되지 않으나, 일 예로서, 총 조성물의 중량 대비 0.0001 내지 50 중량%가 될 수 있고, 다른 예로서, 0.01 중량% 내지 10 중량%가 될 수 있다.
본 발명의 약학 조성물은 약학 조성물의 제조에 통상적으로 사용하는 약학적으로 허용가능한 담체, 부형제 또는 희석제를 추가로 포함할 수 있고, 상기 담체는 비자연적 담체(non-naturally occuring carrier)를 포함할 수 있다.
본 발명의 용어, "약학적으로 허용가능한"이란, 상기 조성물에 노출되는 세포나 인간에게 독성이 없는 특성을 나타내는 것을 의미한다.
구체적으로, 상기 약학 조성물은, 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 본 발명에서, 상기 약학 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(calcium carbonate), 수크로스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스티레이트, 탈크 같은 윤활제들도 사용된다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는 데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명의 일 실시예에 의하면, 본 발명에서 제공하는 6SL-PAMAM 결합체가 바이러스의 감염에 어떠한 영향을 미치는지 전자현미경을 통해 분석한 결과, 상기 6SL-PAMAM 결합체가 바이러스의 표면에 존재하는 헤마글루티닌(hemagglutinin, HA)과 결합하여 바이러스의 표면에 소포를 형성하고, 이로 인하여 바이러스의 부착, 세포내 진입 및 엔도좀 융합/방출 과정을 억제할 것으로 분석되었다(도 4). 또한, 면역형광분석을 통하여 상기 6SL-PAMAM 결합체가 바이러스의 세포내 진입에 관여함을 알 수 있었다(도 5). 아울러, 상기 6SL-PAMAM 결합체는 바이러스의 감염에 의하여 증가되는 NA의 발현수준을 저하시키는 효과를 나타냄을 확인하였다(도 6).
이에, 생체내에서 바이러스의 감염에 대한 6SL-PAMAM 결합체의 효과를 분석한 결과, 감염된 숙주의 폐조직내의 바이러스 역가를 감소시키고(도 7a), 바이러스 감염에 의한 조직병리학적 증상을 감소시키며(도 7b), 바이러스 감염에 의한 사이토카인(IL-6 및 CCL2)의 수준을 감소시키고(도 7c), 감염된 숙주의 생존율을 증가시키며(도 7d), 바이러스 감염에 의하여 감소된 체중을 회복시키는 효과를 나타냄(도 7e)을 확인하였다.
끝으로, 항 바이러스 제제에 대하여 내성을 나타내는 바이러스의 감염에 대한 6SL-PAMAM 결합체의 효과를 분석한 결과, 현존하는 항 바이러스 제제에 대하여 내성을 나타내는 인플루엔자 바이러스의 감염에 대하여도, 본 발명의 6SL-PAMAM 결합체가 억제활성을 나타냄을 알 수 있었다(도 8a 내지 8c).
또 다른 실시양태로서, 본 발명은 상기 결합체 또는 상기 결합체를 포함하는 약학 조성물을, 인플루엔자 바이러스 감염증의 발병이 의심되거나 또는 인플루엔자 바이러스 감염증이 발병된 개체에 투여하는 단계를 포함하는, 인플루엔자 바이러스 감염증을 예방 또는 치료하는 방법을 제공한다.
본 발명의 용어 "개체"란, 인플루엔자 바이러스 감염증이 발병하였거나 발병할 수 있는 살아있는 유기체를 의미하는데, 일 예로서, 호흡기관을 포함하는 고등 척추동물이 될 수 있고, 다른 예로서, 포유동물이 될 수 있으며, 또 다른 예로서, 영장류가 될 수 있고, 또 다른 예로서, 인간을 포함한 쥐, 생쥐, 가축 등이 될 수 있다.
본 발명의 용어 "투여"란, 적절한 방법으로 환자에게 소정의 물질을 도입하는 행위를 의미하며, 유효성분을 인체 또는 수의용으로 제형화시킨 후, 다양한 경로로 투여할 수 있다. 본 발명의 결합체를 포함하는 약학 조성물의 투여경로는 특별히 이에 제한되지 않으나, 일 예로서, 비경구 경로, 예컨대 혈관내, 정맥내, 동맥내, 근육내 또는 피하 등의 경로로 투여될 수 있고, 다른 예로서, 경구, 비강, 직장, 경피 또는 에어로졸을 통한 흡입 경로로 투여될 수도 있으며, 또 다른 예로서, 볼루스로 투여하거나 또는 서서히 주입할 수도 있다.
본 발명의 결합체 또는 이를 포함하는 약학 조성물은 약학적으로 유효한 양으로 투여될 수 있다.
본 발명의 용어, "약학적으로 유효한 양"이란, 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분하며 부작용을 일으키지 않을 정도의 양을 의미하며, 유효 용량 수준은 환자의 성별, 연령, 체중, 건강상태, 질병의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 방법, 투여 시간, 투여 경로, 및 배출 비율, 치료 기간, 배합 또는 동시에 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 당업자에 의해 용이하게 결정될 수 있다.
일 예로서, 본 발명의 결합체는, 일 예로서, 1회 투여시 1×107 내지 1×1011개, 다른 예로서 1×108 내지 5×1010개, 또 다른 예로서 5×108 내지 2×1010개가 투여될 수 있으나, 이에 제한되지 않는다.
다른 예로서, 본 발명의 약학 조성물을 기준으로 하는 경우에는, 고형분을 기준으로 1일 0.0001 내지 100 mg/체중 kg으로, 더욱 구체적으로 0.001 내지 100 mg/체중 kg으로 투여할 수 있다. 투여는 상기 권장 투여량을 하루에 한 번 투여할 수도 있고, 수회 나누어 투여할 수도 있다.
본 발명의 인플루엔자 바이러스 감염증의 예방 또는 치료 방법에서, 상기 약학 조성물을 투여하는 투여 경로 및 투여 방식은 특별히 제한되지 않으며, 목적하는 해당 부위에 상기 결합체를 포함하는 약학 조성물이 도달할 수 있는 한 임의의 투여 경로 및 투여 방식에 따를 수 있다. 구체적으로, 상기 약학 조성물은 경구 또는 비경구의 다양한 경로를 통하여 투여될 수 있으며, 그 투여 경로의 비제한적인 예로는, 구강, 직장, 국소, 정맥내, 복강내, 근육내, 동맥내, 경피, 비측내 또는 흡입 등을 통하여 투여되는 것을 들 수 있다.
또 다른 실시양태로서, 본 발명은 상기 결합체를 포함하는 인플루엔자 바이러스 감염증 예방 또는 개선용 식품 조성물을 제공한다.
본 발명의 용어 "개선"이란, 본 발명의 결합체를 포함하는 조성물의 투여로 치료되는 상태와 관련된 파라미터, 예를 들면 증상의 정도를 적어도 감소시키는 모든 행위를 의미한다.
본 발명의 용어 "식품"이란, 육류, 소시지, 빵, 초콜릿, 캔디류, 스낵류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알코올음료, 비타민 복합제, 건강 기능 식품 및 건강 식품 등이 있으며, 통상적인 의미에서의 식품을 모두 포함한다.
본 발명의 식품 조성물은, 일상적으로 섭취하는 것이 가능하기 때문에 높은 수준으로 인플루엔자 바이러스 감염증을 개선시킬 수 있는 효과를 기대할 수 있으므로, 건강 증진 목적으로 매우 유용하게 사용될 수 있다.
상기 건강 기능(성) 식품(functional food)이란, 특정보건용 식품(food for special health use, FoSHU)과 동일한 용어로, 영양 공급 외에도 생체조절기능이 효율적으로 나타나도록 가공된 의학, 의료효과가 높은 식품을 의미한다. 여기서 '기능(성)'이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건용도에 유용한 효과를 얻는 것을 의미한다. 본 발명의 식품은 당 업계에서 통상적으로 사용되는 방법에 의하여 제조가능하며, 상기 제조시에는 당 업계에서 통상적으로 첨가하는 원료 및 성분을 첨가하여 제조할 수 있다. 또한 상기 식품의 제형 또한 식품으로 인정되는 제형이면 제한 없이 제조될 수 있다. 본 발명의 식품용 조성물은 다양한 형태의 제형으로 제조될 수 있으며, 일반 약품과는 달리 식품을 원료로 하여 약품의 장기 복용 시 발생할 수 있는 부작용 등이 없는 장점이 있고, 휴대성이 뛰어나므로, 본 발명의 식품은 인플루엔자 바이러스 감염증 개선 효과를 증진시키기 위한 보조제로 섭취가 가능하다.

구체적으로, 상기 건강 기능 식품은 본 발명의 화합물을 음료, 차류, 향신료, 껌, 과자류 등의 식품 소재에 첨가하거나, 캡슐화, 분말화, 현탁액 등으로 제조한 식품으로, 이를 섭취할 경우 건강상 특정한 효과를 가져오는 것을 의미하나, 일반 약품과는 달리 식품을 원료로 하여 약품의 장기 복용 시 발생할 수 있는 부작용이 없는 장점이 있다.
상기 식품 조성물은 생리학적으로 허용 가능한 담체를 추가로 포함할 수 있는데, 담체의 종류는 특별히 제한되지 않으며 당해 기술 분야에서 통상적으로 사용되는 담체라면 어느 것이든 사용할 수 있다.
또한, 상기 식품 조성물은 식품 조성물에 통상 사용되어 냄새, 맛, 시각 등을 향상시킬 수 있는 추가 성분을 포함할 수 있다. 예들 들어, 비타민 A, C, D, E, B1, B2, B6, B12, 니아신(niacin), 비오틴(biotin), 폴레이트(folate), 판토텐산(panthotenic acid) 등을 포함할 수 있다. 또한, 아연(Zn), 철(Fe), 칼슘(Ca), 크롬(Cr), 마그네슘(Mg), 망간(Mn), 구리(Cu), 크륨(Cr) 등의 미네랄; 및 라이신, 트립토판, 시스테인, 발린 등의 아미노산을 포함할 수 있다.
또한, 상기 식품 조성물은 방부제(소르빈산 칼륨, 벤조산나트륨, 살리실산, 데히드로초산나트륨 등), 살균제(표백분과 고도 표백분, 차아염소산나트륨 등), 산화방지제(부틸히드록시아니졸(BHA), 부틸히드록시톨류엔(BHT) 등), 착색제(타르색소 등), 발색제(아질산 나트륨, 아초산 나트륨 등), 표백제(아황산나트륨), 조미료(MSG 글루타민산나트륨 등), 감미료(둘신, 사이클레메이트, 사카린, 나트륨 등), 향료(바닐린, 락톤류 등), 팽창제(명반, D-주석산수소칼륨 등), 강화제, 유화제, 증점제(호료), 피막제, 검기초제, 거품억제제, 용제, 개량제 등의 식품 첨가물(food additives)을 포함할 수 있다. 상기 첨가물은 식품의 종류에 따라 선별되고 적절한 양으로 사용될 수 있다.
본 발명의 식품 조성물의 일 예로 건강음료 조성물로 사용될 수 있으며, 이 경우 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드; 말토스, 슈크로스와 같은 디사카라이드; 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드; 자일리톨, 소르비톨, 에리트리톨 등의 당알콜일 수 있다. 감미제는 타우마틴, 스테비아 추출물과 같은 천연 감미제; 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 건강음료 조성물 100 mL 당 일반적으로 약 0.01 ~ 0.04 g, 구체적으로 약 0.02 ~ 0.03 g이 될 수 있다.
상기 외에 건강음료 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산, 펙트산의 염, 알긴산, 알긴산의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올 또는 탄산화제 등을 함유할 수 있다. 그 밖에 천연 과일주스, 과일주스 음료, 또는 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 혼합하여 사용할 수 있다. 이러한 첨가제의 비율은 크게 중요하진 않지만 본 발명의 건강음료 조성물 100 중량부당 0.01 내지 0.1 중량부의 범위에서 선택되는 것이 일반적이다.
또 다른 실시양태로서, 본 발명은 상기 결합체 또는 상기 결합체를 인플루엔자 바이러스에 처리하는 단계를 포함하는, 인플루엔자 바이러스의 감염을 억제하는 방법을 제공한다.
상술한 바와 같이, 본 발명의 결합체는 이에 포함된 시알산, 시알릴락토스 또는 이들의 유도체가 인플루엔자 바이러스의 표면에 존재하는 헤마글루티닌과 결합하여 소포를 형성하고, 이로 인하여 상기 바이러스의 부착, 세포내 진입 및 엔도좀 융합/방출 과정을 억제하며, 바이러스의 세포내 진입에 관여하고, 바이러스의 감염에 의하여 증가되는 NA의 발현수준을 저하시키는 효과를 나타낼 수 있다.
따라서, 본 발명의 결합체는 시험관 조건(in vitro), 생체내 조건(in vivo) 또는 탈체 조건(ex vivo)에서 인플루엔자 바이러스의 감염을 억제하기 위하여 사용할 수 있다.
특히, 목적하는 세포에 대한 인플루엔자 바이러스의 감염성 연구를 수행하기 위하여 시험관 조건(in vitro) 또는 탈체 조건(ex vivo)에서 인플루엔자 바이러스의 감염을 억제하고자 하는 경우, 상기 결합체를 적절한 농도로 완충액에 희석시킨 후, 바이러스 용액 또는 바이러스에 감염된 세포에 처리하여, 상기 세포에 대한 인플루엔자 바이러스의 감염을 억제할 수 있다.
이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.

결합체의 합성
에틸렌 디아민 코어(G2 내지 G5)를 갖는 PAMAM 덴드리머의 아미노기로 6SL의 환원당의 알데히드 그룹을 환원성 아미노화 함으로써 6SL-PAMAM 결합체를 제조하였다(도 1a).
대략적으로, 50mM NaCNBH3을 포함하는 0.1M 소듐 보레이트 완충액(pH 9.5)에서, 각각의 PAMAM 덴드리머(20mg)와 서로 다른 양의 6SL 용액과 혼합하였다(표 1).

반응액을 서서히 교반하면서, 암소 및 실온에서 5일동안 연속적으로 반응시켰다.
반응이 종료된 후, 적절한 크기(MWCO 3 K 또는 10 K)의 Amicon centrifugal filters (Millipore)를 사용하여 여과함으로써, 반응하지 않은 제제를 제거하고, 탈이온수로 용매를 교체하였다.
여과 후, 용액을 동결건조시키고, 얻어진 백색 분말의 시료를 사용시까지 건조하에 저장하였다.

MS 분석
337 nm 질소 레이저를 사용하여 MALDI-TOF MS 분석을 수행하였다. 이때, 매트릭스로서 2,5-디히드록시 벤조산(10 mg/㎖)을 포함하는 50% 아세토니트릴을 사용하였는데, 분석물-매트릭스 용액을 1:2(분석물:매트릭스, v/v)의 비율로 제조 하였다. 상기 제조된 분석물-매트릭스 용액(1 ㎕)을 시료용 플레이트에 가하고 진공증발하여 건조시켰다. 또한, 선형 양이온 모드를 사용하여 스펙트럼 당 최소 100 회 촬영하였다. 상기 촬영된 데이터 분석은 Bruker Daltonics Microflex Analysis Software를 사용하여 수행하고(도 1b), 상기 데이터 분석을 통해 PAMAM에 결합된 6SL의 평균값을 산출하였다(표 2).

-NMR 분석
G2-G5 덴드리머, 6SL 및 이들의 결합체에 대한 NMR 스펙트럼을 Bruker AscendTM 500 분광기를 사용하여 중수소 옥사이드에서 측정하고(도 1c), 상기 데이터 분석을 통해 PAMAM에 결합된 6SL의 평균값을 산출하였다(표 3 및 도 1c).
도 1c에서 보듯이, PAMAM 덴드리머의 1H NMR 스펙트럼은 PAMAM 덴드리머 내의 아미드 결합에 인접한 -CH2-에 해당하는 2.45 내지 2.50 ppm에서 신호를 나타내었고, 이러한 전체적인 신호는 G2의 경우 56H, G3의 경우 120H, G4의 경우 248H 및 G5의 경우 504H를 나타내는 내부 표준으로 사용되었으며, 6SL 아세틸기의 -CH3에 해당하는 2.07 ppm에서의 특유의 전체적인 신호는 각 덴드리머상의 6SL 리간드의 수를 결정하는데 사용되었다.

실시예
3: 시험관 조건에서 6SL-
PAMAM
결합체의 인플루엔자 바이러스 감염 억제분석
인플루엔자 감염을 억제하는 6SL-PAMAM 결합체의 능력평가 방법을 이용하여 시험관 조건에서의 감염억제 분석을 수행하였다.
먼저, PBS를 이용하여 시험하고자 하는 결합체의 반-로그 연속 희석액(half-log serial dilution)을 제조하였다. 상기 제조된 희석액 50 ㎕를 동일부피의 1 × 103 TCID50의 H1N1 균주(A/California/04/2009 or oseltamivir-resistant H1N1, A/Gyeongnam/1820/2009)와 혼합하였다. 상기 혼합액을 37℃에서 1시간 동안 배양하고, 이어 1.5 × 105세포수/㎖의 MDCK 세포를 100 ㎕씩 가한 다음, 5% CO2 및 37℃ 조건에서 18시간 동안 추가로 배양하였다. 배양이 종료된 후, 상기 배양된 MDCK 세포에 고정액(아세톤:PBS=4:1, v/v)을 가하고 10 내지 15분 동안 고정시켰다. 이어, 항-NA 마우스 항체(Millipore) 및 항-마우스 염소 항체-HRP 결합체(Jackson Immunoresearch)를 사용한 ELISA 분석을 수행하여, 상기 각 MDCK 세포에 포함된 인플루엔자 핵 단백질(NP)을 검출하였다(도 2a). 이때, 음성 대조군(미감염 세포, 감염율 0%) 및 양성 대조군(억제제 없이 감염된 세포, 감염율 100%)에 대한 흡광도를 사용하여 상기 각 MDCK 세포에 대한 감염율을 산출하고, 상기 산출된 감염율의 통계분석은 GraphPad Prism 6.0 (GraphPad Software, San Diego, CA)을 사용하여 수행하였다.
도 2a에서 보듯이, 크기가 비교적 작은 S1-G2 및 S2-G3의 경우(G2 및 G3 덴드리머는 4 nm 미만의 직경), 감염억제는 관찰되지 않았고, S6-G4를 제외한, G4 및 G5 덴드리머(4.5 nm 초과의 직경)를 포함한 모든 6SL 결합체는 H1N1 감염을 억제함을 확인하였다.
상기 결과는 시험관 조건에서 H1N1 감염을 억제하기 위해서는 일정한 크기의 덴드리머 지지체가 필요함을 시사한다.
또한, 상기 도 2a는 PAMAM 결합체에 결합된 6SL의 수가 증가할 수록 H1N1 바이러스의 감염억제 효율이 감소하는 결과를 보여주었는데, 이는 다가 리간드의 결합 친화력이 리간드 원자가 (예를 들면, 리간드의 수)에 따라 증가한다는 지금까지의 연구결과에 부합되지 않았다.
탄저 독소의 경우, 이에 대한 억제제의 개발에 사용되는 요인으로서 원자가 뿐만 아니라 리간드 간의 간격이 사용된다는 점을 감안하여, 6SL-PAMAM 결합체에 포함된 6SL 리간드 간의 간격이 상기 H1N1 바이러스의 감염억제 효율과 연관되는지 확인하고자 하였다.
이에 따라, 6SL-PAMAM 결합체에 포함된 PAMAM 덴드리머의 직경을 이용하여 상기 덴드리머의 표면적(4πr2)을 산출하고, 상기 표면적을 6SL 리간드의 수로 나누어 6SL 리간드 간의 간격을 산출한 다음, 상기 산출된 6SL 리간드 간의 간격과 H1N1 바이러스의 감염억제 효율 간의 연관성을 분석하였다(표 5, 도 2b).

상기 표 5 및 도 2b에서 보듯이, 6SL 리간드 간의 간격과 H1N1 바이러스의 감염억제 효율이 연관됨을 확인하였다.
구체적으로, G2 및 G3를 포함하는 6SL-PAMAM 결합체(S1-G2 및 S2-G3)의 경우에는 6SL 리간드 간의 간격이 좁고, H1N1 바이러스의 감염억제 효과를 나타내지 않았다. 이에 반하여, G4를 포함하는 6SL-PAMAM 결합체(S3-G4, S4-G4, S5-G4 및 S6-G4)의 경우에는 6SL 리간드 간의 간격이 감소될 수록 H1N1 바이러스의 감염억제 효과가 저하되었다. 또한, G5를 포함하는 6SL-PAMAM 결합체(S7-G5, S8-G5, S9-G5 및 S10-G5)의 경우에도 6SL 리간드 간의 간격이 감소될 수록 H1N1 바이러스의 감염억제 효과가 저하되었다.
그러나, 동일한 6SL 리간드 간의 간격을 갖더라도, 6SL-PAMAM 결합체에 포함된 에틸렌 디아민 코어(G4 또는 G5)에 따라, H1N1 바이러스의 감염억제 효과가 차이를 나타냄을 알 수 있었다.
한편, H1N1 바이러스의 뉴라미니다제(neuraminidase, NA)는 6SL 리간드를 가수분해 할 수 있어, 6SL-PAMAM 결합체의 H1N1 바이러스 감염억제 효과를 제한 시킬 수 있다.
이에, 상기 NA의 억제제인 오셀타미비르 카르복실레이트(oseltamivir carboxylate, OC)의 처리여부가 6SL-PAMAM 결합체의 H1N1 바이러스 감염억제 효과에 영향을 미칠 수 있는지 확인하고자 하였다.
대략적으로, 1μM의 OC를 처리하거나 또는 처리하지 않은 조건에서 6SL-PAMAM 결합체와 H1N1 균주를 혼합하는 단계를 포함하는 것을 제외하고는, 상술한 바와 동일한 실험을 수행하여, 6SL-PAMAM 결합체의 H1N1 바이러스 감염억제 효과를 분석하였다(표 6).

4: HA에 대한 6SL-
PAMAM
결합체의 결합 친화력 분석
표준 아민 커플 링 프로토콜(GE healthcare, Uppsala, Sweden)에 따라 H1N1 바이러스의 표면항원인 HA 단백질을 CM5 칩에 고정시키고, 이로부터 6-SL PAMAM 결합체와 HA 단백질 간의 분자반응/속도 데이터를 수득하는 SPR(Surface plasmon resonance) 분석을 통해, HA와 6SL-PAMAM 결합체의 결합 친화력을 분석하였다.
대략적으로, CM5 칩 표면의 카르복시 메틸기는 N-에틸-N-(디메틸아미노프로필) 카르보디이미드와 N-히드록시석신이미드의 동일몰량 혼합물(최종농도 0.05M) 35㎖을 가하여(유속, 5㎖/분) 활성화시켰다. 그런 다음, HA를 100 mM 소듐아세테이트 완충액(pH 5.0) 완충액에 50㎍/㎖가 되도록 가하고, 상기 활성화된 CM5 칩 표면에 가하였다. 이어, 35㎕의 1M 에탄올아민을 처리하여 상기 CM5 칩 표면의 미반응 부위를 불활성화 시켰다.
한편, N-에틸-N-(디메틸아미노프로필) 카르보디이미드와 N-히드록시숙신이미드(최종농도 0.05M)의 동일몰량 혼합물(최종농도 0.05M) 35㎖을 가한 다음(유속, 5㎖/분), 35㎕의 1M 에탄올아민을 처리하여 대조군을 제작하였다.
다음으로, 6-SL PAMAM 결합체를 HBS-EP 완충액(0.01 M HEPES, 0.15 M NaCl, 3 mM EDTA, 0.005% surfactant P20, pH 7.4)에 희석시켰다. 서로 다른 수준의 6-SL PAMAM 결합체의 희석액을 50㎕/분의 유속으로 상기 HA가 결합된 CM5 칩에 가하였다. 그런 다음, 상기 CM5 칩에 HBS-EP 완충액을 가하여, 해리를 촉진시켰다. 3분이 경과한 후, 상기 CM5 칩에 2 M NaCl 50 ㎕를 가하여, 상기 CM5 칩의 표면을 재생하였다. 반응결과는 BIAcore 3000 control 및 BIAevaluation software (version 4.0.1)를 사용한 BIAcore 3000 (GE healthcare, Uppsala, Sweden)를 이용하여 분석하였고, 25℃에서 시간(센서 그램)의 함수로 표시하였다(도 3a).
도 3a에서 보듯이, 사용된 6SL-PAMAM 결합체 중에서 일부 6SL-PAMAM 결합체(S3-G4, S4-G4, S7-G5 및 S8-G5) 만이 HA 단백질과 높은 결합 친화도를 나타냄을 확인하였다.
특히, 6SL 리간드 간의 간격이 2.4 nm 이상인 6SL-PAMAM 결합체(S3-G4, S4-G4 및 S7-G5)는 모두 HA 단백질과 높은 결합친화도를 나타냄을 확인하였다.
반면, 6SL 리간드 간의 간격이 1.9 nm 보다 작은 6SL-PAMAM 결합체(S2-G3, S6-G4, S9-G5 및 S10-G5)는 HA 단백질과 현저히 낮은 결합친화도를 나타내었고, 이들의 대부분(S6-G4, S9-G5 및 S10-G5)은 상기 6SL 리간드 간의 간격이 2.4 nm 이상인 6SL-PAMAM 결합체(S3-G4, S4-G4 및 S7-G5) 보다도 더 많은 6SL 리간드를 포함하고 있음에도 불구하고, HA 단백질과 현저히 낮은 결합친화도를 나타냄을 확인하였다.
한편, HA 삼량체의 결정을 분석한 결과, H1N1의 삼량체형 HA 내부에 존재하는 6SL 결합 부위간의 거리가 약 4 nm인 것으로 확인되었다(도 3b).
따라서, 상기 결과를 종합하면, 상기 6SL-PAMAM 결합체에 포함된 6SL 리간드 간의 간격이 2.4 내지 4.0nm인 경우, HA 단백질에 효과적으로 결합할 수 있음을 알 수 있었다.
또한, 상기 6SL-PAMAM 결합체는 G4 및 G5의 에틸렌 디아민 코어를 포함하는 형태가 바람직하고, 상기 에틸렌 디아민 코어의 종류에 따라, 적절한 수준의 6SL 리간드를 포함함이 바람직함을 알 수 있었다.
구체적으로, G4를 포함하는 6SL-PAMAM 결합체는 약 20 내지 30개의 6SL 리간드를 포함함이 바람직하고, G5를 포함하는 6SL-PAMAM 결합체는 약 30 내지 40개의 6SL 리간드를 포함함이 바람직함을 알 수 있었다.
상기 6SL-PAMAM 결합체 중에서 S3-G4 및 S6-G4를 사용하여, 상기 분석내용을 도식화하였다(도 3c).
도 3c에서 보듯이, 약 3.1nm의 6SL 리간드 간격을 갖는 S3-G4는 H1N1 바이러스의 표면항원인 HA 단백질에 존재하는 리간드 결합부위 사이의 간격과 상기 S3-G4에 포함된 6SL 리간드 간격이 유사하므로, 상기 S3-G4는 H1N1 바이러스의 표면항원인 HA 단백질에 강하게 결합할 것으로 분석되었다. 이에 반하여, 약 1.6nm의 6SL 리간드 간격을 갖는 S6-G4는 H1N1 바이러스의 표면항원인 HA 단백질에 존재하는 리간드 결합부위 사이의 간격과 상기 S6-G4에 포함된 6SL 리간드 간격이 상이하므로, 상기 S6-G4는 H1N1 바이러스의 표면항원인 HA 단백질에 약하게 결합할 수 밖에 없을 것으로 분석되었다.
실시예
5: 전자현미경 분석
공지된 방법으로 인플루엔자 바이러스(A/California/07/2009)를 정제하였다.
대략적으로, 인플루엔자 바이러스(A/California/07/2009)에 감염된 계란의 난포액을 수집하고 원심분리(3000 rpm, 10 분) 하여 상등액을 수득하였다. 상기 수득한 상등액을 고속원심분리(19,000 x g, 2시간)하여, 침전물을 수득하였다. 상기 수득한 침전물을 TNE 완충액(50 mM Tris-HCl (pH 7.4), 100 mM NaCl 및 1 mM EDTA) 1㎖에 현탁시키고, 3 단계 불연속 수크로스 구배 (50, 40 및 30% 수크로스를 포함하는 TNE 완충액 2㎖) 조건하에서 원심분리하여(36,000 x g, 2시간), 바이러스 분획을 수득하였다. 상기 수득한 바이러스 분획을 PBS에 현탁시키고, 다시 원심분리하여(100,000 x g, 1시간, 4℃), 침전물을 수득한 다음, 이를 PBS 2㎖에 현탁시켰다. 상기 현탁된 바이러스 용액 20㎕와 S3-G4 용액(100 nmol/㎖) 20㎕를 혼합하고, 이를 가속 전압 120kV에서 작동되는 Tecnai G2 spirit TEM(FEI company, Hillsboro, OR)에 적용하여 TEM(Transmission electron microscopy) 분석을 수행하였다(도 4). 이때. 대조군으로는 상기 바이러스 용액 5㎕를 공기 중에서 3분 동안 글로우 방전시킨 탄소코팅 그리드에 가한 후, 2% 우라닐아세테이트를 사용하여 염색한 것을 사용하였다.
도 4에서 보듯이, 약 7nm의 구형(좌측상단 패널)의 S3-G4 입자와, 상기 S3-G4의 결합에 의해 H1N1의 표면에 형성된 소포형성(우측하단 패널)을 확인하였으나, 상기 소포형성은 6SL 리간드가 포함되지 않은 G4 PAMAM 덴드리머를 처리한 경우에는 확인되지 않았다(좌측하단 패널). 상기 소포형성은 S3-G4의 표면 응집에 의한 것으로 예상되며, 이로 인하여 바이러스의 부착, 세포내 진입 및 엔도좀 융합/방출 과정을 억제할 것으로 분석되었다.
실시예
6: 바이러스 감염과정 분석
먼저, 인플루엔자 바이러스(A/PR/8/34)를 정제하고, 1㎖ PBS에 현탁시켜서 바이러스 용액을 수득하였다. 상기 바이러스 용액에 100μM의 R18 용액(6 ㎕)을 가하고, 1시간 동안 실온에서 반응시켜서 R18로 단독 표지하거나; 또는 상기 바이러스 용액에 DiOC18(33μM)과 R18(67μM)를 포함하는 에탄올 용액(6㎕)을 가한 다음, 실온에서 1시간 동안 서서히 진탕시키면서 반응시켜서 DiOC18 및 R18로 이중표지하였다. 반응이 종료된 용액을 0.22㎛ 멤브레인 필터(Millipore)를 사용하여 여과하였다.
다음으로, MDCK 세포를 37℃ 및 5% CO2 조건에서 24시간 동안 배양하였다. 상기 배양된 세포에 상기 여과된 바이러스 용액을 가하고, 4℃에서 20분 동안 반응시켰으며, 반응이 종료된 후, 액상성분을 제거하고, 상기 세포를 차가운 PBS로 3회 세척하였다. 상기 세포를 37℃ 및 5% CO2 조건에서 488 및 594 nm 파장의 공초점 현미경으로(LSM700, Carl Zeiss) 촬영하였다(도 5).
도 5에 도시된 R18로 단독 표지된 바이러스를 이용한 분석결과에서 보듯이, MDCK 세포의 표면에 R18 표지된 바이러스가 부착되는 것을 S3-G4가 억제함을 확인하였다.
또한, 도 5에 도시된 DiOC18과 R18로 함께 표지된 바이러스를 이용한 분석결과에서 보듯이, R18 및 DiOC18로 표지된 대조군 바이러스를 처리한 경우에는 녹색 및 노란색 바이러스 입자가 확인되었으나, S3-G4를 전처리한 바이러스를 처리한 경우에는 녹색 및 노란색 바이러스 입자가 확인되지 않음을 확인하였다.
따라서, S3-G4가 바이러스의 세포내 진입에 관여함을 알 수 있었다.

7:
면역형광분석
MDCK 세포를 48웰 플레이트에 접종하고, S3-G4(10 또는 100 nmol/㎖)를 처리하거나 또는 처리하지 않은 조건에서, 다양한 H1N1 바이러스(A/NWS/33, A/Puerto Rico/8/43 and A/California/07/2009) 또는 H3N2 바이러스(A/Shandong/9/93)를 37℃에서 24시간 동안 감염시켰다. 상기 감염된 세포에 4% 파라포름알데히드를 처리하고 실온에서 10분동안 고정시킨다음, 상기 고정된 세포를 차가운 PBS로 세척하고, 0.25 % Triton X-100를 포함하는 PBS를 가하여 10분동안 반응시킨 후, 다시 PBS로 5분씩 3회 세척하였다. 상기 세척된 세포에 인플루엔자 바이러스 A NP-특이적 단클론 항체를 1시간 동안 처리한 다음, Alexa 488-염소 항-마우스 2차 항체를 가하여 1시간 동안 반응시키고, 역상 형광현미경으로 촬영하였다(도 6). 이때, 상기 세포의 핵은 NucBLue fixed cell stain(Life technologies)을 사용하여 대조염색하고, 상기 세포의 액틴은 ActinRed 555 ReadyProbes Reagent(Life technologies)를 사용하여 염색하였다.
도 6에서 보듯이, H1N1에 감염된 MDCK 세포(대조군)에서는 녹색으로 표시된 NP 단백질의 발현수준이 급격히 증가하였으나, S3-G4 결합체를 처리한 경우에는 NP 단백질의 발현수준이 낮은 수준을 나타냄을 확인하였다

실시예
8-1: 인플루엔자 바이러스에 감염된 마우스의 제작
6주령의 18-20g의 자성 무균 BALB/c 마우스를 사용하여, 6SL-PAMAM 결합체의 인플루엔자 바이러스 감염 억제능을 분석하였다.
상기 마우스에 Avertin(375 mg/kg)을 복강주사하여 마취시키고, S3-G4를 비강투여하였으며, 10분 후에 104 EID50(50% egg infective dose)의 H1N1 바이러스(A/NWS/33)를 비강투여하여 감염시켰다. 이때, S3-G4 및 바이러스의 투여수준에 따라, 4개 시험군으로 분류하였다: 양성대조군(바이러스 단독투여), 음성대조군(S3-G4 단독투여), 비교군(PBS 단독투여) 및 실험군(바이러스 및 50μmol/kg S3-G4 투여). 상기 각 시험군의 마우스를 14일 동안 사육한 다음, 이후의 실험을 수행하였다.

상기 14일 동안 사육한 각 마우스로부터 폐를 적출한 후, 이의 파쇄물을 수득하고, 원심분리(1400 x g, 4℃, 20분)하여 상등액을 수득하였으며, 상기 상등액에 PBS를 가하여 10배 부피로 희석하였다. 상기 희석된 상등액을 MDCK 세포에 처리하여, 상등액에 포함된 바이러스의 역가를 TCID50(tissue culture infective dose)로 산출하였다(도 7a).
도 7a에서 보듯이, S3-G4를 투여한 마우스의 폐에서 검출된 바이러스 역가는 S3-G4를 투여하지 않은 대조군의 것 보다도 10배 낮은 수준을 나타냄을 확인하였다.

폐조직의
조직병리학적 분석
상기 14일 동안 사육한 각 마우스로부터 폐를 적출한 후, 이를 10% 포르말린으로 고정시키고, 이의 조직박편을 수득한 다음, 헤마톡실린과 에오신으로 염색하고, 이의 조직병리학적 검사를 수행하였다(도 7b).
도 7b에서 보듯이, 양성대조군(H1N1)의 경우 폐의 곁에있는 기관지 영역에서 염증세포인 단핵구와 림프구의 침윤이 확인되었고, 폐포벽의 비후와 중증도의 출혈이 확인되었으며, 병변은 전엽 전체에 퍼져있음을 확인하였다. 이에 반하여, 실험군(H1N1+S3-G4)의 경우 염증성 병변이 감소되었고, 폐의 곁에있는 기관지 영역과 폐포 영역에서 단핵구와 림프구의 침윤이 감소됨을 확인하였다.

사이토카인 수준 분석
상기 14일 동안 사육한 각 마우스로부터 폐를 적출하고, 이의 파쇄물에 포함된 사이토카인(IL-6, IL-10, TNF-α, CXCL1, CXCL2 또는 CCL2)의 수준을 측정하고 비교하였다(도 7c).
도 7c에서 보듯이, TNF-α, IL-10, CXCL1 및 CXCL2의 수준은 양성대조군(H1N1)과 실험군(H1N1+S3-G4)에서 별다른 차이를 나타내지 않았으나, IL-6 및 CCL2의 수준은 양성대조군(H1N1)에 비하여, 실험군(H1N1+S3-G4)에서 유의하게 감소됨을 확인하였다.

8-5: 마우스의 생존율 분석
상기 14일 동안 사육한 각 마우스의 생존율 변화를 분석하였다(도 7d).
도 7d에서 보듯이, 음성대조군(S3-G4(50μmol/kg)) 및 비교군(PBS)은 사육기간이 경과하여도 생존율이 변화되지 않았으나, 양성대조군(H1N1)의 경우 6일째 부터 생존율이 감소하기 시작하여 10일이 경과된 시점에서 모두 사멸함을 확인하였다. 이에 반하여, 실험군(H1N1+S3-G4)의 경우 8일째에 생존율이 약 75%로 감소되었으나, 이후에는 상기 생존율이 변화되지 않음을 확인하였다.

8-6: 마우스의 체중변화 분석
상기 14일 동안 사육한 각 마우스의 생존율 변화를 분석하였다(도 7e).
도 7e에서 보듯이, 음성대조군(S3-G4(50μmol/kg)) 및 비교군(PBS)은 사육기간이 경과함에 따라, 체중이 서서히 증가하는 경향을 나타내었으나, 양성대조군(H1N1)의 경우 모두 사멸하기 직전인 9일까지 사육기간이 경과함에 따라 체중이 감소함을 확인하였다. 이에 반하여, 실험군(H1N1+S3-G4)의 경우 9일까지 사육기간이 경과함에 따라 체중이 감소하였으나, 이후에는 사육기간이 경과함에 따라 체중이 회복됨을 확인하였다.

내성 바이러스에 대한
면역형광분석
일반적으로 사용되는 NA 억제성 항 바이러스제의 사용에 의해 오셀타미비르(oseltamivir) 내성 돌연변이가 증가됨이 보고되었는 바, 본 발명에서 제공하는 6SL-PAMAM 결합체의 일종인 S3-G4가 오셀타미비르 내성 H1N1 바이러스의 감염을 억제할 수 있는지 확인하고자 하였다.
대략적으로, 오셀타미비르 및 페라미비르(peramivir) 치료를 받았으나, 입원 중 사망한 1세 여성 환자에게서 분리된 오셀타미비르 내성 H1N1 바이러스(A/Gyeongnam/1820/2009)에 대한 S3-G4의 감염억제능을 면역형광분석방법을 이용하여 평가하였다(도 8a).
도 8a에서 보듯이, 10 nmol/㎖의 S3-G4는 오셀타미비르 내성 H1N1 바이러스의 감염을 90% 억제함을 확인하였다.

내성 바이러스에 대한 MN(

) 분석
바이러스로서 오셀타미비르 내성 H1N1 바이러스(A/Gyeongnam/1820/2009)를 사용하고, 6SL-PAMAM 결합체로서 S3-G4를 사용하는 것을 제외하고는, 상기 실시예 3의 방법을 수행하여, 오셀타미비르 내성 H1N1 바이러스(A/Gyeongnam/1820/2009) 대한 S3-G4의 감염억제 활성을 분석하였다(도 8b).
도 8b에서 보듯이, H1N1 바이러스(A/California/04/2009)에 대한 S3-G4의 억제활성(IC50 = 3.4 μM)과 유사한 수준으로, 오셀타미비르 내성 H1N1 바이러스(A/Gyeongnam/1820/2009)에 대하여도 S3-G4가 억제활성(IC50 = 5.8 μM)을 나타냄을 확인하였다.

항 바이러스제
내성 바이러스에 대한 플라크 감소 분석
S3-G4를 처리하거나 또는 처리하지 않은 H1N1 바이러스(A/Gyeongnam/1820/2009)를 MDCK 세포에 가하여 1시간 동안 감염시키고, PBS로 세척하여 결합되지 않은 바이러스를 제거한 다음, 다양한 농도의 오셀타미비르를 처리하였다, 이어, 상기 세포를 아가로스 고체배지에서 배양하고, 배양된 고체배지에 1% 아가로스를 포함하는 중층배지를 중첩시킨 다음, 37℃에서 48시간 동안 배양하였다. 배양이 종료된 후, 하부의 고체배지를 1% crystal violet으로 염색하고, 플라크를 계수하였다(도 8c).
도 8c에서 보듯이, 오셀타미비르를 단독으로 투여한 경우에 비하여, 오셀타미비르와 S3-G4를 병용 투여할 경우, 오셀타미비르 내성 H1N1 바이러스에 감염된 MDCK 세포에서 플라크 형성이 유의하게 감소됨을 확인하였다.
상기 결과를 종합하면, 항 바이러스제에 대하여 내성을 나타내는 바이러스에 대하여도, 본 발명의 6SL-PAMAM 결합체가 억제활성을 나타냄을 알 수 있었다.
이상의 설명으로부터, 본 발명이 속하는 기술분야의 당업자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시 예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로서 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (19)
- 코어(core)와 그의 표면에 결합된 시알산, 시알릴락토스 또는 이들의 유도체를 포함하고, 상기 시알산, 시알릴락토스 또는 이들의 유도체는 서로 동일한 간격으로 배열된 것을 특징으로 하는, 결합체.
- 제1항에 있어서,상기 코어는 폴리아미도아민, 폴리하이드록시부티레이트, 폴리하이드록시발레르에이트, 폴리라이신, 폴리락트산, 폴리글리콜리드, 폴리카프로락톤, 폴리프로필렌퓨머레이트, 폴리다이옥세논, 폴리뉴클레오티드, 이들의 공중합체 및 이들의 조합으로 구성된 군으로부터 선택되는 물질로 구성된 것인, 결합체.
- 제1항에 있어서,상기 코어는 덴드리머인 것인, 결합체.
- 제1항에 있어서,상기 코어는 3.0 내지 6.0 nm의 직경을 갖는 것인, 결합체.
- 제1항에 있어서,상기 코어는 3.5 내지 5.5 nm의 직경을 갖는 것인, 결합체.
- 제1항에 있어서,상기 코어는 4.0 내지 6.0 nm의 직경을 갖는 것인, 결합체.
- 제1항에 있어서,상기 코어는 4.5 내지 5.4 nm의 직경을 갖는 것인, 결합체.
- 제1항에 있어서,상기 시알산 유도체는 시알릴올리고당(sialyloligosaccharide)인 것인, 결합체.
- 제1항에 있어서,상기 시알산, 시알릴락토스 또는 이들의 유도체는 3'-시알릴락토스(3'-Sialyllactose), 6'-시알릴락토스(6'-Sialyllactose), 시알릴락토-N-테트라오스(sialyl lacto-N-tetraose), 디시알릴락토-N-테트라오스(disialyl lacto-N-tetraose) 및 이들의 조합으로 구성된 군으로부터 선택되는 화합물인 것인, 결합체.
- 제1항에 있어서,상기 시알산, 시알릴락토스 또는 이들의 유도체는 코어의 반응기와 결합하거나 또는 링커를 통해 코어와 결합하는 것인, 결합체.
- 제1항에 있어서,상기 시알산, 시알릴락토스 또는 이들의 유도체는 1.0 내지 4.0 nm의 간격으로 배열된 것인, 결합체.
