精製工程(工程2)では、反応工程(工程1)で得た第1液L1と抽剤とを混合して抽剤にDHI類を抽出した油性の第2液L2を得る。
溶媒置換工程(工程3)では、精製工程(工程2)で得た第2液L2と水Wとを混合して抽剤を留去することにより溶媒置換を行い、水性の第3液L3であるDHI類を含有する水溶液又は水分散液のDHI類液を得る。また、生産時の効率性の観点からは、この溶媒置換と共に濃度調整を行うことが好ましい。
以下の実施例1~4及び比較例1~2のDHI水溶液の製造を行った。なお、いずれの工程も窒素雰囲気下にて行った。それぞれの内容については表1にも示す。
A槽として、アンカー翼を有する撹拌機及び温度調節用のジャケットが設けられた容量が300Lであり、材質がSUS製の反応槽を用い、そのA槽に窒素ガスを継続的に供給し、そこに窒素ガスを吹き込んで溶存酸素濃度を1.0mg/L以下にした水を仕込んだ。撹拌機を起動させてA槽内で水を撹拌しながらDOPAを投入して溶解させることにより、濃度が0.33質量%のDOPA水溶液を175L調製した。このとき、ジャケットによる加熱によりDOPA水溶液の液温を35℃に調整した。
A槽として傾斜パドル翼を有する撹拌機及び温度調節用のジャケットが設けられた容量が8m3であり、材質がSUS製の反応槽を用い、B槽として傾斜パドル翼を有し、材質がSUS製である撹拌機及び温度調節用のジャケットが設けられた容量が6m3の反応槽を用い、A槽に濃度が0.33質量%のDOPA水溶液を4.61m3調製し、また、酸化剤槽に濃度が16.8質量%のヘキサシアノ鉄(III)酸カリウム水溶液を0.61m3調製し、更に、抽剤の酢酸エチルを2.65m3添加したことを除いて実施例1と同様の操作を行うことにより得られたDHI水溶液を実施例2とした。
A槽として、アンカー翼を有する撹拌機及び温度調節用のジャケットが設けられた容量が30Lの、材質がポリテトラフルオロエチレン製である反応槽を用い、A槽に濃度が0.33質量%のDOPA水溶液を17.9L調製し、また、酸化剤槽に濃度が16.8質量%のヘキサシアノ鉄(III)酸カリウム水溶液を2.38L調製し、更に、抽剤の酢酸エチルを10.0L添加し、そして、A槽から排出した第2液を、一旦事前に窒素ガスによって容器内の酸素濃度を0体積%にした容量が1Lの密閉可能なガラス容器に供給して貯留しておき、第2液を排出したA槽を洗浄した後、ガラス容器からA槽に第2液を戻す、つまり、槽内を洗浄したA槽をB槽として用いたことを除いて、実施例1と同様の操作を行うことにより得られたDHI水溶液を実施例3とした。なお、A槽の槽内の洗浄は、第2液を排出したA槽に水を仕込んで60分間撹拌した後、その水を排出することにより行った。また、洗浄後のA槽の槽内の酸素濃度を0体積%とした。
A槽として、アンカー翼を有する撹拌機及び温度調節用のジャケットが設けられた容量が4m3であり、材質がSUS製の反応槽を用い、第2液を貯留する容器として事前に窒素ガスによって酸素濃度を0体積%にした、容量が200Lで窒素ガス流通可能なSUS製オープンドラムを用い、A槽に濃度が0.33質量%のDOPA水溶液を2.36m3調製し、また、酸化剤槽に濃度が16.7質量%のヘキサシアノ鉄(III)酸カリウム水溶液を0.31m3調製し、更に、抽剤の酢酸エチルを1.33m3添加したことを除いて実施例3と同様の操作を行うことにより得られたDHI水溶液を実施例4とした。
A槽に濃度が0.34質量%のDOPA水溶液を17.8L調製し、また、酸化剤槽に濃度が16.8質量%のヘキサシアノ鉄(III)酸カリウム水溶液を2.38L調製し、更に、抽剤の酢酸エチルを10.2L添加し、そして、B槽を用いずに、A槽のみを用いたことを除いて実施例3と同様の操作を行うことにより得られたDHI水溶液を比較例1とした。
A槽に濃度が0.33質量%のDOPA水溶液を175L調製し、また、酸化剤槽に濃度が16.8質量%のヘキサシアノ鉄(III)酸カリウム水溶液を25.2L調製し、更に、抽剤の酢酸エチルを100L添加し、そして、B槽を用いずに、A槽のみを用いたことを除いて実施例1と同様の操作を行うことにより得られたDHI水溶液を比較例2とした。
実施例1~4及び比較例1~2のそれぞれのDHI水溶液について、共立理化学研究所社製の水質測定用試薬セットNo.46全シアン(R-1試薬:酸性粉末、R-2試薬:アルカリ性粉末)及びデジタルパックテスト・マルチSPを用いてピクリン酸法により全シアン濃度を測定した。具体的には、以下の測定操作を行った。

Claims (15)
- 3-(3,4-ジヒドロキシフェニル)アラニン及びその誘導体の群から選ばれる1種又は2種以上とヘキサシアノ鉄(III)酸塩とを反応させてジヒドロキシインドール類を生成させた水性の第1液を得る工程1と、
前記工程1で得た前記第1液と抽剤とを混合して前記抽剤に前記ジヒドロキシインドール類を抽出した油性の第2液を得る工程2と、
前記工程2で得た前記第2液と水とを混合して前記抽剤を留去した水性の第3液を得る工程3と、
を含むジヒドロキシインドール類の製造方法であって、
前記工程2の前記ジヒドロキシインドール類の抽出をA槽で行い、前記A槽から前記第2液を排出した後、前記第2液を、B槽に供給し、前記B槽において前記工程3を行う製造方法。 - 前記B槽が、前記A槽とは別の液槽である、請求項1に記載された製造方法。
- 前記B槽が槽内を洗浄した液槽である、請求項2に記載された製造方法。
- 前記B槽が、前記工程2を行って前記第2液を排出した後に槽内を洗浄した前記A槽である、請求項1に記載された製造方法。
- 前記B槽の槽内の洗浄を水洗で行う、請求項3又は4に記載された製造方法。
- 前記抽剤がオクタノール/水分配係数が0.90未満の有機溶剤を含む、請求項1乃至5のいずれかに記載された製造方法。
- 前記抽剤が酢酸エチルを含む、請求項6に記載された製造方法。
- 前記A槽の材質がステンレスである、請求項1乃至7のいずれかに記載された製造方法。
- 前記B槽の材質がステンレスである、請求項1乃至8のいずれかに記載された製造方法。
- 初回の前記第2液への水の添加を、前記A槽から排出して前記B槽に供給されるまでの間の前記第2液に対して行う、請求項1乃至9のいずれかに記載された製造方法。
- 前記水の添加量が、前記第2液の体積に対して、50体積%以上、150体積%以下である、請求項1乃至10のいずれかに記載された製造方法。
- 前記抽剤を留去させるときの液温が、40℃以上、90℃以下である、請求項1乃至11のいずれかに記載された製造方法。
- 前記第2液への水の添加及び前記抽剤の留去を複数回行う、請求項1乃至12のいずれかに記載された製造方法。
- 前記水の添加及び前記抽剤の留去の後に前記B槽に残留した液体に水及び水溶性溶媒を添加して濃度調整することにより前記第3液を得る、請求項1乃至13のいずれかに記載された製造方法。
- 前記水溶性溶媒がエタノールを含む、請求項14に記載された製造方法。
