Abstract
본 발명은 고순도, 고수율 및 안정성을 갖는 홍합접착단백질을 얻을 수 있는 홍합접착단백질의 생산 방법에 관한 것으로, 미생물 배양을 이용하여, 홍합접착단백질을 발현시키는 단계; 상기 미생물로부터 상기 홍합접착단백질을 포함하는, 봉입체를 수득하는 단계; 및 상기 봉입체로부터 유기산, 알코올, chaotropic agent 및 설폭사이드 화합물 중 하나 이상의 추출 용매를 사용하여, 상기 홍합접착단백질을 수득하는 단계를 포함하는 홍합접착단백질의 생산 방법에 관한 것이다.
Description
본 발명은 홍합접착단백질의 생산 방법에 관한 것으로, 특히 고순도, 고수율 및 안정성을 갖는 홍합접착단백질을 얻을 수 있는 홍합접착단백질의 생산 방법에 관한 것이다.
홍합접착단백질은 도파(L-dihydroxylalanine, DOPA)가 풍부한 단백질로, 접착력이 우수하여, 다양한 표면에 강력한 접착력을 가지며, 인간세포를 공격하지 않고, 면역반응을 일으키지 않는 것으로 알려져 생체조직의 접착 등 의료분야에 응용 가능성이 크다.
하지만, 종래의 홍합접착단백질을 배양 및 정제하는 방법은, 단백질의 회수량이 높지 않고 높은 농도의 산과 염을 이용한 정제방법으로 인하여 회수량이 불규칙하고, 말단에 생리활성 펩티드를 포함한 홍합접착단백질의 경우 정제 과정에서 손실되는 현상이 나타났다. 이는 대한민국 공개특허 제10-2017-0017499호에도 개시되어 있다.
이에 따라, 단백질은 단백질 고유의 성질에 따라 정제 효율이 달라지기 때문에 단백질의 손실을 최소화하는 정제과정의 개발이 필요하게 된다. 종래 대장균을 이용한 유전자 재조합 기술을 이용한 기술이 개발되었으나, 친화 크로마토그래피에 의한 분리로 비용이 많이 든다는 단점이 있으며, 대한민국 등록특허공보 제10-0868047호, 대한민국 등록특허공보 제10-0872847호에도 친화 크로마토그래피 공정으로 고비용이 든다는 단점을 가지는 기술이 개시되어 있다.
본 발명은 상술한 문제를 해결하기 위하여 안출된 것으로, NaCO3를 이용하여 pH를 9.0 내지 10.0으로 맞춘 후 원심분리하여 정제함으로써 고순도, 고효율 및 안정성을 갖는 홍합접착단백질을 얻을 수 있는 홍합접착단백질의 생산방법을 제공하는데 그 목적이 있다.
본 발명의 다른 목적은, Acetic acid, n-propanol 및 DMSO을 사용하여 추출하며, 이온크로마토그래피 공정을 진행하는 단계를 거쳐 고순도, 고수율 및 안정성을 가진 홍합접착단백질을 얻을 수 있는 홍합접착단백질의 생산방법을 제공하는 것에 있다.
상기 목적을 달성하기 위하여 본 발명은, 미생물 배양을 이용하여, 홍합접착단백질을 발현시키는 단계; 상기 미생물로부터 상기 홍합접착단백질을 포함하는, 봉입체를 수득하는 단계; 및 상기 봉입체로부터 유기산, 알코올, chaotropic agent 및 설폭사이드 화합물 중 하나 이상의 추출 용매를 사용하여, 상기 홍합접착단백질을 수득하는 단계를 포함하는, 홍합접착단백질의 생산 방법을 제공한다.
일 실시예에 따르면, 상기 유기산은 아세트산(acetic acid)일 수 있다.
일 실시예에 따르면, 상기 알코올은 n-propanol일 수 있다.
일 실시예에 따르면, 상기 chaotropic agent는 Guanidine hydrochloride일 수 있다.
일 실시예에 따르면, 상기 설폭사이드 화합물은 DMSO(Dimethyl sulfoxide)일 수 있다.
일 실시예에 따르면, 상기 추출 용매는 상기 유기산, 상기 알코올, 상기 chaotropic agent 및 상기 설폭사이드 화합물 중 하나 이상을 포함하며, 추출 용매 총 중량 대비, 상기 유기산은 15 내지 50 중량%, 상기 알코올은 5 내지 15 중량%, 상기 설폭사이드 화합물은 5 중량%로 포함될 수 있다.

본 발명은 미생물의 파쇄액에 염 및 NaCO3를 사용하여 pH를 높여줌으로써 재조합 단백질의 불용성을 높여 봉입체로의 회수율을 90%이상으로 제공하며, Chaotropic agent인 n-propanol을 첨가하여, acetic acid로만 추출하던 종래의 방식에 비해 높은 수율로 단백질을 추출할 수 있으며, 이온 크로마토그래피(ion chromatography) 방법을 사용하여 불순물을 제거하고 단백질의 순도를 높여주어 기존의 고농도 산과 염을 사용한 정제 방법보다 회수되는 단백질의 순도와 안정성이 높은 홍합접착단백질을 얻을 수 있다.
도 1은 본 발명의 일 실시예에 의한 홍합접착단백질 생산 방법의 개요를 나타내는 사진이다.

이하, 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 발명을 더욱 상세히 설명한다.
본 발명의 홍합접착단백질에 관한 생산 방법은, (a) 미생물 배양을 이용하여, 홍합접착단백질을 발현시키는 단계; (b) 상기 미생물을 파쇄하여, 파쇄액을 수득하는 단계; (c) 상기 파쇄액에 염(salt) 및 pH조절제 중 하나 이상을 사용하여 원심분리하여, 상기 봉입체를 수득하는 단계; (d) 상기 봉입체로부터 추출 용매를 이용하여, 상기 홍합접착단백질 추출 용액을 수득하는 단계; (e) 상기 홍합접착단백질 추출 용액에 아세톤을 첨가하여, 홍합접착단백질 침전물을 형성시키는 단계; 및 (f) 상기 홍합접착단백질 침전물을 증류수에 용해시킨 뒤, 이온 크로마토그래피로 정제하여, 상기 홍합접착단백질을 수득하는 단계;를 포함하여, 기존의 고농도 산과 염을 사용한 정제 방법보다 고순도, 고수율 및 안정성을 가진 홍합접착단백질 제공할 수 있는 생산 방법에 관한 것으로 구체적인 방법은 도 1에 도식화 되어 있으며, 구체적인 설명은 하기와 같다.
(a) 미생물 배양을 이용하여, 홍합접착단백질을 발현시키는 단계
본 발명의 홍합접착단백질의 생산 방법은 기본적인 효모 배지에서 홍합접착단백질 및 그와 연결된 생리활성 motif(모티프)를 갖는 홍합접착단백질을 미생물(예를 들어, 대장균)을 이용하여 과다발현시켜 배양액을 수득한다.
또한, 본 발명에서 홍합접착단백질 발현 벡터가 삽입된 미생물, 구체적인 예를 들어, E.coli BL21(DE3)는 통상의 ampicillin을 포함하는 LB 배지 혹은 탄소원과 질소원을 포함하는 배지에서 IPTG를 사용하여 홍합접착단백질이 과다 발현되며, 미생물 배양액을 수득한다.

종래의 재조합단백질 파쇄 방법에는 기계적인 방법 및 비기계적인 방법이 있다. 본 발명의 파쇄 방법은 기계적인 방법에 속하며, 압착기(presses)를 이용하는 고압 파쇄 방법으로 실험실 규모에서 많이 사용되는 스테인레스 스틸로 된 속이 빈 실린더 모양이다. 여기에 파쇄 대상 미생물을 채운 후 고압 하에서 실린더 바닥에 있는 니들 밸브(needle valve)를 통해 대기압 상태로 압출하면 파쇄된다.
본 발명에서의 파쇄 방법은 고압균질기를 사용하며, 최소 800bar이상의 조건에서 미생물을 파쇄하여 홍합접착단백질을 포함하는 봉입체를 포함한 세포 파쇄액을 얻는다.
(c) 상기 파쇄액에 염(salt) 및 pH조절제 중 하나 이상을 사용하여 원심분리하여, 상기 봉입체를 수득하는 단계
미생물을 이용하여 홍합접착단백질을 발현시킬 시, 목적하는 단백질이 대장균 내 용해성(soluble), 비용해성(insoluble) 형태로 같이 발현되어 존재한다. 이는 미생물 파쇄 및 원심 분리 후 정제 타겟을 비용해성 형태 만으로 제한하여, 상층액 내에 존재하는 용해성을 갖는 형태의 단백질은 버려지게 된다.
이에, 파쇄액에 염으로서, 소듐 클로라이드(NaCl) 및 암모늄 설페이트(Ammonium sulfate); 및 pH 조절제로서, 소듐 카보네이트(Sodium carbonate) 및 소듐 보레이트(Sodium borate) 중 하나 이상을 첨가하여, 홍합접착단백질의 등전점인인 pI 9.8에 가깝게, pH 9.0 내지 10.0으로 조절 하여, 용해성 형태의 홍합접착단백질 또한, 비용해성 형태로 전환 시킬 수 있으며, 원심 분리 후 목적하는 단백질을 불용성 응집체 형태로의 회수를 극대화 하여 90% 이상의 봉입체를 회수할 수 있다.
(d) 상기 봉입체로부터 추출 용매를 이용하여, 상기 홍합접착단백질 추출 용액을 수득하는 단계
봉입체에서, 종래의 알려진 기술인 25% 아세트산(Acetic acid)을 사용하여 목적하는 단백질의 추출 시, 추출 효율에 한계가 있다.
이를 개선하기 위하여, 본 발명은 유기산, 알코올, chaotropic agent 및 설폭사이드 화합물 중 하나 이상의 추출 용매를 이용하여, 상기 홍합접착단백질 추출 용액을 수득하며, 기존의 방법 대비 최대 50% 가량 추출 효율 증가를 확인하였다. 이에 대한 자세한 사항은 하기 표 1에서 control(아세트산 25 중량%)의 추출 효율을 100%로 보았을 경우 No.3의 조성에서 150%까지 추출 효율이 증가되는 실험을 통해 알 수 있다.
일 실시예를 들어, 상기 유기산은 아세트산이고, 상기 알코올은 n-propanol이고, 상기 chaotropic agent는 Guanidine hydrochloride이고, 상기 설폭사이드 화합물은 DMSO(Dimethyl sulfoxide)일 수 있다. 상기 설폭사이드 화합물은 단백질의 unfolding을 유도하여, 추출 효율을 향상시킬 수 있다.
상기 추출 용매는 상기 유기산, 상기 알코올, 상기 chaotropic agent 및 상기 설폭사이드 화합물 중 하나 이상을 포함하며, 추출 용매 총 중량 대비, 상기 유기산은 15 내지 50 중량%, 상기 알코올은 5 내지 15 중량%, 상기 설폭사이드 화합물은 5 중량%로 포함될 수 있다.
하기 표 1을 참조하면, 추출 용매로서 아세트산, n-propanol 및 DMSO 중 하나 이상을 혼합하여 봉입체(inclusion body)에 포함된 홍합접착단백질을 추출할 경우, 아세트산 단독으로 사용한 경우와 비교하여 현저히 우수한 추출 효율을 나타내는 것을 확인할 수 있다. 또한, 추출 용매로서 아세트산 30 중량%와 n-propanol 15 중량%를 혼합하여 사용하는 경우, 가장 높은 추출 효율을 나타내는 것을 확인할 수 있다.
(e) 상기 홍합접착단백질 추출 용액에 아세톤을 첨가하여, 홍합접착단백질 침전물을 형성시키는 단계
홍합접착단백질 추출 용액은 아세톤을 이용한 단백질 침전법을 이용하여, pellet 형태의 홍합접착단백 침전물로 회수할 수 있으며 또한 높은 농도로 존재하는 아세트산(acetic acid)의 제거 효과도 가질 수 있다.
종래의 기술된 방법은, 아세트산을 사용하여 추출된 홍합접착단백질을 동결 건조 후, 크로마토그래피 단계로 진입하였으며, 이에 따라 동결 건조에 소요되는 시간적 손실을 초래하였다. 반면, 아세톤을 이용한 단백질 침전법의 경우, 원심분리 후 재 용해 만으로 크로마토그래피 단계로 진입할 수 있는 장점을 가진다.
(f) 상기 홍합접착단백질 침전물을 증류수에 용해시킨 뒤, 이온 크로마토그래피로 정제하여, 상기 홍합접착단백질을 수득하는 단계
상기 홍합접착단백질 침전물을 증류수에 녹여 수용액 상태로 만든 뒤, 이온 크로마토그래피 방법을 이용하여 불순물을 제거할 수 있다. 상기 홍합접착단백질 침전물을 레진(resin) 충진된 컬럼에 흘려, 레진에 결합 시킨 후 1M NaCl을 넣어주게 되면 불순물이 제거되게 되며, 이후 100mM NaOH를 이용하여 상기 홍합접착단백질만 회수할 수 있으며, 회수된 홍합접착단백질은 탈염(desalting) 및 동결 건조될 수 있다.
실시예에 따른 홍합접착단백질의 생산 방법
사용 배지
본 실험에서 액체배지는 글루코스가 들어간 LB배지 (Luria-bertani media)와 직접 제조한 배지 조성 (glucose 20 g/L, yeast extract 5 g/L, potassium phosphate monobasic 5 g/L, potassium phosphate dibasic 3 g/L, ammonium sulfate 1.5 g/L, magnesium sulfate 0.5 g/L)을 사용하였으며 모든 배지는 50 ug/ml 농도의 ampicillin 항생제를 포함시켰다. 이는 본 실험에서 사용한 pET-22b(+) 벡터 내에 포함하고 있는 ampicillin 항생제 내성부위의 존재에 따른 것이다.
종배양 (Seed culture)
2리터 삼각 플라스크에 LB배지 1리터 이외에 추가로 10 gram/L의 glucose를 넣어준 후 멸균처리 하였다. 종균 stock 1ml과 항생제 (ampicillin 50 mg/L)를 넣어 준 후 overnight 으로 진탕 배양하였다. (200 rpm, 37 ℃)
본 배양 (Main culture)
12 리터 배양조 (Vessel)를 준비하여 직접 제조한 배지 9 리터 (glucose 20 g/L, yeast extract 5 g/L, potassium phosphate monobasic 5 g/L, potassium phosphate dibasic 3 g/L, ammonium sulfate 1.5 g/L, magnesium sulfate 0.5 g/L)와 소량의 anti-form (300 ~ 500 ul/L)을 넣어 준 후 pH probe와 DO meter를 셋팅 하고 밀봉한 후 멸균 처리 (121 ℃, 15 min) 하였다. 에어의 주입량은 1 VVM, 배양기의 온도는 37 ℃, 교반기의 속도는 800 rpm으로 맞춰준 후 종배양액 1리터를 넣어준 후 배양을 진행하였다. 배양액의 OD가 5~7 정도 되었을 때 단백질 발현 유도를 위해 IPTG를 0.5 mM 내지는 1mM이 되도록 넣어준 후 다시 3시간 내지 12시간 배양을 추가 진행하였다.
균체 회수 및 단백질 추출
배양액을 원심 분리하여 균체를 회수하고 균체를 증류수에 현탁 하여 균질화 시켰다. 고압균질기(High pressure homogenizer)를 사용하여 균질화 된 용액을 800 내지는 1000 bar에서 2회 파쇄한 후 0.5M의 NaCl과 1~2%의 sodium carbonate를 넣어주어 pH를 9.0 ~ 10.0으로 맞춰준 후 원심 분리하여 pellet을 회수하였다. Pellet에 30%의 Acetic acid와 15%의 n-propanol이 함유된 용액을 넣어서 균질화 시킨 후 30분 이상 고르게 섞어준 후 원심 분리하여 단백질이 추출된 상층액을 회수하였다.
단백질 정제
추출된 상층액의 2배~3배 볼륨의 acetone을 넣어주어 단백질을 침전시킨 후 원심 분리하여 pellet을 회수하고, 증류수에 희석시켜 준 후 acetic acid를 이용하여 pH를 5.0 이하로 맞춰주었다. 수용액을 원심 분리하여 상층액을 회수한 후 20mM sodium acetate buffer(pH 4.5)로 안정화 시킨 Cation exchange resin (SP sepharose)에 로딩하여 binding 시킨 후 동일 buffer로 씻어주고 1M NaCl이 첨가된 20mM sodium acetate buffer (pH 4.5) 용액을 흘려주어 불순물을 제거하였다. 이후 다시 20mM sodium acetate buffer (pH 4.5)로 컬럼을 씻어준 후 100mM NaOH 용액을 흘려주어 단백질을 용출시켜 고순도의 단백질을 회수하였다. 용출된 단백질은 acetic acid를 이용하여 pH를 5.0 이하로 맞춰주고 desalting 처리를 해준 후 동결건조 하였다.
본 발명의 홍합접착단백질의 생산 방법은, 미생물의 파쇄액에 염(salt) 및/또는 pH조절제를 사용하여 pH를 높여줌으로써 단백질의 불용성을 높여 봉입체(inclusion body)로의 회수율을 거의 100%로 높여주었으며, 유기산, 알코올, chaotropic agent 및 설폭사이드 화합물 중 하나 이상의 추출 용매를 사용하여 acetic acid로만 추출하던 종래의 방식에 비해 최대 50% 증가된 높은 수율로 단백질을 추출해 낼 수 있게 되었다. 또한, 종래 크로마토그래피 단계 진입 전 동결 건조 단계를 생략하고, 아세톤 침전 후 재 용해 함으로써 공정 중 시간 절감을 가능하게 하였다. 재용해 된 단백질을 이온 크로마토그래피(ion chromatography) 방법을 사용하여 불순물을 제거하고 단백질의 순도를 높여주었으며, 기존의 고농도 산과 염을 사용한 정제 방법보다 회수되는 단백질의 고순도와 고수율 및 안정성이 높은 효과를 확인하였다.
본 발명은 미생물의 파쇄액에 염 및 NaCO3를 사용하여 pH를 높여줌으로써 재조합 단백질의 불용성을 높여 봉입체로의 회수율을 90%이상으로 제공하며, Chaotropic agent인 n-propanol을 첨가하여, acetic acid로만 추출하던 종래의 방식에 비해 높은 수율로 단백질을 추출할 수 있으며, 이온 크로마토그래피(ion chromatography) 방법을 사용하여 불순물을 제거하고 단백질의 순도를 높여주어 기존의 고농도 산과 염을 사용한 정제 방법보다 회수되는 단백질의 순도와 안정성이 높은 홍합접착단백질을 얻을 수 있다.
Claims (11)
- 미생물 배양를 이용하여, 홍합접착단백질을 발현시키는 단계;상기 미생물로부터 상기 홍합접착단백질을 포함하는, 봉입체를 수득하는 단계; 및상기 봉입체로부터 유기산, 알코올, chaotropic agent 및 설폭사이드 화합물 중 하나 이상의 추출 용매를 사용하여, 상기 홍합접착단백질을 수득하는 단계를 포함하는, 홍합접착단백질의 생산 방법.
- 청구항 1에 있어서,상기 유기산은 아세트산인, 홍합접착단백질의 생산 방법.
- 청구항 1에 있어서,상기 알코올은 n-propanol인, 홍합접착단백질의 생산 방법.
- 청구항 1에 있어서,상기 chaotropic agent는 Guanidine hydrochloride인, 홍합접착단백질의 생산 방법.

- 청구항 1에 있어서,상기 홍합접착단백질을 수득하는 단계는,상기 봉입체로부터 상기 추출 용매를 이용하여, 상기 홍합접착단백질 추출 용액을 수득하는 단계;상기 홍합접착단백질 추출 용액에 아세톤을 첨가하여, 홍합접착단백질 침전물을 형성시키는 단계; 및상기 홍합접착단백질 침전물을 증류수에 용해시킨 뒤, 이온 크로마토그래피로 정제하여, 상기 홍합접착단백질을 수득하는 단계를 포함하는, 홍합접착단백질의 생산 방법.
