Description

金属錯体粒子及びそれを用いた免疫測定用試薬
 本発明は金属錯体粒子及びそれを用いた免疫測定用試薬に関する。
 従来、生体物質を含有する試料、例えば生体サンプル中のタンパク質等を測定又は精製する方法が知られている。例えば、タンパク質が結合し得る粒子表面に生体サンプル中のタンパク質等を結合させ、生体サンプル中の目的タンパク質等以外の不純物を除くために、粒子を洗浄しタンパク質等が結合した粒子を回収して、タンパク質の結合量を測定する方法やタンパク質解離溶液中に解離させて精製する方法が挙げられる。
 また、目的のタンパク質(抗原等)に対して、酵素で標識された抗体を作用させた後、当該酵素と発光試薬の反応による発光を光学的に測定することによって目的タンパク質を検出する方法(ELISA法)が広く利用されている。
 上記ELISA法においては、下記特許文献1、2には、磁力によって容易に分離、回収が可能であることから、磁性を有する粒子が用いられていることが開示されている。
 更に、磁性を有する粒子に蛍光を付与することで、蛍光標識を可能とした粒子が提案されている。例えば、特許文献3には、フェライト粒子表面に形成したポリマー層にユーロピウムキレート錯体を含侵させることで、磁性と蛍光を有する粒子が開示されている。
 また、特許文献3には、ユーロピウムキレート錯体を磁性粒子に内包させた水懸濁液を得る方法として、(1)具体例は示されてはいないが、ポリマー被覆磁性粒子の水懸濁液に、ユーロピウムキレート錯体を溶解させた水との親和性の高い低沸点非水系溶媒(好ましくはアセトン)溶液を添加し、ポリマー層を膨潤させると同時にユーロピウムキレート錯体も取り込ませた後、前記溶媒を蒸発させることで、ユーロピウムキレート錯体含有磁性粒子の水懸濁液を得る方法、(2)水を除去した微架橋ポリマー被覆磁性粒子を、ユーロピウムキレート錯体を溶解させたアセトン溶液に添加し、ポリマー層を膨潤させると同時にユーロピウムキレート錯体も取り込ませた後、アセトンを蒸発させ、水を添加することで、ユーロピウムキレート錯体含有磁性粒子の水懸濁液を得る方法が記載されている。
さらに、特許文献4には、(3)アルコール洗浄等で水を除いた微架橋ポリマー被覆磁性粒子を、ユーロピウムキレート錯体を溶解させたアセトン溶液に添加し、ポリマー層を膨潤させると同時にユーロピウムキレート錯体も取り込ませた後、水を添加し、アセトンを蒸発させることで、ユーロピウムキレート錯体含有磁性粒子の水懸濁液を得る方法が記載されている。

 しかしながら、特許文献3には、記載されている製法では、得られる磁性粒子の最大径が300nmであることから、粒子1個に含まれる蛍光体や磁性体量が少なく、磁石による分離の際の分離速度が遅く、また、フェライトによる蛍光の吸収があるため、十分な蛍
光が得られず、感度が低いという問題があった。さらに特許文献3には、(1)の方法は具体的な例は無く、また、好ましいとされているアセトンを用いた場合、ユーロピウムキレート錯体のアセトン溶液をポリマー被覆磁性粒子水懸濁液に添加した時点で、ユーロピウム錯体が析出し、ポリマー層に必要量を取り込むことは困難であった。ユーロピウムキレート錯体が析出しないように、水に対して大量のアセトンを使用する場合、ユーロピウムキレート錯体の濃度が下がり充分に含有させることが出来なかった。(2)の方法では、アセトンを蒸発させる工程で、粒子同士が合着し、単粒子を得ることが困難であった。(3)の方法においても、水を添加した際、キレート錯体が析出し、取り除くことが極めて困難であった。
 上述の通り、磁石により分離/回収が迅速に行え、かつ、蛍光標識を可能とした、感度の高い磁性と蛍光とを具備した粒子は免疫測定用試薬としては実用化されていなかった。
 本発明は、集磁性に優れた、磁性と蛍光を併せ持つ、金属錯体粒子及びそれを用いた免疫測定用試薬を提供することを目的とする。
 本発明は以下の内容に関する。
(1)磁性体と、疎水性ポリマーを含む粒径調整組成物と、を備える金属錯体粒子であって、前記磁性体は、金属イオンと配位子からなる、磁性及び蛍光の両方を有する金属錯体を含み、前記金属錯体粒子中の前記粒径調整組成物の含有率が60w%未満である、金属錯体粒子。
(2)前記磁性体は蛍光を有さない磁性を有する金属錯体を含まない磁性体である、(1)に記載の金属錯体粒子。
(3)前記磁性体は前記金属錯体のみである、(1)に記載の金属錯体粒子。
(4)前記金属錯体を形成する金属イオンの磁気モーメントが7.5~10.7である、(1)~(3)のいずれか1つに記載の金属錯体粒子。
(5)前記金属錯体を形成する金属イオンがテルビウム、ジスプロシウム、ホルミウム、エルビウムのイオンからなる群から選択される少なくとも一種である、(1)~(4)のいずれか1つに記載の金属錯体粒子。
(6)前記金属錯体を形成する配位子がβ―ジケトン系配位子及び複素環系配位子の両方又はいずれか一方である、(1)~(5)のいずれか1つに記載の金属錯体粒子。
(7)前記金属錯体を形成する配位子が、(1)β―ジケトン系配位子は、ジベンゾイルメタン、又はテノイルフルオロアセトン、(2)複素環系配位子は、2,2´‐ビピリジル、2,2´:6,2´´-テルピリジン、又は1,10-フェナントロリン、
から選ばれる配位子である、(1)~(6)のいずれか1つに記載の金属錯体粒子。
(8)前記金属錯体粒子の平均粒径が1μm以上5μm以下である、(1)~(7)のいずれか1つに記載の金属錯体粒子。
(9)前記金属錯体中の前記金属錯体の含有量が、40w%以上98w%以下ある、(1)~(8)のいずれか1つに記載の金属錯体粒子。
(10)前記金属錯体粒子の表面に担持用物質を備える、(1)~(9)のいずれか1つに記載の金属錯体粒子。
(11)前記担持用物質が、検出対象物質と特異的に反応する抗体または抗原である、(1)~(10)のいずれか1つに記載の金属錯体粒子。
(12)(10)に記載の金属錯体粒子を含む、免疫測定用試薬。
 本発明によれば、集磁性に優れた、磁性と蛍光を併せ持つ、金属錯体粒子及びそれを用いた免疫測定用試薬を提供される。

 本発明者らは、上記課題を解決するべく鋭意検討した結果、複数ある希土類錯体の中に、磁性及び蛍光の両方を有する金属錯体が数種あることを知見した。そして、この金属錯体を粒径分布が揃った粒子として調製することにより、集磁性に優れ、かつ、蛍光発光特性を有する金属錯体粒子が得られることを見いだした。さらに、得られた粒子を免疫測定用試薬に用いることにより、高い集磁性と測定精度(視認性)の向上が図られ、上述の課題が解決されることとなった。以下に、実施形態を挙げて本発明の説明を行うが、本発明は以下の実施形態に限定されるものではない。
[金属錯体粒子]
 本発明は、磁性体と、疎水性ポリマーを含む粒径調整組成物と、を備え、
 磁性体は、金属イオンと配位子からなる、磁性及び蛍光の両方を有する金属錯体を含み、 金属錯体粒子中の前記粒径調整組成物の含有率が60w%未満である、金属錯体粒子に関する。金属錯体粒子は、磁性を有する物質として金属錯体のみを含有することが好ましい。
 本発明の金属錯体粒子は、磁性を有する金属錯体の含有率が高いため、高い集磁性を有すると同時に、粒子1個あたりの磁性量が均一となるため、磁力による分離の際、個々の粒子の磁力への応答が均一となり、分離の際のばらつきが小さくなる。更に、金属錯体が磁性の他に蛍光特性も有するため、この金属錯体粒子を免疫測定用試薬に用いることで、検出感度が高くなる。また、金属錯体種を変えることで、金属錯体粒子の蛍光スペクトルを変えることが可能となり、金属錯体粒子の色が変わるため、複数の検体を測定する際、試薬の取り違い等に起因する誤診断を回避できる。
本発明の金属錯体粒子は、磁性体と蛍光体をそれぞれ含有する粒子に比べ、同一金属錯体にて磁性と蛍光を有するため、磁性発現物質と蛍光発現物質との負の相互作用がなく、磁性、蛍光とも効果的に発現できる。また、本発明の金属錯体粒子は、金属錯体の高含有化が可能であるため、高い集磁性を発現することが可能である。さらに、本発明の金属錯体粒子は、粒径が揃っているので、磁石への応答性も均一となるため、均一な集磁性を発現する。
 上記粒径調整組成物は、重合性モノマー(以下、単に「モノマー」ともいう)を重合(重縮合も含む)して得られるポリマー(以下、単に「ポリマー」ともいう)を主成分とし、上記ポリマーは一種または二種以上の重合性モノマーのコポリマーも含む。
 上記ポリマーとしては、例えば、(不)飽和炭化水素、芳香族炭化水素、(不)飽和脂肪酸、芳香族カルボン酸、(不)飽和ケトン、芳香族ケトン、(不)飽和アルコール、芳香族アルコール、(不)飽和アミン、芳香族アミン、(不)飽和チオール、芳香族チオール、有機ケイ素化合物の1種以上の重合性モノマーからなるポリマーが挙げられる。
 上記ポリマーとしては、例えば、ポリエチレン、ポリプロピレン、ポリブチレン、ポリメチルペンテン、ポリブタジエン等のポリオレフィン類;ポリエチレングリコール、ポリプロピレングリコール、テトラメチレングリコール等のポリエーテル;ポリスチレン;ポリ(メタ)アクリル酸;ポリ(メタ)アクリル酸エステル;ポリビニルアルコール;ポリビニルエステル;ポリビニルエーテル;フェノール樹脂;メラミン樹脂;ベンゾグアナミン樹脂;アリル樹脂;フラン樹脂;(不)飽和ポリエステル;エポキシ樹脂;ポリシロキ
サン;ポリアミド;ポリイミド;ポリウレタン;テフロン(登録商標);アクリロニトリル/スチレン樹脂;スチレン/ブタジエン樹脂;ハロゲン化ビニル樹脂;ポリカーボネート;ポリアセタール;ポリエーテルスルホン;ポリフェニレンオキシド;ポリエーテルエーテルケトン;糖;澱粉;セルロース;ポリペプチド、及び、重合性官能基を有する種々の重合性モノマーを1種もしくは2種以上重合させて得られるポリマー等が挙げられる。これらのポリマーは単独で用いても、2種以上が併用されても良い。
 上記ポリマーは、ラジカル重合、イオン重合、配位重合、開環重合、異性化重合、環化重合、脱離重合、重付加、重縮合、付加縮合により製造される。
 上記重合性モノマーは、一種単独であってもよく、二種以上の任意の割合の組み合わせでもよい。また、二種以上の重合性モノマーを用いる場合、二種以上の重合性モノマーの組み合わせは、二種以上の単官能重合性モノマーの組み合わせ、二種以上の多官能重合性モノマーの組み合わせ、一種以上の単官能重合性モノマーと一種以上の多官能重合性モノマーとの組み合わせ、のいずれであってもよい。これらの重合体の分子量は、例えば80以上30,000以下であるが、特に限定されるものではない。尚、本発明及び本明細書において分子量とは、多量体については、ゲル浸透クロマトグラフィー(GPC)によりポリスチレン換算で測定される重量平均分子量をいうものとする。後述する、重量平均分子量は、下記測定により測定された値である。

 上記重合性モノマー(以下、単に「モノマー」ともいう)とは、一分子中に一つ以上の重合性官能基を含む化合物であれば特に限定されず、重合性官能基を一分子に一つ含む単官能モノマーであっても、重合性官能基を一分子中に二つ以上含む多官能モノマーであっても良い。また、一分子中に重合性官能基を一つ以上含むオリゴマーやプレポリマー等であっても良い。
 上記重合性官能基は、ラジカル重合性官能基、イオン重合性官能基、配位重合性官能基であってもよく、ラジカル重合性官能基が好ましい。重合反応の反応性の観点からは、エチレン性不飽和結合含有基、エポキシ基、オキセタン基、メチロール基等の重合性基を挙げることができ、エチレン性不飽和基がより好ましい。エチレン性不飽和基としては、(メタ)アクリロイルオキシ基、(メタ)アクリロイル基、ビニル基、芳香族ビニル基、アリル基を挙げることができ、(メタ)アクリロイル基、ビニル基、芳香族ビニル基がより好ましい。なお、本発明及び本明細書において、「(メタ)アクリロイル基」との記載は、アクリロイル基、とメタクリロイル基の少なくともいずれかの意味で用いるものとする。「(メタ)アクリロイルオキシ基」、「(メタ)アクリレート」、「(メタ)アクリル」等も同様である。多官能モノマーについては、化合物に含まれる重合性基の数は1分子中に2つ以上である。
 上記エチレン性不飽和基を有する単官能モノマーの具体例としては、特に限定されず、
 例えば、(メタ)アクリロイル基を有する単官能モノマーとしては、メチル(メタ)アクリレート、エチル(メタ)アクリレート、プロピル(メタ)アクリレート、ブチル(メタ)アクリレート、2-エチルヘキシル(メタ)アクリレート、ラウリル(メタ)アクリレート、セチル(メタ)アクリレート、ステアリル(メタ)アクリレート、シクロヘキシル(メタ)アクリレート、イソボルニル(メタ)アクリレート、2-ヒドロキシエチル(メタ)アクリレート、グリセロール(メタ)アクリレート、ポリオキシエチレン(メタ)アクリレート、グリシジル(メタ)アクリレート、トリフルオロメチル(メタ)アクリレート、ペンタフルオロエチル(メタ)アクリレート、3-(メタ)アクリロキシプロピルメチルジメトキシシラン、3-(メタ)アクリロキシプロピルトリメトキシシラン、3-(メタ)アクリロキシプロピルメチルジエトキシシラン、3-(メタ)アクリロキシプロピルトリエトキシシラン等の(メタ)アクリレートモノマーが挙げられる 
 ビニル基を有する単官能モノマーとしては、ジイソブチレン、イソブチレン、リニアレン、エチレン、プロピレン等のオレフィンモノマー;イソプレン、ブタジエン等の共役ジエンモノマー;メチルビニルエーテル、エチルビニルエーテル、プロピルビニルエーテル等のビニルエーテルモノマー;酢酸ビニル、酪酸ビニル、ラウリン酸ビニル、ステアリン酸ビニル等のビニルエステルモノマー;塩化ビニル、フッ化ビニル等のハロゲン含有ビニルモノマー;
(メタ)アクリロニトリル等のニトリル含有ビニルモノマー;ビニルトリメトキシシラン、ビニルトリエトキシシラン、ジメトキシメチルビニルシシラン、ジメトキシエチルビニルシラン、ジエトキシメチルビニルシラン、ジエトキシエチルビニルシラン、メチルビニルジメトキシシラン、エチルビニルジメトキシシラン、メチルビニルジエトキシシラン、エチルビニルジエトキシシラン等のシランアルコシキシド含有ビニルモノマーが挙げられる。
 芳香族ビニル基を有する単官能モノマーとしては、スチレン、α-メチルスチレン、クロルスチレン、ビニルナフタレン、4-ビニルビフェニル、p-スチリルトリメトキシシラン、等の芳香族ビニルモノマーが挙げられる。
 上記エチレン性不飽和基を有する多官能モノマーの具体例としては、特に限定されず、
 例えば、(メタ)アクリロイル基を有する多官能モノマーとしては、トラメチロールメタンテトラ(メタ)アクリレート、テトラメチロールメタントリ(メタ)アクリレート、テトラメチロールメタンジ(メタ)アクリレート、トリメチロールプロパントリ(メタ)アクリレート、ジペンタエリスリトールヘキサ(メタ)アクリレート、ジペンタエリスリトールペンタ(メタ)アクリレート、グリセロールトリ(メタ)アクリレート、グリセロールジ(メタ)アクリレート、(ポリ)エチレングリコールジ(メタ)アクリレート、(ポリ)プロピレングリコールジ(メタ)アクリレート、(ポリ)テトラメチレングリコールジ(メタ)アクリレート、1,4-ブタンジオールジ(メタ)アクリレート、9,9-ビス[4-(2-アクリロイロキシエチロキシ)]フェニルフルオレン等の多官能(メタ)アクリレート系モノマー等が挙げられる。
 ビニル基を有する多官能モノマーとしては、1,4-ジビニロキシブタン、ジビニルスルホン、9,9´-ビス(4-アリルオキシフェニル)フルオレン、エチルメチルジビニルシラン等が挙げられる。
 芳香族ビニル基を有する多官能モノマーとしては、ジビニルベンゼン、ジビニルナフタレン、ジビニルビフェニル等の芳香族ビニルモノマーが挙げられる。
 アリル基を有する多官能モノマーとしては、トリアリル(イソ)シアヌレート、トリアリルトリメリテート、ジアリルフタレート、ジアリルアクリルアミド、ジアリルエーテル、9,9‘-ビス(4-アリルオキシフェニル)フルオレン等のアリルモノマーが挙げられる。
 上記粒径調整組成物としては、疎水性ポリマーを含んでいることが好ましい。尚、本明細書において、「疎水性ポリマー」とは、25℃での水への溶解度が5w%以下のポリマーをいい、親水性官能基を有する、又は、親水性のブロックセグメントを有するブロックポリマーも含まれる。また、所定の確率で、水中に分散され得るものも含まれる。
 磁性体としては、金属イオンと配位子からなる金属錯体であることが好ましい。金属錯体を構成する金属や配位子としては以下に挙げるものを用いることが好ましい。
 金属錯体を構成する金属は、後述の配位子が結合した、金属錯体として磁性特性および蛍光特性を有するものであれば特に限定されないが、金属錯体粒子として高い集磁性を実現するため、金属イオンの磁気モーメントが7.5以上の金属であることが好ましい。更
に、9.0以上であることが好ましい。磁気モーメントが7.5以上の金属としては、テルビウム(Tb)、ジスプロシウム(Dy)、ホルミウム(Ho)、エルビウム(Er)、ツリウム(Tm)等が例示される。中でも、Tb、Dy、Ho、Er等が磁気モーメントが9.0以上であるため、特に好ましい。これらは単独で用いても良く、また2種以上を併用しても良い。
 金属錯体を形成する配位子は、上記金属と錯体を形成し、磁性と蛍光を発現するものであれば特に限定されないが、前記粒径調整組成物、特に、前記粒径調整組成物中の疎水性ポリマーとの疎水性相互作用によって、金属錯体粒子を形成させやすいことから、式(X)で示されるβ-ジケトン系配位子及び複素環系配位子、リン系疎水性中性配位子が好ましい。これらは、単独で用いても良く、また2種以上を併用しても良い。より強い磁性と蛍光を得るためにはβ-ジケトン系配位子及び複素環系配位子を併用または、複素環系配位子を用いる事が特に好ましい。
 β-ジケトン配位子としては、次式(X)で表されるものを用いることができる。

(上記式(X)中、R1及びRは、同一でも異なっていても良く、チエニル基、トリフルオロメチル基、フルオロメチル基、フリル基、フェニル基、ナフチル基、ベンジル基、ter‐ブチル基、メチル基等を表す。)
 このようなβ-ジケトン配位子として、例えばテノイルトリフルオロアセトン、ナフトイルトリフルオロアセトン、ベンゾイルトリフルオロアセトン、フロイルトリフルオロアセトン、ピバロイルトリフルオロアセトン、ヘキサフルオロアセチルアセトン、トリフルオロアセチルアセトン、フルオロアセチルアセトン、ジベンゾイルメタン、ジテノイルメタン、およびフロイルトリフルオロアセトンを挙げることができる。なかでも、テノイルトリフルオロアセトン(TTA)、ジベンゾイルメタン(DBM)が好ましい。
 複素環(ヘテロ環を持つ陰イオン)系配位子
 窒素や酸素などのヘテロ原子を含むヘテロ環を持つ複素環系配位子として、例えば、2,2´‐ビピリジル、4,4´-ジメチル-2,2´-ビピリジル、6,6´-ジメチル-2,2´-ビピリジル、5,5´-ジメチル-2,2´-ビピリジル、2,2´-ビピリミジル、4,4´-ジエチル-2,2´-ビピリジル、8-キノリノール、2,2´:6,2´´-テルピリジン、4,4´-ジノニル-2,2´-ビピリジル、ジ-tert-ブチルビピリジン、1,10-フェナントロリン、3,4,7,8-テトラメチル-1,10-フェナントロリン、バトフェナントロリン、5,6-ジメチル-1,10-フェナントロリン、フタロシアニン、5,15-ジフェニルポルフィリン、テトラフェニルポルフィリン、5,10,15,20-テトラ(4-ピリジル)ポルフィリン、3-ピラゾロン等が挙げられる。なかでも、2,2´‐ビピリジル、2,2´:6,2´´-テルピリジン、1,10-フェナントロリンが好ましい。
 リン系疎水性中性配位子
 リンを有する疎水性中性配位子として、トリオクチルホスフィンオキシド、トリ-n-オクチルホスフィン、リン酸トリブチル等が挙げられる。なかでも、トリオクチルホスフィンオキシド、トリ-n-オクチルホスフィンが特に好ましい。
 粒子中の金属錯体の含有率A(w%)は、不活性ガス中で1000℃まで加熱した際の、重合体及び配位子を分解した残渣から式(1)~式(3)によって計算される。
 粒子X(g)を計量し、空気中1000℃で焼成後、分子量377.86(g/mol)の酸化ホルミウムがY(g)残存したとすると、粒子中に含まれる酸化ホルミウムの物重量Z(mol)は、式(1)により計算される。

使用した錯体の分子量をM(g/mol)とすると、粒子中に含まれる金属錯体量a(g)は、式(2)により計算される。

よって粒子中に含まれる金属錯体の含有率A(w%)は、式(3)により算出される。

 また前記残渣はTG-DTA6300(日立ハイテック社製)装置を用いて、粒子を35℃から1000℃までを5℃/minで昇温した後、5分間1000℃で維持させ、その際に残った量より求める事が出来る。
 金属錯体粒子の好ましい1態様としては、粒径調整組成物としてスチレン系又は(メタ)アクリレート系の疎水性ポリマー、金属錯体としてホルミウムイオン、テルビウムイオン又はジスプロシウムイオンと、β-ジケトン系配位子及び複素環系配位子の少なくともいずれか一方、又は両方の配位子との金属錯体からなる金属錯体粒子が挙げられる。金属錯体の含有量が、粒子の合計重量に対して40w%以上98w%以下であることが好ましい。
 金属錯体の含有量は、その種類にもよるが、40w%以上98w%以下であることが好ましい。40w%以上の場合、充分な集磁性が発現する。98w%以下であると、粒子としての形状を維持できる。より好ましくは、50w%以上、更に好ましくは60w%以上である。
 本発明の金属錯体粒子の粒径は特に限定されないが、高い集磁性を実現するため、粒径は500nm以上10μm以下が好ましい。500nm以上であれば、磁石による分離速度が速く、また10μm以下であれば、粒子が沈降しにくい。粒径は1μm以上5μm以下がより好ましい。
 なお、粒子の平均粒径は、粒子を走査型電子顕微鏡で観察し、観察された画像における任意に選択した50個の各粒子の最大径を算術平均することにより求めた。
 本発明の金属錯体粒子の変動係数(CV値)は、測定結果の再現性を良好にするため、20%以下が好ましい。20%を超えると、集磁性にバラツキが生じ、測定結果の再現性が低下することがある。より好ましくは10%以下である。
 なお、上記変動係数は、粒子を走査型電子顕微鏡で観察し、観察された画像における任意に選択した50個の各粒子の粒径の標準偏差を求め、下記式により粒子の粒径のCV値を求めた。

 本発明の金属錯体粒子の製造方法としては、以下の製法が例示される。
(1)A:粒径調整組成物と金属錯体とを双方溶解可能な疎水性溶剤に溶解させる工程、B:工程Aで得られた溶液を、界面活性剤及び/または分散安定剤が溶解した水溶液中に
添加して、乳化、転層乳化、懸濁等により、液滴を形成する工程、C:疎水性溶剤を加熱や減圧下にて留去する工程、D:フィルトレーション、分級等で粒径を揃える工程、による方法。
(2)A:重合性モノマーと疎水性重合開始剤と金属錯体を溶解させる工程、B:界面活性剤及び/又は分散安定剤が溶解した水溶液中にて、乳化、懸濁等後により液滴を形成する工程、D:加熱及び/又は光照射により、金属錯体存在下で粒径調整組成物を重合する工程、E:フィルトレーション、分級等で粒径を揃える工程、による方法。尚、工程Aにおいて、必要に応じて重合性モノマーと金属錯体の双方が溶解可能な疎水性溶剤を添加しても良い。その場合、工程Dの後、疎水性溶剤を加熱や減圧下にて留去する工程が追加される。また、重合性モノマーとしては、前述の単官能モノマーであっても、多官能モノマーであってもよく、両方を併用しても良い。
(3)A:界面活性剤及び/又は分散安定剤が溶解した水に、テンプレートとなる粒径調整組成物からなる粒子(以下、「テンプレート粒子」ともいう)を分散させる工程、B:上記テンプレート粒子と金属錯体の双方を溶解可能な疎水性有機溶剤及び/または重合性モノマーに金属錯体を溶解させた金属錯体溶液を用意する工程、C:工程Aで得られた分散液に、工程Bで得られた金属錯体溶液を添加し、前記テンプレート粒子内に、疎水性有機溶剤及び/または重合性モノマーと伴に金属錯体を吸収させる工程、D:重合性モノマーを加熱及び/又は光照射により重合し、疎水性有機溶剤を加熱や減圧下で留去する工程、による方法。尚、工程Bにおいて、疎水性重合開始剤を添加しても良い。また、工程Bにおいて、重合性モノマーが添加されていない場合は、工程Dにおける重合の工程は不要となる。
等が挙げられる。
 テンプレートとなる粒径調整組成物からなる粒子(テンプレート粒子)に、粒径の揃った粒子を用いることで、得られる金属錯体粒子の粒径を揃えられることと、内包させる金属錯体の量を均一にできることから、(3)の方法が好適に用いられる。
 上記粒径調整組成物からなるテンプレート粒子は、前述のとおり、重合性モノマーを重合して得られるポリマーを主成分とし、上記ポリマーは一種または二種以上の重合性モノマーのコポリマーも含む。
 上記テンプレート粒子を主として構成するポリマーとしては、疎水性ポリマーであれば特に限定されず、上述の単官能重合性モノマーをラジカル重合、イオン重合、配位重合等により、重合して得られるポリマーが用いられる。中でも、後述する疎水性溶媒への溶解性が高いことから、重合性モノマーが芳香族ビニルモノマー、(メタ)アクリルレートモノマー、ビニルエステルモノマーを主成分とするポリマー/コポリマーが好適に用いられる。芳香族ビニルモノマー、(メタ)アクリルレートが更に好ましい。
 上記テンプレート粒子の製法としては、乳化重合、懸濁重合、ソープフリー重合、分散重合、ミニエマルション重合、マイクロサスペンジョン重合;ポリマー溶液を用いた、転相乳化、液中乾燥法等を用いることができるが、粒径が揃ったポリマー粒子が得られるため、ソープフリー重合、分散重合が好適に用いられる。
 上記製法(1)又は(3)の場合、上述の重合性モノマーと開始剤とを添加し、加熱等により重合させても良い。中でも、多官能モノマーを用いた場合、粒子内に架橋構造を導入できるため、好ましい。尚、上記添加された重合性モノマーが加熱等により重合したポリマーは、粒径調整組成物の一部となるため、「第二粒径調整組成物」ともいう。
 上記金属錯体粒子中の粒径調整組成物の含有率は、上限値としては60w%未満が好ましく、50w%未満がより好ましい。下限値としては2w%以上が好ましく、5w%以上がより好ましい。2w%以上であると、粒子の形状を維持しやすく、また、金属錯体の流出を防止することができる。また、本発明の金属錯体粒子は十分な集磁性を発現させるため、金属錯体の含有率は30w%以上が好ましいため、粒径調整組成物の含有率は、必然
的に70w%以下となる。尚、粒径調整組成物の含有率は、前述の式(1)~(3)の金属錯体の含有率を用いて、式(4)により算出される。

 特開平10-55911に開示された従来の磁性粒子においては、図2Aに示すように、断面視において、磁性体(Fe)は粒子の表層部に存在していた。また特願2010-528640(特許第5574492号)に開示された従来の磁性粒子においては、図2Bに示すように磁性体(Fe)は粒子の核付近の樹脂(海)上に磁性粒子(島)が局所的に存在し、蛍光色素(希土類(Eu))は粒子表層部に存在していた。しかし、本願の金属錯体粒子においては図1A,図1Bに示すように、金属錯体そのものにより粒子が形成されていることより、金属錯体粒子の断面視において、粒子の表層(表面)から中心(点)までを、略均一なモルフォロジーが形成されている。つまり、上述のように、金属錯体の含有率が高いため、高い集磁性を有すると同時に、粒子1個あたりの磁性量が均一となるため、磁力による分離の際、個々の粒子の磁力への応答が均一となり、分離の際のばらつきが小さくなる。なお、粒径調整組成物が金属錯体粒子内に局所的に分布することもあり得るが、その場合は金属錯体(海)中に粒径調整組成物(島)が局所的または点在する形態となる。このような形態も本発明の一態様である。
 上述の製法(1)、(2)及び(3)で用いられる、疎水性溶剤としては、特に限定されないが、SP値が10以下で、後に疎水性溶媒を留去するために沸点が水よりも低いことが好ましい。例えば、酢酸エチル(SP値:9.1、沸点:77.1℃)、ベンゼン(SP値:9.2、沸点:80.1℃)、ジイソプロピルエーテル(SP値:6.9、沸点:69℃)、クロロホルム(SP値:9.1、沸点:61.2℃)等が挙げられる。これらは、単独で用いてもよく、2種類以上を併用してもよい。なかでも、酢酸エチルが好ましい。
 SP値とは、沖津俊直、「接着」、高分子刊行会、40巻8号(1996)p342-350に記載された、沖津による各種原子団のΔF、Δv値を用い、下記式(A)により算出した溶解性パラメーターδを意味する。また、混合物、共重合体の場合は、下記式(B)により算出した溶解性パラメーターδmixを意味する。

式中、ΔF、Δvは、それぞれ、沖津による各種原子団のΔF、モル容積Δvを表す。φは、容積分率又はモル分率を表し、φ+φ+・・・φ=1である。
 上記製法(1)、(2)及び(3)に用いられる疎水性重合開始剤としては、上記疎水性溶媒に溶解可能な疎水性重合開始剤であれば特に限定されない。具体的には、ジ-tert-ブチルペルオキシド、tert-ブチルヒドロペルオキシド、過酸化ベンゾイル、メチルエチルケトンペルオキシド、ジ(セカンダリーブチル)パーオキシジカーボネート等の過酸化物、アゾビスイソブチロニトリル、2,2´-アゾビス(2-メチルブチロニトリル)(V‐59)、2,2´-アゾビス(2,4-ジメチルバレロニトリル)(V-65)、2,2´-アゾビス(4-メトキシ-2,4-ジメチルバレロニトリル)(V-70)、ジメチル2,2´-アゾビス(イソブチレート)(V-601)等のアゾ系化合物が挙げられるが、好ましくはtert-ブチル-2-エチルペルオキシヘキサノアート、ジ(セカンダリーブチル)パーオキシジカーボネート、アゾビスイソブチロニトリルが良い。
 上記製法(1)、(2)及び(3)に用いられる、分散剤としては、ポリアルキレングリコール、ポリビニルアルコール、ポリビニルピロリドン、ポリ(メタ)アクリル酸、側鎖に水酸基、カルボキシル基、ポリアルキレングリコール基等の水溶性基を有したポリ(メタ)アクリル酸、ポリ(メタ)アクリルレート等の水溶性ポリマー、コロイダルシリカ、炭酸カルシウム等が挙げられる。これらは単独で用いても、2種以上が併用されても良
い。
 上記製法(1)、(2)及び(3)に用いられる、界面活性剤としては、水溶性であれば特に限定されず、ポリアルキレングリコールアルキルエーテル等のノニオン性界面活性剤;アルキル硫酸塩、アルキルベンゼンスルホン酸塩、アルキルカルボン酸塩、アルキルリン酸エステル塩、アルキル硫酸エステル塩等のアニオン性界面活性剤;アルキルアミン塩、4級アルキルアンモニウム塩等のカチオン性界面活性剤;ベタイン型、スルホベタイン型、アルキルベタイン型等の両性界面活性剤が挙げられる。これらは単独で用いても、2種以上が併用されても良い。
 上記製法(1)、(2)及び(3)においては、必要に応じて、その他添加剤を用いても良い。
  その他添加剤としては、電解質として塩化カリウム、塩化ナトリウム、酢酸ナトリウム、酢酸アンモニウム、クエン酸ナトリウム、硫酸カリウム、硫酸ナトリウム、リン酸二水素ナトリウム、リン酸水素二ナトリウム等が挙げられる。これらは単独で用いても、2種以上が併用されても良い。
 金属錯体粒子の製造方法の1態様として、
 界面活性剤または水溶性ポリマーが溶解した水系媒体(a)を用意する工程と、
 上記水系媒体(a)に粒径調整組成物からなる粒子(テンプレート粒子)を分散させた、水系媒体分散液(A)を用意する工程と、
 粒径調整組成物が溶解可能な疎水性溶剤(b)を用意する工程と、
 上記疎水性溶剤(b)に、磁性及び蛍光の両方を有する金属錯体を溶解させた、金属錯体溶解液(B)を用意する工程と、
 前記、(A)と(B)を混合し、粒径調整組成物と金属錯体と疎水性溶剤(b)からなる膨潤液滴(c)を用意する工程と、
 上記膨潤液滴(c)から疎水性溶剤(b)を除去する工程と、を含む金属錯体粒子の製造方法が挙げられる。
 金属錯体が、ホルミウムイオン、テルビウムイオン又はジスプロシウムイオンと、β-ジケトン系配位子及び複素環系配位子の両方又はいずれか一方とからなることが好ましい。
 疎水性ポリマーは、スチレン系又は(メタ)アクリレート系の疎水性ポリマーからなることが好ましい。
 本発明の蛍光と磁性を併せ持つ金属錯体粒子を担体として、測定対象物質、測定対象物質の類似物質、または測定対象物質と特異的に結合する物質(以下、これらを総称して担持用物質と略記することがある)を固定化することで感作金属錯体粒子を作製することができる。この感作金属錯体粒子もまた、本発明の一つである。
 本発明における測定対象物質としては、通常この分野で測定されるものであれば特に限定はされず、例えば血清、血液、血漿、尿等の生体体液、リンパ液、血球、各種細胞類等の生体由来の試料中に含まれるタンパク質、脂質タンパク質、核酸、免疫グロブリン、血液凝固関連因子、抗体、酵素、ホルモン、癌マーカー、心疾患マーカー及び各種薬物等が代表的なものとして挙げられる。更に具体的には、例えばアルブミン、ヘモグロビン、ミオグロビン、トランスフェリン、C反応性蛋白質(CRP)等のタンパク質;例えば高比重リポ蛋白質(HDL)、低比重リポ蛋白質(LDL)、超低比重リポ蛋白質等のリポ蛋白;例えばデオキシリボ核酸(DNA)、リボ核酸(RNA)等の核酸;例えばアルカリ性ホスファターゼ、アミラーゼ、酸性ホスファターゼ、γ-グルタミルトランスフェラーゼ(γ-GTP)、リパーゼ、クレアチンキナーゼ(CK)、乳酸脱水素酵素(LDH)、グルタミン酸オキザロ酢酸トランスアミナーゼ(GOT)、グルタミン酸ピルビン酸トランスアミナーゼ(GPT)、レニン、プロテインキナーゼ(PK)、チロシンキナーゼ等の酵素;例えばIgG、IgM、IgA、IgD、IgE等の免疫グロブリン(或いはこれらの、例えばFc部、Fab部、F(ab)部等の断片);例えばフィブリノーゲン、フィブリン分解産物(FDP)、プロトロンビン、トロンビン等の血液凝固関連因子;例えば抗ストレプトリジンO抗体、抗ヒトB型肝炎ウイルス表面抗原抗体(HBs抗原)、抗ヒトC型肝炎ウイルス抗体、抗リウマチ因子等の抗体;例えば甲状腺刺激ホルモン(TSH)、甲状腺ホルモン(FT3、FT4、T3、T4)、副甲状腺ホルモン(PTH)、ヒト絨毛性ゴナドトロピン(hCG)エストラジオール(E2)等のホルモン;例えばα-フェトプロテイン(AFP)、癌胎児性抗原(CEA)、CA19-9、前立腺特異抗原(PSA)等の癌マーカー;例えばトロポニンT(TnT)、ヒト脳性ナトリウム利尿ペプチド前駆体N端フラグメント(NT-proBNP)等の心疾患マーカー;例えば抗てんかん薬、抗生物質、テオフィリン等の薬物等が挙げられる。
 本発明における測定対象物質の類似物質(アナログ)は、測定対象物質と特異的に結合する物質(測定対象物質結合物質)が有する測定対象物質との結合部位と結合し得るもの、言い換えれば、測定対象物質が有する測定対象物質結合物質との結合部位を有するもの、更に言い換えれば、測定対象物質と測定対象物質結合物質との反応時に共存させると該反応と競合し得るものであれば何れでもよい。
 本発明における測定対象物質と特異的に結合する物質(測定対象物質結合物質)としては、例えば「抗原」-「抗体」間反応、「糖鎖」-「タンパク質」間反応、「糖鎖」-「レクチン」間反応、「酵素」-「インヒビター」間反応、「タンパク質」-「ペプチド鎖」間反応、「染色体又はヌクレオチド鎖」-「ヌクレオチド鎖」間反応、又は、「ヌクレオチド鎖」-「タンパク質」間反応等の相互反応によって測定対象物質又はその類似物質と結合するもの等が挙げられ、上記各組合せに於いて何れか一方が測定対象物質又はその類似物質である場合、他の一方がこの測定対象物質結合物質である。例えば、測定対象物質又はその類似物質が「抗原」であるとき測定対象物質結合物質は「抗体」であり、測定対象物質又はその類似物質が「抗体」であるとき測定対象物質結合物質は「抗原」である(以下、その他の上記各組合せにおいても同様である)。

 なお、本発明に用いる抗体としては、免疫グロブリン分子自体の他、パパインやペプシン等の蛋白質分解酵素、或いは化学的分解により生じるFab、F(ab’)フラグメント等の分解産物も包含される。また、上記抗体は、ポリクローナル抗体又はモノクローナル抗体のどちらを用いてもかまわない。上記抗体の取得方法も通常使用される方法を用いることができる。
 上記の如き測定対象物質結合物質としては、「抗原」-「抗体」間反応或いは「糖鎖-タンパク質」間反応によって測定対象物質又はその類似物質と結合するものが好ましい。具体的には、測定対象物質又はその類似物質に対する抗体、測定対象物質又はその類似物質が結合する抗原、或いは、測定対象物質又はその類似物質に結合するタンパク質が好ましく、測定対象物質又はその類似物質に対する抗体、或いは測定対象物質又はその類似物質に結合する抗原が更に好ましい。なお、本明細書において「抗原抗体反応」、「抗原」、「抗体」の語を用いる場合、通常の意味に加え、特異的な結合反応により感作金属錯体粒子を凝集させることができる上記の概念・形態のいずれをも含む場合があり、限定的に解釈してはならない。
 本発明の金属錯体粒子に担持用物質を固定化し感作金属錯体粒子を製造する方法は特に限定されない。従来公知の物理的および/または化学的結合によって固定化する方法を用いることができる。化学的結合の例としては、アルデヒド、アルブミン、カルボジイミド、ストレプトアビジン、ビオチン、エポキシドからなる群から選ばれる少なくとも一種の有機化合物、および/または、アミノ基、カルボキシ基、水酸基、メルカプト基、グリシジルオキシ基、トシル基等から選ばれる少なくとも一種の官能基を金属錯体粒子表面にもたせ、それらを介して担持用物質を金属錯体粒子に固定化させる等が挙げられる。このうち、金属錯体粒子自体の安定性および担持用物質固定化効率の観点から、カルボキシ基、トシル基を含む有機化合物が特に好ましい。
 本発明の感作金属錯体粒子における担持用物質の結合量は、用いる担持用物質の種類によって異なり、実験的に最適な量を適宜設定することができる。なお、本明細書において、「担持」「感作」「固定化」は通常の意味を有し、同義に使用している。
 このような方法により得られた本発明の感作金属錯体粒子は、必要に応じて各種高分子化合物やタンパク質(例えば、ウシ血清アルブミン等)で被覆(ブロッキング)処理を施し、適当な緩衝液に分散して感作金属錯体粒子分散液として用いる。感作金属錯体粒子分散液は、免疫測定用試薬として用いることができ、該測定試薬も本発明の一つである。本発明の免疫測定用試薬は、測定に用いる希釈液(緩衝液)や標準物質等を組み合わせて、測定試薬キットとして用いることができ、該キットも本発明の一つである。
 上記希釈液は、測定試料等の希釈に用いられる。上記希釈液としては、pH5.0~9.0の緩衝液であればどのようなものでも用いることができる。具体的には、例えば、リン酸緩衝液、グリシン緩衝液、トリス緩衝液、ホウ酸緩衝液、クエン酸緩衝液、グッド緩衝液等が挙げられる。
 本発明の免疫測定用試薬や希釈液は、測定感度の向上や、測定対象物質と担持用物質間の特異的反応の促進を目的に、種々の増感剤を含有してもよい。上記増感剤としては、例えば、メチルセルロース、エチルセルロース等のアルキル化多糖類や、プルラン、ポリビニルピロリドン等が挙げられる。
 本発明の免疫測定用試薬や希釈液は、測定試料中に存在する測定対象物質以外の物質により起こる非特異反応の抑制や、測定試薬の安定性向上等を目的に、各種高分子化合物やタンパク質、およびその分解物、アミノ酸、界面活性剤等を含有してもよい。こういった添加物の例としては、アルブミン(牛血清アルブミン、卵性アルブミン)、カゼイン、ゼラチン等のタンパク質が挙げられる。
 本発明の測定対象物質測定方法は、上記本発明における金属錯体粒子を用いて行われる以外は特に限定されず、例えば、この分野で通常行われる、文献(例えば、「酵素免疫測定法第2版」石川栄治ら編集、医学書院、1982年)記載のサンドイッチ法、競合法等に準じて行えばよい。
 本発明の測定対象物質測定法において、試料、金属錯体粒子、標識された測定対象物質結合物質、標識測定対象物質又はその類似物質等を接触させる方法としては、通常なされる撹拌、混合等の処理により、金属錯体粒子が分散されればよい。反応時間は、測定対象物質、用いられる測定対象物質結合物質、サンドイッチ法、競合法等の違いに応じて適宜設定されればよい。
 本発明の測定対象物質測定方法におけるB/F分離(結合標識抗体と未結合標識抗体の分離)は、例えば、金属錯体粒子の磁性を利用し、反応槽の外側等から磁石等により金属錯体粒子を集めて、反応液を排出し、洗浄液を加えた後、磁石を取り除き、金属錯体粒子を混合して分散させ、洗浄することによりなされる。上記操作を1~3回繰り返してもよい。洗浄液としては、通常この分野で用いられるものであれば特に限定はされない。
 測定対象物質結合物質、測定対象物質又はその類似物質等を標識するために用いられる標識物質としては、例えば酵素免疫測定法(EIA)に於いて用いられるアルカリホスファターゼ、β-ガラクトシダーゼ、ペルオキシダーゼ、マイクロペルオキシダーゼ、グルコースオキシダーゼ、グルコース-6-リン酸脱水素酵素、リンゴ酸脱水素酵素、ルシフェラーゼ、チロシナーゼ、酸性ホスファターゼ等の酵素類、例えば放射免疫測定法(RIA)に於いて用いられる99mTc、131I、125I、14C、3H、32P等の放射性同位元素、例えば蛍光免疫測定法(FIA)に於いて用いられるフルオレセイン、ダンシル、フルオレスカミン、クマリン、ナフチルアミン或いはこれらの誘導体、グリーン蛍光タンパク質(GFP)等の蛍光性物質、例えばルシフェリン、イソルミノール、ルミノール、ビス(2,4,6-トリフロロフェニル)オキザレート等の発光性物質、例えばフェノール、ナフトール、アントラセン或いはこれらの誘導体等の紫外部に吸収を有する物質、例えば4-アミノ-2,2,6,6-テトラメチルピペリジン-1-オキシル、3-アミノ-2,2,5,5-テトラメチルピロリジン-1-オキシル、2,6-ジ-t-ブチル-α-(3,5-ジ-t-ブチル-4-オキソ-2,5-シクロヘキサジエン-1-イリデン)-p-トリオキシル等のオキシル基を有する化合物に代表されるスピンラベル化剤としての性質を有する物質等が挙げられる。これらの内、感度等の観点から、酵素、蛍光性物質が好ましく、更に好ましいのはアルカリホスファターゼ、ペルオキシダーゼ及びグルコースオキシダーゼであり、特に好ましいのはペルオキシダーゼである。
 上記した如き標識物質を測定対象物質結合物質、測定対象物質又はその類似物質等に結合させるには、通常この分野で用いられる方法、例えば自体公知のEIA、RIA或いはFIA等に於いて一般に行われている自体公知の標識方法[例えば、医化学実験講座、第8巻、山村雄一監修、第1版、中山書店、1971;図説 蛍光抗体、川生明著、第1版、(株)ソフトサイエンス社、1983;酵素免疫測定法、石川栄治、河合忠、室井潔編、第2版、医学書院、1982等に記載のグルタルアルデヒド法、過ヨウ素酸法、マレイミド法又はピリジルジスルフィド法等]等を利用すればよい。
 標識物質の使用量は、用いる標識物質の種類により適宜設定すればよい。例えばトリス緩衝液、リン酸緩衝液、ベロナール緩衝液、ホウ酸緩衝液、グッド緩衝液等の通常この分野で用いられている緩衝液中に含有させて用いればよい。尚、当該緩衝液としては、通常この分野で用いられている、例えばトリス緩衝液、リン酸緩衝液、ベロナール緩衝液、ホウ酸緩衝液、グッド緩衝液等が挙げられ、そのpHは、抗原抗体反応を抑制しない範囲であればよく、通常5~9である。また、このような緩衝液中には、目的の抗原抗体反応を阻害しないものであれば、例えばアルブミン、グロブリン、水溶性ゼラチン、ポリエチレングリコール等の安定化剤、界面活性剤、糖類等を含有させておいてもよい。
 標識物質又はその活性の測定方法としては、通常行われる方法を特に制限なく用いることができる。測定に際し用いる光学機器も特に限定されず、代表的には、臨床検査で広く使用されている生化学自動分析機であればいずれも使用することができる。
 以下に実施例を挙げて本発明を説明するが、本発明は以下の実施例に限定して解釈されることはない。
[調製例1]
金属錯体(a)の合成
 塩化ホルミウム6水和物(HoCl・6HO)のエタノール溶液に、塩化ホルミウム6水和物に対し、1倍等量の2,2’‐ビピリジル(bpy)、3倍等量のジベンゾイルメタン(DBM)を添加し、混和後、更にジエタノールアミンを加えることで、pH6-7に調整した。室温で24時間撹拌後、淡黄色の沈殿物が得られた。得られた沈殿物をろ過後、エタノールで洗浄することで、金属イオンがホルミウム、配位子がbpy及びDBMである、金属錯体(a)を得た。
[調製例2]
金属錯体(b)の合成
 配位子として、DBMの代わりにテノイルトリフルオロアセトン(TTA)を用いた以外は調製例1と同様の操作にて、金属イオンがホルミウム、配位子がbpy及びTTAである、金属錯体(b)を得た。
[調製例3]
金属錯体(c)の合成
 塩化ホルミウム6水和物の代わりに塩化ジスプロシウム(DyCl)を用いた以外は調製例1と同様の操作にて、金属イオンがジスプロシウム、配位子がbpy及びDBMである、金属錯体(c)を得た。
[調製例4]
金属錯体(d)の合成
塩化ホルミウム6水和物の代わりに塩化テルビウム6水和物(TbCl・6HO)を用いた以外は調製例1と同様の操作にて、金属イオンがテルビウム、配位子がbpy及びDBMである、金属錯体(d)を得た。
[調製例5]
金属錯体(e)の合成
 配位子として、bpyの代わりに2,2´:6,2´´-テルピリジン(terpy)を用いた以外は調製例1と同様の操作にて、金属イオンがホルミウム、配位子がterpy及びDBMである、金属錯体(e)を得た。
[調製例6]
金属錯体(f)の合成
 配位子として、bpyの代わりに1,10-フェナントロリン一水和物(phen)を用いた以外は調製例1と同様の操作にて、金属イオンがホルミウム、配位子がphen及びDBMである、金属錯体(f)を得た。
[調製例7]
金属錯体(g)の合成
 塩化ホルミウム6水和物の代わりに塩化ユーロピウム6水和物(EuCl・6HO)を用い、配位子にテノイルトリフルオロアセトン(TTA)を用いた以外は調製例1と同様の操作にて、金属イオンがユーロピウム、配位子がTTAである、金属錯体(g)を得た。
[調製例8]
テンプレート粒子(I)の調整
 反応容器にイオン交換水500g、粒径調整組成物を構成するモノマーとしてメチルメタクリレート(MMA)(和光純薬工業社製)50g、過硫酸カリウム(和光純薬工業社製)0.5gを添加し、窒素雰囲気下、室温で2時間撹拌した。その後70℃に加熱し、24時間重合を行うことで、ポリメチルメタクリレートからなるテンプレート粒子(I)を得た。
 得られたテンプレート粒子(I)の平均粒子径は1.06μmであり、CV値は3.3%であった。
[調製例9]
テンプレート粒子(II)の調整
 粒径調整組成物を構成するモノマーをMMAからスチレン(St)(NSスチレンモノマー社製)に変更した以外は、同様の操作にて、ポリスチレンからなるテンプレート粒子(II)を得た。
 得られたテンプレート粒子(II)の平均粒子径は1.03μmであり、CV値は3.8%であった。
[実施例1]
金属錯体粒子(1)の調製
 イオン交換水100g中に上記テンプレート粒子(I)の含有量が0.1gとなるよう、また、ラウリル硫酸トリエタノールアミンの含有量が0.1gとなるよう、上記テンプレート粒子(I)とラウリル硫酸トリエタノールアミン(エマールTD、花王社製)をイオン交換水へ添加し、テンプレート粒子(I)分散液を得た。次に、金属錯体(a)2.70gを酢酸エチル4.00gに溶解させ調製した金属錯体(a)溶液を、上記テンプレート粒子(I)分散液に添加した。室温で24時間撹拌することで、上記テンプレート粒子(I)と金属錯体(a)との複合液滴を形成した。その後80℃で10時間加熱撹拌を行い、酢酸エチルを除去することで、本発明の金属錯体粒子(1)を得た。
 得られた金属錯体粒子(1)の平均粒子径は3.21μm、CV値は3.8%であり、金属錯体含有率は96.4w%であった。
[実施例2]
 イオン交換水100g中に上記テンプレート粒子(I)の含有量が0.1gとなるよう、また、ラウリル硫酸トリエタノールアミンの含有量が0.1gとなるよう、上記テンプレート粒子(I)とラウリル硫酸トリエタノールアミン(エマールTD、花王社製)をイオン交換水へ添加し、テンプレート粒子(I)分散液を得た。次に、金属錯体(a)2.50g、第二粒径調整組成物用モノマーとしてメチルメタクリレート(和光純薬社製)0.20g、ジ(セカンダリーブチル)パーオキシジカーボネート(ルペロックス225、アルケマ吉富社製)0.02gを酢酸エチル4.00gに溶解させ調製した金属錯体(a)溶液を、上記テンプレート粒子(I)分散液に添加した。室温で24時間撹拌することで、上記テンプレート粒子(I)、金属錯体(a)との複合液滴を形成した。その後80℃で10時間加熱撹拌を行い、メチルメタクリレートを重合すると同時に酢酸エチルを除去することで、第二粒径調整組成物としてのポリメチルメタクリレート(pMMA)を含有する金属錯体粒子(2)を得た。
 得られた金属錯体粒子(2)の平均粒子径は3.20μm、CV値は4.2%であり、金属錯体含有率は89.1w%であった。
[実施例3]
 第二粒径調整組成物用モノマーとして、ジビニルベンゼン(DVB)(DVB960、NSスチレンモノマー社製)を用いたこと以外は実施例2と同様に操作し、第二粒径調整組成物としてのポリジビニルベンゼン(pDVB)を含有する金属錯体粒子(3)を得た。
 得られた金属錯体粒子(3)の平均粒子径は3.18μm、CV値は4.1%であり、金属錯体含有率は88.1w%であった。
[実施例4]
 テンプレート粒子(II)を用い、第二粒径調整組成物用モノマーとして、スチレン(St)(NSスチレンモノマー社製)を用いた以外は、実施例2と同様に操作し、第二粒径調整組成物としてのポリスチレン(pSt)を含有する金属錯体粒子(4)を得た。
 得られた金属錯体粒子(4)の平均粒子径は3.10μm、CV値は4.3%であり、金属錯体含有率は89.0w%であった。
[実施例5]
 金属錯体として、金属錯体(b)を用いた以外は、実施例3と同様に操作し、金属錯体粒子(5)を得た。
 得られた金属錯体粒子(5)の平均粒子径は3.12μm、CV値は4.2%であり、金属錯体含有率は88.2w%であった。
[実施例6]
 金属錯体(a)を2.00g、ジビニルベンゼンを0.70g、ジ(セカンダリーブチル)パーオキシジカーボネートを0.08g用いたこと以外は実施例3と同様に操作し、金属錯体粒子(6)を得た。
 得られた金属錯体粒子(6)の平均粒子径は3.20μm、CV値は3.8%であり、金属錯体含有率は71.4w%であった。
[実施例7]
 金属錯体(a)を1.60g、ジビニルベンゼンを1.10g、ジ(セカンダリーブチル)パーオキシジカーボネートを0.11g用いたこと以外は実施例3と同様に操作し、金属錯体粒子(7)を得た。
 得られた金属錯体粒子(7)の平均粒子径は3.18μm、CV値は3.9%であり、金属錯体含有率は55.6w%であった。
[実施例8]
 金属錯体(a)を1.20g、ジビニルベンゼンを1.50g、ジ(セカンダリーブチル)パーオキシジカーボネートを0.15g用いたこと以外は実施例3と同様に操作し、金属錯体粒子(8)を得た。
 得られた金属錯体粒子(8)の平均粒子径は3.17μm、CV値は4.1%であり、金属錯体含有率は40.2w%であった。
[実施例9]
 金属錯体(a)を2.50g、スチレンを0.20g、クロロホルムを13g、ジ(セカンダリーブチル)パーオキシジカーボネートを0.002g、ポリビニルアルコール(ゴーセノールGH20、日本合成社製)5重量%水溶液を13g、ラウリル硫酸トリエタノールアミンを1.5g、イオン交換水を150g混合し、ホモジナーザーを用いて、2500rpmで撹拌後した。その後、反応槽へ移し、窒素気流下、200rpmで2時間撹拌後、80℃で10時間加熱撹拌を行い、スチレンを重合すると同時にクロロホルムを除去した。反応終了後、遠心分離操作を繰り返すことで、粒径調整組成物としてポリスチレン(pSt)を含有する金属錯体粒子(9)を得た。
 得られた金属錯体粒子(9)の平均粒子径は3.31μm、CV値は12.8%であり、金属錯体含有率は90.2w%であった。
[実施例10]
 実施例9における反応終了後の遠心分離操作の遠心条件を変更して遠心分離操作を繰り返した以外は、実施例9と同様の操作を行い、本発明の金属錯体粒子(10)を得た。
 得られた金属錯体粒子(10)の平均粒子径は3.42μm、CV値は17.8%であり、金属錯体含有率は91.3w%であった。
[実施例11]
 金属錯体として、金属錯体(c)を用いた以外は、実施例3と同様に操作し、金属錯体粒子(11)を得た。
 得られた金属錯体粒子(11)の平均粒子径は3.22μm、CV値は4.8%であり、金属錯体含有率は89.3w%であった。
[実施例12]
 金属錯体として、金属錯体(d)を用いた以外は、実施例3と同様に操作し、金属錯体粒子(12)を得た。
 得られた金属錯体粒子(12)の平均粒子径は3.16μm、CV値は4.4%であり、金属錯体含有率は88.7w%であった。
[実施例13]
 金属錯体として、金属錯体(e)を用いた以外は、実施例3と同様に操作し、金属錯体粒子(13)を得た。
得られた金属錯体粒子(13)の平均粒子径は3.22μm、CV値は5.2%であり、金属錯体含有率は89.2w%であった。
[実施例14]
 金属錯体として、金属錯体(f)を用いた以外は、実施例3と同様に操作し、金属錯体粒子(14)を得た。
 得られた金属錯体粒子(14)の平均粒子径は3.06μm、CV値は4.9%であり、金属錯体含有率は87.5w%であった。
[比較例1]
 金属錯体(a)の仕込量を0.50g、第二粒径調整組成物としてのジビニルベンゼンの仕込量を2.20gとしたこと以外は実施例3と同様に操作し、金属錯体粒子(15)を得た。
 得られた金属錯体粒子(15)の平均粒子径は3.18μm、CV値は5.1%であり、金属錯体含有率は14.8w%であった。
[比較例2]
実施例9における反応終了後の遠心分離操作の遠心条件を変更して遠心分離操作を繰り返した以外は、実施例9と同様の操作を行い、本発明の金属錯体粒子(16)を得た。
得られた金属錯体粒子(16)の平均粒子径は3.33μm、CV値は25.6%であり、金属錯体含有率は90.4w%であった。
[比較例3]
 金属錯体として、金属錯体(g)を用いた以外は、実施例3と同様に操作し、金属錯体粒子(17)を得た。
得られた金属錯体粒子(17)の平均粒子径は3.15μm、CV値は4.8%であり、金属錯体含有率は88.6w%であった。
[評価方法]
 *集磁性評価
 分光光度計(U‐3900H 日立製作所社製)に予めスペーサー(W10mm×D10mm×H10mm)を介し、磁石(3680G、W10mm×D10mm×H5mm)をあてた状態で分光光度計に設置した石英セルに試料(1.2mL)を投入し、5秒後から125秒後の吸光度を測定し、式(6)を用いて集磁率を算出した。
 集磁率(%)=[{(試料投入5秒後の吸光度)-(試料投入125秒後の吸光度)}/(試料投入5秒後の吸光度)]×100  式(6)
[判定基準]
  〇〇〇:集磁率が61%以上
  〇〇:集磁率が41%以上60%未満
  〇:集磁率が21%以上40%未満
  ×:集磁率が20%未満
 得られた結果をまとめて表1、表2、表3に示す。
[参考例1]
 特許文献3(特開2008-127454号公報)の製法では、図2Bに示すように磁性体(Fe)は粒子の核付近に局所的に、蛍光色素(希土類(Eu))は粒子表層
部に存在する。そのため、また粒子の色は、褐色~黒色をとる。また、粒径は約0.2μm、フェライト粒子含有量は約5w%である。120s集磁性は8%であり、相対蛍光強度は36であった。
[参考例2]
 特開平10-55911記載の製法では、図2Aに示すように磁性体(Fe)は粒子の表層部に存在する。また、蛍光色素を含まないため、蛍光を有さない。フェライト含有率は18.3w%であり、120s集磁性は83%であった。
[参考例3]
 参考特許(特表昭59-500691)記載の製法では、磁性体(Fe)は粒子の内部に存在する。また、蛍光色素を含まないため、蛍光を有さない。フェライト含有率は31.2w%であり、120s集磁性は95%であった。

Claims (12)

  1.  磁性体と、疎水性ポリマーを含む粒径調整組成物と、を備える金属錯体粒子であって、
     前記磁性体は、金属イオンと配位子からなる、磁性及び蛍光の両方を有する金属錯体を含み、
     前記金属錯体粒子中の前記粒径調整組成物の含有率が60w%未満である、金属錯体粒子。
  2. 前記磁性体は蛍光を有さない磁性を有する金属錯体を含まない磁性体である、請求項1に記載の金属錯体粒子。
  3.  前記磁性体は前記金属錯体のみである、請求項1に記載の金属錯体粒子。
  4.  前記金属錯体を形成する金属イオンの磁気モーメントが7.5~10.7である、請求項1~3のいずれか1項に記載の金属錯体粒子。
  5.  前記金属錯体を形成する金属イオンがテルビウム、ジスプロシウム、ホルミウム、エルビウムのイオンからなる群から選択される少なくとも一種である、請求項1~4のいずれか1項に記載の金属錯体粒子。
  6.  前記金属錯体を形成する配位子がβ―ジケトン系配位子及び複素環系配位子の両方又はいずれか一方である、請求項1~5のいずれか1項に記載の金属錯体粒子。
  7.  前記金属錯体を形成する配位子が、
    (1)β―ジケトン系配位子は、ジベンゾイルメタン、又はテノイルフルオロアセトン、
    (2)複素環系配位子は、2,2´‐ビピリジル、2,2´:6,2´´-テルピリジン、又は1,10-フェナントロリン、
    から選ばれる配位子である、請求項1~6のいずれか1項に記載の金属錯体粒子。
  8.  前記金属錯体粒子の平均粒径が1μm以上5μm以下である、請求項1~7のいずれか1項に記載の金属錯体粒子。
  9.  前記金属錯体中の前記金属錯体の含有量が、40w%以上98w%以下ある、請求項1~8のいずれか1項に記載の金属錯体粒子。
  10.  前記金属錯体粒子の表面に担持用物質を備える、請求項1~9のいずれか1項に記載の金属錯体粒子。
  11. 前記担持用物質が、検出対象物質と特異的に反応する抗体または抗原である、請求項1~10のいずれか1項に記載の金属錯体粒子。
  12.  請求項10に記載の金属錯体粒子を含む、免疫測定用試薬。
