Abstract

Description

流行性角結膜炎の治療用点眼剤、当該点眼剤を生成するプラズマ活性点眼剤生成装置、及び流行性角結膜炎の治療方法
 本発明は、流行性角結膜炎の新規かつ有効な治療用点眼剤、当該点眼剤を生成するためのプラズマ活性点眼剤生成装置、及び流行性角結膜炎の治療方法に関する。
 従来より、結膜炎は、白目(しろめ)やまぶたの裏側が赤く充血して炎症を起す病気として知られている。結膜炎の種類には色々あり、感染で起こる細菌性結膜炎、ウィルス性結膜炎、アレルギーで起こるアレルギー性結膜炎などさまざまな種類がある。
 結膜炎のうち流行性角結膜炎(EKC:epidemic keratoconjunctivitis)は、ウイルス感染症で、主にD種およびE種のアデノウイルスによる疾患で、主として手を介した接触やくしゃみなどの飛沫により感染する。本疾患は、職場や家庭などの人が濃密に接触する場所、また本患者を扱う医療機関からの感染が多い流行性感染症である。
 流行性角結膜炎の臨床的症状としては、潜伏期が8~14日であり急に発症し眼瞼の浮腫、痛み、流涙を伴う。アデノウイルスは感染力が強いので両眼に感染しやすいが初発眼の症状がより強い。角膜に炎症が及ぶと角膜の透明度が低下し混濁は数年に及ぶことがある。ときに結膜炎が出血性となり、出血性結膜炎や咽頭結膜熱との鑑別を要することがある。流行性角結膜炎の患者数は軽微なケースを含むと日本だけで年間数十万人、世界中では年間二千万人に到ると推測されている。
 ところで、従来より細菌やウイルスを殺菌又は減菌する方法は、熱や圧力による物理的手法と薬剤を用いる化学的手法の2種類に大別されるが、近年、安全性が高く、短時間で殺菌処理のできるプラズマ殺菌技術が普及している。
 プラズマ技術は、電気、化学、材料の各分野に応用され近年、医療応用研究が活発になっている。プラズマの内部では、電子やイオン等の荷電粒子の他に、紫外線やラジカルが発生する。これらには、生体組織の殺菌をはじめとして、生体組織に対する種々の効果があることが分かってきている。例えば、特許文献1(WO2009/041049)には、pHが4.8以下となるように調整された液体にプラズマを照射することにより、液体中の菌を殺菌する技術が開示されている。この特許文献1においては、スーパーオキシドアニオンラジカルやヒドロペルオキシラジカル等が殺菌効果を担っている可能性があると記載されている。また、非特許文献1(H. Yasuda, M. Hashimoto, M. M. Rahman, K. Takashima, A. Mizuno, Plasma Process. Polym. 2010, 7, 301-308.)においてもプラズマ殺菌技術は開示されている。
 特に、解放大気中における非平衡放電プラズマ(低温プラズマ)は大気圧のために多量のラジカルを生成でき、ガス温度が低いために殺菌対象部に熱的ダメージを与えない、排気装置が不要で装置が簡単でコストを要しないなど汎用性が高い。この非平衡放電プラズマは、室温付近でのプラズマプロセスが可能であり、微生物死活化やウィルスの不活化効果も想定され始めている。今後、新型インフルエンザや口蹄疫などによるウイルスの大流行は深刻な問題になると予想され、薬品を使用せずかつ薬品耐性を誘起する事なく短時間でウイルスを不活化できる大気圧低温プラズマを利用した殺菌技術に期待が持たれている。
 このような状況下、現状では、アデノウイルスが引き起こす流行性角結膜炎などのウイルス感染症の全般についても有効な薬剤はない。現状の流行性角結膜炎の対処法は、対症療法的に抗炎症剤の点眼を行い、さらに角膜に炎症がおよび混濁がみられるときにはステロイド剤を点眼する。細菌の混合感染の可能性がある場合には抗菌剤の点眼を行うのが実情である。
 本発明は上記の課題に鑑みてなされたものである、流行性角結膜炎に対する新規かつ有効な治療用点眼剤、当該点眼剤を生成するプラズマ活性点眼剤生成装置、及び流行性角結膜炎の治療方法を提供することを目的とする。
 上記目的を達成するために本発明は、流行性角結膜炎の治療用点眼剤を生成するプラズマ活性点眼剤生成装置であって、電源供給部と、所定の液体を無菌状態で密閉封入する容器本体部を有する点眼容器が挿入される挿入空間の周囲に配置されたプラズマ生成電極と、前記電源供給部に接続されて電源供給を受けると共に、高圧電流を前記プラズマ生成電極に印加する高電圧発生部と、を備え、前記プラズマ生成電極は、前記高電圧発生部から高圧電流を受けて前記所定の液体にプラズマ照射することでプラズマ活性化された流行性角結膜炎の治療用点眼剤であるプラズマ活性点眼剤を生成する、ことを特徴とする流行性角結膜炎の治療用点眼剤を生成するプラズマ活性点眼剤生成装置である。
 本発明に係るプラズマ活性点眼剤生成装置において、前記高電圧発生部は、LF (low frequency)帯交流高電圧発生部であり、前記プラズマ生成電極は、前記LF帯交流高電圧発生部より高圧電流を受けて大気圧非熱平衡プラズマを前記点眼容器に照射することが好ましい。
 本発明に係るプラズマ活性点眼剤生成装置において、前記点眼容器の所定の液体はプラズマ活性化が可能なプラズマ照射溶液であり、前記容器本体部の容器体積の約半分はガスや空気で満たされおり、前記高電圧発生部は、駆動電圧として2kV~10kV、駆動電流として0.1~0.5A、周波数30KHzから80KHzの交流電力を、前記プラズマ生成電極の電極間に印加することで前記プラズマ活性点眼剤を生成することが好ましい。
 本発明に係るプラズマ活性点眼剤生成装置において、前記プラズマ照射溶液は、超純水、ヒアルロン酸ナトリウム溶液及び硝酸銀溶液のいずれか、又はこれらを用いた混合液であることが好ましい。
 本発明に係るプラズマ活性点眼剤生成装置において、前記プラズマ生成電極は、芯材である電極線と、当該電極線の周囲を被覆する誘電体とから構成され、螺旋状に加工され、その中空空間に前記点眼容器が挿入されることが好ましい。
 本発明に係るプラズマ活性点眼剤生成装置において、前記プラズマ生成電極は、平面薄膜状の誘電体の内面及び外面の2か所に平面状の電極が形成され、当該プラズマ生成電極が円筒状に丸められて形成され、前記点眼容器は当該円筒状に形成されたプラズマ生成電極の中空空間に挿入されることが好ましい。
 本発明に係るプラズマ活性点眼剤生成装置において、前記電極線の材料は、柔軟性のある銅線、ステンレス線、アルミ線、鉄線、金線及びこれらの合金線の何れかであり、前記誘電体の材料は、シロキサン結合を有するポリマー又は複数の芳香族がイミド結合したポリマーであることが好ましい。
 本発明に係るプラズマ活性点眼剤生成装置において、前記プラズマ生成電極と、前記点眼容器との間は密着されることが好ましい。
 本発明に係るプラズマ活性点眼剤生成装置において、さらに、絶縁処理された筐体部を備え、当該筐体部には、前記点眼容器が挿入される挿入口と、プラズマ照射を開始するためのスイッチと、少なくともプラズマ照射の完了を示すインジケータとが形成されることが好ましい。
 上記目的を達成するために本発明は、前記プラズマ活性点眼剤生成装置に挿入され、少なくともプラズマが照射される所定の液体を無菌状態で密閉封入する容器本体部と、当該容器本体部に接続されたボトルキャップとを有することを特徴とする点眼容器である。
 本発明に係る点眼容器において、前記所定の液体はプラズマ活性化が可能なプラズマ照射溶液であり、前記容器本体部の容器体積の約半分はガスや空気で満たされ、前記点眼容器は低密度ポリエチレンで形成されることが好ましい。
 本発明に係る点眼容器において、前記プラズマ照射溶液は、超純水、ヒアルロン酸ナトリウム溶液及び硝酸銀溶液のいずれか、又はこれらを用いた混合液であることが好ましい。
 上記目的を達成するために本発明は、流行性角結膜炎の治療用点眼剤であって、前記プラズマ活性点眼剤生成装置を用いて、プラズマ活性化が可能なプラズマ照射溶液に大気圧非熱平衡プラズマを照射して生成されたことを特徴とする。
 本発明に係る流行性角結膜炎の治療用点眼剤において、前記プラズマ照射溶液は、超純水、ヒアルロン酸ナトリウム溶液及び硝酸銀溶液のいずれか、又はこれらを用いた混合液であることを特徴とする。
 上記目的を達成するために本発明は、流行性角結膜炎の治療用点眼剤を生成するプラズマ活性点眼剤生成装置を用いたプラズマ活性点眼剤の生成方法であって、電源供給を受けると共に、高圧電流をプラズマ生成電極に印加する高電圧発生ステップと、所定の液体を無菌状態で密閉封入する容器本体部を有する点眼容器内の液体にプラズマ照射することでプラズマ活性化された流行性角結膜炎の治療用点眼剤であるプラズマ活性点眼剤を生成するプラズマ活性点眼剤生成ステップと、を含むことを特徴とする。
 上記目的を達成するために本発明は、流行性角結膜炎の治療方法であって、前記請求項1に記載の点眼容器を点眼前に前記請求項1記載のプラズマ活性点眼剤生成装置の挿入空間にセットする装着ステップと、前記プラズマ活性点眼剤生成装置の前記点眼容器に対するプラズマ照射を開始するためのスイッチをONとする開始ステップと、プラズマ照射の完了した前記点眼容器を取り出すステップと、前記点眼容器のキャップを外して、前記点眼容器内でプラズマ活性された流行性角結膜炎の治療用点眼剤を、前記プラズマ照射の完了後の所定期間以内に点眼する点眼ステップと、を含むことを特徴とする。
 本発明に係る流行性角結膜炎の治療方法において、前記所定期間内は10分以内であることが好ましい。
 上記目的を達成するために本発明は、前記プラズマ活性点眼剤生成装置と、前記点眼容器のパッケージとから成る、ことを特徴とする流行性角結膜炎の治療用点眼キットである。
 本発明は、流行性角結膜炎の治療用点眼剤を生成するプラズマ活性点眼剤生成装置であって、電源供給部と、所定の液体を無菌状態で密閉封入する容器本体部を有する点眼容器が挿入される挿入空間の周囲に配置されたプラズマ生成電極と、前記電源供給部に接続されて電源供給を受けると共に、高圧電流を前記プラズマ生成電極に印加する高電圧発生部と、を備え、前記プラズマ生成電極は、前記高電圧発生部から高圧電流を受けて前記所定の液体にプラズマ照射することでプラズマ活性化された流行性角結膜炎の治療用点眼剤であるプラズマ活性点眼剤を生成する。この構成により、本発明では、流行性角結膜炎に対する新規かつ有効な治療用点眼剤を生成することができる。

(実施の形態1)
 本発明の実施の形態に係る流行性角結膜炎の治療用点眼剤、当該点眼剤を生成するプラズマ活性点眼剤生成装置、及び流行性角結膜炎の治療方法について図面を参照しながら説明する。
 最初に流行性角結膜炎(EKC:epidemic keratoconjunctivitis)を引き起こすアデノウイルスに関して説明する。アデノウイルス(AD, Adenovirus)は、大きさが直径70nm~90nmであり、DNA遺伝子をもち、カプシド(正20面体)構造体をもっている小さな球形粒子状のウイルスである。アデノウイルス細胞の表面には12本のアンテナ様の突起が見られ、たんぱく質と脂質で構成されるエンベロープは保持しない。
 アデノウイルスは結膜炎、咽頭炎、高熱などの症状を引き起こす原因となり、比較的多く見られるウイルスの一種である。アデノウイルスは、現時点において51種類の血清型が登録されている。これらの中で結膜炎の原因になる血清型は,Ad3,4,7,8,11,19,37, 54, 56型である。これらの血清型は、結膜の上皮細胞に親和性を持ちウイルスのファイバーが宿主細胞のレセプターに接着することにより感染が始まる。その後、核内に封入体が形成され,感染細胞が破壊される。その破壊スピードはそれぞれの血清型によって異なる。
 本発明者は、アデノウイルスに基づく流行性角結膜炎の治療の為にウイルスの不活化作用を有するプラズマ現象に着目した。そして、ある条件下で生成したプラズマを溶液(液体)に照射して、この溶液を角結膜上に点眼することで、角膜や結膜に存在するアデノウイルスを不活化させる可能性に着目し、この為の必要な医薬品構成を見出して本発明に至った。
 次に、本実施の形態に係るEKCの治療に用いる点眼容器の構成に関して図1を参照しながら説明する。図1aは、点眼容器1(UDDC, Unit Dose ophthalmic Dropper Container)の概念図を示しており、容器本体部1a及び容器本体部1aに接続されたボトルキャップ1bを備える。点眼容器1の容器本体部1aは例えば、横幅50mm×奥行10mm×高さ10mm程度の大きさであり、後述するプラズマ活性点眼剤生成装置の内部に装着出来る構造・大きさを有している。
 容器本体部1aの内部には、プラズマが照射されるプラズマ照射溶液(Plasma irradiating solution)が、無菌状態で密閉封入(例えば0.5ml程度の液体封入)され、容器体積の約半分はガスや空気で満たされている。このプラズマ照射溶液は、例えば超純水、ヒアルロン酸ナトリウム溶液及び硝酸銀溶液のいずれか、又はこれらを用いた混合液が封入され滅菌処理されている。当然、プラズマ活性化効果を有する他の液体でも良い。点眼容器1の材料は、例えば低密度ポリエチレン(LDPE)やこれに類似する樹脂である。点眼容器1は、流行性角結膜炎の治療用点眼キットの一部として、例えば図1bに示すようなパッケージとして販売される。
 次に、EKCの治療用の点眼剤のプラズマ活性点眼剤生成装置(Plasma Activated Ophthalmic Solution Generation Device)(以下、生成装置と記載)に関して図2を参照しながら説明する。生成装置2は、点眼容器1にプラズマ照射をして、内部のプラズマ照射溶液をプラズマ活性溶液とすることで流行性角結膜炎の治療用点眼剤を生成する。生成装置2は、非常に小さく、軽量で持ち運び可能な形態を有し、例えば幅50mm×奥行20mm×高さ100mm程度、重量10g程度のものである。
 生成装置2は、点眼容器1をプラズマ照射するために絶縁処理された筐体部2aを備える。この筐体部2aの外装には、図2a及び図2bに示すように、プラズマ照射を開始するためのスイッチ2b、プラズマ照射条件(照射完了など)を示すインジケータ2cと、点眼容器1が挿入される挿入口2dとを備える。
 生成装置2の機能を説明する。生成装置2は、図2cに示すように、プラズマ生成電極2eと、交流電源に接続された電源供給部2fと、LF (low frequency)帯交流高電圧発生部(高電圧発生部)2gと、を備える。プラズマ生成電極2eは、所定の液体を無菌状態で密閉封入する容器本体部1aを有する点眼容器1が挿入される挿入空間Sの周囲に配置され、点眼容器1内の液体にプラズマ照射することでプラズマ活性化された流行性角結膜炎の治療用点眼剤を生成する。電源供給部2fは、筐体部2aの内部に収容されてバッテリーや商用電源に接続される。生成装置2は、バッテリー又は商用電源で駆動される。LF帯交流高電圧発生部2gは、電源供給部2fに接続されて電源供給を受けると共に、高圧電流をプラズマ生成電極2eに印加する。なお、LF帯交流高電圧発生部2gは、プラズマ生成電極2eに間欠的に電界を印加して電子が選択的にエネルギーを受けられる状態とさせ、 電子温度 (電子エネルギー) がガス温度よりも高い状態を作り出すことが出来る。そしてこの状態は、大気圧非熱平衡プラズマ(低温プラズマ)と呼ばれている。
 プラズマ生成電極2eは、金属と誘電体により特徴的に構成され、LF帯交流高電圧発生部2gに接続されて高圧電流を受けて電極間にプラズマを生成する。なお、プラズマ生成電極2eの電極間に点眼容器1が配置される。生成装置2は、アデノウイルスを不活化するプラズマ活性点眼剤を生成するための最適なプラズマ生成方法と条件を有している。
 次に、プラズマ活性点眼剤生成装置2に備わるプラズマ生成回路の概念に関して図3を参照しながら説明する。図3は、このプラズマ生成回路の概念図であり、基本的には、電源供給部2fと、プラズマ生成電極2eとが必要とされる。
 本実施の形態において、プラズマ生成回路は、一対のプラズマ生成電極2eと、この一対の電極2eに交流電圧を印加する電源供給部2fとを有する。ここで、一対の電極2eは、後述するように、表面にポリシロキサン等の誘電体膜が形成されており、電源供給部2fから一対の電極2eに交流電圧が印加されたとき、放電空間内にプラズマ状態が生成される。
 より具体的には、図3に示すプラズマ生成電極2eは、2つの並行平板電極を用いた電極21,22を有しており、電極間には例えばAC 4kV p-p、共振周波数40kHz程度の電圧が電源供給部2fより印加される。高圧電極21と接地電極22との間に点眼容器1を置き、高圧電極21及び接地電極22と点眼容器1との間には2mm程度の空隙たる絶縁バリア2hがある。この絶縁バリア2hがプラズマ照射距離となり、最適なアデノウイルスの殺菌効果を発揮するプラズマ活性点眼剤を生成するにはこの照射距離も重要となる。このため、高圧電極21及び接地電極22と点眼容器1との間をできるだけ密着することも考え得る。
 なお、この絶縁バリア2hに、ガスを封入することも可能であり、例えば、使用するガス種は、空気・ヘリウム・アルゴン・窒素やそれら混合ガス、酸素と窒素の混合ガスをアルゴンやヘリウムで希釈したガスである。絶縁バリア2hを介した電極21,22の間に低温プラズマを発生させ、その中で生成するラジカルが点眼容器1中の液体に注入され、プラズマ活性化された点眼剤が生成される。この際、生成するラジカル種は、使用するガス種により異なるが、例えば酸素ラジカル、ヒドロキシルラジカル、オゾンラジカルである。
 プラズマ活性点眼剤(PAOS; Plasma Activated Ophthalmic Solution)は、滅菌した点眼液を封入した点眼容器1に、生成装置2でプラズマ照射され、生成された溶液である。プラズマ中には、電子,イオン,ラジカル,光,電界等、及び雰囲気中の酸素,窒素,水蒸気等と反応して酸素ラジカル,窒素ラジカル,ヒドロキシルラジカル,一酸化窒素等が生成される。そしてプラズマ活性点眼溶剤の中には、過酸化水素,硝酸イオン,亜硝酸イオン等が生成される。また、溶液中の成分とも反応して反応生成物も生成され、これら生成物がアデノウイルスのカプシドを酸化分解する。酸化分解されたカプシド内のウイルスDNAは酸素ラジカルや紫外線の攻撃を受け、最終的にアデノウイルスが不活性化する。なお、アデノウイルスの不活性化を確かめるため、発明者らは、後述の図8において、誘電体バリア放電方式でプラズマ照射した大腸菌ウイルスであるλファージを用いてウイルスがどのように損傷を受けるかを調べた。
 <変形例1>
 次に、本実施の形態に係る生成装置2に備わるプラズマ生成電極2eの変形例1に関して図4を参照しながら説明する。本変形例1においては、プラズマを発生させるプラズマ生成電極2eを誘電体バリア放電型とし、小型化を可能としている。
 従来より、プラズマを発生させる装置は様々な形態がある。その一つとして電極の表面にガラスやセラミック等の材料で誘電体を形成してプラズマを発生させる誘電体バリア放電が知られている。大気中に存在する窒素分子等は、安定した状態で大気中に存在しており、これらの分子を電離させてプラズマ状態にするには、極めて高い電圧を印加する必要がある。従来のセラミックを用いる方法では、電極形状の自由度が制限され装置の小型化には不適である事、高温焼成にて生成され焼成後の形状安定性が低い事、製造コストが高い事、重量が重くなり軽量化が難しくなる事、さらにセラミックを用いた誘電体バリア放電では、依然として高電圧が必要で消費電力が大きくなるなど使用環境が限られてしまう事などの課題がある。
 一方、本変形例1で使用するプラズマ生成電極2eは、このような従来の課題を解決するものであり、実装自由度が大きく、安価で消費電力の少なく量産性と使用環境への自由度が高いプラズマ発生装置を提供する事が可能となる。
 具体的に、本変形例1に係るプラズマ生成電極2eの電極構造に関して図4を参照しながら説明する。図4a及び図4bは、プラズマ生成電極2eの正面からの概念図であり、例えば、断面形状が円形の線状を有し、一対の電極は撚った構造に加工される。プラズマ生成電極2eは、図4cの断面図に示すように、芯材である電極線23と、電極線23の周囲を被覆する誘電体24とを有する。プラズマ生成電極2eは、図4a及び図4bで示すように螺旋状に加工され、その中空空間に点眼容器1が挿入される構造となるため、プラズマ生成電極2eと点眼容器1との間をできるだけ密着できる。但し、プラズマ生成電極2eの構造は、これに限らず、板材、管材であってもよく、フィルム状に形成された導電体であってもよい。
 電極線23は、図4cに示すように、その周囲に誘電体24の膜が被覆されている。この際、電極線23の断面直径5mmに対して、誘電体24の膜厚は、50~200μm程度である。なお、誘電体24の膜厚を大きくすれば、強度的には高くなるが、その反面、放電し難くなるため高い放電電圧を印加する必要があり、誘電体24の膜厚は可耐性許容範囲内で可能な限り薄くする事が望ましい。
 電極線23の材料は、柔軟性のある銅線、SUS線(ステンレス線)、アルミ線、鉄線、金線及びこれらの合金線等を用いることができる。また、電極線23は、必ずしも直線状のものを螺旋状にする必要はなく、例えば、リング状に形成されていてもよく、図4に示すような形態に限られるものではなく、例えば一対の平板電極でこの平板電極に誘電体膜が形成されていてもよい。
 誘電体24の材料は、図5aに示すようなシリコンと酸素が直鎖状に連なったシロキサン結合を有するポリマー(ポリシロキサン)、又は図5bに示すような複数の芳香族がイミド結合したポリマー(ポリイミド)である。 絶縁耐力と電極と誘電体とのギャップは比例、誘電体の厚みは反比例にあり、可能な限りギャップを無くしかつ厚みを薄くする事で低電力高効率にプラズマを生成出来る事になる。
 図5aはシリコン化学式を示す。R,R′あるいは重合度nを変えることによって,油状(シリコン油),ゴム状(シリコンゴム),樹脂状(狭義のケイ素樹脂,シリコン樹脂)と多様に変化する。図5bはポリイミド化学式を示す。R および R' が芳香族である場合を芳香族ポリイミドと呼び、工業的に利用されるほとんどのものがこの芳香族ポリイミドである。そして、これらポリマーを誘電体24の材料として用いて、電極線23に塗布し塗布後に乾燥させ電極線23の表面に一定の膜厚の誘電体24の膜を形成する。本変形例1では、誘電体24の膜を電極線23の表面上に塗布乾燥することにより形成できるため、電極線23の形状の自由度を高めることが可能になる。なお、誘電体24の膜の生成方法は、これに限らず、例えば、ディッピング、印毛塗り、スプレーによる吹付け、スクリーン印刷による方法も可能である。
 なお、従来の大気圧プラズマ発生装置は、誘電体材料にセラミックやガラスといった固形物が用いられていたために小型化、フレキシブル化、軽量化が困難であり、ひいてはセラミックやガラスを用いるため、放電させるには高電圧を必要としていた。これに対し、本変形例1に係る発生装置2に用いるプラズマ生成電極2eでは、誘電体24の形成を塗布乾燥という工程で行うことができ、乾燥後に形成される誘電体24の厚みは約100μm程度であり、電極の形状にこだわらず実行放電距離を小さくすることに非常に有利となる。実行放電距離を小さくすることが出来れば、放電に要するエネルギーも少なくでき、数W程度の消費電力でプラズマ発生をすることができる。さらに、誘電体24の形成に自由度が高いため、単純な電極構成でプラズマ発生が可能であり、撚線は曲げて形状を変化させることも可能である。
 次に、本変形例1に係る生成装置2を動作させるときの電気的特性について説明する。LF帯交流高電圧発生部2gは、駆動電圧として2~10kV、駆動電流として0.1~0.5A、周波数30KHzから80KHzの交流電力を一対の電極間に印加する。
 <変形例2>
 なお、変形例2として、プラズマ生成電極2eを図6に示すような電極構造とすることも考え得る。この変形例2の場合、プラズマ生成電極2eは、ポリシロキサンやポリイミドから成る平面薄膜状の誘電体24の内面及び外面の2か所に平面状の電極23a,23bが形成され、このプラズマ生成電極2eが図6dに示すように円筒状に丸められて形成される。点眼容器1はこの円筒状に形成されたプラズマ生成電極2eの中空空間に挿入される。
 次に、本実施の形態に係る生成装置2を用いたEKCの治療方法の手順に関して図7に示すフローチャートを参照しながら説明する。
 最初に、EKC感染患者などのユーザは、点眼容器1を点眼前に生成装置2の挿入空間Sにセットする(S101)。次に、生成装置2の点眼容器1に対するプラズマ照射を開始するためのスイッチ2bをONとする(S102)。すると、生成装置2では、生成スイッチ2bをトリガとして、滅菌した点眼液を封入した点眼容器1に対するプラズマ照射を開始して、プラズマ活性点眼剤が点眼容器1内に生成される(S103)。そして、プラズマ活性点眼剤の生成が完了すると(S104でYes)、生成装置2にあるインジケータ2cが点灯又は点滅などしてプラズマ活性点眼剤の生成完了をユーザに伝える。
 次に、ユーザが点眼容器1を生成装置2から取り出し、生成完了後の所定時間(例えば、生成完了直後から10分程度)以内に点眼容器1のキャップ1bを取り外して点眼する(S105)。これは、プラズマ活性点眼溶液のラジカルやイオン等の状態は時間に依存して変化するためであり、この為、発明者らは、点眼少し前にプラズマ活性点眼剤を生成する事が最も安定した状態で点眼出来る方法であると発見し、その為に必要な構成を見出した。そして、最後に、アデノウイルスの疾患の完治治療の為に、数時間ごとに一日数回点眼しこれを数日継続する(S106)。
 以上の説明のように、本実施の形態に係る流行性角結膜炎の治療用点眼剤を生成するプラズマ活性点眼剤生成装置2は、電源供給部2fと、所定の液体を無菌状態で密閉封入する容器本体部1aを有する点眼容器1が挿入される挿入空間の周囲に配置されたプラズマ生成電極2eと、電源供給部2fに接続されて電源供給を受けると共に、高圧電流をプラズマ生成電極2eに印加する高電圧発生部2gとを備える。プラズマ生成電極2eは、高電圧発生部2gから高圧電流を受け、前記所定の液体にプラズマ照射することでプラズマ活性化された流行性角結膜炎の治療用点眼剤であるプラズマ活性点眼剤を生成する。
 この構成により、流行性角結膜炎に対する新規かつ有効な治療用点眼剤、当該点眼剤を生成するプラズマ活性点眼剤生成装置を提供することができる。すなわち、生成装置2は、所定の条件のもとで点眼容器1中のプラズマ照射溶液(或いは生成装置2内の溶液)にプラズマ照射してプラズマ活性点眼剤を生成し、それを所定の条件内で細菌やアデノウイルスに直接投与することで、殺菌とウィルス不活化効果を与えることができる。
 また、本発明者は、眼表面にプラズマを直接照射する事による角膜障害等のリスクを排除する為に、プラズマを照射した液体であるプラズマ活性点眼剤を生体(細胞)の表面に投与して、特にアデノウイルスなどのウイルス不活化効果を得る事を成功したものである。特に、大気圧非熱平衡プラズマを照射して生じる活性酸素種は、ウィルスのカプシドを酸化分解することによりウィルスの不活化(感染能)を亢進する。正常細胞に影響を与えない密度のプラズマ(ラジカル)はカプシドを破壊して抗ウィルス活性を発揮する事が出来る。
 なお、本願発明の供給形態は、プラズマ活性点眼剤生成装置2と、点眼容器1のパッケージから成る流行性角結膜炎の治療用点眼キットとすることができる。
 <実験結果>
 次に、本実施の形態に係るプラズマ活性点眼剤生成装置2のプラズマ発生電極2eの発光レベルに関して検証する。図8は、プラズマ発生装置により大気中でプラズマを発生させたときの発光スペクトルを示し、波長が10-400nm程度の紫外線域に強い輝度の紫外線発光、Nプラズマ、Oプラズマの発光が観測されている。プラズマ活性点眼剤からはこれらの紫外線、Nプラズマ、Oプラズマなどからラジカルが発生してアデノウイルスの殺菌要因となっていると考えられる。
 図9及び図10は、本実施の形態に係るプラズマ活性点眼剤の殺菌力を検証した実験結果を示す。なお、試験資料には、大腸菌: E. coli ATCC13706 とファージ(細菌に感染するウイルス): ΦX174 phageを使用した。なお、図中のCFUとは、Colony Forming Unitの略称で菌量の単位を現している。例えば 20CFU/mlとは1ml中に菌が20個存在することを表している。大腸菌やファージはプラズマ照射後に殺菌されるので縦軸は時間経緯時点で生き残った菌数を照射直前の値を1としてその相対値で示している。
 <実験例1>
 この実験では直接照射における効果確認を目的している。条件としては、MilliQ水に、大腸菌(E.coli ATCC13706) 1.E+08を含む懸濁液 0.5mL をシャーレに入れ、試料溶液濃度を2×108/mLとして、放電装置の電極を懸濁液と2mmの距離に置き、照射時間後 0,1,2,4,8,16,32分に菌数を計測した。その結果を図9(〇)に示す。
 次に、同様な条件で、ΦX174 phageにて行いファージを計測した結果が図10(〇)となる。
 本図に示すように、直接照射においては高速の滅菌効果が確認された。これは、生成装置2で試料溶液に生成されたラジカルにより大腸菌やΦX174 phageの細胞壁やカプシドが破壊され、内部のDNAが破壊されて結果として菌やウイルスの不活性化に至っていることが分かる。
 <実験例2>
 この実験では間接照射における効果確認を目的としている。指定した照射時間照射したPTW(Milli-Q水にプラズマ照射したもの)から135uL採取し、大腸菌(E.coli ATCC13706)を含む菌液(約 2×108/mL) 15uLと混合(約10倍希釈)し、混合後20分間、20℃で保存し寒天培地で段階的希釈し、照射時間後 0,1,2,4,8,16分に菌数を計測した。その結果を図9(×)に示す。 
 次に、同様な条件で、ΦX174 phageにて行いファージを計測した結果が図10(×)となる。
 本図に示すように、間接照射においても、直接照射よりは遅いが高速での滅菌効果が確認された。即ち、本実施の形態に係るプラズマ活性点眼剤が、アデノウイルスの不活性化に非常に有効な効果があることがこの実験により確認された。
 なお、本発明は、上記各実施の形態の構成に限られず、発明の趣旨を変更しない範囲で種々の変形が可能である。例えば、EKCに対する抗アデノウイルス剤としての使用例以外の方法でも、異なる溶液や容器材料を用いてプラズマ活性された溶液でアデノウィルス不活効果を与える事ができる。また、特にラジカル活性溶液は、体の器官の表面に存在する細菌やウイルスの殺菌剤や殺菌方法として用いることができ、別な用途として口内にPTWを含む事で口内細菌を殺菌する事も考え得る。
 また、本発明は、このような生成装置として実現することができるだけでなく、このような生成装置が備える特徴的な手段をステップとするプラズマ活性点眼剤の生成方法として実現したり、それらのステップをコンピュータに実行させるプログラムとして実現できることはいうまでもない。

Claims (18)

  1.  流行性角結膜炎の治療用点眼剤を生成するプラズマ活性点眼剤生成装置であって、
     電源供給部と、
     所定の液体を無菌状態で密閉封入する容器本体部を有する点眼容器が挿入される挿入空間の周囲に配置されたプラズマ生成電極と、
     前記電源供給部に接続されて電源供給を受けると共に、高圧電流を前記プラズマ生成電極に印加する高電圧発生部と、を備え、
     前記プラズマ生成電極は、前記高電圧発生部から高圧電流を受けて前記所定の液体にプラズマ照射することでプラズマ活性化された流行性角結膜炎の治療用点眼剤であるプラズマ活性点眼剤を生成する、ことを特徴とする流行性角結膜炎の治療用点眼剤を生成するプラズマ活性点眼剤生成装置。
  2.  前記高電圧発生部は、LF (low frequency)帯交流高電圧発生部であり、
     前記プラズマ生成電極は、前記LF帯交流高電圧発生部より高圧電流を受けて大気圧非熱平衡プラズマを前記点眼容器に照射する、ことを特徴とする請求項1記載のプラズマ活性点眼剤生成装置。
  3.  前記点眼容器の所定の液体はプラズマ活性化が可能なプラズマ照射溶液であり、
     前記容器本体部の容器体積の約半分はガスや空気で満たされおり、
     前記高電圧発生部は、駆動電圧として2kV~10kV、駆動電流として0.1~0.5A、周波数30KHzから80KHzの交流電力を、前記プラズマ生成電極の電極間に印加することで前記プラズマ活性点眼剤を生成する、ことを特徴とする請求項2記載のプラズマ活性点眼剤生成装置。
  4.  前記プラズマ照射溶液は、超純水、ヒアルロン酸ナトリウム溶液及び硝酸銀溶液のいずれか、又はこれらを用いた混合液である、ことを特徴とする請求項3記載のプラズマ活性点眼剤生成装置。
  5.  前記プラズマ生成電極は、芯材である電極線と、当該電極線の周囲を被覆する誘電体とから構成され、螺旋状に加工され、その中空空間に前記点眼容器が挿入される、ことを特徴とする請求項1記載のプラズマ活性点眼剤生成装置。
  6.  前記プラズマ生成電極は、平面薄膜状の誘電体の内面及び外面の2か所に平面状の電極が形成され、当該プラズマ生成電極が円筒状に丸められて形成され、
     前記点眼容器は当該円筒状に形成されたプラズマ生成電極の中空空間に挿入される、ことを特徴とする請求項1記載のプラズマ活性点眼剤生成装置。
  7.  前記電極線の材料は、柔軟性のある銅線、ステンレス線、アルミ線、鉄線、金線及びこれらの合金線の何れかであり、前記誘電体の材料は、シロキサン結合を有するポリマー又は複数の芳香族がイミド結合したポリマーである、ことを特徴とする請求項5又は6記載のプラズマ活性点眼剤生成装置。
  8.  前記プラズマ生成電極と、前記点眼容器との間は密着される、ことを特徴とする請求項5又は6記載のプラズマ活性点眼剤生成装置。
  9.  前記プラズマ活性点眼剤生成装置は、さらに、絶縁処理された筐体部を備え、
     当該筐体部には、前記点眼容器が挿入される挿入口と、プラズマ照射を開始するためのスイッチと、少なくともプラズマ照射の完了を示すインジケータとが形成される、ことを特徴とする請求項1記載のプラズマ活性点眼剤生成装置。
  10.  前記請求項1記載のプラズマ活性点眼剤生成装置に挿入され、少なくともプラズマが照射される所定の液体を無菌状態で密閉封入する容器本体部と、当該容器本体部に接続されたボトルキャップとを有する、ことを特徴とする点眼容器。
  11.  前記所定の液体はプラズマ活性化が可能なプラズマ照射溶液であり、前記容器本体部の容器体積の約半分はガスや空気で満たされ、前記点眼容器は低密度ポリエチレンで形成される、ことを特徴とする請求項10記載の点眼容器。
  12.  前記プラズマ照射溶液は、超純水、ヒアルロン酸ナトリウム溶液及び硝酸銀溶液のいずれか、又はこれらを用いた混合液である、ことを特徴とする請求項11記載の点眼容器。
  13.  流行性角結膜炎の治療用点眼剤であって、
     前記請求項1記載のプラズマ活性点眼剤生成装置を用いて、プラズマ活性化が可能なプラズマ照射溶液に大気圧非熱平衡プラズマを照射して生成された、ことを特徴とする流行性角結膜炎の治療用点眼剤。
  14.  前記プラズマ照射溶液は、超純水、ヒアルロン酸ナトリウム溶液及び硝酸銀溶液のいずれか、又はこれらを用いた混合液である、ことを特徴とする請求項13記載の流行性角結膜炎の治療用点眼剤。
  15.  流行性角結膜炎の治療用点眼剤を生成するプラズマ活性点眼剤生成装置を用いたプラズマ活性点眼剤の生成方法であって、
     電源供給を受けると共に、高圧電流をプラズマ生成電極に印加する高電圧発生ステップと、
     所定の液体を無菌状態で密閉封入する容器本体部を有する点眼容器内の液体にプラズマ照射することでプラズマ活性化された流行性角結膜炎の治療用点眼剤であるプラズマ活性点眼剤を生成するプラズマ活性点眼剤生成ステップと、を含むことを特徴とする流行性角結膜炎の治療用のプラズマ活性点眼剤の生成方法。
  16.  流行性角結膜炎の治療方法であって、
     前記請求項1に記載の点眼容器を点眼前に前記請求項1記載のプラズマ活性点眼剤生成装置の挿入空間にセットする装着ステップと、
     前記プラズマ活性点眼剤生成装置の前記点眼容器に対するプラズマ照射を開始するためのスイッチをONとする開始ステップと、
     プラズマ照射の完了した前記点眼容器を取り出すステップと、
     前記点眼容器のキャップを外して、前記点眼容器内でプラズマ活性された流行性角結膜炎の治療用点眼剤を、前記プラズマ照射の完了後の所定期間以内に点眼する点眼ステップと、を含むことを特徴とする流行性角結膜炎の治療方法。
  17.  前記所定期間内は10分以内である、ことを特徴とする請求項16に記載の流行性角結膜炎の治療方法。
  18.  前記請求項1記載のプラズマ活性点眼剤生成装置と、前記点眼容器のパッケージとから成る、ことを特徴とする流行性角結膜炎の治療用点眼キット。
