Description
B 세포 성숙 항원(B Cell Maturation Antigen: BCMA) 단백질에 특이적으로 결합하는 항체-약물 접합체, 이를 제조하는 방법, 및 그의 용도에 관한 것이다.
B 세포 성숙 항원(B Cell Maturation Antigen: BCMA)은 약 20kDa의 분자량을 가지는 단백질로서 종양괴사인자 수용체(Tumor Necrosis Factor receptor: TNFR)에 에 속하는 단백질이다. 병리적 상황에서 BCMA는 다발성 골수종(multiple myeloma: MM) 환자의 종양 형질 세포(neoplastic plasma cell)에서 발현하며, 다발성 골수종 환자군의 생존율은 BCMA 발현이 높을수록 낮아진다(Moreaux et al., Eur J Haematol 2009; 83:119-129, Novak et al., Blood. 2004; 103:689-694, Sanchez et al., Br J Haematol 2012;158:727-738).
다발성 골수종은 형질세포의 단클론성의 증식으로 인하여 발생하는 종양성 질환으로 2000년도 이후 탈리도마이드(thalidomide), 보르테조밉(bortezomib), 레날리도마이드(lenalidomide) 등의 약제가 개발되어 환자들의 기대수명 증가에 기여하였다. 그러나 대부분의 환자에서 결국에는 재발이 되고 재발 시 이들 약제에 대한 반응성이 현저하게 낮아져 이들 약을 더이상 쓸 수 없어 재발성/난치성 환자에 대한 치료약 개발이 시급하다.
항체-약물 접합체는 단독 항체가 가진 단점인, 저분자 약물 대비 낮은 치료 효과를 약물과의 결합을 통하여 개선하고자 개발되었다. 저분자 합성 의약품 항암제의 경우 직접적으로 세포 사멸을 유도할 수 있으나 항체는 그 자체로는 표적 단백질에 결합을 할 뿐이고 약리적 효과를 일으키기 위해서는 면역세포나 표적자체의 성질에 의존해야 하는 수동적 물질이다. 이러한 한계점을 극복하기 위해 항체에 약물을 결합시켜 이 결합체가 능동적으로 약리적 효과를 일으키게 만든 것이 항체-약물 복합체이다. 접합된 약물은 항체의 약리적 특징을 강화시켜서 항체-약물 접합체는 화학요법 약제와의 병용투여 없이 단독 요법으로도 충분한 항암능력을 보일 수 있다.
그러나 항체-약물 접합체는 높은 역가로 인해 단독 항체에 비해서 일반적으로 독성이 높으며 항체-약물 접합체를 제작하는 방식에 의해서도 이러한 독성은 차이가 난다. 따라서 이러한 독성을 낮추는 것이 항체-약물 접합체 개발 시 고려해야할 문제이다. 항체 약물 접합체의 독성을 유발하는 요인으로는 생체 내 안정성이 있으며 생체 내 안정성에 영향을 주는 요소로는 약물의 결합 비율(Drug Antibody Ratio: DAR) 항체에 결합한 소수성 약물의 영향, 약물의 결합부위 등이 있다. 이러한 요인들이 결합하여 생체내 반감기로 대표되는 생체 내 안정성에 영향을 주고 독성을 유발한다. 여기에서부터 다양한 기술적 문제가 도출된다. 먼저, 표적 특이적 치료를 위하여 항체의 항원 결합능 및 Fc 기능을 방해하지 않아야 하며, 항체 대 약물의 결합 비율을 높임으로써 치료 효과를 증가시키고, 항체 대 약물의 결합 비율에 따라 항체-약물 결합체의 생체 내 안정성, 즉 혈중 반감기가 감소하지 않아야 한다. 암세포 표면 항원의 발현 정도가 낮은 경우를 고려하면, 높은 세포 독성을 유지하기 위해서는 최대한 높은 DAR을 유지해야 하나, DAR이 8에 이른 경우, 항체에 결합한 소수성 약물의 영향으로 혈중 반감기가 감소하여 독성은 증가하고 생체 내 효과(in vivo efficacy)가 감소할 수 있는 단점이 있다.
따라서, 기존 약물대비 항암 효과가 우수하면서 독성이 없는 다발성 골수종 치료제를 개발할 필요가 있다.

상기 항체 또는 그의 항원 결합 단편은 서열번호 9 내지 14로 이루어진 군으로부터 선택된 하나 이상의 아미노산 서열을 포함하는 중쇄 가변 영역, 서열번호 15 내지 20 및 30 내지 34로 이루어진 군으로부터 선택된 하나 이상의 아미노산 서열을 포함하는 경쇄 가변 영역, 또는 상기 중쇄 가변 영역 및 상기 경쇄 가변영역을 포함할 수 있다.
상기 중쇄(heavay chain)는 5가지(γ, δ, α, μ, ε) 종류가 있으며 중쇄가 항체의 종류를 결정짓는다. α와 γ는 450개, μ 와 ε는 550개의 아미노산으로 구성되어 있다. 중쇄는 두 영역 즉 가변 영역과 불변 영역이 있다.
상기 경쇄(light chain)는 λ, κ 2가지 종류가 있으며 대략 211 내지 217개의 아미노산으로 구성되어 있다. 사람의 항체 각각에는 모두 동일하게 1가지의 쇄만이 존재한다. 경쇄는 불변 영역과 가변 영역이 연속적으로 이루어져 있다.
상기 가변 영역(variable region)은 항체에서 항원이 결합하는 영역을 말한다.
상기 중쇄 가변 영역은 서열번호 9 또는 10으로 이루어진 아미노산 서열을 포함하는 상보성 결정 영역(complementarity determining region: CDR)-H1; 서열번호 11 또는 12로 이루어진 아미노산 서열을 포함하는 CDR-H2; 및 서열번호 13 또는 14로 이루어진 아미노산 서열을 포함하는 CDR-H3을 포함할 수 있다. 용어 "상보성 결정 영역(Complementarity-determining region: CDR)"은 항체의 가변 부위 중에서 항원과의 결합 특이성을 부여하는 부위를 말한다. 예를 들어, 상기 중쇄 가변 영역은 서열번호 5 또는 6으로 이루어진 아미노산 서열을 포함할 수 있다.

이하 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1. 항-BCMA 항체의 준비 및 항-BCMA 항체의 효과
1. 항원의 제조
항-BCMA 항체 제조를 위해 항원을 다음과 같이 제조하였다. 인간 BCMA(NP_001183.2, 서열번호 1)의 아미노산 서열에서 N-말단으로부터 5 내지 54번째 잔기, 1 내지 54 번째 잔기, 및 4 내지 48 번째 잔기를 포함하는 항원을 각각 사용하였다.
구체적으로, 인간 BCMA 5 내지 54번째 잔기(Genscript®Z02731)로 구성된 항원("인간 BCMA-His(5-54)"), 인간 BCMA 1-51(자체 제작, CHO 세포에서 발현시킴)를 인간 IgG1의 Fc 영역에 융합시킨 항원("인간 BCMA-Fc(1-51)), C 말단에 Fc 영역 및 His 태그가 융합된 인간 BCMA 1-51 항원(10620-H03H, Sino Biological Inc.)("인간 BCMA-Fc/His(1-51)"), 및 인간 BCMA 4-48(자체 제작, HEK293 세포에서 발현시킴)을 Fc 영역에 융합시킨 항원("인간 BCMA-Fc(4-48)")을 준비하였다.
인간 BCMA-Fc(4-48)는 하기와 같이 제조하였다. 인간 BCMA 4 내지 48 번째 잔기를 암호화하는 폴리뉴클레오티드를 CMV 프로모터를 포함하는 동물세포 발현용 벡터인 pAB1-Fc에 클로닝하였다. 클로닝된 벡터를 HEK293E 세포에 형질전환시키고, 단백질 A 친화성 크로마토그래피를 사용하여 인간 BCMA-Fc(4-48)를 정제하였다.
또한, 종간 교차반응성을 확인하기 위해, 인간 IgG1의 Fc 영역이 융합된 원숭이(Rhesus) BCMA(1-53)(90103-C02H, Sino Biological Inc.), 마우스 BCMA(1-49)(50076-M01H, Sino Biological Inc.), 및 래트 BCMA(1-49)(80156-R01H, Sino Biological Inc.)를 사용하였다. 원숭이 BCMA(1-53), 마우스 BCMA, 및 래트 BCMA의 아미노산 서열을 하기 표 1에 기재하였다.

2. 파아지 디스플레이 패닝(phage display panning) 및 단일클론 파아지 항체 선별(single clone screening) 다양한 항원에 대해 결합 가능성을 가지는 인간 유래 ScFv(Single-chain variable fragment) 파아지 라이브러리 세포(Mol. Cells OT, 225-235, February 28, 2009)를 준비하였다. 인간 BCMA, 인간 BCMA 및 원숭이 BCMA에 교차 반응성이 있는 항체를 스크리닝하기 위해, 준비된 파아지 라이브러리를 사용하여 총 2회 패닝을 수행하였다. BCMA 단백질에 결합하는 BCMA-특이적인 파아지를 회수하고, K12 ER2738 대장균에 감염시키고, 다시 파아지를 회수하는 과정을 4회 반복하여 파아지를 패닝하였다. 패닝 라운드가 진행될수록 PBS-T를 이용한 세척 회수를 증가시켜 항원-특이적 파아지를 증폭 및 농축하였다.
수득한 파아지 풀(phage pool)을 순차 희석하고, LB-테트라사이클린/카베니실린을 함유한 고체 배지에서 배양하여 단일 콜로니를 확보하였다. 각 콜로니로부터 얻은 파아지를 사용하여 재조합 BCMA에 대한 ELISA를 수행하였다. 각 웰 당 100 ng의 재조합 BCMA를 ELISA 플레이트에 코팅하고, 비특이적 결합을 방지하기 위해 3% BSA로 코팅한 후 플레이트를 PBS로 세척하였다. 준비된 단일 클론 파아지를 각 웰에 가한 후 37℃에서 1시간동안 인큐베이션하고, 플레이트를 PBS-T로 3회 세척하였다. HRP(horseradish peroxidase)가 접합된 항-HA(hemagglutinin) 항체와 TMB(Tetramethylbenzidine, Sigma, T0440)를 사용하여 ELISA를 수행하였다. 450 nm에서의 흡광도가 0.5 이상이면서, 항-HA HRP 단독인 대조군의 흡광도와 비교하여 5배 이상의 흡광도가 증가된 클론을 선별하였다. 이에 따라, 인간 BCMA에 특이적으로 결합하는 항체 클론 2종 B58 및 5D5를 선별하였다.
선별된 항체를 암호화하는 핵산 서열로부터 항체 중쇄 가변영역의 아미노산 서열(서열번호 5 내지 6) 및 경쇄 가변영역의 아미노산 서열(서열번호 7 내지 8)을 분석하고, Kabat 정의에 따라 상보성 결정 영역(complementarity determining region: CDR)을 결정하였다. 결정된 중쇄 및 경쇄의 CDR 아미노산 서열(N->C)을 각각 표 2 및 표 3에 나타내었다.

3. 선별된 항-BCMA phage로부터 항-BCMA IgG 항체의 생산
실시예 1.3에서 선별된 항체를 암호화하는 핵산 서열을 갖는 폴리뉴클레오티드를 합성하였다. 준비된 폴리뉴클레오티드를 동물세포 배양용 벡터(중쇄 발현 벡터: pAB1-HC, 경쇄 발현 벡터: pAB1-LC)에 클로닝하였다. 각 2종의 항체 클론에 대하여 각각 중쇄 및 경쇄를 암호화하는 폴리뉴클레오티드를 포함한 총 4개의 벡터를 제조하였다. 준비된 벡터는 IgG1 타입 서열을 포함한다.
CHO-S 세포를 CD-CHO(Gibco, 10743) 배지에서 배양하고, 준비된 벡터는 폴리에틸렌이민(polyethylenimine: PEI)을 사용하여 CHO-S 세포에 도입하였다. 형질도입된 CHO-S 세포를 CD-CHO 배지에서 8% CO2, 37℃, 및 110 rpm의 조건에서 약 4일 동안 배양하였다.
평형화 완충액(50 mM Tris-HCl, pH7.5, 100 mM NaCl)으로 평형화된 MabSelect SuRe 컬럼(GE healthcare, 5 ㎖)에 준비된 CHO-S 세포 배양액을 통과시켜, 발현된 항체가 컬럼에 결합하도록 하였다. 항체를 50 mM Na-시트르산염(pH 3.4) 및 100 mM NaCl의 용액으로 용출시킨 후, 1M Tris-HCl(pH 9.0)으로 중화시켜 최종 pH가 7.2가 되게 하였다. 이후 완충액을 PBS(pH 7.4)로 교환하여 사용시까지 항-BCMA IgG 항체인 B58 및 5D5를 4℃에서 보관하였다.
4. 5D5의 돌연변이의 제조
선별된 5D5 항체의 생산성을 개선하기 위해, 표 4의 핵산 서열을 이용하여, 항체의 경쇄 CDR 중 1개의 아미노산 잔기를 돌연변이시킨 돌연변이 항체를 제조하였다.
5D5 돌연변이 항체의 경쇄 가변영역(서열번호 25 내지 29) 중 CDR-L1, CDR-L2, 및 CDR-L3의 아미노산 서열을 표 5에 나타내었다. 표 5에서, 밑줄 및 굵은 글씨로 표시된 아미노산 잔기가 돌연변이된 부분이다(WT: 야생형, LM: 경쇄 돌연변이).

5. 항-BCMA IgG 항체의 BCMA에 대한 결합력의 확인(1) 재조합 BCMA에 대한 결합력의 확인
실시예 1.3에서 분리된 항-BCMA IgG 항체의 재조합 BCMA 단백질에 대한 특이적 결합력을 ELISA 방법으로 분석하였다.
항원으로는 재조합 인간 BCMA를 사용하고, 2차 항체로서 HRP-접합된, 인간 Fab 특이적 염소 다중클론 항체 시약(Pierce, 31414)을 사용하여, 실시예 1.2에 기재된 바와 같이 ELISA를 수행하였다. 항체의 농도에 따른 450 nm에서 흡광도를 도 1a에 나타내었다. 도 1a는 B58 및 5D5 항체의 인간 BCMA에 대한 결합력을 나타낸 그래프이다.
도 1a에 나타난 바와 같이, B58 항-BCMA 항체 및 5D5 항-BCMA 항체 모두 인간 BCMA에 대해 농도 의존적으로 결합함을 확인하였다.
(2) 세포 표면의 BCMA에 대한 결합력의 확인
선별된 항-BCMA IgG 항체들이 세포 표면에 발현된 BCMA에 결합하는 정도를 FACS으로 분석하였다.
BCMA가 발현되는 것으로 알려진 다발성 골수종 암세포인 H929(ATCC, CRL-9068TM) 및 OPM-2 세포주를 준비하였다. 비교군으로 BCMA 발현을 하지 않는 Raji(B-lymphocyte cancer cell line)(ATCC, CCL-86TM) 세포주를 사용하였다.
상기 준비된 세포에 실시예 1.3에서 정제된 7종(B58, 5A6, 5D5, 5B5, 1H, 2G 및 5G)의 IgG 항체 10 ㎍/㎖를 가하고, 4℃에서 1시간 동안 인큐베이션 한 후, PBS 완충액으로 2회 세척하였다. 항-인간 Fc FITC를 1:400으로 희석하여 다시, 4℃에서 1시간 동안 인큐베이션 한 후 PBS 완충액으로 희석하는 과정을 반복하였다. FACSCalibur(BD Bioscience) 기기를 이용하여 세포의 형광 강도를 측정하여 도 3에 나타내었다(MFI: 평균 형광 강도).
도 1b에 나타난 바와 같이, B58 항-BCMA 항체 및 5D5 항-BCMA 항체 모두 세포 표면에 발현된 BCMA에 특이적으로 결합하고, BCMA가 발현되지 않는 세포에는 결합하지 않음을 확인하였다. 따라서, B58 항-BCMA 항체 및 5D5 항-BCMA 항체는 재조합 BCMA 단백질 뿐만 아니라 세포 표면에 발현된 BCMA의 세포외 도메인에 특이적인 결합능이 있고, 이 항-BCMA 항체를 이용하여 BCMA 발현 암세포주를 선택적으로 타겟팅 할 수 있음을 확인하였다.
6. 항-BCMA IgG 항체의 BCMA에 대한 친화도 분석
선별된 2종의 항-BCMA 항체의 인간 BCMA에 대한 친화도를 분석하였다.
Biosensor 트레이 케이스에 96 웰 블랙 마이크로플레이트를 장착하고 8개 웰 각각에 1XKB 200 ㎕를 넣은 후, Ni-NTA biosensor(Fortebio) 8개를 꽂아서 Hydration을 10분간 수행하였다. 항원 고정을 위하여, 5 ㎍/㎖의 재조합 인간 BCMA-His(Sino Biological Inc.)을 1X KB를 사용하여 희석하였다. 고정화는 역치 0.5 내지 1.0 nm로 고정하여 실험하였다. Octet Data Acquisition 9.0 소프트웨어를 활성화하여 Octet 프로그램 템플레이트를 작성하였다. 첫 번째 단계는 Baseline 1로 두었다. 두 번째 단계는 적재 단계(loading)이며 역치를 0.5 내지 1.0 nm로 고정하였다. 세번째 단계는 Baseline 2로 두었다. 다음으로 5분 동안 결합(Association) 및 20분 동안 해리(Dissociation)를 수행하였다. 플레이트의 온도는 30℃로 고정하였다. Octet 프로그램 템플레이트에 맞추어 새로운 96 웰 블랙 마이크로플레이트에 준비된 완충액을 순서에 맞게 넣었다. Baseline1로 사용되는 1XKB, 200 ㎕를 넣었다. 적재할 항원으로서 재조합 인간 BCMA-Fc/His을 5 ㎍/㎖로 희석하여 200 ㎕ 넣었다. Baseline2로 사용되는 1X KB를 200 ㎕ 넣었다. 항원과 반응시킬 항체를 200 ㎕씩 넣고, 기기를 작동시켰다. 실험이 종료 후 Octet Analysis 9.0 소프트웨어로 분석하여, 각 항체에 대한 흡착율 상수(association constant: kon), 분리율 상수(dissociation constant: kdis) 및 평형 분리 상수(equilibrium dissociation constant: KD)를 산출하였다. 산출된 값을 표 6에 나타내었다.

표 6에 나타난 바와 같이, B58 항체의 친화도는 약 10-11의 KD 값을 갖고, 5D5 항체가 약 10-10의 KD 값을 가졌다. 따라서, B58 항-BCMA 항체 및 5D5 항-BCMA 항체는 인간 BCMA 단백질에 대해 높은 결합력을 갖는 것을 확인하였다.
7. 항-BCMA IgG의 BCMA에 대한 특이도 및 에피토프 확인
(1) 항-BCMA IgG의 BCMA에 대한 특이도
BCMA는 B 세포의 성숙화 과정에 관여한다고 알려져 있고, 이 성숙화 과정에 TACI 및 BAFF-수용체가 관여하는 것으로 알려져 있다. 선별된 항체가 BCMA 관련 단백질에 결합되는지 여부를 ELISA 기법을 통해 분석하였다.
구체적으로, 인간 BCMA-Fc(R&D system,193-BC-050), TACI-Fc(R&D system, 174-TC), 및 BAFF-수용체(R&D system, 1162-BR)를 PBS 버퍼를 이용하여 희석한 후, ELISA 플레이트에 웰 당 100 ng씩 코팅하였다. 1차 항체로서 선별된 항-BCMA IgG 항체 및 2차 항체로서 항-인간 Fab HRP(1:20000 희석)을 사용하고, 실시예 1.2에 기재된 바와 같이 ELISA 분석법을 수행하였다. 비교군으로서 항-BCMA 모노클론 항체 J6M0(GSK)를 사용하였다. 마이크로플레이트 리더에서 측정된 450 nm에서의 흡광도를 도 2a에 나타내었다.
도 2a에 나타난 바와 같이, B58 항체 및 5D5 항체는 TACI 및 BAFF-수용체에는 결합하지 않으나, BCMA에만 결합하였다. 따라서, B58 항체 및 5D5 항체는 BCMA에 특이적으로 결합함을 확인하였다.
(2) 항-BCMA IgG의 에피토프 확인
제작된 B58 항체의 BCMA 에피토프를 확인하기 위헤 에피토프 맵핑을 수행하였다.
구체적으로, BCMA 4-48번 잔기를 GST에 융합시키고 BCMA A20D 점 돌연변이, BCMA I22K 점 돌연변이, 및 BCMA N31 결실 돌연변이를 제작하였다. 제작된 BCMA 돌연변이를 비환원 조건 하에서 전기영동하고, B58 항체를 1차 항체로 사용하여 면역블로팅을 수행하였다. 면역블로팅 이미지를 도 2b에 나타내었다.
도 2b에 나타난 바와 같이, BCMA I22K 점 돌연변이에서만 밴드가 나타나지 않았다. 따라서, BCMA의 22번째 아미노산인 이소류신이 B58 항체의 BCMA에 대한 에피토프인 것으로 추정되었다.
실시예 2. 항-BCMA 항체-약물 접합체(ADC)의 준비 및 ADC의 효과
1. ADC의 제조 및 확인
(1) 항체의 N-말단에 약물을 접합시킨 ADC의 제조
항체의 N-말단에 아민 결합을 통해 약물로서 모노메틸 아우리스타틴 F(Monomethyl auristatin F: MMAF)을 결합시켰다.
구체적으로, 실시예 1.3에서 준비된 B58 항체 및 5D5 항체를 한외여과/투석여과(Ultrafiltration/Diafiltration: UF/DF)를 통해 반응버퍼 A(100 mM 인산칼륨, pH 5.49)로 교체하고 8 ㎎/㎖ 이상의 농도로 농축하였다. 알데히드-MMAF(Xcessbio Bioscience)는 디메틸술폭시드(dimethyl sulfoxide: DMSO)에 녹여 5 mg/ml의 농도로 준비하였다. 준비된 항체, 알데히드-MMAF, 1M 소듐 시아노보로하이드리드(sodium cyanoborohyride: SCB)를 섞어 반응을 시켰으며 최종 반응 조건은 항체:MMAF=1:12 (몰비), 5 ㎎/㎖ 항체, 20 mM SCB, 및 14%(v/v) DMSO였다. 이 반응액을 4℃에서 16시간 동안 반응 시켰다. 반응물은 PBS로 평형화한 Hiprep Desalting 컬럼(GE Healthcare)으로 탈염하여 남아있는 알데히드-MMAF를 제거하여 반응을 종결시켰다.
제조된 항체는 '항체-N-MMAF'로 명명하였다.
ADC의 약물-항체의 비(drug-to-antibody ratio: DAR)는 액체 크로마토그래피-질량분석(LC/MS) 방법으로 결정하였다. 1 ㎎/㎖의 ADC(PBS 중) 항체 100 ㎍ 당 1 유닛의 PNGaseF(NEB)를 가하고 37℃에서 15 시간 동안 인큐베이션하여 N-글리칸을 제거하였다. Waters UPLC I-class 장비와 Waters Synapt G2-S로 구성된 LC/MS 장비에 Acquity UPLC BEH200 SEC 1.7 ㎛(4.6*150 mm) 컬럼을 장착한 후 30%(v/v) 아세토니트릴, 0.1%(v/v) 포름산, 및 0.05% 트리플루오로아세트산(Trifluoroacetic acid: TFA)의 이동상으로 평형화시켰다. 여기에 N-글리칸이 제거된 시료 5 ㎍을 로딩하여 LC/MS를 수행하였다. LC/MS 결과로 얻은 분자량 분포로부터 각 화학종의 상대함량을 가중평균하여 DAR를 결정하였다. 제조된 ADC의 DAR은 3.16으로, 항체 당 약 3.2개의 약물 분자가 결합한 것으로 나타났다.
제조된 ADC의 순도는 크기 배제-고성능 액체크로마토그래피(SE-HPLC)로 측정하였다. Agilent 1200 시리즈 HPLC 기기에서 Tosoh TSKgel G3000SWxl 컬럼(Tosoh bioscience)을 사용하여 측정하였고, 각 시료가 용출되는 위치와 곡선하면적(area under the curve: AUC)을 비교하여 각 시료의 순도를 결정하였다. ADC의 주요 피크의 순도는 99.5%였고, 전체 공정 수율(항체 기준)은 92.5%임을 확인하였다. 또한, 1x PBS(pH 7.4) 완충액 중 52.4 ㎎/㎖ ADC를 용해시키고, 3회 냉해동 시험 시, 주요 피크의 순도가 약 0.5% 감소하였다.
메탄올 침전법을 이용한 C18 컬럼 분석에서 유리 약물(free drug)은 약 3.2%이었고, 내독소(endotoxin)는 1.0 EU/㎎ 미만임을 확인하였다.
한편 MMAF 외에, 모노메틸 아우리스타틴 E(Monomethyl auristatin E: MMAE) 및 피롤로벤조디아제핀(pyrrolobenzodiazepine: PBD)을 사용하여 ADC를 제조하였다. 제조된 항체는 '항체-X-MMAE' 및 '항체-X-PBD'로 명명하였다.
(2) 항체의 티올기에 약물을 접합시킨 ADC의 제조
항체의 시스테인 잔기에 이황화 결합을 통해 MMAF를 결합시켰다.
구체적으로, B58 항체 및 5D5 항체의 버퍼를 티올 반응 버퍼 B(250 mM 붕산나트륨(pH 8.0), 125 mM NaCl, 7.5 mM EDTA)에 10배 희석한 후, 11 ㎎/㎖ 이상으로 농축하였다. 준비된 항체에 1 mM 트리스(2-카르복시에틸)포스핀(tris(2-carboxyethyl)phosphine: TCEP)를 항체:TCEP 몰비가 1:2.75가 되도록 가하고, 37℃에서 2시간 동안 반응시켜 항체 내의 이황화 결합을 환원시켰다. 이러한 티올기를 갖는 항체는 말레이미드, 할로겐알칸, 디술피드 또 티올 화합물과 반응할 수 있다.
그 후 반응물을 얼음으로 옮겨 냉각시킨 후 0.6배 부피의 반응 버퍼 C(100mM 인산 나트륨(pH 6.5), 10 mM NaCl, 및 1.5 mM EDTA)를 가하였다. 아세토니트릴에 5 mg/ml 농도로 녹아있는 말레이미도 카프로일(maleimido caproyl: mc)-MMAF(Xcessbio Bioscience)를 몰비 1:8이 되도록 가하되, 아세토니트릴의 최종 농도가 20%(v/v)가 되도록 100% 아세토니트릴을 먼저 추가해 주었다. 이후 30분간 얼음위에 방치하여 반응을 시킨 후 50 mM 시스테인을 최종 농도 5 mM이 되게 가하여 반응을 중단시켰다. 반응물은 PBS로 평형화한 Hiprep Desalting 컬럼(GE Healthcare)으로 탈염하여 반응을 종결시키고 미반응물을 제거하였다.
제조된 항체-약물 접합체는 '항체-C-MMAF'로 명명하였다.
중성(Native) LC/MS 조건에서 DAR이 3.87로, 항체 당 약 4개의 약물 분자가 결합한 것으로 나타났다.
SE-HPLC 분석에서 주요 순도가 99.3%였고, 전체 공정 수율(항체기준)은 80.0%임을 확인하였다. 또한, 1x PBS(pH 7.4) 완충액 중 50.8 ㎎/㎖ ADC를 용해시키고, 3회 냉해동 시험 시, 주요 피크의 순도가 약 0.1% 감소하였다.
메탄올 침전법을 이용한 C18 컬럼 분석에서 유리 약물은 검출되지 않았고, 내독소는 1.0 EU/㎎ 미만임을 확인하였다.
2. ADC의 항원에 대한 결합력
제조된 ADC가 항원에 대한 결합력을 갖는지 확인하였다.
실시예 2.1(1) 및 2.1(2)에 기재된 바와 같이 제조된 ADC와 B58 항체를 사용하여, 실시예 1.2에 기재된 바와 같이 ELISA를 수행하였다. 처리한 항체의 농도에 따른 450 nm에서 흡광도를 도 3a 및 도 3b에 나타내고, 산출된 50% 효과 농도(Half maximal effective concentration: EC50)(nM)을 표 7 및 표 8에 나타내었다. 도 3a는 B58 항체, B58-N-MMAF ADC, 및 B58-C-MMAF ADC의 인간 BCMA에 대한 결합력을 나타낸 그래프이고, 도 3b는 5D5 항체 및 5D5 N-MMAF ADC의 인간 BCMA에 대한 결합력을 나타낸 그래프이다.

도 3a 및 도 3b에 나타난 바와 같이, ADC 제조 후에도 항원인 인간 BCMA에 대한 결합력은 유지되는 것을 확인하였다.
3. ADC의 시험관 내 세포독성 평가
제조된 ADC의 세포 사멸능력을 평가하기 위해, 세포에 ADC를 가하고 세포의 대사활성을 측정하는 방법으로 세포의 사멸 능력을 비교하였다.
구체적으로, RPMI(ATCC), 10% FBS(Gibco), 0.05 mM 베타-메르캅토에탄올, 및 Antibiotic-Antimycotic(Gibco)를 함유한 배지에서 배양한 H929 세포주(ATCC, CRL-9068TM) 또는 Raji 세포주(ATCC, CCL-86TM)를 20,000개씩 96 웰의 각 웰에 50 ㎕ 부피로 분주한 후 같은 배양 배지를 사용하여 여러 농도로 희석한 ADC를 50 ㎕씩 분주하였다. ADC의 농도는 연속희석을 통해서 200 nM에서 5 pM이었다. 그 후 세포를 37℃, 5% CO2에서 약 6일간 배양하였다. 배양이 종료된 후 10 ㎕의 WST-8(Dojindo)을 각 웰에 가하고 8시간 동안 추가 배양하였다. SpectraMax 플레이트 리더를 사용하여 450 nm 파장에서 흡광도를 측정하였다. 흡광도와 ADC 농도간 반응곡선은 4PL 커브 피팅을 수행하여 50% 세포 사멸 농도인 IC50 값(nM)을 산출하였다.
처리한 항체 혹은 ADC의 농도에 따른 450 nm에서 흡광도를 도 4a 내지 도 4d에 나타내고, 산출된 IC50 값(nM)을 표 9에 나타내었다. 도 4a 내지 도 4c는 B58 항체, B58-N-MMAF ADC, 5D5 항체, 또는 5D5 N-MMAF ADC의 H929 세포에 대한 세포독성을 나타낸 그래프이고, 도 4d는 B58-N-MMAF ADC 및 5D5 N-MMAF ADC의 Raji 세포에 대한 세포독성을 나타낸 그래프이다.

도 4a 내지 도 4d 및 표 9에 나타난 바와 같이, BCMA를 발현하는 H929 세포에 대해 B58-N-MMAF ADC는 약 0.75 nM의 IC50이었고, 5D5 N-MMAF ADC는 약 9.95 nM의 IC50이었다. 그러나, BCMA를 발현하지 않는 Raji 세포에 대해서는 시험내 최고 농도에서도 세포가 사멸되지 않았다. 또한, 약물이 접합되지 않은 B58 항체 및 5D5 항체는 동일 실험 조건에서 세포사멸을 유도하지 않았다. 따라서, ADC에 의한 세포 사멸 효과는 BCMA와 이에 결합하는 ADC에 의한 것임을 확인하였다.한편, B58-N-MMAF ADC 외에 B58-X-MMAE ADC 및 B58-X-PBD ADC에 대해서도 동일한 방법으로 세포독성을 측정하고, 그 결과를 도 4e에 나타내었다. 도 4e에 나타난 바와 같이, MMAF 뿐만 아니라 MMAE 및 PBD를 항체에 접합시킨 경우에도 세포사멸 효과가 있음을 확인하였다.
4. ADC의 세포자살 유도 능력 평가
제조된 ADC가 세포자살(apoptosis)를 유도하는 효과가 있는지 여부를 확인하였다.
배양된 H929 세포에 1 ㎍/㎖의 농도로 B58 항체, B58-N-MMAF ADC, 또는 B58-C-MMAF ADC를 가한 후 3일간 배양하였다. 이후 세포들을 차가운 PBS로 2회 세척 후 100 ㎕의 부피 중 100,000개의 세포를 준비하였다. 준비된 세포에 FITC-아넥신(Annexin) V Apotosis Dectection Kit I (BD bioscience, Cat# 556547)에 포함된 5 ㎕의 FITC-아넥신 V와 5 ㎕의 프로피디움 아이오디드(Propidium Iodide: PI)를 가하였다. 빛을 차단한 상태에서 상온에서 15분간 반응 시킨 후 키트에 포함된 400 ㎕의 결합 완충액을 추가하고, BD FACScalibur 모델로 1시간 이내에 분석하였다. 아넥신 V에 와 PI의 형광강도 과 그 빈도를 도 5에 나타내었다.
도 5에 나타난 바와 같이, B58 항체는 형광 강도가 증가하지 않으나, B58-N-MMAF 및 B58-C-MMAF는 아넥신 V에 표지된 형광과 및 PI의 형광 강도 및 빈도 증가하여 세포막의 위상변화와 세포막 결손이 관찰되었다. 따라서, B58-N-MMAF 및 B58-C-MMAF는 세포자살에 의해 세포사멸을 유도하는 것으로 확인되었다.
5. 마우스 이종이식 모델에서 ADC의 생체 내 효능 평가
생체 내에서 ADC의 효능을 평가하기 위해서 마우스 이종이식(xenograft) 모델을 수립하고 ADC를 정맥투여하여 종양억제능을 확인하였다.
구체적으로, 7주령의 암컷 Fox chase SCID 마우스(CB17/Icr-Prkdcscid/IcrIcoCrl 마우스)에 1x107 개의 H929 세포를 동일 부피의 Matrigel(Corning)과 섞어 옆구리 부분 피하에 투여하였다. 종양의 평균 크기가 약 200 ㎣가 되면 순차 배정 방식으로 군 분리를 수행하였다(각 군에서, n=8).
각 군별로 4 mpk(milligrams per kilograms) 또는 2 mpk의 약물을 하기와 같이 꼬리정맥을 통해 투여하였고, 음성 대조군으로 PBS를 투여하였다. 반복 투여는 제1일, 제4일, 제7일, 및 제10일의 총 4회에 걸쳐 투여하였으며 단회 투여 시험에서는 제1일에 1회만 투여하였다.

버니어 캘리퍼스를 이용하여 종양의 장축, 단축의 길이를 측정한 후 하기 식을 이용하여 최종 종양 크기를 계산하였다.종양 크기(mm3)=(0.5)x(장축)x(단축)2
종양 크기는 약물 투여 후 7주간 관찰하였고, 반복 투여 및 단회투여 시험에 따른 종양의 크기를 각각 도 6a 및 도 6b에 나타내었다(화살표: 약물 투여일).
도 6a에 나타난 바와 같이, 4 mpk B58-N-MMAF, 4 mpk B58-C-MMAF, 또는 2 mpk B58-N-MMAF의 반복 투여시 종양이 모두 소실되었다. 또한, 도 6b에 나타난 바와 같이, 5 mpk B58-N-MMAF 또는 5 mpk 5D5-N-MMAF의 단회 투여시에도 종양이 모두 소실되었다. 따라서, B58-N-MMAF, B58-C-MMAF, 및 5D5-N-MMAF는 종양 성장 억제 효과가 있음을 확인하였다.
6. ADC의 생체 내 안정성의 확인
ADC의 생체내 안정성을 평가하기 위해, 래트에서 약물동태학 시험을 수행하였다.
구체적으로, 7주령의 암컷 SD 래트에 4 mpk B58 항체 또는 4 mpk B58-N-MMAF ADC를 꼬리정맥으로 투여하고, 목정맥을 통해 채혈하여 시료를 확보하였다. 투여 후 0.5 hr, 1 hr, 3 hr, 6 hr, 18hr, 24 hr, 제2일, 제3일, 제7일, 제14일, 제21일, 제28일 및 제35일에 채혈하였다. 혈액에서 혈장을 분리한 후 다음 ELISA 방법으로 혈장내 항체 또는 ADC의 농도를 결정하였다.
항체 및 총 ADC: 접합된 약물인 MMAF의 DAR에 상관 없이, 혈중에 남아있는 ADC를 항원-결합력 기반의 ELISA 방법으로 그 농도를 측정하였다. 이를 위해 ELISA 플레이트에 PBS 완충액으로 희석한 인간 BCMA-Fc(R&D system, 193-BC-050)를 50 ng/웰의 양으로 코팅하였다. 실시예 1.2에 기재된 바와 같이, ELISA를 수행하고, 450 nm에서의 흡광도를 측정하였다.
접합된 ADC: 혈중 남아있는 ADC 중 MMAF가 접합된 접합 항체만의 농도를 선택적으로 측정하였다. 토끼에 MMAF를 면역하여 얻은 항-MMAF 토끼 IgG(영인프런티어)를 100 ng/웰의 양으로 코팅하였다. 이후 시료를 래트 혈청이 10% 함유된 PBSB로 연속희석하였다. 이 시료를 100 ㎕씩 웰에 넣은 후 37℃에서 1시간 반응시켰다. 상등액을 제거한 후, PBS 완충액으로 5회 세척하였다. 20 ng/㎖ 농도의 비오틴화된-BCMA(ACRO Bioscience, BC7-H82F0)를 웰 당 100 ㎕씩 가한 후 37℃에서 1시간 반응시켰다. 상등액을 제거한 후, PBS 완충액으로 5회 세척하였다. 마지막으로 100 ng/㎖ 농도의 스트렙타비딘-HRP를 100 ㎕씩 분주하고, 다시 37℃ 1시간 반응 후 세척을 반복하였다. TMB(Tetramethylbenzidine, Sigma, T0440)를 사용하여 발색시킨 후 0.5 mol/L의 황산을 가하여 반응을 중지시켰다. SpectraMax 마이크로 플레이트 리더기(Molecular device)를 사용하여 450 nm에서 흡광도를 측정하였다.
약물 투여 후 시간에 따른 혈장 중 남아있는 항체, 총 ADC, 및 약물이 접합된 ADC의 농도를 도 7에 나타내었다.
도 7의 시간에 따른 농도 변화 그래프를 WinNolin 소프트웨어(Certara)로 분석하여, 총 항체 및 접합된 항체의 혈중 반감기 등 약물동력학적 지표를 산출하고, 그 결과는 표 11에 기재하였다.

도 7 및 표 11에 나타난 바와 같이, 약물이 접합되지 않은 B58 항체는 최소 약 10일의 긴 반감기를 가졌다. B58-N-MMAF ADC는 약물인 MMAF의 유무에 상관없이 평가했을 경우(총 ADC)에는 최소 7일을 반감기를 가졌고, 약물이 부착되어 있는 물질만 특이적으로 검출했을 경우에는 6일 정도의 반감기를 가졌다. ADC로 제작했을 경우 급격한 반감기의 감소가 보이지 않고 일반적인 항체들과 유사한 반감기를 보였다. 따라서, B58 항체 기반의 ADC는 생체 내에서 안정함을 확인하였다.
7. ADC의 설치류 단회 투여 독성시험
ADC의 생체내 안전성을 평가하기 위해, 래트에 ADC를 단회 투여한 후 독성의 양태를 관찰하였다.
구체적으로, 7주령의 암컷 SD 래트에 대해서 25 mpk 또는 50 mpk의 B58-C-MMAF, 또는 31.25 mpk 혹은 62.5 mpk의 B58-N-MMAF를 단회 투여하고 3주간 체중을 측정하여 독성의 발현 여부를 관찰하였다. 한편 약물 투여전 제3일, 투여후 제4일, 제11일, 및 부검 당일(제22일)에 채혈을 하여 AST(aspartate aminotransferase) 및 ALT(alanine aminotransaminase)의 수치를 측정하였다. 음성 대조군으로 PBS를 사용하였다. 약물 투여 후 체중 변화, 혈중 AST 농도(IU/L), 및 혈중 ALT 농도(IU/L)를 각각 도 8a 내지 도 8c에 나타내었다.
도 8a 내지 도 8c에 나타난 바와 같이, 25 mpk 또는 50 mpk 용량을 단회 투여한 결과 체중 감소는 관찰되지 않았다. 또한, 간 효소인 AST 및 ALT의 값이 증가하였으나, 제22일차에 감소하는 경향을 보여 회복의 양태를 보였다. 따라서, 투여 용량까지 ADC에 의한 회복불가능한 독성은 나타나지 않는 것으로 확인되었다.
8. ADC와 항암제와의 병용 투여
ADC와 항암제를 병용 투여한 경우 상승작용이 있는지 여부를 확인하기 위해, H929 이종이식 마우스 모델에서 ADC와 보르테조밉을 병용투여하였다.
구체적으로, 7주령의 암컷 Fox chase SCID 마우스에 1x107 개의 H929 세포를 동일부피의 Matrigel(Corning)과 섞어 옆구리 부분 피하에 투여하였다. 종양의 평균 크기가 약 200 ㎣가 되면 순차 배정 방식으로 군 분리를 수행하였다(각 군에서, n=8).
각 군별로 ADC(개체 당 100 ㎕)와 보르테조밉(bortezomib, VELCADE®Takeda Oncology)을 표 11과 같이 꼬리정맥을 통해 투여하였고, 음성 대조군으로 PBS를 투여하였다. ADC는 제1일에 1회 단회 투여하였고, 보르테조밉은 주 2회씩 총 6회 투여하였다.

도 9에 나타난 바와 같이, B58-N-MMAF ADC 및 5D5-N-MMAF ADC는 모두 보르테조밉에 비해 종양의 크기가 유의하게 감소하여, B58-N-MMAF ADC 및 5D5-N-MMAF ADC의 항종양 효과가 우수한 것으로 확인되었다. 한편, 병용 투여시에도 체중감소와 같은 독성이 관찰되지 않아 ADC와 보르테조밉은 병용투여가 가능함을 확인하였다. 이는 보르테조밉에 대한 반응성이 떨어지기 시작하는 환자에게 ADC를 추가로 투여할 수 있을 것이라는 가능성을 보여주고 있다.
9. 다발성 골수종 동물모델에서 ADC의 효력 평가
5주령의 암컷 NOG 마우스에 마우스에 1 Gy의 감마선을 조사하고 1x107 개의 U266 다발성 골수종 암세포주를 꼬리 정맥을 통해 투여하였다. 이후 1주일 간격으로 혈액내 M-단백질의 농도를 평가한 후 평균 농도가 최초로 10,000 ng/㎖ 이상이 되었을 때 순차 배정 방식으로 7군으로 나누어 군 분리를 수행하였으며 이때 혈중 M-단백농도의 평균은 12,800 ng/㎖ 이었다(비유도군(sham)에서 n=3, 나머지 각 군에서 n=7, 총 n=45).
각 군별로 ADC(개체 당 100 ㎕) 또는 보르테조밉(bortezomib, VELCADE®Takeda Oncology)을 표 13과 같이 꼬리정맥을 통해 투여하였다. 비유도군(sham)은 물질 투여를 하지 않았고, 음성 대조군은 자외선은 조사하였으나, 약물 대신 PBS를 투여하였다. ADC는 제1일에 1회 단회 투여하였고, 보르테조밉은 주 2회씩 총 6회 투여하였다.
약물의 효과는 다음과 같이 평가하였다. 주 2회 보행 등 운동능력을 평가하여 질환의 이행을 관찰하였다. 주 1회 채혈을 하여 혈중 M-단백질을 평가하여 종양 세포의 성장 저해 여부를 관찰하였다. 최종적으로 약물 투여 3주 후 마우스를 희생하여 넙적다리뼈를 수득하여 골수와 조직 분석을 수행하였고 정강이뼈를 수득하여 마이크로-CT로 뼈조직의 특징을 분석하였다.

마우스의 행동 평가로서 뒷다리 마비 시험을 수행하였다. 정상인 경우 0점, 보행 장애(전조증상)이 있는 경우 0.5점, 한쪽 뒷다리가 마비된 경우 1점, 및 양쪽 뒷다리가 마비된 경우 2점을 기준으로 행동 평가 점수를 매겼다. 뒷다리 마비 시험 점수의 평균값을 표 14에 나타내었다.

또한, 혈중 M-단백질의 양을 ELISA로 측정하고, 그 결과를 도 10a에 나타내었다. 도 10a에 나타난 바와 같이, 혈중 M-단백질의 농도가 유의하게 감소하여 다발성 골수종 세포가 투여 약물에 의해 감소하였음을 알 수 있다.종양 발발 조직인 골수내에서의 다발성 골수종 세포의 증식 혹은 억제 정도를 정량적으로 분석하기 위해 골수를 채취하고 골수종 세포의 바이오마커인 CD138이 발현되는 세포의 비율을 FACS로 확인하고, 그 결과를 표 15 및 도 10b에 나타내었다.

마우스 넙적다리 뼈를 해모토실린(haemotoxylin)과 에오신(eosin)으로 염색하여 조직검사를 수행하고, 그 염색 이미지를 도 10c에 나타내었다. 조직염색을 통해 골조직 손상 정도와 골수종 세포의 침투 정도를 볼 수 있었다. 음성 대조군의 경우 대조그룹에 비해 대퇴골에 전체적으로 골수종세포가 침윤되어 있었으며 골수종 세포 침윤 부위에서 뼈잔기둥(bone trabeculae)의 뚜렷한 감소가 관찰되었다. 5D5-N-MMAF 2 mpk를 투여한 저용량 그룹의 일부 개체에서 뼈끝부위(epiphysis)의 골수 및 뼈잔기둥은 감소가 없었지만(intact), 뼈몸통끝부위(metaphysis)에서 상당 부분 골수종세포의 침윤과 함께 뼈잔기둥 감소가 관찰되었다. 그러나 나머지 B58-N-MMAF 2 mpk 또는 4 mpk 투여군과 5D5-N-MMAF 2 mpk 또는 4 mpk 투여군에서는 현미경적으로 인정할 만한 골수종세포의 침윤이나 뼈잔기둥의 조직학적 변화는 관찰할 수 없었다.마이크로 CT를 통한 뼈의 해부학적 변화에 대한 정량적 평가 결과는 도 10d에 표시하였다. 도 10d에 나타난 바와 같이, 골수강 단면적, 치밀골 밀도 면에서 ADC 투여군[B_2mg: B58-N-MMAF (2 mg/kg), B_4mg: B58-N-MMAF (4 mg/kg), D_2mg: 5D5-N-MMAF (2 mg/kg), D_4mg: 5D5-N-MMAF (4 mg/kg)]은 비처치군(담체, vehicle)에 비해서 비유발군(음성 대조군, control)과 유사한 결과를 보여주고 있다. 이는 골수종 질환 모델에서 투여한 ADC에 의해서 골수종 세포의 증식이 억제되었음을 보여주고 있다.
10. 원숭이 예비독성 시험
원숭이에 대하여 ADC의 독성 시험을 수행하였다.
수컷 게잡이 원숭이 6마리를 두 그룹으로 나누고(군당 3마리), 음성 대조군 없이 B58-N-MMAF 또는 5D5-N-MMAF을 투여하였다. 각 개체에 대해 3 mg/kg의 용량으로 시작시 1회 투여하고, 제4주 후 10 mg/kg의 용량으로 1회 투여하고, 제8주 후 30 mg/kg의 용량으로 1회 투여하였다. 매 물질 투여 후 3일, 14일, 21일차에 채혈하여 혈액검사와 혈액생화학검사를 수행하였으며 매 물질 투여 직전 3일, 투여 후 7일차에 네 가지 항목에 대해 조사하여 안구 독성 발현 여부를 평가하였다(도 11a).
원숭이의 상대적 체중 변화를 도 11b에 나타내었다(1M: B58-N-MMAF 투여, 2M: 5D5-N-MMAF 투여, 01 내지 03: 개체 일련번호). 도 11b에 나타난 바와 같이, ADC 투여에 의한 급격한 체중 변화는 관찰되지 않았다.
혈액학적 변화를 확인하기 위해, 혈중 백혈구(WBC)의 수, 혈중 혈소판의 수, 혈장 중 ALT 농도(IU/L), 및 혈장 중 AST 농도(IU/L)를 측정하고, 그 결과를 도 11c에 나타내었다. 도 11c에 나타난 바와 같이, 혈중 백혈구의 수나 혈소판의 수는 큰 변화가 없었고, 고용량의 ADC를 투여하였음에도 혈장 중 ALT 농도 및 AST의 농도는 낮게 유지되었다. 따라서, B58-N-MMAF ADC 및 5D5-N-MMAF ADC는 영장류에서도 독성이 크게 나타나지 않는 것으로 확인하였다. 이는 아마니틴(amanitin) 접합체 ADC인 HDP-101이 원숭이에서 3 mpk 투여시 급성 반응으로 ALT가 400 IU/L 이상, AST가 250 IU/L 이상 오르는 것에 비해 독성이 낮은 것이다.
11. 5D5 클론 돌연변이의 결합능 분석
실시예 1.4에서 제조한 5D5 돌연변이 항체의 결합능을 분석하였다. 먼저 항원에 대한 결합능을 ELISA 방법으로 분석하여 비교하고, 그 결과를 도 12a에 나타내었다. BCMA 과발현 세포주인 H929에 대한 결합 정도를 유세포 계수기를 사용하여 분석하고 이를 도 12b에 나타내었다.
도 12a에 나타난 바와 같이, 5D5 LM4 돌연변이의 항원 결합능은 ELISA EC50 값으로 약 0.237 nM로 산출되었고, 5D5 WT 항체의 EC50 값은 약 0.16 nM로 산출되었다. 따라서, 5D5 LM4 돌연변이의 항원 결합능은 5D5 WT에 비하여 크게 떨어지지 않는 것으로 확인되었다.
또한, 도 12b에 나타난 바와 같이, 세포 표면 항원에 대한 결합에 있어서는 5D5 LM4 돌연변이의 결합 신호 크기는 약 11.9이었고, 5D5 WT 항체의 결합 신호 크기는 약 5.56이었으므로, 5D5 LM4 돌연변이는 5D5 WT에 비해 세포 표면 항원에 대한 결합력이 우수한 것으로 확인되었다.
12. 5D5 클론 돌연변이 ADC의 세포독성 평가
결합능이 개선된 5D5 클론 돌연변이의 세포독성을 평가하기 위하여, 실시예 2.1에 기재된 바와 같이 ADC를 제조하고, 실시예 2.3에 기재된 바와 같이 H929 세포에 대한 세포독성을 평가하였다.
ADC의 농도에 따른 450 nm에서 흡광도를 도 13에 나타내고, 이로부터 산출된 IC50 값(nM)을 표 16에 나타내었다.

도 13 및 표 16에 나타난 바와 같이, 5D5 LM4 돌연변이 기반의 ADC가 5D5 WT 기반의 ADC에 비해서 유의한 세포독성을 유도하였다.
13. 5D5 돌연변이 ADC의 약리 효과
5D5 돌연변이 ADC가 어떤 기작으로 세포독성을 나타내는가를 연구하기 위해 세포주기 변화양상을 분석하였다.
우선, H929 세포에 시험물질들을 5 ㎍/㎖의 농도로 가하고, 2일간 인큐베이션하였다. 그 후, H929 세포에 70%(v/v) 에탄올을 가하고 -70℃에서 약 1일간 보관하여 세포를 고정시켰다. 고정된 세포를 PBS로 세척한 후 RNase A를 가하여 RNA를 제거하였다. 세포에 프로피디움 아이오디드(PI, Abcam, ab189818)를 가하여 DNA를 염색하였다. 염색된 세포는 BD Calibur 유세포분석기로 분석하고, 그 결과를 도 14a에 나타내었다.
도 14a에 나타난 바와 같이, ADC 처리군은 대조군에 비해 G1 기의 세포수가 줄어들고 S 기와 G2 기의 세포수가 증가하였다. 따라서, ADC에 의해 세포 주기에서 G2/M 정지(arrest)가 발생하고, 세포 분열과 생장이 저해됨을 확인하였다.
ADC에 의해 세포자살(apoptosis)이 유도되는지 확인하였다. 구체적으로, H929 세포에 5 ㎍/㎖ 또는 1 ㎍/㎖ 농도의 ADC를 가하고, Incucyte Caspase-3/7 green reagent(satorius, Cat# 4440)를 가하였다. 그후, 세포를 적정 조건에서 배양하면서 Incucyte S3(satorius) 장비를 이용하여 배양 시간에 따른 형광 강도를 측정하고, 그 결과를 도 14b에 나타내었다. 도 14b는 세포자살과 관련된 카스파제-3 또는 caspase-7의 활성화 수준을 나타내는 그래프로서, ADC에 의해 세포자살이 시간에 따라 얼마나 유도되는지를 보여주고 있다.
도 14b에 나타난 바와 같이, ADC 처리 후 약 16시간부터 세포사멸 현상이 두드러지기 시작했으며, 시간에 따라 증가하는 양상을 보였다. 그러나 증가 양상은 ADC 종류마다 차이가 있었고, B58-N-MMAF, 5D5(LM4)-N-MMAF, 5D5(WT)-N-MMAF 순으로 형광 신호가 빠르게 증가하였다.
14. 5D5 돌연변이 ADC의 생체 내 효능 평가
5D5 돌연변이 기반의 ADC의 생체내 효능을 평가하기 위해, 실시예 2.5에 기재된 바와 같은 방법으로 H929 이종이식 모델을 수립하였다. B58-N-MMAF("D20101"), 5D5-N-MMAF("D20108"), 및 5D5-LM4-N-MMAF("D20110")의 3종의 ADC를 3 mpk 용량으로 단회 투여하였다(각 군에서, n=8). 투여 시간에 따른 종양 크기를 도 15에 나타내고, 그 결과로부터 ADC의 종양 성장 억제능을 비교하였다.
도 15에 나타난 바와 같이, 3종의 ADC 모두 3 mpk 용량에서 종양의 성장을 효과적으로 억제하였다. 또한, B58-N-MMAF 투여군(D20101)에서는 8 마리 중 5 마리, 5D5-N-MMAF 투여군(D20108)은 8 마리 중 7 마리가, 5D5-LM4-N-MMAF 투여군(D20110)은 8 마리 모두 종양이 소실된 것을 확인하였다.
따라서, B58 항체-기반 ADC 뿐만 아니라, 5D5 항체-기반 ADC 및 5D5 항체 돌연변이-기반 ADC는 우수한 종양 억제 효능이 있음을 확인하였다.
Claims (19)
- B 세포 성숙 항원(B Cell Maturation Antigen: BCMA)에 특이적으로 결합하는 항체 또는 그의 항원 결합 단편에 세포독성 약물이 접합된 항체-약물 접합체(antibody-drug conjugate: ADC)로서, 상기 항체 또는 그의 항원 결합 단편은서열번호 9 내지 14로 이루어진 군으로부터 선택된 하나 이상의 아미노산 서열을 포함하는 중쇄 가변 영역,서열번호 15 내지 20 및 30 내지 34로 이루어진 군으로부터 선택된 하나 이상의 아미노산 서열을 포함하는 경쇄 가변 영역,또는 상기 중쇄 가변 영역 및 상기 경쇄 가변영역을 포함하는 것인 항체-약물 접합체.
- 청구항 1에 있어서, 상기 중쇄 가변 영역은서열번호 9 또는 10으로 이루어진 아미노산 서열을 포함하는 상보성 결정 영역(complementarity determining region: CDR)-H1;서열번호 11 또는 12로 이루어진 아미노산 서열을 포함하는 CDR-H2; 및서열번호 13 또는 14로 이루어진 아미노산 서열을 포함하는 CDR-H3을 포함하는 것인 항체-약물 접합체.
- 청구항 2에 있어서, 상기 중쇄 가변 영역은 서열번호 5 또는 6으로 이루어진 아미노산 서열을 포함하는 것인 항체-약물 접합체.
- 청구항 1에 있어서, 상기 경쇄 가변 영역은서열번호 15, 16, 및 30 내지 34로 이루어진 군으로부터 선택된 하나 이상의 아미노산 서열을 포함하는 CDR-L1;서열번호 17 또는 18로 이루어진 아미노산 서열을 포함하는 CDR-L2; 및서열번호 19 또는 20으로 이루어진 아미노산 서열을 포함하는 CDR-L3을 포함하는 것인 항체-약물 접합체.
- 청구항 4에 있어서, 상기 경쇄 가변 영역은 서열번호 7, 8, 및 25 내지 29로 이루어진 군으로부터 선택된 아미노산 서열을 포함하는 것인 항체-약물 접합체.
