Abstract

본 발명은 페놀산(Phenolic acid) 형태의 파이토케미컬(Phytochemical)이 프럭토올리고당 주쇄를 구성하는 일부 당에 결합된 구조를 가지는 신규 파이토케미컬-프럭토올리고당 결합체를 제공한다. 본 발명에 따른 신규 파이토케미컬-프럭토올리고당 결합체는 이에 대응되는 페놀산 파이토케미컬에 비해 구강-위장관 통과 조건에서 거의 분해되지 않고 대부분이 대장에 도달할 수 있으므로 경구 투여시 대장에서 우수한 생체이용성을 갖는다. 특히, 본 발명에 따른 페룰산-프럭토올리고당 결합체는 상업적인 대장암 치료용 약물과 비교하였을 때 대장암 세포에 대한 선택적 사멸 효과가 우수하여, 대장암을 예방, 개선 또는 치료하는데에 유용한 식의약 소재로 사용될 수 있다.

파이토케미컬-프럭토올리고당 결합체, 이의 제조방법 및 이의 용도
본 발명은 파이토케미컬-프럭토올리고당 결합체 등에 관한 것으로서, 더 상세하게는 프럭토올리고당의 주쇄를 구성하는 일부 당에 페놀산 형태의 파이토케미컬이 측쇄 결합되어 형성된 신규 물질, 이의 제조방법 및 이의 생리활성에 기반한 다양한 용도에 관한 것이다.

본 발명은 종래의 기술적 배경하에서 도출된 것으로서, 본 발명의 목적은 페놀산 형태의 파이토케미컬과 동등하거나 향상된 생리활성을 가지면서 동시에 경구 투여시 구강, 위 및 소장 환경에서 거의 분해되지 않고 대부분이 대장에 도달할 수 있는 신규 파이토케미컬-프럭토올리고당 결합체를 제공하는데에 있다.
또한, 본 발명의 목적은 신규 파이토케미컬-프럭토올리고당 결합체의 제조방법을 제공하는데에 있다.
또한, 본 발명의 목적은 신규 파이토케미컬-프럭토올리고당 결합체의 생리활성에 기반한 다양한 의약 용도, 건강기능식품 용도 또는 화장품 용도를 제공하는데에 있다.

본 발명의 신규 파이토케미컬-프럭토올리고당 결합체는 페놀산(Phenolic acid) 형태의 파이토케미컬(Phytochemical)이 프럭토올리고당 주쇄를 구성하는 일부 당에 결합된 구조를 가진다. 본 발명에 따른 신규 파이토케미컬-프럭토올리고당 결합체는 이에 대응되는 페놀산 파이토케미컬에 비해 구강-위장관 통과 조건에서 거의 분해되지 않고 대부분이 대장에 도달할 수 있으므로 경구 투여시 대장에서 우수한 생체이용성을 갖는다. 또한, 본 발명에 따른 신규 파이토케미컬-프럭토올리고당 결합체는 식품원에서 유래한 천연물질들로 구성되어 있기 때문에 인체에 안전하다. 특히, 본 발명에 따른 페룰산-프럭토올리고당 결합체는 상업적인 대장암 치료용 약물과 비교하였을 때 대장암 세포에 대한 선택적 사멸 효과가 우수하여, 대장암을 예방, 개선 또는 치료하는데에 유용한 식의약 소재로 사용될 수 있다. 또한, 본 발명에 따른 신규 파이토케미컬-프럭토올리고당 결합체는 우수한 항산화 활성을 가지기 때문에 노화 방지 등에 유용한 식의약 소재 또는 화장품 소재로 사용될 수 있다.

이하, 본 발명에서 사용한 용어를 설명한다.
본 발명에서 사용되는 용어 "약학적으로 허용가능한" 및 "식품학적으로 허용가능한"이란 생물체를 상당히 자극하지 않고 투여 활성 물질의 생물학적 활성 및 특성을 저해하지 않는 것을 의미한다.
본 발명에서 사용되는 용어 "대장암"은 맹장, 결장 또는 직장에 생기는 악성 종양을 의미한다.
본 발명에서 사용되는 용어 "예방"은 본 발명의 조성물의 투여로 특정 질환의 증상을 억제하거나 진행을 지연시키는 모든 행위를 의미한다.
본 발명에서 사용되는 용어 "치료"는 본 발명의 조성물의 투여로 특정 질환의 증상을 호전 또는 이롭게 변경시키는 모든 행위를 의미한다.
본 발명에서 사용되는 용어 "개선"은 치료되는 상태와 관련된 파라미터, 예를 들면 증상의 정도를 적어도 감소시키는 모든 행위를 의미한다.
본 발명에서 사용되는 용어 "투여"는 임의의 적절한 방법으로 개체에 소정의 본 발명의 조성물을 제공하는 것을 의미한다. 이때, 개체는 본 발명의 조성물을 투여하여 특정 질환의 증상이 호전될 수 있는 질환을 가진 인간, 원숭이, 개, 염소, 돼지 또는 쥐 등 모든 동물을 의미한다.
본 발명에서 사용되는 용어 "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜 또는 위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 이는 개체의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출비율, 치료기간, 동시에 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
이하, 본 발명을 구체적으로 설명한다.
본 발명의 일 측면은 파이토케미컬과 동등하거나 향상된 생리활성을 가지면서 동시에 경구 투여시 구강, 위 및 소장 환경에서 거의 분해되지 않고 대부분이 대장에 도달할 수 있는 신규 파이토케미컬-프럭토올리고당 결합체에 관한 것이다. 본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체는 하기의 일반 구조식으로 표시되는 프럭토올리고당(Fructooligosaccharide) 주쇄; 상기 주쇄의 말단 일 측에 존재하는 글루코스와 에스테르 결합에 의해 연결된 페놀산(Phenolic acid) 형태의 파이토케미컬(Phytochemical); 및 상기 주쇄에 존재하는 프럭토스와 에스테르 결합에 의해 선택적으로 연결된 페놀산 형태의 파이토케미컬로 이루어진다.
[프럭토올리고당 주쇄의 일반 구조식]
Glu-(Fru) j-Fru
상기 프럭토올리고당 주쇄의 일반 구조식에서 'Glu'는 글루코스를 나타내고, 'Fru'는 프럭토스를 나타내고, j는 글리코시딕 결합을 통해 연결된 프럭토스의 수로서 1 내지 59의 정수에서 선택된다. 상기 프럭토올리고당 주쇄의 일반 구조식에서 j는 프럭토올리고당의 난소화성 또는 파이토케미컬-프럭토올리고당 결합체의 제조 용이성 등을 고려할 때 4 내지 48의 정수에서 선택되는 것이 바람직하고 8 내지 40의 정수에서 선택되는 것이 바람직하다.
본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체의 일 구성성분인 페놀산 형태의 파이토케미컬은 의약 또는 건강기능식품 용도로 사용될 수 있는 생리활성을 나타내는 것이라면 그 종류가 크게 제한되지 않으며, 예를 들어 페룰산(Ferulic acid), 카페익산(Caffeic acid), 신남산(Cinnamic acid), 클로로겐산(Chlorogenic acid), 쿠마린(Coumarin), 시나픽산(Sinapinic acid), 치코르산(Cichoric acid), 다이페룰산(Diferulic acid), 쿠마르산(Coumaric acid), 살리실산(Salicylic acid), 3-하이드록시벤조산(3-Hydroxybenzoic acid), 4-하이드록시벤조산(4-Hydroxybenzoic acid), 바닐산(Vanillic acid), 갈산(Gallic acid) 또는 엘라그산(Ellagic acid)에서 선택될 수 있다.
또한, 본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체에서 파이토케미컬은 바람직하게는 주쇄의 글루코스 6번 탄소에 위치하는 하이드록실기와 파이토케미컬에 존재하는 카르복실기 사이의 에스테르 결합에 의해 주쇄에 필수적으로 연결된다. 또한, 본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체에서 파이토케미컬은 바람직하게는 주쇄의 프럭토스 6번 탄소에 위치하는 하이드록실기와 파이토케미컬에 존재하는 카르복실기 사이의 에스테르 결합에 의해 주쇄에 선택적으로 연결된다.
또한, 본 발명의 바람직한 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체는 하기의 화학식 Ⅰ또는 화학식 Ⅱ로 표시되는 화합물에서 선택된다.

또한, 본 발명의 더 바람직한 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체는 하기의 화학식 Ⅲ 또는 화학식 Ⅳ로 표시되는 페룰산-프럭토올리고당 결합체에서 선택된다. 본 발명의 더 바람직한 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체에서 페놀산 형태의 파이토케미칼은 페룰산(Ferulic acid)이다. 본 발명의 더 바람직한 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체는 페룰산(Ferulic acid)과 비교하였을 때 동등하거나 향상된 항암 활성(특히 항-대장암 활성)을 가지면서 동시에 경구 투여시 구강, 위 및 소장 환경에서 거의 분해되지 않고 대부분이 대장에 도달하기 때문에 대장에서의 생체 이용성이 우수하다.

또한, 본 발명의 가장 바람직한 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체는 하기 화학식 Ⅴ로 표시되는 페룰산-프럭토올리고당 결합체로 이루어지거나 이를 주성분으로 포함한다. 본 발명의 발명자들은 하기 화학식 Ⅴ로 표시되는 화합물을 주성분으로 포함하는 페룰산-프럭토올리고당 결합체를 'FA-FOS Ⅰ'로 명명하였고 물리적 특성을 분석한 결과 물에 대해 불용성이고 수계 환경에서 마이셀(Micelle) 구조의 구형 입자를 형성하는 것으로 나타났다. 또한, 페룰산-프럭토올리고당 결합체 FA-FOS Ⅰ은 구강, 위 및 소장 환경에서 거의 분해되지 않고 대부분 대장에 도달하며 대장 환경에서 점액(Mucus)을 통과하여 산성 조건의 암세포에 부착된 후 암세포의 사멸을 유도하는 것으로 나타났다. 특히, 페룰산-프럭토올리고당 결합체 FA-FOS Ⅰ은 전이가 가능한 대장암 세포만을 선별적으로 사멸시킬 수 있기 때문에 대장암의 전이 억제에 매우 유용하다.

또한, 본 발명의 가장 바람직한 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체는 하기 화학식 Ⅵ으로 표시되는 페룰산-프럭토올리고당 결합체로 이루어지거나 이를 주성분으로 포함한다. 본 발명의 발명자들은 하기 화학식 Ⅵ으로 표시되는 화합물을 주성분으로 포함하는 페룰산-프럭토올리고당 결합체를 'FA-FOS Ⅱ'로 명명하였고 물리적 특성을 분석할 결과 수용성이고, 구강, 위 및 소장 환경에서 거의 분해되지 않고 대부분 대장에 도달하는 것으로 확인되었다. 또한, 페룰산-프럭토올리고당 결합체 FA-FOS Ⅱ는 인간 대장 미생물총(Human microbiota)에 의해 모사된 대장 환경 조건에서 분해되어 페룰로일 글루코스(Feruloyl glucose) 형태로 방출되고 대장암의 예방, 개선 또는 치료에 매우 유용하다.

본 발명의 일 측면은 전술한 신규 파이토케미컬-프럭토올리고당 결합체의 약학적 또는 식품학적으로 허용가능한 염에 관한 것이다. 본 발명에서 신규 파이토케미컬-프럭토올리고당 결합체의 약학적 또는 식품학적으로 허용가능한 염으로는 유리산 (free acid)에 의해 형성된 산부가염이 유용하다. 산부가염은 통상의 방법, 예를 들면 화합물을 과량의 산 수용액에 용해시키고, 이 염을 메탄올, 에탄올, 아세톤 또는 아세토니트릴과 같은 수혼화성 유기 용매를 사용하여 침전시켜서 제조한다. 동 몰량의 화합물 및 물 중의 산 또는 알코올(예, 글리콜 모노메틸에테르)을 가열하고 이어서 상기 혼합물을 증발시켜서 건조시키거나, 또는 석출된 염을 흡인 여과시킬 수 있다. 이때, 유리산으로는 유기산과 무기산을 사용할 수 있으며, 무기산으로는 염산, 인산, 황산, 질산, 주석산 등을 사용할 수 있고, 유기산으로는 메탄설폰산, p-톨루엔설폰산, 아세트산, 트리플루오로아세트산, 시트르산, 말레인산 (maleic acid), 숙신산, 옥살산, 벤조산, 타르타르산, 푸마르산, 만데르산, 프로피온산 (propionic acid), 젖산 (lactic acid), 글리콜산 (glycollic acid), 글루콘산 (gluconic acid), 갈락투론산, 글루탐산, 글루타르산 (glutaric acid), 글루쿠론산 (glucuronic acid), 아스파르트산, 아스코르빈산, 카본산, 바닐릭산, 히드로 아이오딕산 등을 사용할 수 있다. 또한, 염기를 사용하여 약학적 또는 식품학적으로 허용 가능한 금속염을 제조할 수 있다. 알칼리 금속 또는 알칼리토 금속염은, 예를 들면 화합물을 과량의 알칼리 금속 수산화물 또는 알칼리토 금속 수산화물 용액 중에 용해하고, 비용해 화합물염을 여과한 후 여액을 증발, 건조시켜 얻는다. 이때, 금속염으로서는 특히 나트륨, 칼륨 또는 칼슘염을 제조하는 것이 적합하며, 또한 이에 대응하는 은염은 알칼리 금속 또는 알칼리토 금속염을 적당한 은염(예, 질산은)과 반응시켜 얻는다.
본 발명의 일 측면은 신규 파이토케미컬-프럭토올리고당 결합체의 제조방법에 관한 것이다. 본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체의 제조방법은 (a) 반응 용매에 페놀산(Phenolic acid) 형태의 파이토케미컬(Phytochemical) 및 에스테르화 반응용 촉매를 첨가 및 용해하고 가열하여 파이토케미컬의 활성화 반응을 진행하고 활성화된 형태의 파이토케미컬을 포함하는 1차 반응 혼합물을 수득하는 단계; 및 (b) 상기 1차 반응 혼합물에 하기의 일반 구조식으로 표시되는 프럭토올리고당(Fructooligosaccharide)을 첨가하고 불활성 가스 분위기하에서 가열하여 프럭토올리고당(Fructooligosaccharide)과 파이토케미컬(Phytochemical) 간의 에스테르화 반응을 진행하고 파이토케미컬-프럭토올리고당 결합체를 포함하는 2차 반응 혼합물을 수득하는 단계를 포함한다. 또한, 본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체의 제조방법은 바람직하게는 (c) 상기 2차 반응 혼합물을 냉각 및 방치하여 파이토케미컬-프럭토올리고당 결합체를 침전시키고 원심분리 및 세척을 순차적으로 진행하여 정제된 파이토케미컬-프럭토올리고당 결합체를 수득하는 단계를 더 포함할 수 있다.
[프럭토올리고당의 일반 구조식]
Glu-(Fru) k
상기 프럭토올리고당의 일반 구조식에서 'Glu'는 글루코스를 나타내고, 'Fru'는 프럭토스를 나타낸다. 또한, 상기 k는 글리코시딕 결합을 통해 연결된 프럭토스의 수로서 2 내지 60의 정수에서 선택되고, 5 내지 49의 정수에서 선택되는 것이 바람직하고 9 내지 41의 정수에서 선택되는 것이 바람직하다.
본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체의 제조방법에서 최종적으로 수득되는 파이토케미컬-프럭토올리고당 결합체의 구조적 특징은 전술한 부분을 참조하며 구체적인 설명을 생략한다.
본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체의 제조방법에서 상기 (a) 단계의 파이토케미컬 활성화 반응은 45~120℃에서 2~20 hr 동안 진행되는 것이 바람직하고, 50~80℃에서 5~18 hr 동안 진행되는 것이 더 바람직하다. 또한, 상기 (a) 단계에서 사용되는 반응 용매는 페놀산(Phenolic acid) 형태의 파이토케미컬(Phytochemical), 에스테르화 반응용 촉매 및 프럭토올리고당(Fructooligosaccharide)을 용해할 수 있는 것이라면 그 종류가 크게 제한되지 않으며, 유기합성 분야에서 사용되는 공지의 다양한 유기 용매로부터 선택될 수 있다. 예를 들어 상기 반응 용매는 극성 유기 용매에서 선택될 수 있고, 구체적인 종류로는 다이메틸설폭사이드(Dimethylsulfoxide), 다이메틸포름아마이드(Dimethylformamide), 헥사메틸포스포르아마이드(Hexamethylphosphoramide), N-메틸피롤리돈(N-methylpyrrolidone), 테트라하이드로푸란(tetrahydrofuran), 에틸아세테이트(ethyl acetate), 아세톤(acetone), 아세토니트릴(acetonitrile), 프로필렌 카보네이트(propylene carbonate) 등이 있다. 또한, 상기 (a) 단계에서 사용되는 에스테르화 반응용 촉매는 카르복실산(Carboxylic acid)과 알코올 간의 에스테르화 반응 또는 카르복실산과 하이드록실기 간의 에스테르화 반응에 사용되는 다양한 공지의 촉매에서 선택될 수 있고, 구체적인 종류로는 카보닐다이이미다졸(N,N'-carbonyldiimidazole, CDI), 다이시클로헥실카보다이이미드(Dicyclohexylcarbodiimide), 2-브로모-1-메틸피리듐아이오다이드(2-bromo-1-methylpyridinium iodide) 등이 있다.
또한, 본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체의 제조방법에서 사용된 페놀산(Phenolic acid) 형태의 파이토케미컬(Phytochemical), 에스테르화 반응용 촉매 및 프럭토올리고당의 몰 비는 1:1:1 내지 40:40:1의 범위에서 선택되는 것이 바람직하고, 2:2:1 내지 35:35:1의 범위에서 선택되는 것이 바람직하다. 예를 들어, 페룰산-프럭토올리고당 결합체 FA-FOS Ⅰ을 제조하는 경우 페룰산, 에스테르화 반응용 촉매 및 프럭토올리고당의 몰 비는 20:20:1 내지 35:35:1인 것이 바람직하다. 또한, 페룰산-프럭토올리고당 결합체 FA-FOS Ⅱ를 제조하는 경우 페룰산, 에스테르화 반응용 촉매 및 프럭토올리고당의 몰 비는 2:2:1 내지 10:10:1인 것이 바람직하다.
또한, 본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체의 제조방법에서 상기 (b) 단계의 에스테르화 반응은 70~150℃에서 2~15 hr 동안 진행되는 것이 바람직하고, 80~120℃에서에서 3~10 hr 동안 진행되는 것이 더 바람직하다.
또한, 본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체의 제조방법에서 상기 (c) 단계의 파이토케미컬-프럭토올리고당 결합체를 세척하기 위해 탄소수 2 내지 5의 저급 알코올 용매를 사용할 수 있다. 예를 들어, 상기 저급 알코올 용매로는 에탄올, 이소프로필 알코올 등이 있다.
본 발명의 일 예에 따른 신규 파이토케미컬-프럭토올리고당 결합체의 제조방법에서 페놀산(Phenolic acid) 형태의 파이토케미컬(Phytochemical)로 페룰산을 사용하고 에스테르화 반응용 촉매로 카보닐다이이미다졸(N,N'-carbonyldiimidazole, CDI)을 사용하는 경우 상기 (a) 단계는 페룰산의 활성화 단계로서 하기 화학 반응식 1과 같이 나타낼 수 있고, 상기 (b) 단계는 활성화된 페룰산을 프럭토올리고당에 그래프팅 하는 단계로서 하기 화학 반응식 2와 같이 나타낼 수 있다.

본 발명의 일 측면은 신규 파이토케미컬-프럭토올리고당 결합체의 생리활성에 기반한 다양한 의약 용도, 건강기능식품 용도 또는 화장품 용도에 관한 것이다. 본 발명의 일 예는 신규 파이토케미컬-프럭토올리고당 결합체 또는 이의 약학적으로(또는 식품학적으로) 허용가능한 염을 유효성분으로 하는 항암용 조성물을 제공한다. 또한, 본 발명의 일 예는 신규 파이토케미컬-프럭토올리고당 결합체 또는 이의 약학적으로(또는 식품학적으로) 허용가능한 염을 유효성분으로 하는 대장암 예방, 개선 또는 치료용 조성물을 제공한다. 또한, 본 발명의 일 예는 신규 파이토케미컬-프럭토올리고당 결합체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 하는 대장암의 전이 억제용 조성물을 제공한다. 또한, 본 발명의 일 예는 신규 파이토케미컬-프럭토올리고당 결합체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 하는 항산화용 조성물을 제공한다.
본 발명의 일 예에 따른 항암용 조성물에서 유효성분인 신규 파이토케미컬-프럭토올리고당 결합체는 전술한 FA-FOS Ⅰ 또는 FA-FOS Ⅱ인 것이 바람직하다. 또한, 본 발명의 일 예에 따른 대장암 예방, 개선 또는 치료용 조성물에서 유효성분인 신규 파이토케미컬-프럭토올리고당 결합체는 전술한 FA-FOS Ⅰ 또는 FA-FOS Ⅱ인 것이 바람직하고, FA-FOS Ⅱ인 것이 더 바람직하다. 또한, 본 발명의 일 예에 따른 대장암의 전이 억제용 조성물에서 유효성분인 신규 파이토케미컬-프럭토올리고당 결합체는 전술한 FA-FOS Ⅰ 또는 FA-FOS Ⅱ인 것이 바람직하고, FA-FOS Ⅰ인 것이 더 바람직하다. 또한, 본 발명의 일 예에 따른 항산화용 조성물에서 유효성분인 신규 파이토케미컬-프럭토올리고당 결합체는 전술한 FA-FOS Ⅰ 또는 FA-FOS Ⅱ인 것이 바람직하고, FA-FOS Ⅰ인 것이 더 바람직하다.
본 발명의 일 예에 따른 항암용 조성물, 대장암 예방, 개선 또는 치료용 조성물, 대장암의 전이 억제용 조성물 또는 항산화용 조성물은 사용 목적 내지 양상에 따라 약학 조성물, 식품 첨가제, 식품 조성물(특히 건강 기능 식품) 또는 사료 첨가제 등으로 구체화될 수 있다. 또한, 본 발명의 일 예에 따른 항산화용 조성물은 피부 노화방지를 위한 화장료 조성물로 구체화될 수 있다.
또한, 본 발명의 일 예에 따른 다양한 조성물에서 유효성분인 신규 파이토케미컬-프럭토올리고당 결합체의 함량도 조성물의 구체적인 형태, 사용 목적 내지 양상에 따라 다양한 범위에서 조정될 수 있다.
본 발명에 따른 약학 조성물에서 유효성분인 신규 파이토케미컬-프럭토올리고당 결합체의 함량은 크게 제한되지 않으며, 예를 들어 조성물 총 중량을 기준으로 0.01~99 중량%, 바람직하게는 0.5~50 중량%, 더 바람직하게는 1~30 중량%일 수 있다. 또한, 본 발명에 따른 약학 조성물은 유효성분 외에 약학적으로 허용가능한 담체, 부형제 또는 희석제와 같은 첨가제를 더 포함할 수 있다. 본 발명의 약학 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 또한, 본 발명의 약학 조성물은 신규 파이토케미컬-프럭토올리고당 결합체 외에 항암 활성, 특히 항-대장암 활성을 갖는 공지의 유효성분을 1종 이상 더 함유할 수 있다. 본 발명의 약학 조성물은 통상의 방법에 의해 경구 투여를 위한 제형 또는 비경구 투여를 위한 제형으로 제제화될 수 있고, 제제화할 경우 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다. 경구 투여를 위한 고형 제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형 제제는 유효성분에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(Calcium carbonate), 수크로스(Sucrose), 락토오스(Lactose) 또는 젤라틴 등을 섞어 조제될 수 있다. 또한, 단순한 부형제 이외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용될 수 있다. 경구 투여를 위한 액상 제제로는 현탁제, 내용액제, 유제 및 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제, 좌제가 포함될 수 있다. 비수성용제, 현탁용제로는 프로필렌글리콜(Propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다. 더 나아가 당 분야의 적정한 방법으로 또는 Remington's Pharmaceutical Science(최근판), Mack Publishing Company, Easton PA에 개시되어 있는 방법을 이용하여 각 질환에 따라 또는 성분에 따라 바람직하게 제제화할 수 있다. 본 발명의 약학 조성물은 목적하는 방법에 따라 인간을 포함한 포유류에 경구 투여되거나 비경구 투여될 수 있으며, 비경구 투여 방식으로는 피부 외용, 복강내주사, 직장내주사, 피하주사, 정맥주사, 근육내 주사 또는 흉부내 주사 주입방식 등이 있다. 본 발명의 약학 조성물의 투여량은 약학적으로 유효한 양이라면 크게 제한되지 않으며, 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설율 및 질환의 중증도에 따라 그 범위가 다양하다. 본 발명의 약학 조성물의 통상적인 1일 투여량은 크게 제한되지 않으나 바람직하게는 유효성분을 기준으로 할 때 10 내지 9000 ㎎/㎏이고, 더 바람직하게는 100 내지 5000 ㎎/㎏이며, 하루 1회 또는 수회로 나누어 투여될 수 있다.
또한, 본 발명에 따른 식품 조성물에서 유효성분인 신규 파이토케미컬-프럭토올리고당 결합체의 함량은 조성물 총 중량을 기준으로 0.01~99 중량%, 바람직하게는 0.1~50 중량%, 더 바람직하게는 0.5~25 중량%이나, 이에 한정되는 것은 아니다. 본 발명의 식품 조성물은 환제, 분말, 과립, 침제, 정제, 캡슐, 또는 액제 등의 형태를 포함하며, 구체적인 식품의 예로는 육류, 소시지, 빵, 초콜릿, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 기능수, 드링크제, 알코올음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강기능식품을 모두 포함한다. 본 발명의 식품 조성물은 유효성분 외에 식품학적으로 허용 가능한 담체, 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 또한, 본 발명의 식품 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다.
또한, 본 발명에 따른 화장료 조성물은 당업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있으며, 예를 들어, 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이 등으로 제형화될 수 있으나, 이에 한정되는 것은 아니다. 구체적으로, 본 발명의 화장료 조성물은 유연 화장수, 영양 화장수, 영양 크림, 마사지 크림, 에센스, 아이 크림, 클렌징 크림, 클렌징 폼, 클렌징 워터, 팩, 스프레이 또는 파우더의 제형으로 제조될 수 있다.
이하, 본 발명을 실시예를 통하여 보다 구체적으로 설명한다. 다만, 하기 실시예는 본 발명의 기술적 특징을 명확하게 예시하기 위한 것일 뿐, 본 발명의 보호범위를 한정하는 것은 아니다.
1. 페룰산-프럭토올리고당 결합체의 제조 및 치환도 분석
(1) FA-FOS Ⅰ의 제조
반응용기에 다이메틸설폭사이드(Dimethyl Sulfoxide, DMSO) 및 페룰산을 첨가하고 페룰산을 다이메틸설폭사이드(Dimethyl Sulfoxide, DMSO)에 용해시켜 5%(w/v) 농도의 페룰산 용액을 제조하였다. 이후, 반응용기에 카보닐다이이미다졸(N,N'-carbonyldiimidazole, CDI)을 첨가하고 약 60℃에서 약 16 hr 동안 교반하여 페룰산을 활성화시켰다. 상기 페룰산의 활성화 형태는 페룰로일 이미다졸(Feruloyl imidazole)이다. 이후, 반응용기에 프럭토올리고당[약 2~60개의 D-프럭토스 잔기들이 β(2→1) 결합에 의해 연결되어 있고 말단 일 측에 1개의 D-글루코스가 D-프럭토스에 α(1→2) 결합에 의해 연결된 형태의 중합체임]을 첨가하고 용해시킨 후 반응용기를 밀봉시켰다. 밀봉된 반응용기 내에 존재하는 페룰산, 카보닐다이이미다졸 및 프럭토올리고당의 몰 비는 32:32:1이었다. 이후, 반응용기의 온도를 90℃로 증가시키고, 불활성 가스(질소) 분위기하에서 약 7 hr 동안 연속적으로 교반하여 프럭토올리고당에 활성화된 페룰산을 그래프트팅(Grafting) 시키는 반응을 진행하였다. 반응 종료 후, 반응 혼합물을 실온으로 냉각시키고 여기에 얼음처럼 차가운 이소-프로필 알코올을 반응 혼합물의 약 3배 부피의 양으로 첨가하고 약 4℃에서 하룻밤 동안 방치하여 페룰산-프럭토올리고당 결합체(FA-FOS)을 침전시켰다. 이후, 반응 혼합물을 원심분리하여(4000×g, 20분) 펠렛을 수득하고, 수득한 펠렛을 이소-프로필 알코올로 약 3번 세척하여 미반응 페룰산, 카보닐다이이미다졸 및 페룰로일 이미다졸을 제거하고, 다시 증류수로 세척하여 최종 반응 산물인 페룰산-프럭토올리고당 결합체 Ⅰ('FA-FOS Ⅰ'로 약칭함)을 수득하였다. 최종 반응 산물인 FA-FOS Ⅰ의 제조 수율은 약 89% 이었다.
(2) FA-FOS Ⅱ의 제조
반응용기에 다이메틸설폭사이드(Dimethyl Sulfoxide, DMSO) 및 페룰산을 첨가하고 페룰산을 다이메틸설폭사이드(Dimethyl Sulfoxide, DMSO)에 용해시켜 5%(w/v) 농도의 페룰산 용액을 제조하였다. 이후, 반응용기에 카보닐다이이미다졸(N,N'-carbonyldiimidazole, CDI)을 첨가하고 약 60℃에서 약 16 hr 동안 교반하여 페룰산을 활성화시켰다. 상기 페룰산의 활성화 형태는 페룰로일 이미다졸(Feruloyl imidazole)이다. 이후, 반응용기에 프럭토올리고당[약 2~60개의 D-프럭토스 잔기들이 β(2→1) 결합에 의해 연결되어 있고 말단 일 측에 1개의 D-글루코스가 D-프럭토스에 α(1→2) 결합에 의해 연결된 형태의 중합체임]을 첨가하고 용해시킨 후 반응용기를 밀봉시켰다. 밀봉된 반응용기 내에 존재하는 페룰산, 카보닐다이이미다졸 및 프럭토올리고당의 몰 비는 4:4:1이었다. 이후, 반응용기의 온도를 90℃로 증가시키고, 불활성 가스(질소) 분위기하에서 약 7 hr 동안 연속적으로 교반하여 프럭토올리고당에 활성화된 페룰산을 그래프트팅(Grafting) 시키는 반응을 진행하였다. 반응 종료 후, 반응 혼합물을 실온으로 냉각시키고 여기에 얼음처럼 차가운 이소-프로필 알코올을 반응 혼합물의 약 3배 부피의 양으로 첨가하고 약 4℃에서 하룻밤 동안 방치하여 페룰산-프럭토올리고당 결합체(FA-FOS)를 침전시켰다. 이후, 반응 혼합물을 원심분리하여(4000×g, 20분) 펠렛을 수득하고, 수득한 펠렛을 이소-프로필 알코올로 약 3번 세척하여 미반응 페룰산, 카보닐다이이미다졸 및 페룰로일 이미다졸을 제거하고, 최종 반응 산물인 페룰산-프럭토올리고당 결합체 Ⅱ('FA-FOS Ⅱ'로 약칭함)을 수득하였다. 최종 반응 산물인 FA-FOS Ⅱ의 제조 수율은 약 89% 이었다.
(3) FA-FOS Ⅰ 및 FA-FOS Ⅱ의 페룰산 치환도 분석
FA-FOS Ⅰ 및 FA-FOS Ⅱ에서 페룰산의 치환도(The degree of substitution)는 프럭토올리고당 기본 단위 당 부착된 치환기 페룰산의 평균 수로 정의된다. 2M 농도의 NaOH 수용액에 FA-FOS Ⅰ 및 FA-FOS Ⅱ를 각각 첨가하고 2 hr 동안 배양하여 결합체로부터 페룰레이트기(Ferulate group)를 방출시켰다. 이후, NaOH 수용액에 6N 농도의 HCl 수용액을 첨가하여 pH를 2로 조정하고 방출된 페룰레이트기(Ferulate group)를 자유 페룰산(Free ferulic acid) 형태로 산성화시켰다. 이후, 에틸아세이트로 페룰산을 3번 추출하고 방출된 페룰산의 양을 HPLC로 정량화하였고, 하기의 식으로 페룰산의 치환도(The degree of substitution)를 계산하였다.

상기 표에서 보이는 바와 같이 FA-FOS Ⅰ의 페룰산 치환도는 FA-FOS Ⅱ 보다 약 10배 가깝게 높았다. FA-FOS Ⅰ은 물 및 유기 용매에 용해되지 않는 반면, FA-FOS Ⅱ는 물 및 유기 용매에 즉시 용해되었다. 페룰산의 치환도가 높으면 높을수록 FA-FOS의 물에 대한 용해도는 낮아지는 것으로 확인되었다. FA-FOS Ⅰ은 양쪽 친매성 때문에 물에 대해 불용성이었고, 테트라부틸 포스포늄 아세테이트(Tetrabutyl Phosphonium Acetate), 테트라부틸 암모늄 아세테이트(Tetra butyl ammonium acetate)와 같은 이온성 액체에 즉시 용해되었다.
2. 페룰산-프럭토올리고당 결합체의 구조적 특징 분석
(1) FA-FOS Ⅰ
도 1은 페룰산(FA), 프럭토올리고당(FOS) 및 FA-FOS Ⅰ을 FTIR 분광기(Fourier-transform infrared spectroscopy)로 분석한 스펙트럼 결과이다. 도 1에서 FA-FOS Ⅰ은 'FA FOS Ⅰ'로 표시되었다. 도 1에서 보이는 바와 같이 프럭토올리고당(FOS)의 3400 ㎝ -1에서의 완만한 피크는 하이드록실기의 수가 감소함에 따라 FA-FOS Ⅰ에서 강도가 크게 감소하였다. FA-FOS Ⅰ의 1726 ㎝ -1에서의 피크는 치환 또는 에스테르화 반응을 나타내고, 2947 ㎝ -1, 3008 ㎝ -1, 및 2842 ㎝ -1에서 발생하는 2개의 피크는 페룰로일(Feruloyl) 치환과 관련된 메틸 및 메틸렌 C-H 스트레칭에 기인한다. FOS의 900~1200 ㎝ -1 사이의 피크는 C-O 스트레칭에 의한 것이고, FA-FOS Ⅰ의 3074 ㎝ -1에서의 피크는 링의 메탄하이드로겐 관련 C-H 스트레칭에 기인한 것이다.
또한, 페룰산(FA), 프럭토올리고당(FOS) 및 FA-FOS Ⅰ을 13C(L armor frequency: 400.25 ㎒) 조건의 고체상태 핵자기 공명(Solid state NMR) 분광기로 분석하였다. FA-FOS Ⅰ은 다이메틸설폭사이드(DMSO) 등의 유기 용매에 용해되지 않기 때문에 그 구조를 설명하기 위하여 고체상태 핵자기 공명(Solid state NMR)을 수행하였다. FA-FOS Ⅰ의 13C 고체상태 핵자기 공명(Solid state NMR) 스펙트럼에서 페룰산의 방향족 탄소는 100 ppm 내지 150 ppm 사이에서 관찰되었고 페룰산의 카보닐 탄소(-COOH) 이동이 150 ppm 초과 영역에서 관찰되었다. 순수한 페룰산의 피크 이동은 173.13 ppm에서 관찰되지만, FA-FOS Ⅰ의 경우 프럭토올리고당(FOS)과 페룰산의 에스테르 결합 형성으로 인해 피크 이동이 164.17 ppm에서 관찰되었다. 또한, 순수한 프럭토올리고당(FOS)에서 지방족 탄소는 50 ppm 내지 100 ppm에 걸쳐 다수의 좁은 피크들로 관찰되었으나 FA-FOS Ⅰ의 경우 프럭토올리고당의 지방족 탄소는 55.17 ppm에서 단일의 넓은 피크로 관찰되었다. 이러한 결과는 FA-FOS Ⅰ에서 프럭토올리고당(FOS)의 말단 글루코스와 페룰산이 결합하고 프럭토올리고당(FOS)의 프럭토스가 4개 간격으로 페룰산과 결합하기 때문이다. FA-FOS Ⅰ의 13C 고체상태 핵자기 공명(Solid state NMR) 스펙트럼에서 관찰된 다중선(multiplets)은 다음과 같다. 13C NMR (101MHz) : d = 263.9, 251.1, 233.4, 223.3, 217.4, 180.4, 164.4, 151.9, 148.1, 144.7, 142.2, 133.6, 124.2, 117.6, 109.0, 76.7, 65.0, 55.3, 43.1, 34.1, 24.4, 18.5 ppm
또한, 페룰산(FA), 프럭토올리고당(FOS) 및 FA-FOS Ⅰ을 1H(L armor Frequency: 400.66 ㎒, spin rate: 14KHZ) 조건의 고체상태 핵자기 공명(Solid state NMR) 분광기로 분석하였다. FA의 1H 고체상태 핵자기 공명(Solid state NMR) 스펙트럼에서 13.19 ppm의 피크는 페룰산에 존재하는 카르복실산(R-COOH)의 화학적 이동에 해당한다. 반면, FA-FOS Ⅰ에서는 페룰산이 프럭토올리고당(FOS)의 당 분자들인 글루코스 및 프럭토스와 결합하기 때문에 상기 13.19 ppm의 피크는 존재하지 않고 4.94 ppm에서 강한 피크가 관찰되었다. 상기 4.94 ppm에서 강한 피크는 FA-FOS Ⅰ에서 페룰산의 방향족기에 결합되어 있는 -OH 기를 나타낸다. 또한, FA-FOS Ⅰ에서 관찰되는 6.94 ppm에서의 피크는 페룰산의 방향족 고리에 결합된 -H에 해당하며, 순수한 프럭토올리고당의 NMR 스펙트럼에는 존재하지 않는다. 또한, FA-FOS Ⅰ에서 관찰되는 2.56 ppm의 확연한 피크는 페룰산이 프럭토올리고당에 치환되어 존재하는 카보닐기에 기인한 것이며, 이는 페룰산과 프럭토올리고당(FOS)의 컨쥬게이션(conjugation)에 대한 강력한 증거가 된다.
또한, 페룰산(FA), 프럭토올리고당(FOS) 및 FA-FOS Ⅰ을 XRD(X-ray Diffraction) 분광기로 분석하였다. 그 결과, FA FOS I은 약 17.65°(2θ)와 25.55°(2θ) 에서 강한 회절 피크를 나타 내고 10.5°, 12.70°, 20.31°, 23.73°, 28.07°(2θ) 에서는 작고 넓은 피크를 나타냈다. FA FOS I은 A형 결정질상과 비결정질상의 혼합이라고 보이며 FOS의 피크들과 비교하면 FOS에 비해 더 많은 결정 구조를 갖는다. 위의 결과로부터, FA FOS I에 존재하는 선형의 프럭토스들은 비정질 영역에 기여하고 말단 글루코스에서 페룰산과의 컨쥬게이션 및 4번째 프럭토스에서 프럭토스와의 컨쥬게이션은 결정영역에 기여한다고 볼 수 있다.
또한, FA-FOS Ⅰ을 UV-Vis 분광기(Ultraviolet-visible spectroscopy) 및 형광 분광광도계(Fluorescence spectrophotometer)로 분석하였다. 그 결과, FA-FOS Ⅰ은 280 ㎚에서 광을 흡수하고 310㎚에서 광을 방출하였다.
또한, 페룰산(FA), 프럭토올리고당(FOS) 및 FA-FOS Ⅰ의 열 중량 분석(Thermogravimetric analysis, TGA)을 수행하였다. 그 결과, FOS는 238℃에서 약 41.06%의 중량 손실을 나타냈고, FA-FOS Ⅰ은 400℃에서 약 41.5%의 중량 손실을 나타냈다. 이는 FA-FOS Ⅰ이 FOS와 비교하여 더 높은 열 안정성을 가지는 것으로 볼 수 있다.
또한, FA-FOS Ⅰ의 제타 전위를 측정하고 동적광산란법(Dynamic Light Scattering, DLS)을 이용하여 입자 크기를 측정하였다. 그 결과, FA-FOS Ⅰ의 제타 전위는 -14.5±4.88 ㎷ 인 것으로 나타났고, 입자 평균 직경은 1.839±0.339 ㎛인 것으로 나타났다. FA-FOS Ⅰ의 낮은 제타 전위는 페룰산의 그래프팅에 의해 야기된 것으로 보이며, FA-FOS Ⅰ은 물(water)에서 균질한 현탁액을 생성하는 고차 구조로 자체응집(self aggregation) 되어 있음을 알 수 있다. 또한, DLS를 통해 측정한 FA-FOS Ⅰ의 다분산 지수(polydispersity index)는 0.243로서, 균질함(homogeneity)을 나타낸다.
도 2는 FA-FOS Ⅰ을 SEM(Scanning Electron Microscopy)으로 분석한 이미지 결과이다. 도 2의 i은 디스크형 구조를 보여주는 저배열 이미지이고, ii는 디스크형 입자의 단면 이미지이고, iii은 디스크형 미세입자를 보여주는 고배율 이미지이다. 도 2에서 보이는 바와 같은 FA-FOS Ⅰ은 자체응집(self aggregation)에 의해 평균 직경이 약 2㎛인 디스크 형태의 구조를 가진다. 또한, 도 2의 iii에 의하면 디스크 형태의 구조를 가지는 FA-FOS Ⅰ은 공동(cavity)을 가진 층상의 쉬트 유사 조립화에 의해 형성된다.
FA-FOS Ⅰ의 구조적 특징을 분석한 결과로부터, FA-FOS Ⅰ의 주성분은 하기의 화학식 Ⅴ로 표시되는 페룰산-프럭토올리고당 결합체인 것을 알 수 있다.

(2) FA-FOS Ⅱ
도 3은 페룰산(FA), 프럭토올리고당(FOS) 및 FA-FOS Ⅱ를 FTIR 분광기(Fourier-transform infrared spectroscopy)로 분석한 스펙트럼 결과이다. 도 3에서 FA-FOS Ⅱ는 'FA FOS Ⅱ'로 표시되었다. 도 3에서 보이는 바와 같이 프럭토올리고당(FOS)의 3400 ㎝ -1에서의 완만한 피크는 하이드록실기의 수가 감소함에 따라 FA-FOS Ⅱ에서 강도가 감소하였다. FA-FOS Ⅱ의 1717 ㎝ -1에서의 작은 피크는 에스테르화된 카보닐기를 나타내고, 900~1200 ㎝ -1 사이의 피크는 FOS 분자에서 유래하는 C-O 스트레칭에 의한 것이다.
또한, FA-FOS Ⅱ를 13C 조건의 핵자기 공명(NMR) 분광기 및 1H 조건의 핵자기 공명(NMR) 분광기로 분석하였다. FA-FOS Ⅱ의 13C 핵자기 공명(NMR) 스펙트럼에서 관찰된 다중선(multiplets)은 다음과 같다. 13C NMR (DMSO-d6) δ: 104.25, 103.52, 103.38, 81.89, 76.88, 74.46, 62.33, 61.79, 40.64, 25.76. FA-FOS Ⅱ의 1H 핵자기 공명(NMR) 스펙트럼에서 관찰된 다중선(multiplets)은 다음과 같다. 1H NMR (DMSO-d6) δ: 6.82-6.61 (m, 0H), 5.11 (d, J = 5.7 Hz, 11H), 4.67 (d, J = 6.5 Hz, 10H), 4.56 (t, J = 5.3 Hz, 11H), 4.01 (t, J = 7.3 Hz, 10H), 3.82-3.72 (m, 12H), 3.59 (d, J = 8.5 Hz, 11H), 3.55 (s, 6H), 3.43 (d, J = 19.1 Hz, 15H), 3.35 (s, 11H), 2.47 (d, J = 1.8 Hz, 0H), 2.46 (s, 1H), 2.45 (d, J = 1.9 Hz, 0H), 1.00 (s, 1H), 0.98 (s, 1H).
FA-FOS Ⅱ의 구조적 특징을 분석한 결과로부터, FA-FOS Ⅱ의 주성분은 하기의 화학식 Ⅵ로 표시되는 페룰산-프럭토올리고당 결합체인 것을 알 수 있다. 또한, FA-FOS Ⅱ를 LC-MS(Liquid Chromatography-Mass Spectrometry) 및 MS-MS(Mass spectroscopy-mass spectroscopy)로 분석한 결과, FA-FOS Ⅱ의 주쇄에 존재하는 프럭토스의 수는 3~24개로 다양하게 분포되어 있었다.

3. 페룰산-프럭토올리고당 결합체의 구강-위장관 모사 조건 등에서의 안정성 및 분해 특성
(1) 구강-위장관 모사 조건에서의 안정성 실험
FA-FOS Ⅰ 및 FA-FOS Ⅱ를 각각, 모사된 침액(Simulated Salivary fluid, SS), 모사된 위액(Simulated Gastric Fluid, SGF) 및 모사된 소장액(Simulated Small Intestine fluid, SSI)에 넣고 37℃에서 0분, 15분, 30분, 그리고 1 hr 내지 4 hr 동안 배양하였을 때의 안정성 특성을 HPLC로 측정하였다. 페룰산-프럭토올리고당 결합체의 구강-위장관 모사 조건에서의 안정성 특성 측정 실험에 사용한 모사된 침액(Simulated Salivary fluid, SS), 모사된 위액(Simulated Gastric Fluid, SGF) 및 모사된 소장액(Simulated Small Intestine fluid, SSI)의 조성은 다음과 같다.
<모사된 침액 조성>
100㎖를 기준으로 CaCl 2·2H 2O 0.0228%, NaCl 0.1017%, Na 2HPO 4 0.0204%, MgCl 2 0.0061%, K 2CO 3 0.0603%, Na 2HPO 4 0.0273%, α-amylase 200 units, Lysozyme 0.0015%로 구성되고 pH는 7.4임
<모사된 위액 조성>
100㎖를 기준으로 Sodium taurocholate 80 μM, Lecithin 20 μM, Pepsin 0.01%, Lysozyme 0.01%, Sodium Chloride 34.2 mM로 구성되고, pH는 1.6임
<모사된 소장액 조성>
100㎖를 기준으로 Sodium taurocholate 3 mM, Lecithin 0.2 mM, Maleic acid 19.12 mM, Sodium hydroxide 34.8 mM, Sodium Chloride 68.62 mM, Pancreatin (4 USP/mg) 0.05g으로 구성되고 pH는 6.5임
(2) 인간 미생물총에 의해 모사된 대장 환경 조건에서의 소화 특성 실험
멸균된 혐기성 최소 기초 배지(anaerobic minimal basal medium) 10㎖가 수용된 각각의 바이얼에 멸균된 FA-FOS Ⅰ, FA-FOS Ⅱ 및 프럭토올리고당(FOS)을 0.05g 첨가하고 혐기 조건에서 인간 대변을 1%(w/v)의 양으로 접종하고 밀봉하였다. 이후, 밀봉된 바이얼을 37℃에서 12 hr, 24 hr, 그리고 36 hr 내지 48 hr 동안 배양하고 간헐적으로 교반하였다. 배양이 완료된 후 배양된 배지를 원심분리하여 상등액을 수거하고 수거한 상등액에 6N 농도의 HCl 수용액을 첨가하여 pH를 2로 조정하고 2배 부피의 에틸 아세테이트로 자유 페룰산을 3번 추출하였다. 이후, 추출액을 질소 분위기하에서 증발시켜 에틸 아세테이트를 제거하고 50% 메탄올 수용액에 용해시켜 분석시료를 준비하였다. 이후, 분석시료를 HPLC로 분석하여 모사된 대장 환경 조건에서의 자유 페룰산 방출량을 결정하였다. 상기 실험에 사용한 혐기성 최소 기초 배지(anaerobic minimal basal medium)의 조성은 다음과 같다.
<혐기성 최소 기초 배지의 조성>
100㎖를 기준으로 Peptone 0.2g, Yeast extract 0.1g, NaCl 0.01g, K 2HPO 4 0.004g, KH 2PO 4 0.004g, MgSO 4·7H 2O 0.001g, CaCl 2·2H 2O 0.001g, NaHCO 3 0.2g, Bile salts 0.05g, L-Cysteine HCl 0.05g, Tween 80 0.2㎖, 0.05% Resazurin solution A 0.1㎖, Hemin 0.0005g, 0.02 Mm Vitamin K 1 10 μl로 구성됨
(3) 장내 소화 효소에 대한 안정성 실험
250㎎의 FA-FOS Ⅰ 및 FA-FOS Ⅱ를 각각 장내 소화 효소액에 첨가하고 37℃에서 12 hr, 24 hr, 36 hr, 그리고 48 hr 동안 배양하였다. 장내 소화 효소액은 0.02%(w/v) 소듐아자이드(Sodium azide) 용액에 대장 내 서식 세균에 의해 생산되는 Cellulase, Endo-galactouranase, endo-carbohydrase, exo-glycosidase 및 Feruloyl esterase를 첨가하여 제조한 액상 혼합 효소액이고 pH는 6이다. 구체적으로, 상기 장내 소화 효소액은 pH 6.0의 MOPS buffer 5㎖에 Driselase(Amano enzymes, Japan) 50㎎, Protease M Amano(Amano enzymes, Japan) 50㎎, DEPOL 670L(Biocatalyst, UK) 50 μl, DEPOL 740L(Biocatalyst, UK) 50 μl 및 0.02%(w/v) 소듐아자이드(Sodium azide)가 용해된 것이다. 이후, 배양액을 원심분리하여 상등액을 수거하고, 수거한 상등액에 6N 농도의 HCl 수용액을 첨가하여 pH를 2로 조정하고 3배 부피의 에틸 아세테이트로 자유 페룰산을 3번 추출하였다. 이후, 추출액을 질소 분위기하에서 증발시켜 에틸 아세테이트를 제거하고 50% 메탄올 수용액에 용해시켜 분석시료를 준비하였다. 이후, 분석시료를 HPLC로 분석하였다.
(4) 실험 결과
하기 표 2는 FA-FOS Ⅰ의 구강-위장관 모사 조건에서의 안정성 실험, 인간 미생물총에 의해 모사된 대장 환경 조건에서의 소화 특성 실험 및 장내 소화 효소에 대한 안정성 실험 결과를 나타낸 것이다. 또한, 하기 표 3은 FA-FOS Ⅱ의 구강-위장관 모사 조건에서의 안정성 실험, 인간 미생물총에 의해 모사된 대장 환경 조건에서의 소화 특성 실험 및 장내 소화 효소에 대한 안정성 실험 결과를 나타낸 것이다.

상기 표 2 및 표 3에서 보이는 바와 같이 구강-위장관 모사 조건에서의 FA-FOS Ⅰ 및 FA-FOS Ⅱ의 안정성 및 반감기 결과에 의하면, FA-FOS Ⅰ 및 FA-FOS Ⅱ는 모사된 침액(Simulated Salivary fluid, SS), 모사된 위액(Simulated Gastric Fluid, SGF) 및 모사된 소장액(Simulated Small Intestine fluid, SSI)에서 거의 분해되지 않았다.
상기 표 2에서 보이는 바와 같이 FA-FOS Ⅰ은 인간 대장 미생물총(Human microbiota)에 의해 거의 분해되지 않는 것으로 나타났다.
도 4는 인간 대장 미생물총(Human microbiota)에 의해 모사된 대장 환경 조건에서 FA-FOS Ⅱ를 배양하였을 때의 분해 거동을 HPLC로 분석한 결과이다. 상기 표 3 및 도 4에서 보이는 바와 같이 FA-FOS Ⅱ는 인간 대장 미생물총(Human microbiota)에 의해 페룰레이트(Ferulate) 형태로 분해되어 방출되는 것으로 나타났다. 또한, 인간 대장 미생물총(Human microbiota)에 의해 모사된 대장 환경 조건에서 FA-FOS Ⅱ를 배양하였을 때의 최종 대사 산물을 LC-MS(Liquid Chromatography-Mass Spectrometry) 및 MS-MS(Mass spectroscopy-mass spectroscopy)로 분석한 결과, 인간 대장 환경에서 FA-FOS Ⅱ의 최종 대사 산물은 페룰로일 글루코스(Feruloyl glucose)인 것으로 확인되었다.
상기 표 2 및 표 3에서 보이는 바와 같이 FA-FOS Ⅰ은 FA-FOS Ⅱ에 비해 장내 소화 효소에 대해 더 저항성을 가지며 방출되는 자유 페룰산의 양도 매우 적었다.
4. 페룰산-프럭토올리고당 결합체의 생체외(In-Vitro) 실험에서의 항암 활성
(1) 암세포 사멸 유도 경과 관찰 및 분석
CCD 18Co(Normal Human colon fibroblast primary cell line)와 같은 인간 정상 대장 세포주, HT-29(Colorectal adenocarcinoma, capable of inducing tumor in mice)와 같은 대장암 세포주 또는 Lovo(Duke’s type C, grade IV, Colorectal adenocarcinoma capable of metastasis)와 같은 전이성 대장암 세포주에 약물로 옥살리플라틴(Oxaliplatin), 5-플루오로우라실(5-Fluorouracil), FA-FOS Ⅰ, FA-FOS Ⅱ 및 페룰산(Ferulic acid, FA)을 처리하고 세포 사멸 유도 경과를 관찰하였다. 구체적으로 96-well 플레이트의 각 웰에 배양된 세포주를 웰당 18,000 세포수의 밀도로 분주하고 하룻밤 동안 배양한 후, 배양 배지에 다양한 농도로 용해한 약물로 세포를 처리하고 가습 배양기 내에서 37℃의 온도 및 5% CO 2 조건으로 72 hr 동안 배양하였다. 배양 완료 후 배지를 제거하고 100 μl의 PBS로 대체한 후 여기에 Cell Counting Kit-8(CCK-8; Dojindo, Japan) 용액 10 μl를 첨가하고 3 hr 동안 배양하여 반응시켰다. 이후, 반응액의 발색 정도를 450 ㎚에서의 흡광도로 측정하고 제조사의 지시에 따라 세포 증식 수를 계산하였다.
HT-29 세포는 McCoy’s 5A 배지에서 배양되었고, LoVo 세포는 Ham’s F-12K(Kaigh’s) 배지에서 배양되었고, CCD 18Co 세포는 DMEM에서 배양되었고, 모든 배지에는 사용전에 10% Fetal bovine serum 및 100 U/ml penicillin, streptomycin이 첨가되었다.
도 5는 HT-29 세포주 및 Lovo 세포주에 옥살리플라틴(Oxaliplatin), FA-FOS Ⅰ, FA-FOS Ⅱ 및 페룰산(Ferulic acid, FA)을 처리하였을 때의 암세포 사멸 유도 경과를 Annexin Ⅴ 및 Propidium iodide로 염색 후 형광 현미경으로 관찰한 결과이다. 또한, 도 6은 HT-29 세포주 및 Lovo 세포주에 FA-FOS Ⅰ 및 FA-FOS Ⅱ를 처리하였을 때의 암세포 사멸 유도 경과를 TUNEL 어세이(terminal deoxynucleotidyl transferase-dUTP nick end labeling) 및 추가적인 Hoechst 33342 염색 후 형광 현미경으로 관찰한 결과이다. 또한, 하기 표 4에 처리한 약물의 다양한 세포주에 대한 IC 50(half maximal inhibitory concentration) 값과 선택되 지수(Selectivity Index, SI) 값을 나타내었다.

도 5, 도 6 및 상기 표 4에서 보이는 바와 같이 FA-FOS Ⅰ 및 FA-FOS Ⅱ는 우수한 대장암 세포 사멸 효과를 보였다. 특히, FA-FOS Ⅰ은 상업적 약물인 옥살리플라틴(Oxaliplatin) 및 5-플루오로우라실(5-Fluorouracil)과 비교하였을 때 더 높은 선택도 지수를 가지는 것으로 확인되었다. 또한, FA-FOS Ⅰ은 FA에 비해 더 낮은 IC 50 값을 보였다.
(2) 페룰산-프럭토올리고당 결합체 FA-FOS Ⅰ의 세포사멸 유도 기전 분석
FA-FOS I에 의한 세포사멸 유도 기전을 이해하기 위해 LoVo 세포주에 FA-FOS I을 0.198 ㎍/㎖의 농도로 12 hr 동안 처리한 후 유전자 발현을 프로파일링 하였다. 처리된 세포로부터의 총 m-RNA를 Qiagen m-RNA 추출 및 정제 키트를 사용하여 추출하고, Qiagen RT2 Profiler PCR array Human apoptosis kit을 이용하여 분석하였다. 하기 표 5에 FA-FOS I으로 처리된 LoVo 세포주에서 세포사멸과 관련하여 발현된 유전자의 프로파일링 결과를 나타내었다.

상기 표 5에서 보이는 바와 같이 LoVo 세포주에 FA-FOS I 처리시 12 개의 유전자가 업레귤레이션 되고 7 개의 유전자가 다운 레귤레이션 되었다. 상기 표 5에서 업레귤레이션된 유전자인 TNFRSF9는 TNF-수용체 수퍼 패밀리에 속하는 CD137로도 불리며, T 세포(면역 세포)의 클론 확장, 생존 및 발달에 관여하고, T 세포를 활성화시켜 면역 반응을 일으키고, IL-2(Inter Lukin 2)의 분비와 면역 세포의 침윤을 유도하고 종양을 제거하는것으로 알려져있다. TNFRSF9의 업레귤레이션은 FA-FOS I 가 면역 세포 매개를 통해 생체 내 종양/암 세포를 제거하는 가능성에 대한 실마리를 제공한다
세포주기 정지(Cell Cycle Arrest)는 사이클린(Cyclin)이라 불리는 세포주기 체크 포인트 조절제를 조절함으로써 종양 세포의 증식을 검사하는 것으로 항암제의 기능 중 하나이다. 세포주기 정지에서 FA-FOS I의 기능을 조사하기 위해, LoVo 세포주와 HT-29 세포주 각각을 배양 플레이트에 시딩하고 FBS가없는 배양 배지를 사용하여 48 hr 동안 굶주리게 하여 동기화시킨 후 FA-FOS I과 FA를 여러 농도로 처리하고 LoVo 세포주에 대해 24 hr 동안 배양하고 HT-29 세포주에 대해 72 hr 동안 배양한 후 증식된 세포를 DAPI로 착색 후 세포수를 FACS로 분석하였다. FA-FOS I은 세포주기의 GO 단계에서 용량 의존적으로 세포주기를 정지시켰다. 구체적으로, HT-29 세포주에 FA-FOS I을 0.198 ㎍/㎖의 농도로 처리하는 경우 약 91.22%가 GO상에서 정지되었고, HT-29 세포주에 FA-FOS I을 0.988 ㎍/㎖의 농도로 처리하는 경우 약 94.16%가 GO상에서 정지되었다. 또한, LoVo 세포주에 FA-FOS I을 0.198 ㎍/㎖의 농도로 처리하는 경우 약 62.74%가 GO상에서 정지되었고, LoVo 세포주에 FA-FOS I을 0.988 ㎍/㎖의 농도로 처리하는 경우 약 79.52%가 GO상에서 정지되었으며, 이러한 결과는 대조군(무처리)의 48.64%에 비해 매우 높은 값이다.
또한, 세포주기 정지에 대한 FA-FOS I의 기능을 검증하기 위해 LoVo 세포주로부터 총 단백질을 추출하고, 사이클린 B1, E 및 사이클린 의존성 키나제 억제 단백질 p21의 발현 프로파일링을 밀도계(densitometry)로 분석하였다. 그 결과, FA-FOS I은 세포주기를 조절하는 모든 사이클린을 억제하는 p21의 발현을 업레귤레이션(Up-regulation)하고 G2 기에서 유사 분열로의 세포 프로그레션(cell progression)에 관여하는 사이클린 B1의 발현을 다운 레귤레이션(Down-regulation) 하며 G0/G1 상에서 S 상으로의 세포의 프로그레션에 관여하는 Cyclin E를 다운 레귤레이션 함으로써 세포주기 정지를 유도하는 것으로 나타났다.
또한, FA-FOS I을 0.198 ㎍/㎖의 농도로 24 hr 동안 처리한 LoVo 세포주를 사용하여 세포주기 관련 유전자의 발현 향상을 조사하였다. 유전자 발현량은 Qiagen의 RT2 Prolifer PCR Array Human Cell cycle를 사용하여 측정하였다. 하기 표 6에 FA-FOS I으로 처리된 LoVo 세포주에서 세포주기와 관련하여 발현된 유전자의 프로파일링 결과를 나타내었다. 하기 표 6에서 보이는 바와 같이 세포주기 진행 및 증식에 직접적으로 관여하는 9개의 주요 업레귤레이션 유전자 및 4개의 주요 다운레귤레이션 유전자가 있는 것으로 관찰되었다.

AOM(Azoxymethane)-DSS(Dextran Sodium sulfate)로 결장암이 유발된 마우스 모델을 이용하여 FA-FOS I 및 FA-FOS Ⅱ의 항암 활성을 평가하였다. 5~6 주령된 C57BL/6 암컷 마우스를 각각 7마리씩 아래의 6개군에 무작위로 할당하고 각 군에 필요한 처리를 하였다.
(i) 정상군(Naive)
(ii) 대조군(Conrtol) : AOM-DSS에 의해 결장암이 유발된 마우스로서, 별도의 약물을 투여하지 않음
(iii) 옥살리플라틴 처리군(Oxaliplatin) : 옥살리플라틴을 물(water)에 용해시켜 약물을 준비하고, 옥살리플라틴 양을 기준으로 마우스에 30 mg/㎏(b.w)의 용량으로 경구투여함
(iv) 페룰산 처리군(FA) : 페룰산을 물에 용해시켜 약물을 준비하고, 페룰산 양을 기준으로 마우스에 100 mg/㎏(b.w)의 용량으로 경구투여함
(v) FA-FOS I 처리군(FA FOS-I) : 페룰산-프럭토올리고당 결합체 FA-FOS I을 물에 현탁하여 약물을 준비하고, FA-FOS I 양을 기준으로 500 mg/㎏(b.w)의 용량으로 경구투여함
(vi) FA-FOS Ⅱ 처리군(FA FOS-Ⅱ) : 페룰산-프럭토올리고당 결합체 FA-FOS Ⅱ를 물에 용해시켜 약물을 준비하고, FA-FOS Ⅱ 양을 기준으로 3890 mg/㎏(b.w)의 용량으로 경구투여함
참고로, 페룰산-프럭토올리고당 결합체 FA-FOS I 및 FA-FOS Ⅱ를 페룰산의 양으로 환산하여 비교하면, FA-FOS I 500 mg 및 FA-FOS Ⅱ 3890 mg은 페룰산 100 mg에 해당한다.
도 7은 AOM-DSS에 의해 결장암이 유발된 마우스 모델을 이용하여 페룰산-프럭토올리고당 결합체의 항암 활성을 평가하기 위한 실험 진행 과정을 개략적으로 나타낸 것이다.
도 8은 AOM-DSS에 의해 결장암이 유발된 마우스 모델을 이용하여 페룰산-프럭토올리고당 결합체의 항암 활성을 평가한 실험 결과 중 (a) 결장에서의 종양 병변의 총 수, (b) 콜론 인덱스, (c) 약물 치료 종료시 마우스의 체중 및 (d) 4 주의 치료 요법 동안 마우스의 체중 증가율을 나타낸 것이다. 도 8에서 보이는 바와 같이 FA-FOS Ⅱ 처리군에서 종양 병변의 수가 대조군에 비해 77.1% (p <0.001) 감소되었다. 또한, FA-FOS Ⅱ 처리군은 옥살리플라틴 처리군 및 페룰산 처리군에 비교하여 더 높은 종양 감소를 보였다. 또한, 마우스 결장 지수는 FA-FOS Ⅱ 처리군과 정상군 간에서 유의한 차이가 없었다.
도 9는 AOM-DSS에 의해 결장암이 유발된 마우스 모델을 이용하여 페룰산-프럭토올리고당 결합체의 항암 활성을 평가한 실험 결과 중 면역학적 파라미터 결과를 나타낸 것이다. 도 9에서 (a)는 전체 혈액속의 백혈구(WBC) 수, (b)는 총 골수 세포 수, (c)는 흉선 지수 (흉선/체중), (d)는 비장 지수 (비장/체중) (e)는 혈장 내 인터루킨 3 농도, 그리고 (f)는 혈장 내 인터페론 감마 농도 결과이다. 도 9에서 보이는 바와 같이 면연 관련 파라미터 중 WBC의 수는 대조군과 비교할 때 FA-FOS I 처리군에서 약 6.90% 감소하였고(p <0.01) FA-FOS Ⅱ 처리군에서 약 5.38% 감소하였다(p < 0.05). 그러나 골수 세포 수, 흉선 지수 및 비장 지수 등을 고려할 때, FA-FOS I 처리 및 FA-FOS Ⅱ 처리에 의해 유도된 면역 억제는 없는 것으로 판단된다. 한편, 마우스의 혈장에 존재하는 IL-3 농도는 FA-FOS Ⅱ 처리군에서 대조군에 비해 약 15% 증가하였다(p <0.05). 이러한 결과는 FA-FOS Ⅱ에 의해 T 세포의 종양 항원에 대한 자극/감작이 존재함을 시사한다.
(2) 래트(rat)에서 FA-FOS Ⅱ의 생체내 약동학
FA-FOS Ⅱ의 생체내 약동학을 평가하기 위해 래트(rat)의 결장에서 FA-FOS Ⅱ로부터 유리된 페룰산(FA)의 방출 프로파일을 조사하였다. 구체적으로, 체중이 각각 180-250g 인 수컷 스프라그 다울리(Sprague Dawley) 래트를 아래와 같이 3개의 군으로 무작위 배정하였다.
(i) 정상군 : 별도의 약물을 투여하지 않음
(ii) 페룰산(FA) 투여군 : 페룰산(FA)을 물에 용해시켜 약물을 준비하고 소정의 용량으로 경구투여함
(iii) FA-FOS Ⅱ 투여군 : FA-FOS Ⅱ를 물에 용해시켜 약물을 준비하고 소정의 용량으로 경구투여함
페룰산(FA)은 멸균수에 용해된 상태에서 페룰산(FA)의 양을 기준으로 100mg/㎏ 체중의 용량으로 경구투여 되었다. 또한, FA-FOS Ⅱ는 멸균수에 용해된 상태에서 FA-FOS Ⅱ의 양을 기준으로 3.89g/㎏ 체중(페룰산의 양으로 환산하면 100mg/㎏ 체중에 해당함)의 용량으로 경구투여 되었다. 경구 섭식을 통한 단일 용량 투여 후, 래트 혈액을 0, 0.5, 1, 3, 6, 8, 12, 24 hr에 걸쳐 연속적으로 샘플링하였다. 샘플링한 시료에 대해 물질 분자량 193.0/133.9 Da(FA의 intact mass/fragment mass)의 다중 반응 모니터링(MRM) 방식으로 LC MS/MS 분석을 수행하여 혈장에서 유리된 FA를 정량화했다.
도 10은 래트(Rat) 혈장에서 FA-FOS Ⅱ로부터 유리된 페룰산(FA)의 약동학적 방출 프로파일을 나타낸 것이다. 도 10에서 보이는 바와 같이 FA-FOS Ⅱ 투여군에서 FA-FOS Ⅱ로부터 유리된 FA의 Tmax는 약 30분이었다. 반면, 페룰산(FA) 투여군에서 Tmax는 약 5분이었다. 또한, 도 10에서 보이는 바와 같이 FA-FOS Ⅱ 투여군에서 FA-FOS Ⅱ로부터 유리된 FA의 평균 체류 시간은 약 240분으로, 페룰산(FA) 투여군에서 T 1/2이 약 30분인 것과 비교할 때 비교적 긴 시간이었다. FA-FOS Ⅱ 투여 후 관찰된 유리 FA의 Cmax는 27.08 μM이었다. AUC(Area Under Curve)는 약물 분자가 신체에 노출 된 정도(혈장 내 약물 분자의 퍼짐)를 나타내는데, FA-FOS Ⅱ 투여군에서 AUC(Area Under Curve)는 6234.77 μM*Min/L 이었다. 이는 페룰산(FA)을 기준으로 동일 용량의 페룰산(FA)을 투여한 군에서의 AUC보다 약 2.02배 높은 값이다. 따라서, 페룰산(FA) 분자의 생체 이용률은 FA-FOS Ⅱ를 투여하였을 때 페룰산(FA) 자체를 투여한 것에 비해 약 2배가 되는 것을 알 수 있다.
(3) HT-29 종양(인간 종양) 보유 이종 이식 마우스 모델에서 FA-FOS Ⅱ의 항암 활성
HT-29 종양을 보유한 이종 이식 마우스를 사용하여 FA-FOS Ⅱ의 항암 활성을 평가하였다. 구체적으로 BALB/c 누드 마우스를 아래의 4개군에 무작위로 할당하고 각 군에 필요한 처리를 하였다. 또한, HT-29 종양을 보유한 이종 이식 마우스 모델은 다음과 같은 방법으로 제조하였다. 먼저 HT-29 세포주를 5% CO 2 조건으로 10% FBS를 갖는 RPMI 1640 배지에서 배양하고 수확하였다. 수확한 HT-29 세포주 배양액을 희석하고 1×10 7 cells/0.2㎖/mice의 양으로 BALB/c 누드 마우스의 우측 측면 피하 영역에 주사한 후 종양 부피가 150-200 ㎜ 3에 도달 할 때까지 기다렸다.
(i) 정상군(Naive)
(ii) 대조군(conrtol) : HT-29 종양을 보유한 이종 이식 마우스로서, 별도의 약물을 투여하지 않음
(iii) 5-플루오로우라실 처리군(5-Fu) : 5-플루오로우라실(5-Fluorouracil)을 0.9% 식염수에 용해시켜 약물을 준비하고, 복강 내 IP 투여함/ 0.2㎖ 이하로 1주일 동안 5일 연속으로 하루에 한 번 투여하고, 1주일 휴식하고, 5-플루오로우라실(5-Fluorouracil) 양을 기준으로 마우스에 1회당 15 mg/㎏(b.w)의 용량으로 2주 동안 1주일에 2회 IP 투여함(이러한 특정 치료 요법은 연구자에 의해 관찰된 5-FU의 부작용으로 인해 선택됨)
(iv) FA-FOS Ⅱ 처리군(FA FOS Ⅱ) : 페룰산-프럭토올리고당 결합체 FA-FOS Ⅱ를 증류수에 용해시켜 약물을 준비하고, FA-FOS Ⅱ 양을 기준으로 3890 mg/㎏(b.w)의 용량으로 4주 동안 매일 경구투여함
도 11은 HT-29 종양 보유 이종 이식 마우스 모델을 이용하여 페룰산-프럭토올리고당 결합체 FA-FOS Ⅱ의 항암 활성을 평가한 실험 결과 중 (a) 4 주 치료 후 종양 부피, (b) 종양 성장률, (c) 4 주 치료 후 종양 무게, (d) 4 주 치료 후 체중 , (e) 치료 과정 동안 매일 체중 증가율 및 (f) 치료 동안 체중 증가 프로파일을 나타낸 것이다. 도 11에서 보이는 바와 같이 FA-FOS Ⅱ 처리군에서 종양 부피는 대조군과 비교할 때 약 50% 감소하였다(p <0.05). 또한, FA-FOS Ⅱ 처리군에서 평균 종량 중량은 대조군과 비교할 때 약 37.34% 감소하였다. 약물 치료의 독성 또는 부작용을 간접적으로 알아보기 위해 측정한 체중은 FA-FOS Ⅱ 처리군에서 대조군과 비교할 때 약 6% 증가하였다. 또한, 매일 체중 증가율도 FA-FOS Ⅱ 처리군에서 대조군과 비교할 때 약 41.47% 증가했다. 전체 치료 기간 동안 FA-FOS Ⅱ 처리군에서의 체중 증가가 대조군보다 더 높은 것을 보여주었고 이러한 결과는 페룰산-프럭토올리고당 결합체 FA-FOS Ⅱ가 부작용을 유발하지 않는다는 점을 시사한다.
6. 페룰산-프럭토올리고당 결합체의 항산화 활성
FA-FOS Ⅰ의 항산화 활성을 ABTS[2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)] 양이온 라디칼 소거능 분석법을 이용하여 측정하였다. ABTS 양이온 라디칼 소거능 분석법은 Arda Serpen et al., 2007에 의거하여 수행되었다. 구체적으로, 에펜도르프 튜브(eppendorf tube) 내에 동결건조된 FA-FOS Ⅰ을 다양한 양으로 첨가한 후 여기에 MnO 2를 사용하여 화학적으로 미리 생성한 ABTS 양이온 라디칼 시약 1.7㎖를 첨가한 후 2분 동안 교반하여 균일하게 혼합하고 6분 동안 반응을 진행하였다. 이후, 반응 혼합물을 원심분리하여 상등액을 수거하고, UV-VIS 플레이트 판독기를 이용하여 상등액의 흡광도를 734 ㎚에서 측정하였다. 도 12는 FA-FOS Ⅰ의 항산화 활성을 ABTS[2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)] 양이온 라디칼 소거능 분석법을 이용하여 측정한 결과이다. 도 12에서 'FA FOS Ⅰ'은 FA-FOS Ⅰ을 나타낸다. 또 12에서 보이는 바와 같이 FA-FOS Ⅰ은 우수한 항산화 활성을 보였고 FA-FOS Ⅰ의 EC 50은 약 7.16㎎ 이었다.
이상에서와 같이 본 발명을 실시예를 통해 설명하였지만 본 발명이 반드시 여기에만 한정되는 것은 아니며 본 발명의 범주와 사상을 벗어나지 않는 범위 내에서 다양한 변형실시가 가능함은 물론이다. 따라서, 본 발명의 보호범위는 본 발명에 첨부된 특허청구의 범위에 속하는 모든 실시 형태를 포함하는 것으로 해석되어야 한다.

Claims (15)

  1. 하기 화학식 Ⅱ로 표시되는 화합물에서 선택되는 파이토케미컬-프럭토올리고당 결합체 :
    [화학식 Ⅱ]
    PhA-Glu-(Fru) n-Fru
    상기 화학식 Ⅱ에서,
    'PhA'는 페놀산 형태의 파이토케미컬을 나타내고, 'Glu'는 글루코스를 나타내고, 'Fru'는 프럭토스를 나타내고;
    'PhA'와 'Glu'는 에스테르 결합에 의해 연결되고, 'Glu'와 'Fru'는 글리코시딕 결합에 의해 연결되고, 'Fru'와 'Fru'는 글리코시딕 결합에 의해 연결되고;
    n은 글리코시딕 결합에 의해 연결된 'Fru'의 수로서 1 내지 59의 정수에서 선택된다.
  2. 제1항에 있어서, 상기 페놀산 형태의 파이토케미컬은 페룰산(Ferulic acid), 카페익산(Caffeic acid), 신남산(Cinnamic acid), 클로로겐산(Chlorogenic acid), 쿠마린(Coumarin), 시나픽산(Sinapinic acid), 치코르산(Cichoric acid), 다이페룰산(Diferulic acid), 쿠마르산(Coumaric acid), 살리실산(Salicylic acid), 3-하이드록시벤조산(3-Hydroxybenzoic acid), 4-하이드록시벤조산(4-Hydroxybenzoic acid), 바닐산(Vanillic acid), 갈산(Gallic acid) 또는 엘라그산(Ellagic acid)에서 선택되는 것을 특징으로 하는 파이토케미컬-프럭토올리고당 결합체.
  3. 제1항에 있어서, 상기 페놀산 형태의 파이토케미컬은 글루코스 6번 탄소에 위치하는 하이드록실기와 파이토케미컬에 존재하는 카르복실기 사이의 에스테르 결합에 의해 연결되는 것을 특징으로 하는 파이토케미컬-프럭토올리고당 결합체.
  4. 제1항에 있어서, 하기 화학식 Ⅳ로 표시되는 페룰산-프럭토올리고당 결합체에서 선택되는 파이토케미컬-프럭토올리고당 결합체 :

  5. 제4항에 있어서, 하기 화학식 Ⅵ로 표시되는 페룰산-프럭토올리고당 결합체로 이루어지거나 이를 주성분으로 포함하는 파이토케미컬-프럭토올리고당 결합체.

  6. 제1항 내지 제5항 중 어느 한 항에서 선택되는 파이토케미컬-프럭토올리고당 결합체의 약학적으로 허용가능한 염.
  7. (a) 반응 용매에 페놀산(Phenolic acid) 형태의 파이토케미컬(Phytochemical) 및 에스테르화 반응용 촉매를 첨가 및 용해하고 가열하여 파이토케미컬의 활성화 반응을 진행하고 활성화된 형태의 파이토케미컬을 포함하는 1차 반응 혼합물을 수득하는 단계; 및
    (b) 상기 1차 반응 혼합물에 하기의 일반 구조식으로 표시되는 프럭토올리고당(Fructooligosaccharide)을 첨가하고 불활성 가스 분위기하에서 가열하여 프럭토올리고당(Fructooligosaccharide)과 파이토케미컬(Phytochemical) 간의 에스테르화 반응을 진행하고 제1항 내지 제5항 중 어느 한 항의 파이토케미컬-프럭토올리고당 결합체를 포함하는 2차 반응 혼합물을 수득하는 단계를 포함하는 파이토케미컬-프럭토올리고당 결합체 제조방법 :

    상기 프럭토올리고당의 일반 구조식에서 'Glu'는 글루코스를 나타내고, 'Fru'는 프럭토스를 나타내고, k는 글리코시딕 결합을 통해 연결된 프럭토스의 수로서 2 내지 60의 정수에서 선택된다.
  8. 제7항에 있어서, 하기의 단계를 더 포함하는 파이토케미컬-프럭토올리고당 결합체 제조방법 :
    (c) 상기 2차 반응 혼합물을 냉각 및 방치하여 파이토케미컬-프럭토올리고당 결합체를 침전시키고 원심분리 및 세척을 순차적으로 진행하여 정제된 파이토케미컬-프럭토올리고당 결합체를 수득하는 단계.
  9. 제7항에 있어서, 상기 (a) 단계의 파이토케미컬 활성화 반응은 45~120℃에서 2~20 hr 동안 진행되는 것을 특징으로 하는 파이토케미컬-프럭토올리고당 결합체 제조방법.
  10. 제7항에 있어서, 상기 (b) 단계의 에스테르화 반응은 70~150℃에서 2~15 hr 동안 진행되는 것을 특징으로 하는 파이토케미컬-프럭토올리고당 결합체 제조방법.
  11. 제7항에 있어서, 상기 에스테르화 반응용 촉매는 카보닐다이이미다졸(N,N'-carbonyldiimidazole, CDI)인 것을 특징으로 하는 파이토케미컬-프럭토올리고당 결합체 제조방법.
  12. 제7항에 있어서, 상기 파이토케미컬-프럭토올리고당 결합체를 제조하기 위해 사용된 페놀산(Phenolic acid) 형태의 파이토케미컬(Phytochemical), 에스테르화 반응용 촉매 및 프럭토올리고당의 몰 비는 2:2:1 내지 10:10:1의 범위에서 선택되는 것을 특징으로 하는 파이토케미컬-프럭토올리고당 결합체 제조방법.
  13. 제4항 또는 제5항 중 어느 한 항의 파이토케미컬-프럭토올리고당 결합체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 항암용 조성물.
  14. 제4항 또는 제5항 중 어느 한 항의 파이토케미컬-프럭토올리고당 결합체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 대장암 예방 또는 치료용 약학 조성물.
  15. 제4항 또는 제5항 중 어느 한 항의 파이토케미컬-프럭토올리고당 결합체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 대장암 전이 억제용 조성물.
