Abstract
본 발명의 일 실시예는, (A) 합성 고분자 수지를 용융 및 성형하여 베이스 필름을 제조하는 단계; (B) 상기 베이스 필름을 가압하여 제1 고분자 필름을 제조하는 단계; 및 (C) 상기 제1 고분자 필름 및 천연 고분자 수지를 포함하는 제2 고분자 필름을 교차적으로 적층 및 접합하는 단계;를 포함하고, 하기 조건을 만족하는 치조골 재생 차폐막의 제조방법을 제공한다. 상기 베이스 필름 및 상기 제1 고분자 필름의 평균 기공 크기는 각각 D 1 및 D 2이고, 0.1 ≤ D 2/D 1 ≤ 0.5 이다.
Description
본 발명은 치조골 재생 차폐막의 제조방법에 관한 것으로, 더 상세하게는, 성형이 용이하고, 성형 후 공간유지능이 우수하여 시술 편의성이 향상된 치조골 재생 차폐막의 제조방법에 관한 것이다.
인구 노령화로 인해 치아의 기능이 상실하거나 치아결손부의 장기적인 방치로 인해 치조골이 소실되거나 그 역할을 제대로 하지 못하는 경우가 증가하고 있다. 치아 기능만이 상실된 경우 임플란트 시술을 통해 인공치아를 식립하여 기능을 회복할 수 있으나, 치조골이 소실된 경우에는 골이식술을 통해 새로운 치조골을 형성시킨 후 인공치아를 식립해야 한다.
구체적으로, 교정 시술이나 임플란트 시술 시 뼈의 질이 좋지 않거나 뼈의 양이 부족한 경우 또는 치주염으로 인해 치조골이 파괴, 소실된 경우에는 치조골을 증가시키는 시술이 필요한데, 이와 같이 골재생(골조성)이 필요한 경우에 골유도 재생술(guided bone regeneration, GBR)이나 골이식술(bone grafting)이 수행되고 있다. 상기 골유도 재생술은 골재생 부위에 차폐막을 덮고 미분화된 섬유아세포의 침입을 차단하면서 치조골의 생장을 위한 방어벽을 형성하는 치조골 재생술이다. 상기 골유도 재생술의 경우, 골재생 부위에 적절한 공간을 부여하기 위한 차폐막이 필요하다.
대표적으로 사용되는 차폐막으로는 PTFE(Polytetrafluoroethylene), 콜라겐(Collagen), 티타늄(Titanium) 등이 있다.
콜라겐 차폐막은 일반적으로 동물 유래로 얻어진 콜라겐을 이용한 흡수성 차폐막으로 골재생의 높은 성공률과 제거 수술이 불필요한 장점이 있으나, 대부분의 흡수성 차폐막은 막 자체의 견고성이 떨어져 골 이식재를 이식부에 고정이 용이하지 않고, 탄성에 의해 성형 후의 유지성이 불량할 수 있으며 찢기거나 밀려 정확한 위치에 고정하기 어려워 이들 틈으로 골 이식재가 탈락되거나 이동할 수 있다는 문제점이 있다. 또한, 흡수성 차폐막은 조기에 흡수될 수 있어 골조직 재생 효과가 저하될 수 있다.
티타늄 차폐막은 비흡수성 차폐막으로 금속 고유의 견고성에서 유래한 공간 유지력이 우수하여 다량의 골이식이 가능한 것으로 알려져 있다. 다만, 티타늄 차폐막은 환자 개별의 치주 형태에 따라 추가적인 성형이 필요하고, 치조골 재생 후 별도의 2차 수술을 통해 제거되어야 하는 불편함이 있다. 특히, 상기 성형 과정에서 절곡 부위가 국부적으로 과도하게 돌출되는 현상으로 인해 골재생 유도 기간 동안 돌출부가 치은을 뚫고 나와 외부로부터 감염을 유발할 수 있다.
또한, PTFE 차폐막은 비흡수성 차폐막으로 생체적합성이 우수하여 인체에 유해하지 않고, 차폐 안정성이 우수하고 기존 뼈와 인공 뼈를 본래의 형태로 유지시켜주는 장점이 있다. 그러나, PTFE 차폐막은 티타늄 차폐막과 마찬가지로 2차 수술을 통해 제거하여야 하고, 차폐막의 노출 부위에 치태 등이 침착되어 감염을 유발할 수 있다.
이에 대해, 한국등록특허 제10-1649125호는 생분해성 고분자에 생체 재료를 적층시키는 치조골 재생 차폐막의 제조방법을 제공하며, 이를 통해 높은 공간유지능을 구현하였다. 다만, 상기 치조골 재생 차폐막은 벤딩 구조로 열성형된 상태로 제공되어 환자의 구강 구조 및 형상에 맞추어 시술자가 차폐막을 성형하기 어렵기 때문에 시술 편의성의 측면에서 한계가 있다.
본 발명은 전술한 종래 기술의 문제점을 해결하기 위한 것으로, 본 발명의 목적은 성형이 용이하고, 성형 후 공간유지능이 우수하여 시술 편의성이 향상된 치조골 재생 차폐막의 제조방법을 제공하는 것이다.
본 발명의 일 측면은, (A) 합성 고분자 수지를 용융 및 성형하여 베이스 필름을 제조하는 단계; (B) 상기 베이스 필름을 가압하여 제1 고분자 필름을 제조하는 단계; 및 (C) 상기 제1 고분자 필름 및 천연 고분자 수지를 포함하는 제2 고분자 필름을 교차적으로 적층 및 접합하는 단계;를 포함하고, 하기 조건을 만족하는 치조골 재생 차폐막의 제조방법을 제공한다. 상기 베이스 필름 및 상기 제1 고분자 필름의 평균 기공 크기는 각각 D

일 실시예에 있어서, 상기 합성 고분자 수지는 PLA(polylactic acid), PGA(polyglycolic acid), PLGA(poly lactic-co-glycolic acid), PLLA(poly(L-lactic acid)), PCL(polycaprolactone), PLCL(poly lactide-co-caprolactone), PHB(polyhydroxybutyrate), PHV(polyhydroxyvalerate), PDO(polydioxanone), PTMC(Polytrimethylenecarbonate) 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나일 수 있다.
일 실시예에 있어서, 상기 (A) 단계에서, 상기 성형은 적층 가공(additive manufacturing)으로 수행될 수 있다.
일 실시예에 있어서, 상기 적층가공은 하나 이상의 노즐을 포함하는 FDM(Fused Deposition Modeling) 방식의 3D 프린터를 이용하여 수행될 수 있다.
일 실시예에 있어서, 상기 노즐의 직경은 0.1~0.5mm일 수 있다.
일 실시예에 있어서, 상기 (B) 단계에서, 상기 가압은 3~50mPa의 압력으로 수행될 수 있다.
일 실시예에 있어서, 상기 천연 고분자 수지는 콜라겐, 키토산, 히알루론산, 카르복시메틸셀룰로오스, 헤파란설페이트, 덱스트란, 알지네이트 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나일 수 있다.
일 실시예에 있어서, 상기 제2 고분자 필름은 하기 단계를 포함하는 방법에 따라 제조될 수 있다. (b1) 제2 고분자 수지를 산 용액에 용해시키는 단계; (b2) 상기 (b1) 단계의 생성물에 염을 투입하여 상기 제2 고분자 수지를 섬유화하는 단계; (b3) 상기 (b2) 단계의 생성물을 건조시키는 단계; (b4) 상기 (b3) 단계의 생성물에 가교제를 투입하여 상기 제2 고분자 수지를 가교시키는 단계; 및 (b5) 상기 (b4) 단계의 생성물을 세척 및 건조시킨 후 가압하는 단계.
일 실시예에 있어서, 상기 산 용액은 인산, 황산, 염산, 질산, 아세트산 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나를 포함할 수 있다.
일 실시예에 있어서, 상기 염은 인산염, 칼슘염, 나트륨염 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나일 수 있다.
일 실시예에 있어서, 상기 가교제는 포름알데히드(formaldehyde), a-하이드록시아디프알데히드(a-hydroxyadipaldehyde), 글루타르알데히드(glutaraldehyde), EDC/NHS(1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide/N-hydroxy succinimide) 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나일 수 있다.
일 실시예에 있어서, 상기 (C) 단계는 하기 단계를 포함할 수 있다. (c1) 상기 제1 및 제2 고분자 필름을 교차적으로 적층하는 단계; 및 (c2) 상기 (c1) 단계의 생성물을 30~80℃의 온도로 가열하고, 3~30mPa의 압력으로 접합하는 단계.
일 실시예에 있어서, 상기 (C) 단계에서, 상기 제1 고분자 필름의 적어도 일 면에 상기 천연 고분자 수지를 포함하는 상기 제2 고분자 필름을 적층 및 접합할 수 있다.
본 발명의 일 측면에 따르면, 복수의 층이 적층된 구조의 치조골 재생 차폐막에서 기저층 또는 코어층에 사용되는 합성 고분자 필름의 제조 시 가압에 따른 평균 기공 크기를 미리 정해진 범위로 조절하고, 이를 천연 고분자 필름과 적층 및 접합함으로써 제조된 치조골 재생 차폐막의 성형성과 성형 후 유지능, 즉, 공간유지능을 균형적으로 구현할 수 있고, 그에 따른 시술자의 시술 편의성을 극대화할 수 있다.
본 발명의 효과는 상기한 효과로 한정되는 것은 아니며, 본 발명의 상세한 설명 또는 청구범위에 기재된 발명의 구성으로부터 추론 가능한 모든 효과를 포함하는 것으로 이해되어야 한다.

이하에서는 첨부한 도면을 참조하여 본 발명을 설명하기로 한다. 그러나 본 발명은 여러 가지 상이한 형태로 구현될 수 있으며, 따라서 여기에서 설명하는 실시예로 한정되는 것은 아니다. 그리고 도면에서 본 발명을 명확하게 설명하기 위해서 설명과 관계없는 부분은 생략하였으며, 명세서 전체를 통하여 유사한 부분에 대해서는 유사한 도면 부호를 붙였다.

상기 치조골 재생 차폐막의 제조방법은, 실질적으로, 상기 제1 고분자 필름을 제조하는 단계, 상기 제2 고분자 필름을 제조하는 단계, 및 상기 제1 및 제2 고분자 필름을 적층 및 접합하는 단계를 포함할 수 있다.
제1 고분자 필름의 제조
상기 제1 고분자 필름은, (A) 합성 고분자 수지를 용융 및 성형하여 베이스 필름을 제조하는 단계, 및 (B) 상기 베이스 필름을 가압하여 제1 고분자 필름을 제조하는 단계에 의해 제조될 수 있다.
상기 합성 고분자 수지는 PLA(polylactic acid), PGA(polyglycolic acid), PLGA(poly lactic-co-glycolic acid), PLLA(poly(L-lactic acid)), PCL(polycaprolactone), PLCL(poly lactide-co-caprolactone), PHB(polyhydroxybutyrate), PHV(polyhydroxyvalerate), PDO(polydioxanone), PTMC(Polytrimethylenecarbonate) 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나일 수 있고, 바람직하게는, PCL(polycaprolactone)일 수 있으나, 이에 한정되는 것은 아니다.
상기 (A) 단계에서, 상기 성형은 캐스팅, 용융압착, 압출, 사출, 적층 가공(additive manufacturing) 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나, 바람직하게는, 적층 가공, 즉, 3D 프린팅 방식으로 수행될 수 있다. 예를 들어, 상기 3D 프린팅은 하나 이상의 노즐을 포함하는 FDM(Fused Deposition Modeling) 방식의 3D 프린터를 이용하여 수행될 수 있고, 상기 노즐의 직경은 0.1~0.5mm일 수 있다. 이 때, 상기 노즐의 직경은 0.1 mm 이상, 0.2 mm 이상, 또는 0.3 mm 이상이고, 0.5 mm이하일 수 있다. 상기 노즐의 직경은 상기 제1 고분자 필름의 기공 특성, 구체적으로, 기공율 및/또는 기공 크기에 직, 간접적으로 영향을 미치는 인자이며, 예를 들어, 상기 노즐이 직경이 클수록 상기 베이스 필름의 평균 기공 크기가 커질 수 있다.
상기 (B) 단계에서, 상기 가압은 3~50mPa의 압력으로 수행될 수 있다. 이 때, 상기 (B) 단계에서 가해지는 압력은 3 mPa 이상, 또는 30 mPa 이상이고, 50 mPa 이하일 수 있다. 상기 가압에 의해 상기 베이스 필름의 평균 기공 크기를 축소시킬 수 있고, 축소 가능한 범위는, 예를 들어, 상기 베이스 필름 및 상기 제1 고분자 필름의 평균 기공 크기가 각각 D

상기 제1 고분자의 필름의 제조방법은, 예를 들어, 합성 고분자 수지 원료를 평균 입도가 0.1~2mm인 분말상으로 준비하는 단계; 상기 합성 고분자 수지 원료를 0.1~0.5mm의 두께, 0.1~3mm 의 평균 기공 크기를 가지는 베이스 필름으로 제조하는 단계; 상기 베이스 필름을 3분~3시간 동안 냉각하고, 3~50mPa로 가압하여 상기 베이스 필름의 평균 기공 크기와 두께를 축소시키는 단계;를 포함할 수 있으나, 각 단계 및 조건이 이에 한정되는 것은 아니다.
제2 고분자 필름의 제조
도 2는 본 발명의 일 실시예에 따른 제2 고분자 필름의 제조방법을 도식화한 것이다. 도 2를 참고하면, 상기 제2 고분자 필름은, (b1) 제2 고분자 수지를 산 용액에 용해시키는 단계; (b2) 상기 (b1) 단계의 생성물에 염을 투입하여 상기 제2 고분자 수지를 섬유화하는 단계; (b3) 상기 (b2) 단계의 생성물을 건조시키는 단계; (b4) 상기 (b3) 단계의 생성물에 가교제를 투입하여 상기 제2 고분자 수지를 가교시키는 단계; 및 (b5) 상기 (b4) 단계의 생성물을 세척 및 건조시킨 후 가압하는 단계;를 포함하는 방법에 따라 제조될 수 있다.
상기 (b1) 단계에서, 상기 산 용액은 인산, 황산, 염산, 질산, 아세트산 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나, 바람직하게는, 인산을 포함할 수 있으나, 이에 한정되는 것은 아니다.
상기 (b2) 단계에서, 상기 염은 인산염, 칼슘염, 나트륨염 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나, 바람직하게는, 상기 (b1) 단계와의 호환성, 상용성을 고려하여 인산염일 수 있으나, 이에 한정되는 것은 아니다.
상기 (b4) 단계에서, 상기 가교제는 포름알데히드(formaldehyde), a-하이드록시아디프알데히드(a-hydroxyadipaldehyde), 글루타르알데히드(glutaraldehyde), EDC/NHS(1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide/N-hydroxy succinimide) 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나일 수 있고, 바람직하게는, 글루타르알데히드일 수 있으나, 이에 한정되는 것은 아니다.
상기 천연 고분자 수지는 콜라겐, 키토산, 히알루론산, 카르복시메틸셀룰로오스, 헤파란설페이트, 덱스트란, 알지네이트 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나, 바람직하게는, 콜라겐일 수 있으나, 이에 한정되는 것은 아니다. 상기 콜라겐은 porcine skin, porcine pericardium, porcine tendon, bovine skin, bovine pericardium, bovine tendon 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나로부터 추출한 것일 수 있으나, 이에 한정되는 것은 아니다.
상기 (b5) 단계에서, 상기 세척은 정제수, PBS(phosphate buffered saline), 에탄올 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나를 사용하여 수행될 수 있다.
상기 제2 고분자의 필름의 제조방법은, 예를 들어, 천연 고분자 수지와 산 용액을 0.3~3w/v%(w/v%=mg/ml% 또는 kg/L%)의 농도로 혼합하는 단계; 혼합된 상기 용액을 염을 포함하는 용액으로 20~37℃에서 12~48시간 동안 섬유화시키는 단계; 섬유화된 생성물을 24~72시간 동안 동결건조시켜 베이스 필름을 얻는 단계; 가교제를 이용하여 상기 베이스 필름을 멤브레인을 6~48시간 동안 화학적으로 가교시키는 단계; 가교 후 세척액을 이용하여 20~37℃에서 12~24시간 동안 1~5회 세척하는 단계; 20~37℃ 에서 3~24시간 동안 풍건하여 건조시키는 단계; 상기 건조된 필름를 20~80℃에서 30분~12시간 동안 가압하는 단계;를 포함할 수 있으나, 각 단계 및 조건이 이에 한정되는 것은 아니다.
제1 및 제2 고분자 필름의 적층 및 접합
상기 (C) 단계에서는, 상기 제1 고분자 필름 및 천연 고분자 수지를 포함하는 제2 고분자 필름을 교차적으로 적층 및 접합하여 최종 제품인 치조골 재생 차폐막을 제조할 수 있다.
상기 (C) 단계는, (c1) 상기 제1 및 제2 고분자 필름을 교차적으로 적층하는 단계; 및 (c2) 상기 (c1) 단계의 생성물을 30~80℃의 온도로 가열하고, 3~30mPa의 압력으로 접합하는 단계를 포함할 수 있다. 이 때, 상기 (c2) 단계의 가열 온도는 30℃ 이상, 40℃ 이상, 또는 50℃ 이상이고, 80℃ 이하, 70℃ 이하, 또는 60℃ 이하일 수 있다. 또한, 상기 (c2) 단계에서 가해지는 압력은 3 mPa 이상, 7 mPa 이상, 또는 11 mPa 이상이고, 30 mPa 이하, 26 mPa 이하, 또는 22 mPa 이하일 수 있다.
또한, 상기 (c1) 단계에서, 상기 제1 고분자 필름의 적어도 일 면, 바람직하게는, 양 면에 천연 고분자 수지를 포함하는 상기 제2 고분자 필름을 적층 및 접합할 수 있다.
도 3은 본 발명의 일 실시예에 따른 합성 고분자 필름 및 천연 고분자 필름의 적층 및 접합방법을 도식화한 것이다. 도 3을 참고하면, 상기 제1 및 제2 고분자 필름의 적층 및 접합방법은, 예를 들어, 제1 및 제2 고분자 필름을 2~10층으로 교차 적층하고, 30~80℃로 가열하는 단계; 3~30mPa의 압력으로 가압하여 제1 및 제2 고분자 필름을 접합하는 단계; 접합된 차폐막을 3분~3시간 동안 냉각시키는 단계; 냉각된 차폐막을 적절한 크기로 재단하는 단계; 및 재단된 차폐막을 멸균한 후 포장하는 단계;를 포함할 수 있으나, 각 단계 및 조건이 이에 한정되는 것은 아니다.
이하, 본 발명의 실시예에 관하여 상세히 설명하기로 한다.
실시예 1
콜라겐 필름의 제조
인산 수용액 1L에 콜라겐 20g을 용해시키고, 이를 PBS(phosphate buffered saline) 용액으로 25℃에서 20시간 동안 처리하여 상기 콜라겐을 섬유화시킨 다음, 40시간 동안 동결건조시켜 시트를 제조하고, 가교제인 글루타르알데히드를 이용하여 상기 시트를 화학적으로 가교시켰다. 가교 후 시트에 잔류하는 가교제를 제거하기 위해 시트를 세척하고, 25℃에서 12시간 동안 바람을 이용하여 온풍 건조시켰다. 건조된 시트를 50℃에서 2시간 동안 가압하여 두께가 0.2mm인 콜라겐 필름을 얻었다.
고분자 필름의 제조
폴리카프로락톤(PCL)을 직경이 0.2mm인 3D 프린터 노즐에 공급하고, 상기 노즐을 130℃로 가열하여 폴리카프로락톤을 용융시킨 후, 300kPa의 압력으로 사출하여 평균 기공 크기가 1.2mm인 베이스 필름을 제조하였다. 사출된 베이스 필름을 25℃에서 5분 간 냉각한 다음, 30mPa의 압력으로 가압하여 평균 기공 크기가 0.5mm인 고분자 필름을 얻었다.
치조골 재생 차폐막의 제조
상기 고분자 필름의 양 면에 상기 콜라겐 필름을 적층한 다음, 판상형 열압착기(큐머시스, QM900M)로 80℃ 및 20mPa의 조건하에서 5분 간 열압착하여 콜라겐/폴리카프로락톤/콜라겐(Col/PCL/Col)의 3층 구조를 가지며 평균 기공 크기 및 두께가 각각 0.2mm 및 0.4mm인 치조골 재생 차폐막을 제조하였다.
실시예 2
고분자 필름의 제조 시 3D 프린터 노즐의 직경을 0.4mm로 변경한 것을 제외하면, 상기 실시예 1과 동일한 방법으로 치조골 재생 차폐막을 제조하였다.
실시예 3
치조골 재생 차폐막의 제조 시 열압착기의 압력을 7mPa로 변경하여 치조골 재생 차폐막의 평균 기공 크기를 0.4mm로 조절한 것을 제외하면, 상기 실시예 1과 동일한 방법으로 치조골 재생 차폐막을 제조하였다.
실시예 4
치조골 재생 차폐막의 제조 시 상기 고분자 필름의 일 면에 상기 콜라겐 필름을 적층한 것을 제외하면, 상기 실시예 1과 동일한 방법으로 치조골 재생 차폐막을 제조하였다.
실시예 5
치조골 재생 차폐막의 제조 시 열압착기의 온도 및 압력을 각각 50℃ 및 5mPa로 변경한 것을 제외하면, 상기 실시예 1과 동일한 방법으로 치조골 재생 차폐막을 제조하였다.
실시예 6
치조골 재생 차폐막의 제조 시 열압착기의 온도 및 압력을 각각 50℃ 및 30mPa로 변경한 것을 제외하면, 상기 실시예 1과 동일한 방법으로 치조골 재생 차폐막을 제조하였다.
실시예 7
치조골 재생 차폐막의 제조 시 열압착기의 압력을 5mPa로 변경한 것을 제외하면, 상기 실시예 1과 동일한 방법으로 치조골 재생 차폐막을 제조하였다.
실시예 8
치조골 재생 차폐막의 제조 시 열압착기의 압력을 30mPa로 변경한 것을 제외하면, 상기 실시예 1과 동일한 방법으로 치조골 재생 차폐막을 제조하였다.
비교예 1
기존에 상용화되어 있는 콜라겐 소재의 치조골 재생 차폐막(Bio-Gide
TM, Geistlich社, 두께 0.5mm)을 준비하였다.
비교예 2
기존에 상용화되어 있는 콜라겐 소재의 치조골 재생 차폐막(OssGuide
TM, SK바이오랜드社, 두께 0.5mm)을 준비하였다.
비교예 3
고분자 필름의 제조를 다음과 같이 변경한 것을 제외하면, 상기 실시예 1과 동일한 방법으로 상기 실시예 1과 동일한 방법으로 치조골 재생 차폐막을 제조하였다.
PLGA를 사출성형기(Fanuc, i30A-injection molding machine)의 배럴 내로 이동하기 전에 200℃온도로 용융시킨 다음, 시트 형태의 몰딩부에 넣고 1,800bar의 압력 및 200℃의 온도로 사출성형하였다. 사출성형된 PLGA 를 25℃에서 10초 간 냉각한 다음, 몰딩부에서 떼어내어 PLGA 판재를 얻었다.
비교예 4
고분자 필름의 제조 시 베이스 필름을 2mPa의 압력으로 가압한 것을 제외하면, 상기 실시예 1과 동일한 방법으로 치조골 재생 차폐막을 제조하였다.
비교예 5
고분자 필름의 제조 시 베이스 필름을 60mPa의 압력으로 가압한 것을 제외하면, 상기 실시예 1과 동일한 방법으로 치조골 재생 차폐막을 제조하였다.
상기 실시예 및 비교예에 따라 제조된 각 구성 및 치조골 재생 차폐막의 구조적 특징을 하기 표 1 내지 표 3에 나타내었다.

실험예 1: 공간유지능 및 성형성 평가
실시예 및 비교예에 따른 치조골 재생 차폐막의 공간유지능 및 성형성을 비교, 평가하였다. 공간유지능은 도4의 push-pull gauge(HG-500)를 이용하여 굽힘강도를 통해 평가하였고, 성형성은 도 5의 만능 재료 시험기(INSTRON 3365)를 이용하여 탄성회복률을 통해 평가하였다.
하기 표 4 및 도 6은 실시예 및 비교예에 따른 치조골 재생 차폐막의 공간유지능 및 성형성 평가결과를 나타낸다.

도 6을 참고하면, 기존 콜라겐 소재의 치조골 재생 차폐막인 비교예 1, 2는 모두 1N 이하의 낮은 굽힘강도를 나타내었으나, 실시예 1~4에 따른 차폐막의 굽힘강도는 9N 이상으로 나타나 비교예 1, 2에 비해 향상된 고정력을 구현할 수 있다.
또한, 적층된 차폐막의 접합 시 압력 및 온도를 조절하여 차폐막의 기공 크기를 미리 정해진 범위로 조절한 실시예 5~8의 경우 12N 이상의 굽힘강도를 가짐과 동시에 90% 이하의 탄성회복률을 나타내어 상온에서 충분한 성형성을 가짐을 알 수 있다.
반면, 비교예 3~5의 경우 공간유지능 및 성형성을 균형적으로 구현하지 못한 것으로 나타났다. 구체적으로, D
2/D
1의 값이 0.5 초과인 비교예 3, 4의 차폐막은 성형성은 양호하나 공간유지능이 현저히 불량하며, D
2/D
1의 값이 0.1 미만인 비교예 5의 차폐막은 공간유지능은 양호하나 탄성회복률이 100%로 나타나 성형성이 현저히 불량하다.
상기 결과를 통해, 실시예 1~8, 특히, 실시예 5~8의 차폐막은 비교예 1~5의 차폐막에서 나타나는 공간유지능 및 성형성 간의 트레이드-오프(trade-off)를 유효 적절하게 해소하여 이들을 동시에 균형적으로 구현할 수 있음을 알 수 있다.
실험예 2: 차폐막 유효성 평가
실시예 및 비교예에 따른 치조골 재생 차폐막의 영양물질 투과능을 확인하기 위해 도 7과 같이 diffusion cell을 이용하여 실험을 진행하였다. 용기의 상부층에 485nm에서 강한 파장을 나타내는 BSA-FITC 용액을 넣고 차폐막을 투과하여 아래층으로 이동하는 정도를 흡광도 측정기를 이용하여 분석하였고, 그 결과를 도 8에 나타내었다.
도 8을 참고하면, 기존 콜라겐 소재의 치조골 재생 차폐막인 비교예 1, 2에 비해 실시예 1, 2의 차폐막은 영양물질에 대한 투과능이 동등하거나 그 이상이며, 특히, 약 40시간이 경과한 이후 투과능의 차이가 더 현저하게 확인된다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다. 예를 들어, 단일형으로 설명되어 있는 각 구성 요소는 분산되어 실시될 수도 있으며, 마찬가지로 분산된 것으로 설명되어 있는 구성 요소들도 결합된 형태로 실시될 수 있다.
본 발명의 범위는 후술하는 청구범위에 의하여 나타내어지며, 청구범위의 의미 및 범위 그리고 그 균등 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (13)
- (A) 합성 고분자 수지를 용융 및 성형하여 베이스 필름을 제조하는 단계;(B) 상기 베이스 필름을 가압하여 제1 고분자 필름을 제조하는 단계; 및(C) 상기 제1 고분자 필름 및 천연 고분자 수지를 포함하는 제2 고분자 필름을 교차적으로 적층 및 접합하는 단계;를 포함하고,하기 조건을 만족하는, 치조골 재생 차폐막의 제조방법:상기 베이스 필름 및 상기 제1 고분자 필름의 평균 기공 크기는 각각 D 1 및 D 2이고, 0.1 ≤ D 2/D 1 ≤ 0.5 이다.
- 제1항에 있어서,상기 합성 고분자 수지는 PLA(polylactic acid), PGA(polyglycolic acid), PLGA(poly lactic-co-glycolic acid), PLLA(poly(L-lactic acid)), PCL(polycaprolactone), PLCL(poly lactide-co-caprolactone), PHB(polyhydroxybutyrate), PHV(polyhydroxyvalerate), PDO(polydioxanone), PTMC(Polytrimethylenecarbonate) 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나인, 치조골 재생 차폐막의 제조방법.
- 제1항에 있어서,상기 (A) 단계에서, 상기 성형은 적층 가공(additive manufacturing)으로 수행되는, 치조골 재생 차폐막의 제조방법.
- 제3항에 있어서,상기 적층가공은 하나 이상의 노즐을 포함하는 FDM(Fused Deposition Modeling) 방식의 3D 프린터를 이용하여 수행되는, 치조골 재생 차폐막의 제조방법.
- 제4항에 있어서,상기 노즐의 직경은 0.1~0.5mm인, 치조골 재생 차폐막의 제조방법.
- 제1항에 있어서,상기 (B) 단계에서, 상기 가압은 3~50mPa의 압력으로 수행되는, 치조골 재생 차폐막의 제조방법.
- 제1항에 있어서,상기 천연 고분자 수지는 콜라겐, 키토산, 히알루론산, 카르복시메틸셀룰로오스, 헤파란설페이트, 덱스트란, 알지네이트 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나인, 치조골 재생 차폐막의 제조방법.
- 제1항에 있어서,상기 제2 고분자 필름은 하기 단계를 포함하는 방법에 따라 제조되는, 치조골 재생 차폐막의 제조방법:(b1) 제2 고분자 수지를 산 용액에 용해시키는 단계;(b2) 상기 (b1) 단계의 생성물에 염을 투입하여 상기 제2 고분자 수지를 섬유화하는 단계;(b3) 상기 (b2) 단계의 생성물을 건조시키는 단계;(b4) 상기 (b3) 단계의 생성물에 가교제를 투입하여 상기 제2 고분자 수지를 가교시키는 단계; 및(b5) 상기 (b4) 단계의 생성물을 세척 및 건조시킨 후 가압하는 단계.
- 제8항에 있어서,상기 산 용액은 인산, 황산, 염산, 질산, 아세트산 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나를 포함하는, 치조골 재생 차폐막의 제조방법.
- 제8항에 있어서,상기 염은 인산염, 칼슘염, 나트륨염 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나인, 치조골 재생 차폐막의 제조방법.
- 제8항에 있어서,상기 가교제는 포름알데히드(formaldehyde), a-하이드록시아디프알데히드(a-hydroxyadipaldehyde), 글루타르알데히드(glutaraldehyde), EDC/NHS(1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide/N-hydroxy succinimide) 및 이들 중 2 이상의 조합으로 이루어진 군에서 선택된 하나인, 치조골 재생 차폐막의 제조방법.
- 제1항에 있어서,상기 (C) 단계는 하기 단계를 포함하는, 치조골 재생 차폐막의 제조방법:(c1) 상기 제1 및 제2 고분자 필름을 교차적으로 적층하는 단계; 및(c2) 상기 (c1) 단계의 생성물을 30~80℃의 온도로 가열하고, 3~30mPa의 압력으로 접합하는 단계.
- 제1항에 있어서,상기 (C) 단계에서, 상기 제1 고분자 필름의 적어도 일 면에 상기 천연 고분자 수지를 포함하는 상기 제2 고분자 필름을 적층 및 접합하는, 치조골 재생 차폐막의 제조방법.
