편도 오가노이드의 제조방법 및 이의 용도
편도 오가노이드의 제조방법 및 이의 용도에 관한 것이다.
편도선(편도)은 인두 편도선, 두 개의 관 편도선, 두 개의 구개 편도선 및 설측 편도선을 포함한 발다이어 편도선환(Waldeyer 's tonsillar ring)으로 알려진 호흡기 및 소화관의 관문에서 점막과 관련된 림프 조직의 모음이다. 각 편도선은 림프 모낭을 덮고 있는 상피로 구성되며 박테리아나 바이러스와 같은 섭취 또는 흡입 된 병원체에 대한 첫 번째 방어선 역할을 한다. 편도 상피의 선와(crypt)는 입을 통해 유입되는 병원체를 인식하고 신호를 전송하여 B 세포, T 세포, 대 식세포 및 수지상 세포에 의한 면역 반응을 자극한다. 편도선은 세균성 및 바이러스 성 감염에 취약하여 감염될 시 편도선염, 편도선 비대 및 인두 암으로 이어질 수 있다.
이전 연구에 따르면 편도선의 세균성 및 바이러스성 감염은 만성 편도선 장애의 주요 원인이며 약리학적, 영양소 및 다형성 요인도 만성 편도선 장애의 중요한 원인으로 간주된다. 그러나 병리 생리학은 여전히 불분명하고 논쟁의 여지가 있다. 따라서 이런 병리학적 연구를 위하여 인체 편도를 재현하기에 적합한 체외 모델이 없으며, 실험 동물 모델을 사용할 수도 없으므로 편도에 대한 병리 생리학을 연구하기가 어렵다.
이러한 문제점에 기하여 인간 표본 또는 상피 줄기 세포로부터의 장기 3D 배양에서의 최근의 진보는 인간 상피 오가노이드의 생성을 가능하게 하였다. 이 3D 배양 모델은 인간 상피 오가노이드가 모든 조직-특이적 상피 세포 유형 및 상피 줄기 세포로부터 분화되고 증식 된 구조를 포함하기 때문에 인간 생체 내 생리학을 나타낼 수 있다. 지금까지, 이 방법은 상피 조직 관련 질환 모델을 생성하고 상피 생물학을 연구하기 위해 광범위하게 사용되어오고 있으나(등록 특허 10-18719490000), 세포 구성 및 미세한 구조와 같은 편도 상피의 필수 기능을 유지하는 편도 오가노이드의 제조 방법은 아직 미미한 실정이다.

다른 양상은 상기 인간 편도 오가노이드 제조용 배지는 HGF, FGF10, FGF2, FGF 베이직, PGE2, A83-01, 니코틴아미드(Nicotinamide), 노긴(Noggin), R-스폰딘1(R-spondin1), 항생-항진균제, 글루타맥스(glutamax), B27, SB202190, 페니실린 스트렙토마이신, EGF 및 HEPES를 포함하는 것인, 편도 오가노이드 제조용 배지를 제공하는 것이다.

본 명세서 내 용어 "오가노이드(organoid)"는 3D 입체구조를 가지는 세포를 의미하며, 동물 등에서 수집, 취득하지 않은 인공적인 배양 과정을 통해 제조한 간과 유사한 모델을 의미한다. 이를 구성하는 세포의 유래는 제한되지 않는다. 2D 배양과는 달리, 3D 세포 배양은 체외에서 세포가 모든 방향으로 성장할 수 있는바, 상기 오가노이드는 생체 내에서 상호작용 하고 있는 장기를 유사하게 모사하여 질병의 치료제 개발 등에 이용될 수 있다. 또한, 편도 오가노이드를 배양함으로써 기존의 동물 모델의 한계인 인간의 생리학적 및 진화적 차이를 극복할 수 있다.
본 명세서 내에서 “편도”란 편도, 편도선 또는 편도상피를 의미하며, 이는 림프소절의 집합체이고, 인두편도 (아데노이드, adenoid, pharyngeal tonsil), 귀인두관편도(tubal tonsil), 목구멍편도(구개편도, palatine tonsil) 또는 혀편도 (설편도, lingual tonsil)를 모두 포함한다.
본 명세서 내에서 “편도 오가노이드”란 상기 편도, 편도선 또는 편도상피를 포함하는 편도 유래 세포로부터 유래된 오가노이드를 의미한다. 상기 편도 오가노이드는 편도 상피의 특성을 그대로 발현할 수 있다. 예컨대, 상기 편도 오가노이드는 주화성 분석, qRT-PCR, 싸이토카인 분석 및 면역형광분석법 등의 분석 방법을 통하여 편도 상피의 특성을 나타내는 것으로 공지된 마커들이 발현되는 것일 수 있다.
본 발명에서 용어, "배지"는 인 비트로(in vitro)에서 줄기세포의 성장, 생존 및 분화를 지지할 수 있게 하는 배지를 의미하고, 해당분야에서 사용되는 줄기세포의 배양 및 분화에 적절한 통상의 배지를 모두 포함한다. 세포의 종류에 따라 해당분야의 기술적 수준에서 배지의 종류와 배양 조건을 선택할 수 있다. 배양에 사용되는 배지는 구체적으로 세포 배양 최소 배지(cell culture minimum medium, CCMM)로, 일반적으로 탄소원, 질소원 및 미량원소 송분을 포함한다. 상기 세포 배양 최소 배지에는 예를 들어, DMEM(Dulbecco's Modified Eagle's Medium), MEM(Minimal essential Medium), BME(Basal Medium Eagle), RPMI1640, F-10, F-12, α-MEM(α-Minimal essential Medium), GMEM(Glasgow's Minimal essential Medium), IMDM(Iscove's Modified Dulbecco's Medium) 등이 있으나, 이에 제한되는 것은 아니다. 또한, 상기 배지는 페니실린(penicillin), 스트렙토마이신(streptomycin), 겐타마이신(gentamicin) 또는 이들의 2 이상의 혼합물 등의 항생제를 포함할 수 있다.
본 명세서 내에서 용어 “TFM”이란 편도 형성 배지(Tonsil Formation Medium)를 의미하며, 인간 결장 오가노이드 배지 (HCM) 또는 인간 전립선 오가노이드 배지 (HPM)에서 EGF(epidermal growth factor)를 제거한 조성물을 의미한다.
다른 양상은 상기 인간 편도 오가노이드 제조용 배지는 HGF, FGF10, FGF2, FGF 베이직, PGE2, A83-01, 니코틴아미드(Nicotinamide), 노긴(Noggin), R-스폰딘1(R-spondin1), 항생-항진균제, 글루타맥스(glutamax), B27, SB202190, 페니실린 스트렙토마이신, EGF 및 HEPES를 포함하는 것인, 편도 오가노이드 제조용 배지를 제공하는 것이다.
다른 양상은 상기 HGF는 전체 편도 오가노이드 제조용 배지 조성물 중 20 내지 80ng/ml의 농도로 포함하는 것인, 편도 오가노이드 제조용 배지를 제공하는 것이다.
일 구체예에 있어서, 상기 배지 조성물 중 HGF의 총 농도는 10 내지 100ng/ml, 10 내지 90ng/ml, 10 내지 80ng/ml, 10 내지 70ng/ml, 10 내지 60ng/ml, 20 내지 80ng/ml, 20 내지 70ng/ml, 20 내지 60ng/ml, 30 내지 80ng/ml, 30 내지 70ng/ml, 30 내지 60ng/ml, 40 내지 80ng/ml, 40 내지 80ng/ml, 40 내지 70ng/ml, 40 내지 50ng/ml일 수 있다.
다른 양상은 상기 편도 오가노이드 제조용 배지에 의하여 제조된 편도 오가노이드는 CD44, 신경 성장 인자 수용체 (Nerve growth factor receptor), 시토 케라틴 5 (Cytokeratin 5), 시토 케라틴 13 (Cytokeratin 13), 인테그린 알파 6 (Integrin alpha 6) 및 상피 세포 접착 분자 (epithelial cell adhesion molecule) 유전자를 발현하는 것인, 편도 오가노이드 제조용 배지를 제공하는 것이다.
다른 양상은 상기 편도 오가노이드 제조용 배지에 의하여 제조된 편도 오가노이드는 TLR(toll-like receptor) 2, 3, 4 및 6을 발현하는 것인, 편도 오가노이드 제조용 배지를 제공하는 것이다.
다른 양상은 상기 편도 오가노이드 제조용 배지에 의하여 제조된 편도 오가노이드는 2세대 이상의 계대배양이 가능한 것인, 편도 오가노이드 제조용 배지를 제공하는 것이다.
본 발명에서 용어, "계대"는 세포, 구체적으로 오가노이드를 건강한 상태로 지속적으로 장기간 배양하기 위해 세포의 대를 계속 이어서 배양하는 방법에서, 배양용기를 교체하는 것 또는 세포군을 나누어 배양하는 것을 의미한다. 한 차례 배양용기 교체 또는 세포군을 나누어 배양하는 것을 1 계대라고 한다. 본 발명에서 상기 계대는 세대와 혼용되어 사용될 수 있다.
일 실시예에 있어서 상기 편도 오가노이드 제조용 배지에 의하여 제조된 편도 오가노이드는 실제 인간 편도 오가노이드와 동일한 마커 및 TLR을 발현하였으며, 1세대부터 5세대, 즉 1계대부터 5계대까지 총 50여일까지 정상적인 배양 및 분화가 가능함을 확인하였다.
또 다른 양상은 TFM에서 HGF를 추가로 포함하고 4-[4-(4-Fluorophenyl)-5-(4-pyridinyl)-1H-imidazol-2-yl]phenol를 포함하지 않게 하여 편도 오가노이드 제조용 배지를 제조하는 단계; 및
상기 편도 오가노이드 제조용 배지에 배양하는 단계를 포함하는 편도 오가노이드의 제조 방법을 제공하는 것이다.
일 구체예에 있어서 상기 편도 오가노이드의 크기는 100 내지 300, 150 내지 300, 200 내지 300, 100 내지 250, 100 내지 200, 150 내지 250 마이크로미터일 수 있다.
또 다른 양상은 편도 오가노이드에 염증 유발 물질을 접촉시키는 단계;
상기 염증 유발 물질이 처리된 편도 오가노이드에 시험 물질을 접촉시키는 단계;
상기 시험 물질을 접촉한 편도 오가노이드의 사이토카인 발현량이 대조군에 비하여 감소했을 경우 해당 시험 물질을 항염제로 분류하는 단계를 포함하는 편도 오가노이드 항염제의 스크리닝 방법을 제공하는 것이다.
상기 염증 유발 물질은 편도 오가노이드에 염증을 유발하는 물질로서, 예를 들어, 임의의 물질(substance), 분자(molecule), 원소(element), 화합물(compound), 실재물(entity) 또는 이들의 조합을 포함할 수 있다. 예를 들어, 단백질, 폴리펩티드, 저분자 유기화합물(small organic molecule), 다당류(polysaccharide), 폴리뉴클레오티드 등을 포함할 수 있다. 또한, 자연 산물(natural product), 합성 화합물 또는 화학 화합물 또는 2개 이상의 물질의 조합일 수 있다. 더 구체적으로는 LPS, TNF-알파 및 기타 알레르기 유발 물질 등일 수 있다.
본 발명에서 "사이토카인(cytokine)"은 세포 신호 전달에서 중요한 역할을 하는 단백질을 의미한다. 사이토카인의 방출은 사이토카인 주위의 세포의 행동에 영향을 미친다. 구체적으로, 사이토카인은 면역조절제로서 자가분비 신호, 파라크린 신호 전달 (paracrine signaling) 및 내분비 신호 전달에 관여한다. 또한, 사이토카인은 대식세포, B 림프구, T 림프구 및 비만 세포와 같은 면역세포뿐만 아니라 내피 세포, 섬유 아세포 및 다양한 기질 세포를 포함하여 광범위한 세포에 의해 생성된다. 사이토카인은 체액성 면역 반응과 세포 기반 면역 반응 사이의 균형을 조절하고 특정 세포 집단의 성숙, 성장 및 반응을 조절하는 역할을 한다.
상기 염증 유발 물질이 처리되어 발현량의 변화를 측정하는 대상인 사이토카인은, IL-2(interleukin-2), IL-8(interleukin-8), TNFα(Tumor Necrosis Factor-α), IL-22(interleukin-22), IL-6(interleukin-6), IL-1β(interleukin-1β), IL-11(interleukin-11), EGF(epidermal growth factor), OSM(oncostatin M), IL-10(interleukin-10), IL-1α(interleukin-1α), CXCL1(Chemokine (C-X-C motif) ligand 1), CXCL8(Chemokine (C-X-C motif) ligand 2), IL-1ra(interleukin-1ra), MIF(migration inhibition factor) 또는 PAI-1(plasminogen activator inhibitor 1)을 포함한다.
상기 시험 물질은 편도 오가노이드의 항염 작용에 효과를 나타낼 것으로 기대되는 물질로서, 예를 들어, 임의의 물질(substance), 분자(molecule), 원소(element), 화합물(compound), 실재물(entity) 또는 이들의 조합을 포함할 수 있다. 예를 들어, 단백질, 폴리펩티드, 저분자 유기화합물(small organic molecule), 다당류(polysaccharide), 폴리뉴클레오티드 등을 포함할 수 있다. 또한, 자연 산물(natural product), 합성 화합물 또는 화학 화합물 또는 2개 이상의 물질의 조합일 수 있다.
본 명세서 내 용어, “대조군”이란, 염증 유발 물질을 처리 후 항염용 시험 물질을 처리하지 않은 편도 오가노이드를 의미하며, 항염용 시험 물질을 처리한 편도 오가노이드와 사이토카인 분비량을 비교하기 위해 사용된다. 사이토카인 분비량의 감소량은 시험 물질의 항염 효과와 비례한다. 이때, 상기 사이토카인 분비량이 대조군에 비해 감소한 경우, 상기 시험 물질이 편도 오가노이드의 항염증용 약물인 것으로 판단할 수 있다.
일 양상에 따른 편도 오가노이드 제조용 배지에 의해 제조된 편도 오가노이드는 안정적인 장기 계대배양이 가능하며, 실제 편도 상피와 동일한 마커 및 TLR을 발현하여 신체 내와 유사한 면역 반응을 나타낼 수 있다.

이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
모든 통계적 분석은 GraphPad Prism 5를 사용하여 수행 되었다. 정량의 통계 분석은 두 그룹의 비교를 위해 쌍을 이루지 않은 양측 t- 검정에 의해, 그리고 다중 그룹의 비교를 위해 Newman-Keuls 다중 비교 테스트를 통한 일원 분산 분석에 의해 수행되었다. 통계적 유의성에 대한 역치는 P <0.05였다. 표본 크기를 미리 결정하기 위해 통계적 방법을 사용하지 않았다. 실험의 샘플 크기는 NIS 이미징 소프트웨어를 사용하여 추정되었다. 표본 크기, 통계 분석에 대한 모든 세부 사항은 평균 ± SEM이다.
실시예 1: 편도 오가노이드의 생성 및 편도 형성 배지(Tonsil Formation Medium, TFM)의 제조
인간의 구개골 편도(palatine tonsil)에서 오가노이드를 생성하기 위해, 인간의 결장 오가노이드 배지(human colon organoid medium, HCM)나 인간 전립선 오가노이드 배지(human prostate organoid medium, HPM)를 사용할 때의 통상적인 배양 프로토콜을 수정했으며 하기와 같다.
편도 상피 조직을 가위로 잘게 자른 다음 1X 인산염 완충 용액 (PBS, Welgen)을 사용하여 세척한다. 잘게 잘린 편도 상피 조직을 37 ℃에서 2 시간 동안 500ug / ml 콜라게나제 type2 (Gibco)를 포함하는 Advanced DMEM / F12 (Gibco)를 사용하여 효소적으로 해리시켰다.
효소적으로 해리 된 인간 구개골 세포를 수집하고, 펠릿 화하고, 마트리겔(matrigel, Corning)에 임베딩한 후 48- 웰 플레이트 (SPL)에 시딩 하였다. 편도 세포는 1X 항생-항진제 (Thermo Fisher Scientific), 1X 글루타맥스(Thermo Fisher Scientific), 1X B27 (Invitrogen), 10 % R-스폰딘-1(R-spondin-1, RSPO1) 조건 배지 및 성장 인자를 포함하는 Advanced DMEM / F12 (Gibco)에서 배양되었다. 상기 R-spondin1 조건 배지의 추가적인 조성물은 다음과 같다: FGF10 (50 ng/ml), FGF2(= FGF basic, 20 ng/ml), PGE2 (10 μM), A83-01 (200 μM), SB202190 (=4-[4-(4-Fluorophenyl)-5-(4-pyridinyl)-1H-imidazol-2-yl]phenol, 10 μM), 니코틴아미드 (Nicotinamide, 10 mM), 노긴 (Noggin, 100 ng/ml).
뉴레글린-1 (Neuregulin-1, NRG1, Peprotech)은 인두 편도에만 첨가되었다. 계대 배양 후, 10uM Y-27632 (Tocris)를 2 일 동안 배양 배지에 첨가 하였다.
효소적으로 해리 된 인간 구개골 세포를 마트리겔에 현탁시키고, 도 1a에 나타난 바와 같이, EGF, 노긴 (Noggin), FGF2, FGF10 및 R-스폰딘-1을 함유하는 인간 결장 오가노이드 배지 (HCM) 또는 인간 전립선 오가노이드 배지 (HPM)에서 성장시켰다. 도 1b에 나타난 바와 같이, 10일째에 HPM에서 배양 된 구개골 편도 세포는 HCM에 배양된 구개골 편도 세포에 비하여, 많은 수의 3D 편도 오가노이드를 생성했다. 가장 생산적인 배지 조성물을 스크리닝하기 위해, 해리 된 편도 세포를 HPM의 각 성분을 제거하거나 첨가함으로써 생성 된 다양한 조건에서 플레이팅 하였다. 도 1c에 나타난 바와 같이 HPM에서 EGF를 제거한 배지는 편도 오가노이드 배양에 가장 효과적이었으며 이를 편도 형성 배지(TFM)로 명명했다.
실시예 2: 편도 오가노이드의 장기 계대배양 및 발현마커의 확인
위, 장, 자궁 내막 및 간과 같은 성인 상피 조직의 오가노이드는 몇 개월에 걸쳐 계대배양하며 성장할 수 있다. TFM에서 생장한 편도 오가노이드는 10 일마다 1 : 3의 계대 비율로 배양되었으나, 이들 오가노이드를 2계대 이상 계대 배양하는 것은 불가능하였다.
띠라서 본 실시예에서는 편도 오가노이드가 안정적으로 장기간 계대배양할 수 있게끔 하는 배지 조성물을 확인하기 위하여 다양한 배지 조성물에 편도 오가노이드를 배양하고 qRT-PCR 을 수행하였다.
qRT-PCR은 하기와 같이 실시되었다. 오가노이드를 마트리겔에서 분리하고 PBS로 세척하고 스핀다운 한 후, 프로토콜에 따라 RNA 추출 키트 (Bioneer)를 사용하여 RNA 분리를 수행 하였다. TP950 Thermal Cycler Dice Real Time SysTEM III(TAKARA)을 사용하여 샘플에 대해 qPCR을 수행 하였다. 표 1에 사용 된 프라이머 목록이 포함되어 있다.

장기 계대 배양 조건을 개선하기 위해 배지 조성물에 HGF 및/또는 SB의 제거 및/또는 첨가를 진행 하였다. HGF의 첨가는 50ng/ml의 농도로 이루어졌다.
도 2a에 나타난 바와 같이, 장기 계대배양(long-term passage)하기 위해 TFM에서 배양 된 오가노이드는 편도 상피와 같은 계층적 구조를 나타냈으나 2번째 계대배양에서 생장이 제한되었다. 또한 p38 경로를 활성화시키기 위한 SB의 제거 및 HGF의 첨가는 초기에 확립 된 오가노이드의 형성 및 생장 효율을 변화시키지 않았다. 그러나, HGF를 첨가하고 SB를 제거한 TFM에서 배양 된 오가노이드는 5 계대 또는 60 일 이상으로 유지될 수 있었다.
상기 장기 계대배양을 가능케 한, HGF를 첨가하고 SB를 제거한 TFM을 편도 오가노이드 제조용 배지(편도 확장 배지, Tonsil expansion medium, TEM)로 정의하였다. TEM에서 배양 된 오가노이드를 편도 상피 조직 마커인 CD44, 신경 성장 인자 수용체 (Nerve growth factor receptor, NGFR), 시토케라틴 5 (Cytokeratin 5, CK5), 시토케라틴 13 (Cytokeratin 13, CK13), 인테그린 알파 6 (Integrin alpha 6, ITGA6) 및 상피 세포 접착 분자 (epithelial cell adhesion molecule, EpCAM)를 통하여 유전자 발현 패턴을 분석 하였다. 오가노이드의 편도 상피 조직 마커의 mRNA 수준을 5 내지 15 일 동안 모니터링하고, 실제 인간 편도 조직의 상피의 마커와 비교 하였다.
도 2b에 나타난 바와 같이, 편도 상피 조직 마커인 CD44, NGFR, CK5, CK13, ITGA6 및 EpCAM은 모두 TEM- 성장(grown) 오가노이드에서 발현되었다. 오가노이드에서의 NGFR 발현은 편도 상피보다 낮았지만, EpCAM, CD44, KRT13, KRT5 및 ITGA6 발현은 편도 상피보다 높았다. 특히, 세포 분화 마커 CK13의 수준은 배양 기간이 길수록 증가 하였다.
이러한 실시예에 의하여, HGF의 추가 및 SB을 제거한 TFM으로 정의되는 인간 편도 확장 배지 (TEM)에 배양된 편도 오가노이드는 장기 계대 배양이 가능함을 확인할 수 있었다.
실시예 3: 편도 오가노이드의 구조적 특징의 확인
구개골 편도의 관강 표면(luminal surface)은 기저막 상에 층으로 배열 된 전형적인 비-각질화 층화 편평 상피로 덮여있다. 기저막에 가까운 세포일수록 자가 재생 특성이 강하고 입방형 형태가 많은 반면, 표면 근처의 세포는 평평한 모양으로 분화되는 특성을 가지고 있다.
상기 실시예 2의 TEM에서 배양된 편도 오가노이드의 구조적 특징을 확인하기 위하여, H&E 염색 및 면역 형광법을 실시하였다.
H&E 염색은 다음과 같이 이루어졌다. 편도 오가노이드를 4 % 파라 포름 알데히드 (PFA, bio solution)에 고정시킨 다음 편도 오가노이드를 Cryo 몰드 (10 x 10 x 5 mm, k4555, SungWon Medical Company)에서 2 % 아가로스 겔과 혼합하고 실온에서 고형화시켰다. 그런 다음 각 단계마다 파라핀 임베딩에 조직 처리기 (tissue processor, TP1020, Leica)를 사용했다: 1 시간 동안 70 % EtOH, 1 시간 동안 80 % EtOH, 1 시간 동안 90 % EtOH, 1 시간 동안 95 % EtOH, 1 시간 동안 100 % EtOH 2번, 1 시간 동안 자일렌 2 회 및 진공에서 2 시간 동안 파라핀 2 회. 파라핀 섹션은 5 ㎛의 두께로 절단하였다. 그 후, 상기 파라핀 섹션 샘플을 3 분 동안 크실렌에서 3 회 탈파라핀 화하고 3 분 동안 100 % EtOH, 3 분 동안 95 % EtOH, 3 분 동안 90 % EtOH, 3 분 동안 80 % EtOH 및 3 분 동안 70 % EtOH에서 재수화시켰다. 증류수에서 3분 동안 3번 헹구고, Harris Hematoxylin (SH3777, Cancer Diagnostics)에서 5 분 동안 핵을 염색하고, 증류수에서 3 분 동안 3번 헹구고, 1 % 산성 알코올에서 1 초 동안 구별하고, 증류수에서 3분 동안 3번 헹구고, Eosin Y (EM500G, Cancer Diagnostics)에서 1 분 동안 세포질을 염색하고, 증류수로 3 분 동안 3번 동안 세척 하였다. 상기 염색 과정을 거친 샘플을 3 분 동안 70 % EtOH, 3 분 동안 80 % EtOH, 3 분 동안 90 % EtOH, 3 분 동안 95 % EtOH, 3 분 동안 100 % EtOH 2 회, 3 분 동안 자일렌 2 회 탈수 시켰다. 덮개를 씌우고 실온에서 보관하며 현미경으로 관찰했다.
면역 형광법은 다음과 같이 이루어졌다. 편도 조직 및 오가노이드를 4 % 파라 포름 알데히드 (PFA, 바이오 용액)에 고정시켰다. 오가노이드를 Cryo 몰드 (10 x 10 x 5 mm, k4555, SungWon Medical Company)에서 2 % 아가로스 겔과 혼합하고 실온에서 고화시켰다. 그런 다음 각 단계마다 파라핀 임베딩에 조직 처리기 (tissue processor, TP1020, Leica)를 사용했다. 1 시간 동안 70 % EtOH, 1 시간 동안 80 % EtOH, 1 시간 동안 90 % EtOH, 1 시간 동안 95 % EtOH, 1 시간 동안 100 % EtOH 2 회, 1 시간 동안 자일렌 2 회, 진공에서 2 시간 동안 2 회 파라핀 처리하였다. 파라핀 섹션은 마이크로톰 (RM2235, Leica)을 사용하여 5um의 두께로 절단되었다. 그 후, 샘플을 크실렌에 3 분 동안 2 회 침지시키고, 자일 렌과 100 % EtOH을 1 : 1로 3 분 동안, 100 % EtOH 3 분 동안 2 회 침지시키고, 상기 파라핀 섹션을 재수화하기 위해 3 분 동안 95 % EtOH, 3 분 동안 70 % EtOH, 및 3 분 동안 50 % EtOH, 이어서, 95 ℃ 수조에서 소듐 시트 레이트 완충제 (0.05 % 트윈 20을 갖는 10mM 시트르산 나트륨, pH6)를 사용하여 항원 복구를 수행 하였다. 샘플을 5 분 동안 TBS 중의 0.025 % Tx-100을 사용하여 5 분 동안 2 회 세척하고, 2 시간 동안 실온에서 차단 용액 (TBS 중 1 % BSA를 갖는 10 % NGS)으로 차단 하였다. 샘플을 얼음상에서 1 % BSA로 TBS 중에 희석 된 1 차 항체와 함께 밤새 인큐베이션 하였다. 샘플을 TBS 중의 0.025 % Tx-100을 사용하여 세척 한 후, 1 시간 동안 1 % BSA로 TBS에 희석 된 2 차 항체와 함께 인큐베이션 하였다. 샘플을 TBS를 사용하여 5 분 동안 2 회 세척 한 후, 샘플을 PBS에 희석 된 Hoechst 용액으로 배양 하였다. PBS를 사용하여 샘플을 2 회 세척 하였다. 형광 현미경으로 슬라이드를 조사했다.
도 3a에 나타난 바와 같이, 편도 조직의 계층화 된 상피와 유사한 것인 조직화 된 다층 구조가 H & E 염색 된 배양 오가노이드에서 항상 관찰되었다.
도 3b에 나타난 바와 같이, 상피 마커인 e-cadherin을 기준으로 하여, 편도 오가노이드의 외부 층은 편도 조직의 기저 층에 상응하는 NGFR에 의해 염색되었다. 대조적으로, Muc1은 편도 오가노이드의 내부 층에서 염색되어 검출되는 반면, 이는 편도 조직의 부기저층에서 상층에 이르는 범위였다. 이는 편도 오가노이드 구조가 편도 상피와 유사한 구조를 가졌다는 것을 보여준다.
편도 오가노이드에서 증식 세포의 이동을 시험하기 위해, 10 일째에 BrdU를 2 일 동안 편도 오가노이드에 처리 한 다음 15 일째에 Ki67, NGFR, CK5, CK4 및 브로모 데옥시 우리딘 (BrdU)으로 이중 면역 형광 염색을 수행 하였다.
도 3c에 나타난 바와 같이, NGFR 및 Ki67 염색 된 세포는 편도 오가노이드의 외부 층, 즉 조직의 기저층에 상응하는 부분에서 검출되는 반면, Muc1가 염색 된 세포는 편도 오가노이드의 내부층, 즉 조직의 상부 세포에 상응하는 부분에서 검출되었다. BrdU 염색 된 세포는 기저층에 포함 된 몇 가지 추가 층에서 검출되었다. 결과적으로, 10일째에 BrdU 처리 된 세포가 편도 오가노이드의 중심을 향해 이동한 바 분화가 내향 방향으로 이루어짐을 확인 하였다.
실시예 4: HPV16 E6/E7 바이러스가 편도 오가노이드의 구조적 변화를 일으키는지 여부
구개 편도선에서 두경부암으로 인한 HPV 바이러스가 편도 오가노이드의 구조적 변화를 생성하는지 확인하기 위하여 관찰하였다.
Lentiviral 벡터를 이용한 감염은 다음과 같이 수행되었다. 간단히, 5 일째 배양된 편도 오가노이드를 0.25 % 트립신 -EDTA (25200-072, GIBCO)에 의해 37 ℃ 수조에서 3 분 동안 단일 세포 내로 해리시키고 10 % FBS로 중화시켰다. 중화하는 동안, 100 ul의 차가운 액체 마트리겔 (354230, BD)을 24 웰 플레이트에 첨가하고 37 ℃에서 20 분 동안 배양하여 마트리겔을 고화시킨다. 125 ㎕의 바이러스 입자 및 125 ㎕의 2X 편도 오가노이드 배양 배지에서 1x105 단일 세포 현탁액을 1.5 ㎖ 에펜도르프 튜브에서 혼합 하였다. 그리고 10μM Y27632 (Y0503, Sigma Aldrich), 2.5μM CHIR-99021 (SML1046, Sigma Aldrich) 및 8μg / ml 폴리브렌 (H9268, Sigma Aldrich)을 첨가하고 탭핑하여 잘 혼합 하였다. 이어서, 바이러스 입자 및 단일 세포의 250 ㎕ 혼합물을 고화 된 마트리겔에 첨가하고 37 ℃에서 밤새 인큐베이션 하였다. 다음날 (~ 16 시간), 세포를 수거하고, 바이러스-함유 배지를 제거하고 배지 1 : 마트리겔 1의 비율로 48 웰 플레이트상에서 돔 형태로 배양 하였다. 세포를 처음 3 일 동안 10 uM Y-27632 (Y0503, Sigma Aldrich)가 보충 된 편도 오가노이드 배지에서 배양 하였다. 감염 3 일 후, 형질 도입 된 세포를 0.125ug / ml 푸로 마이신으로 선별하고 선별된 오가노이드를 15 일 동안 배양 하였다.
도 4a 및 b에 나타난 바와 같이, 직경 측정으로 E6 / E7 형질 도입 된 오가노이드가 대조군보다 더 큰 직경을 가지는 것을 확인했다. E6/E7을 도입한 오가노이드가 구조적으로 어떻게 변화하는지 확인하기 위해 CD44, ITGA6, NGFR, MUC1 안티바디를 사용하여 형광 항체법(immunocytochemistry, ICC)을 진행하였다. 그 결과 도 4c에 나타난 바와 같이, 기저층을 이루는 세포들과 기저층 바로 윗부분 층을 이루는 세포가 대조군에 비해서 E6/E7을 도입한 오가노이드에서 더 염색 된 것이 확인되었다. 이로써 E6/E7이 오가노이드의 구조적 변화를 일으킨 것을 확인할 수 있었다.
실시예 5: 편도 오가노이드의 사이토카인 방출 여부 확인
본 발명의 편도 오가노이드에서도 편도 상피처럼 사이토카인 분비 기능이 존재한다는 것을 입증하기 위해 편도 오가노이드로부터 방출 된 여러 사이토 카인의 상대적인 분비량을 측정하였다.
사이토카인의 분석은 다음과 같이 수행되었다. 제조사의 프로토콜에 따라 Proteome Profiler 인간 사이토 카인 어레이 키트 (ARY005B, R & D SysTEMs)를 사용하여 수행되었다. 편도 오가노이드 배양 상청액은 처리되지 않은 구개골 편도 오가노이드 및 10 ug / ml LPS로 처리 된 구개골 편도 오가노이드로부터 제조되었다. 피펫 2.0 mL의 어레이 완충제 4를 4- 웰 멀티-디쉬에 넣고 막을 4- 웰 멀티-디쉬의 웰에 두었다. 막을 1 시간 동안 인큐베이션 한 후, 1 시간 동안 인큐베이션 한 오가노이드 배양 상청액, 어레이 버퍼 4 및 1 시간 동안 인큐베이션 된 인간 사이토 카인 어레이 검출 항체 칵테일과의 혼합물을 4- 웰 멀티-디쉬에 첨가 하였다. 다음으로, 4- 웰 멀티-디시 함유 막 및 혼합물을 밤새 2-8 ℃에서 인큐베이션 한 다음, 1X 세척 완충제로 막을 10 분 동안 3 회 세척 하였다. 어레이 완충액 5 2mL로 희석 된 스트렙 타비딘 -HRP를 4- 웰 멀티-디시에 첨가하고 30 분 동안 인큐베이션 하였다. 이어서, 막을 1X 세척 완충제로 10 분 동안 3 회 세척 하였다. 막을 플라스틱 시트에 놓고 1mL의 케미 시약 혼합물을 막에 첨가 하였다. LAS4000을 사용하여 화학 물질 처리 된 막을 Chemi 시약 혼합물로 검출 하였다.
도 5a에 나타난 바와 같이, 자극되지 않은 편도 오가노이드인 대조군에 비해 LPS 자극 된 편도 오가노이드는 IL-1α, CXCL1 및 CXCL8을 더 많이 방출했다. IL-1ra, MIF 및 PAI-1은 대조군보다 자극 된 편도 오가노이드에서 더 낮게 방출됨을 확인할 수 있었다.
실시예 6: 편도 오가노이드에서 호중구의 주화성 확인
편도 오가노이드에서 호중구의 주화성(chemotaxis)를 확인하기 위하여 주화성 분석 및 정량적 PCR을 실시하였다.
호중구와 같은 특성을 갖는 HL-60 세포를 사용하여 화학 주화성 분석을 다음과 같이 수행 하였다. HL-60 세포는 한국 세포주 은행에서 구입했다. 이들은 10 % 소 태아 혈청 (FBS, Gibco)이 보충 된 RPMI1640 배지 (25mM HEPES 및 L- 글루타민, Hyclone)에서 배양되었다. 배양된 세포를 5 % CO2를 함유 한 가습 대기에서 37 ℃로 보관하였다. 실험에 사용하기 위해, 7 일 동안 1.3 % 디메틸 설폭 사이드 (DMSO, Duchefa)를 첨가하여 HL-60 세포를 과립구로 분화시켰다.
도 5b에 나타난 바와 같이, dHL-60 세포(diffrenciated-HL 세포, 과립구)는 다른 그룹보다 LPS로 처리 된 오가노이드 그룹에서 더 큰 이동성을 보였다. 이러한 결과는 편도 오가노이드가 사이토카인을 방출하고, 방출 된 사이토 카인이 dHL-60 세포의 이동 활성을 증가 시킨다는 것을 확인할 수 있었다.
실시예 7: 편도 오가노이드에서 TLR의 확인
편도 오가노이드에는 편도 조직과 동일한 TLR(toll-like receptor)이 존재하며, 병원체의 신호를 림프구로 보낸다는 것을 확인하기 위하여 TLR2, 3, 4 및 6의 발현량을 qRT-PCR로 분석하였다.
qRT-PCR은 하기와 같이 실시되었다. 오가노이드를 마트리겔에서 분리하고 PBS로 세척하고 스핀다운 한 후, 프로토콜에 따라 RNA 추출 키트 (Bioneer)를 사용하여 RNA 분리를 수행 하였다. TP950 Thermal Cycler Dice Real Time SysTEM III(TAKARA)을 사용하여 샘플에 대해 qPCR을 수행 하였다. 표 1에 사용 된 프라이머 목록이 포함되어 있다.
도 5c에 나타난 바와 같이 TLR2, 4 및 6의 발현량은 계대 3과 비교하여 계대 0에서 변하지 않았지만, 계대 0과 비교하여 계대 3에서 TLR3 발현 수준이 증가 하였다. 결과적으로, 편도 오가노이드는 TLR을 가지며, 편도 상피와 같은 면역 자극 반응으로 사이토카인을 방출함을 확인할 수 있었다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.

Claims (8)

  1. 편도 형성 배지(TFM)에서 HGF(Hepatocyte Growth Factor)를 추가로 포함하고 4-[4-(4-Fluorophenyl)-5-(4-pyridinyl)-1H-imidazol-2-yl]phenol를 포함하지 않는 것인 편도 오가노이드 제조용 배지.
  2. 청구항 1에 있어, 상기 편도 오가노이드 제조용 배지는 HGF, FGF10, FGF2, FGF 베이직, PGE2, A83-01, 니코틴아미드(Nicotinamide), 노긴(Noggin), R-스폰딘1(R-spondin1), 항생-항진균제, 글루타맥스(glutamax), B27, SB202190, 페니실린 스트렙토마이신, EGF 및 HEPES를 포함하는 것인, 편도 오가노이드 제조용 배지.
  3. 청구항 2에 있어, 상기 HGF는 전체 편도 오가노이드 제조용 배지 조성물 중 20 내지 80ng/ml의 농도로 포함하는 것인, 편도 오가노이드 제조용 배지.
  4. 청구항 1에 있어, 상기 편도 오가노이드 제조용 배지에 의하여 제조된 편도 오가노이드는 CD44, 신경 성장 인자 수용체 (Nerve growth factor receptor), 시토 케라틴 5 (Cytokeratin 5), 시토 케라틴 13 (Cytokeratin 13), 인테그린 알파 6 (Integrin alpha 6) 및 상피 세포 접착 분자 (epithelial cell adhesion molecule)로 구성된 군에서 하나 이상의 유전자를 발현하는 것인, 편도 오가노이드 제조용 배지.
  5. 청구항 1에 있어, 상기 편도 오가노이드 제조용 배지에 의하여 제조된 편도 오가노이드는 TLR(toll-like receptor) 2, 3, 4 및 6으로 구성된 군에서 선택된 하나 이상의 것을 발현하는 것인, 편도 오가노이드 제조용 배지.
  6. 청구항 1에 있어, 상기 편도 오가노이드 제조용 배지에 의하여 제조된 편도 오가노이드는 2세대 이상의 계대배양이 가능한 것인, 편도 오가노이드 제조용 배지.
  7. TFM에서 HGF를 추가로 포함하고 4-[4-(4-Fluorophenyl)-5-(4-pyridinyl)-1H-imidazol-2-yl]phenol를 포함하지 않게 하여 편도 오가노이드 제조용 배지를 제조하는 단계; 및
    상기 편도 오가노이드 제조용 배지에 배양하는 단계를 포함하는 편도 오가노이드의 제조 방법.
  8. 청구항 7의 제조방법으로 제조된 오가노이드에 염증 유발 물질을 접촉시키는 단계;
    상기 염증 유발 물질을 접촉시킨 편도 오가노이드에 시험 물질을 접촉시킨 후 사이토카인 수준을 측정하는 단계; 및
    상기 측정된 사이토카인 수준을 상기 시험 물질과 비접촉된 대조군의 염증 수준과 비교하는 단계를 포함하는, 항염제를 스크리닝하는 방법.
