노화 세포를 선택적으로 사멸시킴으로써 노화 관련 질환 및 장애를 치료 또는 예방할 수 있는 약학적 조성물 또는 건강기능식품 조성물의 제공에 관한 것이다.

Description

TPP 유도체 화합물 및 이를 포함하는 노화 또는 노화 관련 질환의 치료 또는 예방을 위한 약학적 조성물
TPP 유도체를 포함하는 노화 또는 노화 관련 질환의 치료 또는 예방을 위한 약학적 조성물 또는 건강기능식품 조성물에 관한 것이다. 본 발명은 과학기술정보통신부의 지원 하에서 과제번호 2017R1E1A1A01073964 (과제명: 노화망막세포 제거를 통한 노년황반변성 치료제 개발), 및 NRF-2018R1E1A2A02058946(과제명: 미토콘드리아 안에서의 펩타이드 자기조립에 의한 선택적 노화세포 제거를 통한 생체조직의 재생능력 회복)의 결과물이다.
노화과정이란 시간이 흐름에 따라 세포의 노화현상과 더불어 다양한 신체의 생화학적 또는 생리적인 기능이 저하되는 것으로써 나이가 들어감에 따라 노화 세포가 생체 내에 축적되고 노화와 관련한 질병들과 밀접한 영향을 미치면서 인간의 수명을 제한할 수 있다. 세포의 노화란 세포가 더 이상 분열하지 않게 되는 것으로, 세포 분열시 염색체 말단의 염기서열 부위인 텔로미어 길이의 소진으로 인하여 세포가 더 이상 분열하지 않게 되는 복제노화와 자외선, 바이러스 감염 및 화학물질 등과 같은 외부의 자극이나 스트레스에 의한 암 발생을 억제하는 기작으로써 발생하는 스트레스에 유도된 세포노화가 알려져 있다. 이런 노화세포는 노화세포의 분비 활성(SASP)이라 불리는 특정 성장인자나 생리활성 물질들을 정상세포에 비해 더 많이 분비하여 인접한 세포조직이나 세포외 기질의 미세환경에 영향을 미치면서 염증반응, 생체항상성 및 다양한 생리활성 반응에 관여할 수 있다.
일반적으로 염증반응과 관련된 분비 활성을 분비하는 노화세포들은 다양한 생리학적인 역할을 수행한 후에는 면역 시스템에 의해 제거되어 생체항상성을 유지하도록 하여 생명체가 건강한 생명활동을 수행하면서 안정적인 상태로 활동하게 되지만, 연령이 증가함에 따라서 면역 시스템이 퇴화하고 세포의 자가 치유 능력이 감소하게 됨으로써 노화세포들은 점차 몸에 축적되고 결국 생물체는 만성적으로 염증과 사는 비정상적인 상태에 이르게 되며 암 또는 다양한 노인성 질환에 걸릴 확률이 높아지게 된다.
노인성 질환 중의 하나인 황반 변성은, 65세 이상 노년층에서 약 5-10% 유병률(2014년 기준 국내 121만명)을 보이며, 고령사회의 진입에 따라 이미 급격하게 유병률이 증가하고 있는 심각한 안질환 중의 하나이다. 노년 황반 변성은 건성(dry, 80-90%)과 습성(wet, 10-20%)로 두 가지 종류가 있으며, 이 두 가지 황반 변성의 공통된 병리기전은 망막색소상피(retinal pigment epithelium, RPE) 조직의 퇴화(degeneration)로 망막색소상피에 인접해 있는 광수용체세포 (photoreceptor)가 사멸하는 것이 특징이다. 노년 황반 변성의 병인은, 유전적 요인에 기반한 환경인자의 복잡한 기전에 의해 일어날 수 있는데, 2005년에 GWAS (genome-wide association study)기법을 통하여 complement factor H (CFH) 유전자의 특정 단일염기다형성(single-nucleotide polymorphism : SNP)이 노년 황반 변성과 매우 놀랄만큼 높은 연관성을 보인다는 연구결과가 발표되었다(Klein et al. Science 2005). 그 이래로 현재까지 약 50여개의 위험 allele이 보고되었는데(Fritsche et al. Nat Genet 2016), 이를 5개의 카테고리로 나누었을 때 다수를 차지하는 것은 염증과 면역 반응 관련 유전자였다. 따라서, 노년 황반 변성은, 노화와 외부의 유해한 환경(만성적 산화스트레스 등) 및 이와 동반된 만성 염증에 의해 유발된다고 정리할 수 있다.
따라서 축적된 노화세포들을 선택적으로 제거할 수 있다면 생체항상성 또는 생체조직 재생능력을 향상시켜 생물체의 만성적 염증반응을 회복시켜 노화로 발생하는 황반 변성을 포함한 다양한 질병을 치료하는데 획기적인 방법이 될 수 있다. 최근 노화세포를 제거하는 약물들(senolytics)을 이용하여 퇴행성 관절염, 심혈관질병 등의 퇴행성 질환들을 완화시킬 수 있다는 가능성이 보고되었으나 현재까지 발굴된 senolytic은 5개 안팎 정도이며, 노화세포 클리어런스 분자적 기작이 명확하지 않아 다양한 임상학적 가치를 지닌 노화세포만 제거할 수 있는 의약 후보 물질 발굴연구가 절실하게 필요한 실정이다.
일 양상은 화합물 또는 이의 약학적으로 허용가능한 염을 제공하는 것이다.

상기 노화 관련 질환은 노인성 심혈관 질환 및 장애, 노인성 폐질환, 특발성 폐 섬유증, 만성 폐쇄성 폐질환, 골관절염, 노인 안구 질환 및 피부 노화로 이루어진 군으로부터 선택된 하나 이상인 것일 수 있다. 일반적으로 염증반응과 관련된 분비 활성을 분비하는 노화세포들은 다양한 생리학적인 역할을 수행한 후에는 면역 시스템에 의해 제거되어 생체항상성을 유지하도록 하여 생명체가 건강한 생명활동을 수행하면서 안정적인 상태로 활동하게 되지만, 연령이 증가함에 따라서 면역 시스템이 퇴화하고 세포의 자가 치유 능력이 감소하게 됨으로써 노화세포들은 점차 몸에 축적되고 결국 생물체는 만성적으로 염증과 사는 비정상적인 상태에 이르게 되며 암 또는 다양한 노인성 질환에 걸릴 확률이 높아지게 된다. 이에 따라 약학조성물 내지 화합물에 의하여 축적된 노화세포들을 선택적으로 제거할 수 있다면 생체항상성 또는 생체조직 재생능력을 향상시켜 생물체의 만성적 염증반응을 회복시켜 상기 노화 관련 질환을 치료하는 유효한 물질이 될 수 있다. 일 실시예에 따르면, 상기 약학 조성물은 노화세포의 미토콘드리아에 축적되어 미토콘드리아의 막의 무결성(integrity)을 불안정화, 미토콘드리아에서 superoxide의 생성 및 세포사멸(apoptosis)을 유도할 수 있어, 노화세포를 선택적으로 제거할 수 있다. 따라서, 상기 약학 조성물은 노화 관련 질환의 예방 또는 치료를 위한 유효물질로 사용될 수 있다.
상기 약학 조성물은 목적하는 방법에 따라 경구 투여(경구제)하거나 비경구투여(예를 들어, 주사제로서 정맥 내, 피하, 복강 내 또는 국소에 적용)할 수 있으며, 예를 들어, 피하(즉, 피부 외용제) 또는 경구(즉, 경구제) 투여일 수 있다. 또한, 점안제일 수 있다. 상기 용어 "점안제"는 점안액, 안환류액, 안연고제 또는 동결건조제의 제형, 즉 눈의 치료에 관한 모든 제형을 포함한다. 상기 점안제는 임의 제형으로 각결막 상피세포 장해의 치료 또는 예방약으로서 사용되나 특히 점안액, 안환류액, 크림, 안연고제 또는 동결건조제의 제형으로 하는 것이 바람직할 수 있으나 이것으로 한정되는 것은 아니다. 점안액, 안연고제, 크림으로 하는 경우, 첨가제로서 데히드로초산나트륨, 파라옥시안식향산메틸 등의 보존제, 염화나트륨, 글리세린 등의 등장화제(等張化劑), 카르복시메틸셀루로스, 폴리비닐알콜 등의 증점제, EDTA, 다당류 등의 안정화제의 일반적인 것을 사용하는 것이 가능하나, 이들로 한정하는 것은 아니며, 생리적으로 허용되는 것이면 된다. 상기 각종 제형에 첨가되는 완충액으로는, 등장(isotonic)으로 pH 4-9, pH 5-9의 무자극인 것을 사용하는 것이 바람직하다. 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 시간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다.
상기 약학 조성물은 약학적으로 유효한 양으로 투여할 수 있다. 용어 "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명에 다른 약학 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
상기 약학 조성물의 유효량은 환자의 연령, 성별, 상태, 체중, 체내에 활성 성분의 흡수도, 불활성율 및 배설속도, 질병종류, 병용되는 약물에 따라 달라질 수 있으며, 일반적으로는 하루에 체중 1 kg 당 0.1 내지 500 mg의 범위 내일 수 있고, 매일 또는 격일로 투여되거나, 1일 1 내지 5회의 범위 내로 나누어 투여될 수 있다. 그러나 투여 경로, 비만의 중증도, 성별, 체중, 연령 등에 따라서 증감 될 수 있으므로 상기 투여량이 어떠한 방법으로도 그 범위를 한정하는 것은 아니다.

상기 식품 조성물은 지시된 비율로 필수 성분으로서 상기 유효성분을 함유하는 것외에 추가되는 다른 성분에는 특별한 제한이 없으며 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물의 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨, 소르비톨, 에리트리톨 등의 당알코올이다. 상술한 것 이외의 향미제로서 천연 향미제(타우마틴, 스테비아 추출물(예를 들어 레바우디오시드 A, 글리시르히진등)) 및 합성 향미제(사카린, 아스파르탐 등)를 유리하게 사용할 수 있다. 상기 천연 탄수화물의 비율은 당업자의 선택에 의해 적절하게 결정될 수 있다.
상기 외에도, 식품 조성물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율 또한 당업자에 의해 적절히 선택될 수 있다.
일 실시예에 따른 상기 식품 조성물은 노화된 안구세포의 미토콘드리아에서 superoxide의 생성시키고, 세포사멸(apoptosis)를 유도함에 따라, 노화된 안구세포를 선택적으로 제거할 수 있다. 따라서, 일 실시예에 따른 상기 식품 조성물은 안구 건강의 개선를 위한 유효물질로 사용될 수 있다.
상기 식품 조성물에서 언급된 용어 또는 요소 중 청구된 화합물 또는 약학 조성물에 대한 설명에서 언급된 것과 같은 것은, 앞에서 청구된 화합물 또는 약학조성물에 대한 설명에서 언급된 바와 같은 것으로 이해된다.
또 다른 양상은 일 양상의 화합물 또는 이의 염을 유효성분으로 포함하는, 황반 변성의 개선을 위한 건강기능성 식품 조성물을 제공한다. 일 구체예에서 상기 황반 변성은 노화로 인해 발생하는 것일 수 있다.
상기 황반 변성의 개선을 위한 식품 조성물에서 언급된 용어 또는 요소 중 청구된 화합물, 약학 조성물 및 식품 조성물에 대한 설명에서 언급된 것과 같은 것은, 앞에서 청구된 것에 대한 설명에서 언급된 바와 같은 것으로 이해된다.
출원에서 개시된 각각의 설명 및 실시형태는 각각의 다른 설명 및 실시 형태에도 적용될 수 있다. 즉, 본 출원에서 개시된 다양한 요소들의 모든 조합이 본 출원의 범주에 속한다. 또한, 하기 기술된 구체적인 서술에 의하여 본 출원의 범주가 제한된다고 볼 수 없다.
일 양상에 따른 화합물 및 이를 포함하는 약학 조성물 내지 건강기능식품 조성물은, 노화 세포의 미토콘드리아 선택적으로 축적되어, 세포 독성 효과를 발생시켜 이를 제거할 수 있어 황반 변성을 포함하는 노화와 관련한 질환의 증상을 예방, 개선 또는 치료할 수 있는 바, 기존의 치료제를 대체할 수 있는 유효 물질로 활용될 수 있을 것이다.

이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1. 화합물 합성
1.1. Ada-C3-TPP의 합성
1,3-디브로모 프로판(1,3-Dibromopropane) 3.8mmol(0.769g) 및 트리페닐 포스핀(Triphenylphosphine) 3.8 mmol(1.0g)을 10 mL의 톨루엔에 용해시켰다. 반응을 110 ℃에서 3일간 가열한 후, 침전된 백색을 여과하고 추가로 50mL의 톨루엔 및 에테르로 세척하였다. 그리고 생성된 3-브로모프로필-트리페닐포스페이트 (3-Bromopropyl-TPP)를 건조시켰다. 생성물은 백색 고체였다.
상기 3-브로모프로필-TPP 1.30mmol(0.5g) 및 아다만틸 아민 1.5mmol을 12mL의 ACN에 용해시켰다. 반응을 110℃에서 24시간 동안 가열한 후, 혼합물을 HPLC로 정제하여 최종 생성물인 Ada-C3-TPP을 생성하였다. 생성물은 황색의 오일이었다. 상기 Ada-C3-TPP를 NMR을 통하여 확인하였다. 상기 화학 반응에 대한 대략적인 모식도와 상기 NMR 결과는 도 1a에 나타내었다.
1.2. C12-DA-C6-DA-C3-TPP 합성
3-브로모프로필-TPP 1.30mmol(0.5g) 및 N,N,N',N'-테트라메틸-1,6-디아 미노헥산 6.5mmol(1.2g)을 12mL의 아세토니트릴(acetonitrile : ACN)에 용해시켰다. 110 ℃에서 24시간 동안 가열한 후, 혼합물을 HPLC로 정제하여 (6-디메틸아미노헥실)-3-디메틸아미노-프로필-트리페닐포스페이트((6-dimethylaminohexyl)-3-dimethylammonio-propyl-TPP)를 생성하였다. 상기 생성물은 황색 오일이었다.
1-브로모도데칸 0.63mmol(187μL) 및 상기 (6-디메틸아미노헥실)-3-디메틸아미노-프로필-트리페닐포스페이트 0.21mmol(0.1g)를 5mL의 ACN에 용해시켰다. 110 ℃에서 24 시간 동안 가열한 후, 혼합물을 HPLC로 정제하여, 최종으로 C12-DA-C6-DA-C3-TPP를 생성하였다. 상기 생성물은 백색 고체였다. 상기 C12-DA-C6-DA-C3-TPP를 NMR을 통하여 확인하였다. 상기 화학 반응에 대한 대략적인 모식도와 상기 NMR 결과는 도 1b에 나타내었다.
참고예 1. 노화세포의 유도
우선, 정상 IMR90세포에 독소루비신(doxorubicin) 250nM를 처리하여 24시간 동안 배양하였다. 7일 동안 신선한 DMEM 배지에서 배양한 후, 노화세포를 확인하였다. 그 결과 정상 IMR90 세포의 90% 이상이 노화세포화 되었음을 확인하였다. 해당 결과는 도 2에 나타내었다.
실험예 1. 세포 생존률 실험
본 실험예에서는 상기 실시예 1에서 제조된 두 화합물이 참고예 1에서 배양된 노화세포의 세포생존률(cell viability)에 끼치는 영향을 알아보고자 하였다.
실시예 1에서 생성된 각 화합물을 참고예 1에서 배양한 노화세포와 비노화세포 각각에 처리하였고, 24, 48,및 72시간에 걸쳐 세포수를 각 계수하였다. 해당 결과는 도 3a 및 3b에 각 나타내었다.
도 3a 및 3b에 나타낸 바와 같이, 두 화합물 모두, 비노화세포 보다 노화세포에서의 세포 생존률이 낮아졌음을 확인할 수 있었다. 이는 상기 두 화합물이 비노화세포보다 노화세포에 선택적으로 작용하여 사멸시킬 수 있음을 나타낸다.
실험예 2. TRMR 실험
본 실험예에서는 상기 실시예 1에서 제조된 두 화합물이 참고예 1에서 배양된 노화세포의 미토콘드리아 막 전위차에 끼치는 영향을 확인하고자 하였다. 그러기 위하여 미토콘드리아의 막 전위차 변화를 나타내게 하는 Tetramethylrhodamine, methyl ester (TMRM) 염색과 세포핵으로서의 투과성을 확인하기 위해 4',6-diamidino-2-phenylindole(DAPI) 염색을 수행하였다. 정상 세포와 노화 세포에 아무것도 처리하지 않은 대조군과, 상기 두 화합물 각각을 처리한 후, 염색을 통해 미토콘드리아 막을 확인하였다. 해당 결과는 도 4에 나타내었다.
도 4에 나타낸 바와 같이, 두 화합물은 선택적으로 노화세포의 미토콘드리아 막(membrane)의 전위차를 낮추어 주고 있음을 확인하였다. 이것은 미토콘드리아를 지향하는 두 가지 화합물이 비노화세포 보다 노화세포의 미토콘드리아 막의 무결성(integrity)을 더 방해할 수 있음을 나타낸다.
실험예 3. Superoxide 생성 실험
본 실험예에서는 상기 두 화합물이 노화세포의 미토콘드리아 내 Superoxide의 생성에 끼치는 영향을 확인하고자 하였다. 실험은 상기 실험예 2에서와 같이 진행하였고, MitoSOX쪠 Red reagent을 사용해 형광 현미경으로 가시화하였다. 또한, 별도로 세포의 DNA에 삽입되면 핵을 밝은 적색으로 염색하는 DHE 실험을 수행하였다. 해당 결과는 도 5a 및 5b에 각 나타내었다.
도 5a 및 5b에 나타낸 바와 같이, Superoxide의 생성량이 많을수록, MitoSOX와 DHE의 붉은색이 더 강하게 나타나는데, 두 화합물은 모두 비노화세포에 비해 노화세포에서 Superoxide의 생성을 강화시켰다. 이를 통해, 두 화합물은 노화세포에 선택적으로 세포독성의 효과를 나타내는 것을 알 수 있다.
실험예 4. 세포 사멸 효과 실험
본 실험예에서는 세포 독성을 일으키는 다른 방법중의 하나인 세포사멸(apoptosis) 유도 효과를 두 화합물이 발생시키는지 여부를 확인하고자 하였다. Anexin V 실험을 통해 이를 확인하였고, 해당 결과는 도 6에 나타내었다.
도 6에 나타낸 바와 같이, 두 화합물 모두 비노화 정상 세포보다 노화세포에서 세포사멸을 더 유도하는 것으로 나타났다. 이는 상기 실시예들에서 설명한 바와 같이, 두 화합물이 비노화세포보다 노화세포에서 선택적으로 세포독성을 일으킬 수 있음을 의미한다.
실험예 5. 동물 모델
본 실험예에서는 상기 화합물이 동물의 노화세포에서도 세포독성을 발생시키는지, 그리고 화합물의 농도에 의존적인지 여부를 확인하고자 하였다. 이에, 도 7a에 나타낸 바와 같이, 8주령의 C57BL/6 마우스의 안구에 100ng/ul의 독소루비신을 투여하고, 동시에 90.9 ng/ml의 Ada-C3-TPP와 129.0 ng/ml의 C12-DA-C6-DA-C3-TPP를 투여하였다. 3일 후 다시 같은 농도의 독소루비신과 두 화합물을 투여하였다. 같은 방법으로, 도 8a에 나타낸 바와 같이, 화합물의 농도를 각 363.6ng/ml(Ada-C3-TPP), 516.0ng/ml(C12-DA-C6-DA-C3-TPP)로 증량하여, 각 투여하였다. 그리고 7일째 날에 β-galactosidase 염색법을 통해 그 화합물의 효과를 확인하였다.
도 7b 및 도 7c는 상대적으로 낮은 농도의 화합물을 투여하였을 때의 결과를 나타낸다. 도 7b 및 도 7c가 나타내는 바와 같이, 대조군에서 β-galactosidase 녹색염색 발현은 안구세포의 노화가 유도되어 있음을 나타내고, 두 화합물을 투여하였을 때 β-galactosidase 녹색염색 발현이 현저하게 줄어들었는데, 이것은 두 화합물이 안구내 노화세포에 세포독성 효과를 보였음을 의미한다.
도 8b 및 도 8c는 상대적으로 높은 농도의 화합물을 투여하였을 때의 결과를 나타내는데, 이 또한, 두 화합물을 투여하였을 때 β-galactosidase 녹색염색 발현이 현저하게 줄어들어 두 화합물이 노화된 안구세포에 세포독성 효과를 보였음을 나타내고 있다. 따라서, 상기 결과들은 두 화합물이 노화된 안구세포에서 성공적으로 세포 독성 효과를 발생시켰음을 의미한다.

  8. 청구항 7에 있어서, 상기 조성물은 황반 세포의 미토콘드리아를 표적으로 하는 것인, 황반 변성의 예방 또는 치료를 위한 약학적 조성물.
