脂質粒子
 本発明は、脂質粒子等に関する。
 脂質ナノ粒子(LNP:Lipid Nano Particle)は、種々の薬剤を内包することが可能であり、ドラッグデリバリー担体として用いられている。近年のCOVID-19の流行に伴い、LNPを利用した感染症予防用ワクチンが広く利用されるに至っている。
 現在ワクチンに利用されているLNPにおいては、粒子の凝集を阻害するためにポリエチレングリコール(PEG)修飾脂質が含まれている(特許文献1)。PEG修飾脂質を使用しない場合はLNPを得ることができないのが現状である。しかしながら、PEGはアレルゲン性を有しており、また日常的に又は医療目的で使用される各種製品に含まれているため接触頻度が高いので、PEGに起因するアナフィラキシー反応の懸念がある。

 本発明は、PEG修飾脂質に依拠せずに製造可能であり、又は、PEG修飾脂質に依拠した脂質粒子よりも薬効、安全性、又は安定性に優れ、若しくはPEG修飾脂質に依拠した脂質粒子とは免疫動態が異なる脂質粒子を提供することを課題とする。
 本発明者は上記課題に鑑み得て鋭意研究を進めた結果、驚くべきことに、脂質粒子であって、脂質粒子構成脂質としてイオン化脂質、リン脂質、及びステロールを含有し、且つ疎水性基を含む修飾多糖類を含有する、脂質粒子、であれば、上記課題を解決できることを見出した。本発明者はこの知見に基づいてさらに研究を進めた結果、本発明を完成させた。即ち、本発明は、下記の態様を包含する。
 項1. 脂質粒子であって、脂質粒子構成脂質としてイオン化脂質、リン脂質、及びステロールを含有し、且つ疎水性基を含む修飾多糖類を含有する、脂質粒子。
 項2. 前記イオン化脂質の含有量が、前記脂質粒子構成脂質100モル%に対して35~70モル%であり、
前記リン脂質の含有量が、前記脂質粒子構成脂質100モル%に対して4~25モル%であり、且つ
前記ステロールの含有量が、前記脂質粒子構成脂質100モル%に対して15~55モル%である、
項1に記載の脂質粒子。
 項3. 前記修飾多糖類の構成多糖類がプルランを含む、項1又は2に記載の脂質粒子。
 項4. 前記疎水性基がステロール骨格を有する疎水性基を含む、項1~3のいずれかに記載の脂質粒子。
 項5. 前記修飾多糖類の重量平均分子量が5000~2,000,000である、項1~4のいずれかに記載の脂質粒子。
 項6. 薬剤を含有する、項1~5のいずれかに記載の脂質粒子。

 1.脂質粒子
 本発明は、その一態様において、脂質粒子であって、脂質粒子構成脂質としてイオン化脂質、リン脂質、及びステロールを含有し、且つ疎水性基を含む修飾多糖類を含有する、脂質粒子(本明細書において、「本発明の脂質粒子」と示すこともある。)、に関する。以下にこれについて説明する。
 イオン化脂質は、脂質粒子を構成可能であり且つ脂質粒子内で正電荷を示す脂質である限りにおいて特に制限されない。イオン化脂質としては、例えば[4-ヒドロキシブチル)アザンジイル]ジ(ヘキサン-6,1-ジイル)ビス(2-ヘキシルデカノエート)(ALC-0315)、9-ヘプタデカニル 8-{(2-ヒドロキシエチル)[6-オキソ-6-(ウンデシロキシ)ヘキシル]アミノ}オクタノエート(SM-102)、(6Z,9Z,28Z,31Z)-Heptatriaconta-6,9,28,31-tetraen-19-yl 4-(dimethylamino)butanoate (D-Lin-MC3-DMA)、1,2-ジリノレイルオキシ-N,N-ジメチルアミノプロパン(DLinDMA)、1,2-ジリノレニルオキシ-N,N-ジメチルアミノプロパン(DLenDMA)、2,2-ジリノレイル-4-(2-ジメチルアミノエチル)-[1,3]-ジオキソラン(DLin-K-C2-DMA;「XTC2」)、2,2-ジリノレイル-4-(3-ジメチルアミノプロピル)-[1,3]-ジオキソラン(DLin-K-C3-DMA)、2,2-ジリノレイル-4-(4-ジメチルアミノブチル)-[1,

 アルキル基、アルケニル基は、直鎖状又は分岐鎖状であり、特に好ましくは直鎖状である。アルキル基、アルケニル基の炭素原子数の下限は、例えば1、2、3、4、5、6、7、8、9又は10であることができる。アルキル基、アルケニル基の炭素原子数の上限は、例えば30、25、20、18、16、15、14、13、12、11、10、9、8、7、6、5、又は4であることができる。炭素原子数は、上記の上限及び下限を任意に組み合わせてなる範囲であることができる。
 R1で示されるアルキル基の炭素原子数は、一般式(1)の化合物が脂質となる限りにおいて特に制限されず、例えば3~20である。当該炭素原子数は、上記集積性の観点から、好ましくは3~12、より好ましくは3~10、さらに好ましくは4~8、よりさらに好ましくは5~7、特に好ましくは6である。

 R7で示されるアルキル基の炭素原子数は、一般式(2)の化合物が脂質となる限りにおいて特に制限されず、例えば12~24である。
 R8で示されるアルキル基の炭素原子数は、一般式(2)の化合物が脂質となる限りにおいて特に制限されず、例えば12~24である。
 非芳香族窒素含有ヘテロ環基、アミノ基が置換されている場合、その置換基はアルキル基であることが好ましい。当該アルキル基としては、例えば炭素原子数1~4のアルキル基が挙げられる。
 非芳香族窒素含有ヘテロ環基は非芳香族窒素含有ヘテロ環から一つの水素原子を除いてなる一価の基である。非芳香族窒素含有ヘテロ環としては、例えばピロリジニル基、ピペリジニル基、モルホリニル基、ピペラジニル基等が挙げられる。
 Eで示されるリンカーとしては、鎖構成原子数が例えば1~10、好ましくは3~8のリンカーであることができる。当該リンカーは、好ましくは鎖中に-O-C(=O)-又は-C(=O)-O-が介在していてもよいアルキル基である。
 イオン性脂質は、1種単独であることができ、2種以上の組み合わせであることもできる。
 イオン性脂質の含有量は、脂質粒子形成性、上記集積性、安定性等の観点から、前記脂質粒子構成脂質100モル%に対して、好ましくは35~70モル%、より好ましくは40~65モル%、さらに好ましくは45~60モル%、よりさらに好ましくは45~55モル%である。
 イオン性脂質は、市販品を購入して準備することができるし、公知の方法に従って又は準じて製造することができる。製造については、例えばWO2018/230710、WO2010/129687を参照することができる。
 リン脂質としては、脂質粒子を構成可能なものである限り特に制限されないが、例えばホスファチジルコリン、ホスファチジルエタノールアミン、ホスファチジルセリン、ホスファチジルイノシトール、ホスファチド酸、リゾホスファチジルコリン、リゾホスファチジルエタノールアミン等が挙げられる。より具体的な例としては、ジステアロイルホスファチジルコリン(DSPC)、ジパルミトイルホスファチジルコリン(DPPC)、ジオレオイルホスファチジルエタノールアミン(DOPE)、パルミトイルオレオイル-ホスファチジルコリン(POPC)、パルミトイルオレオイル-ホスファチジルエタノールアミン(POPE)、パルミトイルオレオイル-ホスファチジルグリセロール(POPG)、ジパルミトイル-ホスファチジルエタノールアミン(DPPE)、ジミリストイル- ホスファチジルエタノールアミン(DMPE)、ジステアロイル-ホスファチジルエタノールアミン(DSPE)、モノメチル-ホスファチジルエタノールアミン、ジメチル-ホスファチジルエタノールアミン、ジエライドイル-ホスファチジルエタノールアミン(DEPE)、ステアロイルオレオイル-ホスファチジルエタノールアミン(SOPE)、卵ホスファチジルコリン(EPC)等が挙げられる。
 リン脂質は、脂質粒子形成性、安定性等の観点から、好ましくはホスファチジルコリン、より好ましくはDSPC、DPPC等、さらに好ましくはDSPCである。
 リン脂質は、1種単独であることができ、2種以上の組み合わせであることもできる。
 リン脂質の含有量は、脂質粒子形成性、安定性等の観点から、前記脂質粒子構成脂質100モル%に対して、好ましくは4~25モル%、より好ましくは4~20モル%、さらに好ましくは4~15モル%、よりさらに好ましくは6~12モル%、とりわけ好ましくは8~11モル%である。
 ステロールとしては、ステロール骨格を有し、脂質粒子を構成可能なものである限り特に制限されないが、例えばコレステロール、コレステリルヘミスクシネート、ラノステロール、ジヒドロラノステロール、デスモステロール、ジヒドロコレステロール、フィトステロール、フィトステロール、スチグマステロール、チモステロール、エルゴステロール、シトステロール、カンペステロール、ブラシカステロール等が挙げられる。また、これら以外にも、コレスタノール、コレスタノン、コレステノン、コプロスタノール、コレステリル-2'-ヒドロキシエチルエーテル、コレステリル-4'-ヒドロキシブチルエーテル等のコレステロールの誘導体も挙げられる。
 ステロールは、脂質粒子形成性、安定性等の観点から、好ましくはコレステロールである。
 ステロールは、1種単独であることができ、2種以上の組み合わせであることもできる。
 ステロールの含有量は、脂質粒子形成性、安定性等の観点から、前記脂質粒子構成脂質100モル%に対して、好ましくは15~55モル%、より好ましくは25~50モル%、さらに好ましくは30~45モル%、よりさらに好ましくは35~45モル%である。
 修飾多糖類は、多糖類が修飾してなる化合物であって、修飾基として疎水性基を含む限り、特に制限されない。本発明では、修飾多糖類を用いることにより、PEG修飾脂質に依拠せず(好ましくは当該脂質を使用しないものであるが、PEGに起因する副反応等を回避したものであれば多少のPEGを含んでいてもよい)、脂質粒子(特に、各脂質を上記含有量で含有する脂質粒子)を形成可能であることを見出した。
 修飾多糖類を構成する多糖類(すなわち、修飾前の多糖類)としては、糖残基がグリコシド結合した高分子であれば特に限定されることはない。多糖類を構成する糖残基としては、例えば、グルコース、マンノース、ガラクトース、フコース等の単糖類、または二糖類またはオリゴ糖類などの糖類に由来する残基を採用することができる。糖残基は1,2-、1,3-、1,4-又は1,6-グリコシド結合していてもよく、その結合はα-またはβ-型結合のいずれであってもよい。また、多糖類は直鎖状でも分枝鎖状のいずれでもよい。糖残基としてはグルコース残基が好ましく、多糖類としては、例えば天然または合成由来のプルラン、マンナン、デキストラン、アミロース、アミロペクチン等が挙げられる。これらの中でも、脂質粒子形成性、安定性等の観点から、好ましくはプルラン、マンナン、デキストラン等、より好ましくはプルラン、マンナン等、特に好ましくはプルラン等が用いられる。
 多糖類の重量平均分子量は、修飾多糖類がゲル化粒子を構成可能な限り特に制限されないが、例えば5,000~2,000,000である。該重量平均分子量は、好ましくは10,000~1,000,000、より好ましくは20,000~500,000、さらに好ましくは40,000~250,000、よりさらに好ましくは80,000~125,000である。
 多糖類としては、市販品を使用することができ、或いは公知の製造方法に従って得られたものを使用することができる。
 疎水性基は、疎水性を有する基である限り特に制限されない。疎水性基としては、脂質粒子形成性、安定性等の観点から、好ましくはステロール骨格を有する疎水性基、炭化水素基等が挙げられ、特に好ましくはステロール骨格を有する疎水性基が挙げられる。
 ステロール骨格は、式(I)に示すシクロペンタヒドロフェナントレン環にヒドロキシ基が結合したアルコールである。式(I)のA~Dの記号は、シクロペンタヒドロフェナントレン環を構成する各環を表す。

 ステロール骨格においては、シクロペンタヒドロフェナントレン環に二重結合を有していてもよく、水酸基の結合する位置も特段限定されない。好ましくは、C-3位の位置にヒドロキシ基が結合し、B環に二重結合を有するステロール類、又は、C-3位の位置にヒドロキシ基が結合し、飽和環で構成されたスタノール類である。ステロール骨格を有する疎水性基は、ステロール骨格が修飾されてなる、例えば、環構成炭素において炭化水素基(例えば炭素原子数1~20の直鎖状又は分岐鎖状のアルキル基等)に置換されてなる化合物に由来する基が挙げられる。ここで、「由来する基」とは、ある化合物において、水素原子、又は水酸基等の官能基が除かれてなる基を示す。
 ステロール骨格を有する疎水性基としては、例えばコレステロール由来の基、コレスタノール由来の基、ラノステロール由来の基、エルゴステロール由来の基、β-シトステロール由来の基、カンペステロール由来の基、スティグマステロール由来の基、ブラシカステロール由来の基等が挙げられる。これらの中でも、好ましくはコレステロール由来の基、コレスタノール由来の基、ラノステロール由来の基、エルゴステロール由来の基等のステロール由来の基が挙げられ、より好ましくはコレステロール由来の基が挙げられる。
 疎水性基としての炭化水素基としては、特に制限されず、例えば炭素原子数8~50(好ましくは10~30、より好ましくは12~20)の鎖状(好ましくは直鎖状)炭化水素基(好ましくはアルキル基)が挙げられる。
 修飾多糖類の重量平均分子量は、修飾多糖類がゲル化粒子を構成可能な限り特に制限されないが、例えば5,000~2,000,000である。該重量平均分子量は、好ましくは10,000~1,000,000、より好ましくは20,000~500,000、さらに好ましくは40,000~250,000、よりさらに好ましくは80,000~125,000である。
 修飾多糖類が含む疎水性基の数は、修飾多糖類がゲル化粒子を構成可能な限り特に制限されず、例えば多糖類を構成する糖残基100個当たり、例えば1~10個、好ましくは1~5個である。
 疎水性基は、多糖類に、直接又は間接的に(例えばリンカーを介して)連結することができる。
 修飾多糖類としては、例えば、多糖類を構成する糖残基100個当たり例えば1~10個(好ましくは1~5個)の糖単位の1級水酸基が、式(II):-O-(CH2)xCONH(CH2)yNH-CO-O-R (II)(式中、Rはステロール骨格を有する疎水性基又は炭化水素基を示し;xは0又は1を示し;yは任意の正の整数を示す)で表されるものが好ましい。yは好ましくは1~8である。
 修飾多糖類は、公知の方法(例えば国際公開第WO00/12564号)に従って又は準じて、合成することができる。一例として次の方法は挙げられる。最初に、炭素数12~50の水酸基含有炭化水素又はステロールと、OCN-RA NCO(式中、RAは炭素数1~50の炭化水素基である。)で表されるジイソシアナート化合物を反応させて、炭素数12~50の水酸基含有炭化水素又はステロールが1分子反応したイソシアナート基含有疎水性化合物を製造する。次いで、得られたイソシアナート基含有疎水性化合物と多糖類とをさらに反応させて、疎水性基として炭素数12~50の炭化水素基又はステリル基を含有する疎水性基含有多糖類を製造する。得られた反応生成物をケトン系溶媒で精製して高純度疎水性基含有多糖類の製造が可能である。
 修飾多糖類は、1種単独であることもできるし、2種以上の組合せであることもできる。
 修飾多糖類の含有量は、脂質粒子形成性、安定性等の観点から、前記脂質粒子構成脂質100モル%に対して、例えば0.5~15モル%、好ましくは1~10モル%である。
 本発明の脂質粒子は、粒子の凝集を阻害する複合化脂質(以下、単に「複合化脂質」と示す。)を含有することができる。複合化脂質としては、脂質粒子の凝集を阻害することができ、且つ脂質粒子を構成可能なものである限り特に制限されないが、例えばポリエチレングリコール(PEG)-脂質コンジュゲート、ポリアミド(ATTA)-脂質コンジュゲート、陽イオン性ポリマー-脂質コンジュゲート(CPL)等が挙げられる。より具体的には、例えば、PEG-ジアシルグリセロール(DAG)、PEGジアルキルオキシプロピル(DAA)、PEG-リン脂質、PEG-セラミド(Cer)、またはそれらの混合物を含めたPEG-脂質を含みうる。PEG-脂質コンジュゲートのPEG部分の分子量は、特に制限されないが、例えば200~10000、好ましくは500~7000、より好ましくは500~4000、さらに好ましくは1000~3000、よりさらに好ましくは1500~2500である。
 複合化脂質は、1種単独であることもできるし、2種以上の組合せであることもできる。
 複合化脂質の含有量は、脂質粒子形成性、安定性及びアナフィラキシーショックを回避する等の観点から、前記脂質粒子構成脂質100モル%に対して、例えば0~2モル%である。当該含有量は、好ましくは0~1モル%、より好ましくは0~0.5モル%、さらに好ましくは0~0.2モル%、よりさらに好ましくは0~0.1モル%、とりわけ好ましくは0~0.01モル%、特に好ましくは0モル%(複合化脂質を含有しないこと)である。
 本発明の脂質粒子は、薬剤を含有することが好ましい。薬剤としては、特に制限されないが、例えば核酸アジュバント、及び抗がん剤等が挙げられ、それらを組み合わせて含有することができる。
 本発明の好ましい一態様において、薬剤は、本発明の脂質粒子の内側に(脂質粒子の外部と隔てられた状態で)配置されている。好ましくは、薬剤(特に、核酸)は、本発明の脂質粒子の内側に、イオン化脂質と複合化した状態で配置されている。
 本発明の核酸は特に制限されず、例えばmRNA、免疫刺激性二本鎖RNA (poly-IC RNAなど), siRNA、miRNA、リボザイム、CpGオリゴDNA、アンチセンスオリゴDNA、核酸アプタマー、デゴイ、遺伝子発現用二本鎖DNA、及びジヌクレオチド等が挙げられる。核酸は、DNA、RNAのみならず、医薬有効成分と成り得るものであればそれでよく、これらに、次に例示するように、公知の化学修飾が施されたものも包含する。ヌクレアーゼなどの加水分解酵素による分解を防ぐために、各ヌクレオチドのリン酸残基(ホスフェート)を、例えば、ホスホロチオエート(PS)、メチルホスホネート、ホスホロジチオネート等の化学修飾リン酸残基に置換することができる。また、各リボヌクレオチドの糖(リボース)の2位の水酸基を、-OR(Rは、例えばCH3(2´-O-Me)、CH2CH2OCH3(2´-O-MOE)、CH2CH2NHC(NH)NH2、CH2CONHCH3、CH2CH2CN等を示す)に置換してもよい。さらに、塩基部分(ピリミジン、プリン)に化学修飾を施してもよく、例えば、ピリミジン塩基の5位へのメチル基やカチオン性官能基の導入、あるいは2位のカルボニル基のチオカルボニルへの置換などが挙げられる。さらには、リン酸部分やヒドロキシル部分が、例えば、ビオチン、アミノ基、低級アルキルアミン基、アセチル基等で修飾されたものなどを挙げることができるが、これに限定されない。また、「核酸」の語は、BNA(Bridged Nucleic Acid)、LNA(Locked Nucleic Acid)、PNA(Peptide Nucleic Acid)等の何れも包含する。
 本発明の核酸は、好ましくはCpGオリゴDNA、又はmRNA、特に後述の微生物抗原、がん抗原をコードするmRNAであることができる。
 CpGオリゴDNAの具体例は以下の通りである。
クラスA:D35-CpG、ODN1585、ODN2216、ODN2336等
クラスB:K3-CpG、ODNBW006、ODN D-SL01、ODN1668、OND1826、OND2006(CpG7909、PF-3512676)、ODN2007、ODN684等

が挙げられる。
 また、その他、例えばCpG-28、CpG-685(GNKG-168)、CpG-ODN C274、KSK-13(KSK-CpG)、CpG ODN 10104(CpG-10104)、CpG ODN-1585、ODN-5890、1018-ISS、EMD-1201081(HYB-2055、IMO-2055)等が挙げられる。
 CpGオリゴDNAとしては、市販品を使用することができ、或いは公知の製造方法に従って得られたものを使用することができる。例えば、US2022/0111061 A1の内容に基づき、ステロール骨格を有する疎水性基Aを含む修飾されたものであっても良いが、好ましくはステロール骨格を有する疎水性基Aを含む修飾されたものではない。
 mRNAの場合にそれがコードする微生物抗原としては、例えばウイルス抗原、菌抗原等が挙げられる。ウイルス抗原の由来ウイルスとしては、特に制限されないが、例えばインフルエンザウイルス(例えばA型、B型等)、風疹ウイルス、エボラウイルス、コロナウイルス、麻疹ウイルス、水痘・帯状疱疹ウイルス、単純ヘルペスウイルス、ムンプスウイルス、アルボウイルス、RSウイルス、SARSウイルス、肝炎ウイルス(例えば、B型肝炎ウイルス、C型肝炎ウイルス等)、黄熱ウイルス、エイズウイルス、狂犬病ウイルス、ハンタウイルス、デングウイルス、ニパウイルス、リッサウイルス等のエンベロープウイルス(エンベロープを有するウイルス); アデノウイルス、ノロウイルス、ロタウイルス、ヒトパピローマウイルス、ポリオウイルス、エンテロウイルス、コクサッキーウイルス、ヒトパルボウイルス、脳心筋炎ウイルス、ライノウイルス等の非エンベロープウイルス(エンベロープを有さないウイルス)等が挙げられる。菌抗原の由来菌としては、特に制限されないが、例えば百日咳菌、破傷風菌、ジフテリア菌、サルモネラ菌、ピロリ菌、ウエルシュ菌、ボツリヌス菌、カンピロバクター、大腸菌、黄色ブドウ球菌、レンサ球菌、セレウス菌、腸炎ビブリオ、アクネ菌、フェカーリス菌、ディフィシル菌、肺炎球菌、インフルエンザ桿菌、モラキセラ菌、肺炎桿菌、コイネバクテリウム、溶連菌、緑膿菌、ブドウ球菌、マイコプラズマ、カンジダ菌、アスペルギルス菌等が挙げられる。

 対象がんとしては、特に制限されず、例えば白血病(慢性リンパ性白血病、急性リンパ性白血病を含む)、リンパ腫(非ホジキンリンパ腫、ホジキンリンパ腫、T細胞系リンパ腫、B細胞系リンパ腫、バーキットリンパ腫、悪性リンパ腫、びまん性リンパ腫、濾胞性リンパ腫を含む)、骨髄腫(多発性骨髄腫を含む)、乳がん、大腸がん、腎臓がん、胃がん、卵巣がん、膵臓がん、子宮頚がん、子宮内膜がん、食道がん、肝臓がん、頭頚部扁平上皮がん、皮膚がん、悪性黒色腫、尿路がん、前立腺がん、絨毛がん、咽頭がん、喉頭がん、きょう膜腫、男性胚腫、子宮内膜過形成、子宮内膜症、胚芽腫、線維肉腫、カポジ肉腫、血管腫、海綿状血管腫、血管芽腫、網膜芽腫、星状細胞腫、神経線維腫、稀突起謬腫、髄芽腫、神経芽腫、神経膠腫、横紋筋肉腫、謬芽腫、骨原性肉腫、平滑筋肉腫、甲状肉腫及びウィルムス腫瘍等が挙げられる。
 アジュバントとしては、主剤の有効成分と併用することにより有効成分が有する本来の機能を補助したり増強したりするアジュバント機能を発揮するものであれば特に制限されないが、具体的には核酸、リポ多糖、リポペプチド、合成低分子化合物などから選択することができ、好ましくはCpGオリゴDNA、ポリIC RNA、イミダゾキノリン(例えばR848やイミキモドなど)、STINGアゴニスト(例えばサイクリックdi-GMPなど)、モノホスホリルリピッドやリポペプチドなどが用いられる。また、上記以外にも、アジュバントとしては、例えば、タキサン系の薬物、アントラサイクリン系の薬物、JAK/STATの阻害物質、インドールデオキシゲナーゼ(IDO)の阻害物質、トリプトファンデオキシゲナーゼ(TDO)の阻害物質などが挙げられる。これらの阻害物質には、当該因子に対して拮抗作用を持つ化合物の他、当該因子の中和抗体やsiRNAやアンチセンスDNAも含まれる。
 抗がん剤としては、例えばアルキル化剤、代謝拮抗剤、微小管阻害剤、抗生物質抗がん剤、トポイソメラーゼ阻害剤、白金製剤、分子標的薬、ホルモン剤、生物製剤などが挙げられる。
 薬剤は、1種単独であることもできるし、2種以上の組合せであることもできる。
 本発明の脂質粒子において、脂質粒子構成成分であるCHPに対する薬剤の質量比(CHP/mRNA質量)は、200以下、より好ましくは、100以下、さらに好ましくは90以下である。当該質量比の上限は、例えば70、50、40、30、20、10、又は8であることができ、当該質量比の下限は、例えば0.01、0.02、0.05、0.1、0.2、0.5、又は1であることができる。上記の上限及び下限は任意に組み合わせることができる。
 本発明の一態様において、本発明の脂質粒子を含む液体は、薬剤(好ましくは核酸、より好ましくはmRNA)を、例えば1~1000μg/mL、好ましくは5~500μg/mL、より好ましくは15~400μg/mL、さらに好ましくは30~300μg/mL、よりさらに好ましくは50~200μg/mLの濃度で含有することができる。
 本発明の脂質粒子は、上記以外にも他の成分(溶媒以外)を含んでいてもよい。他の成分としては、脂質粒子に配合することが公知の各種成分が挙げられ、具体的には例えば抗酸化剤、膜タンパク質等が挙げられる。
 抗酸化剤は、膜の酸化防止のために含有させることができ、膜の構成成分として必要に応じて使用される。膜の構成成分として使用される抗酸化剤としては、例えば、ブチル化ヒドロキシトルエン、没食子酸プロピル、トコフェロール、酢酸トコフェロール、濃縮混合トコフェロール、ビタミンE、アスコルビン酸、L-アスコルビン酸ステアリン酸エステル、パルミチン酸アスコルビン酸、亜硫酸水素ナトリウム、亜硫酸ナトリウム、エデト酸ナトリウム、エリソルビン酸、クエン酸等が例示される。
 膜タンパク質は、膜への機能付加又は膜の構造安定化を目的として含有させることができ、膜構成成分として必要に応じて使用される。膜タンパク質としては、例えば、膜表在性タンパク質、膜内在性タンパク質、アルブミン、組換えアルブミン等が挙げられる。
 他の成分の含有量は、本発明の脂質粒子100質量%に対して、例えば10質量%以下、好ましくは5質量%以下、より好ましくは2質量%以下、さらに好ましくは1質量%以下である。
 本発明の脂質粒子の質量平均粒子径は、例えば200nm以下、好ましくは10~200nm、より好ましくは20~200nm、さらに好ましくは40~190nm、よりさらに好ましくは40~180nm、とりわけ好ましくは40~160nmである。該粒子径は、動的光散乱法により測定することができる。
 本発明の脂質粒子は、通常、水系溶液中で形成されている。水系溶液としては、例えば各種緩衝液(例えば、酢酸緩衝液、リン酸緩衝液、ギ酸緩衝液、ヒスチジン緩衝液等)が挙げられる。
 本発明の脂質粒子は、好適には、脂質粒子構成脂質を含有するアルコール溶液と、修飾多糖類を含有する(好ましくはさらに薬剤を含有する)水系溶液とを混合する工程、又は脂質粒子構成脂質を含有するアルコール溶液と、水系溶液(好ましくはさらに薬剤を含有する)と、修飾多糖類を含有する水系溶液とを混合する工程(工程1)を含む方法によって、製造することができる。当該方法を採用することにより、簡便且つ効率的に、PEG修飾脂質に依拠せずに、又は、PEGに起因する副反応等を回避した本発明の脂質粒子を得ることができる。
 アルコール溶液の溶媒であるアルコールとしては、脂質を溶解可能なアルコールである限り特に制限されない。アルコールとしては、エタノールが好ましく挙げられる。
 アルコール溶液は、脂質粒子構成脂質を上述のモル比となるように含有することが好ましい。これにより、脂質粒子構成脂質の上述のモル比で含む脂質粒子を得ることができる。
 アルコール溶液中の脂質濃度は、例えば2~20mM、好ましくは4~12Mである。
 水系溶液は、溶媒が水を含有する(好ましくは水の含有割合が80%以上、90%以上、95%以上、又は100%である)溶媒である溶液である。
 水系溶液中の修飾多糖類の濃度は、例えば0.2~5mg/mL、好ましくは0.4~3mg/mL、より好ましくは0.6~1.5mg/mLである。
 水系溶液とアルコール溶液との混合比(水系溶液/アルコール溶液、v/v)は、例えば1~20、好ましくは2~4である。
 混合態様は、脂質粒子の形成が可能な態様である限り特に制限されないが、通常は、ボルテックス等で激しく撹拌する態様である。或いは、マイクロ流路を用いた反応系で行う場合は、反応系内で混合される。
 工程1は、通常、室温下又は加温下で実行される。
 本発明の脂質粒子は、PEG修飾脂質に依拠せずに製造可能であり、及び/又は、PEG修飾脂質に依拠した脂質粒子よりも薬効、安全性、又は安定性に優れ、若しくはPEG修飾脂質に依拠した脂質粒子とは免疫動態が異なる点において有用である。
 また、本発明の脂質粒子の別の態様では、PEG修飾脂質等の粒子の凝集を阻害する複合化脂質をさらに含むことができるが、PEG修飾脂質に依拠した脂質粒子よりも薬効、安全性、又は安定性に優れ、若しくはPEG修飾脂質に依拠した脂質粒子とは免疫動態が異なる点において有用である。
 安全性は、例えば抗PEG抗体誘導の低下、投与部位の炎症の低下、投与後発熱の低下等であることができる。
 安定性は、製剤としての安定性であり、例えば凍結融解による粒子径の増大が抑制されている。具体的には、COVID-19ワクチンに代表される従来のPEG修飾脂質等を含む脂質粒子は、凍結融解に耐えることが出来ず、等張化剤(Sucrose)の有無に関わらず、凍結融解を繰り返すことで、粒子径が大きくなるが、本発明の脂質粒子は、CHP等の修飾多糖類を含むことにより、脂質粒子製剤の安定性を向上させることができ、また、後述の実施例4(Method2)の作製方法で作られたPEG修飾脂質を含まない本発明の脂質粒子は、凍結融解に対する安定性は、さらに向上し、Sucroseが無くても、粒子径増大を抑えることが出来る。
 免疫動態は、例えば誘導される細胞免疫の質の違い等であることができる。
 2.用途
 本発明の脂質粒子は、薬剤を内包することができ、またDC-SIGN(CD209)発現細胞(例えばM2様マクロファージや抗原提示細胞)へ選択的集積性を有する。このため、本発明の脂質粒子は、例えば医薬、試薬等(本明細書において、「本発明の薬剤」と示すこともある。)に、より具体的には、例えば、がん、免疫疾患、糖尿病起因性疾患、中枢性等の治療薬や、感染症ワクチン又はがんワクチンに利用することが可能である。感染症ワクチンとしては、感染症に対して使用されるワクチンであれば特に制限されず、予防用のワクチンだけでなく、治療用のワクチンも含まれる。
 本発明の薬剤は、本発明の脂質粒子を含有する限りにおいて特に制限されず、必要に応じてさらに添加剤を含んでいてもよい。添加剤としては、例えば基剤、担体、溶剤、分散剤、乳化剤、緩衝剤、安定剤、賦形剤、結合剤、崩壊剤、滑沢剤、増粘剤、保湿剤、着色料、香料、キレート剤等が挙げられる。
 本発明の薬剤の使用態様は、特に制限されず、その種類に応じて適切な使用態様を採ることができる。本発明の薬剤は、その用途に応じて、例えばin vitroで使用する(例えば、培養細胞の培地に添加する。)こともできるし、in vivoで使用する(例えば、動物に投与する。)こともできる。
 本発明の薬剤の適用対象は特に限定されないが、哺乳動物では、例えば、ヒト、サル、マウス、ラット、イヌ、ネコ、ウサギ、ブタ、ウマ、ウシ、ヒツジ、ヤギ、シカ等が挙げられる。また、細胞としては、動物細胞等が挙げられる。細胞の種類も特に制限されず、例えば血液細胞、造血幹細胞・前駆細胞、配偶子(精子、卵子)、線維芽細胞、上皮細胞、血管内皮細胞、神経細胞、肝細胞、ケラチン生成細胞、筋細胞、表皮細胞、内分泌細胞、ES細胞、iPS細胞、組織幹細胞、がん細胞等が挙げられる。
 本発明の薬剤は、任意の剤形、例えば錠剤(口腔内側崩壊錠、咀嚼可能錠、発泡錠、トローチ剤、ゼリー状ドロップ剤などを含む)、丸剤、顆粒剤、細粒剤、散剤、硬カプセル剤、軟カプセル剤、ドライシロップ剤、液剤(ドリンク剤、懸濁剤、シロップ剤を含む)、ゼリー剤などの経口製剤形態や、注射用製剤(例えば、点滴注射剤(例えば点滴静注用製剤等)、静脈注射剤、筋肉注射剤、皮下注射剤、皮内注射剤)、外用剤(例えば、軟膏剤、パップ剤、ローション剤)、坐剤吸入剤、点眼剤、眼軟膏剤、点鼻剤、点耳剤、リポソーム剤等の非経口製剤形態を取ることができる。
 本発明の薬剤の投与経路としては、所望の効果が得られる限り特に制限されず、経口投与; 経管栄養、注腸投与等の経腸投与; 経静脈投与、経動脈投与、筋肉内投与、心臓内投与、皮下投与、皮内投与、腹腔内投与、経鼻投与、気道内投与、点眼投与等の非経口投与等が挙げられる。
 本発明の薬剤中の本発明の脂質粒子の含有量は、使用態様、適用対象、適用対象の状態等に左右されるものであり、限定はされないが、例えば0.0001~100重量%、好ましくは0.001~50重量%とすることができる。
 本発明の薬剤を動物に投与する場合の投与量は、薬効を発現する有効量であれば特に限定されず、通常は、本発明の脂質粒子の重量として、一般に経口投与の場合には一日あたり0.1~1000 mg/kg体重、好ましくは一日あたり0.5~500 mg/kg体重であり、非経口投与の場合には一日あたり0.01~100 mg/kg体重、好ましくは0.05~50 mg/kg体重である。上記投与量は、年齢、病態、症状等により適宜増減することもできる。
 以下に、実施例に基づいて本発明を詳細に説明するが、本発明はこれらの実施例によって限定されるものではない。
 調製例1.プルランナノゲルの調製
 既報(Macromolecules 1993, 23, 3062-3068)の記載に従って、重量平均分子量100,000のプルランに100単糖あたりコレステロールが1.2個導入されたコレステロール修飾プルラン(CHP)を作製した。作製されたCHPを用いて、既知の方法で、プルランナノゲル溶液を調整した。
 参考例1.LNPの作製1(mRNA:オボアルブミン(OVA) mRNA)
 OVA mRNA水溶液(mRNA濃度:70μg/mL、溶媒:25mM NaAc, 154mM NaCl, pH=4.5)を調製した(A液)。ALC-0315([(4-Hydroxybutyl)azanediyl]di(hexane-6,1-diyl) bis(2-hexyldecanoate)):DSPC(Distearoylphosphatidylcholine):DMG-PEG200(1,2-Dimyristoyl-sn-glycero-3-methoxypolyethylene glycol):Cholesterol=50:10:1.5:38.5の割合(モル比)で混合した脂質のエタノール溶液(脂質濃度:8 mM)を調製した(B液)。調製したそれぞれの溶液を、iLiNPマイクロ流路チップ(ライラックファーマ)、或いは、NanoAssemblr(Precision NanoSystems)を用いて、流速500μL/min(iLiNPマイクロ流路チップ)あるいは、流速12mL/min (NanoAssemblr)(Flow Rate Ratio;A:B=3)で混合した。混合した溶液を、透析膜(MWCO;3.5K)を用いて、PBSへ溶媒置換した。回収した溶液を、アミコンチューブ(MWCO; 30K)を用いて5倍濃縮し、0.45μmのフィルターに通し、LNP(OVA)溶液を得た(質量平均粒子径:81nm、ELSZ-2000ZS大塚電子株式会社)。Ribo Green Assay (Invitrogen) を行い、mRNA濃度として、185μg/mLを算出した。
 参考例2.LNPの作製2(mRNA:EGFP mRNA)
 OVA mRNAに代えて蛍光タンパク質であるEGFPのmRNAを用いる以外は参考例1と同様にしてLNP(EGFP)溶液を得た(質量平均粒子径:75nm、mRNA濃度:186μg/mL)。
 参考例3.LNPの作製3(mRNA:HPV E7 mRNA)
 OVA mRNAに代えてヒトパピローマウイルスのE7タンパク質のmRNAを用いる以外は参考例1と同様にしてLNP(HPV E7)溶液を得る。
 実施例1.CHP:LNPの作製1(mRNA:OVA mRNA)
 LNP(OVA)溶液(参考例1)1mLに対し、同量の10mg/mL CHP水溶液を加え、30分間攪拌した。0.45μmのフィルターに通し、CHP:LNP(OVA)溶液を得た(質量平均粒子径:122nm、mRNA濃度:93μg/mL)。(Method 1) 。
 実施例2.CHP:LNPの作製2(mRNA:EGFP mRNA)
 LNP(OVA)溶液(参考例1)に代えてLNP(EGFP)溶液(参考例2)を用いる以外は実施例1と同様にしてCHP:LNP(EGFP)溶液を得た(質量平均粒子径:117nm、mRNA濃度:95μg/mL)。(Method 1) 。
 実施例3.CHP:LNPの作製3(mRNA:HPV E7 mRNA)
 LNP(OVA)溶液(参考例1)に代えてLNP(HPV E7)溶液(参考例3)を用いる以外は実施例1と同様にしてCHP:LNP(HPV E7)溶液を得る。(Method 1) 。
 実施例4.CHP:LNPの作製4(mRNA:OVA mRNA)
 OVA mRNA水溶液(mRNA濃度:70μg/mL、溶媒:1mg/mL CHP in 25mM NaAc, 154mM NaCl, pH=4.0)を調製した(A液)。MC3:DSPC:Cholesterol=50:10:40の割合(モル比)で混合した脂質のエタノール溶液(脂質濃度:8mM)を調製した(B液)。調製したそれぞれの溶液を、iLiNPマイクロ流路チップ、或いは、NanoAssemblrを用いて、流速500μL/min(iLiNPマイクロ流路チップ)、或いは、流速12mL/min(NanoAssemblr)(Flow Rate Ratio;A:B=3)で混合した。混合した溶液を、透析膜(MWCO;3.5K)を用いて、PBSへ溶媒置換した。回収した溶液を、アミコンチューブ(MWCO; 30K)を用いて5倍濃縮し、0.45μmのフィルターに通し、CHP :LNP(OVA)溶液を得た(質量平均粒子径:155nm、mRNA濃度:70μg/mL)。(Method 2) 。
 実施例5.CHP:LNPの作製5(mRNA:EGFP mRNA)
 EGFP mRNA水溶液(mRNA濃度:70μg/mL、溶媒:2mg/mL CHP in 25mM NaAc, 154mM NaCl, pH=4.0)を調製した(A液)。ALC-0315:DSPC:Cholesterol=45.5:9:45.5の割合(モル比)で混合した脂質のエタノール溶液(脂質濃度:8mM)を調製した(B液)。調製したそれぞれの溶液を、iLiNPマイクロ流路チップ、或いは、NanoAssemblrを用いて、流速500μL/min(iLiNPマイクロ流路チップ)、或いは、流速12mL/min(NanoAssemblr)(Flow Rate Ratio;A:B=3)で混合した。混合した溶液を、透析膜(MWCO;3.5K)を用いて、PBSへ溶媒置換した。回収した溶液を、アミコンチューブ(MWCO; 30K)を用いて5倍濃縮し、0.45μmのフィルターに通し、CHP:LNP(EGFP)溶液を得た(重量平均粒子径:135nm、mRNA濃度:82μg/mL)。(Method 2) 。
 実施例6.CHP:LNPの作製6(mRNA:HPV E7 mRNA)
 OVA mRNAに代えてHPV E7 mRNAを用いる以外は実施例4と同様にしてCHP:LNP(HPV E7)溶液を得る。(Method 2) 。
 実施例7~23.CHP: LNPの作製7
 以下の表1に示す方法(Method1、Method2)、核酸、イオン化脂質、脂質割合を採用して、CHP: LNPを作製した。実施例1~23で作製したCHP:LNPの作製組成と物性値を以下の表に示す。

 実施例58.CHP:LNPの作製8(mRNA:mCherry mRNA)
 mCherry mRNA水溶液(mRNA濃度:70μg/mL、溶媒 25mM NaAc, 154mM NaCl, pH=4.0)を調製した(A液)。SM-102:DSPC:Cholesterol=45.5:9:45.5の割合(モル比)で混合した脂質のエタノール溶液(脂質濃度:8mM)を調製した(B液)。さらに、調整例1で示した方法で、プルランナノゲル溶液(CHP濃度:2mg/mL、溶媒 PBS)を調整した(C液)。調製したそれぞれの溶液を、NanoAssemblr(Precision NanoSystems)を用いて、流速12mL/min(Flow Rate Ratio;A:B=1:3:2.46)で混合した。混合した溶液を、透析膜(MWCO;10K)を用いて、PBSへ溶媒置換した。回収した溶液を、アミコンチューブ(MWCO; 100K)を用いて7倍濃縮し、0.45μmのフィルターに通し、CHP:LNP(mCherry)溶液を得た(重量平均粒子径:131nm、mRNA濃度:135μg/mL)。(Method 3)。
 実施例24~57及び59~62. CHP:LNPの作製9
 以下の表2及び表3に示す作製方法(Method1、Method2、Method3)、核酸、イオン化脂質、リン脂質、PEG修飾脂質、脂質割合、CHP濃度を採用して、CHP: LNPをそれぞれ作製した。実施例24~62で作製したCHP:LNPの作製組成と物性値を、実施例1~23と併せて以下の表に示す。

 なお、特にPEG修飾脂質を加えない、或いは少量の場合は、析出して製造することができなかったり、粒子径が200nm以上の非常に大きな粒子となることが多く、体内への投与に資するLNPが作成できないが、予想外にも、CHPを付与することによって、本願発明の実施例1~62では何れも理想的な粒子径のLNPを製造することができた。
 試験例1.マウスへの免疫による抗原性評価試験
 C57BL/6マウス(日本エスエルシー)に、CHP:LNP(OVA)(実施例1、実施例4)、またはLNP(OVA)(参考例1)を、mRNA量として10μgずつ皮下投与を行う。また、比較対象として、別のマウスに、それぞれ同用量にて筋肉内投与を行う。3週後に再び同じ用量と投与方法で投与を行い、さらに1 週間後に採血と脾臓細胞、所属リンパ節を採取する。
 速やかに脾臓細胞を回収し、OVAのCD4エピトープペプチド(ROCOLAND、Ovalbumin 323-339-OH)とOVAのCD8エピトープペプチド(ROCOLAND、OVA 257-264)で刺激し、分泌されるIFN-γをELISA法で、細胞内のIFN-γ、IL-2、TNFαを細胞内サイトカイン染色法で測定する。
また、所属リンパ節由来細胞と回収した脾臓細胞について、複数細胞表面マーカー(CD3、CD4、CD8、CD127、CD62L、CD44、KLRG1、Tim-3、Lag-3、 PD-1)で発現解析を行い、T細胞の状態を解析する。
 OVA抗原とMHC複合体を認識する抗体(Biolegend)を用いて、抗原提示細胞の頻度と提示されたOVA抗原量を測定する。この際、細胞表面マーカーとしてCD11b, CD11c, F4/80, B220, CD8aなどを共染色することで抗原提示細胞の特定を行う。また、SIGN-R1に対する抗体を用いて抗原提示細胞への取り込みが受容体依存的であるか評価する。
 血清中のOVA特異的抗体をELISA法で測定する。
 試験例2.CHP:LNP(OVA)とLNP(OVA)の毒性比較試験
 (1)サイトカイン放出症候群
 C57BL/6マウスに、CHP:LNP(OVA)(実施例1、実施例4)、またはLNP(OVA)(参考例1)を、mRNA量として10μgずつ皮下投与を行う。経時的に採血を行い、IL-6やIL-1β等の血中濃度をELISA法またはmultiplex bead-based assay panel(BioLegend)で測定する。
 (2)投与部位局所の皮膚炎症
 C57BL/6マウスに、CHP:LNP(OVA)(実施例1、実施例4)、またはLNP(OVA)(参考例1)を、mRNA量として10μgずつ皮下投与を行う。24時間後に投与部位の皮膚を摘出する。皮膚の紅潮を観察した後、浸潤しているリンパ球数を測定する。
 (3)体重変動
 C57BL/6マウスに、CHP:LNP(OVA)(実施例1、実施例4)、またはLNP(OVA)(参考例1)を、mRNA量として10μgずつ皮下投与を行う。その後、経時的に体重変化を測定する。
 (4)体温変動
 C57BL/6マウスに、CHP:LNP(OVA)(実施例1、実施例4)、またはLNP(OVA)(参考例1)を、mRNA量として10μgずつ皮下投与を行う。その後、経時的に直腸体温を測定する。
 試験例3.マウスへの免疫によるワクチン評価試験
 C57BL/6マウスに、CHP:LNP(HPV E7)(実施例6)、またはLNP(HPV E7)(参考例3)を、mRNA量として10μgずつ皮下投与にて免疫する。2または3週間後、同用量にて2回目の免疫を行い、その同日に1x105個のHPV E7タンパク安定発現マウス肺癌細胞株(TC-1細胞)を皮下に移植する。その後、腫瘍の大きさを経時的に測定する。
 試験例4.免疫原性試験
 C57BL/6マウスに、CHP:LNP(OVA)(実施例1、実施例4)、またはLNP(OVA)(参考例1)を、mRNA量として10μgずつ皮下投与する。その後、2または3週間隔で同用量にて複数回(2回~5回)の皮下投与を行う。その後、血中の抗PEG抗体量と抗CHP抗体量をELISA法で測定する。
 試験例5.ヒト末梢血リンパ球由来マクロファージでの機能評価試験
 ヒトの末梢血からCD14陽性単球を単離し、24穴Tissue culture plateに、2 x 105 個/ウェルで500 μLの基礎培地(RPMI、10%ウシ胎児血清、50 ng/mL M-CSF)で播種し、37℃、5%CO2のインキュベーターで3日間培養した。その後、500μLの基礎培地で交換してさらに3日間培養した。M0マクロファージの分化誘導は、500μLの基礎培地で交換し、一方、M2様マクロファージへの分化誘導は、基礎培地に50 ng/mL IL-4、50 ng/mL IL-6、50 ng/mL IL-10、50 ng/mL IL-13を添加した、500 μLの分化誘導用培地で交換し、二日間培養した。mRNA量として0.5μgずつ含有したCHP:LNP(EGFP)(実施例2、実施例5)、またはLNP(EGFP)(参考例2)を、基礎培地に添加し、4℃条件のみ、2時間培養後、500 μLの培地に置換し、一晩37℃、5%CO2でインキュベートした。その後、PBSで洗浄後、200 μLのCell Dissociation 溶液(Biological Industries)を添加して、20分間、37℃でインキュベートし、200 μLのWash溶液(Miltenyi)を加え、ピペッティングにて細胞を剥がし、500gで5分間遠心して細胞を回収した。その後、PB450標識抗CD206抗体(BD Biosciences)と、APC標識抗DC-SIGN抗体(R&D Systems)で染色し、フローサイトメーターで解析した。
 <試験結果>
 CHP:LNP(EGFP)(実施例2)またはLNP(EGFP)添加後、4℃での処理を経ずに37℃一晩インキュベートした場合の結果を図1に示す。また、CHP:LNP(EGFP)(実施例2)またはLNP(EGFP)添加後、4℃で2時間処理後、培地を交換し、37℃一晩インキュベートした場合の結果を図2に示す。また別実験で、CHP:LNP(EGFP)(実施例2)、PEGを含まないCHP:LNP(EGFP)(実施例5)またはLNP(EGFP)添加後、37℃一晩インキュベートした場合の結果を図3に示す。
 試験例6.マウス皮下投与による抗原提示細胞での発現確認実験
 C57BL/6マウスに、CHP:LNP(EGFP)(実施例5)、またはLNP(EGFP)(参考例2)を、mRNA量として10μgずつ皮下投与を行う。24時間後に鼠経リンパ節を摘出し、細胞を回収して、フローサイトメーターで解析を行う。
 試験例7. 凍結融解前後の粒子径の測定
 CHP:LNP(実施例19, 20, 23, 26~54, 57~62)を-30℃で一度凍結し、その後融解させ、凍結融解前後での粒子径を測定した。また、その比較対象としてCHPを含まない同じ組成のLNP(参考例19, 26, 32, 36, 39, 42, 44~50, 54,60)についても同様に、-30℃で一度凍結し、その後融解させ、凍結融解前後の粒子径を測定した。
 <試験結果>
 得られた結果を以下の表4に示す。比較対象のCHPを含まないLNPは、凍結融解を一度行うだけで粒子径が増大するが、予想外にも、CHP:LNPは凍結融解前後で粒子径の増大が抑制されていた。以上から、CHPはLNPの安定性を向上する効果があるといえる。

 試験例8.ヒト末梢血リンパ球由来マクロファージでの機能評価試験-2
 試験例5と同様の方法で、M0マクロファージとM2様マクロファージを準備し、mRNA量として0.5μgずつ含有したCHP:LNP(mCherry)(実施例51、58、61)、または、LNP(mCherry)(参考例50)を基礎培地に添加し、一晩37℃、5%CO2でインキュベートした。その後、PBSで洗浄後、200 μLのCell Dissociation 溶液(Biological Industries)を添加して、20分間、37℃でインキュベートし、200 μLのWash溶液(Miltenyi)を加え、ピペッティングにて細胞を剥がし、500gで5分間遠心して細胞を回収した。その後、PB450標識抗CD206抗体(BD Biosciences)と、APC標識抗DC-SIGN抗体(R&D Systems)で染色し、フローサイトメーターで解析した。
 <試験結果>
 結果を図4に示す。CHPを付与したLNP(CHP:LNP)でのみ、DC-SIGN陰性細胞であるM0マクロファージに比べ、DC-SIGN陽性細胞であるM2様マクロファージで高発現していた。以上から、CHPを付与したLNPは、イオン化脂質に依らずDC-SIGN陽性細胞に選択的にmRNAの送達が可能であり、搭載したmRNAからタンパク質を発現できる性能があることが判った。
 試験例9. 免疫原性試験-2
 C57BL/6マウスに、CHP:LNP(mCherry)(実施例40)、PEG修飾脂質を含まないCHP:LNP(mCherry)(実施例57)、または、LNP(mCherry)(参考例39)を、mRNA量として10μgずつ皮下投与をした。試験群は各n=5とした。さらに3週間後に同用量にて再度皮下投与を行った。その1週間後に、血中の抗PEG抗体の量をELISA法(Life Diagnostics)で測定した。陰性コントロールとしては溶媒のみ(PBS)を投与した。
 <試験結果>
 得られた結果を図5に示す。陰性コントロール、CHPを付与したLNP(CHP:LNP(mCherry))、CHPを付与したPEG修飾脂質を含まないLNP(CHP:LNP(mCherry)PEG Free)投与群と比較して、LNP(mCherry)投与群は、有意に抗PEG抗体(IgM)が検出された(*;p <0.05, unpaired t検定)。以上の結果から、CHPを付与したLNPは、PEG修飾脂質を含んでいても、抗PEG抗体に起因する副反応、例えば、アナフィラキシーや、繰り返し投与によって薬効が減弱するAccelerated blood clearance(ABC)現象を回避できる可能性を示唆している。 
 試験例10. マウス皮下投与による近傍リンパ節での発現確認実験-1
 C57BL/6マウスに、CHP:LNP(mCherry)(実施例40)、またはLNP(mCherry)(参考例39)を、mRNA量として5μgずつを、右側背臀部周囲の皮内に投与した。投与後17時間目にin vivo イメージング機器(Vilber Bio Imaging)を用いて、mCherryタンパク質の蛍光強度を検出し、投与局所および近傍リンパ節(鼠経リンパ節)での発現を測定した。また、摘出した近傍リンパ節(鼠経リンパ節)を、同機器を用いて、同波長で蛍光タンパクを検出した。
 <試験結果>
 投与局所の代表例の結果を図6に示す。また、摘出した近傍リンパ節(鼠経リンパ節)の代表例の結果を図7に示す。投与局所では、LNP(mCherry)は広く発現が確認できたが、CHP:LNP(mCherry)の発現は低く、一方、近傍リンパ節では、CHP:LNP(mCherry)のほうがLNP(mCherry)に比べて発現が高かった。また、摘出した近傍リンパ節では、LNP(mCherry)に比して、CHP:LNP(mCherry)で高い発現が確認できた。以上の結果は、皮内投与した場合、CHPを付与したLNPは、LNPに比べて近傍のリンパ節への送達能が優れていることを示している。
 試験例11. 投与部位局所の皮膚炎症評価-2
 C57BL/6マウスを剃毛後、CHP:LNP(EGFP)(実施例2)、またはLNP(EGFP)(参考例2)を、mRNA量として9.8μgずつを、背臀部周囲の皮内に投与した。投与後3、5日目後に、それぞれ皮膚組織を採取し、4%パラホルムアルデヒドで48時間固定後、70%エタノール溶液に置換した。その後、HE染色と抗IBA1抗体による免疫染色を行い、病理観察を行った。   
 <試験結果>
 得られた結果を表5に示す。LNP(EGFP)の投与群では、炎症性細胞の浸潤、活性化マクロファージの指標であるIBA1陽性細胞の増加が、CHP:LNP(EGFP)投与群に比べて高かった。以上の結果は、脂質ナノ粒子の局所投与によって誘発される炎症が、CHPを付与したLNP(CHP:LNP)ではLNPに比べて抑制されることを示している。

 試験例12. 体重に及ぼす影響の評価
 C57BL/6マウスに、CHP:LNP(OVA)(実施例1)、または、LNP(OVA)(参考例1)を、mRNA量として10μgずつを皮下投与した(0日目)。さらに、初回投与から3週間後(21日目)に、同用量にて再度皮下投与した。体重測定は、各投与日とその直後2日間は連続して測定し、さらに初回投与から28日目まで、週一回の頻度で行った。試験群は各n=3とした。比較対象群は薬液を何も投与を行わなかった。
 <試験結果>
 得られた結果を図8に示す。各群の投与初日(0日目)の体重を100%と換算し、各群の平均値を用いてグラフ化した。なお、図中の矢印はLNPを投与した日を示す。その結果、LNP投与群では、各投与の翌日に体重減少が認められたが、一方、CHPを付与したLNP投与群では、その変化は軽微であり、薬剤未投与群と近い体重推移を示した。以上から、LNPにCHPを付与することによって、LNPに起因する体重減少という副作用を軽減できることが示された。

Claims (19)

  1. 脂質粒子であって、脂質粒子構成脂質としてイオン化脂質、リン脂質、及びステロールを含有し、且つ疎水性基を含む修飾多糖類を含有する、脂質粒子。
  2. 前記イオン化脂質の含有量が、前記脂質粒子構成脂質100モル%に対して35~70モル%であり、
    前記リン脂質の含有量が、前記脂質粒子構成脂質100モル%に対して4~25モル%であり、且つ
    前記ステロールの含有量が、前記脂質粒子構成脂質100モル%に対して15~55モル%である、
    請求項1に記載の脂質粒子。
  3. 前記修飾多糖類の構成多糖類がプルランを含む、請求項1に記載の脂質粒子。
  4. 前記疎水性基がステロール骨格を有する疎水性基を含む、請求項1に記載の脂質粒子。
  5. 前記修飾多糖類の重量平均分子量が5000~2,000,000である、請求項1に記載の脂質粒子。
  6. 薬剤を含有する、請求項1に記載の脂質粒子。
  7. 前記薬剤が核酸である、請求項6に記載の脂質粒子。
  8. 前記核酸が、mRNA、免疫刺激性二本鎖RNA、 siRNA、miRNA、リボザイム、CpGオリゴDNA、アンチセンスオリゴDNA、核酸アプタマー、デゴイ、遺伝子発現用二本鎖DNA、及びジヌクレオチドから選択される少なくとも1つである、請求項7に記載の脂質粒子。
  9. 前記核酸がmRNAまたはCpGオリゴDNAである、請求項7に記載の脂質粒子。
  10. 前記核酸がmRNAである、請求項7に記載の脂質粒子。
  11. 前記核酸がCpGオリゴDNAである、請求項7に記載の脂質粒子。
  12. 前記イオン化脂質が一般式(1):

  15. 前記複合化脂質がポリエチレングリコール-脂質コンジュゲートである、請求項13に記載の脂質粒子。
  16. 質量平均粒子径が10~200nmである、請求項1に記載の脂質粒子。
  17. 請求項1~16のいずれかに記載の脂質粒子を含有する、医薬。
  18. がん、免疫疾患、中枢性疾患等の治療薬、感染症ワクチン又はがんワクチンである、請求項17に記載の医薬。
  19. 皮下投与、経静脈投与、筋肉内投与、経鼻投与又は気道内投与に用いるための、請求項17に記載の医薬。
