Abstract

La divulgation concerne un peptide antigénique, en particulier pour le traitement d'individus souffrant d'un cancer de l'ovaire, de préférence sur la base d'une mutation c. 5470_5477del8 du gène BRCA1. Il est choisi parmi les séquences d'acides aminés de SEQ ID NO. 1 à SEQ ID NO. 6, ou dérivé par substitution, délétion et/ou addition d'au moins un acide aminé. Les polypeptides néoantigéniques de la présente divulgation peuvent activer de manière significative des lymphocytes T spécifiques de la mutation c. 5470_5477del8 du gène BRCA1 in vitro, stimulant la libération de la cytokine IFN-y, indiquant une immunogénicité notable. Ceci améliore la capacité des lymphocytes T à cibler et tuer des cellules cancéreuses provenant de patientes atteintes d'un cancer de l'ovaire porteuses de la mutation c. 5470_5477del8 du gène BRCA1. De plus, le peptide antigénique peut activer et étendre des lymphocytes T humains spécifiques de la mutation c. 5470_5477del8 du gène BRCA1 in vitro pour une thérapie cellulaire adoptive. Le peptide antigénique de la présente divulgation comble le manque de thérapies peptidiques antigéniques personnalisées pour des patientes atteintes d'un cancer de l'ovaire présentant les mutations somatiques c. 5470_5477del8 du BRCA1.
