Abstract
L'invention concerne des cellules ou des lignées cellulaires qui expriment un complexe de médiateur d'activation synergique (SAM) associé à des courtes répétitions palindromiques groupées et régulièrement espacées (CRISPR) (Cas) (complexe SAM CRISPR) qui comprennent un ARN guide (ARNg) qui cible spécifiquement un élément régulateur GJB2 (par exemple, un promoteur ou un activateur GJB2), dans lequel GJB2 n'est pas normalement exprimé dans ces cellules ou lignées cellulaires. L'invention concerne également des procédés de mesure de la capacité d'un vecteur à transférer une molécule d'acide nucléique dans ces cellules ou lignées cellulaires.
