Die Verbindung 5-[2-[[2-(2-Ethoxyphenoxy)ethyl]amino]propyl]-2-Methoxybenzen-Sulfonamid der Formel (1)Background of the Invention The present invention relates to a process for producing optically active 5- [2 - [[2- {2-ethoxyphenoxy) ethyl] amino] propyl] -2-methoxybenzene sulfonamide, also called tamsulosin, and to a enantiomerically enriched tamsulosin or enantiomerically pure tamsulosin prepared by this method. The compound 5- [2 - [[2- (2-ethoxyphenoxy) ethyl] amino] propyl] -2-methoxybenzene sulfonamide of the formula (1)
(5) dargestellt.is a commercially-available pharmaceutically active substance useful for the treatment of heart failure and benign prostatic hyperplasia. It has been described in EP34432 and US 4731478. The molecule, hereinafter referred to as "tamsulosin," has an asymmetric carbon atom (denoted by * in formula (1) above), so as to allow the presence of two enantiomers conventionally known as (R) - or (S) Enantiomers. The free base and the acid addition salts thereof may contain either one or both enantiomers. The individual enantiomers have a specific optical activity in polarized light and they also differ in their pharmacological effects. The commercially-available product is the hydrochloride salt of the (R) -enantiomer of tamsulosin, which is levorotatory or (R) (-) - tamsulosin hydrochloride. EP34432 / US 4731478 describe two general methods by which tamsulosin can be formed. A general method (hereinafter called method A) consists of a reductive amination of a benzyl methyl ketone compound with a substituted phenoxyethyl amine. For the production of tamsulosin, the corresponding species would be represented by formula (4) and (5), respectively.
However, none of these compounds nor the details of the actual production processes that result in tamsulosin using this process are described. Instead, method A has been exemplified only as an example of alkylsulfonamide derivatives. The compounds (4) and (5) are also not prepared as chemical entities. Further, the exemplified products of Method A, see Examples 4 and 5, are crystallized as the hydrochloride salt rather than as the free base. When Method A is used for the synthesis of tamsulosin, it would allow the conclusion that a racemic tamsulosin would be produced, however isolated in the form of a hydrochloride salt and not as a free base. The second method (hereinafter called "method B") generally teaches the conversion of a hydroxylated analogous compound into the desired sulfonamide via a chloro analogue. For the preparation of tamsulosin, the hydroxy analog would be a compound of formula (8).

It has been described that the analogous hydroxy starting compounds can be prepared according to the methods of GB 2006772, which corresponds to DE 2843016 and US 4217305. However, none of these documents specifically describes the formation of this hydroxytamsulo-sin intermediate. It also appears that in both US 4731478 and also in 5 GB 2006772 only racemic tamsulosin or racemic hydroxy tamsulosin are prepared by the described methods. None of the examples in US 4731478 show the details of the formation of tamsulosin via this process. Moreover, the compositions herein are prepared by Process B in Examples 1-3 of US 4731478 isolated as HCl salts via crystallization and not as the free base. While US 4731478 discloses racemic tamsulosin in Example 20, it does not show how the composition was prepared. Apparently, the composition was isolated as the HCl salt and not as the free base.
Siehe Beispiele 33(a) und 33(b). A third method (hereinafter called "method C") consisting of optically pure 5 - ((2) -amino-2-methyl) ethyl) -2-methoxybenzenesulfonamide with 2- (o -ethoxyphenoxy) ethyl-15-bromide is reacted to form corresponding (R) - and (S) -tamsulosin, has been described only in the examples of US 4731478 and not in the text. See Examples 33 (a) and 33 (b). In this procedure, a single enantiomer of tamsulosin was isolated as the free base of CHCl3 -methanol (9: 5) as crude crystals and then converted to HCl salt. The known process for the preparation of optically pure amine, i. the methods described in JP 58-18353, EP257787, JP 02-679248, are tedious and complicated. Further, if the optical purity in the amine is inadvertently insufficient, the resulting tamsulosin also becomes visually impure. There is no known method of purifying optically impure tamsulosin.
Therefore, the methods described above neither teach the formation of racemic tamsulosin as the free base in the solid state nor are they an excitation for the formation of a sol-25 chen substance.
ZUSAMMENFASSUNG DER ERFINDUNGSUMMARY OF THE INVENTION
The present invention relates to tamsulosin in the solid state. One aspect of the present invention relates to racemic tamsulosin as the free base in the solid state. The free base is typically a precipitate and is preferably crystalline in form. The crystalline tamsulosin as the free base of the present invention is polymorphic, two specific polymorphic forms, hereinafter called Form 1 and Form 2, are preferred aspects of the present invention.
Another aspect of the present invention relates to a process for the preparation of racemic tamsulosin as a solid free base from a solution containing racemic tamsulosin as the free base in a solvent, the solvent consisting of at least one of water and a lower alcohol. Preferred solvents are water, methanol, water / methanol mixtures and ethyl acetate / methanol mixtures. The tamsulosin solution can be formed by dissolving a tamsulosin residue in the solvent or by treating a tamsulosin acid addition salt with a base. However, the isolation of tamsulosin as a free base in the solid state is not limited to the racemate. Another aspect of the present invention relates to a process for isolating tamsulosin as a free base which consists of treating an acid addition salt of tamsulosin in a solvent with a base and precipitating tamsulosin as the free base from the solvent, the solvent being water , a lower alcohol 45 or both. The tamsulosin as the free base may or may not be racemic. The acid addition salt is normally but not limited to the hydrochloride salt and includes e.g. the camphor-10-sulfonic acid salt thereof.
KURZE BESCHREIBUNG DER ZEICHNUNGEN:An additional aspect of the present invention relates to a sulfonic acid salt of 2- (o -ethoxyphenoxy) ethylamine, especially the methanesulfate or tosylate salts thereof, and 50 (3-aminosulfonyl-4-mehoxy) phenylacetone. These compounds are novel intermediates useful for preparing tamsulosin as the free base. BRIEF DESCRIPTION OF THE DRAWINGS:

DETAILLIERTE BESCHREIBUNG DER ERFINDUNGDETAILED DESCRIPTION OF THE INVENTION
Racemic tamsulosin as the solid free base was not prepared in any of the known methods described above. The present invention is based, in part, on the discovery that racemic free base can be formed and isolated in the solid state. The solid form is preferably a precipitate of a solution. Even better, the solid form is a crystalline form. The crystalline form includes all polymorphic modifications unless otherwise stated and includes hydrates and solvates thereof. In particular, two polymorphic forms of racemic tamsulosin as the solid state free base, designated Form 1 and Form 2, have now been discovered as part of the present invention, and these are described below. Free base racemic tamsulosin can be isolated in solid state in high purity, including at least 80% purity, preferably 90% purity, and more preferably at least 95% purity. Relatively pure precipitates are white or almost white microcrystalline substances that are sparingly soluble in water but are soluble in alcohols such as methanol and ethanol and in chlorinated carbohydrates. 20 Racemic tamsulosin as a free base can occur in various polymorphic modifications. One of these modifications is referred to herein as Form 1. This form of free base leads to an X-ray diffraction recording, which essentially corresponds to FIG. 1, to an IR spectrum which substantially corresponds to FIG. 3 and to a DSC curve which corresponds essentially to FIG. 5. Form 1 generally has a melting point of about 127-129 ° C. Form 1 as a poly-25-morphic form can be obtained, inter alia, by crystallization of tamsulosin as the free base from a methanol / ethyl acetate mixture, but is not limited thereto.
Another such modification is referred to herein as Form 2. This form of the X-ray diffraction pattern, which essentially corresponds to FIG. 2, has an IR spectrum that substantially corresponds to FIG. 4 and a DSC curve that substantially corresponds to FIG. 6. A comparison 30 of the X-ray diffraction patterns in FIGS. 1 and 2 shows that tamsulosin differs as Form 1 free base of tamsulosin as Form 2 free base. This difference is also confirmed by the differences in the IR spectra. Generally, mold 2 has a melting point of about 124-126 ° C, and Fig. 6 shows a single endothermic melt value for mold 2 of about 125 ° C. The polymorphism of Form 2 is, inter alia, obtainable by precipitation of tamsulosin as free base 35 after neutralization of tamsulosin hydrochloride by sodium hydroxide in a methanol / water mixture, but not limited thereto.
Tamsulosin as a solid state free base, especially as a racemic free base, can surprisingly be obtained by precipitating the free base from a solution in which the solvent for the solution consists of at least one of either water or a lower alcohol 40. A "lower alcohol" refers to an alcohol of 1 to 4 carbon atoms, and is preferably, but not limited to, methanol or ethanol, an ester, such as ethyl acetate, an aliphatic ketone, such as acetone or methyl isobutyl ketone, or an ether, in particular water-miscible ethers such as dioxane or tetrahydrofuran. Preferred solvents are water, a mixture of water and methanol and a mixture of ethyl acetate and methanol.
The precipitation may be carried out by any conventional method, including reducing the solution temperature, removing a portion of the solvent, etc. In some embodiments, water may be used as a counter-solvent in a manner such that, upon addition of water, optionally in conjunction with reduction in the 50 solution temperature precipitation takes place. In a solvent system of water and methanol, tamsulosin is precipitated as the free base in the solid state at room temperature or less less in the solution.
4 AT 007 563 U1The precipitation of tamsulosin as the free base can be recovered by filtration, optionally dried. If desired, solid tamsulosin may also be crystallized as free base 55 from a suitable solvent. 4 AT 007 563 U1
The solution of tamsulosin can be prepared or obtained in various ways. For example, a residue from the synthesis of racemic tamsulosin can be dissolved in a solvent. A "residue" refers to the tamsulosin material obtained by evaporation of all or most of the solvent, e.g. of the solvent used to synthesize the tamsulosin molecule. Racemic tamsulosin as free base residue may e.g. dissolved in water, typically at temperatures above room temperature, i. at over 50 ° C to form the solution from which racemic tamsulosin can be precipitated as the free base. Similarly, a residue of racemic tamsulosin as the free base may be dissolved in a mixture of ethyl acetate and methanol to form the solution from which racemic tamsulosin may be precipitated as the free base. Alternatively, the solution of tamsulosin may be formed as the free base as a result of its synthesis, i. the solvent in which tamsulosin is obtained as a free base is suitable as it is or after the addition of water or a lower alcohol for use in the precipitation process according to the invention. Alternatively, the solution of racemic tamsulosin can be formed from a solution of an acid addition salt of racemic tamsulosin. The process involves treating its acid addition salt of racemic tamsulosin with a base in a solvent containing either water or a lower alcohol to form the solution of racemic tamsulosin. The base may be an organic or inorganic base, such as an Alka-20 metal hydroxide, especially sodium hydroxide, ammonia or an organic amine. The amount of base is preferably equimolar. The preferred reaction temperature ranges from room temperature to reflux temperature. The solvent in which the treatment is carried out may be formed in situ, i. Water can be added simultaneously with the base in a solution of tamsulosin as the free base in an organic solvent. A specific embodiment of this process comprises treating an acid addition salt of tamsulosin in a solvent with base and precipitating tamsulosin as the free base from the solvent, which solvent contains water, a lower alcohol, or both. This method is useful for both racemic tamsulosin such as racemic tamsulosin HCl and nonracemic or enantiomerically enriched camphor 10-30 sulfonate salts of tamsulosin. In any case, a free base is obtained in solid form. The base and the treatment conditions are the same as described above.
The solution containing the tamsulosin salt can be formed by synthesis of tamsulosin or by dissolving the available solid form of the salt, e.g. a commercially available tam-sulosine salt in a solvent. The precipitation of tamsulosin as the free base is generally formed with high purity, typically at least 80%, more typically at least 90%, and preferably at least 95% purity. The purity is based on the isolated and dried solid precipitate, as is common in the industry. If desired, tamsulosin can be recrystallized as the free base in solid form to increase its purity. As an example, tamsulosin may be recrystallized as free base 40 from a solvent containing a lower alcohol such as methanol, e.g. from a mixture of methanol and ethyl acetate, or from a mixture of methanol and water.
Racemic tamsulosin or its salt can be prepared by a suitable synthetic method. More specifically, three preferred methods will be described in more detail below. In a first method ("Method A"), racemic tamsulosin may be prepared as the free base by reductive amination of the ketone of Formula (4) by the amine of Formula (5).

30The 2- (o -ethoxyphenoxy) ethylamine (5) can be prepared by a method described in BE 668124. In this procedure, 2-ethoxyphenol (10) reacts with chloroacetonitrile for 16-24 hours by refluxing acetone in the presence of potassium carbonate. After filtration and evaporation of the solvent, the oily residue is dissolved in a suitable solvent such as ethanol or ether, treated with water and with a base such as ammonia or sodium hydroxide, and crude o-ethoxyphenoxyacetonitrile (11) is removed by evaporation of the solvent or crystallization from Solvent received. The crude product (11) can be recrystallized from a suitable solvent such as an ethanol-water mixture. The compound (11) is catalytically hydrogenated under elevated pressure in a suitable solvent such as toluene or a toluene / triethylamine solution using Raney cobalt or any other catalyst. Free base of 2- (o-ethoxyphenoxy) ethylamine (5) is obtained as a crude product after filtration and evaporation of the solvent as an oil. 30

It has now been found that the resulting free base of (5) is contaminated by one by-product and should be purified for the next step. However, normal alkaline extraction surprisingly resulted in too high a loss. The amine (5) has an unexpectedly high water solubility for an amine compound. It has been found that an effective solution for purifying the amine is the formation of a sulfonic acid salt of the amine, especially methanesulfonate or tosylate. The salt, especially the methanesulfonate salt of (5), can be isolated in the solid, preferably crystalline state by conventional methods and used as a beneficial substrate in the next reaction step. The conversion of the free base of (5) to an acid addition salt and the isolation of said salt in the solid state improves the purity of the compound (5), particularly removing the by-products resulting from the hydrogenation such as 2-ethoxyphenol. The 2- (o -ethoxyphenoxy) ethyl-methanesulfonate therefore forms a specific aspect of the present invention.
AT 007 563 U1The racemic tamsulosin is obtained by the method of US 4558156 by reductive aminolysis of the ketone (4) with the free base of amine (5). When a salt of the amine (5) is used for the reaction, this salt is first treated by treatment with a suitable base, e.g. Sodium methoxide in methanol, converted into a free base. First, an imino compound is formed by contacting both components in methanol. A hydrogenation catalyst such as platinum oxide or palladium / carbon is added to the reaction mixture and the mixture is hydrogenated by gaseous hydrogen, most preferably under elevated AT 007 563 U1
Druck. Print. After filtering the catalyst, the reaction mixture is acidified with an acid, preferably alcoholic or aqueous HCl, to form an acid addition salt of the racemic tamsulosin, preferably tamsulosin hydrochloride. The racemic tamsulosine salt is isolated in solid, preferably crystalline form by evaporation of the solvent or by recrystallization from the solvent.
The tamsulosin salt is then treated with base, and tamsulosin as the free base is precipitated as described above.
In a second method ("Method B") racemic tamsulosin hydrochloride may be prepared in the form of a hydrochloride salt from the hydroxytamsulosin of Formula (8) by the method of EP 1034342. As starting material, hydroxytamsulosin (8) can be prepared from the amine compound (5) according to US 4,217,305. The process comprises, in a first step, conversion of the hydrochloride salt of hydroxytamsulosine (8) to a chlorotamsulosine hydrochloride (12) by reaction of (8) with thionyl chloride in acetonitrile and isolation of the product from the reaction mixture after spontaneous crystallization by the reaction mixture , Compound (12) is then dehalogenated, e.g. by catalytic hydrogenation using palladium or carbon as catalyst at normal temperatures and pressures. Upon concentration of the reaction mixture, crystalline tamsulosin hydrochloride is obtained and this can be recrystallized from a mixture of methanol and ethanol.
In order to obtain tamsulosin as the free base in the solid state, the process provided for in the present invention should be applied as described above.
In einem dritten Verfahren („Verfahren C„) kann racemisches Tamsulosin als freie Base direkt 30 durch Kondensation des racemischen Amins der Formel (6) mit einer BromverbindungIn a third method ("Method C") racemic tamsulosin may be used as free base directly by condensation of the racemic amine of formula (6) with a bromo compound
The conditions for the preparation process have been described in US 5447958. However, the starting amine (6) used was optically active. Tamsulosin as the free base (optically active) was obtained after separation of the reaction mixture by column chromatography in the solid state and converted into hydrochloride. However, by column chromatography this method becomes unsuitable for use in the production of racemic tamsulosin as the free base using industrial standards.
However, it has been found that the process can be modified by using a dipolar aprotic solvent, such as dimethylformamide, to condense (6) with (7) acid, which is generally based on well-known methods. Examples of such salts are hydrochloride, hydrobromide, acetate, fumarate, maleate, citrate or methanesulfonate. If the optical purity of the resulting tamsulosin product is insufficient, the resolution of the racemate can be improved by repeating the process. Similarly, a second diastereomeric tamsulosin salt pair may be the same or different than the first pair. In one embodiment, the second pair is different than the first one, using an optically active sulfonic acid that differs in the direction of rotation from the first acid. In certain embodiments it is advantageous that in the first precipitate preferably the (S) -tamsulosin-containing diastereomers and in the second precipitation from a second solvent, preferably the R-tamsolusin-containing diastereomer is precipitated in the 7 AT 007 563 U1 first precipitation. In this embodiment, the enriched solute from the first precipitate is used either with or without liberation of tamsulosin as the free base to produce the second precipitate solution.
Optically pure or substantially pure (R) -tamsulosin, acid addition salts thereof, and more particularly (R) -tamsulosin hydrochloride prepared by the process of the invention, are useful in the preparation of medicaments for the treatment of various diseases or conditions including heart failure and benign prostatic hyperplasia, as well as others, useful. It can be used alone or in combination with other active ingredients. Such medicaments may be formulated for peroral, transdermal or parenteral use, e.g. in the form of tablets and capsules. The formulations contain therapeutically effective amounts of the active ingredient as well as pharmaceutically acceptable carriers or diluents and can be prepared by conventional methods.
The present invention will now be described and explained in more detail by way of the following examples. It should be understood, however, that the present invention is not limited to these examples and that various changes and modifications may be made without departing from the scope of the present invention.
