Description

   <Desc/Clms Page number 1>

 <Desc/Clms Page number 2>

 <Desc/Clms Page number 3>

12) On économise beaucoup de temps et d'argent, car de nombreuses opérations de purification, telles que des distil- lations fractionnées, des cristallisations, etc. deviennent inutiles ;
22) du fait que les opérations de purification mention- nées sous 12) sont devenues superflues, il résulte que le rendement en différents produits intermédiaires et finis est considérablement plus grand que celui qui était normalement possible jusqu'ici ;
32) on a constaté de façon inattendue que le produit final, c'est-à-dire le mélange de composés obtenu en' dernier lieu,'est beaucoup plus actif contre les parasites du sang que les différents-composés élémentaires, On a constaté que ces derniers renforcent mutuellement leurs activités respec- tives ;

48) dans certains cas on a constaté aussi que les élé- ments constitutifs des produits finaux sont mutuellement des antagonistes, c'est-à- dire que les propriétés toxiques nui sibles des éléments constitutifs du mélange sont plus ou moins entravées ;

 <Desc/Clms Page number 4>

   It has already been proposed to use as drugs against malaria many aromatic and heteronucleic ring systems containing an amino group substituted by an aliphatic amino compound of the general formula NR-N R1R2, in which the first nitrogen atom is linked to (or part of an aromatic or heteronucleic radical, while R denotes an aliphatic radical, R1 and R2 denoting an alkyl radical which may or may not be substituted.

   It is further known that, in order to obtain these drugs against malaria, it is necessary to introduce the mentioned aliphatic radical R. above ... in such a state that coupling with the aromatic or heteronucleic ring system, on the one hand, and with the substituted or unsubstituted amino group, on the other hand, is possible.

  Process for the preparation of drugs against malaria.

 <Desc / Clms Page number 2>

   The known methods described above are long and expensive, so that the cost price of the finished products having activity against blood parasites is extremely high. These disadvantages are due, in large part, to the fractional distillations which must be carried out. carried out several times, most often under a high vacuum, in order to separate the mixture of raw materials into its constituent parts and to obtain the raw materials and intermediate products in as pure a state as possible. In addition, it has often been found that fractional distillations do not give the desired result, requiring the application of special purification procedures, for example to separate two or more members of a homologous series.

   However, it has been found that drugs which are extremely active against malaria are obtained by not separating the raw materials into their constituent elements and by using them, on the contrary, in the form of mixtures.

   In accordance with the present invention, the castor oil or other oils (such as peanut oil, olive oil, almond oil, coconut oil or other oils are decomposed, optionally with oxidation). equivalent) to a mixture of compounds preferably containing 7 to 12 carbon atoms, then this mixture is converted in the usual way into compounds whose extreme carbon atoms bear substituents which can be replaced by amino groups and on the one hand these compounds are reacted on the one hand with an amino group of an aromatic or heteronucleic amine, and on the other hand an amino group which may or may not be substituted by groups is introduced therein. - alkyl pes.

   The process has important advantages, namely:

 <Desc / Clms Page number 3>

12) A lot of time and money is saved, because many purification operations, such as fractional distilations, crystallizations, etc. become unnecessary;
22) the fact that the purification operations mentioned under 12) have become superfluous, it follows that the yield of various intermediate and finished products is considerably greater than that which was normally possible heretofore;
32) it was unexpectedly found that the final product, that is to say the mixture of compounds obtained 'lastly,' is much more active against blood parasites than the different elementary compounds. that the latter mutually reinforce their respective activities;

48) in certain cases it has also been found that the constituent elements of the final products are mutually antagonists, that is to say that the harmful toxic properties of the constituent elements of the mixture are more or less hampered;
52) unlike most of the drugs heretofore known to control malaria, some of the mixtures produced have been shown to be active against all forms of the malaria parasite, and
62) The final products can be separated into their constituent parts by distillation or by crystallization of their salts, which is useful for identification purposes.

   Thus, while, on the one hand, the medicaments according to the invention can be produced at very low cost (as it follows from paragraphs 12) and 22 above), it follows on the other hand from paragraphs 32) , 42) and 52) above that very small amounts of these drugs are sufficient to

 <Desc / Clms Page number 4>

 achieve a satisfactory therapeutic effect. Therefore, the chemical-therapeutic indices which have been obtained with some of the mixtures prepared as indicated above are favorable and unprecedented.

   An embodiment of the manufacturing process according to the invention will be found below. This example is, of course, given as non-limiting, it relates to the preparation of a mixture of methoxy-6 (8-diethylamino) -8 quinoline and of methoxy-6 (9-diethylamino nonylamino) -8 quinoline. If one boils 1.5 kg of castor oil - using a reflux condenser .. for 32 hours with 6 kg of nitrogenous acid of density 1.25, two liquid layers are formed which the it is separated (while they are still hot) in a decanter.

  The acidic lower layer is subjected to steam distillation to remove the volatile decomposition products (heptylic acid). The residue crystallizes out to an abundant white product which is extracted and washed with cold water to rid it of the lower dicarboxylic acids which have formed. These acids are mainly oxalic acid and malonic acid which, being byproducts, can also be used for the production of drugs against malaria.

  The rest, which weighs 325 g, consists mainly of suberic acid and azelaic acid. These acids are not separated (which in itself represents a very considerable saving of time and money. ), but they are esterified together by boiling them with absolute methanol in which 2 to 3% gaseous hydrochloric acid has been dissolved. The esterification mixture obtained is subjected only once to fractional distillation, but this operation is carried out very exactly and the fraction boiling between 135 C and 1500 C

   By reduction in the known manner, a mixture of octane diol and nonane diol is obtained (the boiling point of which is 150-160 C under a pressure of 10 mm, these glycols are converted into chloro-octanol and in chlorononanol by heating them to 80 to 100 C with ten times their quantity of concentrated hydrochloric acid. Starting with castor oil (or other fatty oils such as peanut oil, olive oil, almond oil, coconut oil, etc.) can prepare, in addition to suberic acid and azelaffinic acid, many other dibasic acids which lend themselves admirably to the preparation of analogous compounds.

  As an example, the formation of sebacinic acid, nonane dicarboxylic acid, decane and undecane, dicrboxylic acid from undecylenic acid obtained as an almost worthless by-product in the preparation of. oenanthal (by vacuum distillation of castor oil).

   It has been found that the decomposition products of oils which cause the formation of compounds having chains of 7 to 12 carbon atoms are of remarkable importance, because with these compounds the mentioned advantages

ABSTRACT 1, Process for the preparation of medicaments against malaria, characterized in that, optionally, with oxidation, castor oil or other equivalent oils are decomposed into a mixture of compounds preferably containing from 7 to 12 atoms of carbon, then that this mixture is converted in the known manner into compounds whose extreme carbon atoms bear substituents which can be replaced by amino groups, and that on the one hand these compounds are reacted on a EMI9.3 amino group of an aromatic or heteronucleic amine, and that on the other hand an amino group which may or may not be substituted with alkyl groups is introduced therein.
