On sait que certains produits d'oxydation de l'adré- naline manifestent une activité pharmacodynamique et notamment , hémostatique analogue à celle de l'adrénaline même ou de ses

dérivés, tels Itaclrénalone. Parmi ces produits d'oxydation, l'adrénoohrome ou N-méthyl..3..hydroxy-2.3.d3h9dro-5,6.quinone-in- ' dol se distingue par son efficacité comme hémostatique à des doses extrèmement faibles, ( de l'ordre de p.ex. 10-6 gr. chez le lapin) et par son action plus rapide et aussi intense que colle de l'adrénaline.
Par rapport à cette dernière, elle pré- sente même l'avantage de ne pas modifier le rythme cardiaque et de ne provoquer aucune hypertension ni hémorragies internes, Grâce à ces propriétés, 11 adrénochrome conviendrait donc pour de nombreuses applications thérapeutiques, p.ex. dans les in- terventions chirurgicales, dans le traitement d'hémorragies internes, etc. Malheureusement, sa très grande instabilité, en @ @ solution aqueuse ou alcoolique, rend pour ainsi dire impossible

son utilisation en pratique médicale.
L'adrénochrome se décompose en effet en 35 minutes en solution aqueuse d'un pH de 7,3 -- en 4 minutes en solution chlorhydrique à 1% et en 40 secondes en solution ammoniacale à 1%.
Cette instabilité de l'adrénochrome est attribuée, en ordre principale à la fonction quinonique du noyau benzénique de sa molécule. Aussi la demanderesse stest-elle attachée à bloquer la fonction quinonique.
Considérant les orthoquinones, telles l'adrénochrome, comme des dicétones, elle a fait appel, à cet effet, à des ré- actifs spécifiques des cétones, en l'espèce plus particulière- ment aux réactifs aminés, représentés par les sels d'hydroxyl- amine, de semicarbazide, de phénylhydrazine et de diverses ni- trophénylhydrazines. Tous ces réactifs se condensent aisément avec l'adrénochrome, en formant par substitution d'un des ato- mes d'oxygène quinoniques, les monoxime, semicarbazone, phényl- hydrazone et nitrophénylhydrazones correspondantes. Ces pro- duits de condensation, beaucoup moins solubles que l'adrénochro- me et faciles à obtenir, à l'état pur, sous forme de cristaux, sont en effet beaucoup plus résistants vis-à-vis de la transfor- mation ou de la décomposition spontanées que l'adrénochrome même.
Leurs solutions, même très diluées dans l'eau ou dans d'autres solvants, tels l'alcool éthylique, sont également dtune grande stabilité. Malgré leur inertie chimique relative- ment grande, elles conservent, dans une mesure plus ou moins prononcée dans chaque cas, les facultés hémostatiques précieu- ses des solutions d'adrénochrome. Cette constatation, essentiel- le au point de vue thérapeutique, a été mise à profit par la demanderesse pour la préparation de solutions hémostatiques stables susceptibles d'être conservées en ampoules.
A l'expérimentation clinique des différents dérivés mentionnés, on s'est aperçu que l'intensité et la spécificité de l'action hémostatique diffèrent d'une façon plutôt inatten- due d'un produit à l'autre.

Dans la médication hémostatique chez l'homme, c'est la monosemicarbazone d'adrénochrome qui est particulièrement indiquée. Les effets vaso-constricteurs qu'elle produit l'appa- rentent à tel point à l'adrénoohrome qu'elle en constitue en quelque sorte une forme stabilisée, rendant pratiquement possi- bles les applications thérapeutiques de celle-oie
L'exemple pratique ci-dessous illustre l'application de l'invention à ce cas particulièrement intéressant de la mono- semicarbazone d'adrénochrome, sans cependant s'y limiter.
En apportant au mode opératoire indiqué et aux proportions des ré- actifs employés, les modifications dictées par la nature diffé- rente de ces réactifs, on prépare, de façon analogue et sans sortir du cadre de l'invention, les agents hémostatiques stables @ correspondants, à base de monoxime, monophénylhydrazone et mono- nitrophénylhydrazones d'adrénochrome.
Exemple.
On agite, pendant une dizaine de minutes, une suspen- sion de 1 partie en poids d'adrénaline et de 2 à 6 parties en poids d'oxyde d'argent dans 150 à 250 parties en poids d'alcool méthylique ou éthylique, sépare par essorage la solution aloooli- que d'adrénochrome obtenue et évapore rapidement le filtrat à siccité à basse température, sous le vide.
On reprend par 45 à 55 parties en poids d'eau les cristaux rouges dtadrénochrome formés et ajoute à cette solution 2 parties d'acétate de sodium dissous dans 2 à 3 parties dteau et 2 parties de chlorhydrate de semicarbazide dissous dans 2 à 3 parties deau. On sépare par filtration le précipité formé, consistant en aiguilles prismati- ques de couleur rouge-orangé, et le recristallise dans de l'al- cool éthylique dilué. On obtient 0,30 à 0,40 partie en poids/Se mono-semicarbazone d'adrénocliromep fondant à 203 C. en se décom- posant.
La solution aqueuse isotonisée de la semiearbazone à 1 pour 10.QOO, de pH = 7, conservée p.ex. en ampoules scellées, est indéfini.ment stable et peut être administrée par voie sous- cutanée ou par voie intraveineuse.

Injectée à la dose de 1 ce (0,1 mgr,) une heure avant une intervention chirurgicale, elle réduit au maximum l'intensité et la durée des hémorragies, sans produire d'effets nuisibles ou toxiques.
Il va de soi que pour préparer, suivant l'invention, un dérivé hémostatique stable d'adrénochrome, sous forme de mono- semicarbazone, on ne doit pas nécessairement, comme dans l'exemple ci-dessus, s'adresser à un produit obtenu à partir dtadrénaline au moyen d'oxyde d'argent, L'adrénochrome, stade intermédiaire instable entre l'adrénaline et la mono-semicarbazone d'adrénochrome peut, au contraire, être préparée de toute autre manière, p.ex. par oxydation diastasique de l'adrénaline,
REVENDICATIONS.
L'invention a pour objet:
1) Un procédé de préparation de dérivés stables de l'adrénochrome, doués de propriétés hémostatiques, caractérisé en ce que, par condensation avec des réactifsaminés, spécifiques des cétones, représentés par des sels d'hydroxylamine, de semicarbazide, de phénylhydrazine et de n@trophénylhydrazines, on forme les monoxime, monosemicarbazone, monophénylhydrazone et mononitrophénylhydrazones correspondantes d'adrénochrome, qu'après séparation on recristallise éventuellement dans des solvants appropriés.

Adrenochrome breaks down in 35 minutes in aqueous solution with a pH of 7.3 - in 4 minutes in 1% hydrochloric solution and in 40 seconds in 1% ammoniacal solution.
This instability of adrenochrome is attributed, in the main order to the quinone function of the benzene nucleus of its molecule. The applicant is therefore committed to blocking the quinone function.
Considering orthoquinones, such as adrenochrome, as diketones, it has used, for this purpose, specific ketone reagents, in this case more particularly amino reagents, represented by hydroxyl salts. - amine, semicarbazide, phenylhydrazine and various ni-trophenylhydrazines. All these reagents condense easily with adrenochrome, forming, by substitution of one of the quinone oxygen atoms, the corresponding monoxime, semicarbazone, phenylhydrazone and nitrophenylhydrazones. These condensation products, much less soluble than adrenochromic and easy to obtain, in the pure state, in the form of crystals, are in fact much more resistant to transformation or spontaneous decomposition than adrenochrome itself.
Their solutions, even very diluted in water or in other solvents, such as ethyl alcohol, are also very stable. Despite their relatively great chemical inertia, they retain, to a greater or lesser extent in each case, the precious hemostatic properties of adrenochrome solutions. This finding, essential from a therapeutic point of view, has been taken advantage of by the applicant for the preparation of stable hemostatic solutions capable of being stored in ampoules.
On clinical experimentation with the various derivatives mentioned, it has been found that the intensity and specificity of the haemostatic action differ rather unexpectedly from one product to another.

In haemostatic medication in humans, adrenochrome monosemicarbazone is particularly indicated. The vasoconstrictor effects which it produces are so similar to adrenohroma that it constitutes in a way a stabilized form, making the therapeutic applications of this goose practically possible.
The practical example below illustrates the application of the invention to this particularly interesting case of adrenochrome mono-semicarbazone, without however being limited to it.
