 <Desc/Clms Page number 5>

 <Desc/Clms Page number 6>

 EMI6.1

 <Desc/Clms Page number 7>

 <Desc/Clms Page number 9>

 <Desc/Clms Page number 10>

 EMI10.1

 <Desc/Clms Page number 11>

 <Desc/Clms Page number 12>

 <Desc/Clms Page number 13>

 EMI13.1

 <Desc/Clms Page number 14>

 Y is an aldehyde or acetal group. The esterifying group of the carboxylic acid ester can vary widely and can include alkyl, aryl, aralkyl, alkaryl and other groups, such as methyl, ethyl, propyl, phenyl, benzyl, toluyl; etc.

  Of these, lower alkyl groups are preferred. Similar variations are possible in the carbonamide groups. R2 can be virtually any substituted amine group such as pht @ alimido; N alkyl, for example N-methyl, etc; N aryl, for example N-phenyl, etc .; N-acyl for example NOCCH3, NOCC6 H5, etc .; N sulfonyl, for example NS 02 06 H5; N carbobenzoxy, for example NCOOCH2 06 H5; and the like. It will appear that R2 can be monosubstituted or can be bisubstituted by any group or combination of groups of the above type. These compounds can be made in a number of ways which will be indicated more fully below.

   It is therefore an object of the present invention to provide novel aldehyde compounds which are useful as intermediates particularly for the synthesis of amino acids. Another object is to provide new methods for producing these compounds. Yet another object of the present invention is to provide novel amino acid syntheses employing the novel aldehyde compounds described herein. These and other objects of the invention will become more fully apparent from the following description with particular reference to specific examples, which are to be understood as illustrative examples only and not as limiting the invention.

   These aldehyde compounds can be prepared in a number of ways. One method includes the following reactions:

 <Desc / Clms Page number 6>

 EMI6.1
 where R, R1 and R2 are retorted previously described in (1), with the following exceptions. R is preferably not H during the addition of acrylonitrile as some difficulty may be encountered with the addition of a single acrylonitrile molecule.

  During reduction; R and R1 cannot be CH since it is not possible to control the nitrile group which would be reduced. In this and other methods of making the aldehyde compounds, both R and R should preferably not both be amide groups in view of the reduced reactivity of these groups.

   The addition of acrylonitrile to. a typical example of this kind of reaction is acetanidomalonic ester.

  The acetamidomalonic ester is easily prepared according to the numerous syntheses described in the literature. Favorable conditions for the addition of acrylonitrile are given in the review by Albertson et al (Journal of American Chemical Society, Vol. 67, p. 2043-1945). Reduction of cyanobutyrate can be achieved by means of selective reduction in the presence of stannous chloride and hydrochloride.

 anhydrous gene. An example of this synthesis is the following: EXAMPLE 1
10.4 parts of anhydrous stannous chloride are suspended in 80 parts of anhydrous ether, and anhydrous hydrogen chloride gas is passed through the resulting mixture. After 2/5 hours, the formation of two liquid layers is noted.

   The bottom layer is very viscous. The resulting reaction mixture is stirred vigorously when a solution of ethyl alpha-carbethoxy-alpha-acetamido-gamma-cyanobutyrate chloroform (6.7 g of nitrile in 25 parts of chloroform) is added. Vigorous stirring is continued for four hours until the bottom, very viscous / layer is noticed not to solidify and stirring is continued for approximately 16 hours. However, the viscous bottom layer still persists. The reaction mixture can stand at room temperature for 5 days. The aldimine complex is resistant to crystallization.

  The volatile upper layer is removed by decantation, and the viscous lower layer is washed twice with anhydrous ether. The residual oily material is mixed with 100 parts of water and heated in a water bath to 70 C. The reaction mixture is neutralized to a pH of 6 by the addition of sodium hydrate and sodium carbonate. . A large, white precipitate forms rapidly which is essentially inorganic. The entire reaction mixture is extracted with chloroform and the resulting mixture is subjected to centrifugation and filtration. The chloroform layer is collected and is approximately water-clear in color. The chloroform solution is then dried over anhydrous sodium sulfate and after filtration, the chloroform is removed by evaporation in a vacuum.

   A very viscous (syrupy) residue remains, which partially crystallizes on reaction. Gamma butyraldehyde, gamma-dicarbethoxy-gam-

   hy my desired acetamido is isolated as phenylhy drazone according to conventional methods. Crude phenylhydrazone usually melts at 133-137 ° C. and after crystallization melts at 138-140 ° C. The phenylhydrazone of this desired aldehyde compound contains 11.4% nitrogen instead of the calculated 11.58% nitrogen.

   EXAMPLE 2
41.6 parts of anhydrous stannous chloride is suspended in 320 parts of anhydrous ether. Anhydrous hydrogen chloride is passed through the reaction mixture, until the formation of the two layers is noticed. The chloroform solution of the nitrile (26.8 parts nitrile in 125 parts chloroform) is added slowly over a period of 1 hour while the reaction mixture is stirred and hydrogen is continuously passed through. Chloride. After complete addition of the nitrile, hydrogen chloride is passed through for a period of four hours with vigorous stirring. The introduction of chlorinated hydrogen is stopped, and stirring is continued overnight.

   The following day, the stirring and the introduction of hydrogen chloride are continued for 18 hours, at which time a few crystals are noticed. The reaction mixture can then react for an additional period of five days, after this total reaction time of about a week, the aldimi- complex has formed a precipitate as a crystalline product. White. The crystalline aldimine complex is collected by filtration and washed with anhydrous ether. The product is dried in a vacuum and after drying weighs 65 parts.

   There is a slight odor of chlorinated hydrogen, fifteen parts of the above crystal complex are mixed with 100 parts of water and heated to 50 C. This results in a clear, aqueous solution which is extracted with chloroform, and the solvent. is removed by evaporation in a vacuum to produce an oil.

  A small portion of the residual oil is treated with

 <Desc / Clms Page number 9>

 phenylhydrazine in a conventional manner, and the resulting phenylhydrazone which precipitates from the alcoholic medium is collected by filtration and dried. It melts at 138-139 C and does not lower the melting point of the phenylhydrazone obtained in Example 1. (Further extractions of the aqueous solution with chloroform are carried out. The third and fourth extractions produce a crystalline residue. The crude residue melts on a wide temperature scale of 135-155 ° C. This crude crystalline product crystallizes like white needles from ethanol and melts at 181-182 ° C.).

     During the reduction, various modifications are possible. For example, other solvents such as dioxane, chloroform and other closely related organic solvents can be employed instead of ether. It is also possible to employ other stannous halides such as bromide, in which case the corresponding hydrogen halide is also to be employed similarly, time and temperature, concentration of reactants and the like may vary. .

   Another method for preparing the desired aldehyde compounds is represented by the following reaction:
 EMI9.1
 wherein R, R1 and R2 are as previously described in (1) with the following exceptions. R and R1 should not be COOH or H, since a free carboxyl group can intervene with the catalyst and since the addition of a single acrolein molecule is not very easily controlled where two active hydrogens are present. present.

 <Desc / Clms Page number 10>

 EMI10.1

  The reaction between ethyl acetamidomalonate and acrolein in the presence of any of a series of alkaline catalysts is a typical example of such reactions.

  The substituted malonate adds to the acrolein conjugate system to form aldehydes of the type defined above. This reaction is also applicable to alpha, beta-carboxyl unsaturated ether compounds in addition to aldehydes such as ketones; For example, methyl vinyl ketone, crotonaldehye, and the like, addition to acrolein is very simple to perform. Acetamidomalonate is added to an alcoholic solution containing a catalytic amount of sodium ethoxide.

  The resulting reaction mixture is cooled in an ice bath when the unsaturated alpha, beta-carboxyl compound is added dropwise. drop. After complete reaction, the catalyst is destroyed by the addition of glacial acetic acid.

  Aldehyde can now be isolated and characterized as simple-
 EMI10.2
 ment like phenylhydrazonej by adding phenyihydrazine to the mixture.

   The aldehyde can be isolated as a viscous liquid from the crude reaction mixture simply by removing the solvent by evaporation in a vacuum. Aldehyde isolated in this way readily forms phenylhydrazone which usually melts at 133-137 C. Putified phenylhydrazone melts at 138-140 C. This phenylhydrazone does not lower the melting point of the phenylhydrazone prepared from the resulting aldehyde. by the reduction previously described.

   EXAMPLE 3
100 parts of absolute ethyl alcohol are treated with 0.05 part of metallic sodium. When the sodium reaction is complete, 43.7 parts of ethyl acetamidomalonate are added. The resulting reaction mixture is a thick slurry and is cooled to 3 ° C in an ice bath. Then 12.9 parts of acrolein are introduced

 <Desc / Clms Page number 11>

 drip. After the addition of about 4 parts of the acrolein, the reaction temperature increases to 13 C.

  The introduction of acrolein is stopped, and the reaction temperature decreases to 8 C. The remainder of the acrolein is added at a rate such that the reaction temperature is maintained at 8-10 C. After complete addition. acrolein, the reaction mixture is stirred for an additional 30 minutes. The solution is clear and light brown in color. During the addition of acrolein, the solid material initially present has disappeared. This reaction appears to be carried out extremely gently. The clear, slightly colored solution is cooled in an ice bath for a further hour, while the catalyst is neutralized by the addition of 1.5 parts of glacial acetic acid dissolved in a small amount of ethanol.

   The pH of the reaction mixture at this point is between 4 and 6. The resulting mixture is placed in a container filled with ice overnight.

  After reacting overnight, the solution is filtered and the solvent is removed by evaporation in a vacuum. The residual oil is very clear and has a slightly brownish-yellow color. The residual oil is very viscous. The phenylhydrazone of the reaction product is prepared in a conventional manner. The crude phenyl hydrazone melts at 133-135 C. The phenyl hydrazone prepared from this aldehyde in the aforementioned manner does not lower the melting point of the phenyl hydrazone cited in the previous example where the aldehyde was produced by the reduction of nitrile.

   While gamma butyraldehyde, gamma-dicar = bethoxy-gamma-acetamido is prepared in the example above, it appears that all other substituted butyraldehydes falling within the general formula stated at the beginning of this request can be made by this method. Instead of sodium ethoxide, as a catalyst, other aikoxides, oxides

 <Desc / Clms Page number 12>

 and alkali metal hydroxides and various organic bases also such as quaternary ammonium hydroxides, piperidine and the like can be employed.

   Another method for preparing aldehyde compounds is represented by the following equation:
 EMI12.1
 in which R, R1 and R2 can be as previously described with the exception that R and R1 must not be free carboxyl groups. R5 is a lower aliphatic group such as methyl, ethyl, propyl, etc., and X is an alkali metal.

   EXAMPLE 4
To a solution of 2.88 g of sodium in 230 ml of absolute alcohol, 27.2 g of ethyl acetamidomatonate are added. The resulting reaction mixture is heated on a steam bath with stirring while 24.3 g of betak-bromopropionaldehyde acetal are added dropwise.

   The reaction mixture is refluxed for an additional period of time (1 to 3 hours) after the complete addition of the bro @ o compound. Most of the alcohol is removed by distillation, and the reaction mixture is subjected to. hydrolysis to produce the aldehyde compound which, like phenylhydrazone, melts at 130-139 c and does not lower the IC point (3 melting of the examples described previously.

   Another method for producing these aldehyde compounds .. is represented by the following equation:

 <Desc / Clms Page number 13>

 EMI13.1
 where R, R1 and R2 can be as described previously, with the exception that R and Rl must not be free carboxyl groups. The group, -CH = CHCH2CH2Hal, can be provided by compounds such as CH2 = CH-CH2CH2 B. or any compound containing the group described above, which on ozonolysis produces the desired aldehyde.

   EXAMPLE 5 A solution of 2.88 g of sodium in 250 ml of 100% ethanol, 27.2 g of ethyl acetamidomalonate are added. After stirring the resulting reaction mixture, 16.9 g of 4-bromobutene-1 are added dropwise, with stirring, after complete addition of the bromo compound, the reaction mixture is subjected to µ reflux for an additional period of 4-6 hours. Most of the alcohol is removed by distillation and the desired condensation product is extracted and purified. The purified product is subjected to ozonolysis and the resulting ozonyde is decomposed into the aldehyde. After isolation, the aldehyde compound is isolated as phenylhydrazone.

  This phenylhydrazone melts at the same temperature as the previously described phenylhydrazones.

   The above new aldehyde compounds readily undergo reactions such as the formation of phenylhydrazone, oxine, aldimine, semicarbozone, and analogs which are reactive compounds susceptible to

 <Desc / Clms Page number 14>

The crystals thus precipitated are collected by filtration.

 resulting reaction mixture is heated until reflux temperature is reached. The reflux temperature is maintained for a period of 7-14 hours in an oil bath. The reaction mixture is then cooled and neutralized by the gradual addition of 2.4 g of glacial acetic acid.

  As the addition of glacial acetic acid progresses, an abundant white precipitate is noticed. The reaction mixture is cooled in an ice bath and the precipitated product is collected by filtration. The crude dl-tryptophan thus obtained melts at 255-263 C (decomposition). Subsequent purification of the material increases the melting point to 270-280 C with decomposition.

   In the preparation of tryptophan, it is not necessary to isolate the aldehyde compound but it can be converted directly to phenylhydrazone in a crude reaction mixture. In the case where the aldehyde compound is prepared by addition of acrolein and the addition of acrolein is carried out in an alcoholic solution such as methyl alcohol, it is not even necessary to isolate the phenyl hydrazone. when the alcoholic solution is suitable for direct cycling. In this case, the first isolation can take place directly after cycling.
