 <Desc/Clms Page number 11>

 <Desc/Clms Page number 15>

Another main object (the present invention is to provide a method of applying immune bodies originating from the mammary glands of animals, in medicine and in breeding,
Another object of the present invention is (to provide it, as a new compound, a milk of high titer, rich in specific protective principle against any one of a wide series of antigens, or combinations of antigens. .

Yet another object of the present invention is to provide a method of isolating and separating antibodies or protective principles from milk originating from mammary glands stimulated by ungulates.

Other objects of the invention will become apparent from the following description.

In order to achieve the foregoing and other related objects, the present invention comprises the characteristics described in more detail below and repeated in the claims, the following description giving certain forms of non-limiting application of the invention, the principles of this invention. it can be applied in many other ways.

     Production of the protective principle
A very specific antibody in the milk of ungulates (especially cows, goats, sheep, etc.) is produced against any antigen by introducing such an antigen into the udder of the animal. A specific antibody in milk is produced by introducing the antigen into the teat ducts of the animal at any time. However, the highest antibody reaction occurs after the introduction of the antigen during the dry period of the animal and especially after a series of introductions, known as reinforcements, spaced over a period of time. time.

Although the amounts of antigen introduced, the frequency (time interval) and the number of back-up doses can vary widely, the highest antibody reaction results from the injection of a series of doses of increasing amounts over time.

 tation.

Thus, for example, the initial jtgeotions can be given about two to eight weeks before parturition.
Reinforcement injections, when given, may also be spaced at the convenience of the operator, except that
 EMI5.4
injections should be given frequently enough so that an antiphylactic reaction does not occur. For most species, this time is less than about 10 to 14 days. To avoid local irritation and congestion, it is usually preferred that back-up injections not be given more.

frequently than every day. The antigenic substances are suspended in a liquid medium, for injection, such as, for example, sterile physiological saline solution.

The injection is made into the udder duct system through the teat meatus and into the glandular reservoir. The udder can, if desired, be massaged in order to achieve better penetration of the antigen into the udder duct system.

worse. The protective principle appears in the milk approximately, two to two days after the initial injection, we can see that shortly after the particular antigen used, For example, streptococcus agalactica was noted in the milk on the tenth day after the inoculation of 'a cow give milk. the antigenic substances which are used in the. Examples of the present invention for the production of a protective principle are bacteria, viruses, proteins, mycotic molds and fungi, tissues, pollen, and the like which are antigens.

Examples of antigens are for bacteria: Salmonella pullorum ,. Salmonella typhi, Salmonella paratyphi, Staphylococcus aureus, staphylococcus albus, staphylococcus pyogenes, pneumococci, streptococci, etc; for viruses: Influenza Type A, chickenpox, smallpox, herpes simplex, etc; for proteins: egg albumin, mouse tissue, embryonic egg tissue, etc; for tissues: blood and semen. It should be understood that these materials are merely examples of the almost infinite number and varieties of antigenic substances against which antibodies or specific protective principles may be produced in the udder of ungulates.

The terms "antigenic substance" and "antigenic material" are used to denote materials which are antigenic in themselves and also non-antigenic materials but which act as antigens in the presence of adjuvants. Antigenic disease organizations include included in these expressions.

     EXAMPLE 1
The experimental subject was a jersey cow, 5 weeks before parturition. The antigen was dead Salmonella pullorum.

The initial dose of antigen was injected into the teat canal: 1 ml of antigen per quarter, containing about 5 trillion organisms. The first reinforcing dose was -Injected one week later, namely 1 ml of antigen per quarter, containing approximately 10 trillion organisms. The antibody or protective principle can be stored in pasteurized milk, condensed milk, dried milk and in gamma globulin isolated from milk. Pasteurization temperatures must be carefully controlled. Normal pasteurization (ie, at 1 F for 30 minutes) does not adversely affect the protective principle. Care must be taken to keep the temperature from rising for an appreciable period of time. For example, when pasteurizing at 155 ° F, it has been found that the protective principle in the milk falls in half. The cooling of pasteurized milk must be rapid in order to maintain the protective principle.

Pasteurization can be carried out by known "flash" or "holding" methods, the milk containing a protective principle can be condensed under a

temperatures to avoid destruction of the. protective principle. the desiccation can be carried out by the processes with ', ion or rollers under conditions suitably controlled in order to maintain the protective principle.

High temperatures are not in themselves disadvantageous to the protective principle, except when maintained for a period of minutes. This is how milk can be dried in a desiccator in which temperatures of 300 to 400 F are reached but the milk is only at these temperatures for a moment.

A proliferating virus can be delivered to the udder by a two-step operation. The first inoculation is done in the normal way. The second inoculation is then carried out in the milk resulting from the first, the concomitant tissue antigens being removed by dilution.

This sieving process is illustrated below.

   EXEMP XII
A cow was infused into the teat ducts with a Herpes Simplex virus suspended in the brains of mice. The cow was lactating and was milked regularly thereafter. The next day, the vahhe showed a positive Whiteside test. On the second day after inoculation, the milk was used for injection into the udder of a dry cow through the teat ducts.

The result was the transfer of the virus without the antigens from the mouse brains,
Application of the supporting principle
The antibody or protective principle, which is the product of the present invention, is of interest in a number of ways. 0 has found that the protective principle or antibody is absorbed into the system after milk from udder stimulas has been ingested, or administered, by protoclysis. The isolated and separate protective principle can be administered orally, rectally, parenterally or locally. The protective principle is of interest in the immunization and treatment of humans and animals.

 <Desc / Clms Page number 11>

The application of the protective principle of the present invention is illustrated in the following examples.

   EXAMPLE XITI
The experimental subjects were four adult males.

The protective principle was generally prepared according to the procedure of Example 1, the resulting milk containing the antibody against Salmonella pullorum bacteria. All subjects were negative on an agglutination test before ingestion of milk.

The subjects consumed about half a liter to one liter of the milk containing the protective principle. All of them absorbed the antibody from their digestive tract into their bloodstream, as shown by the agglutination test. One of the subjects consuming one liter of milk per day developed an agglutination titer in the blood, ten times more diluted.

   EXAMPLE XIV
The protective principle against Salmonella pullorum was also applied to poultry. The experimental subjects were six adult hens, all negative on the Salmonella antibody test. After feeding milk containing the protective principle, all the hens developed a strongly positive reaction.

   EXAMPLE XV
Another application of the protective principle of the present invention was made to chickens and took in the positive protection provided by the protective agent. 20-day-old chickens were divided into an experimental group of ten, and a control group of ten. The experimental chickens were fed with milk produced according to the present invention and having a high titer (1/1000) against Salmonella pullorum: Salmonella pullorum is a disease organism of poultry. The chickens in the control group were fed with milk which did not contain the specific antibody, during the first day of the experiment. both groups were injected into the heart with an infusion of live, cultured Salmonella pullorum.

' invention, a high titer fraction was isolated by cold precipitation with alcohol.

15 g of this fraction were administered in 50 M1 of water. by infusion into the rectum of a two week old calf. The anus was tied off with stitches. The calf was then bled and its serum showed, by positive agglutination against Salmonella pullorum, that absorption into the bloodstream had occurred.

Experiments with humans, calves and pigs show that with continued ingestion of milk containing the antibody, levels of the protective agent continue to increase in the blood. Experiments with humans and calves also show that the protective principle is absorbed from the rectum. The degree of absorption from the rectum is less than that from the part. system earlier. rectal absorption may, however, be used if oral administration is contraindicated. The material containing the antibody or the protective principle may be applied topically (anointing) in the treatment of local irritations due to allergy.

The Salmonella pullorum antigen was used in the experimental work described in the previous examples because it is relatively harmless to the experimental subject; it is readily available, easily identified, and its absorption in the

 sir to limit the inventive.

It is recognized that the reduced gamut globulin in one animal is antigenic under certain circumstances in other species. For example, gamma globulin from 1 che is antigenic in non-bovine species. There are opportunities, however, to use isolated gamma globulin from cows with a high protective principle or specific antibody parenterally in other species. For example, gamma globulin from cow's milk containing a protective principle against procine plague can be administered to pigs and pigs.

The potently protective gamma globulin produced according to the present invention has effective use in the treatment of other animals of the same species, in which it is not antigen; for example, separated gamma glubulin from cows is useful for calves or other livestock.

With the discovery that the protective principle or antibody can be absorbed from the digestive tract, and that it is possible to produce a specific antibody in milk against most, if not all, antigens by proper treatment of ungulates to stimulate The development of this antibody or protective agent has opened up new avenues for the prevention and treatment of diseases in humans and animals.

The initial phase is the production of a specific protective principle against a mixture of all antigens known to humans or lower animals, or the production of a specific protective principle, such as an antibody, against a particular antigen, for example, from Jacobean pollen.

Like the mammary glands, especially of the cow, will produce an antibody or a specific protective principle against any bacterial, virus, mycotic or protein antigens. etc., leit can be prepared with a useful protective principle against infectious disease organisms and other antigens of

 <Desc / Clms Page number 15>

hummins and lower animals. In particular, prepared milk containing specific antibodies can be produced under specific, although rare, diseases and other conditions which produce specific antigens.

For example, protection for those suffering from hay fever from ragweed pollen is permitted by the consumption of fruit having a high protective principle against this ragweed pollen.

As the literature indicates the antigenicity of tumors, when such an antigen is isolated and injected into the udder. a specific antibody can be produced against this antigen. When milk containing such a specific protective principle is inserted or otherwise administered, it can delay or even cause the complete involution of such a tumor.

Dried milk containing a specifically produced antibody or protective principle can be made into poultices in the treatment of contact allergies. Isolated gamma globulin from milk containing its protective principle can be moistened and similarly applied in the form of a poultice.

For example, a milk is produced containing an antibody against the toxicodendron antigen, and it is applied to the surface of the skin which has come into contact with the antigen, to neutralize the toxic principle.

An important application of the protective principle of the present invention in the field of animal treatment is the provision of protective colostrum for calves. Currently, it has been estimated that about 20 of the calves born alive were given the first few weeks of life. Their death is caused mainly by organisms infectious against such antibodies can be developed. Mixtures of antigens of all common infectious diseases which kill calves can be suitably prepared and injected into the udders of dry cows.

Colostrum from these stipulated udders is dried, packaged and thus made available for feeding to the calves immediately after birth, thereby giving the

 EMI16.5
In particular, the protective principle can be mixed with other dairy products, such as ice cream drinks, ice creams, sorbets, cheeses, etc.

In the treatment of naive animals and humans, parenteral administration of globulin range having a particular potent anti / infection antibody has been shown to be effective. The protective principle isolated from milk from stimulated udders, according to the invention, is useful for parenteral administration to calves, in order to bring the specific antibody to high levels in the blood immediately after administration. .
