   Ltinvention est relative à de nouveaux agents thérapeutiques utiles ainsi qu'à un procédé pour leur préparation ; elle concerne, plus spécialement, des compo- sés qui sont des régulateurs efficaces du système nerveux central. Ces composés sont particulièrement utiles com- me agents calmants et peuvent également être utilisés pour renforcer les effets d'anesthésiques, d'hypnotiques, de narcotiques et d'analgésiques. 



   Les composés suivant l'invention, sont les 
2-(1-naphtylamino)-oxazolines substituées ou non, leurs sels d'addition acides pharmaceutiquement tolérables et les N-(1-naphtyl)-N-(éthyl substituées en   /))-urées.   



   Ces derniers composés sont non seulement des produits intermédiaires pour la préparation   d'oxazolines,   mais sont également utiles comme calmants et comme agents ren forçant l'effet d'anesthésiques, d'hypnotiques, de narco- . tiques et d'analgésiques, comme expliqué avec plus de détails ci-après. 



   Les N-(l-naphtyl)-N'-(éthyl substituées en /3 )urées sont préparées par la réaction entre un isocyana te de 1-naphtyle et une alcoylamine avec certains substi- tuants, spécifiés plus loin, dans une position   /3 par   rapport au groupement amino. Suivant une variante, les produits sont préparés par la réaction entre une 1-naph- tylamine et un isocyanate d'alcoyle avec des substituants analogues dans une position ss par rapport au groupement isocyanate.

   Ainsi, il est possible, pour préparer la 
N-(1-naphtyl)-N'-ss -brométhyl-urée par la réaction entre l'isocyanate de 1-naphtyle et   le. /3   -brométhylamine ou par la réaction entre la 1-naphtylamine et   l'isocyanate   de

 ss-brométhyle, Dans l'un ou l'autre cas, les oxazolines, préparées selon l'invention, sont obtenues par l'élimina- tion du bromure d'hydrogène hors de la brométhyl urée.

   Pour la préparation des composés actifs d'urée conformément à l'invention, le brome peut être remplacé dans les réactions ci-dessus par d'autres halogènes tels que le chlore ou l'iode ou par des groupements sulfony- les alcoylés ou arylés, tels que le méthanesulfonyloxy ou le p-toluènesulfonyloxy.    La réaction entre l'isocyanate et l'amine, conformément à l'invention, a avantageusement lieu dans du benzène ou dans un solvant aromatique équivalent. Des solvants aliphatiques, plus particulièrement ceux qui sont   halogénés   et nitrés, sont également utiles. On peut ci- ter à titre d'exemples le chloroforme, le tétrachlorure de carbone, le bichlorure d'éthylène,   la     chlorobenzène,   le toluène, le xylène, le nitrobenzène et le   nitrotoluène.   



   La réaction entre une amine et un isocyanate a généralement lieu sans intervention de chaleur. En réa- lité, avec certains des composés plus réactifs, il vaut mieux effectuer la réaction dans un bain de glace. Tou- tefois, comme pour d'autres réactions de ce genre, l'évo- lution de la réaction est accélérée par un apport de cha- leur, Pour certains des réactifs les plus inactifs, il peut même être nécessaire de faire intervenir de la cha-

 leur. On a constaté que des températures de 0 à 110  sont utiles et que la plupart des amines et des isocyana- tes, utilisés selon l'invention, réagissent au-dessous de 50 . 



   La formation d'un sel dtaddition acide à l'oxa- zoline par l'élimination d'un acide à partir du composé urée a lieu, dans les meilleures conditions, dans une solution aqueuse soumise à un reflux, mais des alcools aliphatiques inférieurs, tels que le méthanol, l'éthanol, le propanol ou le butanol, peuvent être utilisés. 



   Bien que les deux réactions de synthèse aient lieu aisément à la pression atmosphérique, il peut être avantageux, pour certaines applications, de faire inter- venir une pression accrue. Des pressions aussi élevées qu'une centaine d'atmosphères peuvent être utilisées. 



   L'expérience suivante est citée à titre d'exem- ple de l'effet calmant obtenu avec les agents thérapeu- tiques obtenus selon l'invention. On injecte dans les veines de trois singes 1   mg/kg     d'un,   des composés préparés conformément à l'invention. Avant l'injection les singes s'opposaient vigoureusement à l'approche de   l'homme   et quand ils étaient attrapés essayaient de mordre et de griffer les personnes qui les tenaient. Par contre, peu de temps après l'injection, on constatait que les   animaux   se laissaient approcher et caresser sans difficulté. 



  Quand on les laissait seuls, ces animaux sommeillaient un peu mais, quand on s'en approchait ils réagissaient et acceptaient des nourritures. 



   Bien que les composés, obtenus selon l'inven- tion, ne paraissent pas avoir un effet anesthésique par eux-mêmes,ils renforcent l'effet des anesthésiques. Par exemple, on a constaté que des rats restaient anesthésiés

 pendant une heure par l'effet de 50 mg/kg de l'anesthé- sique formé par le succinate de 21-hydroxy-pregnanedione- sodium quand ce composé est injecté par voie intravei- neuse. Par contre, la durée est presque doublée quand on injecte dans les veines des rats 10 mg/kg d'un des composés obtenus selon l'invention, 20 minutes avant   l'in<   jection du succinate de   21-hydroxy-pregnanedione-sodium.   



  Quand on utilise seulement 10 mg/kg du même composé, les rats ne sont pas anesthésiés. 



   Chacun des types de composés selon l'invention, a ses avantages spécifiques. Par exemple, bien que les urées et les oxazolines soient thérapeutiquement actifs, les oxazolines produisent leur effet en moins de temps. 



  Par contre, les urées, bien qu'elles demandent un temps un peu long pour faire leur effet, sont moins toxiques et peuvent donc être administrées avec des doses plus, grandes. Ceci implique que les deux agents peuvent être administrés simultanément pour avoir à la fois un effet rapide et prolongé dans certains cas. 



   Les urées sont neutres et ne forment pas des   wels   d'addition acides. Les oxazolines, par contre, sont faiblement acides et forment des sels d'addition acides qui, à cause de leur solubilité dans l'eau, sont utilisables plus aisément pour la préparation de solu- tions aqueuses parentérales. 



   En ce qui concerne les oxazolines selon l'in- vention, on envisage à'inclure spécifiquement dans les limites selon l'invention les sels d'addition acides que ces composés forment avec des acides contenant un anion pharmaceutiquement tolérable. L'expression "anion phar-   maceutiquement   tolérable" a une signification bien   défi-   nie pour les spécialistes. Elle vise un anion non toxi- que d'un quelconque des acides simples utilisés couram-

 ment en thérapeutique pour neutraliser les agents médici- naux basiques quand le sel de ces agents doit être admi- nistré à des êtres humains. Ces acides comprennent, par exemple, l'acide chlorhydrique, bromhydrique, iodhydri- que, sulfurique, succinique, maléique, tartrique, citri- que ou glycolique et d'autres.

   L'activité pharmaceutique de la molécule est principalement une fonction du cation l'anion servant principalement à produire la neutralité électrique. 



   Les composés d'urée selon/l'invention, sont remarquables à cause de leur faible toxicité. On a dé- couvert, par exemple, qu'avec un de ceux-ci, le taux de mortalité TM 50 pour des souris ayant absorbé le produit par la bouche était d'environ 875 mg/kg et que le TM 50, après administration intrapéritonale, était dtenviron 800 mg/kg. Pour les rats le TM 50 après administration par la bouche, était de 930 mg/kg et, après administra-   tion intrapéritonale,   le TM 50 était de 660 mg/kg. Ce faible taux de mortalité rend possible l'administration de doses thérapeutiques des produits par voie buccale ou par injection sans que l'on s'approche du niveau de toxicité. 



   Les agents selon l'invention, peuvent être ad- ministrés seuls mais ils sont généralement administrés avec un support pharmaceutique choisi en tenant compte de la voie utilisée pour l'administration et la pratique pharmaceutique ordinaire. Par exemple, ils peuvent être administrés par la bouche sous forme de comprimés ou de cachets contenant des excipients tels que   l'amidon,   le sucre de lait, certains genres d'argiles, etc. Ils peuvent être administrés par la bouche sous forme d'eli- xirs ou de suspensions qui peuvent contenir des agents colorants et édulcorants. 



   Ils peuvent être injectés parentéralement et,

 pour cet usage, on peut les préparer sous forme de solu- tions aqueuses stérilisées contenant   d'autres   produits solubles tels qu'une solution saline ou du glucose en quotité suffisante pour rendre la solution isotonique. 



  Pour les injections intramusculaires, les compositions des produits obtenus conformément   à   l'invention peuvent être préparées dans un support huileux, tel que l'huile d'arachides ou de sésame. 



   Le médecin ou le vétérinaire détermine la dose convenant le mieux dans un cas particulier et, comme on peut s'y attendre, cette dose varie avec le produit par- ticulier choisi et l'effet que l'on veut obtenir.   On   constate généralement que, lorsque les produits sont ad- ministrés par la bouche, une plus grande quantité est nécessaire pour produire le même effet qu'une quantité plus petite administrée par injection. On a constaté que l'administration par injection de 0,5 à 250 mg de l'agent actif procure généralement l'effet désiré. 



   Les compositions selon l'invention, peuvent se présenter sous diverses formes. Différents diluants peuvent être utilisés et le pourcentage des ingrédients actifs peut varier. Il est nécessaire que l'ingrédient actif forme une proportion telle, de la composition, qu'une dose appropriée soit obtenue. Il est évident que plusieurs doses unitaires peuvent être administrées à peu près en même temps. Bien que des compositions con- tenant moins de 0,005 % en poids des ingrédients actifs puissent convenir, il est préférable d'utiliser des com- positions ne contenant pas moins de 0,005 % de l'agent actif car, sans cela, la quantité du support devient ex- cessivement grande.

   L'activité augmente avec la concen- tration de l'agent   actif..   Le pourcentage en poids de l'agent actif peut être   de   10,50, 75, 95 %   ou   même plus.

  Des doses unitaires peuvent être préparées, avec une pro- portion moindre d'un support et une proportion plus grande de matières actives et vice-versa. 



   Les exemples ci-dessous ne sont donnés qu'à ti- tre illustratif et ne limitent en rien l'objet de l'in- vention car bien des variantes sont possibles sans sor- tir du domaine de protection de celle-ci. 



   EXEMPLE I 
Préparation de la N-(1-naphtyl)-1'-ss -brométhyl urée. 



   Une solution à   50 %   d'hydroxyde de potassium (150 g) refroidie à la glace est agitée avec une suspen- sion de 123 g de bromhydrate de ss-brométhylamine dans   420   ml de benzène. La couche de benzène, contenant la base libre, est séparée et séchée sur une petite quanti- té d'hydroxyde de potassium. L'agent de séchage est en-      levé par filtration. A cette solution on ajoute, goutte à goutte et en refroidissant, une solution benzénique équimoléculaire d'isocyanate de 1-naphtyle. On laisse la solution au repos pendant une heure durant laquelle le produit se sépare sous forme d'une poudre blanche. Il est recristallisé hors d'acétate d'éthyle et fond à 146,8-147,2  
L'analyse du C13H13ON2Br donne par calcul C=53,26;   H=4,47;   N=9,56; Br=27,26.

   On trouve C=53,05;   H=4,48;   N=9,76; Br=27,11. 



     EXEMPLE   II Préparation de la   2-(1-naphtylamino)-oxazoline.   



   On fait bouillir un litre d'eau et on y ajoute 15 g de N-(1-nahtyl)-N'-ss -brométhyl urée. A la fin d'une période de reflux de 15 minutes toute la matière est dissoute à l'exception d'une très petite quantité. 



  Le flacon est refroidi dans un bain de glace et la petite

 quantité de matière insoluble, ainsi qu'une.quantité addi- tionnelle de matière qui se sépare-par le refroidissement, sont enlevées par filtration. Une solution à 14 % d'hy- droxyde d'ammonium (30 mls) est ajoutée lentement et il se forme un précipité gommeux. Le précipité est extrait avec trois portions de 250 ml de chloroforme et les ex- traits mélangés sont lavés avec de l'eau. La couche orga- nique est séchée sur du sulfate de sodium anhydre et con- centrée sous vide. Un résidu cristallisé rosâtre est for- mé et ce résidu est trituré avec de l'éther et recristal- lisé hors d'isopropanol. Le composé N-(1-naphtyl)-N'-ss -brométhyl urée est administré, par injection intraveineuse et à des doses de 1 à 5 mg/kg, à deux groupes de singes. 



   Après cette injection, les animaux se laissent caresser et on peut jouer avec eux à des degrés variables, Si on les laisse seuls, les signes somnolent d'une manière normale. Quand on s'en approche, ils ont conscience de la présence de la personne et acceptent et mangent de la nourriture. La durée de l'effet calmant.est d'environ deux à cinq heures. On constate que les animaux sont de- venus normaux à la fin d'une période de 24 heures. 



   Un effet analogue est obtenu quand le composé 2-(1'-naphtyl-amino)-oxazoline est administré d'une   maniés   re similaire mais le début de la période de tranquilisa- tion est plus rapide. 



   EXEMPLE XVIII

   The reaction between an amine and an isocyanate generally takes place without the intervention of heat.    Each of the types of compounds according to the invention has its specific advantages. For example, although ureas and oxazolines are therapeutically active, oxazolines produce their effect in less time.



  On the other hand, ureas, although they take a little long time to take effect, are less toxic and can therefore be administered in larger doses. This implies that the two agents can be administered simultaneously to have both a rapid and a prolonged effect in some cases.



   Ureas are neutral and do not form acid addition wels. Oxazolines, on the other hand, are weakly acidic and form acidic addition salts which, because of their solubility in water, are more readily usable in the preparation of parenteral aqueous solutions.



   With regard to the oxazolines according to the invention, it is intended to include specifically within the limits according to the invention the acid addition salts which these compounds form with acids containing a pharmaceutically tolerable anion. The term "pharmaceutically tolerable anion" has a well-defined meaning for those skilled in the art. It is aimed at a non-toxic anion of any of the simple acids commonly used.

   The physician or veterinarian will determine the most suitable dose for a particular case and, as might be expected, this dose will vary with the particular product chosen and the effect desired. It is generally found that when the products are administered by mouth a greater amount is required to produce the same effect than a smaller amount administered by injection. It has been found that administration by injection of 0.5 to 250 mg of the active agent generally provides the desired effect.



   The compositions according to the invention can be provided in various forms. Different diluents can be used and the percentage of active ingredients can vary. It is necessary that the active ingredient form such a proportion of the composition that an appropriate dose is obtained. It is obvious that several unit doses can be administered at about the same time.    An ice-cold 50% potassium hydroxide solution (150 g) is stirred with a suspension of 123 g of ss-bromethylamine hydrobromide in 420 ml of benzene. The benzene layer, containing the free base, is separated and dried over a small amount of potassium hydroxide. The drying agent is removed by filtration. To this solution is added, dropwise and with cooling, an equimolecular benzene solution of 1-naphthyl isocyanate. The solution is left to stand for one hour during which the product separates out in the form of a white powder. It is recrystallized from ethyl acetate and melts at 146.8-147.2
Analysis of C13H13ON2Br gives by calculation C = 53.26; H = 4.47; N = 9.56; Br = 27.26.

   We find C = 53.05; H = 4.48; N = 9.76; Br = 27.11.

   One liter of water is boiled and 15 g of N- (1-nahtyl) -N'-ss -bromethyl urea are added thereto. At the end of a 15 minute reflux period all material is dissolved except for a very small amount.

A pinkish crystalline residue is formed and this residue is triturated with ether and recrystallized from isopropanol.
