The present invention relates to a process for the preparation of novel antibiotic substances. The new antibiotics are prepared from strains of thermophilic actinomycetes isolated for the first time. Samples of the new strains have been deposited with the American Type Culture Collection * under the numbers ATCC 14760, ATCC 14761 and ATCC 14762.

The new strains of thermophilic aotinomycetes can be isolated from culture media by the usual bacteriological methods of isolating cultures. When these microorganisms are cultivated in a storage medium such as Tendler's and Burkholder's 1a medium ( Applied Miorobiology 394-399 (September 1961)], they show short, spiraling sporophores; when cultured, on the other hand, in a medium of the type described below, they show either single sports formations or% two spores. .

The spores are white in color; during aging, the coloring turns to cream, then to gray, and finally to black Color on the reverse side turning from yellow to gray, then black *.

Excenzymatic spectrum: caseinase, gelatinase, amylase, lipase.

The new antibiotic substances can be prepared by culturing microorganisms belonging to one of the strains of the group ATCC 14760, ATCC 14761 or Arec 14762 at a temperature of about 37-560 in a sterilized liquid culture medium containing assimilable sources of carbon @ and nitrogen, and separating, where appropriate, the substances from

The active substances can be isolated * by precipitation. According to one of the negatives in operation, the active substances can be isolated from the filtered or centrifuged oiling medium, or from thawed fractions of the frozen culture medium, by releasing them, for example by means of ammonium sulphate.

The antibiotic substances obtained by the process of the invention exert an oncostatic and oncolytic action in humans and animals and can therefore be used as antitumor agents.

The following example is intended to illustrate the manufacture, by culturing ATCC 14760, of anti-tumor preparations. @ Example
The culture medium is inoculated with a culture of the microorganism, then allowed to ferment for approximately 17 1/2 hours to 48 hours. With constant stirring, the filtered or centrifuged culture medium is almost completely frozen and then allowed to thaw to about 1/10 of the total volume, the thawed portion separated by decantation or filtration and then olarified by centrifugation. The supernatant solution is treated with about 30% by weight of ammonium sulfate.

The resulting precipitate is dissolved in physiological saline solution. We adjust to the desired titer, based on the activity

antibiotic against Staphylccocus aureus ATCC 13680 (determined by the disc titration method).

A preparation, adjusted for an inhibition zone of 4 mm (by the disc titration method) is sterilized.

Similar antitumor preparations can be obtained by culturing strains ATCC 14761 and ATCC 14762 in the same manner. No general toxic effects were observed.
