Abstract

Procédé pour distinguer des fragments d'ADN qui contiennent des faux appariements individuels des bases à partir de leurs homologues parfaitement appariés. Des régions à brin unique à l'intérieur d'un fragment duplex sont modifiées par un carbodiimide, qui réagit avec les résidus de guanine (G) et de thymine (T) non appariés dans l'ADN, Les molécules d'ADN duplex linéaire ne réagissent pas, alors que les molécules d'ADN avec des faux appariements individuels des bases réagissent de façon quantitative avec le carboiimide. A la suite de la réaction avec le carbodiimide, les molécules d'ADN sont fractionnées sur des gels de polyacrylamide de pourcentage élevé, de telle sorte que les fragments modifiés et non modifiés puissent etre distingués clairement.
