Abstract

"PROTEìNA". A presente invenção se relaciona a um método para produzir uma enzima variante glicolipídio aciltransferase compreendendo: (a) selecionar uma enzima original que é a enzima glicolipídio aciltransferase caracterizada pelo fato de que a enzima compreende um sequência de aminoácidos do motivo GDSX, em que X é um ou mais dos seguintes resíduos de aminoácido L, A, V, I, F, Y, H, Q, T, N, M ou S; (b) modificar um ou mais aminoácidos para produzir a variante glicolipídio aciltransferase; (c) testar a variante glicolipídio aciltransferase para atividade transferase, para, opcionalmente, atividade hidrolítica, em um substrato galactolipídio, e opcionalmente um substrato fosfolipídio e/ou opcionalmente um substrato triglicerídio; (d) selecionar uma enzima variante com uma atividade melhorada para galactolipídios comparada com a enzima original; e opcionalmente (e) preparar uma quantidade de uma enzima variante. A presente invenção também se refere a uma enzima variante lipídio aciltransferase caracterizada pelo fato de que a enzima compreende a seqüência de aminoácidos do motivo GDSX, em que X é um ou mais dos seguintes resíduos de aminoácido L, A, V, I, F, Y, H, Q, T, N, M ou S, e na qual a enzima variante compreende uma ou mais modificações de aminoácidos comparado com uma seqüência original em qualquer um ou mais dos resíduos de aminoácido definidos nos Conjunto 2, Conjunto 4, Conjunto 6 ou Conjunto 7.
