Abstract
XILANASES MODIFICADAS QUE EXIBEM EXPRESSãO MELHORADA. Uma enzima xilanase da Família 11 modificada compreendendo uma seqüência que introduz um sítio de glicosilação de consenso funcional é fornecida. Os exemplos não limitantes de sítios de glicosilação introduzidos incluem a mutação do aminoácido nas posições 34, 131, 180, 182, ou uma combinação destas, para uma asparagina. A posição de aminoácido indicada na xilanase da Família 11 é determinada a partir do alinhamento de seqüência da xilanase de interesse com aquele de uma seqüência de aminoá-cido da xilanase II de Trichoderma reesei.O sítio de glicosilação de consenso introduzido facilita a eficiência de expressão aumentada da xilanase modificada quando comparada á eficiência de expressão de uma xilanase correspondente a partir da qual a xilanase modificada foi derivada, usando cepas hospedeiras e condições de crescimento similares.
