Abstract

INCORPORAçãO SìTIO-ESPECìFICA DE AMINOáCIDOS ARTIFICIAIS CONTENDO áTOMOS PESADOS EM PROTEìNAS PARA DETERMINAçãO DE ESTRUTURA CRISTALINA. São providos sistemas de tradução e outras composições incluindo aminocil tRNA-sintetase ortogonais que carregam preferencialmente um tRNA ortogonal com um aminoácido iodado ou bromado. ácidos nucléicos codificando tais sintetases também são descritos, como o são métodos e kits para produzir proteínas incluindo aminoácidos contendo átomo pesado, p. ex., aminoácidos bromados ou iodados. Também são descritas métodos para determinar a estrutura de uma proteína, p. ex., uma proteína em que um átomo pesado tenha sido incorporado sítio-especificamente através do uso de um par ortogonal de tRNA/aminoacil tRNA-sintetase.
