Abstract
ORGANISMO RECOMBINANTE GENETICAMENTE MODIFICADO PARA CONVERTER SEMIALDEÍDO DE GLUTARATO OU 5-AMINOPENTANOATO EM UM MONÔMERO, POLÍMERO OU COPOLÍMERO COM 5 CARBONOS DO MESMO, MÉTODO PARA A PRODUÇÃO DE MONÔMEROS COM BASE EM 5 CARBONOS, POLÍMEROS OU CO-POLÍMEROS DOS MESMOS, MÉTODO PARA A SUPER PRODUÇÃO DE GLUTARATO A PARTIR DE SUBSTRATOS DE CARBONO RENOVÁVEIS, 1,5-PENTANODIOL Trata-se de hospedeiros recombinantes para a produção de poli(hidróxi alcanoatos) e de métodos de produção de poli(hidróxi alcanoatos) a partir de substratos de carbono renováveis. Também são apresentados determinados hospedeiros recombinantes que produzem substâncias químicas com 5 carbonos como o 5-amino-pentanoato (5AP), 5-hidróxi-valerato (5HV), glutarato e 1,5-pentanodiol (PDO). Uma modalidade confere um hospedeiro recombinante expressando um gene que codifica uma enzima heteróloga selecionada do grupo que consiste em uma poli(hidróxi alcanoato) sintetase e uma 5-hidróxi-valerato-CoA (5HV-CoA) transferase, em que o hospedeiro produz um polímero que contém 5-hidróxi-valetato. Preferencialmente, o hospedeiro expressa tanto uma poli(hidróxi alcanoato) sintetase quanto uma 5HV-CoA transferase. O hospedeiro pode ser procariítico ou eucariótico. Um hospedeiro procariótico preferencial é a E. coli. Os polímeros produzidos pelos hospedeiros recombinantes podem ser homopolímeros ou copolímeros de 5-hidróxi-valerato. Um copolímero preferencial é o poli(3-hidróxi-butirato-5-hidróxi-valerato).
