Abstract

Une protéine qui répond à tous les critères biologiques qui sont caractéristiques de l'inhibine a été isolée d'une source gonadique. La purification et la caractérisation d'inhibine et l'utilisation de la matière purifiée pour produire des anticorps, l'utilisation d'inhibine et dudit antisérum dans un dosage radio-dosage quantitatif, et les applications in vitro et in vivo d'anticorps d'inhibine et de l'inhibine, sont décrites. Il est fourni une protéine purifiée, l'inhibine, caractérisé en ce que : a. le poids moléculaire apparent tel que déterminé par SDS-PAGE est de 56.000 ~ 1.000 b. le point isoélectrique est compris entre 6.9 et 7.3 c. la protéine peut se lier spécifiquement à la concanavaline A - Sepharose d. la protéine est constituée de deux sous-unités, caractérisées en ce que: i. leurs poids moléculaires apparents comme déterminé par SDS-PAGE sont respectivement de 44.000 ~ 3.000 et 14.000 ~ 2.000. ii. le point isoélectrique de la sous-unité de poids moléculaire 44 000 est dans la plage de 6,0 à 7,0 iii. les séquences d'acides aminés N-terminales des deux sous-unités sont telles que décrites ici e. la protéine peut supprimer l'hormone folliculo-stimulante (FSH), mais pas l'hormone lutéinisante (LH), l'hormone stimulant la thyroïde ou la prolactine dans un système de dosage biologique in vitro f. la protéine peut être marquée à l'iode radioactif. Il est également prévu un procédé pour isoler et purifier l'inhibine de liquide folliculaire ovarien de mammifère, caractérisé par a. une ou plusieurs étapes de chromatographie par perméation de gel, b. une ou plusieurs étapes de chromatographie liquide haute performance en phase inversée ; c. une ou plusieurs étapes de préparation polyacrylamide d'électrophorèse sur gel  ; d. une élution électrophorétique de l'inhibine purifiée.
