Abstract
La présente invention concerne des vecteurs dérivés de rétrovirus dans lesquels la glycoprotéine d'enveloppe rétrovirale a été remplacée par la glycoprotéine G du virus de la stomatite vésiculaire, ainsi que l'utilisation de ces vecteurs dans le transfert des gènes exogènes dans les cellules d'une grande variété d'organismes non mammifères. On décrit également un procédé de production de vecteurs rétroviraux à titres élevés, procédé selon lequel on utilise une lignée cellulaire hôte, stable, de recombinaison qui abrite le vecteur rétroviral concerné sans la protéine d'enveloppe. On démarre la production des vecteurs rétroviraux à titres élevés en introduisant l'acide nucléique codant une protéine fonctionnelle associée à une membrane dans la lignée cellulaire. Les vecteurs selon la présente invention peuvent être concentrés par ultracentrifugation jusqu'à l'obtention de titres supérieurs à 10<9> cfu/ml qui sont notamment utilisés dans des études thérapeutiques des gènes humains, et peuvent aussi infecter des cellules, telles que les cellules du hamster et du poisson, qui résistent généralement à l'infection par des vecteurs contenant la protéine d'enveloppe rétrovirale.
