Abstract

Procédés pour la transformation de Pichia méthanolica, et molécules d'ADN utiles dans cette transformation. En présence de molécules d'ADN, les cellules de P. méthanolica sont exposées à un champ électrique pulsé dont l'intensité varie entre 2,5 et 4,5 kV/cm, moyennant une durée d'impulsion comprise entre 1 et 40 millisecondes, et les molécules d'ADN sont ainsi introduites dans les cellules. Ces molécules d'ADN peuvent comprendre une unité d'expression renfermant un promoteur de transcription de gène P. méthanolica lié de manière opérationnelle à un segment qui code un polypeptide ou une protéine présentant de l'intérêt. Les molécules d'ADN peuvent aussi coder un marqueur sélectionnable, par exemple le gène ADE2 de P. méthanolica. Il est possible d'utiliser les cellules ainsi transformées dans les systèmes de production servant à préparer les protéines qui ont une certaine importance commerciale.
