Abstract
L'invention concerne des canaux (14) microfluidiques à l'échelle embryonnaire conçus pour manipuler et mettre en place des embryons, permettant de mieux évaluer et traiter des embryons. Un écoulement fluidique est utilisé pour acheminer et mettre en place les embryons à l'intérieur de canaux microfluidiques; des géométries de canaux pouvant être utilisées pour placer des embryons dans des emplacements spécifiques. Les propriétés de surface et les propriétés de compliance (déformation) des embryons sont évaluées pour servir d'indicateur prévisionnel de la viabilité. Les canaux microfluidiques permettent les réglages précis de la pression de manière à diriger diverses évaluations à des forces sensiblement inférieures à celles qui pourraient endommager les embryons. Le procédé décrit dans l'invention permet également d'évaluer la distance et/ou les embryons qui circulent dans un canal microfluidique à un gradient de pression identique et qui fournissent des informations, les embryons sains se déplaçant plus lentement ou sur une distance plus courte étant donné qu'ils présentent un frottement plus important contre les parois du canal. Placés à une constriction (14a, 14b, 24, 26), les embryons sains semblent, en outre, se déformer moins que les embryons malades qui sont plus rapidement attirés dans une constriction. De plus, les embryons sains semblent reprendre plus facilement leur forme. Le fluide provenant des canaux microfluidiques est facilement récupéré en aval sans endommager l'environnement embryonnaire, ce qui permet une meilleure analyse chimique dudit fluide en comparaison avec les techniques de manipulation et d'échantillonnage manuelles classiques. Le retrait de la zone pellucide des embryons mammifères est effectué au fur et à mesure que les embryons sont acheminés à travers l'écoulement vers un emplacement précis; un agent lytique pouvant être retiré de la surface de l'embryon afin de compléter le retrait de la zone pellucide. Le retrait du cumulus est effectué à l'aide d'une série de constrictions permettant de couper le cumulus, puis des écoulements fluidiques sont appliqués de manière à retirer le cumulus coupé de l'embryon.
