Abstract

La présente invention concerne une technique permettant l'intégration d'éléments génétiques étrangers dans un chromosome bactérien sans recourir à une étape de clonage. Cette technique repose sur la présence du système de recombinase ?-Red Dans un hôte bactérien expert en recombinaison. Au moins deux éléments ou construits de recombinaison linéaire sont transformés ensemble dans l'hôte expert en recombinaison et sont assemblés dans l'orientation de correction dans le chromosome bactérien par recombinaison homologue. Des marqueurs sélectionnables utilisés la sélection de transformants sont excisés ensuite par l'action d'une recombinaison spécifique du site.
