Abstract

Des approches réductrices et correctives permettant de limiter le bruit parasite d'autofluorescence sont appliquées dans les systèmes analytiques reposant sur la mesure sensible de signaux fluorescents émis par des réseaux de sources de signaux fluorescents. Ces systèmes sont particulièrement utiles dans le séquençage basé sur la fluorescence par des systèmes d'intégration reposant sur des petits effectifs ou des molécules fluorescentes individuelles qui détectent l'intégration de nucléotides dans des réactions d'extension d'amorces. L'invention concerne des systèmes et des procédés permettant d'analyser des réseaux d'échantillons extrêmement multiplexés en utilisant des systèmes optiques de haute densité, fortement multiplexés, pour éclairer les réseaux d'échantillons de haute densité et/ou obtenir une détection et de préférence une détection confocale sans signaux émanant de ces réseaux de haute densité. Les systèmes et les procédés sont appliqués selon diverses opérations analytiques différentes, y compris l'analyse de réactions biologiques et biochimiques, y compris la synthèse d'acides nucléiques et la dérivation d'informations de séquences provenant de cette synthèse.
